The prospects for the success of malaria control depend, in part, on the basic reproductive number for malaria, R0. Here, we estimate R0 in a novel way for 121 African populations, and thereby increase the number of R0 estimates for malaria by an order of magnitude. The estimates range from around one to more than 3,000. We also consider malaria transmission and control in finite human populations, of size H. We show that classic formulas approximate the expected number of mosquitoes that could trace infection back to one mosquito after one parasite generation, Z0(H), but they overestimate the expected number of infected humans per infected human, R0(H). Heterogeneous biting increases R0 and, as we show, Z0(H), but we also show that it sometimes reduces R0(H); those who are bitten most both infect many vectors and absorb infectious bites. The large range of R0 estimates strongly supports the long-held notion that malaria control presents variable challenges across its transmission spectrum. In populations where R0 is highest, malaria control will require multiple, integrated methods that target those who are bitten most. Therefore, strategic planning for malaria control should consider R0, the spatial scale of transmission, human population density, and heterogeneous biting. 相似文献

A simple, visual representation of spatial aspects of malaria transmission in successive snap-shots in time, is presented. The spatial components of the simulation involve (i) the identification of mosquito vector breeding sites of defined shape and area, (ii) the identification of a zone of malaria transmission determined by the shapes and areas of the vector breeding sites and the distance from these sites that the mosquitoes disperse, (iii) a human population dispersed in relation to the malaria transmission zone, (iv) perimeters around each individual human within which his or her infection can be transmitted by the local vector mosquitoes. The intensity of transmission within a malaria transmission zone is given by a number which is the number of new cases of malaria that each existing case will distribute through the human population within the duration of an infection. The simulation has been used here to examine the effects of vaccination against malaria transmission. Different levels of vaccine coverage are represented under endemic and epidemic malaria. The consequences of full or partial coverage of a zone of malaria transmission are also examined. The results are numerically compatible with the predictions of previous simple mathematical simulations of malaria transmission and interventions. The present simulation allows the nature of malaria transmission and the effects of interventions to be communicated easily and directly to an audience. It could have practical value in discussions of malaria control strategies with health planners. 相似文献

Seasonal variation of adults and larvae of anophelines was studied during 2003 and 2004 in Roraima State, located in the Northern region of Brazilian Amazon. Species diversity increased with distance of capture to human dwellings. Greater diversity was found in extradomiciliary collections than in peridomiciliary or intradomiciliary. A significant association between Anopheles darlingi Root and An. albitarsis (s.l.) Arribálzaga (Diptera: Culicidae) breeding sites and the proximity to human dwellings was observed. Malaria Sporozoite Antigen Panel Assay (VectestTM Malaria) indicated An. albitarsis s.l. as one of the local vectors in the studied area. Social insect colonies are characterized by extensive interactions among individuals, exchanges that can also potentially transmit pathogens. The large majority of these social interactions in a honeybee colony result from food transfer among individuals. Since colony hunger is likely to have a significant influence on these interactions, we investigated its effect on the distribution of food within the colony. By pulsing two colonies having different amounts of stored food with a radioactive label, we found that a starved colony sent out a larger number of foragers, brought in more food, and stored more of it than the satiated colony. We also found that the food brought into a starved colony was distributed more uniformly within each age class than that in the satiated colony. The queen and the young individuals received the lowest exposure to the label even though the label entered different regions of the colony at the same rate. The satiation level of the colony did not influence the relative exposures of different age groups to the label but a higher amount of it was stored in the hungry colony. We discuss the significance of these results in terms of the role played by the organizational structure of the honeybee colony on the transmission dynamics of an infectious disease. 相似文献

ABSTRACT: BACKGROUND: A dramatic decline in the incidence of malaria due to Plasmodium falciparum infection in coastal East Africa has recently been reported to be paralleled (or even preceded) by an equally dramatic decline in malaria vector density, despite absence of organized vector control. As part of investigations into possible causes for the change in vector population density, the present study analysed the Anopheles gambiae s.l. sibling species composition in north-eastern Tanzania. METHODS: The study was in two parts. The first compared current species complex composition in freshly caught An. gambiae s.l. complex from three villages to the composition reported from previous studies carried out 2-4 decades ago in the same villages. The second took advantage of a sample of archived dried An. gambiae s.l. complex specimens collected regularly from a fourth study village since 2005. Both fresh and archived dried specimens were identified to sibling species of the An. gambiae s.l. complex by PCR. The same specimens were moreover examined for Plasmodium falciparum and Wuchereria bancrofti infection by PCR. RESULTS: As in earlier studies, An. gambiae s.s., Anopheles merus and Anopheles arabiensis were identified as sibling species found in the area. However, both study parts indicated a marked change in sibling species composition over time. From being by far the most abundant in the past An. gambiae s.s. was now the most rare, whereas An. arabiensis had changed from being the most rare to the most common. P. falciparum infection was rarely detected in the examined specimens (and only in An. arabiensis) whereas W. bancrofti infection was prevalent and detected in all three sibling species. CONCLUSION: The study indicates that a major shift in An. gambiae s.l. sibling species composition has taken place in the study area in recent years. Combined with the earlier reported decline in overall malaria vector density, the study suggests that this decline has been most marked for An. gambiae s.s., and least for An. arabiensis, leading to current predominance of the latter. Due to differences in biology and vectorial capacity of the An. gambiae s.l. complex the change in sibling species composition will have important implications for the epidemiology and control of malaria and lymphatic filariasis in the study area. 相似文献

A study was conducted in three villages in western Venezuela between February 1988 and October 1989 to determine the biting and resting activity of anophelines in relation to human habits, rainfall and fenitrothion spraying of houses. Mosquitoes were collected landing on humans inside and outside experimental huts throughout the night. Only three mosquitoes were found resting in the huts in the morning, but 2470 were collected resting on vegetation in the early morning. The collections yielded eleven species of anopheline, the most abundant being Anopheles nuneztovari, comprising over 75% of the total anophelines collected, followed by An.triannulatus, An.albitarsis s.l. and An.oswaldoi. The four most abundant species showed different diel patterns of biting. For An.nuneztovari the peak of activity was close to midnight indoors and outdoors, for An.triannulatus between 19.00 and 20.00 hours outdoors, for An.albitarsis mainly before midnight indoors and outdoors and for An.oswaldoi outdoors at 19.00 hours, with an additional smaller peak indoors at midnight. Most of the human population use bednets, go to bed before 22.00 hours and are therefore most exposed to mosquitoes that bite outdoors early in the night. Fenitrothion house-spraying failed to prevent large mosquito populations developing in the wet season, presumably because of their exophilic resting habits. The possible advantages of impregnation of existing bednets with pyrethroids, and provision of nets for people who do not have them, are discussed. 相似文献

The microcirculation system and its regulation at rest and after gradually growing physical activity in subjects living in different environmental regions was studied using laser Doppler flowmetry. Three main types of microcirculation were found. The activity of regulation mechanisms depended on the microcirculation type, residence region, and level of physical activity. Subjects living in relatively clean regions displayed stable regulation mechanisms irrespective of the level of activity. Subjects with the mesoemic type of microcirculation from regions with a high toxic contamination, high radiation background, or exposed to combined adverse factors, as well as subjects with the hypoemic type exposed to toxic and combined factors displayed a decreased contribution of VLF and LF oscillations (p < 0.05), and subjects with the hyperemic type from toxically contaminated regions displayed a decreased contribution of LF oscillations (p < 0.05). The results indicate that, under environmental pressure, regulatory mechanisms are exerted to the extent verging on adaptation failure. 相似文献

Despite considerable success of genome wide association (GWA) studies in identifying causal variants for many human diseases, their success in unraveling the genetic basis to complex diseases has been more mitigated. Pathogen population structure may impact upon the infectious phenotype, especially with the intense short-term selective pressure that drug treatment exerts on pathogens. Rigorous analysis that accounts for repeated measures and disentangles the influence of genetic and environmental factors must be performed. Attempts should be made to consider whether pathogen diversity will impact upon host genetic responses to infection.We analyzed the heritability of two Plasmodium falciparum phenotypes, the number of clinical malaria episodes (PFA) and the proportion of these episodes positive for gametocytes (Pfgam), in a family-based cohort followed for 19 years, during which time there were four successive drug treatment regimes, with documented appearance of drug resistance. Repeated measures and variance components analyses were performed with fixed environmental, additive genetic, intra-individual and maternal effects for each drug period. Whilst there was a significant additive genetic effect underlying PFA during the first drug period of study, this was lost in subsequent periods. There was no additive genetic effect for Pfgam. The intra-individual effect increased significantly in the chloroquine period.The loss of an additive genetic effect following novel drug treatment may result in significant loss of power to detect genes in a GWA study. Prior genetic analysis must be a pre-requisite for more detailed GWA studies. The temporal changes in the individual genetic and the intra-individual estimates are consistent with those expected if there were specific host-parasite interactions. The complex basis to the human response to malaria parasite infection likely includes dominance/epistatic genetic effects encompassed within the intra-individual variance component. Evaluating their role in influencing the outcome of infection through host genotype by parasite genotype interactions warrants research effort. 相似文献

The concentration of metallothionein (MT), a low-molecular-weight protein, is regulated by many factors, primarily metals (zinc, cadmium, copper), cytokines, glucocorticoides and free radicals. These factors are determined by such aspects of human biology as gender, pregnancy and age, as well as by environmental factors including the use of oral contraceptives and cigarette smoking, all which may affect MT levels in the body.The aim of this study was to investigate the influence of these biological and environmental factors on MT concentrations in erythrocyte lysate and in plasma.MT concentrations were determined by a two-step direct enzyme-linked immunosorbent assay. Evaluation of exposure to cigarette smoking was performed by checking cotinine levels in the plasma of subjects.The studies showed higher MT concentrations in both the erythrocyte lysate and plasma of women when compared to men. Furthermore, pregnancy causes an increase of MT concentration in plasma, while oral contraceptives cause an elevated concentration of MT in erythrocyte lysate. Age impacts plasma MT concentrations in men, whereas it does not affect concentrations of MT in erythrocyte lysate. 相似文献

Late-glacial vegetation changes were studied at Rieme, NW Belgium. Human occupation of this cover sand area occurred from the Final Palaeolithic onwards. The research area is situated on the northern side of a large cover sand ridge in an undulating landscape with small ridges and depressions. The past landscape was reconstructed using a multi-disciplinary approach, including geomorphological, sedimentological, loss-on-ignition, botanical (micro- and macrofossil) and zoological analyses. AMS 14C dating provided an accurate chronology for the sediments. Analyses were performed on three sequences located ~200?C300?m apart. Our study shows that during the B?lling (GI-1e) wet meadows developed on the sandy soils and groundwater levels increased probably as result of permafrost melting. Shallow pools formed in depressions. During the Older Dryas (GI-1d) shrubs with juniper, sea-buckthorn and willow developed. Many shallow depressions were overblown with sand and deposition of organic material almost ceased. In the early Aller?d (GI-1c) open birch woodlands developed. Due to the final melting of permafrost, groundwater levels rose further and ponds with floating-leaved open water vegetation developed. Large water level fluctuations occurred in one of the ponds. Accumulation of organic deposits ceased during the mid-Aller?d. Indirect evidence for human occupation during the Aller?d (GI-1c) was found in indications of burning of the reed-swamps in combination with the presence of large herbivores. Final Palaeolithic people probably used the northern side of the cover sand ridge as hunting area, while they settled their temporary (base) camps on the steep southern side along the extensive and deeper Moervaart lake. 相似文献

Genetic diversity provides Plasmodium falciparum with the potential capacity of avoiding the immune response, and possibly supporting the selection of drug or vaccine resistant parasites. These genetic characters play key roles in the selection of appropriate malaria control measures. Diverse clones of Plasmodium falciparum, often denoted as strains, has been documented, and the degree of genetic diversity supported by several kinds of PCR (polymerase chain reaction) assays. Many studies in different endemic regions with differences in their level of disease transmission have clarified the interactions between the parasite populations and malaria epidemiology. This paper describes recombination events of the malaria parasite life cycle that originate such genetic diversity in P. falciparum, reviewing different studies on this aspect and its implications in the immunity and development of control measures in regions with different degrees of endemicity. 相似文献

The state of microcirculation system and its regulation in peace and after growing physical activity is analyzed on people living in different ecological regions. There are three main types of microcirculation. The level of regulation mechanisms activity depends on the type of microcirculation, region of residence and the value of physical activity. People living in a relatively clean regions have stable mechanisms of regulation despite of the level of activity. Mesoemic type living in toxic radiation regions have reduced contribution of VLF and LF-fluctuation (p < 0.05); hypoemic type living in toxic regions have VLF and LF-fluctuation (p < 0.05); hyperemic type living in toxic regions have LF-fluctuation (p < 0.05). The severe acute respiratory syndrome (SARS) is affected by meteorological parameters such as temperature and humidity. It is also observed that people having asthma are at risk for SARS. Therefore, it is of interest to report the effect of environmental, economic, and health factors on the spread of CoVid-19. We used data reporting CoVid-19 cases from 24 cities in eight different countries for this analysis. Data was analyzed using multiple linear regressions between these parameters. Data shows that temperature has effects on CoVid-19. A one-degree rise in temperature causes a -0.19 decrease in CoVid-19 cases per million people (log natural value per million populations). The effect of humidity is not significant at a p value of 0.26. Moreover, one-unit increase in asthma and GDP cases per million people show 0.06 and 0.46 increases in CoVid-19 cases, respectively. 相似文献
