Distribution of endocrine cells in the human oesophagus was studied histochemically. Large amount of endocrine cells was discovered in terminal parts and excretory ducts of the cardial glands. Endocrine cells of the oesophageal cardial glands are represented, at least, by three types: argentaffine, argyrophil in the reactions of Grimelius and Sevier, Munger and argyrophil in the reaction of Grimelius only. The amount of argentaffine cells in the oesophageal cardial glands was observed to be 5 times as large as that of in the gastric cardial glands. The reason is evidently in functional difference of the oesophageal and gastric cardial glands. The endocrine cells were absent in the mucous glands of the oesophagus.  相似文献

Summary The cellular and subcellular distribution of radioactivity in the mouse thyroid gland different times (20 min — 8 hours) after intravenous administration of 3H-L-DOPA was studied by means of quantitative electron microscopic autoradiography.High concentrations of autoradiographic silver grains occur over parafollicular cells and adrenergic nerves while the labelling of follicular cells and lumina is low or absent and similar to the labelling of connective tissue cells at all observation times.Over the parafollicular cells high levels of radioactivity can be recorded already 20 min after administration of the labelled amino acid. The grain counts are highest at 1 hour and decrease then at 2.5 and 8 hours.The intracellular distribution of label is similar at all observation times; thus, the concentration of silver grains over the typical cytoplasmic granules of the parafollicular cells is 4–5 times higher compared to the concentration over the remainder of the cytoplasm and the nucleus.Treatment with a decarboxylase inhibitor prior to the injection of 3H-L-DOPA results in a low and uniform labelling of all thyroid cells. This finding, taken together with the observation that also pretreatment with reserpine abolishes the autoradiographic reaction over the cytoplasmic granules, gives strong support to the idea that the great majority of silver grains observed over parafollicular cells represents dopamine formed by decarboxylation of the labelled precursor.This study was supported by grant K71-12X-3352-01 from the Swedish Medical Research Council. The mouth, pharynx and oesophagus of Calicotyle are lined by syncytial epithelia, and there are numerous unicellular glands associated with the oesophagus. An infolding of unmodified external tegument lines the mouth cavity and is connected by discrete cytoplasmic processes to subjacent perikarya. It contains two types of secretory body and its luminal surface is invested with a finely filamentous coating. The pharynx and oesophagus are lined by irregularly-folded epithelia that are interconnected by a septate desmosome. Membranous inclusions distinguish the pharynx epithelium and there is a well developed basal lamina for insertion of the pharyngeal muscles. The oesophagus epithelium is perforated by the openings of the oesophageal glands. These lie in the surrounding parenchyma and produce a dense, membrane-bound secretion which is conveyed by duct-like extensions of the glands to the oesophagus lumen. The ducts are supported in places by microtubules and are anchored to the oesophageal epithelium by septate desmosomes. A septate desmosome also marks the junction between the epithelium and the gut caeca.  相似文献

The ultrastructure of the dorsal oesophageal gland ampulla and its relationship with the oesophagus, oesophageal ultrastructure, and control mechanisms in oesophageal activity were studied. Terminal ducts of the sub-ventral glands open through the oesophageal crown at the base of the buccal cavity. The terminal duct of the dorsal oesophageal gland running through the dorsal gutter opens to the exterior at the rim 'groove' of the buccal capsule. The posterior oesophageal region is clavate and the cuticle of the lumen folds to form outlet valves, 'valvulae'. An inconspicuous oesophago-intestinal valve (three lobes) connects oesophagus and intestine and is visualized in the open and shut position. In the female reproductive tract, with the exception of the uterus, the cells lie on a thick, irregular (convoluted) basal lamina. The apical plasma membrane of the uterus, and seminal receptacle, extend into the lumen by microvilli-like projections with which spermatozoa make intimate contact. The lumen of the uterus is filled with oocytes, fertilized and unfertilized. Testicular cells have two parts linked by a rachis. Spermatocytes are elongated with a large nucleus, distinct nuclear membrane, and many granules. The apical membrane of the rachis forms long microvilli-like projections with balloon-like tips. The amoeboid spermatozoa contain membrane specializations, a nucleus devoid of a membrane, and are enclosed by a pseudopodial-like extension.  相似文献

Summary Fucose-3H was injected into the cerebral ventricle of rats that were killed at several time intervals after injection. Semi-thin sections of the supraoptic nucleus and neurohypophysis were processed for radioautography and analysed quantitatively. Silver grains indicating the site of fucose-labeled glycoproteins were first located at the perinuclear region of the secretory neurons. The highest silver-grain density in these cells was observed at 2 h after injection, declining afterwards. Silver grains over the neurohypophysis were observed from 2 h on, reached a peak at 1 day after injection and decreased in the subsequent time intervals. The distributions of the silver grains over the neurohypophysis fitted Poissonian distributions and these were shown to be heterogeneous at the several time intervals. Pituicytes were not labeled. The percentage of silver grains over the Herring bodies increased with time. In rats deprived of water after fucose-3H injection there was a great increase in the release of labeled glycoproteins from the neurohypophysis. These results indicate that the glycoproteins synthesized by the secretory neurons of the hypothalamic nuclei are secreted in the neurohypophysis.  相似文献

Summary Light-microscopic autoradiography was used to localize the cellular sites for neutral amino acid uptake in submandibular and sublingual salivary gland epithelia. The vasculature of isolated glands was perfused for 3–5 min with either L-(3-3H)serine or L-(4-3H)phenylalanine and then fixed by perfusion with buffered glutaraldehyde. In the submandibular gland the small neutral amino acid L-serine and the aromatic amino acid L-phenylalanine were localized to central acinar cells, demilunar cells and ductal cells. In the sublingual gland silver grains associated with each of these tritiated amino acids were localized to central acinar and ductal cells. Perfusion of both submandibular and sublingual glands with unlabelled L-serine (25 mM) or L-phenylalanine (30 mM) resulted in a significant decrease in the silver grain density associated with each labelled amino acid. The absence of silver grains in the lumina of acinar and ductal cells and the presence of tight junctions near the apical surface of the epithelium strongly suggest that the initial uptake of these amino acids was mediated by basolateral plasma membrane carriers.  相似文献

Electron-microscopic examinations of the sturgeon gut were performed. Oesophageal goblet cells were abundant in the stratified epithelium. The ultrastructural features of the secretory granules of the oesophageal and intestinal goblet cells were quite similar to those of other vertebrates. Lobules of multilocular adipose tissue were observed in the deep tunica propriasubmucosa of the oesophagus, in close association with vasculature and large fibre bundles of myelinated and unmyelinated axons. Similarly composed nerve fibre bundles were observed in the cardiac stomach, too. The presence of myelinated axons is an unusual feature in the vertebrate enteric nervous system. Cardiac and fundic zones of the stomach showed an epithelium with columnar ciliated and non-ciliated cells, the latter equipped with fuzzy microvilli. Cells lining the tubular gastric proper glands were markedly granulated. Intestinal superficial epithelium was columnar and contained ciliated, as well as non-ciliated and goblet cells. In the tunica propria all over the intestine, the presence and ultrastructure of granulated cells was in addition described. Intraepithelial granulated leukocytes were seen throughout the alimentary canal. Various types of endocrine cells were seen both in the stomach and in the intestine, the size of their granules was measured and their ultrastructure described and compared to that of mammalian cell types.  相似文献

Anatomical, histological, and ultrastructural studies of the European beaver stomach revealed several unique morphological features. The prominent attribute of its gross morphology was the cardiogastric gland (CGG), located near the oesophageal entrance. Light microscopy showed that the CGG was formed by invaginations of the mucosa into the submucosa, which contained densely packed proper gastric glands comprised primarily of parietal and chief cells. Mucous neck cells represented <0.1% of cells in the CGG gastric glands and 22–32% of cells in the proper gastric glands of the mucosa lining the stomach lumen. These data suggest that chief cells in the CGG develop from undifferentiated cells that migrate through the gastric gland neck rather than from mucous neck cells. Classical chief cell formation (i.e., arising from mucous neck cells) occurred in the mucosa lining the stomach lumen, however. The muscularis around the CGG consisted primarily of skeletal muscle tissue. The cardiac region was rudimentary while the fundus/corpus and pyloric regions were equally developed. Another unusual feature of the beaver stomach was the presence of specific mucus with a thickness up to 950 µm (in frozen, unfixed sections) that coated the mucosa. Our observations suggest that the formation of this mucus is complex and includes the secretory granule accumulation in the cytoplasm of pit cells, the granule aggregation inside cells, and the incorporation of degenerating cells into the mucus.  相似文献

应用石蜡切片和扫描电镜技术对日本沼虾消化道进行了研究。结果表明,食道壁向腔内形成四个纵突,食道上皮由单层柱状细胞构成,上皮下的结缔组织中具有放射肌和皮肤腺,环肌层近于连续。食道和胃连结处的管腔背方具食道瓣,胃内具胃磨、滤器和滤沟等结构,胃的组织学结构中除无皮肤腺分布外与食道相似。中肠较长,约占整个消化道的717%,具一对中肠前盲囊。中肠上皮细胞大致有两种类型,基膜着色深,环肌层连续,纵肌成束分散排列。后肠为一短管,管腔呈迷路状,其中部为一球形膨大的直肠。后肠的组织学结构与前肠相似。  相似文献

Surface architecture of the buccal cavity and the surface organization of the luminal mucosa of the oesophagus, stomach, and intestine of the carnivorous fish M. flavolineatus from the Red Sea were studied by using SEM. The results revealed that M. flavolineatus has four kinds of teeth; curved-blunt, wedge-shaped, flattened crowns, molariform and papilliform. Three types of taste buds (type I, II and III) were recorded in the oropharyngeal cavity. It was observed that taste buds and teeth are co-located in the pharyngeal region. Characteristic patterns of microridges of the surface cells in the oral cavity and oesophagus were observed. Mucous cells are distributed in the lining of the mouth cavity, oesophagus, stomach, and intestine. Characteristic patterns of mucosal folds throughout the alimentary canal, concerning oesophagus, stomach, and intestine were revealed. Numerous gastric pits, which represents the emergence of the gastric glands, were recorded in the anterior and middle regions of the stomach. Complex patterns of the folds and mucous cells were recorded in the intestinal mucosa. These results were discussed with other teleost fishes.  相似文献

An anatomical study of the digestive tract of the channel catfish revealed that the oesophageal mucosa was longitudinally folded and that secondary folds were occasionally located on the primary longitudinal folds. The infoldings were more numerous near the stomach. The stratified squamous epithelium covering the folds was made up of a basal layer, large mucous cells and simple squamous cells on the surface. The epithelium on the side of the folds consisted primarily of mucous secreting cells. Taste buds were observed between mucous cells on the apical portion of the oesophageal folds and were more prevalent in the cranial part of the oesophagus. The remaining layers of the oesophagus were: a lamina propria-submucosa, tunica muscularis and adventitia or serosa.
The J-shaped stomach had two regions: a large sac-shaped region containing gastric glands and a smaller, nonglandular pyloric region. The large rugae of the stomach became gradually smaller near the pylorus. There was a well developed pyloric sphincter. The mucosa included a simple columnar epithelium, a lamina propria and adventitia or serosa.
The intestine could be differentiated into a thick ascending segment, a descending segment, a thin convoluted segment and a thicker terminal segment, the rectum. Many mucosal folds containing branched villi characterized the ascending segment of the intestine. The descending and convoluted segments contained fewer folds with shorter and less-branching villi and were smaller in diameter and thinner walled. Descending and convoluted segments were also mildly convoluted and accounted for 80% of the total length of the intestine. An intestinal valve with a sphincter marked the beginning of the rectum. There was an approximately four-fold increase in the thickness of the tunica muscularis of the terminal segment of the intestine.  相似文献

Using a specific polyclonal antibody raised against rat pancreatic phospholipase A2 (PLA2), we investigated the localization of the enzyme in the rat pancreas and stomach by light and electron microscopy. In the pancreas, the enzyme was localized in the acinar cells, whereas the pancreatic islets showed no immunoreaction. In the stomach, the PLA2 reactive with the anti-pancreatic PLA2 antibody was distributed exclusively in the gastric glands, but not in the gastric pits or the pyloric glands. On the section of the stomach subjected to immuno- and PAS-staining, immunopositive cells were not the PAS-positive cells located in the gastric pit and the neck region of the gastric gland. Immunopositive cells were present from the neck to the bottom of the gastric gland. Immunoelectron microscopic observation revealed that the immunogold-labeled cell had a highly-developed rough endoplasmic reticulum in the basal cytoplasm and characteristic zymogen granules in the apical cytoplasm. Taking into account the cell position in the gastric gland, the immunopositive cell could therefore be identified as a chief cell. Since no double stainability with PLA2 and PAS was observed in the same cell, it is suggested that PLA2 could be used cytochemically as a marker enzyme of the chief cell in the gastric gland at the light-microscopic level. From the immunoelectron microscopic findings, we believe that the PLA2 in the stomach is released into the lumen of the stomach by exocytosis and could function as a digestive enzyme in the alimentary tract, like the PLA2 secreted from the pancreas. Other possible roles of the PLA2 in the stomach are discussed.  相似文献

The gross anatomy and microanatomy of the digestive system of Ichthyophis supachaii were investigated. The microscopic structures of the digestive system are similar to those in other caecilians. Functional and developing teeth are present in adults. The tongue contains the genioglossus muscle. The digestive tract is elongated and consists of the mucosa, submucosa, muscularis and adventitia/serosa. The oesophagus contains longitudinal folds and lacks oesophageal glands. We report for the first time the caecilian gastric rugae and specific localization of oxynticopeptic cells in the anterior gastric region. The intestinal folds are exclusively present in the anterior intestinal region. The liver comprises 30–40 incomplete hepatic lobes, lying in an imbricate manner. Each lobe is enveloped by haematopoietic tissue that produces and delivers blood cells into sinusoids. Hepatic parenchyma is organized into anastomosing, two‐cell‐thick plates, having sinusoids at the basal domain and bile canaliculi at the apical domain of hepatocytes. Pigment cells are scattered inside sinusoids. The pancreas contains pancreatic acini interspersed with islets of Langerhans. The gallbladder proper is thin and continuous with the cystic duct wall. Neutral and carboxylated acid mucosubstances are secreted along the digestive tract, while sulphated mucosubstances are not produced by the stomach and anterior intestinal regions.  相似文献

Membranous organelles, acid glycoconjugates and lipids were characterized in the digestive tract mucosa of Hemisorubim platyrhynchos by cytochemistry techniques. Two types of mucous‐secreting cells were observed in the digestive tract epithelium: goblet cells in the oesophagus and intestine and epithelial cells in the stomach. These cells had a Golgi apparatus more developed than the other cell types. The cytochemical analysis revealed that secretory granules are reactive to acid glycoconjugates, varying in reaction intensity according to the region of the digestive tract. Acid glycoconjugate reactions were also observed in oesophageal epithelial cell microridges and in enterocyte microvilli. In the digestive tract, acid glycoconjugates act to protect the epithelial surface, increasing mucous viscosity, which facilitates the passage of food, prevents the binding of parasites and facilitates their removal. Through lipid staining, a coated membrane was observed around each secretory granule of the oesophageal and intestinal goblet cells, while gastric epithelial cells granules were fully reactive. Oxynticopeptic cells of the gastric glands showed lipid droplets in the cytoplasm and also in the mitochondrial matrix, which act as an energy reserve for these cells that have a high energy demand. Enterocytes showed a well‐developed smooth endoplasmic reticulum, especially in the apical region of the cell, being related to absorption and resynthesis of lipids.  相似文献

The ultrastructure of the amphidial, oesophageal and excretory glands of N. americanus is described. There are two amphidial glands, and each is attached to a lateral hypodermal cord. Anteriorly the glands become associated with the amphidial sense organs. The amphidial glands synthesize complex secretion granules which appear to release their contents into the sense organ. Secretions thus pass over the amphidial cilia and exit via the amphidial pore. It is suggested that the secretory activity of these glands is under direct nervous control. There are three oesophageal glands, and each synthesizes dense secretion granules. The secretions of the oesophageal glands are released into the lumen of the oesophagus and into the buccal capsule. The two excretory glands are ventral in position and connected to the tubular excretory system. These glands synthesize secretion granules of varying density. Secretions from the excretory glands may exit via the excretory pore, or pass back into the tubular excretory system, or both.  相似文献

The gut of adult sturgeon was examined. The oesophageal mucosa contained numerous caliciform cells, synthesizing both neutral and acidic glycoconjugates, the latter of the sialylated type. The deep tunica propria-submucosa contained lobules of multilocular adipose tissue, specially abundant during the cold season. The oesophageal tunica muscularis was made up of a large sheath of striated muscle fibres, arranged orthogonally to a thin, subserous smooth muscle layer. The siphon-shaped stomach showed a ciliated epithelium in cardiac and gastric proper gland zones, where tubular glands were present in the tunica propria. The columnar cells which composed the superficial epithelium and gastric pits were demonstrated to synthesize almost exclusively neutral glycoconjugates. Appendices pyloricae constituted a glandular body equipped with intestinal mucosa. The intestinal mucosa was organized in folds, containing numerous caliciform cells which synthesized neutral or acidic glycoconjugates, the latter either of the sialylated and sulphated type. The sulphoglycoconjugates were more abundant in the caliciform cells of the distal intestinal tracts. The tunica propria-submucosa of the spiral valve (medium intestine) contained lymphatic tissue and large lymphatic follicles. A muscularis mucosae was present only in the rectum, where in addition a peculiar granular cell type was present in the superficial tunica propria-submucosa, possibly related to defensive properties. The subserous connective tissue contained pancreatic lobules all along the stomach and intestine. The enteric nervous system showed some special aspects, the most intriguing of which was the presence of large, longitudinally oriented nerve bundles in the t. propria-submucosa of oesophagus and cardiac stomach. The nerve bundles contained, near unmyelinated nerves, some myelinated nerves, as well as neuronal bodies. Both these aspects are exceptional in vertebrates and obscure in their significance. The structural and histochemical aspects we here describe are in part different from those described for other fish. Some of these special features are possibly related with special functional roles, others require a deeper insight and different approaches to clarify them functionally.  相似文献

The histological characteristics of the digestive tract and the ultrastructure of mucosal cells of the stomach and intestine of rice field eel, Monopterus albus, are described to provide a basis for future studies on its digestive physiology. The digestive tract of the rice field eel is a long and coiled tube composed of four layers: mucosa, lamina propria‐submucosa, muscularis and serosa. The pharynx and oesophagus mucosa is lined with a stratified epithelium. The stomach includes the cardiac and pyloric portions and the fundus. Many gastric pits are formed by invaginations of the mucosal layer and tubular gastric glands formed by the columnar cells in the fundus. The intestine is separated from the stomach by a loop valve and divided into a proximal portion and a distal portion. The proximal intestinal epithelium consists of columnar cells with microvilli towards the lumen and goblet cells. The enterocytes are joined at the apical surface by the junctional complex, including the evident desmosomas. Numerous lysosomes and some vesicles are evident in the upper cytoplasm of the cells, and a moderate amount of endoplasmic reticulum and lysosomes are scattered in the supranuclear cytoplasm. The epithelium becomes progressively thicker and the folds containing large numbers of goblet cells are fewer and shorter in the distal portion of the intestine. At the ultrastuctural level, the columnar cells of the tubular gastric glands have numerous clear vacuoles and channels. A moderate amount of pepsinogen granules are present in the stomach. The enterocytes of the intestinal mucosa display a moderate amount of endoplasmic reticulum and lysosomes, and long and regular microvilli.  相似文献

An immunocytochemical technique using specific antiglucagon serum reveals the presence of glucagon-containing cells situated exclusively in the oxyntic glandular mucosa of the dog stomach. Electron microscope examination of the mucosa demonstrated endocrine cells containing secretory granules with a round dense core surrounded by a clear halo, indistinguishable from secretory granules of pancreatic A cells. Like the alpha granules of pancreatic A cells, the granules of these gastric endocrine cells exhibited a peripheral distribution of silver grains after Grimelius silver staining. Moreover, the granules of these cells were found to be specifically labeled with reaction product, using the peroxidase immunocytochemical technique at the ultrastructural level. Accordingly, these cells were named gastric A cells. These data suggest that the gastric oxyntic mucosa contains cells indistinguishable cytologically, cytochemically, and immunocytochemically from pancreatic A cells. It is believed that gastric A cells are responsible for the secretion of the gastric glucagon.  相似文献

The ontogeny of the digestive tract of the white seabream, Diplodus sargus during the larval development up to day 45 post-hatching (dph) has been studied using histological and histochemical techniques. The oesophageal goblet cells appeared around 6 dph and contained neutral and acid mucosubstances (PAS/diastase-PAS and Alcian Blue pH 2.5 positive reactions). An incipient stomach can be distinguished from 2 dph but the first sign of gastric gland development was detected around 13-15 dph, increasing in number and size by 22-23 dph. Gastric glands were concentrated in the cardiac stomach region and they had a high content of protein rich in tyrosine, arginine and tryptophan. Acidophilic supranuclear inclusions related to pynocitosis of proteins, were already observed in the intestinal cells of the posterior intestine around 4-6 dph (exogenous feeding) and they were present until 25 dph. The intestinal mucous cells appeared between 15-18 dph and contained a mixture of neutral and acid mucosubstances/glycoconjugates, carboxylated ones being more abundant than the sulphated ones. The stomach and gastric glands were fully developed by the first month of life marking the beginning of digestive features characteristic of the juvenile stage. Around 4-6 dph, glycogen, proteins and neutral lipids were observed in the granular cytoplasm of hepatocytes. Strongly acidophilic zymogen granules were also present, at this time, in the basophilic cytoplasm of the exocrine pancreatic acinar cells and contained abundant proteins, especially rich in arginine, tyrosine and tryptophan.  相似文献
