A new aerial alga, Stichococcus ampulliformis S. Handa sp. nov. (Trebouxiophyceae, Chlorophyta) is described based on a clone isolated from the bark of Cephalotaxus harringtonia (Knight ex Forbes) K. Koch collected from Taishaku‐kyo Gorge, Hiroshima Prefecture, south‐west Japan. This alga was examined by light microscopy and transmission electron microscopy and subjected to molecular phylogenetic analysis. Based on its morphological features and life‐cycle, especially short filament formation, the alga was assigned to the genus Stichococcus Nägeli. However, this alga differs from other described Stichococcus species in that it reproduces by a form of ‘budding’, producing two daughter cells of different sizes. The larger cell, enclosed within the mother cell wall, soon reaches the size of a normal vegetative cell. The smaller cell is extruded and takes longer to reach full vegetative size. A phylogenetic tree constructed using 18S rRNA sequences indicated that, within the Trebouxiophyceae, S. ampulliformis is closely related to S. bacillaris Nägeli and some species of Prasiola Meneghini. 相似文献

Summary Mitosis and cytokinesis have been studied in the green algaZygnema C. A. Agardh using interference-contrast light and transmission electron microscopy. At prophase, the nucleolus disintegrates and numerous extranuclear microtubules near the nuclear periphery penetrate into the nucleoplasm. When aligned in the equatorial plane of the open metaphase spindle the chromosomes are coated with persistent nucleolar fragments. At anaphase, vacuoles intrude into the interzonal spindle region and seemingly contribute to the anaphase movement of the chromosomes. At telophase, the spindle is persistent and the reforming nuclei are separated by cytoplasmic strands containing microtubules, interspersed with vacuoles. Extensive bundles of microtubules, dictyosomes and parallel, slightly inflated ER-profiles extend from the poles of the telophase nucleus along the longitudinal side of the chloroplast. Conceivably, these microtubules guide the nucleus during its post-mitotic migration towards its central interphase position between the two halves of the dividing chloroplast. Throughout the mitotic cycle, ubiquitous dictyosomes, positioned near the chloroplast core, seem very active. Arrays of microtubules run towards these dictyosomes and may conduct the dictyosome-vesicles to the cleavage plane. At metaphase, septum growth becomes visible as an annular ingrowth of the plasmalemma. At late telophase or at entering interphase, an extensive clump of vesicles, associated with longitudinal bundles of microtubules, appears between the leading edges of the advanced furrow. Apparent fusion of these vesicles with the head of the centripetally-growing furrow results in its completion. Members of the cosmopolitan streptophycean genus Klebsormidium live in various habitats, including sand dunes and polar/alpine environments. To survive in these harsh conditions they must possess an array of adaptive physiological and structural mechanisms, for example, to deal with chilling and photochilling stresses. Since these mechanisms have not been studied in detail, the objectives of this study were (i) to determine the physiological and biochemical responses of Klebsormidium cf. flaccidum (K. cf. flaccidum) to chilling (low temperature [LT]) and photochilling (LT in combination with high light [HL]) stresses; and (ii) to understand the cross-link between biochemical parameters and cellular ultrastructural changes. The results indicated that 5°C is a temperature threshold (i.e., at 5°C) but not at higher temperatures, physiological changes were observed (Fv/Fm and ETR decreased and energy-partitioning distribution changed, with an increase in Y[NPQ] under LT and an increase in Y[NO] under HL-LT). Also, pigment contents changed significantly, with increased concentrations of photoprotective pigments such as antheraxanthin, zeaxanthin, and total carotenes. All of these responses occurred under LT and, to a greater extent, under LT-HL, indicating that the two stresses (temperature and light) are additive. The cold treatment applied here induced the formation of spores under both LL and HL. The degree of photoinhibition was higher in spores than in vegetative cells, indicating that spores are less susceptible to photodamage. This study demonstrated a broad acclimation potential in different developmental stages of K. cf. flaccidum, which helps to explain the ecological success of this genus. 相似文献

Morphological and molecular phylogenetic studies were carried out on a freshwater green alga from Myanmar. Most specimens exhibited a gross morphology similar to Enteromorpha, however, their thalli were basically monostromatic and chloroplasts were axile and stellate. In addition, the phylogenetic analysis inferred from the 18S rRNA gene strongly supported a monophyly with Prasiola japonica and a more distant relationship with other chlorophytan taxa, including Enteromorpha. From our results, it has been shown that this alga belongs to Prasiola and has a close phylogenetic relationship with P. japonica. 相似文献

Physiological responses of Dunaliella salina and Dunaliella viridis, isolated from solar saltworks on the Yucatan Peninsula, were studied. Optimal growth temperature for D. salina was 22 degrees C (3.06 x 10(6) cells mL(-1)) and 26 degrees C for D. viridis (4.04 x 10(6)cells mL(-1)). Total carotenoid content in D. salina increased with temperature to a maximum of 35.14 pg cell(-1) at 38 degrees C. Dunaliella salina alpha-carotene and beta-carotene content was 0.083+/-0.003 and 0.598+/-0.020 mg 100g dry wt(-1) respectively, whereas lower values were found in D. viridis cultured under same experimental conditions (0.018+/-0.002 and 0.136+/-0.012 mg 100g dry wt(-1) respectively). The highest specific growth rate in D. salina was obtained at 10% NaCl (0.28 d(-1)), while its cell volume increased from 524 to 2066.93 microm(3) when cultured from 10% to 35% NaCl. Maximum photosynthetic rates were attained when increasing from optimal growing temperature to 30 degrees C for D. viridis (108 n mol O(2)microg chl alpha h(-1)) and D. salina (139 n mol O(2)microg chl alpha h(-1)). Photosynthetic responses to temperature variations indicated physiological adjustments in both species, with higher acclimation in D. salina. Evaluation of physiological attributes of these species will be used for to carry out mass cultivation. 相似文献

This paper describes methods of semi-mass culture and the quantification of algal color sprayed on a concrete board of the alga Klebsormidium flaccidum (Küitzing) S. Mattox et Blackwell. The alga was cultured in 100 L tanks normally used for hatching brine shrimp, with aeration from the deepest parts of the slanting sides. Initially mean algal density increased rapidly and attained 9.1 times the original density after 4 weeks. Subsequently, increase of the algal density was slow, and the density became 16.1 times the original level after 8 weeks. The alga cultured in this study was sprayed on to a concrete board using an agricultural sprayer. The relationship between the algal density on the board and its color, expressed as L*a*b* color space, was quantified. The chromaticity was maximum as the algal density on the board was about 5 g/m2, and the color was most vivid. When the algal density was more than 20 g/m2, both the lightness and chromaticity changed slightly, and the color became dark green. 相似文献

One of the most significant processes in the life history of an alga is the colonization of a new substratum. In the present study, we evaluate whether different organic compounds, such as agar, gelatine, chicken albumin, glycerine and polylysine, promote zoospore recruitment and germination in a periphytic, fresh-water green microalga of the genus Pseudulvella(Chlorophyta). Given the low adhesion capacity of its zoospores a series of experiments were conducted in order to find a substance and its optimal concentration that increases zoospore recruitment and allows us to follow the processes of settlement, attachment and germination of zoospores. Polylysine significantly increased the number of zoospores attached with no significant effect on the germination rate. The minimum effective concentration of polylysine for improving zoospore settlement was 0.1%. % 相似文献

Summary The endomembrane system of the chlamydomonad flagellate,Gloeomonas kupfferi (Chlorophyta), is complex. It consists of a proliferating ER network, a perinuclear complex of 14–18 dictyosomes and 8–12 vacuoles and an anterior contractile vacuole complex. The ER network extends from the nuclear envelope outwards, ensheafhs a dictyosome, extends out through a lobe of the chloroplast and terminates in the thin zone of peripheral cytoplasm between the chloroplast and plasmamembrane. The individual dictyosome is polar with distinct cis- and trans-faces. The cis-face is closely associated with transition vesicles emerging from the adjacent ER. Large vesicles emerge from peripheral swellings of terminal cisternae. The dictyosome-associated ER is connected to the peripheral vacuolar system. During cell division and cytokinesis, changes in the endomembrane system occur. Dictyosomes divide and quickly separate to form perinuclear complexes around the daughter nuclei. Each dictyosome undergoes morphological changes during this wall precursor-producing stage. ER lines the furrow zone and is closely associated with phycoplast microtubules. Bulbochaete allahabadensis sp. nov. is described from a temporary pond at Allahabad. Most of its life-cycle stages have been observed. It differs from other known species of the genus Bulbochaete in having characteristic oospores and interesting behaviour of androspores. Andropores after liberation from normal vegetative cells may germinate anywhere and form dwarf males or produce several celled branched germlings with precocious formation of antheridia. Such zoospore-like behaviour of androspores confirms the observations made by other workers under culture conditions. 相似文献

Helicosporidium sp. is a unique, achlorophyllous green alga that has been reported to infect various insect orders, including Lepidoptera, Diptera, and Coleoptera. The infectious cyst stage is ingested by the host, ruptures in the midgut lumen, and releases a filamentous cell. Histopathological examinations using larvae of a susceptible noctuid host, Spodoptera exigua, showed both cysts and filamentous cells affiliated with the microvillar lining of the midgut epithelium. A considerable proportion of the ingested cysts (22-39%) were recovered in feces collected 24 h after ingestion. A small number of filamentous cells passed the midgut epithelium and entered the hemocoel within 4-24 h after cyst ingestion. After 48 h, vegetative cell stages were detected in the hemolymph, followed by a 4- to 5-day period of increasing multiplication. Cyst differentiation in the colonized hemolymph began 6-7 days after the treatment. 相似文献

Members of the genus Klebsormidium have cosmopolitan distribution and occur in a very wide range of freshwater and terrestrial habitats. Due to its simple filamentous morphology, this genus represents a taxonomically and systematically complex taxon in which phylogenetic relationships are still poorly understood. The phylogeny of Klebsormidium and closely related taxa was investigated using new ITS rRNA and rbcL sequences generated from 75 strains (isolated from field samples or obtained from culture collections). These sequences were analyzed both as single-marker datasets and in a concatenated dataset. Seven main superclades were observed in the analyses, which included sixteen well-supported clades. Some species of Klebsormidium, including the type species Klebsormidium flaccidum, were polyphyletic. Interfilum was recovered with high statistical support as sister taxon to a clade of Klebsormidium formed mainly by strains identified as K. flaccidum. Whereas some clades could be easily associated with described species, this was not possible for other clades. A new lineage of Klebsormidium, isolated from arid soils in southern Africa and comprising undescribed species, was discovered. Several morphological characters traditionally used for taxonomic purposes were found to have no phylogenetic significance and in some cases showed intra-clade variation. The capacity to form packet-like aggregates (typical of Interfilum), features of the morphology of the chloroplast and the type of habitat were the main phylogenetically relevant characters. Overall, Klebsormidium and Interfilum formed a more diverse algal group than was previously appreciated, with some lineages apparently undergoing active evolutionary radiation; in these lineages the genetic variation observed did not match the morphological and ecological diversity. 相似文献

Summary Mitosis in a mite-pathogenic species ofNeozygites (Zygomycetes: Entomophthorales) was investigated by indirect immunofluorescence microscopy using an antibody against -tubulin for visualization of microtubules (MTs). DAPI and rhodamine-conjugated phalloidin were used to stain chromatin and actin, respectively. Salient features of mitosis inNeozygites sp. are (1) a strong tendency for mitotic synchrony in any given cell, (2) conical protrusions at the poles of metaphase and anaphase nuclei revealed by actin staining, (3) absence of astral and other cytoplasmic MTs, (4) a spindle that occupies most of the nuclear volume at metaphase, (5) a spindle that remains symmetrical throughout most of mitosis, (6) kinetochore MTs that shorten during anaphase A, (7) a central spindle that elongates during anaphase B, pushing the daughter nuclei into the cell apices, and (8) interpolar MTs that continue to elongate even after separation of the daughter nuclei. Cortical cytoplasmic MTs are present in a few interphasic and post-cytokinetic cells. The data presented show thatNeozygites possesses features unique to this genus and support the erection of theNeozygitaceae as a separate family in theEntomophthorales.Abbreviations DAPI

The effects of salinity on growth, maturation and photosynthesis were examined in the filamentous alga Rhizoclonium sp. (Cladophoraceae, Chlorophyta) growing in a brackish water habitat in a canal draining into Tokyo Bay, Japan. In this habitat Rhizoclonium sp. was exposed to a wide salinity range, both daily, 5–23‰ during November 1996, and hourly, 6–24‰ during the spring tide day. From the results of culture experiments, growth and maturation of Rhizoclonium sp. occurred in the wide salinity range of 10–40‰ at 20 μmol photons m‐2s‐1 at 20°C, but did not occur at salinity of 0‰. Light saturation on the photosynthesis‐irradiance curve at 20°C at 20‰ was reached at 100 μmol photons m‐2s‐1, which is characteristic for shade‐adapted algae. On the photosynthesis‐salinity curve at 20°C at saturated irradiance (160 μmol photons m‐2s‐1), the net photosynthetic rate increased with increasing salinity up to 30‰ but decreased at 40‰. On the photosynthesis–salinity curve at 20°C at 20 μmol photons m‐2s‐1 (at near in situ irradiance), the photosynthetic rates were almost the same in the salinity range from 0 to 40‰. Therefore, this species is able to grow, reproduce and photosynthesize with a relative efficiency in a wide salinity range, which shows that it is well adapted to a brackish water environment. 相似文献

The free-living nematodes Eumonhystera borealis n. sp., Sphaerolaimus occidentalis n. sp., and S. gracilis de Man 1876 from Bothnian Bay in the northern Baltic Sea are described and illustrated. Eumonhystera borealis n. sp. differs from other species by its small body size (314-393 μm), narrow body (a = 37-49), and large anterior amphids. In Sphaerolaimus occidentalis n. sp. the amphids are posterior to the buccal cavity, and it differs from other similar species by having two sclerotized rings in the posterior part of the buccal cavity. An intersex is reported for S. gracilis. Sphaerolaimus gracilis is cannibalistic or a predator of other species, with a preference for E. borealis n. sp. Sphaerolaimus occidentalis n. sp. coexists with S. gracilis at depths of 80 m but not at 12 m. 相似文献

A new heterotrophic flagellate Wobblia lunata gen. et sp. nov. is described. This organism usually attaches to the substratum showing a wobbling motion, and sometimes glides on the substratum or swims freely in the medium. W. lunata has various features characteristic of the stramenopiles. These include a hairy flagellum with tripartite tubular hairs, a mitochondrion with tubular cristae, arrangement of flagellar apparatus components and a double helix in the flagellar transition zone. W. lunata shares a double helix with heterotrophic stramenopiles, including Developayella elegans, oomycetes, hyphochytrids, opalinids and proteromonads, and could be placed in the phylum Bigyra Cavalier-Smith. However, from 18S rDNA tree analysis, these organisms form two distantly-related clades in the stramenopiles, and Wobblia appears at the base of the stramenopiles. Evaluation of morphological features and comparison of 18S rDNA sequences indicate that W. lunata is a member of the stramenopiles, but it is distinct from any other stramenopiles so far described. Its phylogenetic position within the stramenopiles is uncertain and therefore W. lunata is described as a stramenopile incertae sedis. 相似文献
