Stem rot was recorded as one of serious diseases of red‐fleshed dragon fruit, (Hylocereus polyrhizus), in Malaysia. Fusarium fujikuroi was recovered from stem rot lesion of H. polyrhizus and the species was identified using TEF1‐α sequence and mating study. From maximum likelihood phylogenetic tree using combined TEF1‐α and β‐tubulin sequences, the F. fujikuroi isolates from stem rot were grouped according to three geographical locations, namely Peninsular Malaysia, Sabah and Sarawak. Phylogenetic analysis indicated that F. fujikuroi isolates from stem rot of H. polyrhizus were clustered separately from F. fujikuroi isolates from rice because of intraspecific variation. From amplification of MAT allele‐specific primers, 20% of the isolates carried MAT‐1 allele while 80% carried MAT‐2 allele. From isolates that carried MAT‐1 allele, 65% crossed‐fertile with MP‐C (mating population of F. fujikuroi) tester strain while for MAT‐2 allele, 56% crossed‐fertile with MP‐C. None of the isolates were identified as MP‐D (mating population of F. proliferatum). Pathogenicity test conducted on 40 representative isolates showed that the stem rot symptoms were similar with the symptoms observed in the field, and can be categorized as low, moderate and high aggressiveness, which indicated variation in pathogenicity and virulence among the isolates. This study provides novel findings regarding Fusarium species associated with stem rot of H. polyrhizus and indicated that F. fujikuroi as a new causal pathogen of the disease.  相似文献

Red carotenoid colours in birds are widely assumed to be sexually selected quality indicators, but this rests on a very incomplete understanding of genetic mechanisms and honesty‐mediating costs. Recent progress was made by the implication of the gene CYP2J19 as an avian carotenoid ketolase, catalysing the synthesis of red C4‐ketocarotenoids from yellow dietary precursors, and potentially a major mechanism behind red coloration in birds. Here, we investigate the role of CYP2J19 in the spectacular colour diversification of African weaverbirds (Ploceidae), represented by five genera and 16 species: eight red, seven yellow and one without carotenoid coloration. All species had a single copy of CYP2J19, unlike the duplication found in the zebra finch, with high expression in the retina, confirming its function in colouring red oil droplets. Expression was weak or undetected in skin and follicles of pigment‐depositing feather buds, as well as in beaks and tarsi, including those of the red‐billed quelea. In contrast, the hepatic (liver) expression of CYP2J19 was consistently higher (>14‐fold) in seven species with C4‐ketocarotenoid coloration than in species without (including one red species), an association strongly supported by a phylogenetic comparative analysis. The results suggest a critical role of the candidate ketolase, CYP2J19, in the evolution of red C4‐ketocarotenoid colour variation in ploceids. As ancestral state reconstruction suggests that ketocarotenoid coloration has evolved twice in this group (once in Euplectes and once in the Quelea/Foudia clade), we argue that while CYP2J19 has retained its ancestral role in the retina, it has likely been co‐opted for red coloration independently in the two lineages, via increased hepatic expression.  相似文献

植物倍性混合居群的形成和维系常伴随着明显的基因组遗传及表观遗传变异。利用AFLP和MSAP两种分子标记探讨了中华猕猴桃复合体(Actinidia chinensis)倍性混合居群的遗传变异和结构及其基因组甲基化变异方式。结果表明, 该倍性混合居群具有较高的遗传和表观遗传多样性, 但两者之间没有明显的相关性。种群的遗传多样性与海拔呈显著的负相关(P<0.05), 但表观遗传多样性与海拔不具显著相关性。AMOVA分析显示, 主要的遗传和表观遗传分化出现在倍性小种内部(97.65% vs 99.84%, P<0.05); 同时, AFLP邻接聚类分析显示二者存在一定程度的倍性相关性, MSAP分析则未显示有明显的倍性相关性。进一步研究发现, 中华猕猴桃居群的总甲基化程度为24.86%, 且多倍体具有更多的甲基化位点变异。该研究结果为深入探讨猕猴桃倍性混合居群的形成和维系机制奠定了基础。  相似文献

Engineering fluorescent proteins (FPs) to emit light at longer wavelengths is a significant focus in the development of the next generation of fluorescent biomarkers, as far‐red light penetrates tissue with minimal absorption, allowing better imaging inside of biological hosts. Structure‐guided design and directed evolution have led to the discovery of red FPs with significant bathochromic shifts to their emission. Here, we present the crystal structure of one of the most bathochromically shifted FPs reported to date, AQ143, a nine‐point mutant of aeCP597, a chromoprotein from Actinia equina. The 2.19 Å resolution structure reveals several important chromophore interactions that contribute to the protein's far‐red emission and shows dual occupancy of the green and red chromophores.  相似文献

For nearly a century, Alizarin red S (alizarin sodium sulfonate) has been used by morphologists to stain calcified bone matrix. More recently, its traditional use has frequently been replaced by more modern techniques; however, its auto fluorescent property continues to contribute to research well beyond the context of bone development and regeneration. The purpose of this study is to describe detailed methods that can be used to capture the autofluorescence of Alizarin red‐stained mineralized tissues in juvenile zebrafish. These methods allow for in situ analyses of minute skeletal elements, such as pharyngeal teeth, and preclude the need for dissection.  相似文献

Flowers of the kiwifruit species Actinidia chinensis produce a mixture of sesquiterpenes derived from farnesyl diphosphate (FDP) and monoterpenes derived from geranyl diphosphate (GDP). The tertiary sesquiterpene alcohol (E)-nerolidol was the major emitted volatile detected by headspace analysis. Contrastingly, in solvent extracts of the flowers, unusually high amounts of (E,E)-farnesol were observed, as well as lesser amounts of (E)-nerolidol, various farnesol and farnesal isomers, and linalool. Using a genomics-based approach, a single gene (AcNES1) was identified in an A. chinensis expressed sequence tag library that had significant homology to known floral terpene synthase enzymes. In vitro characterization of recombinant AcNES1 revealed it was an enzyme that could catalyse the conversion of FDP and GDP to the respective (E)-nerolidol and linalool terpene alcohols. Enantiomeric analysis of both AcNES1 products in vitro and floral terpenes in planta showed that (S)-(E)-nerolidol was the predominant enantiomer. Real-time PCR analysis indicated peak expression of AcNES1 correlated with peak (E)-nerolidol, but not linalool accumulation in flowers. This result, together with subcellular protein localization to the cytoplasm, indicated that AcNES1 was acting as a (S)-(E)-nerolidol synthase in A. chinensis flowers. The synthesis of high (E,E)-farnesol levels appears to compete for the available pool of FDP utilized by AcNES1 for sesquiterpene biosynthesis and hence strongly influences the accumulation and emission of (E)-nerolidol in A. chinensis flowers.  相似文献

以山梨猕猴桃(Actinidia rufa(Siebold and Zuccarini)Planchon ex Miquel)品种‘63101’和中华猕猴桃(Actinidia chinensis var.chinensis C. F. Liang)品种‘磨山雄7号’杂交产生的400余株F1代个体为材料,连续2~3年对其果实品质和外观特点的9个性状进行测定,分析它们的遗传规律。结果显示,子代群体单果质量连续3年低于双亲中值,遗传传递力连续3年低于100%,优势率连续3年均为负值;可溶性固形物子代中值除2015年略低外,其余2年均高于亲中值12.3%;优势率均为正值,遗传传递力均大于100%;子代果实形状出现分离,与母本圆柱形相同的比例占82%;果皮毛被中,偏离母本的有毛比例为60%;果皮颜色出现绿色、绿褐色、褐色的分离,比例为1∶1.5∶1;果实肩部形状多与母本的方形相似,占比为63%;果肉颜色也多与母本的绿色相似,占比为86%;果实风味中,与父本和母本相似的表型占比均为32%,口感酸的占比18%,超低亲极酸占比1%,超高亲浓甜占比9%。说明杂交子代单果质量和可溶...  相似文献

In many bird species with asynchronous hatching, smaller, later‐hatched nestlings are out‐competed for food by their larger, earlier‐hatched siblings and therefore suffer increased mortality via starvation. It is thought that female birds can either maintain or reduce the survival disadvantage of later‐hatched nestlings by differentially allocating maternal resources across the eggs of a clutch. Carotenoid pigments are an example of resources that female birds allocate differentially when producing a clutch, but laying sequence patterns for these pigments remain poorly studied in North American songbirds. We examined intraclutch variation in yolk carotenoids and egg metrics in 27 full clutches of red‐winged blackbird Agelaius phoeniceus eggs collected from eight wetlands in central Alberta, Canada. We predicted that carotenoids would decrease across the laying sequence, as in this species, later‐hatched, marginal nestlings suffer greater mortality than earlier‐hatched, core nestlings. We found nine carotenoid pigments in red‐winged blackbird egg yolks, including two that have never been described from avian yolks: α‐doradexanthin and adonirubin. As predicted, concentrations and amounts of most carotenoids decreased across the laying sequence, suggesting that female red‐winged blackbirds depleted their carotenoid resources as they laid more eggs. However, egg mass and yolk mass both increased across the laying sequence, suggesting that female red‐winged blackbirds may use other maternal resources to compensate for the size and survival disadvantage experienced by later‐hatched, marginal nestlings.  相似文献

Recently, a consensus has been reached that using lithium metal as an anode in rechargeable Li‐ion batteries is the best way to obtain the high energy density necessary to power electronic devices. Challenges remain, however, with respect to controlling dendritic Li growth on these electrodes, enhancing compatibility with carbonate‐based electrolytes, and forming a stable solid–electrolyte interface layer. Herein, a groundbreaking solution to these challenges consisting in the preparation of a Li2TiO3 (LT) layer that can be used to cover Li electrodes via a simple and scalable fabrication method, is suggested. Not only does this LT layer impede direct contact between electrode and electrolyte, thus avoiding side reactions, but it assists and expedites Li‐ion flux in batteries, thus suppressing Li dendrite growth. Other effects of the LT layer on electrochemical performance are investigated by scanning electron microscopy, electrochemical impedance spectroscopy, and galvanostatic intermittent titration technique analyses. Notably, LT layer‐incorporating Li cells comprising high‐capacity/voltage cathodes with reasonably high mass loading (LiNi0.8Co0.1Mn0.1O2, LiNi0.5Mn1.5O4, and LiMn2O4) show highly stable cycling performance in a carbonate‐based electrolyte. Therefore, it is believed that the approach based on the LT layer can boost the realization of high energy density lithium metal batteries and next‐generation batteries.  相似文献

Background and Aims

Some otherwise promising selections of Actinidia chinensis (kiwifruit) have fruit that are too small for successful commercialization. We have therefore made the first detailed study in diploid kiwifruit of the effects of chromosome doubling induced by colchicine on fruit size, shape and crop loading.

Methods

Flow cytometric analysis of young leaves and chromosome analysis of flower buds and root tips was used to confirm the stability of induced autotetraploids. Fruit weight, size and crop load were measured in the third year after planting in the field and for three consecutive years. DNA fingerprinting was used to confirm the origin of the material.

Key Results

There was a very significant increase in fruit size in induced autotetraploids of different genotypes of A. chinensis. With the commercially important diploid cultivar ‘Hort16A’, most regenerants, Type A plants, had fruit which were much the same shape as fruit of the diploid but, at the same fruit load, were much larger and heavier. Some regenerants, Type B plants, produced fruit similar to ‘fasciated’ fruit. Fruit of the autotetraploids induced from three female red-fleshed A. chinensis selections were also 50–60 % larger than fruit of their diploid progenitors. The main increase in fruit dimensions was in their diameters. These improved fruit characteristics were stable over several seasons.

Conclusions

Chromosome doubling has been shown to increase significantly fruit size in autotetraploid A. chinensis, highlighting the considerable potential of this technique to produce new cultivars with fruit of adequate size. Other variants with differently shaped fruit were also produced but the genetic basis of this variation remains to be elucidated. Autoploids of other Actinidia species with commercial potential may also show improved fruit characteristics, opening up many new possibilities for commercial development.  相似文献

The Actinidia chinensis complex, a group of commercially important fruits (kiwifruit), is a complex of functionally dioecious lianas of variable ploidy. To understand the cytogeography better and to facilitate breeding in this complex, we examined the ploidy and morphological variations in 16 natural populations of A. chinensis var. chinensis and A. chinensis var. deliciosa across an ecogeographical gradient. Four ploidy levels were found, var. chinensis consisting of diploids and tetraploids and var. deliciosa consisting of tetraploids, pentaploids and hexaploids. Hexaploids were centred in the western Yun‐Gui plateau, tetraploids coexisted with hexaploids or diploids in the middle Yun‐Gui plateau and the Wuling‐Xuefeng mountains, and diploids occurred in the eastern Wuling‐Xuefeng mountains and the Hunan foothills. These findings indicate a gradual, clinal transition from hexaploid to diploid across the elevational and longitudinal gradient. The clear geographical segregation of diploids and hexaploids may have arisen from their differential ecological adaptation in response to altitude and climate, whereas the coexistence of cytotypes (2x–4x, 4x–6x and 4x–5x–6x) might be a result of reproductive barriers, with a particular contribution from the postzygotic reproductive isolation between ploidy races. The geographical pattern and morphological variation of cytotypes suggest a hybrid zone between the varieties in the Wuling‐Xuefeng mountains. The differences in cytotypes which have arisen as a result of ecological adaptation, distribution and morphological characteristics will provide important baseline data for the selection of germplasm and the breeding of kiwifruit. © 2010 The Linnean Society of London, Botanical Journal of the Linnean Society, 2010, 164 , 72–83.  相似文献
