Golden hamsters with alimentary hyperlipoproteinaemia (fed on a diet in which 44% of the energy was accounted for by fats, 52 mg cholesterol (CH)/100 g) were given apple pulp (10% of the diet) for six months. From the end of the first month up to the end of the experiment, elevation of serum CH, triacylglycerol (TG) and phospholipid (PL) levels and CH and TG accumulation in the liver were effectively inhibited. At the close of the experiment, hyperlipoproteinaemia was characterized by an increase in the amount of very low density lipoproteins (VLDL), which carried 60-80% of all circulating lipids, accounted for over 70% of the total lipoprotein pool and transported almost 60% of the serum CH (high density lipoproteins--HDL--only 16%). The apple diet markedly reduced (by 50-80%) the amount of all lipids in the lipoproteins within density limits of 1,006-1,063 kg/m3 and lowered the total lipid concentration in these density classes and in the lipoprotein pool as a whole (by 50%). The proportion of the individual lipoprotein classes in the lipoprotein pool was not significantly affected. The apple diet did not affect either the chemical composition of HDL or their participation in the lipoprotein pool, but raised the proportion of serum CH transported in this lipoprotein class. Neither the lipoprotein lipase activity of the adipose tissue nor lecithin:cholesterol acyltransferase activity were influenced by the apple diet.  相似文献

After excision of shell from Pomacea canaliculata, survival rates, repair process, haemocyte response, and calcium content were observed and quantified for three weeks. The following experimental treatments were used: black-colored individuals with large (BL-group) or small (BS-group) shell heights, yellow-colored individuals with large (YL-group) or small (YS-group) shell heights. The survival rates in BL-, YL-, BS-, and YS-groups after making partial excisions of snail shells were, respectively, 93.59, 94.87, 92.31, and 96.15%. Freshly regenerated shell could be seen at 1 day among the four groups after induction of shell regeneration. Regeneration shell filled the wound after 5–10 days. The area of regenerated shell in yellow-colored individuals was larger than that of black-colored individuals, but the thickness of regenerated shell was not significantly different. Multiple (four times) inductions of shell regeneration significantly enhanced the thickness of newly formed shells. Two types of circulating haemocytes (hyalinocytes and granulocytes) were identified under light microscopy based on Wright’s staining. Artificial induction of shell regeneration rendered a significant increase in the number of total circulating haemocytes which was followed by a decrease to pre-wounding levels. This peak value of total circulating haemocytes was 2.86, 2.43, 1.69, and 1.71-fold higher than pre-wounding levels in group BL, YL, BS, and YS, respectively. The number of total circulating haemocytes in yellow-colored individuals was significantly higher than in black-colored. Calcium concentrations in the mantles of snails are favorable for calcium nucleation. It is postulated that calcium content increased in the mantle increase at the start of the experiment and then returned to pre-wounding levels at the end of the experiment. This study shows that apple snails have effective shell regeneration abilities and that haemocytes and calcium transportation appear to play an important role in shell growth and regeneration.  相似文献

Tomatine was present in cultured excised tomato roots but in lower concentrations than in seedling radicles of the same age. The alkaloid was not detected in 'spent' root medium. Newly-initiated callus cultures of hypocotyl, radicle and cotyledon origin produced roots, and tomatine was isolated from both roots and callus. Roots contained more tomatine than callus, but neither contained as much as the organ explants from which the cultures were initiated. The number of roots produced decreased with time, as did also the tomatine content of the callus tissues. After 447 days, when no organized structures were produced by callus cultures, tomatine was not detected. An established hypocotyl callus contained small amounts of tomatine when grown on certain nutrient media, but a chlorophyllous sub-isolate of this callus did not produce detectable quantities of the alkaloid. Tomatine was not detected in an established root callus isolate or in suspension cultures initiated from established, tomatine-containing hypocotyl callus.  相似文献

以高蛋白小麦品种“北农9549”为试材,研究喷施不同浓度脯氨酸(0、1.0、5.0和10.0 mmol·L-1)对镉胁迫下小麦幼苗生长和重金属吸收的影响.结果表明: 以不施镉为对照,1.0 mmol·L-1CdCl2胁迫下,小麦幼苗的根长、株高和干质量分别显著下降24.0%、15.0%和27.5%,叶绿素a、b和类胡萝卜素含量分别显著下降23.3%、6.7%和30.8%,超氧化物歧化酶(SOD)活性降低了18.4%,内源脯氨酸、抗坏血酸和丙二醛(MDA)含量分别显著上升78.6%、31.5%和17.9%,细胞膜相对透性显著升高24.8%,过氧化物酶(POD)活性为对照的2.4倍,并且促进对铜的吸收,抑制锌的吸收.随外源脯氨酸浓度的增加,小麦幼苗的根长、株高、干质量、叶绿素和类胡萝卜素含量均逐渐恢复到对照水平,抗坏血酸、内源游离脯氨酸含量和SOD活性均上升,可溶性蛋白含量先上升后下降,POD活性、MDA含量和细胞膜相对透性下降,而锌积累量升高,镉、铜积累量下降.叶面喷施外源脯氨酸可缓解镉对小麦幼苗生长的胁迫,以喷施5.0~10.0 mmol·L-1外源脯氨酸效果最佳.  相似文献

The relationship between toluene concentration and the rate of leakage of solutes from toluene-treated roots and leaves of Sorghum bicolor, L. Moench, was studied to determine the effect of toluene on plant cell membranes. A threshold concentration of 0.2% toluene was needed to induce leakage. Maximal leakage rates were obtained with 0.5% toluene. Low molecular weight solutes, such as amino acids, sugars, and inorganic ions, leaked from treated tissue, while macromolecules, such as protein were retained. The rates at which the low molecular weight solutes diffused from treated cells decreased with increasing molecular weight. At 25°C, treatment of roots and leaves with 0.5% toluene resulted in the quasi-quantitative leakage of solutes within 180 minutes. At 1°C, roots and leaves differed in their response to toluene. The rates of leakage from roots at 1°C were much lower and the total amounts much smaller than at 25°C, while in leaves the difference between the two temperatures was very small.  相似文献

Excised pieces of vascular bundle and phloem tissue were allowed to accumulate radioactive phosphate and sulfate, and were then sectioned and autoradiographed so as to detect the sites of accumulation. Special methods were needed to prevent any diffusion of the radioisotope. Some autoradiographs obtained are presented. In excised celery vascular bundles, the most radioactive area and hence the most actively accumulating tissue was the young secondary phloem at the sides of the bundle. In intact plants, the same tissue was the most active in translocating. In excised apple phloem there was some variation in behavior, but again the young secondary phloem was generally the most actively accumulating tissue. Accumulation activities of individual cells in the phloem and vascular tissue were compared. It appeared that all cell types, ray, phloem and xylem parenchyma, cambial cells and sieve tubes, accumulated at least 5 times more actively than did the cortical parenchyma cells. The sieve tubes were among the most actively accumulating cells present, accumulating 20 times more actively than the cortical parenchyma cells. It is concluded that accumulation processes have a primary role to play in the mechanism of phloem transport.  相似文献

【背景】福寿螺为世界性恶性入侵水生动物,也是我国公布的第一批外来人侵物种之一。福寿螺大量啃食为害水稻、茭白、白莲等重要农作物,对我国南方各省农业生产造成了巨大威胁,因此,预防和控制福寿螺灾害显得尤为重要。合理利用福寿螺能有效控制福寿螺的数量和危害,是生物防治的一个重要部分。福寿螺卵中含丰富的类胡萝卜素,充分利用螺卵中的类胡萝卜素能拓展福寿螺的利用途径和方法。【方法】为探寻福寿螺卵中类胡萝卜素的提取方法,本研究采用甲醇、无水乙醇、丙酮、乙酸乙酯、二氯甲烷、石油醚等6种常用萃取剂提取福寿螺卵中类胡萝卜素,用紫外可见光分光光度计测定其含量。【结果】结果显示,不同萃取剂中类胡萝卜素的提取量不同,醇类为较适合的提取液(甲醇〉无水乙醇〉丙酮)。【结论与意义】本研究对福寿螺卵中类胡萝卜素的提取方法进行了探索和研究,找出了合适的提取液,为拓展福寿螺的利用途径,以及福寿螺的综合防治提供了参考。  相似文献

We exposed snails of an invasive species of golden apple snail (Pomacea canaliculata) to five artificial sea water treatments at salinity levels of 0, 5, 10, 15 or 20 parts per thousand (ppt) to assess their salinity tolerance. We observed the behaviour, heart rate, total haemocyte counts, haemolymph ionic concentration and Na+/K+-ATPase activity in the mantle at 0, 12, 24, 48, 72 and 96 h post salinity exposures. The heart rate declined with increasing salinity, while Na+/K+-ATPase activity in the mantle presented a reverse trend, possibly to maintain normal osmolality. A trend of rising total haemocyte count was observed from 0 ppt and 5 ppt to 10 ppt salinities, while a sudden increase in the count was observed at 15 ppt and 20 ppt salinity groups. Furthermore, haemolymph Cl?, Na+ and K+ concentrations increased directly with elevated salinity. An additional trial was performed to assess the growth performance of the snails under exposure to low salinities. During a 1 month trial, snails grew better at 5 ppt salinity treatment. Taken together, our results demonstrate that P. canaliculata can tolerate salt stress to some extent. The finding also obviously implies a possible invasive risk to estuaries.  相似文献

An efficient, simple, and small-scale procedure for isolating functional ribonucleic acid (RNA) was successfully applied to many different tissues of grape and apple. These woody plants are rich in polyphenolic compounds and polysaccharides that could impair the RNA extraction. The method chosen is based on the use of hot borate buffer at alkaline pH supplemented with several adjuvants and followed by selective precipitations. Starting with only 0.4 g of fresh tissue and working with small tubes (2 mL), we were able to obtain good yields of high-quality RNA suitable for further applications. The procedure can be proposed for many applications, and it is particularly highly recommended when isolating RNA from a large number of samples.  相似文献

Excised petiolar vascular bundles and excised phloem tissues have been shown to take up phosphate, sulfate and sucrose by a true accumulation process and against high concentration ratios. Phosphate was accumulated principally as inorganic phosphate, and sucrose principally as sucrose. The rates of accumulation of the 3 solutes into the phloem-containing tissues were from 4 to 35 times higher than into comparable parenchyma tissue. It is suggested that this active accumulation mechanism plays an important role in the phenomenon of phloem transport.

The excised vascular, phloem and parenchyma tissues show an aging phenomenon: aerating the excised tissues for 18 hours prior to their use causes marked changes in the accumulatory behavior of the tissue. The data suggest that 1 phosphate accumulation system of low affinity but high capacity exists in fresh tissue, and that aging allows the development of a second, additional phosphate accumulation mechanism of high affinity and low capacity. A possible role in the control of phosphate movement is suggested.

福寿螺是IUCN认定的世界100种恶性外来入侵物种之一,在华南地区已对水稻生产造成严重危害。福寿螺属于热带软体动物,采用万用电表联接热敏电阻法研究了福寿螺在低温胁迫下的过冷却点,并探讨了不同个体福寿螺在过冷却发生后的死亡率和体内组织损伤。结果表明: 1)各种螺高的福寿螺冷却点均值为-6.96 ℃,范围在-6.21--7.32 ℃之间,恢复最高体温均值为-4.07 ℃,范围在-3.07--4.93 ℃之间,过冷却后维持时间均值为45.97 min,范围在18.60-75.34 min;2)福寿螺的过冷却点大小受螺高影响,螺高35 mm≤H<45 mm的福寿螺过冷却点显著高于5 mm≤H<15 mm、15 mm≤H<25 mm、25 mm≤H<35 mm体型的福寿螺,5-35 mm螺高的福寿螺过冷却点变化稳定;3)福寿螺在发生过冷却阶段的死亡率在23.33%-36.67%之间,不同体型的福寿螺之间没有显著性差异;4)过冷却后福寿螺死亡率随暴露时间的延长而提高,0到15 min内由20.9%提高到100%,过冷却后暴露时间对福寿螺死亡率影响较大;5)发生过冷却15 min后取出的福寿螺,染色后其外套膜有少量红色出现,而肾部和消化腺部未呈现明显红色,低温胁迫已经对福寿螺组织造成了显著损伤。本研究结果对于进一步探索福寿螺的生态适应性、扩散北界及福寿螺的越冬机制有一定参考价值。  相似文献

The molluscicidal activity of crude extracts from five highly potential plants, Annona squamosa seed, Nerium indicum Leaves, Stemona tuberose root, Cyperus rotundus corm and Derris elliptica root was assessed to Pomacea canaliculata. D. elliptica root and C. rotundus corm extracts showed the highest toxicity against 3-month old snails which have LC50 as 23.68 +/- 2.96 mg/l and 133.20 +/- 7.94 mg/l, respectively. The C. rotundus corm extracts were chosen for detoxification enzyme in vivo assay which shows esterase and glutathione S-transferase activity in stomach, intestinal tracts and digestive glands of survival treated P. canaliculata were inhibited.  相似文献

Leaves taken from micropropagated shoots of several apple (Malus domestica Borkh.) cultivars were cultured in vitro on Linsmaier & Skoog (LS) medium or the rice anther culture medium of Chu et al. (N6) containing various concentrations of either benzyladenine (BA) or thidiazuron (TDZ) plus naphthaleneacetic acid (NAA). Of the TDZ concentrations tested, 10 M was most effective and it was equivalent to, or better than, 22 M BA for both the percentage of leaves regenerating shoots and number of shoots formed per regenerating leaf in almost every experiment. Lower concentrations of NAA (1.1 and 5.4 M) gave best results with both BA and TDZ. N6 medium gave consistently better results than LS. Lowering total salt concentration or total N concentration of LS to that of N6 did not improve the response nor did changing the NO3:NH4 ratio. The 3–4 leaves on the most distal part of the shoot were most responsive and tended to form the most adventitious shoots. Placing the leaf cultures in the dark for the first 2–3 weeks of the culture period produced the best results. Optimum results were obtained by culturing leaves from the distal part of the shoot in the dark for 2 weeks on N6 medium containing 10 M TDZ and 1.1 or 5.4 M NAA, then moving the cultures to 16 h daylight at a photon flux of 60 mol s-1m-2.  相似文献

Callus formation from stem internodes of the apple rootstocks M.9, M.25, M.26, M.27 and the cherry rootstock Colt, and from pith of Nicotiana tabacum cv. Wisconsin 38 was initiated on 4 -naphthaleneacetic acid (NAA)-based media (2.0–10.0 mg1-1). Transfer of callus to corresponding media lacking NAA allowed regeneration of shoots from callus of M.25, M.27, Colt and tobacco but not of M.9 and M.26. With M.25 phloroglucinol (PG) depressed regeneration from 30 to 10% and no regeneration was observed in cultures grown in the presence of casein hydrolysate (CH) and glutathione (GSH).Organogenesis was also obtained from leaf discs of M.27 employing 6-benzyl-aminopurine (BAP) at 5.0mg 1-1 and 2,4-dichlorophenoxy acetic acid (2,4-D) at 0.1 mg1-1. The regenerated shoots have been multiplied and rooted.Organogenesis also occurred in M.26 from small (1–2mm), green, compact embryoid-like structures derived from stem and leaf surfaces of excised axillary shoots. These structures differentiated into shoots at a low frequency (< 1%) on media containing BAP (1.0mg1-1) and indole-3-butyric acid (IBA) (0.1 mg1-1) and could also be micropropagated by subsequent axillary shoot proliferation.  相似文献

Absorption of ammonium from solutions of ammonium chloride by maize ( Zea mays L. cv. GS-2) tissue was studied. In contrast to an initial rapid phase of absorption in root tissue, a one hour lag period was recorded in leaf tissue. The maximum rate of uptake was observed at 5–10 m M NH4Cl in both tissues. Roots had a Km value of 1.0 m M and Vmax of 24.3 μmol ammonium (g fresh weight)−1 h−1, whereas the leaf tissue had a higher Km (4.1 m M ) and a lower Vmax (8.7 μmol). There was a concentration dependent increase in ethanol soluble and insoluble fractions of organic nitrogen during ammonium supply. The optimum pH for ammonium absorption for both tissues was 7.4. The optimal concentration of CaCl2 for ammonium absorption was 5 m M whereas that of KCl was only 1 m M . In both tissues, the absorption was inhibited substantially by DCMU, DNP, cycloheximide, lincomycin, sodium tungstate, sodium arsenate and to some extent also by the anions nitrate and sulfate. It is suggested that a carrier is involved in an active uptake of ammonium in the leaf tissues.  相似文献

The deep tissue penetration and submicron spatial resolution of multiphoton microscopy and the high detection efficiency and nanometer spectral resolution of a spectrograph were utilized to record spectral images of the intrinsic emission of mouse skin tissues. Autofluorescence from both cellular and extracellular structures, second-harmonic signal from collagen, and a narrowband emission related to Raman scattering of collagen were detected. Visualization of the spectral images by wavelength-to-RGB color image conversion allowed us to identify and discriminate tissue structures such as epidermal keratinocytes, lipid-rich corneocytes, intercellular structures, hair follicles, collagen, elastin, and dermal cells. Our results also showed morphological and spectral differences between excised tissue section, thick excised tissue, and in vivo tissue samples of mouse skin. Results on collagen excitation at different wavelengths suggested that the origin of the narrowband emission was collagen Raman peaks. Moreover, the oscillating spectral dependency of the collagen second-harmonic intensity was experimentally studied. Overall, spectral imaging provided a wealth of information not easily obtainable with present conventional multiphoton imaging systems.  相似文献
