The natriuretic peptide system of a euryhaline teleost, the Japanese eel (Anguilla japonica), consists of three types of hormones [atrial natriuretic peptide (ANP), ventricular natriuretic peptide (VNP), and C-type natriuretic peptide (CNP)] and four types of receptors [natriuretic peptide receptors (NPR)-A, -B, -C, and -D]. Although ANP is recognized as a volume-regulating hormone that extrudes both Na(+) and water in mammals, ANP more specifically extrudes Na(+) in eels. Accumulating evidence shows that ANP is secreted in response to hypernatremia and acts to inhibit the uptake and to stimulate the excretion of Na(+) but not water, thereby promoting seawater (SW) adaptation. In fact, ANP is secreted immediately after transfer of eels to SW and ameliorates sudden increases in plasma Na(+) concentration through inhibition of drinking and intestinal absorption of NaCl. ANP also stimulates the secretion of cortisol, a long-acting hormone for SW adaptation, whereas ANP itself disappears quickly from the circulation. Thus ANP is a primary hormone responsible for the initial phase of SW adaptation. By contrast, CNP appears to be a hormone involved in freshwater (FW) adaptation. Recent data show that the gene expression of CNP and its specific receptor, NPR-B, is much enhanced in FW eels. In fact, CNP infusion increases (22)Na uptake from the environment in FW eels. These results show that ANP and CNP, despite high sequence identity, have opposite effects on salinity adaptation in eels. This difference apparently originates from the difference in their specific receptors, ANP for NPR-A and CNP for NPR-B. VNP may compensate the effects of ANP and CNP for adaptation to respective media, because it has high affinity to both receptors. On the basis of these data, the authors suggest that the natriuretic peptide system is a key endocrine system that allows this euryhaline fish to adapt to diverse osmotic environments, particularly in the initial phase of adaptation.  相似文献

At present, environmental issues associated with rapid economic development are becoming critical concerns that arouse government's and people's particular attention. A large amount of influencing factors and especially their complicated interactions have always thrown confused insights into assessing the dynamic evolvement and sustainable development of urban economy–resource–environment (ERE) system and programming the developing strategies. A combination of system dynamics (SD) and geographic information system (GIS) is expected to explicitly understand the synergic interaction and feedback among a variety of influencing factors in time and space, since SD model can extend the spatial analysis functions of GIS to realize both dynamic simulation and trend prediction of an ERE system development. According to connotation and framework of sustainable development, this study proposes a dynamic combination method of SD–GIS to model and evaluate the urban development in Chongqing city of China suffering from depletion of resource and degradation of environment. To compare different policy inclinations with regard to potential ERE effects, typical scenarios (current, resource, technology and environment scenarios) are designed by adjusting the parameters in the model and changing the specification of some variables. Integrated assessment results indicate that the current ERE system of Chongqing is not sustainable; environment scenario is more effective to sustainable development of urban ERE system in a long run. Under the considerations of development features and regional differences, as well as regular discipline on urbanization, a coordinated combination of environmental, resource and technology scenarios is anticipated to realize sustainable development of urban ERE system.  相似文献

The Epic® system, a high-throughput label-free optical biosensor system, is applied for the biochemical interrogation of phosphor-specific interactions of the 14-3-3 protein and its substrates. It has shown the capability not only for high-throughput characterization of binding rank and affinity but also for the exploration of potential interacting kinases for the substrates. A perspective of biochemical applications for diagnostics and biomarker discovery, as well as cell-based applications for endogenous receptors and viral infection characterization, are also provided.  相似文献

One of the characteristics of all somatic cells is a finite life span. Cells may proliferate until they reach a point after which, although they are metabolically active, they can no longer produce daughter cells. This observation is central to the clonal exhaustion hypothesis, a mechanism cited to explain age-associated immune dysfunction. In this hypothesis, repeated division of lymphocytes leads to a replicative limit, after which they enter the senescent phase but are not lost from the pool of T cells. Advancing age would then be associated with an increase in the number of T cells that are unable to proliferate to a stimulus which induces a proliferative response in T cells from younger individuals. This hypothesis seems both logical and reasonable and is supported by data from both humans and mice with the demonstration of an age-related accumulation of senescent T cells in both species. However, there is an apparent paradox. The paradox arises because the onset of immunosenescence appears to be more closely linked to the life span of the animal rather than the life span of the lymphocyte. The question if mitochondria have some kind of immune system is not trivial. The basis for raising this question is the fact that bacteria, which are progenitors of mitochondria, do have an immune system. The CRISPR system in bacteria based on the principle of RNA interference serves as an organized mechanism for destroying alien nucleic acids, primarily those of viral origin. We have shown that mitochondria are also a target for viral attacks, probably due to a related organization of genomes in these organelles and bacteria. Bioinformatic analysis performed in this study has not given a clear answer if there is a CRISPR-like immune system in mitochondria. However, this does not preclude the possibility of mitochondrial immunity that can be difficult to decipher or that is based on some principles other than those of CRISPR.  相似文献

The authors presented advances in the research on isoform of neural cell adhesion molecule CD56/NCAM, which appears to raise interest not only in biology, but also in clinical medicine. Molecular aspects of its synthesis have been presented and universal features of this protein have been underlined. It appears to play an important role in body homeostasis and especially in the interactions between neural, immune and endocrine systems. In relation to the immune system there are data suggesting significance of local protective and regulatory mechanisms in the liver linked to so called NKT (CD56+) cells. Main sites of prevalence of CD56/NCAM have been indicated both in physiology and pathology, especially in malignancy. CD56/NCAM incidence is common in tumors of neuroectodermal and endocrine origin. Besides it may be expressed on cells of several hemopoetic neoplasms originated both, from lymphoid and myeloid lineage. Moreover, the attention was paid to CD56/ NCAM expression in atypical sites as for example on epithelia of bile ducts in children with extrahepatic biliary atresia and on cells of pancreatic ducts in the course of chronic pancreatitis.  相似文献

The radio frequency (RF)-induced heating is a major concern when patients with medical devices are placed inside a magnetic resonance imaging (MRI) system. In this article, numerical studies are applied to investigate the potentials of using insulated materials to reduce the RF heating for external fixation devices. It is found that by changing the dielectric constant of the insulation material, the RF-induced heating at the tips of devices can be altered. This study indicates a potential technique of developing external fixation device with low MRI RF heating.  相似文献

Hodgkin and Huxley (HH) discovered that voltages control ionic currents in nerve membranes. This led them to describe electrical activity in a neuronal membrane patch in terms of an electronic circuit whose characteristics were determined using empirical data. Due to the complexity of this model, a variety of heuristics, including relaxation oscillator circuits and integrate-and-fire models, have been used to investigate activity in neurons, and these simpler models have been successful in suggesting experiments and explaining observations. Connections between most of the simpler models had not been made clear until recently. Shown here are connections between these heuristics and the full HH model. In particular, we study a new model (Type III circuit): It includes the van der Pol-based models; it can be approximated by a simple integrate-and-fire model; and it creates voltages and currents that correspond, respectively, to the h and V components of the HH system.  相似文献

Environmental variables such as photoperiod, heat, stress, nutrition and other external factors have profound effects on quality and quantity of a dairy cow's milk. The way in which the environment interacts with genotype to impact milk production is unknown; however, evidence from our laboratory suggests that circadian clocks play a role. Daily and seasonal endocrine rhythms are coordinated in mammals by the master circadian clock in the hypothalamus. Peripheral clocks are distributed in every organ and coordinated by signals from the master clock. We and others have shown that there is a circadian clock in the mammary gland. Approximately 7% of the genes expressed during lactation had circadian patterns including core clock and metabolic genes. Amplitude changes occurred in the core mammary clock genes during the transition from pregnancy to lactation and were coordinated with changes in molecular clocks among multiple tissues. In vitro studies using a bovine mammary cell line showed that external stimulation synchronized mammary clocks, and expression of the core clock gene, BMAL1, was induced by lactogens. Female clock/clock mutant mice, which have disrupted circadian rhythms, have impaired mammary development and their offspring failed to thrive suggesting that the dam's milk production was not adequate enough to nourish their young. We envision that, in mammals, during the transition from pregnancy to lactation the master clock is modified by environmental and physiological cues that it receives, including photoperiod length. In turn, the master clock coordinates changes in endocrine milieu that signals peripheral tissues. In dairy cows, it is clear that changes in photoperiod during the dry period and/or during lactation influences milk production. We believe that the photoperiod effect on milk production is mediated, in part by the 'setting' of the master clock with light, which modifies peripheral circadian clocks including the mammary core clock and subsequently impacts milk yield and may impact milk composition.  相似文献

In 2003, a Heat Health Watch Warning System was developed in France to anticipate heat waves that may result in a large excess of mortality. The system was developed on the basis of a retrospective analysis of mortality and meteorological data in fourteen pilot cities. Several meteorological indicators were tested in relation to levels of excess mortality. Computations of sensibility and specificity were used to choose the meteorological indicators and the cut-offs. An indicator that mixes minimum and maximum temperatures was chosen. The cut-offs were set in order to anticipate events resulting in an excess mortality above 100% in the smallest cities and above 50% in Paris, Lyon, Marseille and Lille. The system was extended nationwide using the 98th percentile of the distribution of minimum and maximum temperatures. A national action plan was set up, using this watch warning system. It was activated on 1st June 2004 on a national scale. The system implies a close cooperation between the French Weather Bureau (Météo France), the National Institute of Health Surveillance (InVS) and the Ministry of Health. The system is supported by a panel of preventive actions, to prevent the sanitary impact of heat waves.  相似文献

Ubiquitin–proteasome system (UPS) is a selective proteolytic system that is associated with the expression or function of target proteins and participates in various physiological and pathological processes of breast cancer. Inhibitors targeting the 26S proteasome in combination with other drugs have shown promising therapeutic effects in the clinical treatment of breast cancer. Moreover, several inhibitors/stimulators targeting other UPS components are also effective in preclinical studies, but have not yet been applied in the clinical treatment of breast cancer. Therefore, it is vital to comprehensively understand the functions of ubiquitination in breast cancer and to identify potential tumor promoters or tumor suppressors among UPS family members, with the aim of developing more effective and specific inhibitors/stimulators targeting specific components of this system.  相似文献

 Lotka–Volterra equations are considered a dynamical game, where the phenotypes of the predator and of the prey can vary. This differs from the usual procedure of specifying as a priori laws according to which strategies are supposed to change. The question at stake is the survival of each of the species, instead of the maximization of a given pay-off by each player, as it is commonly discussed in games. The predator needs the prey, while the prey can survive without the predator. These obvious and simplistic constraints are enough to shape the regulation of the system: notably, the largest closed set of initial conditions can be delineated, from which there exists at least one evolutionary path where the population can avoid extinction forever. To these so-called viable trajectories, viable strategies are associated, respectively for the prey or for the predator. A coexistence set can then be defined. Within this set and outside the boundary, strategies can vary arbitrarily within given bounds while remaining viable, whereas on the boundary, only specific strategies can guarantee the viability of the system. Particulate membrane preparations have been isolated from culminatingDictyostelium discoideum cells. The preparations incorporated glucose from uridine 5′-diphosphate-glucose into a glucose polymer or polymers. These have been shown to be homopolymers ofβ-linked glucose. A high percentage (78% by methylation analysis) of the linkages formed are 1,4-linkages and a lower percentage (12%) are 1,3-linkages. The glucan-synthase complex present in the particulate membrane preparation has an apparent Km of 0.28 mM and a Vmax of 1.59 nmol·min?1·(mg protein)?1. The enzyme system is dependent upon Mg2+ and cellobiose for maximal activity, but is inhibited by millimolar levels of Ca2+. Particulate membrane preparations were made from cells at various times during a synchronous developmental time course and demonstrated that the glucan-synthase activity appeared at the tight-aggregate stage of development.  相似文献
