The “Old Sulphur Well” has a subterranean input of water containing 5.5 mM total sulfide, which would be inhibitory to the growth of most bacteria. The obligately chemolithoautotrophic Halothiobacillus neapolitanus is a sulfur bacterium known to tolerate and metabolize high sulfide concentrations, and we report the isolation of H. neapolitanus strain OSWA from this source. Strain OSWA grows well on thiosulfate and tetrathionate as energy sources, and tolerates at least 5 mM sulfide. Its specific growth rates and yields in batch culture were 0.22 h−1 and 5.3 g mol−1 (thiosulfate), and 0.23 h−1 and 9.5 g mol−1 (tetrathionate). Its 16S rRNA gene sequence shows >99% identity to reference sequences of H. neapolitanus, and it shares morphological and physiological characteristics typical of the species. It is one of a very small number of strains of H. neapolitanus described to date, and the first to be isolated from an ancient sulfide-rich natural spa.  相似文献

Deregulated cell turnover in Helicobacter pylori (H. pylori)-colonized gastric mucosa has been suggested to be linked to the gastric carcinogenesis pathway. We previously reported attenuation of apoptosis and enhancement of cellular proliferation in the H. pylori-colonized gastric mucosa of Mongolian gerbils as compared to that in mice, which might reflect a specific link between H. pylori colonization and carcinogenesis in the Mongolian gerbils; the difference between the two strains could be attributable to differences in the host genetic background. Inducible-type nitric oxide synthase (iNOS) is thought to participate in not only the inflammatory response, but also in the regulation of gastric mucosal cell turnover in H. pylori-colonized gastric mucosa. Thus, the present study was designed to examine gastric leukocyte activation and epithelial cell apoptosis in the gastric mucosa following H. pylori inoculation in iNOS-knockout mice. Methods: iNOS-knockout mice (iNOS−/−) and their iNOS+/+ littermates were orally inoculated with the Sydney strain of H. pylori (SS1, 108 colony-forming units [CFU]). H. pylori infection was confirmed by microaerobic bacterial culture. The stomach of each mouse was evaluated 14 weeks and 30 weeks after the inoculation. Gastric mucosal accumulation of polymorphonuclear leukocytes (PMN) was assessed by determining the myeloperoxidase (MPO) activity and histological score based on the updated Sydney system. The level of apoptosis was determined by estimation of the cytoplasmic levels of mono- and oligonucleosomes and by the terminal deoxynucleotidyl transferase-mediated deoxyuridine triphosphate nick-end labeling method. Results: The SS1-inoculated mice showed persistent H. pylori colonization for 12 weeks. While gastric mucosal PMN infiltration increased following SS1 inoculation in both iNOS+/+ and iNOS−/−strains, enhanced DNA fragmentation was observed in only SS1-colonized iNOS+/+ mice, and not in the iNOS−/− mice. In conclusion, although the recruitment of PMN in response to H. pylori was evoked even in the gastric mucosa of iNOS−/− mice, epithelial cell apoptosis induced by H. pylori was attenuated in this strain. These data suggest that iNOS may play an important role in promoting apoptosis in the H. pylori-infected inflamed gastric mucosa, and that persistent inflammation without apoptosis in iNOS−/− mice with H. pylori infection may be linked to preneoplastic transformation.  相似文献

Floating Pennywort (Hydrocotyle ranunculoides L. fil.) is a worldwide distributed aquatic plant. The species is native to North America and quite common also in Central and South America. In Europe, Japan and Australia it is known as an alien plant, sometimes causing serious problems for affected ecosystems and human use of water bodies. Starting from Western Europe with an eastwards directed spread, Floating Pennywort was recorded in Germany in 2004 for the first time. Since then, the species spread out and got established in western parts of Central Europe. For a definite prediction of the potential of a further spread, data about biology, in particular growth and photosynthesis are needed. Here, regeneration capacity, growth at different nutrient availabilities and photosynthesis of H. ranunculoides were investigated. In addition biomass samples were taken in the field. Results show an enormous regeneration capacity (e.g., by forming new shoots from small shoot fragments), increasing growth rates under increasing nutrient availability and a maximum increase of biomass reaching 0.132±0.008 g g−1 dw d−1. Dense populations of H. ranunculoides growing in ponds and oxbows were found at high nutrient content of the substrate, the biomass reaching there up to 532.4±14.2 g dw m−2. Gas exchange analysis showed a physiological optimum of H. ranunculoides CO2 uptake at temperatures between 25 and 35 °C and high photon flux densities (PPFD) above 800 μmol photons m−2 s−1. In comparison, native Hydrocotyle vulgaris showed an optimum of net photosynthesis at 20–30 °C and a light saturation of CO2 gas exchange at 350 μmol photons m−2 s−1.  相似文献

Metal concentrations and population parameters of the seagrass Halodule wrightii were determined at three locations at Rio de Janeiro State, Brazil. The possible increase of metal availability in one of these areas, Sepetiba Bay, as a result of dredging of contaminated bottom sediments which ocurred, was evaluated by analyses of Al, Cd, Cr, Cu, Fe, Ni, Pb and Zn in root, rhizome and shoots. In addition, analyses were carried out in H. wrightii populations from non-contaminated areas located at northwestern (Cabo Frio) and southeastern (Angra do Reis) regions of Rio de Janeiro State. Concurrently, abundance and density data of the seagrass populations were obtained. It was found that concentration from Sepetiba Bay samples up to 1.6 ± 0.4 μg g−1 of Cd, 12 ± 1.0 μg g−1 of Cr, 27 ± 2.4 μg g−1 of Pb, 291 ± 47 μg g−1 of Mn, 128 ± 23 μg g−1 of Zn were significantly higher than that from two other collection sites. An increase in Cd and Zn concentration was observed in H. wrightii from Sepetiba Bay indicating that metal mobilization from contaminated sediments through dredging activities were, at least in part, transferred to the biotic compartment via accumulation by the seagrass. The populations of seagrass within the region demonstrated quite substantial changes in biomass data but not in shoot or rhizome density during the study. Such changes in biomass are to be expected, as these dynamics are typical of the small, isolated monospecific populations of H. wrightii along the Rio de Janeiro coast.  相似文献

Allelopathic effects of the green macroalgae Ulva lactuca on the growth of three species of red tide microalgae, Heterosigma akashiwo, Alexandrium tamarense, and Skeletonema costatum were tested in laboratory co-cultures precluding the nutrient and light limitation and the effect of high pH. The growth of all three species of microalgae was significantly (p < 0.01) inhibited by fresh U. lactuca. In nutrient replete semicontinuous co-cultures with U. lactuca, H. akashiwo was completely dead in 12 days, and the growth of A. tamarense and S. costatum was reduced by 48 and 46%, respectively by U. lactuca within 12 days. The U. lactuca culture filtrate exhibited a significant (p < 0.05) inhibitory effect on the microalgae in the first 1 or 2 days, but growth resumed in the following days, and S. costatum growth was slightly (p > 0.05) promoted from day 3. The results suggested that the allelopathic compounds are quickly degradable and a long-term inhibition might need the continuous addition of compounds originated from macroalgae. Dried U. lactuca also exhibited inhibitory effects on the microalgae, and the normalized mean growth rates of microalgae decreased with the biomass of dried U. lactuca. The dependent relationships were y = −2.1208x2 + 1.0159x + 0.9752 for H. akashiwo, y = 0.7133x2 − 3.5813x + 1.1665 for A. tamarense, and y = −0.2114x2 − 1.063x + 1.0873 for S. costatum, respectively. The potential feasibility of utilization of dried U. lactuca against red tide microalgae was 2.0 g dry wt L−1. The present study shows that U. lactuca exhibits negative allelopathic effects on harmful bloom-forming microalgae.  相似文献

The thermodynamic and spectral properties of the photochemical reaction center components of Heliobacterium chlorum have been examined. The primary electron donor bacteriochlorophyll has Em,7 = +225 mV, and the ‘primary acceptor’ Em,10 = −510 mV. The former has an EPR signal in its oxidised form near G = 2.0025, ΔH = 0.95 mT, reminiscent of the properties of the primary donor in bacteria containing bacteriochlorophyll a. The ‘primary acceptor’ has properties similar to those of the iron-sulfur cluster acceptors of green sulfur bacteria. H. chlorum contains a c-type cytochrome (Em,7 = +170 mV) that donates electrons to the photooxidised primary donor with . The reaction center of H. chlorum is thus very similar to that found in representative green sulfur bacteria, but the cellular architecture and photopigments of this group are quite distinct from those of H. chlorum.  相似文献

This paper presents a model for the continuous production of green cells of the microalga Haematococcus pluvialis, in both indoor and outdoor conditions. To develop this model, the influence of irradiance and dilution rate on the performance of continuous cultures of H. pluvialis was studied in the laboratory but simulating outdoor conditions. Characterization of the cultures included biomass productivity, fluorescence of chlorophylls, pigment content, elemental composition of the biomass, cell density, cell size, homogeneity and nitrate consumption rate. Results showed that the optimal dilution rate was 0.04 h−1, and that higher external irradiance resulted in higher biomass productivity in all cases, with a maximum value of 0.58 g L−1day−1. Continuous cultures were stable for more than 3 months, in spite of photoinhibition at noon, producing homogeneous biomass with a stable biochemical composition and cell morphology at each steady state. Astaxanthin accumulation was not observed in spite of the high levels of irradiance essayed, and cells remained in the flagellated-palmeloids green form whatever the culture conditions. High dilution rates produced small cells of 22 μm diameter, with a high nitrogen content of up to 10.0% d.wt. The average irradiance within the reactor was the main factor determining the behaviour of the cultures, although the external irradiance impinging on the reactor surface also influenced the results, indicating the existence of photoinhibition. The influence of both external and average irradiance on the growth of H. pluvialis was modelized. The accuracy of the model obtained was verified on a 0.22 m3 outdoor tubular photobioreactor operated in both discontinuous and continuous mode, obtaining a maximum biomass productivity of 0.68 g L−1day−1. The model reproduced the experimental data of biomass concentration and productivity, cell size and nitrate consumption, providing to be a powerful tool for optimizing the design and operation of outdoor photobioreactors for the production of green cells of H. pluvialis.  相似文献

Because of its novel bioactive properties the production of gymnodimine for use as a pharmaceutical precursor has aroused interest. The dinoflagellate, Karenia selliformis produces gymnodimine when grown in bulk culture using GP + selenium medium but the growth rates (μ) and levels of gymnodimine are low (μ, 0.05 days−1; gymnodimine 250 μg L−1 max). We describe the effects of organic acid additions (acetate, glycolate, alanine and glutamate additions and combinations of these) in enhancing growth and gymnodimine production in axenic cultures. The most effective organic acid combinations in decreasing order were: glycolate/alanine > acetate > glycolate. Glycolate/alanine optimised gymnodimine production by prolonging growth (maximum cell yield, 1.76 × 105 cells mL−1; gymnodimine, 1260 μg L−1; growth rate (μ), 0.2 days−1) compared to the control (growth maximum cell yield, 7.8 × 104 cells mL−1; gymnodimine, 780 μg L−1; μ, 0.17 days−1). Acetate enhanced gymnodimine by stimulating growth rate (μ, 0.23 days−1) and the large concentration of gymnodimine per cell (16 pg cell−1 cf. 9.8 pg cell−1 for the control) suggests a role for this compound in gymnodimine biosynthesis. Amending culture media with Mn2+ additions resulted in slightly decreased growth in control cultures and increased the gymnodimine while in glycolate/alanine cultures growth was stimulated but gymnodimine production decreased. The results suggest that the organic acid can enhance gymnodimine production by either enhancing growth maximum or the biosynthetic pathway.  相似文献

In different mammalian species, in vitro culture and manipulation can lead to aberrant fetal and peri-natal development. It has been postulated that these diverse abnormalities are caused by epigenetic alterations and that these could affect genes that are regulated by genomic imprinting. To explore this hypothesis relative to somatic cell nuclear transfer in sheep, we investigated whether the ovine H19-IGF2 and IGF2R loci are imprinted and analysed their DNA methylation status in cloned lambs. A comparison between parthenogenetic and control concepti established that imprinting at these two growth-related loci is evolutionarily conserved in sheep. As in humans and mice, IGF2R and H19 comprise differentially methylated regions (DMRs) that are methylated on one of the two parental alleles predominantly. In tongue tissue from 12 out of 13 cloned lambs analysed, the DMR in the second intron of IGF2R had strongly reduced levels of DNA methylation. The DMR located upstream of the ovine H19 gene was found to be similarly organised as in humans and mice, with multiple CTCF binding sites. At this DMR, however, aberrant methylation was observed in only one of the cloned lambs. Although the underlying mechanisms remain to be determined, our data indicate that somatic cell nuclear transfer procedures can lead to epigenetic deregulation at imprinted loci.  相似文献

When cultivated in Murashige & Skoog medium supplemented with 0.2 mg l−1 2,4-dichlorophenoxy acetic acid and 0.5 mg l−1 6-benzyladenine, Perilla frutescens cells in suspension culture grew rapidly reaching about 13.6 g dry wt l−1 after 12 days. The cell line produced both anthocyanin 0.9 g l−1 and triterpenoids: 16 mg l−1 oleanolic acid (OA), 25 mg l−1 ursolic acid (UA) and 14 mg l−1 tormentic acid (TA). When P. frutescens cells of 7-day-old cultures were exposed to a yeast elicitor at 0.5–5% (v/v) for 7 days, it was found that anthocyanin content peaked at 10.2% of dry weight with yeast elicitor at 1% (v/v) whereas the maximum production of oleanolic acid and ursolic acid in cultures treated with 2% (v/v) yeast elicitor was 19 and 27 mg l−1, a 46 and 24% increase over the control, respectively. This is the first report of simultaneous production of both anthocyanin and triterpenoids in a single culture system.  相似文献

Paternal repression of the imprinted H19 gene is mediated by a differentially methylated domain (DMD) that is essential to imprinting of both H19 and the linked and oppositely imprinted Igf2 gene. The mechanisms by which paternal-specific methylation of the DMD survive the period of genome-wide demethylation in the early embryo and are subsequently used to govern imprinted expression are not known. Methyl-CpG binding (MBD) proteins are likely candidates to explain how these DMDs are recognized to silence the locus, because they preferentially bind methylated DNA and recruit repression complexes with histone deacetylase activity. MBD RNA and protein are found in preimplantation embryos, and chromatin immunoprecipitation shows that MBD3 is bound to the H19 DMD. To test a role for MBDs in imprinting, two independent RNAi-based strategies were used to deplete MBD3 in early mouse embryos, with the same results. In RNAi-treated blastocysts, paternal H19 expression was activated, supporting the hypothesis that MBD3, which is also a member of the Mi-2/NuRD complex, is required to repress the paternal H19 allele. RNAi-treated blastocysts also have reduced levels of the Mi-2/NuRD complex protein MTA-2, which suggests a role for the Mi-2/NuRD repressive complex in paternal-specific silencing at the H19 locus. Furthermore, DNA methylation was reduced at the H19 DMD when MBD3 protein was depleted. In contrast, expression and DNA methylation were not disrupted in preimplantation embryos for other imprinted genes. These results demonstrate new roles for MBD3 in maintaining imprinting control region DNA methylation and silencing the paternal H19 allele. Finally, MBD3-depleted preimplantation embryos have reduced cell numbers, suggesting a role for MBD3 in cell division.  相似文献

For most imprinted genes, a difference in expression between the maternal and paternal alleles is associated with a corresponding difference in DNA methylation that is localized to a differentially methylated domain (DMD). Removal of a gene's DMD leads to a loss of imprinting. These observations suggest that DMDs have a determinative role in genomic imprinting. To examine this possibility, we introduced sequences from the DMDs of the imprinted Igf2r, H19, and Snrpn genes into a nonimprinted derivative of the normally imprinted RSVIgmyc transgene, created by excising its own DMD. Hybrid transgenes with sequences from the Igf2r DMD2 were consistently imprinted, with the maternal allele being more methylated than the paternal allele. Only the repeated sequences within DMD2 were required for imprinting these transgenes. Hybrid transgenes containing H19 and Snrpn DMD sequences and ones containing sequences from the long terminal repeat of a murine intracisternal A particle retrotransposon were not imprinted. The Igf2r hybrid transgenes are comprised entirely of mouse genomic DNA and behave as endogenous imprinted genes in inbred wild-type and mutant mouse strains. These types of hybrid transgenes can be used to elucidate the functions of DMD sequences in genomic imprinting.  相似文献

Conidiation and lytic enzyme production by Trichoderma viride at different solids concentration of pre-treated municipal wastewater sludge was examined in a 15-L fermenter. The maximum conidia concentration (5.94 × 107 CFU mL−1 at 96 h) was obtained at 30 g L−1 suspended solids. The maximum lytic enzyme activities were achieved around 12–30 h of fermentation. Bioassay against a fungal phytopathogen, Fusarium sp. showed maximum activity in the sample drawn around 96 h of fermentation at 30 g L−1 suspended solids concentration. Entomotoxicity against spruce budworm larvae showed maximum value ≈17290 SBU μL−1 at 30 g L−1 suspended solids concentration at the end of fermentation (96 h). Plant bioassay showed dual action of T. viride, i.e., disease prevention and growth promotion. The rheological analyses of fermentation sludges showed the pseudoplastic behaviour. In order to maintain required dissolved oxygen concentration ≥30%, the agitation and aeration requirements significantly increased at 35 g L−1 compared to 30 and 25 g L−1. The oxygen uptake rate and volumetric oxygen mass transfer coefficient, kLa at 35 g L−1 did not increase in comparison to 30 g L−1 due to rheological complexity of the broth during fermentation. Thus, the successful fermentation operation of the biocontrol fungus T. viride is a rational indication of its potential for mass-scale production for agriculture and forest sector as a biocontrol agent.  相似文献

Optimal utilisation of tannin-rich browse tree fodders including Acacia spp. foliages as crude protein (CP) supplements to ruminants in the tropics is limited by less available information on their feed nutritive potential. Two studies were conducted to: (1) determine rate and extent of ruminal dry matter (DM) degradability (DMD) and (2) investigate effect of sun-dried Acacia nilotica (NLM), A. polyacantha (PLM) and Leucaena leucocephala leaf meal (LLM) supplementation on growth performance of 20 growing (7–9 months old) Small East African male goats (14.6 ± 0.68 kg) fed on native pasture hay (NPH) basal diet for 84 days in a completely randomised design experiment in north-western Tanzania. The goats were randomised into four treatment groups consisting of five animals each. Three supplement diets: 115.3 g NLM (T2), 125.9 g PLM (T3) and 124.1 g LLM (T4), which was used as a positive control, were supplemented at 20% of the expected DM intake (DMI; i.e., 3% body weight) to the three animal groups fed on NPH (basal diet) compared to the animals in a control group that were fed on NPH without browse supplementation (T1).

NPH had significantly the lowest (P < 0.05) CP of 45.5 g kg−1 DM compared to NLM, PLM and LLM (159, 195 and 187 g kg−1 DM, respectively). NPH had higher (P < 0.05) fibre fractions; lower ruminal DM degradability characteristics and ME than NLM, PLM and LLM. Supplementation of the animals with browse resulted to (P < 0.05) higher average daily weight gains (ADG) of 157.1 g day−1 in T4 than the animals fed on T2 (114.3 g day−1) and T3 (42.9 g day−1), and even to those fed on T1 (control), which lost weight (−71.4 g day−1). Improved weight gains were mainly due to corrected feed nitrogen (N) or CP due to supplementation of the animals with browse fodder. Too low CP of the NPH would not meet the normal requirements of CP (80 g CP kg−1 DM) for optimal rumen microbial function in ruminants. Higher ADG due to LLM (T4) and NLM (T2) supplementation suggest optimised weight gains due to browse supplementation (20% of expected DMI); while lower weight gains from supplementation with PLM (T3) indicate the possible utilisation of A. polyacantha leaves to overcome weight losses especially during dry seasons.  相似文献
