We describe 15 polymorphic microsatellite loci from the white-ruffed manakin Corapipo altera, a common understory bird of Neotropical lowland and montane evergreen forests from eastern Honduras to northwestern Colombia. These markers were developed in order to assess population structure and genetic diversity in a fragmented landscape, and to study gene flow between forest fragments. Primers were tested on a population of 159 individuals from the Coto Brus region of southwestern Costa Rica. We found between four and 23 alleles per locus, and observed heterozygosities ranging from 0.23 to 0.93.  相似文献

Eight microsatellite loci were isolated from the lance‐tailed manakin (Chiroxiphia lanceolata), a polygynous lek‐breeding bird from Central America. Five of these loci were polymorphic (two to seven alleles per locus), with observed levels of heterozygosity ranging from 0.100 to 0.860 (n = 50 individuals). These variable loci provide a valuable tool for assessing patterns of parentage and relatedness within lance‐tailed manakin social groups.  相似文献

The lance‐tailed manakin (Chiroxiphia lanceolata) is a lek‐breeding bird from Central America in which males court females with complex cooperative displays. To resolve detailed patterns of paternity in the wild, we isolated and characterized 12 novel microsatellite loci in this species. Eleven of these loci were polymorphic (five to 14 alleles), with observed heterozygosities ranging from 0.36 to 0.87 (N = 574 individuals). We tested for linkage disequilibrium using randomized subsamples of adults to control for known family structure among long‐lived and sedentary individuals. These loci will be valuable in resolving paternity among many candidate fathers in this species.  相似文献

Determining the genetic structure of tropical bird populations is important for assessing potential genetic effects of future habitat fragmentation and for testing hypotheses about evolutionary mechanisms promoting diversification. Here we used 10 microsatellite DNA loci to describe levels of genetic differentiation for five populations of the lek-mating blue manakin (Chiroxiphia caudata), sampled along a 414-km transect within the largest remaining continuous tract of the highly endangered Atlantic Forest habitat in southeast Brazil. We found small but significant levels of differentiation between most populations. F(ST) values varied from 0.0 to 0.023 (overall F(ST)=0.012) that conformed to a strong isolation by distance relationship, suggesting that observed levels of differentiation are a result of migration-drift equilibrium. Five novel polymorphic microsatellite markers were isolated from enriched libraries in order to examine population structure in the Canary Island endemic Bencomia exstipulata and the related Bencomia caudata. All markers were successfully amplified from both Bencomia species. They possessed an average of 6.8 and 7.0 alleles per locus in B. exstipulata and B. caudata, respectively. Our data are consistent with the tetraploid condition of both Bencomia species. In addition, this set of microsatellite loci offers an efficient tool to clarify the genetic relationships within the Bencomia alliance.  相似文献

The diet of Chiroxiphia caudata and Schiffornis virescens were studied over three years in four forest sites in the southeastern Atlantic Forest of Brazil. Theory suggests that coexistence of similar species is possible by resource-use differences. To test this, patterns of niche overlap and niche breadth in diet were measured in different seasons and forest sites, based on fruit morphological traits and plant life forms. The results suggest seasonal and spatial variation in resource partitioning. During periods of high fruit availability, there was no evidence of fruit trait selection. In contrast, during periods of fruit scarcity in primary forests, there was evidence of niche partitioning, where C. caudata foraged for larger fruits in canopy and S. virescens searched for understory plants. The niche overlap was higher in secondary forest during periods of scarcity, when both species used small understory fruits.

Foi estudada a dieta de Chiroxiphia caudata e Schiffornis virescens durante três anos, em quatro tipos de fisionomias florestais no sudeste da Mata Atlântica, Brasil. A teoria sugere que a coexistência de espécies semelhantes é possível pelas diferenças no uso dos recursos. Assim, nós mensuramos a amplitude e sobreposição de nicho considerando a morfologia dos frutos consumidos e a forma de vida das espécies de plantas utilizadas em quatro tipos florestais e em diferentes estações. Os resultados sugerem mudanças sazonais e espaciais na partilha de recursos. Durante períodos de alta disponibilidade de frutos, não há evidências de partilha de recursos. Já em períodos de escassez de frutos nas florestas primárias, C. caudata consumiu frutos grandes presentes no dossel enquanto S. virescens utilizou os frutos pequenos do sub-bosque. Na floresta secundária, o período de escassez foi marcado por sobreposição intensa de nicho onde ambas as espécies consumiram frutos pequenos de sub-bosque.  相似文献

Guira cuckoos (Guira guira) are communal nesting birds endemic to South America that show high levels of conflict between members of the group over the contribution to the clutch. Adults eject eggs and even nestlings from the communal nest, sometimes leading to the loss of the entire brood. We developed seven polymorphic microsatellites for Guira cuckoos using an enrichment protocol. The number of alleles ranged from 5 to 14 (mean 9.86) and the heterozygosity ranged from 0.41 to 0.89 for the eight to 88 individuals screened. To investigate the mating system of northern flickers (Colaptes auratus), we developed primers for 14 microsatellite loci and screened them in 68 unrelated adults and their offspring. All markers were highly polymorphic with 9 to +36 alleles per locus. One marker was Z‐chromosome linked; one marker exceeded the size standard range and could not be analysed further. We checked the other 12 markers for Mendelian inheritance in 36 broods for which the social parents were known. Seven markers showed evidence for the presence of null alleles, and three of those showed significant deviation from Hardy–Weinberg equilibrium. Markers were generally unlinked.  相似文献

Primer sequences for 16 microsatellite loci were developed from Schizolobium parahyba, a tropical tree species. Twelve loci were found to be polymorphic after screening diversity in individuals from Belize. A total of 39 alleles were found at nine loci. The markers are invaluable tools for studying the population genetics and mating system of the species.  相似文献

Here we presented 10 polymorphic microsatellite loci obtained from scarlet ibis through an enriched genomic library. The analysis of 45 individuals from three Brazilian natural populations showed allelic diversity ranged from three to 17 alleles, observed heterozygosity ranged from 0.03 to 0.92, and expected heterozygosity ranged from 0.06 to 0.92. These highly variable microsatellite loci can provide means for assessing overall genetic variation in its remnant natural populations, which may help the development of effective conservation programs.  相似文献

Epidendrum puniceoluteum is an endemic orchid of Atlantic Rainforest, restricted to few populations only due to the destruction and fragmentation of its native habitat. Here, we report on the development of 10 microsatellite markers isolated from this orchid species. Genetic variability was characterized in two distant populations from Brazil coast. The number of alleles observed for each locus ranged from two to 12 and with an average of 6.4 alleles per locus. These microsatellites should be valuable tools for studying both fine-scale genetic structure of scattered E. puniceoluteum population and patterns will be useful genetic markers for other closely related taxa.  相似文献

The isolation of molecular markers in Asparagus acutifolius, a wild edible plant species, is important to characterize local ecotypes that could be cultivated and preserved. We isolated and characterized polymorphic microsatellite loci from A. acutifolius by constructing and screening an enriched DNA library. Primer pairs were designed for 12 loci. Seven primer pairs worked well during amplification reactions and were tested on a wild population from Pontecagnano (SA), Italy. These loci showed a high level of genetic variability, with the numbers of alleles identified ranging from two to five and observed heterozygosity ranging from 0.20 to 0.73.  相似文献

We characterized eight polymorphic microsatellites in the icefish Chionodraco rastrospinosus (Perciformes, Notothenioidea, Channichthyidae). Microsatellites were isolated from a partial genomic library enriched for an AC motif. Chionodraco rastrospinosus is an endemic species inhabiting southern ocean waters surrounding the Antarctic Peninsula, the South Shetland Islands, and the South Orkney Islands. An excess of homozygotes was observed in seven out of the eight investigated loci; however, presence of null alleles was detected only for three of them suggesting that other factors may act in reducing heterozygosity. These molecular markers will be useful to investigate icefish genetic structure, possibly providing insights on its effective population size and demographic history.  相似文献

We isolated 49 microsatellite loci from a genomic library of Tripterygion delaisi×anthosoma enriched for CA and GA repeats. Ten loci were screened in 30 individuals with high numbers of alleles per locus (averaging 15.5 ± 2.86) and observed heterozygosity (averaging 0.765 ± 0.052). No deviations from Hardy–Weinberg expectations were detected. These highly polymorphic markers will be useful in determining the spatial patterns of genetic diversity between and within subspecies of Tripterygion delaisi.  相似文献

Primers were developed for 10 microsatellite loci for two species of Oceanodroma storm-petrels. Variability was tested in 27 O. castro and 22 O. monteiroi from the Azores, and 24 O. leucorhoa from Norway. At least six loci amplified reliably and were polymorphic in each species. The number of alleles per locus averaged 4.6, and observed heterozygosities averaged 0.41. Most primers also yielded polymerase chain reaction products in O. tethys, O. hornbyi and Pterodroma phaeopygia. Weigela coraeensis var. coraeensis is a deciduous shrub species distributed in Japan on the mainland, Honshu, whereas its variety W. coraeensis var. fragrans is endemic to the Izu Islands located south of Honshu. We isolated eight polymorphic microsatellite loci from the species and characterized these loci for 20 individuals from a population in Honshu. The number of alleles per locus ranged from 7 to 15 and the observed and expected heterozygosities ranged from 0.60 to 0.90 and from 0.65 to 0.90, respectively. These eight polymorphic microsatellites will be useful for examining intraspecific genetic differentiation in W. coraeensis.  相似文献
