Abstract

Natural populations of species in the Acer genus occurring in Campania (southern Italy) were surveyed by screening seven microsatellite loci. Primer pairs for Acer pseudoplatanus L. microsatellite loci were analysed in six different species: Acer lobelii Ten., Acer campestre L., Acer pseudoplatanus L., Acer obtusatum W. et K., Acer neapolitanum Ten. and Acer monspessulanum L. The aim of the present study was to survey the genetic variability and genetic structure of natural populations of the Acer genus in Campania. The high degree of polymorphism observed in six different species of Acer makes these markers useful for investigating genetic variation at various spatial scales, and for the analysis of gene flow and of the mating system.  相似文献

Vegetation experiments were conducted under greenhouse conditions in which long-term effects of simulated acid rain on the shoots, and short-term effects of acidified root medium on the roots of two maple species (Acer pseudoplatanus L. andAcer negundo L.) were studied. Both alternatives of the acidification stress were studied with respect to exogenous applications of auxins and gibberellins. Changes in seedling growth caused by acid rain were not marked, but changes in the levels of endogenous growth regulators were detected, both the long-term exposure ofAcer pseudoplatanus L. seedling shoots to acid rain and the short-term acidification of root environment resulted in enhanced cytokinin activity and in decreased gibberellin activity. But when A.pseudoplatanus L. seedlings were pretreated with gibberellins, their activity was further increased by increased acidity.Acer negundo L. seedlings responded to the acidification of their root environment with enhanced cytokinin activity only when pretreated with exogenous auxins. The differences in the responses ofA. pseudoplatanus L. and A.negundo L. to the acidification can be explained in terms of their distinct physiological properties, including changes recorded in chlorophyll content. The results obtained show that a pretreatment of maple seedlings with exogenous growth regulators can modify the effects of increased acidity in the environment on their physiological status as characterized by the level of endogenous cytokinins and gibberellins.  相似文献

Browsing by ungulates has become a hotly debated issue in many European mountain forests in the past century. Acer pseudoplatanus L. and Fraxinus excelsior L. are broadleaved tree species that are preferentially browsed by roe deer (Capreolus capreolus) in central Europe. We analyzed growth data from shaded saplings of both tree species to quantify the extent to which height growth after game browsing is reduced in subsequent, unbrowsed years in forest stands. Sixty saplings of F. excelsior and A. pseudoplatanus from forest stands at Albisriederberg (Switzerland) were available to us that had been dissected into pieces that then were split in the middle for counting tree rings and assessing ungulate damage. We fitted the von Bertalanffy growth equation to these height growth data and included a reduction factor for winter browsing. Both tree species showed significantly reduced height growth in unbrowsed years after one to several browsing events in winter, and this effect increased with the number of browsing events. Saplings with a high growth rate showed a higher growth reduction. After winter browsing, height growth of A. pseudoplatanus saplings was less affected in unbrowsed years than that of F. excelsior saplings. We conclude that browsed saplings of these species in forest stands are not able to compensate browsing-induced height loss, but that height differences between browsed and unbrowsed saplings probably increase over time. A comparison between our analysis and the parameters estimated using equations published by Eiberle for predicting age at 130 cm height suggests that our parameter values are rather conservative estimates of the growth reduction effect after winter browsing. Neither F. excelsior nor A. pseudoplatanus show a distinct pattern in browsing-induced growth reduction with respect to soil moisture, nutrient level, and altitude. We thus conclude that our results are likely to be valid for a wide range of forested sites.  相似文献

On the arable land of the Rosovka Stream alluvium in the Czech Republic, a windbreak containing native woody species, was planted within a restoration project. This study evaluates the effect of that planting of three different tree sizes, on the rate of growth. The sizes of transplants employed in this study included small (1.0–1.5 meters tall), medium-sized (2.0–2.5 meters tall) and large (with a trunk circumference of 0.1–0.12 meters). The following native tree species, from the Eurosiberian region, were used: Quercus robur L., Carpinus betulus L., Fraxinus excelsior L., Acer campestre L., Acer pseudoplatanus L., Tilia cordata Mill., and Prunus avium L. Growth rates were monitored over a five-year period. Small transplants had the highest growth; they grew approximately 0.4 meters more than transplants from the other size categories. Our results show that the selection of the size of transplant has a significant impact on development and growth. However, differences in the post-transplant responses of individual species were found.  相似文献

The acoustic technique was used in conjunction with the pressure chamber to determine the tensions causing cavitation of xylem sap in leaves of five woody angiosperms (Acer pseudoplatanus L., Alnus glutinosa L. Gaertn., Eucalyptus globulus Labill., Fraxinus excelsior L. and Rhododendron ponticum L.) and three species of herbs (Lycopersicum esculentum Mill., Plantago major L. and Ricinus communis L.). The results showed leaves of most species to suffer considerably from cavitation at sap tensions of 1.6-3 MPa. Two of the herbs, Lycopersicum and Ricinus, cavitated extensively at sap tensions below 1 MPa. Additional evidence is presented that clicks, detected by acoustic amplification, are caused by cavitation of sap in the xylem conduits. A rapid method is suggested for the determination of sap tensions in cavitating leaves and which is suitable for surveys of the critical sap tension in a large number of species.  相似文献

Summary Proteins in sieve tube exudate from Ricinus communis L., Acer pseudoplatanus L., Aesculus hippocastanum L., Cucumis melo L., and two cultivars each of Cucumis sativus L., Cucurbita pepo L. and Cucurbita maxima Duchesne were fractionated and compared using polyacrylamide gel electrophoresis. Striking differences in major exudate proteins were displayed among the genera and species examined. Even cultivars within a single species, although showing general similarities, differed in some prominent proteins. Estimated molecular weights of the major exudate proteins from each plant are presented. The effects of reducing and chaotropic agents on the aggregation and subunit composition of exudate proteins from Cucumis sativus have been investigated. The problems involved in relating structure, function and biochemistry of P-protein are discussed.  相似文献

Changes in the abscisic acid (ABA) levels in embryo axes of seeds, belonging to the orthodox (Norway maple — Acer platanoides L.) and recalcitrant (sycamore — Acer pseudoplatanus L.) categories, were investigated throughout maturation using an ELISA (enzyme-linked immunosorbent assay) test. Concentration of ABA in embryo axes substantially differed depending on species and sampling date. ABA was always higher in Norway maple except at the end of seed maturation when ABA content was similar in both species. During maturation ABA decreased in both species but the decline was more marked in Norway maple than in sycamore (11 vs. 3 fold). These species also differed in the pattern of ABA changes, which in sycamore embryo axes was very regular, while in Norway maple a sharp decrease was recorded after acquisition by the seeds of tolerance to desiccation. Dehydration of embryo axes of Norway maple caused a further significant decrease of ABA level. In contrast, in dehydrated sycamore embryo axes ABA content did not decrease, but slightly increased. The role of ABA in desiccation tolerance and dormancy of Norway maple and sycamore seeds is discussed.  相似文献

The potential use of random amplified polymorphic DNA (RAPD) was evaluated as a source of genetic markers for studying variation among four species of Panicum and within the crop species P. miliaceum and P. sumatrense. Polymorphism in RAPD markers was observed across and within species. The four species were distinct in RAPD patterns and were separated at low correlation values even with small samples involving single genotypes per species. Accessions of P. miliaceum were grouped according to geographical regions of origin. The study demonstrated that unlike isozyme and protein electrophoresis patterns, RAPD markers can be applied to studying genetic diversity, defining gene pools, and identifying cultivars for this group of millets.  相似文献

Membrane phospholipid composition was investigated in seeds of two species from the genus Acer: Norway maple (Acer platanoides L.) — tolerant to desiccation, and sycamore (Acer pseudoplatanus L.) — intolerant to desiccation, during their maturation, from 1 August to 25 September 1995, at weekly intervals. Seeds of Norway maple acquire tolerance to desiccation at the end of August ie. about 125 days after flowering (DAF). Phospholipid composition during development revealed marked differences between studied seeds. Seeds of Norway maple after acquiring tolerance to desiccation contained much more phosphatidylcholine (PC) and phosphatidylethanolamine (PE), compared to sycamore. The ratio of PC/PE in mature Norway maple seeds was evidently higher than those in sycamore. The level of unsaturated fatty acids in the phospholipid fraction substantially increased in Norway maple seeds during development and the saturation of PC and PE was less than in sycamore. The results suggest that phospholipid composition may be involved in desiccation tolerance of Norway maple seeds.  相似文献

We present 16 variable dinucleotide microsatellite markers to quantify genetic variation in the parthenogenetic gecko, Lepidodactylus lugubris. Genetic diversity at these loci was unusually high for an asexual species. Subsets of individuals produced identical genotypes across all loci. Individual loci revealed evidence of polyploidy and marked differences between observed and expected heterozygosity, indicating the presence of null alleles. These patterns conform to prior expectations based on the distant genetic relationships between the presumed sexual progenitors of L. lugubris. Comparisons with sexual relatives will be required to determine the sources of the observed genetic variation.  相似文献

Measurements were made of the levels of key glycolytic intermediates and co-factors during the growth of Acer pseudoplatanus L. cells in batch culture. Mass action ratios were calculated for the enzymes phosphoglucoseisomerase, phosphofructokinase and pyruvate kinase. The ratio for phosphofructokinase was greatly displaced from equilibrium. Major increases in the level of NADPH and ATP and energy charge were observed in the initial or lag period of culture. The data indicate that the energy generating system of the cells is most active in the lag phase.  相似文献

A recent approach to detecting genetic polymorphism involves the amplification of genomic DNA using single primers of arbitrary sequence. When separated electrophoretically in agarose gels, the amplification products give banding patterns that can be scored for genetic variation. The objective of this research was to apply these techniques to cultivated peanut (Arachis hypogaea L.) and related wild species to determine whether such an approach would be feasible for the construction of a genetic linkage map in peanut or for systematic studies of the genus. Two peanut cultivars, 25 unadapted germplasm lines of A. hypogaea, the wild allotetraploid progenitor of cultivated peanut (A. monticola), A. glabrata (a tetraploid species from section Rhizomatosae), and 29 diploid wild species of Arachis were evaluated for variability using primers of arbitrary sequence to amplify segments of genomic DNA. No variation in banding pattern was observed among the cultivars and germplasm lines of A. hypogaea, whereas the wild Arachis species were uniquely identified with most primers tested. Bands were scored (+/–) in the wild species and the PAUP computer program for phylogenetic analysis and the HyperRFLP program for genetic distance analysis were used to generate dendrograms showing genetic relationships among the diploid Arachis species evaluated. The two analyses produced nearly identical dendrograms of species relationships. In addition, approximately 100 F2 progeny from each of two interspecific crosses were evaluated for segregation of banding patterns. Although normal segregation was observed among the F2 progeny from both crosses, banding patterns were quite complex and undesirable for use in genetic mapping. The dominant behavior of the markers prevented the differentiation of heterozygotes from homozygotes with certainty, limiting the usefulness of arbitrary primer amplification products as markers in the construction of a genetic linkage map in peanut.  相似文献

Summary Four species of Geometridae showed similar consumption rates and approximate digestibilities (100 x ingestion-egestion/ingestion) of sycamore leaves (Acer pseudoplatanus L.). A significant decline in E.C.I. (100 x growth/ingestion) and E.C.D. (100x growth/ingestion-egestion) with increasing larval weight is reported. The earlier descent from the sycamore canopy ofErannis defoliaria Cl. andOperophtera brumata L. larvae, to pupate in the soil, in comparison withAlsophila aescularia D. and S. andAgriopis aurantiaria Hb. larvae is discussed in relation to small differences in the utilization efficiencies of final instar larvae.   • 1 The horse‐chestnut leaf miner Cameraria ohridella is an invasive moth in Europe and a serious pest of horse‐chestnut Aesculus hippocastanum. The moth also occasionally attacks sycamore maple Acer pseudoplatanus, when situated beside infested horse‐chestnuts.
  • 2 The main objective of the present study was to provide an overview of the relationship between C. ohridella and A. pseudoplatanus and to determine whether C. ohridella has the potential to shift to this native tree.
  • 3 In the field, females oviposit on different deciduous tree species. Although less frequently attacked than A. hippocastanum, A. pseudoplatanus was clearly preferred for oviposition over 12 other woody species investigated.
  • 4 Surveys in Europe demonstrated that the majority of A. pseudoplatanus trees found beside infested A. hippocastanum had mines of C. ohridella, even though more than 70% of the larvae died within the first two instars. Attack rates and development success greatly varied from site to site. Attack levels on A. pseudoplatanus were not always correlated with those on A. hippocastanum, and mines on A. pseudoplatanus were sometimes observed beside weakly‐infested A. hippocastanum.
  • 5 Field observations, experimental exposure of A. pseudoplatanus saplings and rearing trials in a common garden study showed that individual trees may vary in their susceptibility to C. ohridella, whereas there was no evidence that C. ohridella populations vary in their performance on A. pseudoplatanus.

Flow cytometric analyses of nuclear DNA levels were carriedout during development, stratification and germination of dormantseeds from three tree species with contrasting characteristics.Norway maple (Acer platanoides) and sycamore (Acer pseudoplatanus)have orthodox (desiccation-tolerant) and recalcitrant (desiccation-sensitive)storage behaviours, respectively, and require only a periodof cold to break dormancy, whereas, orthodox cherry (Prunusavium) seeds require an initial warm period before cold stratificationto fully stimulate germination. Whole embryos and radicle tipsof both Norway maple and sycamore were found to have stablehigh levels of 4C DNA during the latter stages of developmentand both contained nuclei arrested at the 2C and 4C levels atmaturity. Mature cherry embryos had nuclei predominantly arrestedat the 2C level. This suggests that the acquisition of desiccationtolerance is not correlated with the arrest of the cell cycleat any particular nuclear DNA level. Neither DNA replicationin radicle cells nor germination occurred when seeds were maintainedmoist at a constant 20 °C. However, in the late stages ofcold treatment during stratification, nuclear DNA levels inradicle cells changed in advance of radicle emergence in theorthodox Norway maple and cherry, whereas in the recalcitrantsycamore, change was not recorded until after radicle emergence.These results show that DNA replication has potential use asan indicator of the progress of tree seeds through stratificationtreatments used to break some types of dormancy. Theory predicts that genetic variation should be reduced at range margins, but empirical support is equivocal. Here, we used genotyping‐by‐sequencing technology to investigate genetic variation in central and marginal populations of two species in the marine gastropod genus Crepidula. These two species have different development and dispersal types and might therefore show different spatial patterns of genetic variation. Both allelic richness and the proportion of private alleles were highest in the most central populations of both species, and lower at the margin. The species with low dispersal, Crepidula convexa, showed high degrees of structure throughout the range that conform to the pattern found in previous studies using other molecular markers. The northernmost populations of the high‐dispersing species, Crepidula fornicata, are distinct from more central populations, although this species has been previously observed to have little genetic structure over much of its range. Although genetic diversity was significantly lower at the range margin, the absolute reduction in diversity observed with these genomewide markers was slight, and it is not yet known whether there are functional consequences for the marginal populations.  相似文献

The role of soluble sugars in desiccation tolerance was investigated in seeds of two species from the genus Acer: Norway maple (Acer platanoides L.) — tolerant and sycamore (Acer pseudoplatanus L.) — intolerant to dehydration. During two years of observations it was found that seeds of Norway maple acquire desiccation tolerance at the end of August i.e. about 125 days after flowering (DAF). During seed development, the transition from intolerant to tolerant state in Norway maple seeds was accompanied by the accumulation in seed tissues of raffinose, stachyose and sucrose. The sucrose/raffinose ratio in Norway maple seeds was lower than in sycamore. In mature Norway maple seeds sucrose and raffinose contents were higher than in sycamore. It was concluded, that soluble sugars such as sucrose, raffinose and stachyose may play an important role in desiccation tolerance and/or intolerance of Norway maple and sycamore seeds. Differential thermal analysis (DTA) was used to study the relationship between desiccation sensitivity and the state of water in seed tissues. The level of non-freezable water was the same in both analysed seed species, but the temperature of water crystallization during desiccation was lower in sycamore seeds.  相似文献

Abstract

Soil microfungi in homogeneous areas of three plant communities of North Italy.—The soil microfungi associated with three different community types in Prealpine region of North Italy were investigated. Frequency values for the taxonomic entities isolated were calculated, based on occurrence at sampling sites, in each community. Most of the entities were quantitatively rare. The microfungal communities associated with Acer pseudoplatanus and Fraxinus excelsior, with Ostrya carpinifolia and with Fagus sylvatica appeared to be a distinctive group, even if many of them contained a substantial number of species wich are common with other vegetation types.  相似文献

Several central European species have failed to reach the British Isles because of the early separation from the continent. The two tree species Fagus sylvatica and Carpinus betulus reached southern England but were unable to spread much further. Other species, such as Acer pseudoplatanus, were only relatively recently introduced. Recent distribution maps for Fagus and Acer show an almost uninterrupted distribution in the whole British Isles. The beech must be considered to be an important element of the present potential natural vegetation; it has been planted widely and regenerates freely. The woodlands of Brittany may provide a model to enable us to visualize the possible species composition and appearance of these potential Atlantic beech woods.Nomenclature of vascular taxa follows Ehrendorfer (1973).  相似文献

Tree species-rich forests are hypothesised to be less susceptible to insect herbivores, but so far herbivory–diversity relationships have rarely been tested for tree saplings, and no such study has been published for deciduous forests in Central Europe. We expected that diverse tree communities reduce the probability of detection of host plants and increase abundance of predators, thereby reducing herbivory. We examined levels of herbivory suffered by beech (Fagus sylvatica L.) and maple saplings (Acer pseudoplatanus L. and Acer platanoides L.) across a tree species diversity gradient within Germany’s largest remaining deciduous forest area, and investigated whether simple beech or mixed stands were less prone to damage caused by herbivorous insects. Leaf area loss and the frequency of galls and mines were recorded for 1,040 saplings (>13,000 leaves) in June and August 2006. In addition, relative abundance of predators was assessed to test for potential top-down control. Leaf area loss was generally higher in the two species of maple compared to beech saplings, while only beech showed a decline in damage caused by leaf-chewing herbivores across the tree diversity gradient. No significant patterns were found for galls and mines. Relative abundance of predators on beech showed a seasonal response and increased on species-rich plots in June, suggesting higher biological control. We conclude that, in temperate deciduous forests, herbivory–tree diversity relationships are significant, but are tree species-dependent with bottom-up and top-down control as possible mechanisms. In contrast to maple, beech profits from growing in a neighbourhood of higher tree richness, which implies that species identity effects may be of greater importance than tree diversity effects per se. Hence, herbivory on beech appeared to be mediated bottom-up by resource concentration in the sampled forest stands, as well as regulated top-down through biocontrol by natural enemies.
