Seven Italian populations of twaite shad Alosa fallax from Northern and Central Italy were investigated to assess genetic diversity by Cytochrome b (Cytb) gene sequencing. The two ecotypes historically referred to landlocked and anadromous populations were investigated for the first time from a genetic point of view, to clarify their phylogenetic relationships. Moreover, results obtained from populations coming from separated Adriatic and Tyrrhenian basins were compared with data assessed in samples of allis shad Alosa alosa from the Atlantic basin. All the Italian samples were recognized at species level as A. fallax, differing for five mutations from A. alosa. The analyses confirmed the occurrence of a single phylogenetic lineage and of a single species within Italian waters, in both landlocked and migratory populations. The minimum spanning network identified six haplotypes for A. fallax and two haplotypes for A. alosa. The neighbour-joining tree and the maximum likelihood on the Cytb gene sequences confirmed two distinct lineages for A. alosa and A. fallax, without evidence of a separation at specific level within the A. fallax group. A weak separation due to incipient population differentiation was detected between anadromous and landlocked Italian populations, supporting the idea of a recent separation. The molecular data herein collected do not support the existence of the already controversial incipient species Alosa agone. Despite this, the two ecotypes could be considered as different management units from a conservation viewpoint.  相似文献

Habitat alteration has been implicated in driving hybridization between the sympatric migratory shads Alosa alosa and Alosa fallax. Morphological and molecular evidence is consistent with hybridization across the overlapping range of these species, but the temporal extent of hybrid occurrence and genetic consequences for populations have not been explored. Using eight nuclear microsatellite loci and samples collected between 1989 and 2008 in the Solway Firth (UK), we genetically identified hybrids, studied temporal changes in their frequency, and explored changes in allele frequencies of parental populations. These molecular data confirmed the hybrid status of individuals identified using morphology (number of rakers on the outer gill arch), and enabled separation of hybrids from purebred individuals. Mitochondrial cytochrome-b sequencing revealed the presence of two haplogroups, each predominantly occurring in one species. Heterospecific haplotypes were found in 22.3 and 12.8% of A. alosa and A. fallax individuals, respectively, consistent with backcrossing and suggesting that hybrids are fertile. On average, microsatellite-identified hybrids comprised 12.7% of all samples, but when individuals with cytonuclear discordance were also considered introgressed on average 25.4% of individuals were of hybrid ancestry. Overall, allelic richness remained largely unchanged within species, but there were declines in the inbreeding coefficient (F IS) of both species and episodes of significant temporal allelic frequency change. Hybrids sampled between 2004 and 2008 showed no evidence of lower fecundity relative to purebred individuals. Together, results suggest that hybridization between shad species in northern Europe is prevalent, and has been ongoing over at least two decades. The challenge is now to understand the extent to which observed patterns are linked to immigration from other populations, and the mechanisms that have prevented species collapse despite apparent hybrid fertility and longstanding introgression of neutral markers.  相似文献

Four Special Areas of Conservation (SACs) have been designated in the Republic of Ireland for Twaite shad, Alosa fallax (Lacépède), based on recent historical information on spawning activity—the estuarine reaches of the Rivers Munster Blackwater, Suir, Barrow–Nore and Slaney. The spawning status of some of these populations is considered to have declined substantially in recent years, and no known spawning areas of Allis shad, Alosa alosa (L.), are known for Ireland. Data are presented confirming the presence of both Allis shad and Twaite shad in all four SACs. The majority of the material has come as by-catch from estuarine commercial salmon netsmen. Angling is the principal source of material from the R. Barrow. Examination of scale and gonad material of Allis shad indicates some degree of readiness for spawning as well as spawning having taken place. Gonadosomatic index (GSI) data from the R. Barrow for Twaite shad in the May spawning period (2000–2002) had mean GSIs of 13.7–15% with maxima in the range of 21–28%. GSI values of 18% and 22% have been recorded in the July period for individual Allis shad from the upper estuaries of the R. Slaney and R. Suir, respectively, with both fish displaying gonad maturity stage V or VI (after Pina, 2000). Low GSI values have been recorded from other Allis shad in the June–July period displaying gonad condition varying from stage III (immature) to stage VIII (spent). These findings point to a possibility that Allis shad do engage in spawning activity, whether mono-specifically or with Twaite shad. The possibility of the latter is supported by gill raker count data presented. These show clearly defined value ranges for Twaite and for Allis as well as a series of fish with intermediate counts. The apparently low population levels of Twaite shad within the SACs identify a need to examine what measures might be undertaken to ensure the favourable conservation status of this species. Mercury concentration [Hg] was assessed in 20 adult Allis shad Alosa alosa (54–59 cm) and 20 adult Twaite shad Alosa fallax (36–44 cm) collected during their spawning migration in the Dordogne and the Garonne rivers (France). [Hg] was measured in the gills, dorsal muscle, liver and kidney. Twaite shad exhibited higher [Hg] than Allis shad. Median [Hg] values were [Hg]Gills = 0.33 μg g−1 (dw), [Hg]Muscle = 1.22 μg g−1, [Hg]Liver = 1.99 μg g−1, [Hg]Kidney = 1.93 μg g−1 for Twaite shad and [Hg]Gills = 0.06 μg g−1, [Hg]Muscle = 0.20 μg g−1, [Hg]Liver = 1.18 μg g−1, [Hg]Kidney = 1.08 μg g−1 for Allis shad. In order to understand such differences, we investigated some life history traits of the two species: migratory history, age (3–6 years), size at age (an expression of growth) and the number of spawning events (0–2 events). The difference in estuarine residence time between the juveniles of both species, which was assumed to influence [Hg], was investigated using otolith Sr:Ca ratio. The microchemical analysis revealed a significant difference in the residence time of juveniles in the estuary (medians are 21 d and 10 d for Twaite shad and Allis shad, respectively) but this residence time seems too short to influence [Hg] (Allis shad: r spearman < 0.128; Twaite shad: r spearman < −0.340). As both species show the same age structure, the influence of age on [Hg] was negligible. The literature shows that the differences in growth and in the number of spawning events reported in our study are in favour of a higher [Hg] for Twaite shad than for Allis shad. Although trophic status was not investigated here, the literature reveals that it is another factor that could produce higher [Hg] in Twaite shad, since its diet includes higher trophic levels than Allis shad. Sixteen microsatellite loci were identified and characterized for American shad (Alosa sapidissima). The number of alleles per locus observed ranged from eight to 32 and averaged 15.4 alleles. Average observed heterozygosity was 81.1%. The markers were screened using four other species from the family Clupeidae. Amplification success among Alosa species was 79.2% with 81.6% polymorphism among those markers that amplified successfully. Amplification success was poor in Dorosoma (31.3%). Due to allelic diversity and estimates of heterozygosity, these markers can be useful in A. sapidissima for population level analyses, parentage assignment and broodstock management.  相似文献

The paper reviews the past, present and future of eight anadromous fish species inhabiting the Lower Rhine (The Netherlands), viz. -sturgeon (Acipenser sturio), whitefish and houting (Coregonus lavaretus, C. oxyrinchus), smelt (Osmerus eperlanus), allis and twaite shad (Alosa alosa, A. fallax), sea trout (Salmo trutta) and salmon (Salmo salar). All species are under threat or became extinct (e.g. sturgeon, allis shad). It is not possible to single out a specific factor for the decline or disappearance. A combination of factors is responsible, as the degradation of the spawning and nursery areas, river correction for shipping, building of sluices and hydropower dams, extraction of sand and gravel and river pollution. The likelihood that a species will return via natural recovery, or restocking is assessed. The return of the sturgeon is unlikely. Present observations of sturgeon can be attributed to releases of unwanted sturgeon hybrids. A natural stock of coregonids in Dutch waters seems not feasible any more due to irreversible habitat degradation. Present day catches originate from German releases. The anadromous smelt, heavily reduced in numbers, still inhabits some of our waters, the non-migratory smelt is still very common. The allis shad is extinct and unlikely to recover. The species never spawned in the Dutch part of the Rhine. Twaite shad, declining in numbers, are still observed in the lower reaches of Rhine and Meuse. Sea trout is presumably still present in the same varying numbers as before. Spawning in our waters has not been documented. The salmon, once fished in large numbers, is now the subject of restocking programmes in Germany. Observations of individuals can partly be attributed to these programmes but also to straying salmon. Restocking programmes should be considerably improved before noticeable success is to be met.  相似文献

Ten polymorphic loci were isolated and characterized from the milkweed, Asclepias syriaca L., of North America. These loci successfully cross-amplified in A. exaltata L. Polymorphism ranged from two to 16 alleles per locus per species in 68 individuals of A. syriaca and 56 individuals of A. exaltata. Expected heterozygosities ranged from 0.017 to 0.851 and significant deviations from Hardy–Weinberg equilibrium were found for two and three loci in A. syriaca and A. exaltata, respectively. No linkage disequilibrium was detected. These markers should prove useful for assessing population genetic structure and interspecific gene flow in these and other species of Asclepias.  相似文献

This study investigates length–weight and length–length relationships of four shad species (Alosa caspia caspia, Alosa immaculata, Alosa maeotica, Alosa tanaica) captured in the western Black Sea coast of Turkey. A total of 686 specimens, 355 females and 331 males, were caught by gill net, trammel net and longline from 2006 through 2007 along the western Black Sea coast of Turkey. The relationships of total length (TL), fork length (FL) and standard length (SL) are presented for inhabiting in the Black Sea four shad species and the relationships between total length (TL) and body weight (BW) from the two localities, Sile and Karasu in the western Black Sea coast of Turkey. The Length–weight relationships generally indicated positive allometric growth in Alosa species for both males and females and an isometric growth for A. immaculata females. The coefficient b ranged from a minimum 2.97 for females of A. immaculata to a maximum 3.75 for both sexes A. c. caspia. Results indicated that the length–length relationship between the three length measurements were highly correlated (r2 > 0.99, P < 0.001).  相似文献

Individual migration behaviour during the juvenile and adult life phase of the anadromous twaite shad Alosa fallax in the Elbe estuary was examined using otolith Sr:Ca and Ba:Ca profiles. Between hatching and the end of the first year of life, juveniles showed two migration patterns. Pattern one exhibited a single downstream migration from fresh water to the sea with no return into fresh water. In contrast, pattern two showed a first migration into the sea, then a return into fresh water and, finally, a second downstream migration into marine water. This first report of migration plasticity for A. fallax points to different exposure times to estuarine threats depending on the migration strategy. In adults, high Sr:Ca and low Ba:Ca in the majority of individuals confirmed prior reports of a primarily marine habitat use. Patterns reflecting spawning migrations were rarely observed on otoliths, possibly due to the short duration of visits to fresh water.  相似文献

After stocking with larvae from the Gironde–Garonne–Dordogne population, in 2013, three young‐of‐the‐year Allis shad Alosa alosa, probably originating from natural reproduction, were documented for the first time in a period of nearly 100 years in the River Rhine. In 2014, a further increase was observed when 57 juveniles and eight adults were caught; seven of these eight adults were derived from stocking, indicating the success of stocking measures within the framework of the EU‐LIFE project.  相似文献

Abstract

Nitrogen deposition may cause shifts in the Sphagnum species composition of bogs, ultimately affecting the conservation value of these systems. We studied the effects of N and P on the expansion of S. fallax and S. flexuosum in bogs. We related historical census data of S. fallax, S. flexuosum, and four of their accompanying species to changes in N deposition. In addition, we conducted two fertilization experiments with N and P; one at a low deposition site with S. flexuosum and one at two high deposition sites with S. fallax. Finally, we related existing data on capitulum N and P concentrations of S. fallax to its abundance in the field.

A relative increase in observed frequency of S. fallax coincided with an historical increase in N deposition in the Netherlands. There was no indication that S. fallax consistently outcompeted one of the other five Sphagnum species; the observed frequency of the Sphagnum species analysed was rather stable with time. The census data on S. flexuosum did not indicate a response to N deposition, but the species expanded at the low N deposition site when extra N was applied. In contrast, the expansion of S. fallax at the high deposition sites was limited by P. Organic nutrient concentrations suggested that when S. fallax can maintain a capitulum N concentration of 7 mg g-1 or higher and a P concentration of 0.7 mg g-1 or higher the species can grow to dominate.

We conclude that S. fallax will gradually colonize an increasing number of new habitats in areas with a low, albeit increasing, N deposition, but will only grow to dominate when P supply is adequate. Then, the expansion of S. fallax may lead to ousting of the other Sphagnum species present.  相似文献

A total of 45 microsatellite loci from yellow perch, Perca flavescens, were isolated and characterized. Among the 45 microsatellite loci, 32 had more than two alleles. A wild population of P. flavescens (n = 48) was used to examine the allele range of the microsatellite loci. Mendelian inheritance of alleles was confirmed by examining the amplified products in pair‐mated families. The number of alleles for the 32 polymorphic loci varied from two to 16, and observed heterozygosity ranged between 0.024 (YP79) and 0.979 (YP60). Cross‐species polymorphic amplification in four other Percidae species was successful for 22 loci.  相似文献

A study was conducted to determine the effects of six constant temperatures (15, 20, 25, 30, 35 and 40°C) on the post‐diapause embryonic development and the hatching time in three grasshopper species –Omocestus haemorrhoidalis (Charp.), Calliptamus abbreviatus Ikonn. and Chorthippus fallax (Zub.) – from the Inner Mongolian steppe. The results indicate that the species differ in the developmental rates, survival curves and cumulative hatching probabilities. The eggs of O. haemorrhoidalis had the fastest developmental rate with a low developmental threshold temperature of 9.9°C and the sum of effective temperature (SET) 211.2 degree‐days (DD). The corresponding values were 10.9°C and 210.6 DD for C. abbreviatus, 10.5°C and 240.2 DD for Ch. fallax respectively. The SET at which 50% of post‐diapause eggs hatched were 252.0 DD for O. haemorrhoidalis, 262.8 DD for C. abbreviatus, and 273.3 DD for Ch. fallax. The predicted maximal hatch ability of O. haemorrhoidalis (91.17%), C. abbreviatus (75.67%) and Ch. fallax (94.07%) occurred at 23.7, 29.0 and 31.3°C, respectively. The thermal death points of each species were reached at 43.3, 45.0 and 48.6°C. The optimal temperature ranges were 12.2–35.2°C for O. haemorrhoidalis, 21.7–36.3°C for C. abbreviatus and 20.9–41.7°C for Ch. fallax respectively. These results suggest that O. haemorrhoidalis adapt to hatch at a lower temperature range, C. abbreviatus adapt to mid‐temperature range, while Ch. fallax adapt to hatch at a higher temperature range. Although the SET of Ch. fallax is more than that of the other two species, it is not sufficient to explain the hatching sequence of the species in springtime. The results also indicate that Ch. fallax and O. haemorrhoidalis have wider adaptive temperature range than C. abbreviatus.  相似文献

Abies firma is a dominant coniferous tree species endemic to Japan. We isolated eight microsatellite loci from needles of this tree species and tested their polymorphism among 26 A. firma individuals. Six of them showed polymorphism, with two to 16 alleles per locus. Their expected heterozygosities ranged from 0.075 to 0.922. Moreover, interspecific amplification among Abies sachalinensis, Abies mariesii and Abies veitchii was successful in majority of the isolated loci, suggesting that these loci may be useful for characterization of other fir species.  相似文献

Nine polymorphic microsatellite loci were identified in the tropical plant Vasconcellea ×heilbornii and used to estimate allelic diversity in two populations of southern Ecuador. Allelic richness ranged from two to five alleles, observed heterozygosity ranged from 0.150 to 0.947 and expected heterozygosity ranged from 0.186 to 0.701. Most of these markers also amplified microsatellite loci in two other Vasconcellea species (Vasconcellea stipulata and Vasconcellea cundinamarcensis). Hence, these markers will be useful for population genetic analysis and the evaluation of genetic diversity and gene flow in these species.  相似文献

Eight microsatellite loci were identified and characterized for the endangered Machilus pseudokobu (Lauraceae), an endemic tree species of the Bonin Islands. The observed number of alleles at each locus ranged from 1 to 20 with an average of 6.2, and the expected heterozygosity ranged from 0.00 to 0.83 with an average of 0.47. All eight loci were screened in cross-amplification tests for two other endemic Machilus species that also inhabit the Bonin Islands. All loci were successfully amplified in these species.  相似文献

We developed microsatellite loci for the southern pine beetle (Dendroctonus frontalis). Twelve microsatellite loci were identified. Eight loci were polymorphic and sufficiently variable in 62 individuals (expected heterozygosity ranged from 0.707 to 0.880) to investigate population structure. All loci conformed to HWE except Dfr‐14, which showed heterozygote excess, and no two loci deviated from linkage equilibrium. The loci were tested for cross‐species amplification in four species of Dendroctonus (D. valens, D. terebrans, D. brevicomis, and D. ponderosae). Seven loci were polymorphic in at least one of the species tested.  相似文献

Kearney's bluestar (Amsonia kearneyana) is a highly endangered herbaceous perennial in the family Apocynaceae. The species is found only in the Baboquivari Mountains of southern Arizona. We report the isolation and development of 12 microsatellite loci for Kearney's bluestar. Numbers of alleles ranged from two to four and observed heterozygosities ranged from 0.20 to 0.80 in the nine loci found to be polymorphic in the test population. All loci were also tested for cross‐amplification in five other Amsonia species representing two subgenera from the southwestern United States. Some loci that were not polymorphic in the Kearney's bluestar were polymorphic in other species.  相似文献

Highly polymorphic genetic markers provide a useful tool for estimating genetic parameters in studies of the evolution of sociality in insects. We isolated and characterized 12 polymorphic microsatellite loci in the invasive ant, Wasmannia auropunctata, and described experimental conditions for PCR (polymerase chain reaction) multiplexing and simultaneously genotyping these loci in two sets of five and seven markers. The number of alleles per locus ranged from two to 14 and the observed heterozygosity ranged from 0.233 to 0.967. Moreover, results of cross‐species amplification tests are reported in three other species of Wasmannia and in two species of the genus Allomerus.  相似文献
