Exoneura bicolor is a univoltine, facultatively social bee exhibiting a solitary/quasisocial/semisocial colony polymorphism (Schwarz, 1986, 1987). Intracolony relatedness in semisocial colonies has been previously estimated at 0.49 ± 0.06 (Schwarz, 1987), although the crucial relatedness between altruists and the brood that they rear will be about half this value. This value is unlikely to be increased by the preferential rearing of only close relatives (Schwarz, 1988a) and no known morphological specializations preclude workers from reproducing in this species. Hamilton (1972, 1975) suggested that relatedness may be increased through population subdivision, if this leads to significant inbreeding and increased between-colony genetic variance. The same process may also operate at higher levels of population structure (e.g., Wade, 1978). Population structure and intracolony relatedness in E. bicolor were investigated in seven localities in southern Victoria, Australia, to determine if inbreeding at any level of population structure was contributing to relatedness between altruists and beneficiaries within these colonies. Population structure was described using hierarchical F-statistics and an identity by descent measure, developed by Queller and Goodnight (1989), was used to estimate intracolony relatedness. It was found that inbreeding was not contributing to between-group genetic variance, at any level, in a consistent manner across localities. Therefore relatedness, considered in isolation, does not seem sufficient to account for the presence of worker behavior. It is suggested that large benefits for group living may be responsible for maintaining altruistic behavior, in part, in this species. Significant heterogeneity among localities for all F-statistics estimated in our analysis was found and this may be attributable to stochastic elements such as cofounding behavior and the low percentage of males in the brood. The possible consequences of such heterogeneity in population structure for the maintenance of altruism in E. bicolor are discussed.  相似文献

Summary Previous studies of the allodapine beeExoneura bicolor have utilised a predominantly social population from tall montane forests. In this report we examine aspects of the sociality and biology of a population from subcoastal heathland in western Victoria, Australia. Entire colonies, nesting inXanthorrhoea minor, were collected at monthly intervals over 12 months between February 1989 and January 1990. In addition, three collections fromMelaleuca squarrosa were made over three consecutive winter months. The heathland population differs from the montane population in several aspects associated with sociality: (i) it shows lower levels of co-occupancy in both newly founded and re-used nests; (ii) foundress dispersal occurs over a larger proportion of the year; and (iii) there may be a partical second brood produced in late summer, during and after adult eclosion of the first brood. The occurrence of a partial second brood gives rise to opportunities for sib-rearing and eusociality. Colony sizes tend to be larger inM. squarrosa thanX. minor, but still smaller than colonies from the montane population. Egg-laying commences several weeks earlier in multifemale colonies than in single female colonies, before spring dispersal begins. It appears that multifemale colonies are more successful at preventing brood loss than single females and that the mean number of brood per female is larger in multifemale colonies than in single female colonies. These two populations, which display differences in several life history traits, provide an ideal system for studying the mechanisms maintaining social behavior.  相似文献

Summary Exoneura bicolor is a univoltine allodapine bee common in montane forests of southern Australia, where it exhibits a semisocial/quasisocial colony organization. Within-nest behaviour in postemergence autumn nests ofExoneura bicolor was recorded with the aim of studying behavioural specialization in pre-reproductive colonies. Ten complete colonies were transferred to purpose-built observation nests shortly before brood eclosion in late summer. Behaviour within observation nests was recorded for periods of up to 44 days after establishment, covering a period when colonies are preparing for overwintering. Dispersal of females and brood rearing do not occur at this time, although some females may become inseminated. Analyses of data using multivariate techniques indicated four distinguishable behavioural castes, designated here as Guards, Nest Absenters, Nest Modifiers and Non-recruits. This represents a higher degree of behavioural specialization than recorded to date for other allodapines. Behaviours performed by Guards and Nest Absenters are likely to involve considerable risks, but benefit the colony as a whole, so that some nestmates in prereproductive colonies exhibit altruism that frequently aids adult siblings or cousins. The males in our study were fed by females via trophallaxis and two of the males participated in nest maintenance tasks. Our results suggest that autumn colonies ofE. bicolor form well-integrated behavioural units even though brood rearing does not commence until the following spring.  相似文献

Montane populations of the Australian allodapine bee, Exoneura bicolor, are characterized by high levels of cooperative nesting and strongly female-biased sex ratios. A conspecific population from heathland shows much lower levels of cooperative nesting and lower levels of female bias. In both habitats, sex-ratio bias is greatest in the smallest brood sizes and becomes successively less biased in larger broods. Parity is approached in the largest heathland colonies, but not for any brood-size category in montane areas. Adult intracolony relatedness is moderately high for colonies in both reused and newly founded nests in the montane habitat, but probably low or zero for newly founded nests in heathland. Colony efficiency, measured as the number of brood per adult, increases with colony size in both habitats, suggesting that cooperation between females increases mean female fitness. It is argued that patterns of sex allocation are consistent with nonlinear fitness-return models, in which the mean reproductive value of daughters increases with the number of daughters produced in a brood. Such increases probably arise from a number of social interactions, including cooperative brood defense, increased task efficiency, and lower per capita costs in nest construction. The term “local fitness enhancement” is introduced here to describe these effects collectively. The female-biased ratios should lower selective thresholds for sib-directed altruism, at least in the earlier stages of colony development. It is argued that local fitness enhancement facilitates eusociality in allodapine bees and could also play a role in other haplodiploid taxa, provided cooperative nesting largely involves sisters, colony efficiency increases with colony size, and optimal colony sizes are only achieved after two or more generations after founding.  相似文献

Despite the role of Australian native bees in important ecological processes, surprisingly little is known of their population structuring. In this study five microsatellite loci were used to investigate genetic structuring of the allodapine bee Exoneura robusta sampled from four locations (maximum pairwise distance c . 35 km) in the Mountain Ash forests of Victoria. Although E. robusta would seem to have high dispersal ability, several analyses show significant population subdivision and a strong pattern of isolation-by-distance from which limited gene flow was inferred. Limited gene flow was not associated with inbreeding at the within-colony level, and within-colony genetic structure implied co-founding, multiple breeding pairs and some degree of reproductive skew. Strong population structure at such fine scales suggests that substantially divergent populations are likely within the extensive distribution currently ascribed to E. robusta .  相似文献

Two previously reported non‐synonymous coding single nucleotide polymorphisms (SNPs) of bovine stearoyl‐CoA desaturase (delta‐9‐desaturase) (SCD) (c.878C>T) and fatty acid synthase (FASN) (g:17924A>G) were assessed for their associations with 72 individual and 12 groups of fatty acids in brisket adipose tissue of 223 Canadian commercial cross‐bred beef steers. It was found that the ‘CC’ genotype of the SCD SNP was significantly associated with lower concentrations of saturated fatty acids (SFA) including 10:0, 14:0 and 20:0, higher concentrations of monounsaturated fatty acids including 9c‐14:1, 12c‐16:1 and 13c‐18:1, higher concentrations of polyunsaturated fatty acids (PUFA) including 9c,15c‐18:2, 10c,12c‐18:2, 11c,13t‐18:2 and 12c,14t‐18:2, but lower concentrations of other PUFA of 9c,13t/8t,12c and 20:2n‐6 (P < 0.05). The ‘AA’ genotype of the FASN SNP was significantly associated with higher concentrations of SFAs of 10:0, 12:0, 13:0, 14:0 and 15:0, lower concentrations of unsaturated fatty acids of 9c‐18:1 and 20:3n‐6, and higher concentrations of unsaturated fatty acids of 9c‐14:1 and 12c‐16:1 (P < 0.05). Significant epistatic effects between the SCD and FASN SNP genotypes were also found for several fatty acids including 10:0, 23:0, 6t/7t/8t‐18:1, 12t‐18:1, 13t/14t‐18:1, 16t‐18:1, total trans18:1 and 9c,13t/8t,12c‐18:2 (P < 0.05). These results further suggest that SCD and FASN are strong candidate genes influencing fatty acid composition in beef cattle.  相似文献

Objective: To survey the present state of undergraduate teaching in the domain of gerodontology in Germany, Switzerland and Austria. Study participants: All universities of Austria (A), Germany (D) and Switzerland (CH). Protocol: A questionnaire on undergraduate teaching in gerodontology was mailed to all Deans (A: n = 3; CH: n = 4; D: n = 31) and all independent departments except paediatric dentistry and orthodontics (A: n = 11; CH: n = 15; D: n = 111). Results: The questionnaires were completed and returned by 29 Deans (A: n = 2; CH: n = 4; D: n = 23) and 102 departments (A: n = 7; CH: n = 8; D: n = 87). In Austria, gerodontology is a very small component of the dental curriculum and the Deans did not want this to be increased. Most German universities claimed to teach some aspects of gerodontology to undergraduate students and 87.4% of the Deans voted for separate lectures in gerodontology. In Switzerland, gerodontology seems well established. The results of questionnaires from the independent departments revealed that in all three countries lectures were more prevalent (A: n = 0; CH: n = 4; D: n = 6) than practical training in nursing homes (A: n = 0; CH: n = 3; D: n = 6). Conclusion: Considering the demographical shift which is leading to an increasing proportion of elderly in the population, the weighting of gerodontology in the undergraduate dental curriculum should be considered for revision in Austria and Germany.  相似文献

The sex pheromone of Phyllonorycter ringoniella (Matsumura) (Lepidoptera: Gracillariidae) has been identified to be a blend of (Z)‐10‐tetradecenyl acetate (Z10‐14:OAc) and E4,Z10‐tetradecadienyl acetate (E4,Z10‐14:OAc) in Japan, Korea, and China. However, the commercial product based on previous results is not attractive enough to be used for monitoring and controlling apple leafminer populations in the field. We re‐investigated the attractiveness of the two pheromone components, singly and in blends, in apple orchards in Shangdong and Shaanxi, the main apple‐growing provinces in China. Our results revealed that Z10‐14:OAc alone was not attractive to Pringoniella male moths in the field, but E4,Z10‐14:OAc alone not only was strongly attractive but caught more males than any of the blends of Z10‐14:OAc and E4,Z10‐14:OAc tested. The most attractive blend ratios differed slightly for the two locations. No clear dose–response relationship was obtained for the 2:8 blend of Z10‐14:OAc and E4,Z10‐14:OAc. However, the dose–response field study of E4,Z10‐14:OAc alone showed that 1 mg per lure achieved the highest moth catch. These findings differ from the previous report of the best pheromone blend in China. Our data showed that E4,Z10‐14:OAc is the major component of the pheromone of Pringoniella.  相似文献

The fae1 gene codes for KCS (β-keto-acyl-CoA synthase), the candidate enzyme for elongation of oleic acid to eicosenoic acid and erucic acid (C22:1) in various oilseed species. Degenerate primers for the fae1 gene were used to amplify and clone fae1 gene homologs in high and zero C22:1 Sinapis alba. Under stringent PCR conditions, a polymorphism was revealed between the two genotypes and was mapped as a fae1 marker in an F2 population derived from a cross between high and zero C22:1 S. alba. The fae1 marker co-segregated with C22:1 content and the C22:1 phenotypic locus. In addition, a set of 11 RAPD markers for C22:1 in S. alba was identified. Cloning and sequencing of the fae1 alleles in high and zero C22:1 S. alba revealed two amino-acid substitutions specific to zero C22:1 S. alba. The underlying nucleotide substitution for one of the amino-acid substitutions and an adjacent silent nucleotide substitution were used to design primers for allele-specific amplicons for both the wild-type and zero C22:1 alleles. The two diagnostic PCR tests are reliable selection tools to identify S. alba carrying one or both of the wild-type and mutant C22:1 alleles of the KCS gene. Abstract— Four glycoprotein:glycosyl transferases (a fetuin:N-acetylglucosaminyl transferase; a bovine submaxillary mucin: N-acetylgalactosaminyl transferase; a collagen: glucosyl transferase and an orosomucoid: galactosyl transferase) were purified 34-, 45-, 37- and 47-fold, respectively, from synaptosomes prepared from guinea pig cerebral cortex. Purifications were achieved by centrifugation and by column chromatography on Sephadex G-100 and G-150 of 0 , 1% (w/v) Triton X-100 extractsof the purified cerebral cortical synaptosomes. The enzymes were separated from endogenous acceptors and were highly specific for specific macromolecular acceptors; small molecules were ineffective as acceptors. The fetuin: N-acetylglucosaminyl transferase functioned only with fetuin minus N-acetylneuraminic acid, galactose and N-acetylglucosamine; the bovine submaxillary mucin: N- acetylgalactosaminyl transferase with bovine submaxillary much minus N-acetylneuraminic acid and N-acetylgalactosamine; the collagen: glucosyl transferase with collagen minus glucose; and the orosomucoid: galactosyl transferase with either orosomucoid minus N-acetylneuraminic acid and galactose or fetuin minus N-acetylneuraminic acid and galactose. Each transferase required a specific (XDP)-monosaccharide for transfer. The transferases were entirely dependent on either Mn2+ or Mg2+ for activation and Fe2+ and Hg2+ inhibited each of the four enzymes. The optimum pH's for the enzymes were: for fetuin: N-acetylglucosaminyl transferase, 7 , 4–8.0; for bovine submaxillary mucin: N-acetylgalactosaminyl transferase, 7 , 7; for collagen: glucosyl transferase, 7 , 7 and for orosomucoid: galactosyl transferase, 6 , 6. The enzymes were distributed subsynaptosomally primarily in the synaptosomal plasma membrane and in the mitochondria of the synaptosome. The respective values for Km (μM) and Vmex (pmoles/h/mg of protein) for the transferases were: fetuin: N-acetylglucosaminyl transferase, 12 and 143; for bovine submaxillary mucin: N-acetylgalactosaminyl transferase, 25 and 166; for collagen: glucosyl transferase, 4 and 10 and for orosomucoid:galactosyl transferase, 8 and 111.  相似文献

Though several genes governing various major traits have been reported in rice, their superior haplotype combinations for developing ideal variety remains elusive. In this study, haplotype analysis of 120 previously functionally characterized genes, influencing grain yield (87 genes) and grain quality (33 genes) revealed significant variations in the 3K rice genome (RG) panel. For selected genes, meta‐expression analysis using already available datasets along with co‐expression network provided insights at systems level. Also, we conducted candidate gene based association study for the 120 genes and identified 21 strongly associated genes governing 10‐grain yield and quality traits. We report superior haplotypes upon phenotyping the subset of 3K RG panel, SD1‐H8 with haplotype frequency (HF) of 30.13% in 3K RG panel, MOC1‐H9 (HF: 23.08%), IPA1‐H14 (HF: 6.64%), DEP3‐H2 (HF: 5.59%), DEP1‐H2 (HF: 37.53%), SP1‐H3 (HF: 5.05%), LAX1‐H5 (HF: 1.56%), LP‐H13 (3.64%), OSH1‐H4 (5.52%), PHD1‐H14 (HF: 15.21%), AGO7‐H15 (HF: 3.33%), ROC5‐H2 (31.42%), RSR1‐H8 (HF: 4.20%) and OsNAS3‐H2 (HF: 1.00%). For heading date, Ghd7‐H8 (HF: 3.08%), TOB1‐H10 (HF: 4.60%) flowered early, Ghd7‐H14 (HF: 42.60%), TRX1‐H9 (HF: 27.97%), OsVIL3‐H14 (HF: 1.72%) for medium duration flowering, while Ghd7‐H6 (HF: 1.65%), SNB‐H9 (HF: 9.35%) were late flowering. GS5‐H4 (HF: 65.84%) attributed slender, GS5‐H5 (HF: 29.00%), GW2‐H2 (HF: 4.13%) were medium slender and GS5‐H9 (HF: 2.15%) for bold grains. Furthermore, haplotype analysis explained possible genetic basis for superiority of selected mega‐varieties. Overall, this study suggests the possibility for developing next‐generation tailor‐made rice with superior haplotype combinations of target genes suiting future food and nutritional demands via haplotype‐based breeding.  相似文献

Abstract

Context: Asthma is multifaceted disease where many targets contribute towards its development and progression. Among these, adenosine receptor subtypes play a major role.

Objective: MCD-KV-10, a novel thiazolo-thiophene was designed and evaluated pre-clinically for its implication in management of asthma.

Materials and methods: This compound showed good affinity and selectivity towards A2A/A3 adenosine receptor (AR) subtypes. Furthermore, MCD-KV-10 was evaluated for in vitro lipoxygenase inhibition activity; in vivo mast cell stabilization potential and in vivo anti-asthmatic activity was done in ovalbumin-induced airway inflammation model in guinea pigs.

Results: The compound showed good (>57%) inhibition of lipoxygenase enzyme and also effectively protected mast cell degranulation (>63%). The compound showed good anti-asthmatic activity as inferred from the in vivo studies.

Discussion: These results indicate that MCD-KV-10 has an inhibitory effect on airway inflammation.

Conclusion: Though, we have identified a potential candidate for management of asthma, further mechanistic studies are needed.  相似文献

Females of the leaf miner moth Phyllonorycter acerifoliella (Z.) [=Ph. sylvella (Hw.)] and Ph. heegerella (Z.) (Lepidoptera: Gracillariidae: Lithocolletinae) release their sex pheromone at the beginning of photophase. The periodicity of the `calling' behaviour of Ph. acerifoliella females was established. Three compounds from calling virgin Ph. heegerella females were collected by the Solid Phase Micro Extraction (SPME) technique and identified as (Z)-8-tetradecenyl acetate (Z8-14:OAc), tetradecyl acetate (14:OAc) and (Z)-8-tetradecenol (Z8-14:OH) in the ratio (88±3):(2±0.6):(10±5) by capillary gas chromatography and mass spectrometry. Field trapping experiments demonstrated that the first two compounds are important for the attraction of conspecific males. Z8-14:OAc was found to be attractive when tested separately, while 14:OAc acted as synergist. The attractivity of the three component blend was reduced by 10% admixture of either (E)-10-dodecenyl acetate (E10-12:OAc) or (Z)-10-tetradecenyl acetate (Z10-14:OAc).Field tests of Z10-, Z8- and E10-14:OAc, identified from Ph. acerifoliella females, demonstrated that the first two compounds were essential for the attraction of conspecific males; so both are sex pheromone components. The attractivity of the three component blend of Z10- Z8- and E10-14:OAc was reduced by 10% admixture of (E)-10-dodecenol (E10-12:OH). The following four semiochemical compounds, Z8-14:OAc, Z8-14:OH, E10-14:OAc and 14:OAc, identified from phyllonoryctid females, as well as two sex attraction antagonists for Ph. acerifoliella and Ph. heegerella males, E10-12:OAc and Z10-14:OAc, are new for the family Gracillariidae. The results of field trapping experiments revealed mechanisms ensuring the specificity of the chemocommunication systems in Ph. acerifoliella, Ph. heegerella and Ph. ulmifoliella (Hb.) moths.  相似文献

Background: Polymorphisms in DNA repair genes have been reported contributing factors in head and neck cancer risk but studies have shown conflicting results.

Objective: To clarify the impact of DNA repair gene polymorphisms in head and neck cancer risk.

Method: A meta-analysis including 30 case–control studies was performed.

Results: Marginally statistically significant association was found for XRCC1 codon 399 (for Caucasians only), XPD Asp312Asn and XRCC1 codon 194 variants and head and neck cancer.

Conclusion: Assessments of the effects of smoking, alcohol, human papillomavirus and race/ethnicity on the association between DNA repair gene polymorphisms and head and neck cancer are needed.  相似文献

A lactate producing derivative of Escherichia coli KO11, strain SZ110, was re-engineered for ethanol production by deleting genes encoding all fermentative routes for NADH and randomly inserting a promoterless mini-Tn5 cassette (transpososome) containing the complete Zymomonas mobilis ethanol pathway (pdc, adhA, and adhB) into the chromosome. By selecting for fermentative growth in mineral salts medium containing xylose, a highly productive strain was isolated in which the ethanol cassette had been integrated behind the rrlE promoter, designated strain LY160 (KO11, Δfrd::celY Ec ΔadhE ΔldhA, ΔackA lacA::casAB Ko rrlE::(pdc Zm -adhA Zm -adhB Zm -FRT-rrlE) pflB + ). This strain fermented 9% (w/v) xylose to 4% (w/v) ethanol in 48 h in mineral salts medium, nearly equal to the performance of KO11 with Luria broth.  相似文献

The ammonia-oxidizing bacterium Nitrosomonas sp. strain ENI-11 contains three copies of the hao gene (hao 1, hao 2, and hao 3) coding for hydroxylamine oxidoreductase (HAO). Three single mutants (hao 1::kan, hao 2::kan, or hao 3::kan) had 68 to 75% of the wild-type growth rate and 58 to 89% of the wild-type HAO activity when grown under the same conditions. A double mutant (hao 1::kan and hao 3::amp) also had 68% of the wild-type growth and 37% of the wild-type HAO activity.  相似文献

The spread of invasive pests is an important topic in pest control and early detection is crucial for containment of invasive aliens. In this study the results of recent surveys on alien insects of ornamental urban plants in Portugal are reported. Overall, 13 alien species associated with allochthonous ornamental plants were recorded. Seven of these pests are new to the Portuguese fauna: Tinocallis kahawaluokalani (Kirkaldy); Illinoia liriodendri (Monell); Appendiseta robiniae (Gillette) (Hemiptera: Aphididae); Acizzia jamatonica (Kuwayama) (Hemiptera: Psyllidae); Dasineura gleditchiae (Osten Sacken); Obolodiplosis robiniae (Haldeman) (Diptera: Cecidomyiidae); and Phytoliriomyza jacarandae Steyskal & Spencer (Diptera: Agromyzidae). Additional distribution data are also provided for other recently introduced alien pests in Portugal: Myzocallis (Lineomyzocallis) walshii (Monell) (Hemiptera: Aphididae); Platycorypha nigrivirga Burckhardt; Glycaspis brimblecombei Moore (Hemiptera: Psyllidae); Calophya schini Tuthill (Hemiptera: Calophyidae); Corythucha ciliata (Say) (Hemiptera: Tingidae); and Lantanophaga pusillidactylus (Walker) (Lepidoptera: Pterophoridae). Three species in particular (Illinoia liriodendri, Obolodiplosis robiniae and Phytoliriomyza jacarandae) were not previously found in the Iberian Peninsula. Details on current distribution, host plants and biological remarks are given for each species. Possible origin areas, pathways and impacts are also discussed.  相似文献

Summary The products of 11 switching (swi) genes are required for efficient mating-type (MT) switching in homothallic (h 90) strains of Schizosaccharomyces pombe. The MT region of h 90 comprises three cassette genes: the expression site mat1: 1 and two silent loci, mat2: 2 and mat3: 3. Besides reducing MT switching, the swi6 mutation leads to deletions in the MT region caused by intrachromosomal cross-overs between two paired cassettes. These deletions only arise if DNA double-strand breaks are present at mat1: 1, which initiate MT switching. Furthermore, swi6 allows meiotic recombination in the K region, a region of 16 kb between mat2: 2 and mat3: 3; in wild-type strains no recombination occurs in K. swi6 also allows the simultaneous expression of two different cassettes in the same haploid cell. Thus swi6 may have an influence on the general chromatin structure in the MT region.  相似文献

We generated a transgenic line Tg(k18:shh:RFP) with overexpression of Sonic hedgehog in the skin epidermis. By 5 day-post-fertilization (dpf), many epidermal lesions were clearly observed, including a swollen yolk sac, epidermis growth malformation around the eyes and at the basement of the pectoral fins. Skin histology revealed embryos derived from Tg(k18:shh:RFP) displayed an elevated Nuclear/Cytoplasmic ratio and pleomorphic nuclei compared to their wild type littermates, suggesting the abnormal growth pattern on the epidermis of Tg(k18:shh:RFP) embryos were dysplasia. Later (by 7 dpf), Tg(k18:shh:RFP) embryos displayed broader pectoral fins which are similar to the polydactyly phenotypes of Nevoid basal cell carcinoma syndrome (NBCCS)/Gorlin patients and polydactylous mice. In addition, treatment with cyclopamine is able to enhance and prolong the survival rates and survival durations of Tg(k18:shh:RFP) embryos. In conclusion, this unique Tg(k18:shh:RFP) fish line, should be an excellent experimental animal for screening for a lower toxicity level of the new Hh-inhibitor and can even be used as a new anti-cancer drug-screening platform.  相似文献

Objective: To study the impact of genetic and lifestyle factors on protein biomarkers and develop personally normalized plasma protein profiles (PNPPP) controlling for non-disease-related variance.

Materials and methods: Proximity extension assays were used to measure 145 proteins in 632 controls and 344 cases with non-communicable diseases.

Results: Genetic and lifestyle factors explained 20–88% of the variation in healthy controls. Adjusting for these factors reduced the number of candidate biomarkers by 63%.

Conclusion: PNPPP efficiently controls for non-disease-related variance, allowing both for efficient discovery of novel biomarkers and for covariate-independent linear cut-offs suitable for clinical use.  相似文献
