To determine the onset of the Cd2+-hyperaccumulating phenotype in Euglena gracilis, induced by Hg2+ pretreatment (Avilés et al. in Arch Microbiol 180:1–10, 2003), the changes in cellular growth, Cd2+ uptake, and intracellular contents of sulfide, cysteine, γ-glutamylcysteine, glutathione and phytochelatins during the progress of the culture were analyzed. In cells exposed to 0.2 mM CdCl2, the Cd2+-hyperaccumulating phenotype was apparent only after 48 h of culture, as indicated by the significant increase in cell growth and higher internal contents of sulfide and thiol-compounds, along with a higher γ-glutamylcysteine synthetase activity. However, the stiochiometry of thiol-compounds/Cd2+ accumulated was similar for both control and Hg2+-pretreated cells. Moreover, the value for this ratio was 2.1 or lower after 48-h culture, which does not suffice to fully inactivate Cd2+. It is concluded that, although the glutathione and phytochelatin synthesis pathway is involved in the development of the Cd2+-hyperaccumulating phenotype in E. gracilis, apparently other pathways and sub-cellular mechanisms are also involved. These may be an increase in other Cd2+ chelating molecules such as di- and tricarboxylic acids, phosphate and polyphosphates, as well as Cd2+ compartmentation into organelles. The glutathione (GSH) status and heavy metal tolerance were investigated in four Paxillus involutus strains isolated from different heavy-metal-polluted and non-polluted regions of Europe. The heavy metal burden in the habitats did not affect significantly either the heavy metal (Cr2O72−, Cd2+, Hg2+, Pb2+, Zn2+, Cu2+) tolerance and accumulation or the GSH production of the strains tested. Exposures to heavy metals increased the intracellular GSH concentrations in 12 from 24 experimental arrangements (four strains exposed to six heavy metals) independently of the habitats of the strains. The importance of GSH in heavy metal tolerance (high MIC values, ability to accumulate heavy metals and to grow in the presence of heavy metals) was thus demonstrated in this ectomycorrhizal fungus.  相似文献

The heavy metal resistant ciliate, Stylonychia mytilus, isolated from industrial wastewater has been shown to be potential bioremediator of contaminated wastewater. The ciliate showed tolerance against Zn2+ (30 μg/mL), Hg2+ (16 μg/mL) and Ni2+ (16 μg/mL). The metal ions slowed down the growth of the ciliate as compared with the culture grown without metal stress. The reduction in cell population was 46% for Cd2+, 38% for Hg2+, 23% for Zn2+, 39% for Cu2+ and 51% for Ni2+ after 8 days of metal stress. S. mytilus reduced 91% of Cd2+, 90% of Hg2+ and 98% of Zn2+ from the medium after 96 h of incubation in a culture medium containing 10 μg/mL of the respective metal ions. Besides this, the ciliate could also remove 88% of Cu2+ and 73% Ni2+ from the medium containing 5 μg/mL of each metal after 96 h. The ability of Stylonychia to take up variety of heavy metals from the medium could be exploited for metal detoxification and environmental clean-up operations.  相似文献

To identify some of the mechanisms involved in the high resistance to Cd2+ in the protist Euglena gracilis, we studied the effect of Cd2+ exposure on its energy and oxidative stress metabolism as well as on essential heavy metals homeostasis. In E. gracilis heterotrophic cells, as in other organisms, CdCl2 (50 μM) induced diminution in cell growth, severe oxidative stress accompanied by increased antioxidant enzyme activity and strong perturbation of the heavy metal homeostasis. However, Cd2+ exposure did not substantially modify the cellular respiratory rate or ATP intracellular level, although the activities of respiratory complexes III and IV were strongly decreased. In contrast, an enhanced capacity of the alternative oxidase (AOX) in both intact cells and isolated mitochondria was determined under Cd2+ stress; in fact, AOX activity accounted for 69-91% of total respiration. Western blotting also revealed an increased AOX content in mitochondria from Cd2+-exposed cells. Moreover, AOX was more resistant to Cd2+ inhibition than cytochrome c oxidase in mitochondria from control and Cd2+-exposed cells. Therefore, an enhanced AOX seems to be a relevant component of the resistance mechanism developed by E. gracilis against Cd2+-stress, in addition to the usual increased antioxidant enzyme activity, that enabled cells to maintain a relatively unaltered the energy status.  相似文献

Seed is a developmental stage that is highly protective against external stresses in the plant life cycle. In this study, we analyzed toxicity of essential (Cu2+ and Zn2+) and non-essential heavy metals (Hg2+, Pb2+ and Cd2+) on seed germination and seedling growth in the model species Arabidopsis. Our results show that seedling growth is more sensitive to heavy metals (Hg2+, Pb2+, Cu2+ and Zn2+) in comparison to seed germination, while Cd2+ is the exception that inhibited both of these processes at similar concentrations. To examine if toxicity of heavy metals is altered developmentally during germination, we incubated seeds with Hg2+ or Cd2+ only for a restricted period during germination. Hg2+ displayed relatively strong toxicity at period II (12–24 h after imbibition), while Cd2+ was more effective to inhibit germination at period I (0–12 h after imbibition) rather than at period II. The observed differences are likely to be due in part to selective uptake of different ions by the intact seed, because isolated embryos (without seed coat and endosperm) are more sensitive to both Hg2+ and Cd2+ at period I. We assessed interactive toxicity between heavy metals and non-toxic cations, and found that Ca2+ was able to partially restore the inhibition of seedling growth by Pb2+ and Zn2+.  相似文献

Biochemical responses to cadmium (Cd2+) and copper (Cu2+) exposure were compared in two strains of the aquatic hyphomycete (AQH) Heliscus lugdunensis. One strain (H4-2-4) had been isolated from a heavy metal polluted site, the other (H8-2-1) from a moderately polluted habitat. Conidia of the two strains differed in shape and size. Intracellular accumulation of Cd2+ and Cu2+ was lower in H4-2-4 than in H8-2-1. Both␣strains synthesized significantly more glutathione (GSH), cysteine (Cys) and γ-glutamylcysteine (γ-EC) in the presence of 25 and 50 μM Cd2+, but quantities and rates of synthesis were different. In H4-2-4, exposure to 50 μM Cd2+ increased GSH levels to 262% of the control; in H8-2-1 it increased to 156%. Mycelia of the two strains were analysed for peroxidase, dehydroascorbate reductase, glutathione reductase and glucose-6-phosphate dehydrogenase. With Cd2+ exposure, peroxidase activity increased in both strains. Cu2+ stress increased dehydroascorbate reductase activity in H4-2-4 but not in H8-2-1. Dehydroascorbate reductase and glucose-6-phosphate dehydrogenase activities progressively declined in the presence of Cd2+, indicating a correlation with Cd2+ accumulation in both strains. Cd2+ and Cu2+ exposure decreased glutathione reductase activity.  相似文献

Heavy metal pollution has become one of the most serious environmental problems today. To develop a more efficient plant to clean up heavy metal contaminated soils, a γ‐glutamylcysteine synthetase (GCS) cDNA, named PaGCS, was isolated by PCR from Phragmites australis. The PaGCS sequence was transformed via agroinfection into the heavy metal intolerant grass Agrostis palustris. Five confirmed transgenic A. palustris plants expressing PaGCS were compared with the wild‐type line for growth and Cd2+ accumulation, as well as for the expression of a number of phytochelatin synthesis and stress‐responsive enzymes when challenged with Cd2+ stress. GCS and phytochelatin synthase (PCS) were up‐regulated in the transgenic lines. All the transgenic lines accumulated more Cd2+ and phytochelatins (PCs) than the wild‐type line, and three of the five lines grew more effectively than the wild‐type after either five or 21 d of Cd2+ stress. Variation among the transgenics was observed for the distribution of Cd2+ in the root, shoot and leaf. The malondialdehyde content of all the transgenic lines was lower than that of the wild type under Cd2+ treatment, while the activity of both superoxide dismutase and peroxidase present in the transgenic lines increased markedly 24 h after Cd2+ stress, and then rapidly declined.  相似文献

We have studied the effects of heavy metals (Hg2+, Cu2+, Cd2+) on growth hormone (GH) activation of tyrosine kinase and Ca2+ signaling in the trout (Oncorhynchus mykiss) hepatoma cell line RTH-149. Molecular cloning techniques using primer designed on Oncorhynchus spp. growth hormone receptor (GHR) genes allowed to isolate a highly homologous cDNA fragment from RTH-149 mRNA. Thereafter, cells were analysed by Western blotting or, alternatively, with Ca2+ imaging using fura-2/AM. Exposure of cells to ovine GH alone produced a stimulation of the JAK2/STAT5 pathway and intracellular free Ca2+ variations similar to what has been observed in mammalian models. Cell pre-exposure to Cu2+, Hg2+ or Cd2+ affected cell response to GH by enhancing (Cu2+) or inhibiting (Cd2+) the phosphorylation of JAK2 and STAT5. Heavy metals induced the activation of the MAP kinase p38, and pre-exposure to Hg2+ or Cu2+ followed by GH enhanced the effect of metal alone. Image analysis of fura2-loaded cells indicated that pre-treatment with Hg2+ prior to GH produced a considerable increase of the [Ca2+]i variation produced by either element, while using Cu2+ or Cd2+ the result was similar but much weaker. Data suggest that heavy metals interfere with GH as follows: Hg2+ is nearly ineffective on JAK/STAT and strongly synergistic on Ca2+ signaling; Cu2+ is activatory on JAK/STAT and slightly activatory on Ca2+; Cd2+ is strongly inhibitory on JAK/STAT and slightly activatory on Ca2+; heavy metals could partially activate STAT via p38 independently from GH interaction.Published online: March 2005  相似文献

The effect of calcium (Ca2+) on Trifolium repens L. seedlings subjected to cadmium (Cd2+) stress was studied by investigating plant growth and changes in activity of antioxidative enzymes. Physiological analysis was carried out on seedlings cultured for 2 weeks on half-strength Hoagland medium with Cd2+ concentrations of 0, 400 and 600 μM, and on corresponding medium supplied with CaCl2 (5 mM). Exposure to increasing Cd2+ reduced the fresh weight of the upper part (stems + leaves) of the seedlings more strongly than that of the root system. In both parts of T. repens seedlings H2O2 level and lipid peroxidation increased. In the upper part, Cd2+ exposure led to a significant decrease in the activity of superoxide dismutase, catalase and glutathione peroxidase and an increase in ascorbate peroxidase activity. In contrast, the roots showed an increase in the activity of antioxidative enzymes under Cd2+ stress. Ca2+ addition to medium reduced the Cd2+ accumulation, and considerably reversed the Cd2+-induced decrease in fresh mass as well as the changes in lipid peroxidation in the both parts of T. repens seedlings. Ca2+ application diminished the Cd2+ effect on the activity of antioxidative enzymes in the upper part, even though it did not significantly affect these enzymes in the roots. So the possible mechanisms for the action of Ca2+ in Cd2+ stress were considered to reduce Cd2+ accumulation, alleviate lipid peroxidation and promote activity of antioxidative enzymes.  相似文献

Intracellular cadmium (Cd2+) ion accumulation and the ability to produce specific Cd2+ ion chelators was studied in the methylotrophic yeast Hansenula polymorpha. Only one type of Cd2+ intracellular chelators, glutathione (GSH), was identified, which suggests that sequestration of this heavy metal in H. polymorpha occurs similarly to that found in Saccharomyces cerevisiae, but different to Schizosaccharomys pombe and Candida glabrata which both synthesize phytochelatins. Cd2+ ion uptake in the H. polymorpha wild-type strains appeared to be an energy dependent process. It was found that Δgsh2 mutants, impaired in the first step of GSH biosynthesis, are characterized by increase in net Cd2+ ion uptake by the cells, whereas Δgsh1met1 and Δggt1 mutants impaired in sulfate assimilation and GSH catabolism, respectively, lost the ability to accumulate Cd2+ intracellularly. Apparently H. polymorpha, similarly to S. cerevisiae, forms a Cd-GSH complex in the cytoplasm, which in turn regulates Cd2+ uptake. Genes GSH1/MET1 and GGT1 are involved in maturation and metabolism of cellular Cd-GSH complex, respectively. Transport of [3H]N-ethylmaleimide-S-glutathione ([3H]NEM-SG) conjugate into crude membrane vesicules, purified from the wild-type cells of H. polymorpha appeared to be MgATP dependent, uncoupler insensitive and vanadate sensitive. We suggest that MgATP dependent transporter involved in Cd-GSH uptake in H. polymorpha, is similar to S. cerevisiae Ycf1-mediated vacuolar transporter responsible for accumulation of organic GS-conjugates and Cd-GSH complex.  相似文献

The effect of different concentrations of Hg2+, Cd2+, and Pb2+ on ultrastructure, growth, respiration, photosynthesis, chlorophyll content, and metal accumulation in Euglena gracilis was examined. The toxicity of the heavy metals was dependent on the culture medium used and whether cells were grown in the dark or under illumination. Hg2+ was the most toxic metal, which showed effects at a concentration as low as 1.5 μM; Cd2+ showed an intermediate toxicity (effects observed above 50 μM); and Pb2+ was almost ineffective up to 1 mM. Cells grown for several weeks in the dark, in the presence of 1.5 μM Hg2+ showed a reduced sensitivity to subsequent exposure to Cd2+ or Pb2+. The Hg2+-pretreated cells also presented an enhanced capacity to accumulate other metals. In comparison, light-grown cells showed a greater Cd2+ accumulation, but a lower Pb2+ uptake than Hg2+-pretreated dark-grown cells. Pretreatment of light-grown cells with Hg2+ did not enhance the accumulation of Cd2+. These results suggest that the capacity to tolerate heavy metals by Euglena may have mechanistic differences when cells are grown in the dark or under illumination.  相似文献

Phytochelatins (PCs) are metal binding peptides involved in heavy metal detoxification. To assess whether enhanced phytochelatin synthesis would increase heavy metal tolerance and accumulation in plants, we overexpressed the Arabidopsis phytochelatin synthase gene (AtPCS1) in the non-accumulator plant Nicotiana tabacum. Wild-type plants and plants harbouring the Agrobacterium rhizogenes rolB oncogene were transformed with a 35S AtPCS1 construct. Root cultures from rolB plants could be easily established and we demonstrated here that they represent a reliable system to study heavy metal tolerance. Cd2+ tolerance in cultured rolB roots was increased as a result of overexpression of AtPCS1, and further enhanced when reduced glutathione (GSH, the substrate of PCS1) was added to the culture medium. Accordingly, HPLC analysis showed that total PC production in PCS1-overexpressing rolB roots was higher than in rolB roots in the presence of GSH. Overexpression of AtPCS1 in whole seedlings led to a twofold increase in Cd2+ accumulation in the roots and shoots of both rolB and wild-type seedlings. Similarly, a significant increase in Cd2+ accumulation linked to a higher production of PCs in both roots and shoots was observed in adult plants. However, the percentage of Cd2+ translocated to the shoots of seedlings and adult overexpressing plants was unaffected. We conclude that the increase in Cd2+ tolerance and accumulation of PCS1 overexpressing plants is directly related to the availability of GSH, while overexpression of phytochelatin synthase does not enhance long distance root-to-shoot Cd2+ transport.  相似文献

The effect of equimolar concentrations of Hg2+ and Cd2+ on the whole cell absorption spectra, absorption spectra of the extracted phycocyanin (PC) and fluorescence emission spectra of phycobilisomes (PBS) was investigated in the cells of Anabaena flos-aquae. The PC component of the PBS was found to be extremely sensitive to the Hg2+ rather than the Cd2+ ions. Further, the results showed that Hg2+ and Cd2+ induced decrease in the rate of Hill activity (H2O - DCPIP) was partially restored by the electron donor NH2OH, not by the diphenyl carbazide. Similarly, chlorophyll a fluorescence emission in the presence of metals showed that addition of NH2OH could effectively reverse the metal induced alterations in the fluorescence emission intensity. These results, together, suggested that Hg2+ and Cd2+ caused damage to the photosystems (PS) II reaction center. However, a relatively higher stimulation of the chlorophyll a emission at 695 nm with a red shift of 4.0 nm in the presence of Hg2+, and Cd2+ induced preferential decrease in the emission intensity at 676 nm as compared with the peak at 695 nm were indicative of the differential action of Hg2+ and Cd2+ on the PS II.  相似文献

Phytochelatins ((γGluCys)nGly) are synthesized from glutathione by plants exposed to metals like Cd2+, Cu2+ and Zn2+. An intracellular complex formed by phytochelatins with Cd2+ and sulfide is thought to detoxify the metal possibly by sequestration in the vacuole. It was found that maize seedlings exposed to Cd2+ produced phytochelatins and two additional families of cysteina-containing peptides, (γGluCys)n and (γGluCys)nGlu. All thiol peptides with n = 2 and 3 were purified and their structure characterized by tandem mass spectrometry. For maize plants exposed to Cd2+ for 7 days, phytochelatins were synthesized preferentially in the first 24 h whereas the amounts of (γGluCys)n and (γGluCys)nGlu were the highest thereafter. This was probably due to an initial large pool of glutathione available in control plants compared with a dearth of γGluCys and no detectable γGluCysGlu. The (γGluCys)nGlu peptides were induced exclusively by Cd2+ as they were below the detection limit in control seedlings that contained low amounts of phytochelatins and (γGluCys)n. Since the Cys moiety of the peptides is essential for binding Cd2+, a role for accumulated (γGluCys)n and (γGluCys)nGlu in detoxifying Cd2+ in plants must be considered.  相似文献

Several Cd2+ hypersensitive and Hg2+ sensitive mutants of Escherichia coli K-12 were isolated after repeated mutagenesis with nitrosoguanidine. The Cd-hypersensitive mutant, CD17P, could not grow in a chemically defined liquid medium containing 0.5 μm Cd2+, and the growth of Hg-sensitive mutant, HG17P-52, was inhibited by 0.9 μm Hg2+. Thus, CD 17P and HG17P-52 were respectively about 1000 fold and 4 fold more sensitive than the parental strain.

Both mutants were also considerably more sensitive to Zn2+ than their parent, but they did not show any appreciable change in sensitivity to other metals tested. Among the various metabolites and chemicals tested, reducing agents such as cysteine, dithiothreitol and vitamin C showed a protective effect against metal toxicity. Reduced glutathione was effective only against Hg2+. EDTA specifically and efficiently reversed only Cd2+ toxicity.  相似文献

Industry residues, phosphate fertilisers and wastewater as a source of irrigation have considerably increased levels of heavy metals in the soil, mainly cadmium (Cd2+). To test the effects of a calcium (Ca2+) treatment on Cd2+ accumulation and plant tolerance to this heavy metal, plants of two citrus genotypes, Cleopatra mandarin (CM) and Carrizo citrange (CC), were watered with increasing concentrations of Cd2+, and phytochelatin (PC) and glutathione (GSH) content were measured. Both genotypes were able to synthesise PCs in response to heavy metal intoxication, although CM seems to be a better Cd2+ excluder than CC. However, data indicate that CC plants had a higher capacity for regenerating GSH than CM plants. In this context, the effects of Ca2+ treatment on Cd2+ accumulation, plant survival and PC, GSH and oxidised glutathione (GSSG) content were assessed. Data indicate that treatment with Ca2+ had two positive effects on citrus physiology: it reduced Cd+2 uptake into roots and also increased GSH content (even in the absence of Cd2+). Overall, the data indicate that although Cd2+ exclusion is a powerful mechanism to avoid heavy metal build‐up into photosynthetic organs, the capacity to maintain optimum GSH levels to feed PC biosynthesis could also be an important factor in stress tolerance.  相似文献

Plants suffer from combined stress of sulfur deficiency and cadmium toxicity in some agricultural lands. However, little is known about the reaction in plants, such as responses in antioxidant enzymes and non-protein thiol compounds, to such combined stress. Therefore, in this study, four treatments, S-sufficiency (TS?Cd), S-deficiency (T?S?Cd), Cd stress (TS+Cd) and combined stress of S-deficiency and Cd stress (T?S+Cd), were set up to investigate (1) the effects of sulfur deficiency or sulfur sufficiency on Cd toxicity to kidney bean cultivar seedlings and the related mechanisms, and (2) the responses of two kidney bean cultivars to combined stress of S-deficiency and Cd-tolerance. The results showed significant increases in hydrogen peroxide (H2O2) and malondialdehyde contents and significant increases in antioxidant enzyme (superoxide dismutase, catalase, peroxidase, and glutathione S-transferase) activities and non-protein thiol compounds (non-protein thiols, reduced glutathione, phytochelatins) synthesis in the plants in TS+Cd and T?S+Cd. On the tissue level, higher proportion of Cd was found to be immobilized/deposited in roots, while on the sub-cell level, higher proportion of Cd was located in cell walls and vacuole fractions with lower in cell organelles. Taken together, the results indicated that Cd detoxification was achieved by the two kidney bean cultivars through antioxidant enzyme activation, non-protein thiol compound synthesis and sub-cellular compartmentalization. In addition, the results indicated that sufficient S supply helped to relieve Cd toxicity, which is of special significance for remediation or utilization of Cd-contaminated soils as S is a plant essential nutrient.  相似文献

Metallothioneins MTT1 and MTT2 from Tetrahymena thermophila have been characterized. The MTT1 contains mainly characteristic Cys-Cys-Cys and Cys-Cys clusters, but MTT2 contains mainly Cys-X-Cys cluster. Cd16-MTT1 mainly consists of α-helix and β-turns, in contrast, Cd11-MTT2 mainly consists of random coils. Reaction of Cd16-MTT1 and Cd11-MTT2 with nitric oxide leads to intramolecular disulfide bond formation, respectively. Binding stabilities of Cd2+, Hg2+ and Zn2+ to MTT1 are stronger than those to MTT2. Cu2+ can not replace Cd2+ from Cd16-MTT1 complex, but can replace Cd2+ from Cd11-MTT2 complex. The analysis of qRT-PCR revealed MTT2 mRNA levels were 31-fold higher than those of MTT1 under basal conditions. These results further suggest MTT1 possibly play a role in the detoxification of heavy metal ions, and MTT2 may be involved in the homeostasis of copper ions.  相似文献
