While stigma anatomy is well documented for a good number of species, little information is available on the acquisition and cessation of stigmatic receptivity. The aim of this work is to characterize the development of stigma receptivity, from anthesis to stigma degeneration, in the pentacarpellar pear (Pyrus communis) flower. Stigma development and stigmatic receptivity were monitored over two consecutive years, as the capacity of the stigmas to offer support for pollen germination and pollen tube growth. In an experiment where hand pollinations were delayed for specified times after anthesis, three different stigmatic developmental stages could be observed: (1) immature stigmas, which allow pollen adhesion but not hydration; (2) receptive stigmas, which allow proper pollen hydration and germination; and (3) degenerated stigmas, in which pollen hydrates and germinates properly, but pollen tube growth is impaired soon after germination. This developmental characterization showed that stigmas in different developmental stages coexist within a flower and that the acquisition and cessation of stigmatic receptivity by each carpel occur in a sequential manner. In this way, while the duration of stigmatic receptivity for each carpel is rather short, the flower has an expanded receptive period. This asynchronous period of receptivity for the different stigmas of a single flower is discussed as a strategy that could serve to maximize pollination resources under unreliable pollination conditions.  相似文献

Silene vulgaris occurs in Israel as a stolonifcrous tetraploid which forms mostly sexually polymorphic populations. Fully hermaphrodite and fully male-sterile plants are linked by a range of intermediate forms which are gynomonoecious and bear hermaphrodite, male-sterile, and partially male-sterile flowers. Male-sterile flowers differ from hermaphrodite ones by having a narrower calyx, a shorter corolla, a thicker style, and a considerably larger stigmatic area with longer papillae which are more markedly bulbous basally. In individual anthers of hermaphrodite flowers, diameters of stained pollen grains showed a continuous variation from 36 to 80 μm with a modal value of 56. About 30% of pollen was found to be non-stainable. Reduction in male fertility occurs at different levels, from small or inviable pollen grains through abortion of single anthers, abortion of entire androecia, and through different degrees of gynomonoecy to totally male-sterile individuals. The proportion of stainable pollen to ovules in hermaphrodite flowers alone was found to be less than 100. From the data presented, less than a third of all ovules produced in the different kinds of flower develop into seeds. This may reflect an inadequacy of pollen, which the local population of S. vulgaris can tolerate because of its stoloniferous habit.  相似文献

对青阳参花(Cynanchum otophyllum)部综合特征、访花昆虫种类、访花行为及传粉过程进行了研究,结果表明,青阳参花结构复杂,两个子房基部离生、花柱联合与雄蕊形成合蕊柱,柱头表面被邻近花药的侧翼紧密包围形成5个柱头腔。青阳参的花粉形成独特的花粉块,一次传粉过程可以转运大量的花粉。东方蜜蜂(Apis cerana)是青阳参的主要传粉昆虫,其传粉包括两个过程:(1)当蜜蜂的口器或足插入着粉腺的槽口后借助蜜蜂的力量将花粉块从花上拔起;(2)当蜜蜂再次访花时将携带的花粉块插入其中一个柱头腔。花粉块里面的花粉粒住柱头腔中萌发出花粉管,然后沿着花柱道向下生长最后进入子房。在整个花期仡粉保持有相对较高的生活力,而其柱头可授性则在7天后逐渐降低。  相似文献

Most species of Dalechampia have expanded stigmatic surfaces that extend from the stylar tip part way down the sides of the elongated styles. Pollen grains landing at the stylar tip grow tubes directly down the style to the ovary. Pollen grains landing on the lateral stigmatic surfaces, however, exhibit indirect pollen-tube growth: the tubes grow first to the stylar tip, bend 180°, and then grow to the ovary. Indirect pollen-tube growth appears to be reproductively important: more than half the arriving pollen lands on the lateral stigmatic surfaces, and virtually all of these grains produce tubes that grow indirectly to the ovary; pollen on the lateral surface fertilizes seeds about as effectively as pollen at the tip (with direct tube growth). Indirect pollen-tube growth may be the result of a correlated response to selection for expanded stigmatic surfaces in Dalechampia, or it may be an adaptation to increase the intensity of pollen competition.  相似文献

In Mutisieae species, the style branches are described as short and the stigmatic areas cover the inside of the style branches. As shown in preliminary observations, the Mutisia speciosa florets had long style branches (7 mm), bifurcate only at the apex (about 1.5 mm) and juxtaposed along the remaining length. The objective was to locate the stigmatic areas in M. speciosa. For this purpose, neutral red, 3% hydrogen peroxide and hand pollinations were used. For the pollinations, florets with cut apical portion of the branches (about 2 mm) and florets with intact branches were used. In the first group of florets, the pollen was deposited along the margins of the juxtaposed branches (about 5 mm), and in the second, the pollen was ventrally deposited on the bifurcated apical portion. Some of these florets were left on the plant until fruiting and others were analyzed under a fluorescence microscope to check pollen tube growth. The neutral red test defined two ventro-marginal bands fused at the apex of each style branch, consisting of papillose cells. Intense bubbling in the hydrogen peroxide test showed that only the bands are receptive. The pollinations resulted in fruit sets and growth of pollen tubes, confirming that the bands are receptive along their entire length. This result is new and indicates the need for further studies on the floral biology of tropical Asteraceae species to improve the understanding of their reproductive attributes.  相似文献

Angiosperm stigmas exhibit high levels of peroxidase activity when receptive to pollen. To explore possible function(s) of this peroxidase activity we investigated amounts of reactive oxygen species (ROS), particularly hydrogen peroxide, in stigmas and pollen. Because nitric oxide (NO) was recently implicated in pollen tube growth, we also investigated amounts of NO in pollen and stigmas. Reactive oxygen species accumulation was assessed with confocal microscopy and light microscopy using ROS probes DCFH2-DA and TMB, respectively. NO was assayed using the NO probe DAF-2DA and confocal microscopy. Stigmas from various different angiosperms were found to accumulate ROS, predominantly H2O2, constitutively. In Senecio squalidus and Arabidopsis thaliana high amounts of ROS/H2O2 were localized to stigmatic papillae. ROS/H2O2 amounts appeared reduced in stigmatic papillae to which pollen grains had adhered. S. squalidus and A. thaliana pollen produced relatively high amounts of NO compared with stigmas; treating stigmas with NO resulted in reduced amounts of stigmatic ROS/H2O2. Constitutive accumulation of ROS/H2O2 appears to be a feature of angiosperm stigmas. This novel finding is discussed in terms of a possible role for stigmatic ROS/H2O2 and pollen-derived NO in pollen-stigma interactions and defence.  相似文献

Plant reproduction is highly vulnerable to environmental conditions such as temperature and, consequently, planet warming may have significant consequences on the reproductive phase with serious implication in agricultural crops. Although pollen tube growth is clearly affected by temperature, little information is available on its effect on the female side and on flower receptivity. In this work, the effect of temperature has been evaluated on stigmatic receptivity of sweet cherry in vivo, in the laboratory, and in planta, in the field. Results herein show that temperature has a clear effect on the duration of stigmatic receptivity. Thus, whereas high temperature reduced stigmatic receptivity, low temperature enlarged it. The stigma lost the capacity to offer support first for pollen penetration, second for pollen germination and, finally, for pollen adhesion. The effect of temperature was more pronounced on pollen germination and penetration than on pollen adhesion. High temperature reduced the germination capacity of the pollen as early as the first day after anthesis, a time when no apparent signs of stigma degeneration are apparent. This clear effect of temperature on stigmatic receptivity and pollen performance may have clear implication in crop performance and in establishing screening criteria of best‐adapted genotypes.  相似文献

The two main factors involved in field-scale production of seed of F1 hybrid Brussels sprouts are pollen availability and honeybee behaviour. Pollen availability depends upon the extent of winter survival of the parent inbreds, their flowering times, plant size and on the number of flowers per plant. Plant losses varied between inbreds and between sites and seasons. Differences in the commencement of flowering time in pairs of inbreds varied from 7 to 21 days, and plant size affected flower number. In hybrid seed-production there was a direct relationship between the number of mature flowers on each inbred and the percentage of non-hybrid seed produced from that inbred. Bees were highly selective in their visits to inbreds, and a mean selfing-to crossing-movement ratio of 30:1 was observed. This behaviour, together with pollen availability, greatly influenced the production of ‘sibs’. Radioactive experiments showed that the amount of cross-pollen carried by a bee decreased by about 30% at each flower visited but radioactive pollen grains were detected on the tenth flower visited. Of a number of factors investigated as possibly influencing bee behaviour, differences in flower colour and plant height were associated with discrimination between inbred lines.  相似文献

BACKGROUND AND AIMS: Fertilization is essential in almond production, and pollination can be limiting in production areas. This study investigated stigma receptivity under defined developmental stages to clarify the relationship between stigma morphology, pollen germination, tube growth and fruit set. METHODS: Light and scanning electron microscopy were employed to examine stigma development at seven stages of flower development ranging from buds that were swollen to flowers in which petals were abscising. Flowers at different stages were hand pollinated and pollen germination and tube growth assessed. Artificial pollinations in the field were conducted to determine the effect of flower age on fruit set. KEY RESULTS: Later stages of flower development exhibited greater stigma receptivity, i.e. higher percentages of pollen germination and more extensive tube growth occurred in older (those opened to the flat petal stage or exhibiting petal fall) than younger flowers. Enhanced stigma receptivity was associated with elongation of stigmatic papillae and increased amounts of stigmatic exudate that inundated papillae at later developmental stages. Field pollinations indicated that the stigma was still receptive and nut set was maintained in older flowers. CONCLUSIONS: Stigma receptivity in almond does not become optimal until flowers are past the fully open stage. The stigma is still receptive and fruit set is maintained in flowers even at the stage when petals are abscising. Strategies to enhance pollination and crop yield, including the timing and placement of honey bees, should consider the effectiveness of developmentally advanced flowers.  相似文献

Anthesis in Nymphaea capensis var. zanzibariensis is diurnal with flowers opening and closing for three consecutive days. On the first day of anthesis, the stigmatic papillae secrete fluid and the outermost anthers are dehiscent. On the second day of anthesis the stamens form a cone above the dry stigmatic cup. The middle stamens open and turn outward. On the third day of flowering, all the stamens open and the dry stigmatic cup is exposed. The flowers are homogamous and not protogynous as the other Nymphaea. The gynoecium of the self-compatible N. capensis var. zanzibariensis , is characterized by a wet papillate stigma, a short hollow style, and secretory cells on the ventral surface of the ovary. The pollen is released on the receptive stigma. Following initial growth in intercellular spaces in the transmitting tract of the stigma, pollen tubes travel through the stylar canal and into the ovary.  相似文献

Low seed ovule ratios have been observed in natural populations of Polygala vayredae Costa, a narrowly endemic species from the oriental pre-Pyrenees. To evaluate physical and nutritional constraints and pollen tube attrition in this endemic species, stigma and style anatomy, as well as pollen tube development along the pistil were investigated using light and fluorescence microscopy. The structural morphology of the stigmatic region was also examined with scanning electron microscopy. Pollen grains that reached the stigmatic papillae came into contact with a lipid-rich exudate and germinated easily. Although a large number of pollen grains reach the stigmatic papillae, few pollen tubes were able to grow into the style towards the ovary. The style was hollow, with the stylar channel beginning a few cells below the stigmatic papillae. Initially, the stylar channel area was small compared to other levels of the style, and was surrounded by lipid-rich, highly metabolic active cells. Furthermore, lipid-rich mucilage was detected inside the stylar channel. At subsequent style levels towards the ovary, no major reserves were detected histochemically. The reduced intercellular spaces below the stigmatic papillae and the reduced area of the stylar channel at its commencement are suggested to physically constrain the number of pollen tubes that can develop. In subsequent levels of the style, the stylar channel could physically support a larger number of pollen tubes, but the lack of nutritional reserves cannot be disregarded as a cause of pollen tube attrition. Finally, the number of pollen tubes entering the ovary was greater than the number of ovules, suggesting that interactions occurring at this level play a major role in the final reproductive outcome in this species.  相似文献

 Cork oak (Quercus suber L.) is a monoecious wind-pollinated species with a protandrous system to ensure cross-pollination. To the best of our knowledge, this report provides the first insight into the sexual reproduction cycle in this species. The cork oak flowering season extends from April until the end of May. Our results show that, at anthesis, the pistillate flower is not completely formed and ovules are just starting to develop. Pollen reaching the dry stigmatic surface adheres to the receptive cells, germinates and penetrates the epidermis in aproximately 24 h, and grows through the intercellular spaces of a solid transmitting tissue. In cross-pollination, a sequential arrest of pollen tubes was observed along the style, providing preliminary evidence for a pollen tube competition mechanism. As a consequence, few pollen tubes reach the basal portion of the style. Furthermore, pollen tube growth is a discontinuous process since tubes are arrested in the basal portion of the style about 10–12 days after pollination. While tubes are latent, the ovarian loculus starts to develop from an emerging mass of sporogeneous cells which later will differentiate into the placenta and ovules. One and a half months after pollination ovules complete their differentiation, tubes resume growth and fertilisation occurs. Background and Aims

Stigmatic receptivity plays a clear role in pollination dynamics; however, little is known about the factors that confer to a stigma the competence to be receptive for the germination of pollen grains. In this work, a developmental approach is used to evaluate the acquisition of stigmatic receptivity and its relationship with a possible change in arabinogalactan-proteins (AGPs).

Methods

Flowers of the domestic apple, Malus × domestica, were assessed for their capacity to support pollen germination at different developmental stages. Stigmas from these same stages were characterized morphologically and different AGP epitopes detected by immunocytochemistry.

Key Results

Acquisition of stigmatic receptivity and the secretion of classical AGPs from stigmatic cells occurred concurrently and following the same spatial distribution. While in unpollinated stigmas AGPs appeared unaltered, in cross-pollinated stigmas AGPs epitopes vanished as pollen tubes passed by.

Conclusions

The concurrent secretion of AGPs with the acquisition of stigmatic receptivity, together with the differential response in unpollinated and cross-pollinated pistils point out a role of AGPs in supporting pollen tube germination and strongly suggest that secretion of AGPs is associated with the acquisition of stigma receptivity.Key words: AGPs, arabinogalactan proteins, apple, Malus × domestica, pollen, pollen tube, stigma, stigmatic receptivity, flower receptivity  相似文献

Our data show that Anemone canadensis L. is xenogamous and has adaptations that should increase the opportunity for sexual selection and minimize interference between female and male sexual function. The flowers of plants within insect exclosures and hand self-pollinated flowers set very few fruits compared to those that were open-pollinated and hand crosspollinated. Examination of styles with uv-fluorescence microscopy showed that self pollen tubes grew into the transmission tissue but rarely entered the ovary, i.e., there is a gametophytic self-incompatibility system. A number of traits should enhance sexual selection. First, pollen grains arrived on the stigmas beginning on day 1, germinated, but the pollen tubes grew very slowly until day 4 or 5 when the transmission tissue matured. This allowed large numbers of pollen grains to reach the stigmas, presumably from a number of pollen parents. Second, basipetal maturation of the transmission tissue, which underlies the elongate stigmatic surfaces, allows pollen tubes originating near the tip of the style to initiate rapid growth prior to those lower in the style. Thus, rapidly growing pollen tubes from the top of the style were able to catch up with those lower in the style. In essence, all the pollen tubes in a style have an equal opportunity to reach the ovule. Large stigmatic pollen loads and high fruit set suggest there is minimal interference between male and female sexual function. This may be a consequence of the spatial separation of the pistils and stamens, which increases during the life of a flower. Synchrony of function among the pistils of a flower in conjunction with large and/or multipatemal pollen loads may reduce the variance in competition among developing embryos and result in fruits of equivalent size.  相似文献

Xerophyllum tenax is a mass-flowering, nectarless herb in which self-pollination is unavoidable as anthers shed pollen onto the three, receptive stigmatic ridges attached to each pistil within a few hours after expansion of the perianth. We compared the pollination system with reproductive success in this species through controlled, hand-pollination experiments. Ovaries of flowers sampled from unbagged inflorescences were visited by pollen-eating flies (primarily members of the family Syrphidae), beetles (primarily Cosmosalia and Epicauta spp.), and small bees, and produced normal-sized capsules and mature seeds. Ovaries of flowers from inflorescences bagged to prevent insect pollination produced small capsules containing undeveloped or no seeds. Epifluorescence analyses suggest that 0.95 of the uncovered flowers are cross-pollinated by insects with pollen tubes penetrating style and ovary tissue. Flowers show a "leaky" but early-acting self-incompatibility system. While hundreds of pollen tubes germinate on each stigmatic surface following self-pollination, few pollen tubes penetrate the stigmatic surface and none penetrate the ovary. In contrast, when stigmas are cross-pollinated by hand with pollen from a second inflorescence pollen tubes were seen penetrating style and ovary. Self-incompatibility in X. tenax parallels that of some species of Trillium, a sister genus within the Melanthiaceae.  相似文献

The pistil of flowers is a specialized organ which contains the female gametophytes and provides the structures necessary for pollination and fertilization. Pollen deposited on the stigmatic surface of a compatible plant germinates a pollen tube which penetrates the stigmatic papillae and grows intercellularly through the style towards the ovules in the ovary. Pollen tube growth is largely restricted to the transmitting tissue in the style. Therefore the stylar transmitting tissue is extremely important for the migration of the pollen cell towards the ovary. We have isolated two related cDNAs, transmitting tissue-specific (TTS)-1 and TTS-2, derived from two proline-rich protein (PRP)-encoding mRNAs that accumulate specifically in the transmitting tissue of tobacco. The deduced PRP sequences share similarities with proline-rich cell wall glycoproteins found in a variety of plants. TTS-1 and TTS-2 mRNAs are induced in very young floral buds, accumulate most abundantly during the later stages of flower development when style elongation is the most rapid, and remain at relatively high levels at anthesis. These mRNAs become undetectable in maturing green fruits. In situ hybridization shows that TTS-1 and TTS-2 mRNA accumulation is restricted to the transmitting tissue of the style. The possible roles that these transmitting tissue-specific PRPs may play in maintaining the structural integrity of the style or in the function of this organ is discussed.  相似文献

Seed set after selfing in E. grandiflorum is often reduced relative to seed set after crossing; however, the compatibility patterns seen are not due to genes of major effect (i.e., S alleles). There is quantitative variation in the proportion of pollen tubes reaching the base of the style after both self- and cross-pollinations. Pollen tubes require between 24 and 72 hr to reach the ovary, but pollen tube growth ceases after 72 hr. When styles were removed from the ovaries 5 days after pollination, between 10 and 80% of the pollen tubes in the stigma had not reached the base of the style. The number of pollen tubes at the base of the style is a much better predictor of seed set than is the number of pollen tubes in the stigma. Pollen tube attrition is not affected by the age of the recipient flower or by the number of pollen donors contributing to the stigmatic pollen pool. The number of pollen tubes reaching the base of the style is dependent upon the source of the pollen and appears to be a decelerating function of the number of pollen tubes present in the stigma.  相似文献

Dichogamy in five taxa of Pulsatilla (P. patens, P. pratensis, P. vernalis, P. vulgaris ssp. gotlandica and P. vulgaris ssp. vulgaris ) is incomplete with overlap between the female and male phases of a flower. The species are protogynous with the duration of the obligate female phase lasting 0 to 10 d and the duration of stigma receptivity of unpollinated flowers lasting 3.5 to 32 d in the greenhouse. The pollen presentation time varied between 1.5 and 10 d. There were significant differences among species in duration of both female and male phases. Hand pollination decreased the duration of stigmatic receptivity and pollen presentation significantly. The spatial separation of pollen presentation and receptive surfaces in a flower changed with time due to the continuing growth of the androecium and the gynoecium, and due to the temporal pattern of pollen presentation within the androecium. Hand pollination experiments indicate that seed set may be partly pollen-limited in a natural population of P. vulgaris ssp. vulgaris. The level of cross-pollination was high in this population as shown by high levels of pollen deposition on the stigmas during the obligate female phase.  相似文献

Stylar micromorphological diversity of 42 Asteraceae species from Argentina was analysed considering species phylogenetic membership and some floral reproductive functions (pollen presentation and pollen reception). In particular, the morphology and organisation of pollen presenter (sweeping hairs) and pollen receptive structures (stigmatic papillae) were described. Results showed that style morphology of the studied species is far more diverse than the categories previously established for Asteraceae, and that it is problematic to relate the sweeping-hair arrangement of species to the only three modes of pollen presentation described for the family, indicating that the hypothesised relationship could be more complex than was formerly thought. For all species with di- or trimorphic florets, the styles of female florets were more slender and without or with more reduced sweeping hairs than the styles of hermaphrodite florets, and divergences of sweeping hair arrangements and morphology were higher among phylogenetically related species. These results suggest that functional aspects of floral morphology seem to be more important than phylogenetic constraints as selective forces determining stylar pollen presentation structures. In contrast, stigmatic-area organisation as well as the morphology of stigmatic papillae remain identical between female and hermaphrodite florets and among phylogenetically related species. Thus, stigmatic papilla morphology seems to be a phylogenetically constrained character in the studied species. The superior performance of F1 hybrids has a significant impact on agricultural productivity. For commercial application, the availability of an efficient system for obtaining male-sterile lines of crops is an essential prerequisite. Here we have investigated the use of RNA interference (RNAi) technology to silence a male-specific gene in the model host tobacco. TA29 is expressed exclusively in anthers at the time of microspore development. About 10 out of 13 tobacco lines transformed with a hairpin RNAi construct containing TA29 sequences were male sterile. Transgenic plants were phenotypically indistinguishable from non-transgenic plants. At the anthesis stage, pollen grains from transgenic, male-sterile plants were aborted and lysed in comparison to the round and fully developed pollen in non-transgenic plants. Microscopic analysis of anthers showed selective degradation of tapetum in transgenic plants with no microspore development. One week after self-pollination, the ovules of non-transgenic plants were double the size of those in transgenic plants, due to successful self-fertilization. Male sterile transgenic plants set seed normally, when cross-pollinated with pollen from non-transgenic plants, confirming no adverse effect on the female parts of the flower. These results show that silencing of male-specific genes by RNAi is potentially a useful tool for generating male-sterile lines for producing hybrid seed.  相似文献
