Summary The diversity of four esterase loci was studied electrophoretically in 690 individual spikes representing 12 populations of wild barley (Hordeum spontaneum C. Koch.) collected from central, peripheral and marginal regions of its distribution in Jordan. A minimum of 6, 10, 5 and 5 alleles were observed at the Est-1, Est-2, Est-4 and Est-5 loci, respectively. Est-2 and Est-4 were the most diverse loci (Hc=0.53±0.05 and 0.46±0.07, respectively). Est-5 was intermediate (Hc=0.33+0.07) and Est-1 was the lowest (Hc=0.22±0.04). Polymorphism was highest in the central populations (He=0.52±0.04), followed by the peripheral (He=0.40±0.05) and the marginal (He=0.22±0.05) populations. Average allelic diversity between (Gst=0.49) and within (Hs=0.51) populations reflects a high allelic differentiation among these populations. Log-linear analyses revealed that four two-locus terms and two three-locus terms were significantly associated (P<0.05). Geographical distances between populations were not significantly correlated with Nei's genetic similarity index (r=0.16; P<0.19). It is postulated that diversifying selection is a major factor in the population genetic differentiation of these esterase loci.  相似文献

Summary The linkage relationship among the loci Hor1, Hor2, Ml-k and Ml-a on the short arm of chromosome 5 was studied by progeny testing the F2 generation of two crosses. The loci Hor1 and Hor2 code for polypeptides of the storage protein hordein (prolamin) and the loci Ml-k and Ml-a determine the resistance reaction with some powdery mildew fungi cultures. The order of the loci is Ml-k, Hor1, Ml-a, and Hor2, the first named being nearest the centromere. The recombination percentage between Hor1 and Hor2 was determined in the F1 and F2 generations in both crosses, the combined estimate being 7.4±0.9 per cent. The recombination percentage estimated between Ml-k and Hor1 was 4.0±1.3, between Hor1 and Ml-a, 5.3±1.1, and between Ml-a and Hor2, 6.1±1.2. The estimates involving the Ml- loci were all probably a little too high.  相似文献

Enzyme electrophoreses were used to estimate genetic variation in five populations of Epipactis helleborine from two National Parks (Biebrza and Wigry) in northeast Poland. It has been proved that populations from these two regions differed in genetic structure, with the populations belonging to the Biebrza group having a higher level of genetic variation than those from Wigry. The number of polymorphic loci (P) ranged from 18.2% to 40.9% and the mean number of alleles per polymorphic locus (Ap) from 2.25 to 2.80. Although the observed heterozygosity (Ho) was lower than the expected one (He) in each population, the values of Ho and He were 2–3 times lower in the populations from Wigry than in those from Biebrza. Excluding the LUK population with the smallest genotypic diversity, the majority of 345 distinct multilocus genotypes occurred only once, sporadically 2–4 times in each population and their frequency ranged from 0.2% to 3.7%. Moreover, factors shaping genetic structure of E. helleborine and their intensity varied in the populations studied. Reproduction from seeds seems to influence greatly the genetic variation of the populations from the Biebrza National Park, while an assortative mating between related individuals or population size appears to be more important in the case of the populations from the Wigry National Park.  相似文献

ISSR markers were used to analyze the genetic diversity and genetic structure of eight natural populations of Cupressus chengiana in China. ISSR analysis using 10 primers was carried out on 92 different samples. At the species level, 136 polymorphic loci were detected. The percentage of polymorphic bands (PPB) was 99%. Genetic diversity (H e) was 0.3120, effective number of alleles (A e) was 1.5236, and Shannon’s information index (I) was 0.4740. At the population level, PPB = 48%, A e=1.2774, H e=0.1631, and I=0.2452. Genetic differentiation (G st) detected by Nei’s genetic diversity analysis suggested 48% occurred among populations. The partitioning of molecular variance by AMOVA analysis indicated significant genetic differentiation within populations (54%) and among populations (46%; P < 0.0003). The average number of individuals exchanged between populations per generation (N m ) was 0.5436. Samples from the same population clustered in the same population-specific cluster, and two groups of Sichuan and Gansu populations were distinguishable. A significantly positive correlation between genetic and geographic distance was detected (r=0.6701). Human impacts were considered one of the main factors to cause the rarity of C. chengiana, and conservation strategies are suggested based on the genetic characters and field investigation, e.g., protection of wild populations, reestablishment of germplasm bank, and reintroduction of more genetic diversity.  相似文献

Noble crayfish (Astacus astacus) is listed as vulnerable by the IUCN Red List of Threatened Species in Europe. However, very little is known about genetic diversity and structuring of noble crayfish populations, mainly because of the lack of informative genetic markers. We describe the isolation and characterization of the first microsatellite markers for this species, which were obtained by screening 4,000 recombinant clones. Eight loci revealed polymorphisms in a panel of 172 individuals from seven populations in Northern Europe. Number of alleles per locus ranged from two to 10 (average 4.4) and heterozygosity levels among populations varied from 0 to 0.80 for H o and from 0 to 0.72 for H e.  相似文献

Plant species in fragmented populations are affected by landscape structure because persistence within and migration among inhabited patches may be influenced by the identity and configuration of surrounding habitat elements. This may also be true for species of the semi-natural vegetation in agricultural landscapes. To determine the effect of landscape elements we analyzed Wood Avens (Geum urbanum L.) populations within three 4×4 km2 agricultural landscapes in Germany, Switzerland and Estonia, which differ in levels of land use intensity and habitat fragmentation. Genetic variation was determined in 15 randomly selected populations in each landscape using 10 microsatellite loci. The landscape structure was assessed at two circles around each population, with radii defined by the range limits of spatial genetic autocorrelation. Multiple regression analysis was used to determine the influence of landscape structure variables for inter- and intrapopulation genetic diversity. Gene diversity was equally high in Germany (He=0.27) and Switzerland (He=0.26) but lower in Estonia (He=0.16). A high overall inbreeding coefficient (FIS=0.89) was found, as expected for a selfing breeding system in G. urbanum. Genetic differentiation among populations was high (overall FST=0.43, 0.48, and 0.45 in Estonia, Switzerland and Germany, respectively), and did not differ among the three landscapes. Only a moderate influence of individual land use types on genetic diversity within and among populations was found with some idiosyncratic relationships. Genetic variation within populations was correlated to the amount of hedgerows positively in Estonia but negatively in Switzerland. The study demonstrates that the distribution of individual land use types affects the genetic pattern of a common plant species. However, different variables were identified to influence the genetic structure in three different landscapes. This indicates a major influence of landscape-specific land use history and stochastic processes determining gene flow and plant population structure.  相似文献

以内蒙古北沙柳(Salix psammophila)国家种质资源库内9个群体(P1~P9)288个无性系为实验材料,利用TP-M13-SSR技术,选取22对具有多态性EST-SSR北沙柳引物,采用毛细管电泳对PCR产物进行检测,分析北沙柳遗传多样性、分化程度和群体遗传结构,为北沙柳种质资源库遗传管理、无性系鉴定、品种选育、遗传改良和构建指纹图谱提供理论依据。结果显示:(1)22对EST-SSR引物共检测到222个等位基因,各位点平均等位基因数(A)为10,四倍体基因型丰富度(G)和特异基因型(G1)总和分别为1 460和802个,平均特异基因型比率(P1)和种质鉴别率(P2)分别为45.86%和13.21%。(2)9个群体平均等位基因数(A)为7.475,基因型丰富度(G)为15.586,观察杂合度(Ho)和期望杂合度(He)分别为0.577和0.638。以期望杂合度He为标准,北沙柳群体遗传多样性水平最低的是P1和P9。(3)北沙柳群体遗传分化系数仅为0.02,AMOVA分子变异分析显示,北沙柳群体大部分遗传变异来自群体内(97%),群体间变异仅为3%。(4)三维主成分、聚类和Structure群体遗传结构分析显示,9个群体被划分为2个组,Mantel检验表明北沙柳遗传距离与地理距离极显著相关(r=0.684 P0.001)。研究表明,北沙柳种质资源具有丰富的遗传多样性,这是其具有耐旱、耐寒、耐高温、耐沙埋和抗风蚀等适应性较强的分子基础;北沙柳的遗传变异集中在群体内;分布区群体呈现由中心向边缘群体扩张分化的趋势。  相似文献

The Caesalpinia hintonii complex is formed by five endemic species (C. hintonii, C. laxa, C. macvaughii, C. melanadenia and C. epifanioi) occurring in central Mexico. This species complex is under incipient genetic divergence as by-product of local adaptations in reproductive and morphological traits to different habitats. We estimate the genetic variation and structure of populations of this species complex to assess the extent of genetic differentiation among populations and related species along its geographic distribution. Estimations of genetic diversity and structure were done based on ten enzymes and 18 loci. Mean number of alleles per locus ranged from 1.5 to 1.9. Polymorphic loci ranged from 42.1 to 68.4. Observed (Ho: range 0.191–0.275) and expected (He: range 0.205–0.317) heterozygosities in this complex were higher compared with other endemic and legume species. Neis genetic diversity estimates showed that most genetic variation was found within (HS = 0.325) rather than among populations (DST = 0.085). Populations of the species C. hintonii showed a considerable genetic differentiation (FST = 0.207). The results of genetic diversity and structure within and among populations are in accord with the great morphological differentiation described for this species complex.  相似文献

The existence of Oryza glumaepatula is threatened by devastation and, thus, the implementation of conservation strategies is extremely relevant. This study aimed to characterize the genetic variability and estimate population parameters of 30 O. glumaepatula populations from three Brazilian biomes using 10 microsatellite markers. The levels of allelic variability for the SSR loci presented a mean of 10.3 alleles per locus and a value of 0.10 for the average allelic frequency value. The expected total heterozygosity (He) ranged from 0.63 to 0.86. For the 30 populations tested, the mean observed (Ho) and expected heterozygosities (He) were 0.03 and 0.11within population, respectively, indicating an excess of homozygotes resulting from the preferentially self-pollinating reproduction habit. The estimated fixation index ( IS ) was 0.79 that differed significantly from zero, indicating high inbreeding within each O. glumaepatula population. The total inbreeding of the species (IT ) was 0.98 and the genetic diversity indexes among populations, ST and ST, were 0.85 and 0.90, respectively, indicating high genetic variability among them. Thus, especially for populations located in regions threatened with devastation, it is urgent that in situ preservation conditions should be created or that collections be made for ex situ preservation to prevent loss of the species genetic variability.  相似文献

白芨(Bletilla striata Rchb.)为中国珍稀濒危植物,重要的药用植物。该研究基于Illumina测序技术构建白芨基因组文库和微卫星文库,设计白芨微卫星引物,用白芨4个野生种群80个个体对引物进行多态性检测,应用4个白芨近缘种中进行引物的通用性检测,并在此基础上分析了白芨的遗传多样性和遗传分化,以探讨白芨的遗传结构和进化潜能。结果表明:(1)白芨基因组中微卫星片断丰富,共检测出17 841条微卫星片断。基于白芨微卫星库对100个位点设计了引物对,经PCR扩增和检测筛选出能够稳定扩增的多态性位点20个,每个位点的复等位基因数(Na)在2~6之间,平均为3.85;20对引物的大部分能够在4个白芨近缘种中成功扩增。(2)白芨在物种水平均有较高的遗传多样性(Na=3.85,I=1.07,H=0.614 7),白芨种群遗传分化强烈(Gst=0.43),居群间的基因流较弱(Nm=0.867 6),居群聚类分析结果均表明地理距离较近的居群具有较近的遗传关系。  相似文献

Freshwater molluscs are globally critically threatened, yet little is known about their genetic diversity. We describe the isolation and characterization of the first microsatellite markers for the European swan mussel, Anodonta cygnea. Nine loci were polymorphic in a panel of 110 individuals from seven German and Estonian populations. Number of alleles ranged from 2 to 14 and heterozygosity levels ranged from 0.021 to 0.498 for H o and from 0.069 to 0.634 for H e. Deficiency of heterozygous genotypes was observed in six loci. The length of the microsatellite repeat motives was positively correlated with the allele numbers in the respective loci.  相似文献

Six polymorphic microsatellites (eight loci) were used to study the genetic diversity and population structure of common carp from Dongting Lake (DTC), Poyang Lake (PYC), and the Yangtze River (YZC) in China. The gene diversity was high among populations with values close to 1. The number of alleles per locus ranged from 2 to 11, and the average number of alleles among 3 populations ranged from 6.5 to 7.9. The mean observed (H O) and expected (H E) heterozygosity ranged from 0.4888 to 0.5162 and from 0.7679 to 0.7708, respectively. Significant deviations from Hardy–Weinberg Equilibrium expectation were found at majority of the loci and in all three populations in which heterozygote deficits were apparent. The analysis of molecular variance (AMOVA) indicated that the percent of variance among populations and within populations were 3.03 and 96.97, respectively. The Fst values between populations indicated that there were significant genetic differentiations for the common carp populations from the Yangtze River and two largest Chinese freshwater lakes. The factors that may result in genetic divergence and significant reduction of the observed heterozygosity were discussed.  相似文献

We examined genetic variation in allozyme loci, nuclear DNA restriction fragment length polymorphisms (RFLPs), and random amplified polymorphic DNAs (RAPDs) in 130 trembling aspen (Populus tremuloides) and 105 bigtooth aspen (P. grandidentata) trees. In trembling aspen 10 out of 13 allozyme loci assayed (77%) were polymorphic (P), with 2.8 alleles per locus (A) and an expected heterozygosity (He) of 0.25. In contrast, bigtooth aspen had a much lower allozyme genetic variability (P=29%; A=1.4; He=0.08). The two species could be distinguished by mutually exclusive alleles at Idh-1, and bigtooth aspen has what appears to be a duplicate 6PG locus not present in trembling aspen. We used 138 random aspen genomic probes to reveal RFLPs in HindIII digests of aspen DNA. The majority of the probes were from sequences of low copy number. RFLP results were consistent with those of the allozyme analyses, with trembling aspen displaying higher genetic variation than bigtooth aspen (P=71%, A=2.7, and He=0.25 for trembling aspen; P=65%, A=1.8, and He=0.13 for bigtooth aspen). The two species could be distinguished by RFLPs revealed by 21 probes (15% of total probes assayed). RAPD patterns in both species were studied using four arbitrary decamer primers that revealed a total of 61 different amplified DNA fragments in trembling aspen and 56 in bigtooth aspen. Assuming a Hardy-Weinberg equilibrium, estimates of P=100%, A=2, and He=0.30 in trembling aspen and P=88%, A=1.9, and He=0.31 in bigtooth aspen were obtained from the RAPD data. Five amplified DNA fragments were species diagnostic. All individuals within both species, except for 2 that likely belong to the same clone, could be distinguished by comparing their RAPD patterns. These results indicate that (1) RFLPs and allozymes reveal comparable patterns of genetic variation in the two species, (2) trembling aspen is more genetically variable than bigtooth aspen at both the allozyme and DNA levels, (3) one can generate more polymorphic and species-specific loci with DNA markers than with allozymes in aspen, and (4) RAPDs provide a very powerful tool for fingerprinting aspen individuals.  相似文献

Rivers in Asturias (northern Spain) constitute the southern limit of the distribution of Atlantic salmon (Salmo salar L.) in Europe, a biological resource facing one of the more serious challenges for conservation today. In this work, eight microsatellite loci have been used to analyse samples collected in 1993 and 1999 from four Asturian rivers (Esva, Narcea, Sella, and Cares), obtaining information about the temporal and the spatial genetic variation in these populations and, in addition, estimations of their effective population sizes. The temporal analysis revealed a general decrease in all the estimated genetic variability parameters when samples from 1993 (mean A (1993) = 6.47, mean H O(1993) = 0.472, mean H E(1993) = 0.530) were compared with those obtained in 1999 (mean A (1999) = 6.16, mean H O(1999) = 0.460, mean H E(1999) = 0.490). This reduction was particularly notable for the case of the Esva river. Our results pointed to a pattern of spatial genetic differentiation inside the Asturian region (F ST (1993) = 0.016 P < 0.01; F ST (1999) = 0.023 P < 0.01). Using the standard Temporal Method we found estimates of N e^ (Esva) = 75.1 (33.2–267.2); N e^ (Cares) = 96.6 (40.0–507.5), N e^ (Sella) = 106.5 (39.1–9396.4) and N e^ (Narcea) = 113.9 (42.0–3693.3). The use of likelihood-based methods for the N e^ estimations improved the results (smaller CIs) for the Esva and Cares rivers (N e^ (Esva) = 63.9 (32.3–165.3); N e^ (Cares) = 76.4 (38.8–202.0) using a Maximum likelihood approach) and suggested the presence of larger populations for the Sella and Narcea rivers (N e^≈200). These results showed that the Asturian Atlantic salmon populations (in particular Esva and Cares river populations) could be close to the conservation genetic borderline for avoiding inbreeding depression although we discuss some implications of the analysis of temporal genetic change in populations with overlapping generations.  相似文献

Delphinium staphisagria is an endemic annual or biennial herb from the Mediterranean Basin, widely distributed in isolated populations of variable size. We evaluated the allozyme diversity of 31 populations along its distribution range via starch gel electrophoresis, assaying 12 enzyme systems and scoring 17 loci. The low levels of genetic variability detected (A = 11.8, A p = 1.6, H o = 0.026, H e = 0.057), are discussed in relation to the life-history traits of the species, such as short life-span, selfing or gravity seed dispersion. Other factors influencing genetic diversity, such as evolutionary history and spreading are also considered. Due to its historical medicinal uses, this plant has probably become widespread in the Mediterranean area. Human-mediated distribution could have promoted few migrant genotypes, recent founder events and long distance dispersal. These events would explain the genetic homogeneity found within and among populations, as well as the absence of a clear biogeographic structure. The limited genetic variability, the high genetic similarity among populations and the dysploidy of this species make it worthy of conservation. Management strategies are proposed mainly to preserve its genetic pool.  相似文献

Allozyme variation at eleven loci encoding seven enzyme systems were examined in 20 populations of diploid (genome AA, 2n = 16)Scilla scilloides in China. In comparison with the average species of seed plants studied, populations of this species display a high amount of genetic variation (A = 2.0, P = 58.6%, Ho = 0.172, and He = 0.185). Allozyme variation pattern revealed predominant outcrossing within populations and considerable differentiation (FST = 0.314) among populations as well as between the subtropic and temperate regions. The wide distribution, long existence and outcrossing are presumably the main factors responsible for the high genetic diversity within populations. But the gravity dispersal of seeds and pollination by small insects set limits to the increase of genetic variation within populations and promote differentiation between populations and regions. In addition, allozyme variation does not distinguishS. scilloides var.albo-viridis and suggests that subtropic populations may be considered as a genetic entity.  相似文献

We tested the cross-amplification of 26 microsatellites developed for passerines and an additional three developed for Gallinula species in eight European Coots from two populations. Sixteen microsatellite markers successfully amplified, of which nine were polymorphic with 2–6 alleles (mean 3.7 alleles) and an expected heterozygosity (H e) ranging from 0.375 to 0.805 (mean H e = 0.589). On average, we found 2.22 alleles/locus and a mean H e of 0.440 in one nest, and 2.56 alleles/locus and a mean H e of 0.494 in the other one. These nine polymorphic markers could be of potential use in studies of genetic variability, population structure and reproductive strategy of European Coots.  相似文献

The evolutionary potential of plant species that reproduce via predominantly clonal means and the conditions under which clonality is favoured are not well known. Long‐term clonal reproduction is expected to result in a number of readily detectable genetic signals not present in populations that reproduce by sexual means. We use a hierarchical sampling strategy to assess genotype probabilities and confirm that two rare sister taxa of Banksia ionthocarpa have contrasting modes of reproduction. Banksia ionthocarpa subsp. chrysophoenix reproduces clonally. Populations had low levels of genotypic diversity and were comprised of large clonal patches consisting of many ramets that covered hundreds of square metres and showed little intermixing. The taxon was genetically depauperate (mean Na = 1.97, mean P = 0.66, mean He = 0.282), had high levels of genetic differentiation between populations (θ = 0.481), and populations exhibited excess heterozygosity and linkage disequilibrium (LD) among loci, suggesting historically high levels of clonality. In contrast, the sister taxon B. ionthocarpa subsp. ionthocarpa, which occurs in an area with more than twice the annual rainfall and less extreme minimum and maximum temperatures, showed no evidence of clonality, high levels of genotypic diversity, greater genetic diversity (mean Na = 3.31, mean P = 0.81, mean He = 0.405), lower levels of genetic differentiation between populations (θ = 0.253) and no evidence of excess heterozygosity or LD among loci. We suggest that the development of clonality in subsp. chrysophoenix is associated with its more marginal environment and enhanced by response to recurrent fires.  相似文献

Theobroma grandiflorum (cupuassu) is an important fruit tree native to the Brazilian Amazon. Establishing the genetic diversity and structure of populations is critical to define long-term strategies for cupuassu conservation presently threatened by rapid deforestation. Three natural populations collected at the putative center of diversity, three groups of accessions established at a germplasm collection, and one derived from commercial plantings were analyzed. The genetic diversity was assessed using 21 polymorphic microsatellite loci originally developed for Theobroma cacao, disclosing a total of 113 alleles. The estimated genetic diversity parameters averaged over cupuassu populations (A = 3.53 alleles per locus; H e = 0.426; H o = 0.346) were lower than the values reported for other Neotropical tree species. The three natural populations presented a positive and significant fixation index (f), ranging from 0.133 to 0.234. Cupuassu apparently adhered to a general pattern of genetic diversity structure of some Neotropical tree species occurring at low densities, with a low intrapopulation genetic diversity and important levels of endogamy, possibly due to biparental inbreeding derived from the presence of spatial genetic structure in the populations. A high level of genetic divergence was detected among the natural populations (θ p = 0.301), a strong differentiation caused by limited gene flow, and suggesting that human interference in spreading and/or stimulating plantings might have had a smaller effect than expected. The approximate location of the T. grandiflorum center of diversity could not be confirmed by analyzing natural populations from the putative region.  相似文献
