Wound healing in the integument of the sea cucumber, Thyone briareus, was studied for up to 50 days after inflicting wide excisional wounds and for 14 days after producing incisional wounds. Rapid re-epithelialization of the wound was effected by the migration of epidermal cells and pigment cells from the periphery of the wound margin. This occurred without apparent evidence of concomitant mitotic activity. Dermal wound healing was completed by the fourteenth day in the incision wounds but occurred very slowly in the broad excision wounds. Morula cells seem to be involved in both epidermal and dermal wound healing, although their precise role is unknown. In excisional wounds the integument was never completely restored to its normal appearance during 50 days of observation.  相似文献

The ultrastructure of the centriolar satellite complex in the spermatozoon of the hydroid, Pennaria, is presented. The complex consists of nine spoke-like arms which emanate from the distal centriole and of nine wedgeshaped connectives which extend into the flagellum joining each of the alpha doublets and the flagellar plasmalemma. Based upon these observations a new model for the structure of the centriolar satellite complex is proposed. In addition, similar centriolar satellite complexes are reported in the spermatozoa of two echinoderms, Ctenodiscus crispatus and Thyone briareus, in support of the proposed model.  相似文献

Recently, an increasing number of studies have shown that Lake Van—the biggest soda lake in the world—is polluted due to an increasing population. Studies have shown abnormalities in the Lake Van fish (Chalcalburnus tarichi), the sole fish species that inhabits the lake. Unlike the vitellogenic and mature oocytes in normal gonads, abnormal gonads show large amounts of connective tissue and young oocytes. In this study, metal levels (nickel [Ni], copper [Cu], cobalt [Co], iron [Fe], zinc [Zn], cadmium [Cd], lead [Pb], and manganese [Mn]) in the muscle, liver, gill, intestine, and gonad of Lake Van fish with normal and abnormal gonads were assessed. Further, the metal contents in the wastewater from the wastewater treatment facility situated near Lake Van in Van City were assessed. All the metal levels, except that of Zn, were high in the Lake Van environment (P?<?0.05). The highest metal content in the tissues was for Fe, while the lowest level was for Co. The Pb level was found to be very high in both fish groups. Cd was not found in the tissues of both fish groups. The levels of Fe, Cu, Pb, and Mn were not significant in the tissues of both normal and abnormal fish groups. Zn level was significantly high in the livers and gonads of fish with abnormal gonads, and Co level was significantly high only in the livers (P?<?0.05). Consequently, high levels of Zn in the liver and gonads and high levels of Co in the liver may be factors causing the abnormal gonads in the Lake Van fish.  相似文献

In recent years, jellyfish blooms have attracted considerable scientific interest for their potential impacts on human activities and ecosystem functioning, with much attention paid to jellyfish as predators and to gelatinous biomass as a carbon sink. Other than qualitative data and observations, few studies have quantified direct predation of fish on jellyfish to clarify whether they may represent a seasonally abundant food source. Here we estimate predation frequency by the commercially valuable Mediterranean bogue, Boops boops on the mauve stinger jellyfish, Pelagia noctiluca, in the Strait of Messina (NE Sicily). A total of 1054 jellyfish were sampled throughout one year to quantify predation by B. boops from bite marks on partially eaten jellyfish and energy density of the jellyfish. Predation by B. boops in summer was almost twice that in winter, and they selectively fed according to medusa gender and body part. Calorimetric analysis and biochemical composition showed that female jellyfish gonads had significantly higher energy content than male gonads due to more lipids and that gonads had six-fold higher energy content than the somatic tissues due to higher lipid and protein concentrations. Energetically, jellyfish gonads represent a highly rewarding food source, largely available to B. boops throughout spring and summer. During the remainder of the year, when gonads were not very evident, fish predation switched towards less-selective foraging on the somatic gelatinous biomass. P. noctiluca, the most abundant jellyfish species in the Mediterranean Sea and a key planktonic predator, may represent not only a nuisance for human leisure activities and a source of mortality for fish eggs and larvae, but also an important resource for fish species of commercial value, such as B. boops.  相似文献

In fish, exposure to waters of low pH causes significant damage to the gill resulting in fatal iono- and osmoregulatory disturbance. In amphibians, exposure to acid waters also disrupts ionic homeostasis, however the extent and nature of injuries to amphibian larvae from acid exposure are poorly understood. Changes in gross morphology and ultrastructure of the gills and integument were examined, together with measures of Na+ efflux/uptake, in larval Litoria fallax (Amphibia: Anura) following acute acid exposure. Examination of tissues revealed significant changes in morphology and ultrastructure of both gills and the integument following acutely lethal exposure to low pH water. Changes to the gills of acid-exposed L. fallax larvae included lifting of the branchial epithelium and opening of tight junctions between pavement cells (with a consequent reduction in tight junction length). Damage to epithelial cell–cell tight junctions was also apparent at the integument along with widespread oncosis and localised epithelial necrosis. Mucous secretory activity at the gills and body surface was largely unaffected by acid exposure, with little or no difference in density, cross-sectional area and number of epithelial mucous secretory vesicles in acid-exposed and control larvae. Changes in morphology and ultrastructure at low pH were accompanied by significant Na+ loss (up to 50% of the total body Na+ content) attributable in large part to increased paracellular ionic efflux across the gills as well as increased transcellular and paracellular efflux of ions across the integument.  相似文献

The daubed shanny, Leptoclinus maculatus, is a common fish species in Arctic and North Atlantic waters and has an important role in high-latitude ecosystems as a link between lower trophic levels and many fish, marine mammal and seabird species. Its biology and ecology have, however, remained largely unstudied. The primary aim of this study was to increase the knowledge about the daubed shanny by analysing total lipids, lipid-classes and the fatty acid composition of liver, muscle and female gonads in adults from the high Arctic archipelago of Svalbard. In female gonads, the triacylglycerols and wax esters in addition to cholesterol esters were dominant among the stored lipids. Triacylglycerols dominated in the liver, whereas structural lipids, such as phospholipids and cholesterols, were the most important lipids in muscles. Phosphatidylcholine and phosphatidylethanolamine were major phospholipids in all organs studied. The fatty acid spectrum of the investigated organs was characterized by a high amount of monounsaturated fatty acids, particularly in the liver. Polyunsaturated fatty acids, particularly 22:6(n-3) and 20:4(n-6), were prevalent in muscle tissues. The lipid and fatty acid spectra in the organs during this period of life are tightly connected with the activation of the liver metabolism and the storage of lipids in the developed female gonads. Lipid accumulation and distribution in gonads are transferred to optimal development of embryos and larvae in Arctic waters.  相似文献

The integument and podia of the sea cucumber Thyone briareus were examined by bright field and electron microscopy. The epidermal surface was found to be covered by an acellular, PAS positive cuticle which appeared to be secreted by the underlying epidermal cells. Although the superficial portion of the cuticle contains numerous fine filaments, their ultrastructure bears no resemblance to collagen fibers. The epidermal cells are widely spaced and have long apical processes that extend along the under surface of the cuticle forming a contiguous epithelium. The apical expansions of the epidermal cells are attached to one another by means of septate desmosomes which may run continuously around all epidermal cells. Special attachment structures within these apical expansions appear to bind the cuticle to the dermis. The epidermal cells and their apical expansions are separated from the dermis by an 800 Å thick basement membrane. Granule containing cells in the upper dermis send processes up to the cuticle where they are bound to the typical epidermal cells by septate desmosomes. The abundant membrane bound granules of the cells enter villous-like processes which pass through the cuticle. The function of these cells may be to produce an adhesive material on the podia or they may be pigment cells. The thick dermis consists of a superficial zone, containing largely ground substance; a middle or laminated zone containing laminae of collagen fibers arranged in an orthogonal fashion; and a hypodermis consisting largely of ground substance and reticular fibers. Fibroblasts are abundant in the superficial dermis and between the collagen laminae. Wandering coelomocytes, or morula cells, accumulate between the collagen laminae and in the hypodermis. They may also become an integral part of the epidermis by forming septate desmosomes with epidermal cells. Morula cells contain highly specialized spherules whose tinctorial properties and electron microscopic appearance suggest that they contain protein and mucopolysaccharide.  相似文献

Germ cell transplantation (GCT) is a promising assisted reproductive technology for the conservation and propagation of endangered and valuable genetic resources. In teleost fish, GCT in adult gonads has been achieved only in male recipients, limiting greatly the usefulness of this technique in situations where both sexes need equal and timely attention for conservation and/or propagation. Here we describe a simplified GCT approach that ultimately leads to production of donor-derived eggs and sperm in considerably short time. Donor germ cells isolated from young pejerrey Odontesthes bonariensis (Atherinopsidae) were transplanted non-surgically through the genital papilla into the sexually mature gonads of Patagonian pejerrey O. hatcheri recipients whose gonads have been depleted of endogenous GCs by heat (26°C) and chemical treatment (four doses of Busulfan at 30 mg/kg and 40 mg/kg for females and males, respectively). Transplanted spermatogonial and oogonial cells were able to recolonize the recipients'' gonads and produce functional donor origin eggs and sperm within 7 months from the GCT. We confirmed the presence of donor-derived gametes by PCR in 17% and 5% of the surrogate O. hatcheri fathers and mothers, respectively. The crosses between surrogate fathers and O. bonariensis mothers yielded 12.6–39.7% pure O. bonariensis and that between a surrogate mother and an O. bonariensis father yielded 52.2% pure O. bonariensis offspring. Our findings confirm that transplantation of germ cells into sexually competent adult fish by non-surgical methods allows the production of functional donor-derived eggs and sperm in a considerably short time. The methods described here could play a vital role in conservation and rapid propagation of endangered fish genetic resources.  相似文献

The aeolid nudibranch genus Phyllodesmium (Mollusca: Gastropoda)is reviewed, three new species are described and further informationon the biology, anatomy and distribution on the eight previouslyknown species is reported. The genus Ennoia Bergh, 1896 is considereda synonym of Phyllodesmium and the type species Ennoia briareusredescribed. The genus Phyllodesmium is unique amongst the aeolidsin feeding on octocoral cnidarians. This has led to the evolutionof nudibranch- zooxanthellae symbioses, zooanthellae being obtainedfrom the octocoral prey. The transplantation of germ cells into adult recipient gonads is a tool with wide applications in animal breeding and conservation of valuable and/or endangered species; it also provides a means for basic studies involving germ cell (GC) proliferation and differentiation. Here we describe the establishment of a working model for xenogeneic germ cell transplantation (GCT) in sexually competent fish. Spermatogonial cells isolated from juveniles of one species, the pejerrey Odontesthes bonariensis (Atherinopsidae), were surgically transplanted into the gonads of sexually mature Patagonian pejerrey O. hatcheri, which have been partially depleted of endogenous GCs by a combination of Busulfan (40 mg/kg) and high water temperature (25°C) treatments. The observation of the donor cells'' behavior showed that transplanted spermatogonial cells were able to recolonize the recipients'' gonads and resume spermatogenesis within 6 months from the GCT. The presence of donor-derived gametes was confirmed by PCR in 20% of the surrogate O. hatcheri fathers at 6 months and crosses with O. bonariensis mothers produced hybrids and pure O. bonariensis, with donor-derived germline transmission rates of 1.2–13.3%. These findings indicate that transplantation of spermatogonial cells into sexually competent fish can shorten considerably the production time of donor-derived gametes and offspring and could play a vital role in germline conservation and propagation of valued and/or endangered fish species.  相似文献

The state of the reproductive system of the pike Esox lucius, descendants of individuals exposed to X-ray irradiation due to the catastrophe at the Chernobyl Nuclear Power Plant in 1986, was studied. The material was collected in water bodies of Ukraine contaminated with radionuclides—Kiev Reservoir, the Teterev River, and Lake Glubokoe in the postcatastrophe period 1999–2004. It was shown that the total number and the extent of disturbances in the pike gonads are positively correlated with the levels of contamination of the water bodies: in the cleanest water body, the Teterev River, the proportion of fish with gonads without considerable deviations was 70%; in the Kiev Reservoir, it was 45%; no individuals with normal gonads were recorded in Lake Glubokoe. The great number of anomalies in sexual cells and gonads has led to a decrease in the reproductive capacity of the pike populations studied. A unique case was revealed of synchronous hermaphroditism in a pike from the Kiev Reservoir that belonged to the fourth generation (F4) of fish that underwent emergency loads. Among postemergency pike generations, the maximum number of disturbances in sexual cells and gonads was recorded in F2–4, which is possibly accounted for by the phenomenon of “prolonged mutagenesis.”  相似文献

Regulatory effect of surfagon—a synthetic analog of gonadotropin-releasing hormone—on the gonad state of rainbow trout (Kamchatka steelhead Parasalmo mykiss (=Oncorhynchus mykiss)) juveniles is investigated. The juveniles were 2.5 months old and were exposed to brief (4 days) heat stress (19–20°C). The increase of water temperature was followed by anomalies of gonads after 1 month (destruction of sex cells and hypertrophy of connective tissue). In females, the increased water temperature activates sex redetermination: in ovaries, cysts containing destroyed spermatogonia are found. In 1.5 months, the exogenous application of surfagon was followed by acceleration of spermatogenesis in gonads of experimental fish (2.5 months old) and by a slight decrease of fraction of anomalies in structure of their testes compared with the gonads of fish not exposed to surfagon. Exposure of rainbow trout to surfagon prior to sex differentiation is more efficient than after it is completed.  相似文献

Sans-fille (SNF) is the Drosophila homologue of mammalian general splicing factors U1A and U2B″, and plays an important role in sex determination in Drosophila melanogaster. In this study, the snf gene from Antheraea pernyi (Lepidoptera: Saturniidae), an economically important insect, was isolated and characterized. The obtained 925 bp cDNA sequence contains an open reading frame of 669 bp encoding a polypeptide of 222 amino acids, showing 78% sequence identity to that from D. melanogaster. A database search revealed that SNF protein homologs are present in many animals, including invertebrates and vertebrates, with more than 70% amino acid sequence identities, suggesting that they were highly conserved during the evolution of animals. Phylogenetic analysis revealed that A. pernyi SNF was closely related to Bombyx mori SNF. Quantitative real-time PCR (qRT-PCR) analysis showed that the A. pernyi snf gene was transcribed during five larval developmental stages, and in six tested tissues (ovaries, testes, silk glands, fat body, integument, and hemolymph), with the most abundance determined in the gonads (ovaries or testes). Investigation of expression changes throughout embryonic development indicated that A. pernyi snf mRNA was expressed at a low level from days 0 to 4, and reached a maximum level at day 10, but decreased to a low level before hatching. These results suggest that the product of the snf gene may play important roles in the development of A. pernyi.  相似文献

Parasitic castration, the specific blocking of host reproductive output by an individual parasite, is a host-parasite interaction common to many invertebrates, particularly crustaceans, echinoderms and molluscs. It can reduce host density, alter host population dynamics and the evolution of host life history traits. Here we show that parasitisation by a single female cymothoid isopod, Anilocra apogonae, castrates its vertebrate host, the five-lined cardinalfish, Cheilodipterus quinquelineatus. Parasitised male fish fail to mouthbrood their young. The gonads of parasitised fish are smaller and parasitised female fish have substantially fewer and smaller ova than do the gonads of unparasitised fish. As for parasitic castrators of invertebrate hosts, A. apogonae on C. quinquelineatus are uniformly dispersed amongst infested hosts (one adult female isopod per host), are site specific, and their body size is highly correlated with that of their host. These isopods are large relative to the body size of their hosts, averaging 3.8% of the weight of the host. Parasitised fish also weigh less and are shorter than unparasitised fish of the same age. Despite the presence of other potential hosts, A. apogonae only infests C. quinquelineatus. The consistency of the ecological correlates amongst known parasitic castrators suggests that the parasitic castrator host-parasite relationship will be recognised for other parasites of vertebrates.  相似文献

N-β-Alanyldopamine (NBAD) and other diphenols in tissues of the fifth larval instar of the tobacco hornworm, Manduca sexta (L.), were analyzed by HPLC with electrochemical detection. NBAD accumulated in the integument during the intermolt feeding period, with maximal levels in the wandering stage (6 mmol/g). It then declined to a low level during apolysis and endocuticle digestion, while hemolymph NBAD increased during the same interval to a peak concentration (3 mM) shortly before pupal ecdysis. Trachea and foregut contained lesser amounts of NBAD (0.5 mmol/g), perhaps associated with cuticle, whereas fat body, muscle, midgut and hindgut had 0.1 mmol/g or less. Dopamine (DA), N-acetyldopamine and 3,4-dihydroxyphenylalanine (DOPA) were at least 10-fold less abundant than NBAD in the integument. NBAD synthase, which catalyzes the formation of NBAD from DA and β-alanine, was assayed in both integument and fat body. Highest activity was detected in the integument, where two peaks were observed, one at day 3 near the end of larval feeding and the other at day 9 as pupal cuticle tanning was initiated. Fat body enzyme was substantially less and was detected only in the pharate pupa. Maximal NBAD synthesis by integument cultured in vitro was dependent upon DA supplementation of at least 1.4 mM. 20-Hydroxyecdysone did not alter NBAD synthesis in vitro in either the integument or the fat body, even though injection of this hormone into isolated larval abdomens induced synthesis and/or transport of integumental NBAD back into the hemolymph. The rate-limiting steps in the NBAD biosynthetic pathway appear to be the production of DOPA and DA, because β-alanine occurs in the hemolymph at relatively high levels throughout larval-pupal development.  相似文献

Although considerable knowledge has been gathered regarding the role of fish in cycling and translocation of nutrients across ecosystem boundaries, little information is available on how the energy obtained from different ecosystems is temporally allocated in fish bodies. Although in theory, limitations on energy budgets promote the existence of a trade-off between energy allocated to reproduction and somatic growth, this trade-off has rarely been found under natural conditions. Combining information on RNA:DNA ratios and carbon and nitrogen stable-isotope analyses we were able to achieve novel insights into the reproductive allocation of diamond mullet (Liza alata), a catadromous, widely distributed herbivorous-detritivorous fish. Although diamond mullet were in better condition during the wet season, most reproductive allocation occurred during the dry season when resources are limited and fish have poorer body condition. We found a strong trade-off between reproductive and somatic investment. Values of δ13C from reproductive and somatic tissues were correlated, probably because δ13C in food resources between dry and wet seasons do not differ markedly. On the other hand, data for δ15N showed that gonads are more correlated to muscle, a slow turnover tissue, suggesting long term synthesis of reproductive tissues. In combination, these lines of evidence suggest that L. alata is a capital breeder which shows temporal uncoupling of resource ingestion, energy storage and later allocation to reproduction.  相似文献
