Pepper seed samples were tested for the infection of tobacco mosaic virus (TMV) and tomato mosaic virus (ToMV) by enzyme linked immunosorbent assay (ELISA). Out of 26 pepper seed samples tested, 17 were infected with TMV and ToMV in ELISA. About 34.7% of pepper seed samples were found to be healthy. Infections of TMV or ToMV were recorded to be 61.53% and 11.5%, respectively of the total tested seed samples.  相似文献

Turnip mosaic virus (TuMV) and cauliflower mosaic virus (CaMV) have been found infecting field crops of winter oilseed rape (Brassica napus ssp. oleifera) in South Warwickshire. Other viruses found include broccoli necrotic yellows virus (BNYV) and a member of the beet western yellows virus group. Systemic leaf symptoms caused by TuMV varied within and between cultivars; the three predominant reaction types were classified as necrotic, mosaic and immune. Some recently introduced cultivars of oilseed rape were more severely affected by TuMV infection than older cultivars. Reactions to CaMV were less varied and immunity was not found. The seed yield from TuMV and CaMV-infected plants was less than that of healthy control plants. This effect was due to infected plants producing either fewer seeds, smaller seeds or both. Germination of seeds from infected plants was unaffected if sown soon after harvest. After storage for one year the germination of seed from a virus infected plant was significantly less than that of seed from a virus-free plant. All commercial cultivars tested were experimentally susceptible to turnip yellow mosaic virus (TYMV) and some American strains of cucumber mosaic virus (CMV).  相似文献

Samples collected in 1994 and 1995 from commercial crops of chickpeas and lentils growing in the agricultural region of south-west Western Australia were tested for infection with alfalfa mosaic (AMV) and cucumber mosaic (CMV) viruses, and for members of the family Potyviridae using enzyme-linked immunosorbent assay (ELISA). In 1994 no virus was detected in the 21 chickpea crops tested but in 1995, out of 42 crops, AMV was found in two and CMV in seven. With lentils, AMV and/or CMV was found in three out of 14 crops in 1994 and 4 out of 13 in 1995, both viruses being detected in two crops in each year. Similar tests on samples from chickpea and lentil crops and plots growing at experimental sites, revealed more frequent infection with both viruses. No potyvirus infection was found in chickpeas or lentils in agricultural areas either in commercial crops or at experimental sites. However, bean yellow mosaic virus (BYMV) was detected along with AMV and CMV in irrigated plots of chickpeas and lentils at a site in Perth. When samples of seed from infected crops or plots of chickpeas and lentils were germinated and leaves or roots of seedlings tested for virus infection by ELISA, AMV and CMV were found to be seed-borne in both while BYMV was seed-borne in lentils. The rates of transmission found through seed of chickpea to seedlings were 0.1–1% with AMV and 0.1–2% with CMV. Seed transmission rates with lentil were 0.1–5% for AMV, 0.1–1% for CMV and 0.8% for BYMV. Individual seed samples of lentil and chickpea sometimes contained both AMV and CMV. With both species, infection with AMV and CMV was sometimes found in commercial seed stocks or seed stocks from multiplication crops of advanced selections nearing release as new cultivars. Seed-borne virus infection has important practical implications, as virus sources can be re-introduced every year to chickpea and lentil crops or plots through sowing infected seed stocks leading to spread of infection by aphid vectors, losses in grain yield and further contamination of seed stocks.  相似文献

Wild plants of Brassica oleracea (wild cabbage) are commonly infected with turnip mosaic poty virus (TuMV), turnip yellow mosaic tymovirus (TYMV) and several other viruses. A field experiment in which plants were inoculated either with TuMV or TYMV showed that virus infection significantly reduced survival, growth and reproduction. Relative to water inoculated-controls, plants infected with TYMV had greater mortality, were shorter, had a smaller leaf area and number, showed a greater amount of damage from herbivory and chlorosis, were less likely to flower and produced fewer pods and lower total seed output. Plants infected with TuMV did not appear to be adversely affected at first; however, mortality after 18 months was higher than control plants. Although TuMV infection had no effect on the number of plants flowering, the infected plants did produce fewer pods and a lower total seed output. We conclude that both viruses can significantly affect vegetative and reproductive performance of wild cabbage and hence that introgression of virus resistance (particularly when conferred by a major gene or a transgene) from a crop might increase plant fitness in natural populations of this species. Ecological risk assessments of virus resistance transgenes must do more than survey adult plants in natural populations for the presence of the target virus. Failure to detect the virus could be due to high mortality on infection with the virus.  相似文献

Cucumber mosaic virus (CMV) accumulation in leaves and stems of infected bell pepper plants at specific symptom stages was evaluated with an emphasis on the transition from full infection to recovery from Cucumber mosaic disease. Four symptom phases occurred in successive order, designated chlorosis (leaves 6–8), mosaic (leaves 9–11/12), leaf distortion (first series of leaves on secondary and tertiary branches) and recovery (progressive recovery with newly emerging leaves in tertiary and younger branches). In situ detection of CMV in leaf tissues revealed widespread occurrence in leaves expressing chlorosis and mosaic symptoms but reduced, localized occurrence in leaves in the recovery phase. Similarly, CMV accumulated to high levels throughout stems expressing chlorosis and mosaic symptoms but with dramatically reduced levels for plants in the recovery symptom phase. Stunting of internodes occurred at all locations above the inoculated leaves by the first expression of systemic symptoms, suggesting an impact on stem growth in response to initial virus invasion of young developing tissues of the stem. Despite the recovery from CMV infection, plant growth was negatively impacted early in the infection process and remained so through the course of the experiment.  相似文献

Commercial formulations of strobilurins (azoxystrobin, kresoxim‐methyl, trifloxystrobin and pyraclostrobin) were evaluated for their efficacy against Bean common mosaic virus (BCMV) in screenhouse and field conditions. Highest seed germination and seedling vigour were recorded with 20 μg/ml pyraclostrobin seed treatment in comparison with the control. In screenhouse studies, 76% protection against BCMV was recorded with pyraclostrobin seed treatment at 10 μg/ml. Under field conditions with natural BCMV inoculum, pyraclostrobin seed treatment resulted in 65% protection against BCMV. The protection offered by strobilurins against BCMV was evaluated by ELISA, with lowest immunoreactive values recorded in common bean seedlings raised from seeds treated with pyraclostrobin and kresoxim‐methyl. Strobilurins in addition to exerting a direct positive physiological effect on common bean plants also protect bean plants against BCMV infection in screen house and field conditions. Thus, it is proposed that these reduced‐risk pesticides are potential inducers against BCMV and growth enhancers and could be a beneficial component of integrated disease management of common bean.  相似文献

Four field trials were done with narrow-leafed lupins (Lupinus angustifolius) in 1988 - 1989, to examine the effect of sowing seed with 5% and 0.5% cucumber mosaic virus (CMV) infection on subsequent virus spread, grain yield and percentage of infection in harvested seed. A proportion of the CM V-infected seed failed to produce established plants and thus, plots sown with 5% and 0.5% infected seed contained 1.5-2.9% and 0.2-0.3% of seed-infected plants respectively. The rate of virus spread by aphids was faster and resulted in more extensive infection at maturity in plots sown with 5% infected seed than with 0.5% infected seed. In three trials, sowing 5% infected seed resulted in yield losses of 34 - 53% and CMV infection in the seed harvested of 6 - 13%. The spread of CMV infection resulting from sowing 0.5% infected seed did not significantly decrease yield. However, late CMV spread in these plots caused > 1% seed infection. In the fourth trial, which was badly affected by drought, CMV spread only slowly, there was no significant effect of CMV on grain yield and the percentage of infected seed harvested was 3–5 times less than that in the seed sown. When CMV-infected seed was sown at different depths, target depths of 8 and 11 cm decreased the incidence of seed-infected plants by c. 15% and c. 50% respectively compared with sowing at 5 cm. However, in glasshouse tests, treatment with the pre-emergence herbicide simazine failed to selectively cull out seed-infected plants. The field trials were colonised by green peach (Myzus persicae), blue-green (Acyrthosiphon kondoi) and cowpea (Aphis craccivora) aphids. When the abilities of these aphid species and of the turnip aphid (Lipaphis erysimi) in transmitting CMV from lupins to lupins were examined in glasshouse tests, short acquisition access times favoured transmission. With 5–10 min acquisition access times, overall transmission efficiencies were 10.8%, 9.4%, 6.1% and 3.9% for the green peach, cowpea, blue-green and turnip aphids respectively.  相似文献

Cucumber mosaic virus (CMV) was transmitted in the seed of infected Stellaria media plants. The rate of seed transmission varied both in manually infected plants (3–21%) and in plants grown from infected seed (21–40%). In naturally infected plants the rates of transmission found were 4–29%. Seeds recovered from field soil carried 4–5% infection and in infected seed placed in the soil the virus persisted for at least 5 months. Seed transmission of CMV also occurred in infected Lamium purpureum (4%), Cerastium holosteoides (2%) and Spergula arvensis (2%) but it could not be demonstrated in six other more common weed species in five botanical families. Seed transmission in Stellaria media occurred with a British (W) and an American (Y) strain of CMV. The virus was shown to occur in S. media pollen. The importance of CMV-infected S. media seed in the soil in relation to the epidemiology of the virus is discussed.  相似文献

The multifunctional protein translationally controlled tumour protein (TCTP) was previously identified as necessary for infection by the potyvirus pepper yellow mosaic virus. Using turnip mosaic virus (TuMV) as a model to study potyvirus biology, we confirmed that TCTP has a positive effect on virus infection. Living cell confocal microscopy demonstrated that TCTP colocalises with 6K2-tagged replication vesicles and with a perinuclear globular structure typically observed during potyvirus infection. Also, TCTP silenced protoplasts showed reduced virus accumulation, quantified by qRT-PCR, which suggests an effect on virus replication, translation or other intracellular process. Finally, TCTP silencing in plants reduced the accumulation of two species belonging to Orthotospovirus and a Begomovirus genus, which are not closely related to potyviruses. The results suggest that TCTP is a general susceptibility factor to several unrelated viruses.  相似文献

Virus-like symptoms were common in British crops of rhubarb. All plants tested of the three main varieties, ‘Timperley Early’, ‘Prince Albert’ and ‘Victoria’, were virus-infected. Turnip mosaic virus and a severe isolate of arabis mosaic virus (AMV) were obtained from ‘Timperley Early’; and ‘Prince Albert’ contained turnip mosaic virus, cherry leaf roll virus (CLRV), a mild isolate of AMV and, infrequently, cucumber mosaic virus (CMV). The main commercial variety ‘Victoria’ contained turnip mosaic virus, CLRV, a mild isolate of AMV and, infrequently, strawberry latent ringspot virus (SLRV). All the viruses were identified serologically. The rhubarb isolates did not differ markedly from other isolates of these viruses in herbaceous host reactions, properties in vitro or particle size and shape. A rhubarb isolate of CLRV was distinguished serologically from a cherry isolate of the virus. Turnip mosaic virus, CLRV and SLRV, were transmitted with difficulty, but AMV isolates were readily transmitted by mechanical inoculation. Turnip mosaic virus was also transmitted to rhubarb by Myzus persicae and Aphis fabae. CLRV was transmitted in 6–8% of the seed of infected ‘Prince Albert’ and ‘Victoria’ rhubarb and in 72% of the seed of infected Chenopodium amaranticolor. Mild isolates of AMV were also transmitted in 10–24% of the seed of infected ‘Prince Albert’ and ‘Victoria’ plants.  相似文献

Soybean crops showing systemic mottling, mosaic and leaf deformation were observed at high disease incidences (25.1–71.0%) in the kharif season of 2011 and 2012 in the experimental farm of the Indian Agricultural Research Institute (IARI), New Delhi. Symptomatic soybean leaves contained flexuous particles (650 × 12 nm), suggesting an infection by a Carlavirus. The causal virus was characterized as a strain of Cowpea mild mottle virus (CPMMV) on the basis of mechanical inoculation, whitefly transmission, seed transmission and sequencing of the viral genome. This is the first report of natural infection by a distinct strain of CPMMV in soybean in India.  相似文献

A severe foliar yellow mosaic disease was observed in horse chestnut trees (Aesculus carnea and A. hippocastanum). Reactions in woody indicator plants grafted with diseased horse chestnut suggested the presence of an ilarvirus. Virus isolates obtained by mechanical inoculation of herbaceous test plants reacted with antisera to apple mosaic virus but not with antisera to its serotype prunus necrotic ringspot virus, or to prune dwarf virus. Yellow mosaic was induced in horse chestnut seedlings grafted with tissues from herbaceous hosts infected with horse chestnut isolates or with the European plum line pattern isolate of apple mosaic virus. Virus was detected by enzyme-linked immunosorbent assay (ELISA) in embryo and endosperm of immature seed from infected trees but not in mature seed, or progeny seedlings. Strawberry latent ringspot virus was detected in one of six A. hippocastanum trees with a leaf vein yellows disease.  相似文献

Aims: To develop a highly sensitive and rapid protocol for simultaneous detection and differentiation of Tobacco mosaic virus (TMV) and Tomato mosaic virus (ToMV) in pepper and tomato. In this study, we use the multiplex PCR technique to detect dual infection of these two viruses. Methods and Results: A multiplex RT–PCR method consisting of one‐tube reaction with two primer pairs targeted to replicase genes was developed to simultaneously detect TMV and ToMV in seed samples of pepper and tomato. Specific primers were designed from conserved regions of each of the virus genomes, and their specificity was confirmed by sequencing PCR products. RT–PCR detected up to 10?6 dilution of total RNA extracted from infected leaves. Multiplex RT–PCR revealed the presence of both TMV and ToMV in three of 18 seed samples of tomato and one of 18 seed samples of pepper. Conclusions: The multiplex PCR assay was a cost effective, quick diagnostic technique, which was helpful in differentiating TMV and ToMV accurately. Significance and Impact of the Study: The multiplex PCR assay described in this study is a valuable tool for plant pathology and basic research studies. This method may facilitate better recognition and distinction of TMV and ToMV in both pepper and tomato.  相似文献

Two thousand sweet-cherry trees (Prunus avium) in English orchards were tested for virus infection by using Lambert and Mazzard F 12/1 as indicators. Most trees of varieties commonly grown before 1920 were infected with more than one virus, usually little cherry (69%) and necrotic ringspot/prune dwarf (56%). Other infection was less prevalent, 35% of these trees having European rusty mottle, 30% ring mottle and 3% necrotic rusty mottle. Most trees of varieties introduced since 1920 were virus-free (61%) but some had become infected with each of these viruses except necrotic rusty mottle. In a field trial of 12 years duration the yield of three varieties was diminished by infection with necrotic ringspot/prune dwarf, rugose mosaic, rusty mottle, ring mottle and necrotic line pattern. The effect of rusty mottle was due to growth suppression resulting in smaller trees, but that of other viruses was also due to impaired fertility. The yield of one variety (Merton Heart) was greatly enhanced by infection with rugose mosaic, rusty mottle and necrotic ringspot/prune dwarf viruses. The high incidence of virus infection and consequent yield depression has probably diminished the potential yield by at least 30% and contributed to the decline in acreage of sweet cherries in England.  相似文献

Soybean cultivars (Glycine max(L.) Merr.) infected with soybean cyst nematode (SCN; Heterodera glycinesIchinohe) often show symptoms similar to K deficiency. The objectives of this experiment were to determine if SCN infection affected macronutrient concentrations in soybean seedling vegetative tissues, determine whether increased K fertility can overcome these possible effects, and to determine if these possible effects are localized at the site of infection or expressed systemically throughout the root system. Soybean plants were grown with root systems split into two halves. This allowed differential K (0.2, 2.4 and 6.0 mM K nutrient solutions) and SCN (0 and 15 000 eggs/plant) treatments to be applied to opposite root-halves of the same plant. Thirty days after plants were inoculated with SCN, macronutrient concentrations of shoot and root tissues were determined. Potassium concentration in leaf blades was not affected; but K concentrations in leaf-petiole and stem tissues were increased with SCN infection. Roots infected with SCN contained lower K concentrations than uninfected roots, but only for the 2.4 mM K treatment. Thus, at the medium level of K fertility, SCN reduced K concentration in soybean roots, and increasing K fertility to the high level overcame the effect. Because K concentrations in the shoot tissues were not reduced by SCN infection, above ground portions of the plant may be able to overcome limitations that occur in roots during the first 30 days of infection. Increasing K fertility level in soybean fields may not benefit vegetative growth of soybean infected with SCN.  相似文献

Delaying the sowing of irrigated field beans in the Sudan after October greatly lowered the yield of seed, and increased both infestation by aphids and the incidence of Sudanese broad bean mosaic virus (SBBMV). Yield per plant was positively correlated with number of pods, but negatively with percentage infection with SBBMV. In greenhouse conditions, SBBMV was readily acquired from diseased plants and inoculated to healthy plants in 5 min feeding periods by both Aphis craccivora and Acyrthosiphon sesbaniae.  相似文献

Expression of double-stranded RNA (dsRNA) homologous to virus sequences can effectively interfere with RNA virus infection in plant cells by triggering RNA silencing. Here we applied this approach against a DNA virus, African cassava mosaic virus (ACMV), in its natural host cassava. Transgenic cassava plants were developed to express small interfering RNAs (siRNA) from a CaMV 35S promoter-controlled, intron-containing dsRNA cognate to the common region-containing bidirectional promoter of ACMV DNA-A. In two of three independent transgenic lines, accelerated plant recovery from ACMV-NOg infection was observed, which correlates with the presence of transgene-derived siRNAs 21–24 nt in length. Overall, cassava mosaic disease symptoms were dramatically attenuated in these two lines and less viral DNA accumulation was detected in their leaves than in those of wild-type plants. In a transient replication assay using leaf disks from the two transgenic lines, strongly reduced accumulation of viral single-stranded DNA was observed. Our study suggests that a natural RNA silencing mechanism targeting DNA viruses through production of virus-derived siRNAs is turned on earlier and more efficiently in transgenic plants expressing dsRNA cognate to the viral promoter and common region.  相似文献

Observations were made on the morphology, seed fertility, pollen viability and meiotic configurations of 22 different interspecific hybrids of 12 North American taxa representative of 3 sections of Ruellia. The hybrids were classified as (1) fully vigorous and fertile; (2) vigorous but with reduced fertility; (3) reduced vigor and fertility; (4) weak and sterile. Hybrids showed meiotic irregularities in microsporogenesis: univalents, chains, precocious separation of bivalents, nondisjunction, and micronuclei were commonly observed. Interspecific pollinations within sect. Dipteracanthus resulted in fertile or partially sterile F1 hybrids. Intersectional crosses produced partially or completely sterile hybrids or failed. Assuming that crossability is correlated with degree of evolutionary relationship, the parental species are interpreted in genecological terms to be grouped in polytypic ecospecies. Hybridization experiments doubtless would be of value in clarifying relationships in tropical and subtropical species of Ruellia.  相似文献

The effects of turnip mosaic virus infection on glucosinolatelevels and catabolism was studied in the mature seeds of rapidcycling Brassica campestris L. plants. Disease infection alteredthe total and soluble N concentrations in seeds. Infected plantssuffered growth reduction and increased concentration of severalseed glucosinolates compared to non-infected plants. The activityof partially purified seed myrosinase was not affected by maternalplant infection. Systemic viral infection did not induce qualitativeor quantitative alterations in the seed glucosinolate degradationprofile. Two major breakdown products, 3-butenylisothiocyanateand 1-cyano-3, 4-epithiobutane, were identified during autolysisand were, presumably, derived from 3-butenyl-GS, the most abundantseed glucosinolate. These results indicate that the glucosinolateconcentration and profile, but not glucosinolate catabolism,is affected in seeds produced on turnip mosaic virus-infectedrapid cycling plants. Four cultivars of cowpea (Vigna unguiculata [L]. Walp.) were infected with cowpea aphid-borne mosaic virus (CABMV) by natural infection in field plots. Seeds taken from these plants were tested for the presence of the virus by ELISA and symptom observation on the plantlets grown from the seeds. A biotin/ streptavidin ELISA technique was used and found to be more sensitive than a standard ELISA protocol for detecting CABMV infection in seed. There was a good correlation between the ELISA detection of CABMV in tissue taken from single cowpea seeds and subsequent development of infected plants grown from the same seeds. The ELISA technique is reliable for selecting CABMV-free stocks of cowpea seeds.  相似文献
