Extracellular production of a heat-stable somatic antigen (HSSA) by Bacillus thuringiensis subsp. dendrolimus strain T84A1-A [flagellar (H) serotype 4a: 4b] was serologically detected. In Ouchterlony tests, the HSSA antiserum gave single precipitin lines against both untreated and heat-treated culture supernatants. These two precipitin lines fused completely. When colonies of strain T84A1-A were grown on nutrient agar plates containing the homologous HSSA antiserum, precipitin halos were formed around the colonies. Of 27 type strains of B. thuringiensis subspecies tested, only the type strains of B. thuringiensis subsp. sotto (H serotype 4a: 4b) and B. thuringiensis subsp. israelensis (H serotype 14) formed [precipitin halos on nutrient agar plates containing antiserum against the HSSA of strain T84A1-A.  相似文献

Bacillus thuringiensis strains non-toxic to Lepidoptera, Bombyx mori and Diptera, Culex pipiens pallens larvae were isolated from Korean soil samples during an investigation of B. thuringiensis isolates highly toxic to insect pests. One of these isolates, NTB-88, produces parasporal inclusions about 138 kDa in size and is non-toxic to 19 insect species of three orders, Lepidoptera, Diptera and Coleoptera, even though it is highly susceptible to tryptic cleavage. Study of flagellar (H) antibodies of 33 B. thuringiensis strains revealed that NTB-88 has an H antigen identical with that of subsp. morrisoni (serotype 8a8b). Comparison of parasporal inclusion proteins and plasmid DNA patterns of strain NTB-88 with B. thuringiensis subsp. morrisoni HD-12 and B. thuringiensis subsp. morrisoni PG-14 showed that the isolate is a novel non-insecticidal B. thuringiensis strain belonging to serotype 8a8b.  相似文献

Bacillus cereus isolates, recovered from natural environments of Japan, were examined for their flagellar (H) antigenicities with the reference H antisera against Bacillus thuringiensis serotypes H1-H55. Of 236 B. cereus isolates tested, 165 (70%) were agglutinated with the reference antisera available. The frequencies of seropositive isolates were: 77% in soils, 68% on phylloplanes, and 60% in animal fecal populations. Among the 45 H serogroups detected, the serovar shandongiensis (H22) was the predominant, followed by the serovars entomocidus (H6), indiana (H16), pakistani (H13), and neoleonensis (H24ab). These five H serovars were commonly distributed in the three populations from different sources.  相似文献

Abstract Flagellar antigen of Bacillus cereus H.1 was purified and tested for serodiagnostic antigen by ELISA. The antibody against the flagellar antigen of B. cereus H.1 reacted not only with the homologous specific antigen but also reacted with the flagellar antigens of 23 strains of B. cereus . This common flagellar antigen of B. cereus was found to be due to 61-kDa protein by SDS-PAGE and immunoblot assay. Monoclonal antibody H15A5 against common antigenic epitope of B. cereus also reacted with flagellar antigens of 21 strains of Bacillus thuringiensis by ELISA. This monoclonal antibody reacted with the 61-kDa protein of the flagella of B. cereus H.1 and H.2 and B. thuringiensis Kurstaki HD1, Alesti and Aizawai juroi by immunoblot analysis. These results indicated that the common antigenic epitope of the 61-kDa protein existed in the flagella both of B. cereus and B. thuringiensis .  相似文献

Abstract An isolate of Bacillus thuringiensis subsp. israelensis (BTI) was obtained from field-collected larvae of the saturniid moth, Hylesia metabus . This isolate was shown to belong to serotype H 14, and to produce a spherical parasporal body identical to that of the type strain of BTI. With an LC50 of 0.764 μg cm−2, this isolate was more toxic to H. metabus than the HD-1 strain of B. thuringiensis subsp. kurstaki (HD-1). These results demonstrate that BTI can be active against lepidopterous insects, a wider spectrum of activity than previously realized.  相似文献

Abstract The strain 273 B, the type strain of a H serotype of Bacillus thuringiensis not yet characterized: B. thuringiensis subsp. cameroun , serotype H32, was isolated from soil samples collected in Cameroon. This strain produces cuboidal parasporal bodies composed of two major proteins of 53 kDa and 35 kDa. N-terminal sequences of the major proteins share no homology with published sequences. Only the 35 kDa protein is susceptible to digestion by trypsin. A complex array of 9 plasmids was revealed.  相似文献

Two new Bacillus thuringiensis strains have been classified by the H antigen of the cells and differentiated by their morphological, biochemical and molecular characteristics. The flagellar agglutination showed that both strains bore specific H antigens which allowed their classification as the new serotypes H57 and H59. The serovar names proposed for the type strains characterized in this work are B. thuringiensis ser. pirenaica, for the H serotype 57, and B. thuringiensis ser. iberica, for the H serotype 59. Further characterization of these strains, by means of SDS-PAGE, Western inmunodetection, plasmid profile and cry -gene identification by polymerase chain reaction, confirmed the originality of the two novel serotypes. Toxicity tests carried out against several insect species, belonging to the orders Lepidoptera, Diptera and Coleoptera, showed no detectable insecticidal activity for either of the B. thuringiensis strains.  相似文献

An improved high-performance liquid chromatography separation was developed to detect and quantify beta-exotoxin production in Bacillus thuringiensis culture supernatants. Exotoxin production was assigned to a plasmid in five strains, from three subspecies (B. thuringiensis subsp. thuringiensis serotype 1, B. thuringiensis subsp. tolworthi serotype 9, and B. thuringiensis subsp. darmstadiensis serotype 10). A new exotoxin, called type II beta-exotoxin in this report, was discovered in B. thuringiensis subsp. morrisoni serotype 8ab, purified, and partially characterized. This material is more specific than type I beta-exotoxin and is very active against the Colorado potato beetle, Leptinotarsa decemlineata.  相似文献

During a survey conducted in Colombia a new isolate of Bacillus thuringiensis that showed toxicity toward Culex quinquefasciatus, Cx. pipiens, Aedes aegypti, and Anopheles stephensi larvae was isolated. Parasporal crystals were spherical in shape and showed a great degree of similarity with those produced by the reference strain of Bacillus thuringiensis subsp. israelensis. Supernatant fraction of the whole culture was not toxic, and heat-stable exotoxin production was negative. Catalase, urease, arginine dihydrolase, amylase, lecithinase, acetyl-methyl-carbinol, and gelatinase production were positive. Hemolysis on sheep blood agar was alpha-type. The isolate 163-131 showed natural resistance to azolocillin and was sensible to cephoperazone, cephalotin, nalidixic acid, and trimetoprin sulfametoxazole. Flagellar agglutination showed a specific H 30 antigen which allows individualization of this strain as a new serotype and the subspecies name of medellin is proposed.  相似文献
