Changes in the protein secondary structure and electron transport activity of the Triton X-100-treated photosystem I (PSI) and photosystem II (PSII) complexes after strong illumination treatment were studied using Fourier transform-infrared (FT-IR) spectroscopy and an oxygen electrode. Short periods of photoinhibitory treatment led to obvious decreases in the rates of PSI-mediated electron transport activity and PSII-mediated oxygen evolution in the native or Triton-treated PSI and PSII complexes. In the native PSI and PSII complexes, the protein secondary structures had little changes after the photoinhibitory treatment. However, in both Triton-treated PSI and PSII complexes, short photoinhibition times caused significant loss of -helical content and increase of -sheet structure, similar to the conformational changes in samples of Triton-treated PSI and PSII complexes after long periods of dark incubation. Our results demonstrate that strong-light treatment to the Triton-treated PSI and PSII complexes accelerates destruction of the transmembrane structure of proteins in the two photosynthetic membranes.  相似文献

Tradescantia albiflora (Kunth) was grown under two different light quality regimes of comparable light quantity: in red + far-red light absorbed mainly by photosystem I (PSI light) and yellow light absorbed mainly by photosystem II (PSII light). The composition, function and ultrastructure of chloroplasts, and photoinhibition of photosynthesis in the two types of leaves were compared. In contrast to regulation by light quantity (Chow et al. 1991. Physiol. Plant. 81: 175–182), light quality exerted an effect on the composition of pigment complexes, function and structure of chloroplasts in Tradescantia: PSII light-grown leaves had higher Chl a/b ratios, higher PSI concentrations, lower PSII/PSI reaction centre ratios and less extensive thylakoid stacking than PSI light-grown leaves. Light quality triggered modulations of chloroplast components, leading to a variation of photosynthetic characteristics. A larger proportion of primary quinone acceptor (QA) in PSI light-grown leaves was chemically reduced at any given irradiance. It was also observed that the quantum yield of PSII photochemistry was lower in PSI light-grown leaves. PSI light-grown leaves were more sensitive to photoinihibition and recovery was slower compared to PSII light-grown leaves, showing that the PSII reaction centre in PSI light-grown leaves was more easily impaired by photoinhibition. The increase in susceptibility of leaves to photoinhibition following blockage of chloroplast-encoded protein synthesis was greater in PSII light-grown leaves, showing that these leaves normally have a greater capacity for PSII repair. Inhibition of zeaxanthin formation by dithiothreitol slightly increased sensitivity to photoinhibition in both PSI and PSII light-grown leaves.  相似文献

The light environment during plant growth determines the structural and functional properties of higher plant chloroplasts, thus revealing a dynamically regulated developmental system. Pisum sativum plants growing under intermittent illumination showed chloroplasts with fully functional photosystem (PS) II and PSI reaction centers that lacked the peripheral chlorophyll (Chi) a/b and Chl a light-harvesting complexes (LHC), respectively. The results suggest a light flux differential threshold regulation in the biosynthesis of the photosystem core and peripheral antenna complexes. Sun-adapted species and plants growing under far-red-depleted illumination showed grana stacks composed of few (3–5) thylakoids connected with long intergrana (stroma) thylakoids. They had a PSII/PSI reaction center ratio in the range 1.3–1.9. Shade-adapted species and plants growing under far-red-enrichcd illumination showed large grana stacks composed of several thylakoids, often extending across the entire chloroplast body, and short intergrana stroma thylakoids. They had a higher PSII/PSI reaction center ratio, in the range of 2.2–4.0. Thus, the relative extent of grana and stroma thylakoid formation corresponds with the relative amounts of PSII and PSI in the chloroplast, respectively. The structural and functional adaptation of the photosynthetic membrane system in response to the quality of illumination involves mainly a control on the rate of PSII and PSI complex biosynthesis.  相似文献

Photosynthetic activities and the redox states of photosystem I (PSI) and photosystem II (PSII) in intact leaves of cucumber plants (Cucumis sativus L.), as well as the sucrose and starch contents were examined under conditions of ongoing soil water deficit imposed by the cessation of watering. As the soil drought progressed, the maximum rate of photosynthetic CO2 fixation was shown to decrease. These changes in the maximum photosynthetic rate occurred synchronously with changes in the maximum quantum yield of photosynthesis. Under soil water deficit, the reduced form of PSII primary acceptor Q A was accumulated only at photon flux densities of about 100 mol/(m2 s). At such photon flux densities, the changes in nonphotochemical quenching (qN) induced by soil water deficit were opposite to changes in photochemical quenching parameter (1 – qP). Irrespective of the duration of soil drought, the relationship between steady-state concentrations of photochemically inactive reaction centers of PSI and PSII (the fractions of P700 and Q A in the oxidized and reduced state, respectively) was almost linear, which provides evidence for the concerted operation of both photosystems. The conditions of soil water deficit promoted sucrose accumulation in the source leaf, which was paralleled by a substantial decrease in the amount of starch in the same leaf. The highest content of sucrose in the leaf after a 7-day drought was correlated with the largest decrease in photosynthetic activity. It is concluded that the progressive drought triggers an endogenous mechanism that regulates photosynthesis through feedback relations, namely, the inhibition of photosynthesis by its end products.  相似文献

为改善玉米群体内光环境,进一步提高玉米单株光合能力以获得高产,本研究以郑单958为试验材料,通过设置种子定向入土方式,研究了定向有序种植条件下群体内光分布特征,以及单株玉米穗位叶花后光合性能,并借助快速叶绿素荧光动力学曲线分析了与叶片光合性能有密切联系的光系统Ⅱ(PSⅡ)的性能特征.结果表明:叶片不同朝向显著改变夏玉米群体内穗位叶处光合有效辐射截获量,朝南处理(S)平均比朝北处理(N)高271.8%.不同朝向的叶片对高光与弱光的利用能力差异显著,朝南处理饱和光强下净光合速率(Pn)显著升高,表明其高光强利用能力显著提升;而朝北处理(N)表观量子效率(α)则随生育期推进显著增加,有利于叶片适应长期弱光环境.生育前期朝南处理PSⅡ电子供体侧和受体侧性能显著提高,进而改善了PSⅡ反应中心性能(PIABS)和荧光光化学淬灭系数(ψo),电子在电子传递链中转移效率(φEo)的提高增强了电子由PSⅡ向光系统Ⅰ(PSⅠ)的传递性能.生育前期叶片性能呈现出朝南>朝东>朝西>朝北的趋势.但成熟末期朝南处理对强光的利用效率显著降低,朝北处理在生育后期表现出较强的弱光利用能力,表观量子效率显著升高,花后40dPn与PSⅡ性能均表现为朝北>朝西>朝东>朝南的趋势.总体上,朝南与朝东处理群体内光环境改善显著,群体穗位层截获光合有效辐射较多,光合能力和干物质生产能力增强,有利于夏玉米产量提高.  相似文献

Plants may experience environmental stress factors operating in nature either simultaneously or in sequence. In the study, we have acclimated the developing primary leaves of wheat seedlings to high light stress and examined their photosynthetic response to polyethylene glycol (PEG) mediated osmotic stress during different developmental phases including senescence. The high light acclimated leaves show higher level of total carotenoids as compared to their non-acclimated counterparts experiencing osmotic stress during senescence. They also exhibit greater membrane stability as indicated by the measurements of fluorescence polarisation and energy transfer efficiency in photosystem I (PSI) and Photosystem II (PSII). From the data of DCPIP photoreduction and pulse amplitude modulated (PAM) fluorimetry, a similar trend is observed for PSII photochemistry of the leaves experiencing osmotic stress during senescence. Our results may suggest that the stress adaptive potential induced by one stress during development is retained by the leaves and helps to mitigate another stress effect operating in sequence during another developmental phase, namely senescence.  相似文献

Plant tolerance to high light and oxidative stress is increased by overexpression of the photosynthetic enzyme Ferredoxin:NADP(H) reductase (FNR), but the specific mechanism of FNR-mediated protection remains enigmatic. It has also been reported that the localization of this enzyme within the chloroplast is related to its role in stress tolerance. Here, we dissected the impact of FNR content and location on photoinactivation of photosystem I (PSI) and photosystem II (PSII) during high light stress of Arabidopsis (Arabidopsis thaliana). The reaction center of PSII is efficiently turned over during light stress, while damage to PSI takes much longer to repair. Our results indicate a PSI sepcific effect, where efficient oxidation of the PSI primary donor (P700) upon transition from darkness to light, depends on FNR recruitment to the thylakoid membrane tether proteins: thylakoid rhodanase-like protein (TROL) and translocon at the inner envelope of chloroplasts 62 (Tic62). When these interactions were disrupted, PSI photoinactivation occurred. In contrast, there was a moderate delay in the onset of PSII damage. Based on measurements of ΔpH formation and cyclic electron flow, we propose that FNR location influences the speed at which photosynthetic control is induced, resulting in specific impact on PSI damage. Membrane tethering of FNR therefore plays a role in alleviating high light stress, by regulating electron distribution during short-term responses to light.

Phycobilisomes (PBS) are the major accessory light-harvesting complexes in cyanobacteria and their mobility affects the light energy distribution between the two photosystems. We investigated the effect of PBS mobility on state transitions, photosynthetic and respiratory electron transport, and various fluorescence parameters in Synechocystis sp. strain PCC 6803, using glycinebetaine to immobilize and couple PBS to photosystem II (PSII) or photosystem I (PSI) by applying under far-red or green light, respectively. The immobilization of PBS at PSII inhibited the increase in cyclic electron flow, photochemical and non-photochemical quenching, and decrease in respiration that occurred during the movement of PBS from PSII to PSI. In contrast, the immobilization of PBS at PSI inhibited the increase in respiration and photochemical quenching and decrease in cyclic electron flow and non-photochemical quenching that occurred when PBS moved from PSI to PSII. Linear electron transport did not change during PBS movement but increased or decreased significantly during longer illumination with far-red or green light, respectively. This implies that PBS movement is completed in a short time but it takes longer for the overall photosynthetic reactions to be tuned to a new state.  相似文献

Effect of UV-B rays (280–320 nm) on photosynthetic electron transport and production of phenolic compounds in tea (Camellia sinensis L.) callus culture grown in white light was investigated. When white light was supplemented with UV radiation, the culture growth was retarded and morphological characteristics were modified. These conditions promoted the formation of chlorophyll-bearing cells and altered the ability of cultured cells to accumulate phenolic compounds, including flavans specific to Camellia sinensis. By the end of the culturing cycle (on the 45th day), the total content of phenolic compounds in the culture grown under supplementary UV irradiation was almost 1.5 times higher than in the control culture. The UV rays greatly stimulated photosystem II (PSII) activity in phototrophic cells of the callus culture, which was indicated by a large increase in the ratio of variable chlorophyll fluorescence to maximal fluorescence. This ratio was as low as 0.19 in cells cultured in white light and increased to 0.53 in the cell culture grown under white and UV light. The kinetics of dark relaxation of chlorophyll variable fluorescence, related to reoxidation of PSII primary acceptor, contained either two or three components, depending on the absence or presence of UV radiation, respectively. An artificial electron acceptor of PSI, methyl viologen modified the kinetics of dark decay of chlorophyll variable fluorescence in a characteristic manner, implying that photosynthetic electron transport was mediated by PSI and PSII in both treatments (culturing in white light with and without UV-B). It is concluded that stimulatory effect of UV rays on the parameters examined in phototrophic regions of Camellia tissue culture is determined by photoexcitation of a regulatory pigment that absorbs quanta in blue and long-wave UV spectral regions.  相似文献

The source-sink ratio of 1-year-old, potted sour cherry trees ( Prunus cerasus L.) was altered by whold-plant partial defoliation or continuous illumination to determine if trees were primarily sink limited and to elucidate the means whereby photosynthetic enhancement or inhibition occurs. Leaf xylem water potential was not affected by either treatment. Although stomatal conductance was reduced by 1 to 3 days of continuous illumination, internal CO2 concentration was not significantly affected indicating that the enhanced physical limitation imposed by the stomata was of no physiological significance. Net CO2 assimilation (A) was significantly higher 4 days after partial defoliation and lower from 1 to 4 days following continuious illumination. The increase in A in partially defoliated plants was associated with reduced leaf starch and increased surose and sorbitol concentrations. The decrease in A in continuously illuminated plants was associated with a decrease in variable fluorescence, photochemical efficiency of photosystem II (PSII) and an increase in instantaneous fluorescence, indicating that leaves were photoinhibited and that irreversible damage had occurred to PSII. In addition, leaves of continuously illuminated plants had 80% more starch and significantly less sucrose and sorbitol. These altered leaf carbohydrate concentrations indicate that the existing sink limitation may have been aggravated by continuous illumination leading to an insufficient utilization of sucrose from the leaf. Whether the altered photochemical and biochemical events occurred simultaneously and/or to the same degree to lead to the observed responses remains equivocal.  相似文献

The photosynthetic acclimation of Tradescantia albiflora (Kunth), a trailing ground species naturally occurring in the deep shade of rainforests, was studied in relation to growth irradiance (glasshouse; direct light and 1 to 4 layers of shade cloth, giving 100 to 1.4% relative growth irradiance). Contrary to other irradiance studies of higher plants grown in natural habitats or controlled light environments, the chlorophyll a/b ratios of Tradescantia leaves were low (∼2.2) and constant. Acclimation to growth irradiance caused no changes in the relative amounts of specific Chl-proteins or the numbers of photosystem I (PSI) and PSII reaction centres on a chlorophyll basis, indicating that the light-harvesting antenna sizes of PSII and PSI, as well as the photosystem stoichiometry, were independent of growth irradiance. However, the amount of cytochrome f and ATP synthase on a chlorophyll basis increased with increasing the relative growth irradiance from 1.4 to 35%, showing acclimation of electron transport and photophosphorylation capacity. The photosynthetic capacity and ribulose 1, 5-bisphosphate carboxylase (EC 4.1.1.39) activity also increased with increase of the growth irradiance to 35%. Beyond that, the inflexible PSII/PSI stoichiometry and shade-type photosystem II/light-harvesting units in Tradescaniia are a disadvantage for long-term exposure to high irradiance since the leaves are more prone to photoinhibition.  相似文献

通过同时测定玉米叶片的叶绿素荧光快速诱导动力学曲线和对820 nm光的吸收、分析叶片的气体交换过程以及叶绿体活性氧清除关键酶的活性,研究了不同浓度的铅(Pb)胁迫对玉米光系统Ⅰ(PSⅠ)、光系统Ⅱ(PSⅡ)的光化学活性和光合作用的影响,并分析了Pb胁迫下两个光系统的相互关系.结果表明:铅胁迫显著抑制了玉米地上部分和地下部分的生长、降低了叶片光合色素含量、并通过非气孔因素限制了光合作用、导致过剩激发能的增加;铅胁迫显著抑制了超氧化物歧化酶(SOD)和抗坏血酸过氧化物酶(APX)的活性、伤害了PSII反应中心、PSII的受体侧和供体侧(放氧复合体)以及PSI光化学活性.  相似文献

Proton gradient regulation 5‐like photosynthetic phenotype 1 (PGRL1)‐dependent cyclic electron transport around photosystem I (PSI) plays important roles in the response to different stresses, including high light. Although the function of PGRL1 in higher plants and green algae has been thoroughly investigated, little information is available on the molecular mechanism of PGRL1 in diatoms. We created PGRL1 overexpression and knockdown transformants of Phaeodactylum tricornutum, the diatom model species, and investigated the impact on growth and photosynthesis under constant and fluctuating light conditions. PGRL1 over‐accumulation resulted in significant decreases in growth rate and apparent photosystem II (PSII) activity and led to an opposing change of apparent PSII activity when turning to high light, demonstrating a similar influence on photosynthesis as a PSII inhibitor. Our results suggested that PGRL1 overexpression can reduce the apparent efficiency of PSII and inhibit growth in P. tricornutum. These findings provide physiological evidence that the accumulation of PGRL1 mainly functions around PSII instead of PSI.  相似文献

Light induced photosystem (PS)II photoinhibition inactivates and irreversibly damages the reaction center protein(s) but the light harvesting complexes continue the collection of light energy. Here we addressed the consequences of such a situation on thylakoid light harvesting and electron transfer reactions. For this purpose, Arabidopsis thaliana leaves were subjected to investigation of the function and regulation of the photosynthetic machinery after a distinct portion of PSII centers had experienced photoinhibition in the presence and absence of Lincomycin (Lin), a commonly used agent to block the repair of damaged PSII centers. In the absence of Lin, photoinhibition increased the relative excitation of PSII and decreased NPQ, together enhancing the electron transfer from still functional PSII centers to PSI. In contrast, in the presence of Lin, PSII photoinhibition increased the relative excitation of PSI and led to strong oxidation of the electron transfer chain. We hypothesize that plants are able to minimize the detrimental effects of high-light illumination on PSII by modulating the energy and electron transfer, but lose such a capability if the repair cycle is arrested. It is further hypothesized that dynamic regulation of the LHCII system has a pivotal role in the control of excitation energy transfer upon PSII damage and repair cycle to maintain the photosynthesis safe and efficient.  相似文献

Cyanobacteria can rapidly regulate the relative activity of their photosynthetic complexes photosystem I and II (PSI and PSII) in response to changes in the illumination conditions. This process is known as state transitions. If PSI is preferentially excited, they go to state I whereas state II is induced either after preferential excitation of PSII or after dark adaptation. Different underlying mechanisms have been proposed in literature, in particular i) reversible shuttling of the external antenna complexes, the phycobilisomes, between PSI and PSII, ii) reversible spillover of excitation energy from PSII to PSI, iii) a combination of both and, iv) increased excited-state quenching of the PSII core in state II. Here we investigated wild-type and mutant strains of Synechococcus sp. PCC 7942 and Synechocystis sp. PCC 6803 using time-resolved fluorescence spectroscopy at room temperature. Our observations support model iv, meaning that increased excited-state quenching of the PSII core occurs in state II thereby balancing the photochemistry of photosystems I and II.  相似文献

Abstract. The physiological characteristics and photo-system composition of the photosynthetic apparatus of Silene dioica , a woodland plant, grown in sun and natural shade are examined. As expected, shade leaves exhibited lower chlorophyll a/b ratios, light saturated rates of CO2 assimilation (Asat), dark respiration (Rd,) and light compensation points ( Г ), with both sun and shade leaves having similar absorptances and quantum yields of CO2 assimilation (φ). Shade leaves were able to utilize far-red light for electron transport and carbon assimilation and reach the compensation point. Sun leaves in far-red light had a rate of carbon assimilation equivalent to their dark respiration rate. Chlorophyll fluorescence kinetics from leaves at 77 K together with analyses of thylakoid contents of photosystems (PS) I and II and the light-harvesting cholorphyll a/b protein complex associated with PSII (LHCII) demonstrated that the antenna size of PSII was similar in thylakoids of sun and shade leaves, but shade leaves contained ca. 20% more PSII and ca. 12% less PSI complexes. The increased PSII/PSI ratio in shade leaves accounted for their ability to achieve the compensation point in far-red light. An important feature of photosynethic shade adaptation in S. dioica is an increase in the PSII/PSI ratio and not an increase in the antenna size of PSII. The adaptive response of sun leaves when placed in a shade environment was rapid and had a half-time of ca. 18h.  相似文献

Oxygenic phototrophs are vulnerable to damage by reactive oxygen species (ROS) that are produced in photosystem I (PSI) by excess photon energy over the demand of photosynthetic CO2 assimilation. In plant leaves, repetitive short-pulse (rSP) illumination produces ROS to inactivate PSI. The production of ROS is alleviated by oxidation of the reaction center chlorophyll in PSI, P700, during the illumination with the short-pulse light, which is supported by flavodiiron protein (FLV). In this study, we found that in the cyanobacterium Synechocystis sp. PCC 6803 P700 was oxidized and PSI was not inactivated during rSP illumination even in the absence of FLV. Conversely, the mutant deficient in respiratory terminal oxidases was impaired in P700 oxidation during the illumination with the short-pulse light to suffer from photo-oxidative damage in PSI. Interestingly, the other cyanobacterium Synechococcus sp. PCC 7002 could not oxidize P700 without FLV during rSP illumination. These data indicate that respiratory terminal oxidases are critical to protect PSI from ROS damage during rSP illumination in Synechocystis sp. PCC 6803 but not Synechococcus sp. PCC 7002.  相似文献
