Powdery mildew infection of barley seedlings was significantly reduced using either pre-inoculation or post-inoculation sprays of the film-forming polymers Nu-Film P, Emerald or Vapor Gard. The greatest reduction in mildew infection was obtained with a pre-inoculation spray of the polymer Vapor Gard. Although the addition of 1 mm α-difluoromethylornithine (DFMO) to a 2% solution of Emerald or Vapor Gard resulted in better control of powdery mildew infection than that obtained using the polymer or DFMO alone, the reductions in infection were not significant. Reduction in mildew infection was greatest when the Vapor Gard + DFMO mixture was applied 1 day prior to inoculation with the fungus. Although substantial reduction in mildew infection was achieved using as little as 0.1 mm DFMO with 2% Vapor Gard, or reducing the polymer concentration to 1% in combination with 1 mm DFMO, best control was achieved using 2% Vapor Gard plus 1 mm DFMO.  相似文献

Pea breeding for rust resistance is hampered by the little resistance available in pea. In an attempt to validate alternative control methods, we evaluated the potential of systemic acquired resistance for rust control in pea by biotic and abiotic inducers. Challenge with a virulent or with an avirulent rust isolate prior to pea rust inoculation did not induce resistance either locally or systemically. Exogenous application of salicylic acid in the range 5–10 m m prior to rust inoculation did not protect against rust locally, but reduced rust infection systemically in first upper leaf node although not in the upper ones. Some phytotoxicity was observed at 10 m m . Exogenous application of benzothiadiazole in the range 1–10 m m provided locally a 30–40% reduction in infection frequency. At least 5 m m was needed to reduce rust infection systemically in first upper leaf, and 10 m m in upper ones. Exogenous application of dl -3-amino- n -butyric acid (BABA) provided locally a 45–58% reduction in infection frequency, while systemically a 33–58 and 49–58% reduction of rust symptoms was achieved on leaves at second and third nodes respectively. BABA application was not associated with symptoms of phytotoxicity.  相似文献

The study reported here primarily focuses on whether fruit extracts of Azadirachta indica Juss. (Neem) can induce systemic acquired resistance (SAR) in Hordeum vulgare against Drechslera graminea. A single leaf from each of the 1-month-old seedlings grown in 50 pots was treated with neem extract. Seven samples were collected at 12-h intervals for estimation of salicylic acid (SA) content and activities of phenylalanine ammonia lyase (PAL) and peroxidase (PO). Disease incidence was recorded on uninoculated leaves after 2 weeks and on newly emerged leaves after 3 weeks of inoculation of spores of the pathogen. Treatment of single leaf of barley seedlings with aqueous fruit extract of neem could protect the untreated and later emerging leaves of these seedlings from infection by leaf stripe pathogen. The concentration of SA and activities of PAL and PO were significantly higher in untreated leaves of seedlings given a single leaf treatment with neem fruit extract. The results show that neem fruit extract induced SAR in barley seedlings against D. graminea. The results of the study are significant for developing an environment-friendly biocide, which could induce SAR in crop plants leading to efficient management of pathogens  相似文献

Lauric Acid Exhibits Antifungal Activity Against Plant Pathogenic Fungi   总被引:3,自引:0,他引:3  
This study aimed at examining the effects of the saturated fatty acid lauric acid on mycelial growth of Rhizoctonia solani and Pythium ultimum and on infection of barley seedlings with Blumeria graminis f. sp. hordei. Mycelial growth of R. solani and P. ultimum in agar culture was significantly reduced by lauric acid at concentrations of 100 μm and above, while no fungal growth occurred in liquid culture at concentrations above 50 μm . Application of lauric acid at concentrations ranging from 250 to 1000 μm to barley leaves before or after inoculation with B. graminis f. sp. hordei led to significant reductions in infection. This study provides the first report of the activity of lauric acid against plant pathogenic fungi and indicates the need for investigation of its mechanism of action.  相似文献

Germination of Erysiphe graminis f.sp. hordei conidia on leaves of several barley cultivars was studied in the laboratory. On both detached leaves and intact plants, within 48 h of inoculation a higher proportion of conidia had germinated on the basal and middle portions of the adaxial leaf surface than on the corresponding portions of the abaxial surface. Such differences between surfaces were not observed near the leaf tip. Similar results were obtained with all the cultivars and growth stages tested, and with five isolates of E. graminis, and are consistent with the observation that there is usually less powdery mildew on the abaxial than the adaxial surface of barley leaves. With most of the barley genotype/mildew isolate combinations tested, within 48 h of inoculation higher proportions of conidia germinated on seedlings and juvenile plants than on older plants. Inherited characteristics which affect spore germination on the leaf surface may be important factors in the development of adult-plant resistance of barley to powdery mildew, particularly in certain genotypes.  相似文献

Drench inoculation of the undisturbed roots of barley seedlings with Fusarium oxysporum f. sp. radicis‐lycopersici (FORL) significantly reduced the primary infection frequency of Blumeria graminis f. sp. hordei (BGH) on the first leaves. The length of secondary hyphae and subsequent conidial production of BGH were also found to be significantly reduced by preinoculation with FORL. The reduction in infection frequency was observed as early as 48 h after inducer treatment, namely when plants were challenge‐inoculated immediately following inoculation with FORL. The induced resistance continued up to 16 days after treatment as indicated by the reduction in infection frequency, up to 22 days after treatment when evaluated as a reduction in the length of secondary hyphae, and up to 35 days after treatment when evaluated as a reduction in conidial production. Characteristics of FORL that may explain its success as an inducer of resistance against barley powdery mildew are discussed.  相似文献

A barley cDNA clone (PRb-1) corresponding to an mRNA differentially induced in resistant compared to susceptible barley cultivars by powdery mildew infection was isolated and characterised. The deduced amino acid sequence revealed 24 amino acids comprising the signal peptide and 140 amino acids of the mature peptide (15 kDa). This showed close homology to PR-1-like proteins, which have been isolated from maize, tobacco, tomato and Arabidopsis thaliana. Northern blot analysis showed accumulation of the corresponding mRNA 12 h after inoculation of resistant barley cultivars with Erysiphe graminis. Increased expression of the PRb-1 gene was also observed in resistant compared with near-isogenic susceptible barley plants following treatment with ethylene, salicylic acid, methyl jasmonate and 2,6-dichloro-isonicotinic acid.  相似文献

为阐明香蕉枯萎病发病机制,研究了尖孢镰刀菌侵染后,香蕉植株中几种对尖孢镰刀菌生长有显著作用的物质(氨基酸、有机羧酸、酚酸)种类和含量的变化。结果表明:(1)病原菌侵染后,伤害逐渐加剧,株高和生物量显著下降。(2)病原菌侵染后,叶片氨基酸总量显著升高,其中丝氨酸、缬氨酸、组氨酸、异亮氨酸和亮氨酸增幅较大,病原菌侵染16 d,其含量分别为侵染前的7.1、6.2、4.4、3.5和2.3倍;而根氨基酸总量开始显著降低,差异逐渐变小。(3)叶片有机羧酸酸含量在病原菌侵染后显著增加,而在根中显著降低。侵染植株叶片中草酸、柠檬酸、苹果酸、琥珀酸和延胡索酸含量分别是未侵染植株叶片的2.6、1.6、1.9、1.8和2.3倍;根中草酸、柠檬酸、苹果酸、琥珀酸和延胡索酸含量分别是未侵染植株的81%、42%、44%、28%和59%。(4)病原菌侵染后,植株叶片和根中酚酸含量都显著升高。叶片中阿魏酸、肉桂酸和水杨酸含量分别是未侵染叶片的2.9、1.7和2.9倍;而根中对羟基苯甲酸和丁香酸含量分别是未侵染根的4.3和1.5倍。研究结果表明,尖孢镰刀菌侵染后,植物与病原菌的相互作用使得植物体内抑菌物质和促菌物质都会相应的增加,植株对病害有一定的抗性,但促菌物质种类和含量较高最终使得感病植株发病。  相似文献

Six benzylic diamines were synthesised and examined for antifungal activity. Four of the compounds, KB 2, KB 4, KB 5 and KB 6, reduced radial growth of the oat leaf stripe pathogen Pyrenophora avenae, the largest reduction obtained with 25 μM KB 4, which reduced radial growth by 47%. Surprisingly, these four amines had no effect against infection of barley seedlings with the powdery mildew fungus Erysiphe graminis f.sp. hordei. Instead, two different amines, KB 1 and KB 3, reduced powdery mildew infection on barley. The greatest reduction was obtained with 25 μM KB 3, which reduced mildew infection by 69%. All of the amines which exhibited antifungal or fungicidal properties perturbed polyamine formation as measured by the incorporation of labelled ornithine into polyamines.  相似文献

In wheat leaves infected with yellow rust (Puccinia striiformis West.) and in barley leaves infected with powdery mildew (Erysiphe graminis DC.) the effect of respiratory inhibitorsin vivo and terminal oxidases activityin vitro were studied. In the experiments the first leaves of seedlings grown in glasshouse were used. The influence of infection was shown by activation of respiration and terminal oxidases (Fe- and Cu-oxidases), first of all cytochrome oxidase and peroxidase. There might be involved also the increased role of ascorbic acid oxidase and phenolase. Peroxidase activation was found to be much higher in susceptible varieties than in resistant ones. Neither in wheat nor in barley the catecholase activity was detected; on the contrary the enzymatic oxidation of floroglucine was found to be also in barley leaves, the intensity of which being dependend on resistance or susceptibility to powdery mildew. Thus, it is not excluded, that in contact establishing between obligate parasite and the host the significant role may be played by specific phenolase and phenolic substances. It suggests itself, that due to the participating in ATP-formation the cytochrome system in terminal oxidation of cereals, infected with obligate parasites, may have centrale position.  相似文献

Two spermidine analogues were synthesised and examined for antifungal activity. Both compounds used as 1 mM post-inoculation sprays reduced infection of barley seedlings by the powdery mildew fungus, Erysiphe graminis f.sp. hordei, infection of broad bean seedlings by the rust fungus, Uromyces viciae-fabae, and infection of apple seedlings by the powdery mildew fungus, Podosphaera leucotricha. Since these fungal pathogens cannot be cultured axenically, the effects of the two spermidine analogues on mycelial growth in vitro, as well as preliminary investigations on polyamine biosynthesis, were undertaken using the oat stripe pathogen, Pyrenophora avenae. Although neither compound affected radial growth of the fungus on plates, both analogues reduced fungal biomass in liquid culture substantially. The two spermidine analogues, used at a concentration of 1 mM, had no significant effect on the conversion of labelled ornithine into polyamines in P. avenae.  相似文献

Treatment of wheat leaves with heptanoyl salicylic acid (HS) and trehalose at concentrations of 0.1 and 15 g l(-1), prior to fungal inoculation, resulted in 40% and 60% protection, respectively, against powdery mildew. The total lipid composition of Blumeria graminis f.sp. tritici (Bgt) conidia, the causal agent of wheat powdery mildew, was compared when produced on wheat leaves, respectively, untreated and treated with the two elicitors, HS and trehalose. An obvious effect was observed on lipid composition (sterol and fatty acid (FA)) of Bgt conidia produced on wheat leaves treated with HS. A total of 16 FA (C12-C24 saturated and unsaturated) as well as unusual methoxylated Fatty Acids (mFA) (3-methoxydocosanoic and 3-methoxytetracosanoic acids) were detected in the conidia. Medium chain FA were predominant in HS treated conidia (64.65%) while long chain fatty acids constituted the major compounds in untreated conidia (62%). The long chain/medium chain FA ratio decreased from 1.8 in the conidia produced on untreated leaves to 0.5 in the conidia obtained from HS treated leaves. When comparing the sterol composition of Bgt conidia produced on leaves treated with HS versus conidia obtained from untreated ones, very important changes within the two major classes can be seen. In particular, 24-methylsterols, e.g., 24-methylenecholesterol and 24-methylcholesta-7,24-dien were reduced by about 82% whereas 24-ethylsterols, e.g., 24-ethylcholesterol and 24-ethylcholesta-5,22-dienol were increased by about 85%. The 24-methylsterols/24-ethylsterols ratio was reduced by ninefold in the conidia produced from HS treated leaves.  相似文献

Field experiments showed that the treatment of winter barley with microbial metabolites produced by a Bacillus subtilis strain led to increased yields in spite of a remaining mildew infection. Disease severity/yield relations obtained on a single tiller basis for either mildew infection at EC 75 or area under disease progress curve (AUDPC) were negatively correlated for untreated plants (R2= 89%, 94%) while this relation did not exist for inducer-treated ones (R2= 10%, 13%). Despite an increasing infection density. yields of main tillers of inducertreated plants were not decreased. On the other hand area under green leaf area curve (AUGLAC) showed a higher correlation with grain yield (R2= 89%) of inducer-treated plants. Possible explanations for the mitigated damaging effect of powdery mildew were expected in carbohydrate metabolism. especially carbohydrate formation and translocation. Assimilation rates of flag leaves of inducer-treated barley with similar infection densities to those of untreated plants were increased over a prolonged period and even exceeded those of non infected ones. In inducer-treated plants the export of 14CO2 from flag leaves into ears remained unimpaired by mildew infection and the allocation of assimilates to grains was highest at late stages of grain filling. Obviously plants were stimulated by inducer treatments to compensate for the damaging effect of powdery mildew and to tolerate mildew infection without yield loss.  相似文献

Arabidopsis thaliana is a host to the powdery mildew Erysiphe cichoracearum and nonhost to Blumeria graminis f. sp hordei, the powdery mildew pathogenic on barley (Hordeum vulgare). Screening for Arabidopsis mutants deficient in resistance to barley powdery mildew identified PENETRATION3 (PEN3). pen3 plants permitted both increased invasion into epidermal cells and initiation of hyphae by B. g. hordei, suggesting that PEN3 contributes to defenses at the cell wall and intracellularly. pen3 mutants were compromised in resistance to the necrotroph Plectosphaerella cucumerina and to two additional inappropriate biotrophs, pea powdery mildew (Erysiphe pisi) and potato late blight (Phytophthora infestans). Unexpectedly, pen3 mutants were resistant to E. cichoracearum. This resistance was salicylic acid-dependent and correlated with chlorotic patches. Consistent with this observation, salicylic acid pathway genes were hyperinduced in pen3 relative to the wild type. The phenotypes conferred by pen3 result from the loss of function of PLEIOTROPIC DRUG RESISTANCE8 (PDR8), a highly expressed putative ATP binding cassette transporter. PEN3/PDR8 tagged with green fluorescent protein localized to the plasma membrane in uninfected cells. In infected leaves, the protein concentrated at infection sites. PEN3/PDR8 may be involved in exporting toxic materials to attempted invasion sites, and intracellular accumulation of these toxins in pen3 may secondarily activate the salicylic acid pathway.  相似文献

Distribution of assimilates in cultivars of spring barley with different resistance against powdery mildew (Erysiphe graminis f. sp. hordei) Transport and distribution of radioactive labelled assimilates in spring barley cultivars with different degrees of resistance to powdery mildew were studied after 14CO2-treatment of single leaves. Plants of the cultivars ‘Amsel’ (susceptible), ‘Asse’ (adult plant resistant), and ‘Rupee’ (resistant) were analyzed at the vegetative growth stage (5. leaf unfolded) and the generative growth stage (anthesis). At the vegetative growth stage the assimilate export from the mildew inoculated 5. leaf of ‘Amsel’ and ‘Rupee’ is decreased; in ‘Asse’, there is no considerable change of assimilate distribution due to infection. At the generative growth stage the assimilate export from the infected flag leaf of ‘Amsel’ is reduced when the fungus, is sporulating. In the cultivar ‘Asse’ the assimilates are bound at the infection site until the seventh day after inoculation, then the transport of assimilates to the ear is increased. In ‘Rupee’ mildew inoculation causes an enhanced assimilate transport to the ear. The changes in assimilate distribution due to mildew inoculation are discussed with respect to the different types of host-parasite-interactions and the source-sink-activities in the different cultivars.  相似文献

Effect of grain soaking presowing in 1 mM salicylic acid (SA) and NaCl (0, 50, 100, 150 and 200 mM) on barley (Hordeum vulgare cv Gerbel) was studied. Increasing of NaCl level reduced the germination percentage, the growth parameters (fresh and dry weight), potassium, calcium, phosphorus and insoluble sugars content in both shoots and roots of 15-day old seedlings. Leaf relative water content (RWC) and the photosynthetic pigments (Chl a, b and carotenoids) contents also decreased with increasing NaCl concentration. On the other hand, Na, soluble sugars, soluble proteins, free amino acids including proline content and lipid peroxidation level and peroxidase activity were increased in the two plant organs with increasing of NaCl level. Electrolyte leakage from plant leaves was found to increase with salinity level. SA-pretreatment increased the RWC, fresh and dry weights, water, photosynthetic pigments, insolube saccharides, phosphorus content and peroxidase activity in the stressed seedlings. On the contrary, Na+, soluble proteins content, lipid peroxidation level, electrolyte leakage were markedly reduced under salt stress with SA than without. Under stress conditions, SA-pretreated plants exhibited less Ca2+ and more accumulation of K+, and soluble sugars in roots at the expense of these contents in the plant shoots. Exogenous application (Grain soaking presowing) of SA appeared to induce preadaptive response to salt stress leading to promoting protective reactions to the photosynthetic pigments and maintain the membranes integrity in barley plants, which reflected in improving the plant growth.  相似文献

Many plant pathogens gain entry to their host via stomata. On sensing attack, plants close these pores to restrict pathogen entry. Here, we show that plants exhibit a second longer term stomatal response to pathogens. Following infection, the subsequent development of leaves is altered via a systemic signal. This reduces the density of stomata formed, thus providing fewer entry points for pathogens on new leaves. Arabidopsis thaliana leaves produced after infection by a bacterial pathogen that infects through the stomata (Pseudomonas syringae) developed larger epidermal pavement cells and stomata and consequently had up to 20% reductions in stomatal density. The bacterial peptide flg22 or the phytohormone salicylic acid induced similar systemic reductions in stomatal density suggesting that they might mediate this effect. In addition, flagellin receptors, salicylic acid accumulation, and the lipid transfer protein AZI1 were all required for this developmental response. Furthermore, manipulation of stomatal density affected the level of bacterial colonization, and plants with reduced stomatal density showed slower disease progression. We propose that following infection, development of new leaves is altered by a signalling pathway with some commonalities to systemic acquired resistance. This acts to reduce the potential for future infection by providing fewer stomatal openings.  相似文献

The presented work was conducted on seedlings of spring barley and meadow fescue which differ in the degree of sensitivity to leaf spot pathogen Bipolaris sorokiniana (Sacc.) Shoem. The seedling reaction to inoculation with mycelium and conidia was examined in glasshouse conditions on the basis of respiration intensity and heat production. The leaf respiration was measured using Clark-type electrode, while heat emission was evaluated by means of isotermic microcalorimeter. The measurements were performed after 1, 3, 6, 10, 24, 48, 72, 168 and 240 hours since the inoculation moment. Leaves of meadow fescue were characterized by the most intense respiration at the 6th hour, while barley leaves at the 24th and 72nd hour after inoculation. In the case of meadow fescue the greatest heat production was noted in the period between 24 and 168 hours after inoculation. Simultaneously, at the 48th hour the smallest rate of respiration was observed. Barley leaves emitted the greatest amount of heat only in the first 3 hours of the pathogenesis. In these hours the smallest respiration rate was noted. The observed, opposing reaction of respiration intensity and heat emission in the infected seedlings of both species may illustrate a disorder in metabolic processes in plants during pathogenesis. The plants studied differed in the time of their reaction to pathogen attack: barley responded earlier in heat production, while fescue extended respiration rate in the first hours after inoculation. This is clearly observable, when coefficients of metabolic inefficiency (heat rates per mole O2) are compared. In the case of barley the highest rates were noticed just after inoculation, whereas in fescue at the 48th hour. In both species attack of pathogen caused high metabolic efficiency.  相似文献

During primary infection by conidia ofErysiphe graminis f. sp.tritici, three mechanisms of resistance operate in first leaves of 8-day-old seedlings of both resistant and susceptible wheats. The first mechanism, operating at the penetration site, is responsible for the failure of penetrations attempted by primary germ tubes (PGT). The second mechanism is concerned with the abortion of haustoria in normal-appearing host cells. The third mechanism relates to the abortion of haustoria and the hypersensitivity of the penetrated host cells.With the inoculum-level of 19–24 conidia/mm2, the three mechanisms together prevented 89.3 % of the attempted penetrations by PGT from producing normal haustoria in resistant wheat Purdue 5752C1-7-5-1 and 37.4 % in the susceptible wheat Vermillion. The first mechanism accounted for the prevention of 73.3 % of the attempted PGT penetrations on Purdue 5752C1-7-5-1 and 36 % on Vermillion. The second mechanism was responsible for stopping 19 % of all the successful penetrations in Purdue 5752C1-7-5-1 and 0.8 % in Vermillion. The third mechanism accounted for the failure of 41 % of all the successful penetrations in Purdue 5752C1-7-5-1 and 1.4% in Vermillion. Thirty-six hours after inoculation, 10.7% of all the attempted PGT penetrations appeared to be developing normally in first leaves of 8-day-old seedlings of resistant wheat Purdue 5752C1-7-5-1 as compared to 62.6 % in the susceptible wheat Vermillion.This appears to be the first report showing the relative effectiveness of various mechanisms of resistance concerning any powdery mildew fungus.  相似文献
