An analysis of the histone H1 subtype, H1-1, in eight legumes belonging to four genera of the tribe Vicieae (Pisum, Lathyrus, Lens, and Vicia), revealed an extended region consisting of the tandemly repeated AKPAAK motifs. We named this region the Regular zone (RZ). The AKPAAK motifs are organized into two blocks separated by a short (two or six amino acids) intervening sequence (IS). The distal block contains six AKPAAK motifs, while the number of repeats in the proximal block varies from six in V. faba to seven in the other species. In V. hirsuta, the first two repeated units of the proximal block are octapeptides AKAKPAAK. The apparent rate of synonymous substitutions in the blocks of RZ is much higher than in the rest of the gene. This can be explained by repeat shuffling within each block. In the C-domain of the orthologous H1 subtype from Medicago truncatula (tribe Trifolieae), a region corresponding to the RZ of Vicieae species was found. It also consists of two blocks of AKPAAK motifs (four and three repeats in the proximal and distal blocks, respectively). These blocks are separated by a 20-amino acid IS. The first 20 amino acids of the Medicago RZ are not part of AKPAAK repeats. We hypothesise that the RZ has most probably evolved as a result of an expansion of AKPAAK repeats from two separate sites in the C-domain. A hypothesis is proposed whereby weedy vetch (Vicia sativa L.) seed moved with seed of the cultivated lentil (Lens culinaris Medikus) as a tolerated weed during the spread of the lentil from the Fertile Crescent in the Near East to its current distribution. As a result, selection occurred in vetch weeds for a reduction in dormancy/hard-seededness, increased competitive ability and biomass, and phenological adaptation to new environments⇆redisposing the weed for domestication. The cropping of common vetch for forage in pure culture followed. Archaeological evidence of admixtures of grass pea (Lathyrus sativus L.) in Neolithic finds of lentil, pea (Pisum sativum L.) and bitter vetch (Vicia ervilia (L.) Wild.) suggests a similar process of selection in grass pea for a weedy habit from which domestication later occurred.  相似文献

The transferability of genome-specific sequence tagged microsatellite site (STMS) primers from field pea (P. sativum) and chickpea (C. arietinum) to other major pulses was examined. Overall, field pea STMS primers amplified products in most of the accessions in comparison to that of the chickpea STMS primers, which amplified products in relatively few accessions. The highest level of successful amplifications with a single primer was 89% for field pea and 33% for chickpea primers respectively. The potential transferability of the STMS primers among species, expressed as the total mean percentage of positive amplifications, was 53% for the field pea STMS primers and 9% for the chickpea STMS primers. The individual mean percentage of successful transferability of field pea STMS primers across lentil, vetch and chickpea/Cicer sp. accessions was 60%, 39% and 62%, respectively. Whereas, for the chickpea STMS primers successful transferability was 5%, 3% and 18% for lentil, vetch and field pea, respectively. The trnasferability of these STMS primers indicates a high level of sequence conservation in these regions across species. Together with their locus-specificity, co-dominant nature and potential to amplify multiple alleles, their transferability makes STMS markers a powerful tool for genetic mapping, diversity analysis and genotyping.  相似文献

Electrospray mass spectrometry was used to identify precisely the proteolytic cleavage points within, and at the C-termini of, the proprotein forms of four Viciae lectins that give rise to their two-chain forms. The lectins examined were the pea and lentil lectins, favin and theLathyrus odoratus lectin, which represent each of the four genera in this tribe. The molecular mass data showed single -chain forms for each lectin, with masses consistent with the available sequence and glycopeptide data, indicating that each came from a single proprotein. In contrast, the pea, lentil andL. odoratus -chains occurred in as many as five forms, due to multiple C-terminal cleavage points. Only favin showed a single -chain form. The -chain mass data were again consistent with the sequence information available, except for the lenti lectin -chain which was re-determined by protein sequencing. The two isolectin forms of this protein were shown to arise from -chain species with and without residue Lys53. The mass spectrum of concanavalin A was also examined and both the single-chain form and the two fragment forms showed no evidence of C-terminal heterogeneity.  相似文献

The X-ray crystal structure of lentil lectin in complex with -d-glucopyranose has been determined by molecular replacement and refined to anR-value of 0.20 at 3.0 Å resolution. The glucose interacts with the protein in a manner similar to that found in the mannose complexes of concanavalin A, pea lectin and isolectin I fromLathyrus ochrus. The complex is stabilized by a network of hydrogen bonds involving the carbohydrate oxygens O6, O4, O3 and O5. In addition, the -d-glucopyranose residue makes van der Waals contacts with the protein, involving the phenyl ring of Phe123. The overall structure of lentil lectin, at this resolution, does not differ significantly from the highly refined structures of the uncomplexed lectin.Molecular docking studies were performed with mannose and its 2-O and 3-O-m-nitro-benzyl derivatives to explain their high affinity binding. The interactions of the modelled mannose with lentil lectin agree well with those observed experimentally for the protein-carbohydrate complex. The highly flexible Me-2-O-(m-nitro-benzyl)--d-mannopyranoside and Me-3-O-(m-nitro-benzyl)--d-mannopyranoside become conformationally restricted upon binding to lentil lectin. For best orientations of the two substrates in the combining site, the loss of entropy is accompanied by the formation of a strong hydrogen bond between the nitro group and one amino acid, Gly97 and Asn125, respectively, along with the establishment of van der Waals interactions between the benzyl group and the aromatic amino acids Tyr100 and Trp128.RL and FC are joint first authors.  相似文献

Lectins from the seeds of broad bean (Vicia faba L.), pea (Pisum sativum L.), common vetch (V. sativa L.), and lentil (Lens culinaris Medik.) were isolated and purified by affinity chromatography. The hemagglutinating activity of lectins was most effectively inhibited by methyl--D-mannopyranoside, trehalose, and D-mannose. Other carbohydrate haptens, such as methyl--D-glucopyranoside, maltose, and alginic and D-glucuronic acids were less effective. Two lectins obtained from different lentil cultivars, unlike other lectins, had a relatively high affinity for melecitose, N-acetyl-D-glucosamine, L-sorbose, and sucrose. Furthermore, these lectins interacted with soluble starch. All the lectins examined had similar, but not identical, carbohydrate-binding properties. Because of their similar D-mannose/D-glucose specificity, these lectins interacted with lipopolysaccharides and exopolysaccharides of Rhizobium leguminosarum bv. viciae, root nodule bacteria that infect broad-bean, pea, common-vetch, and lentil plants with the formation of nitrogen-fixing symbiosis. However, owing to individual distinctions of carbohydrate-binding properties, these lectins showed a higher affinity for the polysaccharides of those microsymbionts within the R. leguminosarum bv. viciae species that were better specialized towards one or the other host plant from the cross inoculation group of legumes.  相似文献

Summary The agglutinin isolated from the seeds of Maclura pomifera (MPA) recognizes a mucin-type disaccharide sequence, Gal13GalNAc (T) on a human erythrocyte membrane. We have utilized the enzyme-linked lectinosorbent assay (ELLSA) and inhibition assay to more systematically analyze the carbohydrate specificity of MPA with glyco-recognition factors and mammalian Gal/GalNAc structural units in lectin–glycoform interactions. From the results, it is concluded that the high densities of polyvalent GalNAc1Ser/Thr (Tn) and Gal13GalNAc1Ser/Thr (T) glycotopes in macromolecules are the most critical factors for MPA binding, being on a nanogram basis 2.0 × 105, 4.6 × 104 and 3.9 × 104 more active than monovalent Gal, monomeric T and Tn glycotope, respectively. Other carbohydrate structural units in mammalian glycoconjugates, such as human blood group Sd (a+) related disaccharide (GalNAc14Gal) and Pk/P1 active disaccharide (Gal14Gal) were inactive. These results demonstrate that the configurations of carbon-4 and carbon-2 are essential for MPA binding and establish the importance of affinity enhancement by high-density polyvalencies of Tn/T glycotopes in MPA–glycan interactions. The overall binding profile of MPA can be defined in decreasing order as high density of polyvalent Tn/T (M.W. > 4.0 × 104) >> Tn-containing glycopeptides (M.W. < 3.0 × 103) > monomeric T/Tn and P (GalNAc13Gal) > GalNAc > Gal >> Man, LAra, DFuc and Glc (inactive). Our findings should aid in the selection of this lectin for elucidating functions of carbohydrate chains in life processes and for applications in the biomedical sciences.  相似文献

The content of -N-oxalyl-L-,-diaminopropionic acid (ODAP) and the activity of glycolate oxidase (GO) were positively correlated in the leaves of grass pea (Lathyrus sativus L.) seedlings. The activity of GO was kept at a steady level under the high light after treatment with ODAP. Although Na2S can activate GO, it cannot maintain the activity of GO under the high light. The content of ODAP increased and the activity of GO decreased with increasing oxalate concentration used for seedling treatment. The GO activity was high enough to keep photosynthesis at a steady level under high light. These findings suggested that Lathyrus sativus, using oxalate as a precursor to produce ODAP, protected the GO activity at high irradiance by scavenging the hydroxyl radicals.  相似文献

Summary Fragments of the E. coli chromosome that carry the dnaB groPB534 or groPB612 alleles have been cloned into a cosmid vector. The resulting recombinant plasmids contained the genes uvrA, groP (B534 or B612), and lexA. Further subcloning into high copy number plasmids, during which the uvrA and lexA genes were removed successively, yielded a groPB534 and groPB612 DNA fragment of about 2.4 kb each. Both fragments contained an overlapping 1.8 kb segment of DNA in which the sites of all restriction enzymes tested were identical. The size of these dnaB gene fragments were further delimited by deletion analysis.In E. coli groPB534 in which wild-type and A mutants do not replicate (Georgopoulos and Herskowitz 1971) phage replication is rescued if the strain contains the groPB534 gene on high copy number plasmids. On the contrary, in E. coli groPB612, which is temperature-sensitive for its groP character, replication of and A is abolished at 30° C if the strain contains the groPB612 recombinant plasmid. On the other hand, replication of B remains unaffected whether or not the groP strains harbor the isogenic dnaB gene-containing plasmid. The results suggest that within the cell not only the quality but also the relative amounts of dnaB and P protein are crucial for phage replication.  相似文献

The polyphagous larvae of click beetles (Coleoptera: Elateridae) are major pests of spring wheat in Montana, USA. Presently available insecticides are unable to provide control over wireworm populations, and the use of natural enemies has not been successful under field conditions. In this study, we examined the effect of seven trap crops: pea, lentil, canola, corn, durum, barley, and wheat, for their attractiveness to wireworms compared to spring wheat. Experimental plots were located in two commercial grain fields in Valier and Ledger, Montana, USA and the trials took place from May to August in 2015 and 2016. Wheat plants damaged by wireworms were recorded and their relative locations in wheat rows and adjacent trap crop rows within a plot were determined using destructive soil samples. In 2016, variable row spacing (0.25, 0.5, 0.75, and 1 m) between the trap crops (pea and lentil) and wheat was assessed. Shade house bioassays were conducted using potted pea, lentil, and wheat plants to support field trial results. Limonius californicus larvae, released at the center of each pot were sampled 4 and 10 days after sowing. Wheat intercropped with pea and lentil had significantly fewer damaged wheat plants. Wireworm numbers were lower in wheat intercropped with pea compared to the control for both locations and years. Shade house results corresponded with field results, with more wireworms collected from pea and lentil than wheat. In the spacing trials, wheat plant counts were also significantly higher when paired with pea and lentil, particularly at 0.5 m spacing. Regardless of inter-row spacing, significantly fewer wireworms were associated with wheat when intercropped with pea and lentil trap crops.  相似文献

Summary The cultivated lentil L. culinaris and the wild lentil L. ervoides are reproductively isolated from one another due to their hybrid embryo breakdowns. Using embryo culture, vegetatively normal hybrids were obtained. One specific hybrid, heterozygous for a reciprocal translocation, had about 50% gamete viability and produced aborted and viable embryos in a 11 ratio. In the F2, vegetatively normal and highly fertile plants were selected. With the aid of embryo culture techniques, L. ervoides can be included in the wild gene pool of the cultivated lentil.  相似文献

Two cDNA clones representing mRNAs, highly expressed in pea root tips, were isolated by mRNA differential display. Ribonuclease protection analyses showed different patterns of expression of these two messages in several pea tissues. Sequence analysis showed that the first clone, PsH1b-40, has 100% homology with a previously isolated H1 histone cDNA, PsH1b. However, it has an additional 30 nt at the 3 end which is absent in PsH1b, suggesting possible multiple polyadenylation sites in the same mRNA. The second clone, PsH1b-41, encodes a deduced 19.5 kDa protein of 185 amino acids with an isoelectric point of 11.5. The putative globular domain of the encoded protein showed 67–71% residue identity with globular domains of 28 kDa pea PsH1b H1 histone and Arabidopsis thaliana H1-1 H1 histone. It has 9 repeating motifs of (T/S)XXK. In the C-terminal domain, there are four lysine-rich repeating motifs of SXK(T/S)PXKKXK which may be involved in chromatin condensation and decondensation. Southern blot analysis of nuclear DNA shows that PsH1-41 belongs to a multigene family.  相似文献

Leaf water potential, leaf osmotic potential, chlorophyll a and b contents, stomatal conductance, net photosynthetic rate, and water use efficiency were determined in two pistachio species (Pistacia khinjuk L. and P. mutica L.) grown under osmotic drought stress induced by a combination of NaCl and polyethylene glycol 6000. A decrease in values for all mentioned variables was observed as the osmotic potential of the nutrient solution (s) decreased. The osmotic adjustment () of the species increased by decreasing s. Reducing losses from pod shatter is a major goal of most lentil (Lens culinaris) improvement programs, however, genetic variability is limited. Recently, a slashed pod trait was suggested to have potential value for reducing losses from shattering, but little was known about the trait. In this study we determined the anatomical features which were associated with the slashed pod trait. Histological specimens from ‘Brewer’ lentil (normal pods) and from germplasm lines expressing the slashed pod trait were compared to each other and to specimens from normal and reduced pod parchment pea (Pisum sativum) lines. Reduced parchment pea pods had less sclerenchyma tissue and fewer fibers than pea pods with normal parchment, but all lentil pods examined had comparable sclerenchyma tissue with similar quantities of fiber. The slashed pod trait was not the result of reduced sclerenchyma tissue (parchment) as had been previously suggested. Apparently, the slashed pod trait resulted from the stresses which develop between fiber cells within the sclerenchyma layer of the pod wall during pod maturation and drying. The trait had little effect on quality of seeds for human utilization in the environments tested but may have an undesirable effect in other environments. Although seed loss due to shatter appeared to be decreased in pods exhibiting the trait, the uncertainty of expression due to environmental influences makes the trait an unlikely candidate for use in lentil improvement programs.  相似文献

Summary The morphological characteristics and lectin-binding properties of mast cell granules from four human neurofibromata are described. Ultrastructural examination of the granules revealed that some contained dense cores, others had membranous configurations and some forms were intermediate between the two. A round electron-lucent area was present in some granules.After treatment with biotinylated lectins (10 g ml–1) followed by an avidin-peroxidase revealing system (5 g ml–1 in 0.125m Tris-buffered saline with 0.347m NaCl, pH 7.6), mast cell granules strongly bound Concanavalin A, garden pea, lentil, wheatgerm, erythro- and leuco-kidney bean lectins. This indicated the presence of abundantN-linked complex-type saccharide sequences. Soybean and peanut lectins showed only weak binding, while the presence of sparse -l-fucosyl terminals was indicated by the weak binding of winged pea lectin. The staining intensity of wheatgerm lectin was considerably reduced when incubated in the presence of its specific competing sugar tri-N-acetylchitotriose.Despite a wide variety of morphological differences between granules, all showed similar staining patterns and all granules within a single cell shared the same binding characteristics.  相似文献

1. The number and distribution pattern of -adrenergic receptors in the brain have been reported to be species specific. The aim of the present study was to describe binding of the -adrenoceptor ligand [125I]iodocyanopindolol in the brain of the tree shrew (Tupaia belangeri), a species which provides an appropriate model for studies of psychosocial stress and its consequences on central nervous processes.2. 125I-Iodocyanopindolol (125ICYP) labeling revealed a high degree of nonspecific binding, which was due mainly to interactions of this ligand with serotonin binding sites. For a quantitative evaluation of 1- and 2-adrenoceptors, serotonin binding sites had to be blocked by 100 M 5HT.3. Binding of the radioligand to 1- and 2-adrenoceptors was characterized using the 1-specific antagonist CGP20712A and the 2-specific antagonist ICI118.551. 1-adrenoceptor binding is present in the whole brain, revealing low receptor numbers in most brain regions (up to 1.5 to 2.7 fmol/mg). A slight enrichment was observed in cortical areas (lateral orbital cortex: 4.0±0.7 fmol/mg) and in the cerebellar molecular layer (8.7±1.0 fmol/mg).4. Competition experiments demonstrated high- and low-affinity binding sites with considerable variations in K i values for CGP20712A, showing that various affinity states of 1-adrenoceptors are present in the brain (K i: 0.61 nM to 67.1 M). In the hippocampus, only low-affinity 1-adrenoceptors were detected (K i: 1.3±0.2 M). Since it is known that 125ICYP labels not only membrane bound but also internalized -adrenoceptors, it can be assumed that the large population of the low-affinity sites represents internalized receptors which may be abundant due to a high sequestration rate.5. High numbers of 2-adrenoceptors are present in only a few brain structures of tree shrews (external layer of the olfactory bulb, 15.8±2.0 fmol/mg; claustrum, 19.3±1.5 fmol/mg; anteroventral thalamic nucleus, 19.4±1.5 fmol/mg; cerebellar molecular layer, 55.0±4.3 fmol/mg). Also for this class of -adrenoceptors, high- and low-affinity binding sites for the 2-selective antagonist ICI118.551 were observed, indicating that 125ICYP labels membrane bound and internalized 2-adrenoceptors. Only in the cerebellar molecular layer was a high percentage of high-affinity 2-adrenoceptors detected (K i for ICI118.551 was 1.8±0.3 nM for 90% of the receptors).6. In conclusion, 1- and 2-adrenoceptor binding can be localized and quantified by in vitro receptor autoradiography in the brains of tree shrews when serotonergic binding sites are blocked. A new virulent bacteriophage, designated RZ1, was isolated from a local pond on the facultative phototrophic bacteriumRhodobacter sphaeroides ZZ101. Electron microscopic studies revealed that, in general morphology, phage RZ1 resembles the bacteriophage ofEscherichia coli. The host range of phage RZ1 is limited to some strains ofR. sphaeroides. The phage genome consists of double-stranded DNA of about 44 kb lacking cohesive ends and seems to present terminal redundancy and cyclic permutation. RZ1 phage may carry out a lytic cycle only in recombination-defective mutants ofR. sphaeroides. Nevertheless, a derivative of the RZ1 phage, termed RW1, able to grow in recombination-proficient strains ofR. sphaeroides, has also been obtained. In vitro restriction analysis of both RZ1 and RW1 phages shows the presence of a rearrangement in their DNA. Generalized transduction of Strr and Rifr chromosomal markers has not been detected with either RZ1 or RW1 phages.  相似文献

Summary The conductance of the Ca2+-activated K+ channel (g K(Ca)) of the human red cell membrane was studied as a function of membrane potential (V m ) and extracellular K+ concentration ([K+]ex). ATP-depleted cells, with fixed values of cellular K+ (145mm) and pH (7.1), and preloaded with 27 m ionized Ca were transferred, with open K+ channels, to buffer-free salt solutions with given K+ concentrations. Outward-current conductances were calculated from initial net effluxes of K+, correspondingV m , monitored by CCCP-mediated electrochemical equilibration of protons between a buffer-free extracellular and the heavily buffered cellular phases, and Nernst equilibrium potentials of K ions (E K) determined at the peak of hyperpolarization. Zero-current conductances were calculated from unidirectional effluxes of42K at (V m –E K)0, using a single-file flux ratio exponent of 2.7. Within a [K+]ex range of 5.5 to 60mm and at (V m –E K) 20 mV a basic conductance, which was independent of [K+]ex, was found. It had a small voltage dependence, varying linearly from 45 to 70 S/cm2 between 0 and –100 mV. As (V m –E K) decreased from 20 towards zero mVg K(Ca) increased hyperbolically from the basic value towards a zero-current value of 165 S/cm2. The zero-current conductance was not significantly dependent on [K+]ex (30 to 156mm) corresponding toV m (–50 mV to 0). A further increase ing K(Ca) symmetrically aroundE K is suggested as (V m –E K) becomes positive. Increasing the extracellular K+ concentration from zero and up to 3mm resulted in an increase ing K(Ca) from 50 to 70 S/cm2. Since the driving force (V m –E K) was larger than 20 mV within this range of [K+]ex this was probably a specific K+ activation ofg K(Ca). In conclusion: The Ca2+-activated K+ channel of the human red cell membrane is an inward rectifier showing the characteristic voltage dependence of this type of channel.  相似文献

Grass pea (Lathyrus sativus L.) family leguminosae is cultivated as an important food and feed crop all over the world. It is very recalcitrant and difficult to regenerate and root under in vitro conditions. In this cotext, the study was carried out in three steps to find out the effects of three auxins [naphthalene acetic acid, indole 3 butyric acid, indole-3-acetic acid (IAA)], four sucrose concentrations and six types of substrate most suited for plant growth and helpful in acclimatisation of grass pea. The results showed that 2 mg L?1 IAA, 3 % sucrose was most suitable for rooting of grass pea. When different concentrations of sucrose were supplied to optimum concentration of IAA in Murashige and Skoog medium, 4.5 % sucrose concentration induced maximum number of 13.70 roots per explants that had positive impact on root length, fresh and dry weight of roots, plant height and morphology of the growing plants. There was 92.66 % acclimatisation and survival rate of these plants using peat moss compared to five other substrates used in this study. The developing plants were vigorous, flowered and set seed contained in pods under glass house conditions. It is concluded that rooting is affected by type and concentration of plant growth regulators and type of substrate has direct bearing on acclimatisation, flowering, pod and seed set of grass pea. As such this paper reports an efficient rooting and acclimatisation system of grass pea that will be very useful in future genetic transformation and breeding for improved characteristics.  相似文献

Globe artichoke (Cynara cardunculus L. var. scolymus L.) is a diploid (2n=2x=34), predominantly cross-pollinated plant native to the Mediterranean basin, and Italy contains the richest primary cultivated gene pool. Commercial production is mainly based on perennial cultivation of vegetatively propagated clones that are highly heterozygous and segregate widely when progeny-tested. Analysis of the artichoke genome by means of molecular markers has been limited to a few studies; here we report on the genetic relatedness among 118 artichoke accessions, including clones belonging to the same varietal type, two accessions of cultivated cardoon (C. cardunculus L. var. altilis DC.) and four accessions of wild cardoon [C. cardunculus L. var. sylvestris (Lamk) Fiori] as measured by amplified fragment length polymorphism (AFLP). Eight primer combinations yielded a total of 667 bands, of which 519 were polymorphic. Genetic similarities among accessions were calculated according to Jaccards Similarity Index and used to construct a dendrogram based on the unweighted pair group method using arithmetic averages. Our results demonstrate that AFLP markers can be useful in evaluating Cynara cardunculus genetic diversity and in classifying accessions to phylogenetic groups based on their genetic similarity values. Genetic variation among artichoke clones belonging to the same varietal type was in some cases higher than that found among accessions differently named and coming from different areas. The lowest Jaccards Similarity Index found within a varietal type can be considered as a threshold for the identification of accessions which share an analogous genetic background. This will enable the selection of representatives in order to develop and manage a germplasm core collection as well as the identification of suitable material for future artichoke breeding efforts.Communicated by J.S.
