The effects of five different virus inocula on three varieties of plum (Prunus domestica L.) were studied in a field trial of 10 years duration. The viruses causing line pattern, ringspot, prune dwarf and bark split had no effect on the growth of Marjorie's Seedling and Cambridge Gage, and only that causing prune dwarf stunted Oullins Gage trees. However, these viruses diminished the fruit yield of one or more varieties, and prune dwarf virus seriously decreased the yield of all three. Decline disease, probably caused by a strain of Prunus necrotic ringspot virus, developed in Marjorie's Seedling but not in the other varieties. The symptoms first appeared after 5 years and then the trees declined progressively with necrotic ‘incompatibility’ between rootstock and scion.  相似文献

Plum pox virus (PPV) was detected by ELISA throughout the year in extracts of root, bark, fruit, flowers and leaves of Prunus species; extracts from healthy plants gave negligible background reactions. In the summer, ELISA values obtained with extracts from infected leaves were variable but samples extracted at 1:50 (w/v) could have been diluted a further five to 110 times before reaching the limit of detection. Using a single antiserum the virus was detected in several hundred trees, suggesting that there was little antigenic variation. PPV was unevenly distributed in leaves and shoots and commonly occurred in only a few branches of an infected tree although it was frequently present in suckers growing from the roots. Virus was detected in the only three trees known to be infected in random leaf samples taken from 530 1-yr-old trees, but some infected trees were missed in samples taken from older trees and from a 7-yr-old rootstock hedge. The main practical use of ELISA for PPV is therefore as a sensitive and highly reliable confirmatory test which greatly facilitates control of the disease by the prompt destruction of infected trees.  相似文献

This is the first report of patch canker disease of rubber trees (clone RRIM600) in China. It is characterized by discrete irregular patches of rotted, discolored bark and wood, accompanied by a decrease in latex flow. A total of seven isolates of Pythium vexans were obtained from the diseased bark of the trunks and roots of rubber trees. Inoculating these isolates into healthy, mature rubber trees resulted in symptoms similar to patch canker and the same fungal species was re-isolated from the diseased tissues. This is also the first record of Py. vexans in Hainan.  相似文献

An extensive survey in 1976-82 indicated that sudden death of cocoa was widespread in Papua New Guinea with losses up to 1% per month in trees over 10-yr-old, especially in neglected plantations. Over 95% trees affected had large Phytophthora palmivora canker lesions and over 90% of the cankers were invaded by bark beetles. In an intensive single year survey of one plot 18-8% of 69 healthy trees died. Ambrosiella spp., P. palmivora and Fusarium solani were isolated from wood surrounding beetle tunnels but only the last two caused extensive lesions when inoculated into the xylem of healthy trees. The evidence obtained indicated that the sudden death syndrome is initiated by P. palmivora cankers on trunks and main branches but bark beetles are a major contributory factor and fungi associated with their tunnels may contribute to the problem. There is a risk of further outbreaks of the disease as cocoa cultivars susceptible to P. palmivora continue to be planted.  相似文献

The deep bark canker of the walnut is a serious threat to walnut plants that had an increasing verge since recent years. This disease is caused by Brenneria rubrifaciens bacteria. Therefore, this research was carried out with an aim to identify the deep bark canker agent. The suspected samples having symptoms were collected from the bark and branches of walnut trees. After purification, the phenotypic and biochemical specification of separations was assessed. Even the genetic diversity of pathogen population was conducted with BOX-polymerase chains reaction (PCR) technique and BOXAIR primer. Based on the phenotypic and biochemical specifications, 30 isolates were identified as B. rubrifaciens. In PCR reaction, the specific amplicons of size 537 and 671?bp in the case study strains were amplified via specific primer pairs. The present study is a prime report on outbreak of this disease in Iran and bacterial pathogenicity evidence of B. rubrifaciens on fruit in the world.  相似文献

A requisite for development of bacterial phloem canker caused by Erwinia rubrifaciens is the presence of the highly susceptible Hartley cultivar; when interplanted with Hartley, the Franquette and Payne cultivars are sometimes attacked by the disease. The recently developed cultivars, Gustine and Howe, developed active cankers when inoculated, but not as extensive as those in Hartley. The age of the plant part is important in the disease. One-year-old Hartley trees and twigs of the current season on mature trees did not develop active cankers. The complete disease syndrome occurs only on trunks and primary (scaffold) branches. Extension of the cankers was most rapid during the summer when the temperature was high. This was correlated with the effect of temperature on growth of the bacterium in culture. At midday in summer the temperature of the cambial area of a shaded trunk was as much as 10d?F (5.6 d?C) below ambient temperature but it was commonly a few degrees above ambient at night. Breaks in the thick phelloderm of the trunks and branches are necessary for penetration of the pathogen to the inner bark. Of the several types of breaks commonly occurring, those produced by mechanical harvesting equipment and by sap-sucking birds were found to be infection sites. Bacteria occur in large numbers in a slimy substance which exudes through cracks and accumulates on the bark of infected trees. They survived for at least 123 days in the exudate and were disseminated laterally as far as 20 ft (6.1 m) in wind-blown rain. Bacteria were also transmitted from tree to tree in exudate contaminating the pads of mechanical harvesting equipment.  相似文献

Coniothyrium canker caused by Coniothyrium zuluense, is a serious stem canker disease of Eucalyptus species in sub-tropical regions of South Africa. This disease is typified by necrotic bark lesions that coalesce to form large kino-impregnated cankers along the stems of trees. The strategy currently used to manage Coniothyrium canker in plantations is to deploy Eucalyptus species or clones that are resistant to the disease. Considerable success has already been achieved in this regard, but the long-term durability of resistance is of concern. Thus, forest managers are interested in the genetic diversity of the pathogen and its potential to overcome disease resistance in planting stock. In this study, 344 isolates of C. zuluense from different plantation regions in South Africa were compared on the basis of colony colour, conidial morphology, growth characteristics on agar and pathogenicity to a susceptible E. grandis clone. Conidia of all C. zuluense isolates measured were similar in size and shape. The fungus is slow growing in culture, which is indicative of its apparent biotrophic habit, with optimum growth observed at 30 °C. Isolates of C. zuluense displayed considerable variation in colony colour and pathogenicity in inoculation trials. Variation in morphology and pathogenicity amongst isolates suggests that C. zuluense has been present in South Africa for an extended period of time, or that it is changing rapidly due to strong directional selection pressures. The pathology and distribution of European beech trees bearing elongated bark lesions (strip-cankers) were investigated. Two types of canker were recognized: those on small trees (<40 cm diameter at breast height (dbh): 1·4 m above ground level) which bore fruit bodies of the xylariaceous ascomycete Biscogniauxia nummularia (Bull.) O. Kuntze, and those on larger specimens (>40 cm dbh) which were consistently associated with the diatrypaceous ascomycete Eutypa spinosa (Pers.) Tul. & C. Tul. All cankers were strictly annual, having formed during single growing seasons following periods of low water availability. The regional and local distribution of trees bearing lesions also appeared to be correlated with environmental conditions, being most severe where low rainfall or high temperatures had occurred. Population studies of the associated fungi, generally considered as saprotrophs, indicated the presence of unique genotypes within individual cankered trees and provided no evidence for the existence of pathotypes within either species. Within the decay columns which underlay canker surfaces, both B. nummularia and E. spinosa formed longitudinally extensive genets, implying non-mycelial spread in colonization. Suppression of both inter- and intraspecific incompatibility between fungi occurred in regions of canker decay columns with elevated water contents. The possible significance of coexistence between the ascomycetes B. nummularia and ' Hypoxylon purpureum ' ( sensu Sharland & Rayner, 1989 b ) and of the formation of heterokaryons by E. spinosa is discussed.  相似文献

In the southeastern United States, the establishment of short-rotation intensively cultured plantations of hybrid poplar has been hindered by its susceptibility to stem cankers. We evaluated the tradeoffs between biomass yield and disease tolerance in hybrid poplar genotypes belonging to P. deltoides × P. maximowiczii (DM), P. deltoides × P. nigra (DN), P. trichocarpa × P. maximowiczii (TM), and P. deltoides × P. deltoides (DD) taxa. We hypothesized that canker resistant genotypes will have thicker bark but bark thickness and biomass yield will be negatively correlated. After two growing seasons, the DD genotypes developed thicker bark compared to the genotypes of other taxa and bark thickness was not correlated with biomass yield in the DD genotypes (R2 = 0.002). However, in the TM, DM, and DN genotypes, bark thickness was negatively correlated with biomass yield (R2 = 0.33–0.77). Disease incidence studies revealed that the DM genotypes were most susceptible to canker whereas no disease was detected in DD genotypes. Furthermore, bark analysis conducted by Fourier transform infrared spectroscopy coupled with multivariate analysis showed that that DD genotypes to be chemically separate from the three hybrid genotypes and that bark chemistry was correlated with canker disease incidence. Taken together, these results reveal that it is possible to generate hybrid poplar genotypes with thicker bark, disease resistance, and higher biomass yields. This insight should guide further efforts to develop genetically improved hybrid poplar genotypes, both in terms of biomass yield and disease tolerance, for cultivation in the southeastern United States. Hybrid poplar cultivation in southeastern United States is hindered by its susceptibility to stem cankers. We evaluated tradeoffs between yield and canker disease resistance in various hybrid poplar genotypes. After two growing seasons, the DD genotypes showed disease resistance and developed thicker bark that was chemically distinct from the other genotypes. Bark thickness was not correlated with yield in the DD genotypes but was negatively correlated with yield in the other genotypes. These results will guide the development of hybrid poplar genotypes that are both disease resistant and high yielding for cultivation in the southeastern United States.  相似文献

Chrysophorte cubensis induced canker occurs in nearly all tropical and subtropical regions where eucalypts are planted, causing losses in both wood quality and volume productivity, especially so in the warmer and more humid regions of Brazil. The wide inter and intra-specific genetic variability of resistance to canker among Eucalyptus species facilitates the selection of resistant plants. In this study, we evaluated resistance to this pathogen in five Eucalyptus grandis (G) and 15 E. urophylla (U) trees, as well as in 495 individuals from 27 progenies derived from crosses between the trees. In the field, six-months-old test seedlings were inoculated with C. cubensis. Lesion length in the xylem and bark was measured eight months later. The results demonstrated that xylem lesions could preferentially be used for the selection of resistant clones. Eight trees (7 U and 1 G) were susceptible, and the remainder (8 U and 4 G) resistant. Individual narrow and broad sense heritability estimates were 17 and 81%, respectively, thereby suggesting that canker resistance is quantitative and highly dependent on dominance and epistasis.  相似文献

The activity of metalaxyl, fosetyl-Al, dimethomorph, and cymoxanil against Phytophthora crown rot of peach trees was examined. Application of fosetyl-Al or metalaxyl by painting the trunk (150 g/l) of 3-year-old PR204 trees was inhibitory to growth of the fungus but neither dimethomorph nor cymoxanil were effective. Application of metalaxyl as a soil drench suppressed canker development when the treated trees were subsequently inoculated with Phytophthora cactorum . Fosetyl-Al significantly reduced the growth of fungus compared with cymoxanil, dimethomorph and control but it was not as effective as metalaxyl. Again, dimethomorph and cymoxanil did not influence canker development. Both metalaxyl and fosetyl-Al were active for at least 21 days after applications. Strips of trunk bark were removed from trees, that had been drenched with the tested chemicals 20 days before, and inoculated on the cambium side with P. cactorum . Metalaxyl was the most effective fungicide and fosetyl-Al significantly reduced the development of fungus compared with dimethomorph, cymoxanil and the untreated strips. Colonization of strips treated with dimethomorph was significantly less than untreated strips. In contrast, cymoxanil did not inhibit the growth of fungus. Application of metalaxyl or fosetyl-Al as trunk paint or a soil drench appear to be effective procedures for preventing Phytophthora rot of peach trees.  相似文献

1 Sudden oak death is caused by the apparently introduced oomycete, Phytophthora ramorum. We investigated the role of bark and ambrosia beetles in disease progression in coast live oaks Quercus agrifolia. 2 In two Marin County, California sites, 80 trees were inoculated in July 2002 with P. ramorum and 40 were wounded without inoculation. Half of the trees in each group were sprayed with the insecticide permethrin [cyclopropanecarboxylic acid, 3‐(2,2‐dichloroethenyl)‐2,2‐dimethyl‐(3‐phenoxyphenyl) methyl ester] to prevent ambrosia and bark beetle attacks, and then were sprayed twice per year thereafter. After each treatment, sticky traps were placed on only the permethrin‐treated trees. Beetles were collected periodically in 2003. 3 Inoculated trees accounted for 95% of all beetles trapped. The ambrosia beetles Monarthrum scutellare and Xyleborinus saxeseni and the western oak bark beetle Pseudopityophthorus pubipennis were the most abundant of the seven species trapped. 4 Permethrin treatment delayed initiation of beetle attacks and significantly reduced the mean number of attacks per tree. Beetles did not attack any wounded or noncankered inoculated trees. 5 Trees with larger cankers trapped more beetles early in the disease. Once permethrin lost effectiveness, the number of beetle entrance tunnels was a more reliable predictor of subsequent trap catch than was canker size. 6 Beetles were initially attracted to P. ramorum cankers in response to kairomones generated in the host‐pathogen interaction. After beetles attacked the permethrin‐treated trees, aggregation pheromones most probably were the principal factor in beetle colonization behaviour.  相似文献

Nematode pathogens cause wilt diseases in conifers and deciduous trees. The longhorn beetles (Coleoptera: Cerambycidae) and bark beetles (Coleoptera: Curculionidae: Scolytinae) act as nematode vectors spreading the invasive juvenile stages during their maturation feeding or during oviposition on the plant hosts. There are numerous reviews of nematodes associated with bark beetles on conifers, while little attention has been paid to the nematodes of deciduous trees. The development of Dutch elm disease and ash dieback is mainly caused by fungal pathogens transmitted by bark beetles; the latter act as vectors of not only fungi but also nematodes enclosed in nematangia under their elytra, and also in the tracheae and Malpighian canals. Apart from phytopathogenic nematodes, bark beetles transmit mycophagous and bacterivorous nematodes and own parasites of bark beetles. The ecological groups of nematodes associated with Scolytinae are reviewed; the known records of associations of nematodes with bark beetles are listed for coniferous host plants of Russia and neighboring countries; the world-wide list of these associations for deciduous plant hosts is given.

Canker disease of Theobroma cacao L. caused by Phytophthora palmivora (Butler) Butler gives rise to serious damage in Fiji and is common in the S.W. Pacific area. Fusarium decemcellulareBiick (imperfect stage of Calonectria rigidiuscula (Berk. & Br.) Sacc.) was constantly associated with P. palmivora in disorders of cocoa but was not found to be a primary pathogen. Most Amelonado cocoa trees were resistant to canker but there were differences in the reactions of other Forastero and of Trinitario types. The F1 generation of a local Trinitario selection, itself resistant, showed segregation resulting in a proportion of very susceptible plants. Isolates of P. palmivora from black pod, canker and green twigs were equally pathogenic, both to pods and seedling stems. Sporangial dimensions, particularly the length/breadth ratio, differed from those reported elsewhere. It is uncertain whether the reason that cocoa canker is more serious in some countries than others is because of differences in the pathogenicity of P. palmivora strains to bark or the genetic differences of the cocoa grown.  相似文献

The bacterial die-back and canker of poplar is characterized by dark necrotic areas in the leaves, die-back accompanied by cracks in the bark of 1–4-year-old branches, and by cankers on the trunk and branches of different ages. The cankers may be 'closed' or 'open' and when moist exude slime containing bacteria.
The disease is caused by Pseudomonas syringae f. sp. populea , a new forma specialis closely resembling Ps. syringae in bacteriological characters but differing from it in parasitism. The association of the slime with the bacteria is essential for the production of typical lesions.
Inoculations with the pathogen alone, or in association with other organisms produce atypical symptoms.
The bacteria attack the cortical parenchyma and occasionally invade the xylem vessels.  相似文献

One-yr-old loblolly pine seedlings of two half-sib families, grown in sand, were fertilized three times per week with nutrient solution containing 20 μg/ml (low) or 80 μg/ml (high) nitrogen. Nitrogen concentration in the nutrient solution was either constant throughout the experiment, or interehanged after the inoculation of stems or shoots with Fusarium subglutinans, 55 days after initiation of fertilization. Growth was suppressed by a weekly excision of shoots branching from the stem apex. Either high nitrogen nutrition or shoot excision generally enhanced canker elongation on stem inoculated plants; the combination of both was extremely conducive for disease development. With intact plants of family 8–68, interchange of pre-inoculation low nitrogen nutrition with high nitrogen after inoculation enhanced canker elongation and rate of wilt. Nitrogen content varied in wood, bark and needles, as well as with time intervals, but was consistently in accordance with nitrogen level in the nutrient solution. In shoot excised plants, nitrogen content was higher than in the respective treatment without shoot excision. The higher nitrogen nutrient accelerated disease development on inoculated shoots, compared to low nitrogen, on both pine families. With respective treatments, stem cankers were larger and rates of shoots exhibiting lesions or wilt were higher on plants of family 8–68 than on 8–61. It is postulated that the disease enhancing effect associated with higher nitrogen content in stem tissues results from an increased nitrogen availability to the pathogen.  相似文献

Chestnut blight is controlled in Europe by using Cryphonectria hypovirus CHV1, a non-encapsulated RNA virus. The chestnut blight fungus, Cryphonectria parasitica, is weakened by the virus, and healing tissue growth occurs in the host tree. Transmission of this cytoplasmic hypovirus is restricted by the incompatibility system of the fungus, so that the hypovirus can be transmitted only between isolates of the same or closely related vegetative compatibility (vc) types. Hypovirulent isolates of C. parasitica (all of the French subtype CHV1-F1) from Castilla y León (NW Spain) were compared with virulent isolates in both laboratory (cut stems) and field inoculations (in two orchards in the province of León and one orchard in the province of Zamora). The tests were performed with the most common vc types in the region, EU1 and EU11. The cut stem assay revealed that the hypovirulent isolates of vc type EU1 did not reduce the growth of virulent cankers. By contrast, four hypovirulent strains H1, H4, H5 and H6 (all vc type EU11) reduced the growth of virulent isolates in the cut stem assay. Field tests showed that hypovirulent isolates of EU1 and EU11 were effective in reducing canker in both orchards in León with all treatments tested; however, in Zamora, where only EU11 was tested, all the treatments failed except H1, which was able to reduce growth of the canker eighteen months after the inoculation. The development of hypovirulence suggests that hypovirus subtype F1 is well adapted in the province of León. Both naturally extended and inoculated hypoviruses appear to have reduced the incidence of the canker, thus improving chestnut stands. However, the inoculations were not as effective in the orchards in Zamora. This indicates that the disease could be controlled in Castilla y León by inoculation of trees with hypovirulent strains, but that more tests should be done in provinces where the hypovirus is still not present.  相似文献

Association of Tobacco Ringspot Virus with a Union Incompatibility of Apple   总被引:1,自引:0,他引:1  
A virus-like disease of apple was observed for the first time in Canada in 1974 in a three year old planting. The disease was characterised by stem pitting, necrosis and breaking or separation of scion/rootstock at the graft union. Foliage was sparse and leaves were chlorotic and diffusely mottled. Using both frozen and freshly harvested leaves of infected apple trees from rootstock suckers as inoculum, a virus was transmitted to herbaceous indicator hosts. Based on host range symptoms, serology and electron microscopy, this virus was identified as tobacco ringspot virus. Initial counts of nematodes in the rhizophere of apple trees affected with union necrosis showed high densities of Xiphinema americanum Cobb., a known vector of nepoviruses. Reports of similar symptoms have indicated that the disease, apple union necrosis and decline was associated with the recovery of tobacco ringspot virus from infected rootstocks. This is the first report associating a union incompatibility condition of apple and tobacco ringspot virus.  相似文献
