A comparison is made between the fine structure of yellow corpuscles and white corpuscles located within the kidneys of the holostean fish, Amia calva L. The yellow corpuscles are composed of epithelial cells possessing all the features of steroid-producing tissues, namely an abundance of vacuoles, tubular smooth endoplasmic reticulum, and mitochondria with tubular cristae. The Golgi apparatus is also a conspicuous component of their cytoplasm. These cells are homologous to adrenocortical cells of higher vertebrates and they have cytoplasmic projections which extend into the lumina of surrounding sinusoids. The white corpuscles possess epithelial cells of variable appearance but all cells contain secretory granules and an extensive rough endoplasmic reticulum. The secretory granules appear to originate at the Golgi apparatus and occasionally are observed intact in the intercellular space. However the method of release of these granules was not clearly defined. These corpuscles are similar to the corpuscles of Stannius which have been described in modern bony fish. The presence of multivesicular bodies and smooth endoplasmic reticulum in some cells may reflect the origin of the corpuscles of Stannius from the tubular nephron. A. calva appears to be a suitable organism for comparative studies into the function of the adrenocortical homolog and corpuscles of Stannius in “primitive” fish.  相似文献

The identity, distribution and structure of the adrenocortical homolog (AH) was made in Polypterus palmas Ayres using routine light and electron microscopy and histochemistry for the enzyme δ5-3β-hydroxysteroid dehydrogenase (3β-HSD). The AH is confined to yellow corpuscles which are positioned near the posterior cardinal and renal veins in the anterior two-thirds of each kidney. The 46–57 spherial to cigar-shaped corpuscles are placed end to end and are equally distributed in the kidneys. The tortuous cords of epithelial cells of each corpuscle are surrounded by sinusoids and are completely delimited from the haemopoietic and renal tissue of the kidneys. The steroidogenic nature of the cells is demonstrated by their 3β-HSD activity and their ultrastructure, namely lipid droplets, tubular smooth endoplasmic reticulum and mitochondria with tubulovesicular cristae. Giant mitochondria and gap junctions are notable features of AH cells in this fish. The yellow corpuscles of the kidneys in P. palmas represent the AH and this tissue has a distribution which is in accordance with the taxonomic position of Polypteriformes.  相似文献

We have studied the ultrastructure of the testis and interstitial tissue of lizard Lacerta muralis sacrificed in spring, summer and autumn, with special emphasis on the morphological changes of Leydig cells. From the autumn to the end of spring, Leydig cells showed a large smooth endoplasmic reticulum and mitochondria with tubular cristae. These features correlate with the synthesis and secretion of androgens. At the end of spring and after mating, the amount of smooth endoplasmic reticulum decreased and the mitochondria showed laminar cristae. Both features are typical of a latent period, during which time secretion of androgens remains inactive until the end of summer. The possible role of other cell organelles, such as Golgi complex and lysosome-like electron dense bodies, during both secreting and resting periods, is also discussed. Finally, we consider the relationship between seasons, secretory activity of Leydig cells and the spermatogenic cycle.  相似文献

Tissues slices superfused with medium containing no ACTH released only traces of corticosterone. Addition of ACTH to the medium caused the rate of corticosterone release to increase to a maximum about 45 min after the addition of ACTH, after which time it either remained constant or started to wane slightly. The rate of release was affected by tissue thickness; the maximum rate of corticosterone release occurred when the tissue slices were 200 microns. Stimulated adrenocortical cells had large spherical nuclei, numerous mitochondria with tubular cristae, numerous lipid droplets, and a large amount of smooth endoplasmic reticulum. Many cells had an extensive network of microfilaments adjacent to the plasma membrane and some microtubules. Prolonged superfusion caused degenerative changes in some of the cells. Both cytochalasin B and cytochalasin D, dissolved in DMSO before addition to the superfusion medium, inhibited the corticotropic responsiveness in a dose-dependent manner. Control tissue samples superfused with medium containing DMSO, but no ACTH and no cytochalasin, released significantly more corticosterone than corresponding unstimulated samples. Few or no microfilaments were observed in adrenocortical cells after treatment with cytochalasin. Neither colchicine nor vinblastine had any discernible effect on the corticotropic responsiveness. After treatment with colchicine, adrenocortical cells had an ultrastructure characteristic of inner zone stimulated cells except that they were mainly devoid of microtubules.  相似文献

Summary A method of primary tissue culture involving both disaggregation of cells by repeated exposure of small tissue fragments to a solution of trypsin, collagenase and hyaluronidase and explantation of the residual tissue fragments intermingled with isolated cells onto polyethylene discs, has been shown to be adequate for the prolonged maintenance (up to 30 days) in vitro of cells arising from decapsulated adult human adrenocortical tissue. The technique and its critical points are discussed.Adrenocortical cells were organized both as outgrowing columns from microexplants or as variously sized islets of monolayered cells. The ultrastructural features of ACTH-deprived adrenocortical cells (i.e., mitochondria with laminar cristae, endoplasmic reticulum mainly consisting of rough profiles, abundance of lipid droplets and -glycogen particles) suggest that the cells dedifferentiate and retain practically no steroidogenic activity.After 2 days of ACTH-treatment, cultured parenchymal cells were found to be quite similar to the zona fasciculata elements of the normal human adrenal cortex. They were grouped in islets of about 50–100 cells. Rough endoplasmic reticulum had decreased, but smooth endoplasmic reticulum showed focal proliferation. The pleomorphic mitochondria with laminar cristae, transformed into a homogeneous population of round or ovoid mitochondria containing tubulo-vesicular cristae. Lipid droplets and glycogen particles were decreased in number.After 7 days of daily treatment with ACTH, the cortical elements, whose nucleus and cytoplasm seemed to be enlarged, were arranged in clusters formed by up to 300 monolayered elements, in which dividing cells were consistently observed. Their cytoplasm was filled with a meshwork of smooth reticulum tubules, in which scantly ribosome-studded profiles and occasional small stacks of granular cisternae were embedded. Mitochondria were similar to those of the 2 days ACTH-treated cultures. Lipid droplets and glycogen particles were absent. The functional significance of these structural changes as well as the possible mechanism underlying the differentiative effect of ACTH are discussed.Primary cultures of human adult adrenals are proposed as a new tool for studies into the physiopathology of the adrenocortical cells under carefully controlled experimental conditions.mis|It is a pleasure to acknowledge our thanks to Drs. The adrenocortical homolog (AH) was identified in the reed-fish ( Calamoichthys calabaricus Smith) using histochemistry for the enzyme δ5-3β-hydroxysteroid dehydrogenase (3β-HSD) and its distribution and structure were examined with both light and electron microscopy. AH appears as yellow corpuscles which extend as isolated bodies throughout the length of both kidneys in close apposition to the posterior cardinal veins. The yellow corpuscles are composed of convoluted cords of epithelial cells and tortuous sinusoids which empty directly into the cardinal veins. Wide lateral intercellular spaces and accompanying microvilli give the impression of pseudotubules or pseudofollicles to the cords of epithelial cells. The extensive network of tubules of smooth endoplasmic reticulum, the many mitchondria with tubulo-vesicular cristae, the conspicuous Golgi apparatus, lipid droplets, many pleomorphic granules and the presence of 3β-HSD are indicative of the steroidogenic nature of the cells. Large, pleomorphic mitochondria and gap junctions are also typical of most cells. The fine structure of the cells in the AH of the reed-fish most closely resembles that of other Polypteriformes, but the tissue is more widely distributed in the kidneys than in any osteichthylian so far reported.  相似文献

(Ramsay Wright Zoological Laboratories, Department of Zoology, University of Toronto, and Division of Endocrinology, Toronto Western Hospital, and Department of Medicine, University of Toronto, Canada.) — Acta zool. (Stockh.) 61(1): 9–21. The ultrastructural morphology of adrenocortical cells of adult rats maintained in culture for four days with and without corticotropin (ACTH) was studied in comparison to freshly dissociated cells. Identification of cells belonging to zona glomerulosa, zona fasciculata, and zona reticularis was made on the characteristics of the mitochondria and smooth endoplasmic reticulum (SER), and was confirmed by comparison with cellular layers from intact adrenals. Without ACTH, glomerulosa and fasciculata cells showed disappearance of SER, atrophied Golgi apparatus (GA), and a striking proliferation of granular reticulum. Fascicular cells had a loss in mitochondrial matrix density and the mitochondrial cristae showed a tendency to convert to lamellar glomerulosa-type cristae. Zona reticularis cells were not strikingly altered. Maintenance with ACTH led to increased cell size and islet formation of cortical cells. Glomerulosa cells had normal appearance and fasciculata cells manifested a pronounced development of GA and SER, and a normal configuration of mitochondria. The effects of ACTH on glomerulosa cells suggest a trophic influence of ACTH on these cells in vitro. The alterations observed in fascicular cell mitochondria and SER can be explained in terms of known concepts of steroid biosynthesis and basically agree with the patterns of steroid synthesis observed in these cells previously (Price et al. 1975).  相似文献

The cytology and ultrastructure of the hypertrophied special zone, which is formed spontaneously in the adrenal cortex of adult female brush-tailed possums (Trichosurus vulpecula), was compared to the adrenocortical tissue in adult males in which the special zone, normally absent, was induced following castration alone or by additional treatment with follicle-stimulating hormone (FSH). The special zone in females was situated between the zona fasciculata and the zona reticularis, the latter being a rudimentary zone in this species. Special zone tissue extended as a broad band parallel to and on one side of the adrenal medulla. In the luteal phase of the reproductive cycle, the special zone cells showed ultrastructural features commonly associated with steroidogenic tissues, with many mitochondria and compact masses of smooth endoplasmic reticulum. Cytoplasmic lipid inclusions were rarely observed. In lactating females, however, the special zone cells exhibited cytological and ultrastructural features suggestive of a transformation in their morphology broadly divided into two types of cells: (1) cells at the periphery of the special zone (closest to the zona fasciculata) showed variable quantities of lipid inclusions, mitochondria with dispersed cristae, and segregation of the smooth endoplasmic reticulum into compact masses; (2) cells within the more central regions showed an increasing abundance of lipid inclusions which in many cells became the dominant feature of the cytoplasm. These special zone cells contained very little smooth endoplasmic reticulum and their mitochondria contained few cristae together with amorphous granular material within the matrix. In castrated males, special zone tissue developed between the zona fasciculata and the zona reticularis, appearing initially as focal islands of cells (8 months postcastration) and later (11 months postcastration) expanding into a single zone, probably via the proliferation and differentiation of adjacent cells of the zona fasciculata and longitudinal growth of the special zone. Similar focal aggregations of special zone cells were induced after 14 days of FSH treatment given to 2-month castrated males. In all castrated and FSH-treated castrated males, the ultrastructure of special zone cells was similar to that of special zone cells in luteal-phase female possums. The findings suggest that the formation and cellular composition of the special zone is associated with changes in the pituitary-gonadal axis and that FSH plays a primary role in the differentiation of this tissue.  相似文献

Summary The ultrastructure of the interrenal (adrenocortical) cells of trout (Salmo fario L.) was studied after dexamethasone treatment. A procedure for identifying and isolating interrenal tissue fragments from the surrounding head kidney tissue prior to their preparation for electron microscopy is described. The peripheral plasma cortisol concentrations were measured in order to evaluate the steroidogenic activity of this tissue.The interrenal cells of control animals contain numerous mitochondria with tubular cristae, and a well developed and highly organized smooth endoplasmic reticulum (SER). The scarcity, or absence, of lipid droplets contrasts markedly with the abundance of SER.Treatment with dexamethasone results in a decrease steroidogenic activity of the interrenal cells, as indicated by the fall in plasma cortisol concentrations. The interrenal cells are small, but still contain numerous mitochondria. The SER is poorly developed, but masses of densely intermeshed smooth cisternae subsist. Lipid droplets do not accumulate in these cells; this peculiarity is discussed in connection with the virtual absence of liposomes in teleost interrenal cells.  相似文献

Summary The ovary of the liver fluke has been studied with light and electron microscopy. The organ consisted of germ cells and a layer of peripheral cells suggested to be nurse cells, and was surrounded by a capsule containing muscular tissue. The peripheral cells rested on a thick basement membrane and were irregular in outline. Their nuclei were of irregular shape, the mitochondria were dark with few cristae and the endoplasmic reticulum was tubular or vesicular and partly studded with ribosomes. The germ cells were rounded or polyhedral except in the outer part of the ovary where some of them showed irregular processes. The germ cells of the outer region (oogonia) were relatively small and in close contact with the cells suggested to be nurse cells. The inner germ cells (oocytes) were large and loosely packed. Their nuclei were irregular and contained round distinct nucleoli. The nuclear envelopes showed numerous pores. The endoplasmic reticulum was very sparse, but free ribosomes were abundant in the cytoplasm. This corresponded with a strong basophilia removable with RNase. In addition round basophilic bodies formed by densely packed ribosomes and membraneous material occurred in close spatial relation to mitochondria. The latter contained dense granules and few cristae. Groups of vesicles and membraneous lamellae were found in the cytoplasm, but they were considerably smaller than vertebrate Golgi complexes. Numerous dense spherical granules were found mainly in the periphery of the large germ cells. The granules were strongly osmiophilic except in the terminal part of the ovary. Slices of whole adrenal gland tissue, incubated in vitro in the presence of ACTH for 1 h and 2 h produced corticosterone and aldosterone in constant ratio (16:1). Tangential slices taken from the region immediately below the connective tissue capsule and slices taken from deeper regions of the gland consisted primarily of cells conforming to the distinct structural characteristics of the subcapsular zone (SCZ) and inner zone (IZ) tissues respectively. When samples were incubated in the presence of ACTH for 1 h and 2 h, the interrenal cells of the SCZ produced relatively more aldosterone than cells taken from the IZ of the gland. The corticosterone: aldosterone ratio for the IZ after 1 h (68:1) and after 2 h (102:1) were ten times greater than the ratios for the SCZ after 1 h (7:1) and after 2 h (10:1). The SCZ slices were not more than 60 cells thick and consisted of cells arranged in cords. These cells contained irregular nuclei, mitochondria with shelf-like cristae and a moderate abundance of smooth endoplasmic reticulum. In contrast, the production of large amounts of corticosterone by the cells of the IZ was associated with tissue containing more vascular space than the SCZ and the cells contained large round nuclei surrounded by an abundance of smooth endoplasmic reticulum and the mitochondria had tubular rather than shelf-like cristae.  相似文献

Summary There are two regions of steroidogenic cells in the duck adrenal gland. An outer, subcapsular zone (SCZ), consisting of cells with irregularly shaped nuclei, shows relatively little smooth endoplasmic reticulum and mitochondria with shelf-like cristae. This region surrounds the inner zone (IZ) of the gland which is comprised of smaller cells with rounded nuclei, a more abundant smooth endoplasmic reticulum and mitochondria with tubular cristae. When samples of tissue from these distinct regions of the gland are superfused in vitro with media containing concentrations of 1–24 ACTH ranging from 100 to 1000 ng per ml (0.034 to 0.34 M) the steroidogenic cells in both zones release corticosterone in a dose-dependent manner. The dose-responsiveness of both the SCZ and the IZ cells over this range is a complex quadratic function of the 1–24 ACTH concentration in the medium and the semilogarithmic linear portions of the dose-response curves are restricted to a narrow midrange of ACTH concentrations. Throughout the dose-response range, however, the steroidogenic cells of the IZ are more responsive to corticotropic stimulation than are the cells of the SCZ. The cells of the two zones are further distinguished by their responses to a challenge for a second time with medium containing 1–24 ACTH; the responses of the IZ cells to a second challenge were greater than those of the SCZ cells, and at a high concentration of ACTH the SCZ slices showed no significant second response.This work was supported by a grant from the National Science Foundation (PCM 79-15777) to James Cronshaw and W.N. The ultrastructure of the interstitial cells of the glandular tissue of Pleurodeles waltlii was studied in testis of animals obtained in early breeding season (January) under gonadotropic releasing hormone (GNRH) treatments and controls. These cells (parenchymal or Leydig-like cells) displayed the structural characteristics of steroid-producing cells. GNRH administration for 24 hours induced a significant decrease of both medial volume and volume density of lipid droplets. On the other hand, cell volume, nucleus, mitochondria, mitochondrial cristae and tubules of smooth endoplasmic reticulum were increased. Summary The fine structure of the interstitial gland of the ovary was studied in hypophysectomized rats and in hypophysectomized rats after denervation of the ovary or stimulation of the ovarian plexus. Hypophysectomized rats were used to eliminate gonadotropic influences on interstitial cells. In hypophysectomized rats, there was a large amount of intercellular space and cells had irregularly shaped nuclei and a large nuclear-cytoplasmic ratio. Prominent cytoplasmic features included small mitochondria with an electron-dense matrix, rough and smooth endoplasmic reticulum, polysomes and large osmiophilic lipid droplets. Interstitial cells from stimulated ovaries had reduced intercellular space and a reduced nuclear-cytoplasmic ratio. Mitochondria had tubular cristae; smooth endoplasmic reticulum-surrounded lipid droplets, and large polysomes were present. After section of the ovarian plexus, intercellular space was increased and filopodia were numerous. Cytoplasmic features included mitochondria with a dense matrix and indistinct cristae, large electronlucent lipid droplets, and variously sized multivesicular structures. These observations suggest that stimulation of the ovarian plexus in hypophysectomized rats causes regressed cells to assume the fine structural features of active steroidogenic cells. In contrast, interruption of the ovarian nerve supply causes a qualitative and quantitative increase in ultrastructural features characteristic of regressed steroidogenic cells. These responses of interstitial gland cells to denervation and stimulation provide morphological evidence for a functional role for the adrenergic nerves to this ovarian compartment.  相似文献

The structure of the rat parietal cell was examined by electron microscopy. The intercellular and intracellular canalculi are lined by microvilli which are more numerous and larger than those of other gastric cells. The numerous mitochondria have closely packed cristae and a dense matrix containing opaque particles. The cytoplasmic vacuoles typical of parietal cells are part of a network of smooth surfaced tubules and vacuoles (the endoplasmic reticulum) which is intimately associated with the mitochondria and probably connected with the lumen of the canaliculi. Only a few dense particles are found attached to the surface of these tubules. The structure of the parietal cell is compared with that of other cells whose function also is transport of inorganic ions and water. Evidence is presented supporting the hypothesis that parietal cells differentiate from a less structurally specialized cell in the neck region of the gastric gland.  相似文献

The zona fasciculata of the rat adrenal cortex synthesizes and secretes glucocorticoids. As observed after aldehyde fixation, the cells in this zone contain an extensive endoplasmic reticulum (ER), a small Golgi apparatus, a moderate number of lipid droplets, and abundant mitochondria with tubulovesicular cristae. Numerous areas within the endoplasmic reticulum and mitochondrial cristae appear clear. In addition, a small percentage of mitochondria encompasses large, clear areas. After immersion of finely minced adrenal cortex in unbuffered 2% OsO4 (40–48 hr at 40°C), deposits of osmium are seen within the Golgi apparatus, the entirety of the ER, and occasionally within mitochondria. In some mitochondria, the deposits are within cristae; in others, within vacuoles; in still others, in both cristae and vacuoles. These localizations correspond best to the clear areas found in aldehyde-fixed tissue. Osmium is not deposited in lipid droplets, in bar-containing inclusions, in mitochondrial matrix inclusions, or in the peripheral, outer mitochondrial spaces. Addition of zinc-iodide to OsO4 increases the amount of Golgi apparatus and mitochondrial staining. Adrenocorticotropin (ACTH) does not affect the localization of deposits; hypophysectomy decreases mitochondrial staining. This study (a) emphasizes the necessity for electron microscopic confirmation of osmium localization when this technique is used as a Golgi apparatus stain; and (b) suggests that the ER-staining pattern may be consistent in cells actively synthesizing steroids or steroid-like compounds.  相似文献

The paired Y-organs of crustaceans control the molting process. In males of C. antennarius, these glands are opalescent, lobulated, epithelioid structures embedded in brown fatty tissue. Cells in the periphery extend processes to the connective tissue capsule, an arrangement that suggests increased surface area for metabolic exchange. The processes contain mitochondria and are tipped distally with electron dense material. The cytoplasm, scarce relative to nuclear volume, contains vesicles, polymorphic mitochondria with tubular cristae, and numerous free ribosomes, but little in way of smooth or rough endoplasmic reticulum or Golgi complexes. Progressing from intermolt to the premolt stage, mitochondria, as well as vesicles, and electron-dense particles in peripheral processes increase somewhat in number. Also, heterochromatin masses concentrate adjacent to the nuclear envelope. Eyestalk removal, which induces premolt stages in some species, did not produce consistent change in Y-organ substructure in C. antennarius. Although evidence is accumulating that Y-organs secrete a steroid molting hormone during late intermolt-premolt, the substructure of the glands exhibits neither (a) striking changes with the molt cycle, nor (b) all the characteristics typical of vertebrate steroid hormone synthesizing glands. Nevertheless, the structural features, respectively, are consistent with biochemical evidence that Y-organs (a) rapidly take up and convert sterol precursor and secrete a product without its accumulation or change in total sterol pool size, and (b) apparently cannot synthesize the sterol precursor. Y-organ cytology closely resembles that of some vertebrate steroid hormone secreting glands in which this synthetic capacity is minimal.  相似文献

Summary Ultrastructure of the granulosa lutein cells of the raccoon from throughout pregnancy has been described. The lutein cells often from epithelial cords which are separated by the connective tissues, capillaries and lymphatics. Based on the arrangements and modifications of the cytoplasmic organelles and inclusions, three types of lutein cells have been recognized. The type I lutein cells predominantly contain tubular, agranular endoplasmic reticulum, juxtanuclear Golgi complexes, a few round to rod-shaped mitochondria, some free ribosomes, and occasional lipid droplets. Occasionally the tubular cristae of mitochondria and tubular smooth endoplasmic reticulum appear contiguous. The type II cells contain abundant lace-like and/or stacked fenestrated endoplasmic reticulum cisternae that frequently form membranous whorls, some tubular, agranular endoplasmic reticulum, mitochondria, and lipid droplets. Mitochondria are usually small, but unusual large ones also occur. The small, rod-to round-shaped mitochondria usually have tubular cristae; but the large, oval, elongate, and cup shaped mitochondria possess tubular, lamellar, plate like, and whorl-like cristae. The plasma membranes of the cells are complexly elaborated and folded, especially when apposing each other. In favorable sections, strands of fenestrated cisternae appose the folds of the plasma membranes. In general, the amount of cytoplasmic organelles and inclusions vary greatly in the cells. The type III cells predominantly contain lipid droplets and sparse cytoplasmic organelles. The type I and II cells are found throughout pregnancy, but the type III cells are observed from mid gestation to term. Taste buds in foliate papillae of the rhesus monkey were examined by electron microscopy. Three distinct cell types were identified. Type I cells were narrow elongated cells containing an oval nucleus, bundles of intermediate filaments, several Golgi bodies, and characteristic apical membrane-bounded dense granules. These cells exhibited morphological variations: some had a moderately dense cytoplasm, perinuclear free ribosomes, and flattened sacs of rough endoplasmic reticulum; others had a more lucent cytoplasm, dilated irregular rough endoplasmic reticulum, lysosome-like dense bodies, and lipid droplets. Type II cells typically contained a spherical, pale nucleus, a prominent nucleolus, supranuclear and infranuclear Golgi bodies, mitochondria with tubular cristae, and one or two centrioles. This cell type, too, showed some variation in the relative amounts of ribosomes and smooth endoplasmic reticulum, which varied inversely with each other. Type III cells were characterized by a clear apical cytoplasm essentially devoid of ribosomes and containing microtubules. In a few type III cells, the peri- and infranuclear regions contained many ribosomes and some rough endoplasmic reticulum. In most Type III cells, there were large numbers of dense and clear vesicles in the peri- and infranuclear regions; some of the vesicles were grouped in synapse-like arrangements with adjacent nerves. The morphological variations exhibited by all three cell types could be accounted for by age differences in each of the cells. This would be consistent with the notion that cell renewal occurs in each of the three cell populations.  相似文献
