We explored a benthic community living on stones in an acidic (pH2) stream of active volcanic origin from Patagonia, Argentina, by combining in situ measurements (temperature, pH, conductivity, dissolved oxygen), photosynthesis of intact biofilms (measured with microsensors by the light–dark shift method), pure-culture experiments on isolated algae, and confocal laser scanning microscopy on the biofilms. The epilithon of the Agrio River was dominated (99% of total biomass) by one species: Gloeochrysis (Chrysophyceae). This species was observed as brown, mucilaginous, 200-m-thick films on stones, growing in clumps in a dense matrix of fungal hyphae, bacteria, and inorganic particles held together by extracellular polymeric substances. Gloeochrysis was isolated and cultivated. The photosynthetic rate measured at saturation irradiance was 120 mol oxygen (mg chlorophyll a)–1h–1 under laboratory conditions, and the saturation rate of photosynthesis by carbon dioxide was 90 mol oxygen (mg chlorophyll a)–1 h–1 for oxygen evolution. Photosynthetic activity of the biofilm was light-dependent and saturated above 200 mol photons m–2 s–1. In the dark, the stone surface became anoxic. Our data suggest that primary production in the Agrio River was not limited by light, carbon, or phosphorus but instead, nitrogen-limited.  相似文献

Laurencia brongniartii is usually found at depths below 4 m, but can be found in shallow subtidal areas in crevices and on the walls of a coral reef in Amami Oshima Island, Kagoshima Prefecture, Japan, where irradiances were significantly lower than those at similar depths in open water. In preparation for the possible cultivation of this species for its antibiotic compounds, the effects of temperature and irradiance on photosynthesis and growth were measured. Photosynthesis and growth rates of L. brongniartii explants were highest at 26 and 28 °C, which closely corresponded to temperatures found during August to late December when it was most abundant. The estimated maximum photosynthesis rate (P max) was 4.41 mol photon m–2 s–1 at 26 °C and 4.07 mol photon m–2 s–1 at 28 °C. Saturating irradiance occurred at 95 mol photon m–2 s–1 at 26 °C and 65 mol photon m–2 s–1 at 28 °C. In contrast, growth experiments at 41.7 mol photon m–2 s–1 caused bleaching of explants and the maximum growth rate observed during the study was 3.02 ± 0.75% day–1 at 28 °C and 25 mol photon m–2 s–1. The difference in the saturating irradiance for photosynthesis and the irradiance that caused bleaching in growth experiments suggests that long-term exposure to high irradiance was detrimental and should be addressed before the initiation of large scale cultivation.  相似文献

Oscillations of photosynthesis induced in leaves of Vicia faba L. were accompanied by oscillations not only in the pH of the chloroplast stroma, but also by pH oscillations in the cytosol and in the vacuole of leaf mesophyll cells. Cytosolic pH oscillations were in phase with stromal oscillations, but antiparallel to vacuolar pH oscillations. During maxima of photosynthesis, the cytosolic pH exhibited maxima and the vacuolar pH minima. Vacuolar acidification is interpreted to be the result of energized proton transport from the cytosol into the vacuole. Since the ratio of dihydroxyacetone phosphate to phosphoglycerate is maximal during the peaks of photosynthesis (Stitt et al., 1988, J. Plant Physiol. 133, 133–143; Laisk et al., 1991, Planta 185, 554–562), while the activity of NADP-malic dehydrogenase is highest during minima of photosynthesis (Scheibe and Stitt, 1988, Plant Physiol. Biochem. 26, 473–481), the present data indicate in agreement with earlier observations (Yin et al., 1991, Planta 184, 30–34) that light-dependent cytosolic energization is brought about by the oxidation of dihydroxyacetone phosphate rather than of malate. They also indicate that the over-reduction of the electrontransport chain observed during minima of photosynthesis is relieved not predominantly by oxaloacetate reduction and export of the resulting malate from the chloroplasts but by another reaction, presumably oxygen reduction.Abbreviations CDCF 5-(and 6-)carboxy-2,7-dichlorofluorescein  相似文献

Removal of inorganic nitrogen sources by cells of the aerial microalga Trentepohlia aurea grown on the surface of substrate, such as filter paper, has been investigated in a batch system. When the alga grew on the paper dampened with medium, it actively ingested inorganic nitrogenous compounds in the medium. Immobilized cells on the filter papers were called algal biofilm in this study. When the algal biofilms were soaked in modified Bold's Basal medium (using 1 g NH4Cl l–1 as a N source), the removal rate was 4.25 mg ammonium-N l–1 day–1 in 40 days. In modified medium with added 26 mg nitrite-N, the removal rate of the total inorganic N ion by the biofilms reached 5.11 mg N l–1 day–1. This removal rate of total N ion was higher than that in the medium by addition of 26 mg nitrate-N. In addition, we tried to examine simultaneous removal of ammonium, nitrate, and nitrite ions and growth inhibition of cyanobacteria in the medium by using the algal biofilms. Consequently, it was demonstrated that the algal biofilms of T. aurea could be utilized as a biofunctional material for the purification of wastewater.  相似文献

This study reports the effects of light availability during the acclimatization phase on photosynthetic characteristics of micropropagated plantlets of grapevine (Vitis vinifera L.) and of a chestnut hybrid (Castanea sativa × C. crenata). The plantlets were acclimatized for 4 weeks (grapevine) or 6 weeks (chestnut), under two irradiance treatments, 150 and 300 mol m–2 s–1 after in vitro phases at 50 mol m–2 s–1. For both treatments and both species, leaves formed during acclimatization (so-called `new leaves') showed higher photosynthetic capacity than the leaves formed in vitro either under heterotrophic or during acclimatization (so-called `persistent leaves'), although lower than leaves of young potted plants (so-called `greenhouse leaves'). In grapevine, unlike chestnut, net photosynthesis and biomass production increased significantly with increased light availability. Several parameters associated with chlorophyll a fluorescence indicated photoinhibition symptoms in chestnut leaves growing at 300 mol m–2 s–1. The results taken as a whole suggest that 300 mol m–2 s–1 is the upper threshold for acclimatization of chestnut although grapevine showed a better response than chestnut to an increase in light.  相似文献

The combined use of antibiotics with low levels of electrical current has been reported to be more effective in controlling biofilms (the bioelectric effect) than antibiotics alone. An electrical colonisation cell was designed to study the effect of antibiotics on biofilms formed on a dialysis membrane away from the electrode surface. To avoid the electrochemical generation of toxic products,Pseudomonas aeruginosa biofilms were formed in minimal salts medium that excluded chloride-containing compounds. Under these conditions, electrical currents of up to 20 mA cm–2 did not prevent biofilm formation or have any detrimental effect on an established biofilm. Tobramycin alone at concentrations of 10 g ml–1 did not affect the biofilm, but were significantly enhanced by 9 mA cm–2. The effect of tobramycin concentrations of 25 g ml–1 were enhanced by a 15 mA cm–2 electrical current. In both cases higher levels of electrical current, up to 20 mA cm–2, did not further enhance the effect of the antibiotic. The possible mechanisms of action of the bioelectric effect have been reported to involve electrophoresis, iontophoresis and electroporesis, thus overcoming the biofilm biomass and cell wall barriers. Our results suggest that other factors may also be important, such as the metabolic activity and growth rate of the bacteria. Such factors may be critical in maximising antibiotic efficacy.  相似文献

Cultures in vitro of Betula pendula Roth were subjected to light of different spectral qualities. Photosynthetic capacity was highest when the plantlets were exposed to blue light (max recorded photosynthesis, 82 mol CO2 dm–2 h–1) and lowest when irradiated with light high in red and/or far-red wave lengths (max recorded photosynthesis, 40 mol CO2 dm–2 h–1). Highest chlorophyll content (2.2 mg dm–2 leaf area) was found in cultures irradiated with blue light, which also enhanced the leaf area. Morphometric analysis of light micrographs showed that the epidermal cell areas were largest in plantlets subjected to blue light and smallest in those subjected to red light. Morphometric analysis of electron micrographs of palisade cells, showed that the functional chloroplast area was largest in chloroplasts of leaves subjected to blue light and smallest in those exposed to red light. We suggest that light quality affects photosynthesis both through effects on the composition of the photosynthetic apparatus and on translocation of carbohydrates from chloroplasts.  相似文献

Summary Experiments were undertaken on the fresh water algaChara corallina to determine the form of inorganic carbon (CO2 or HCO 3 ) which enters the cell during photosynthesis at alkaline pH. Recent proposals have centered on the possibility that proton efflux in alkaline solution is able to generate, in the immediate vicinity of the cell, a sufficiently low pH to raise the partial pressure of CO2, and hence facilitate its passive permeation into the cell. Predictions have been made by modelling this situation (N.A. Walker, F.A. Smith & I.R. Cathers, 1980,J. Membrane Biol. 5751–58, J.M. Ferrier, 1980,Plant Physiol. 661198–1199), and these were tested by placing recessed-tip pH microelectrodes in the unstirred layer surrounding cells in stagnant solution (bulk pH 8.2, buffered only with 1mm HCO 3 ). Even as close as 2 m from the cell wall, the pH was typically 7.2 to 7.6 in the acid band center — over 1 pH unitgreater than that suggested by the models for CO2 entry at the necessary rate for C-fixation. Further evidence for the entry of HCO 3 , rather than CO2, at high solution pH was obtained from experiments in which the radial pH gradient in the unstirred layer was reduced. Buffer solutions containing 5mm phosphate or 5mm HEPES, raised the pH at the cell surface in the acid regions from around 7.2 to 7.8 or higher. This pH increase (reduction in acid gradient) would have greatly reduced the CO2 level at the cell surface and should, therefore, have greatly reduced the CO2-related14C-influx. However,14C-fixation was reduced by only 31% (phosphate) or 15% (HEPES), compared with buffer-free controls. Reduction of the unstirred layer thickness by fast solution flow resulted in a stimulation, and not a reduction, of14C-fixation. The similarity of our radial pH profiles near the wall with that predicted by the model (Walker et al., 1980) assuming H+–HCO 3 cotransport, together with the effects of buffer, and the results of increased solution flow rate, lead to the conclusion that cotransport of HCO 3 with H+ is the likely method of entry of inorganic carbon. Longitudinal pH profiles of theChara cell were obtained at a distance of 25 m from the wall. These revealed much sharper delineation of the acid and alkaline bands than has previously been possible with miniature pH electrodes. Profiles of local electric field, obtained with a vibrating probe, were in excellent agreement with the high resolution pH profiles. This supports the hypothesis that membrane proton transport has a role (direct) in the generation of the extracellular currents.  相似文献

A photobioreactor was constructed using either anchored polyurethane foam strips (1 × 1 × 40 cm, PU-strips) fixed on a stainless-steel ring to prevent flotation, or free-floating polyurethane foam blocks (1 × 1 × 1 cm, PU-blocks) as biomass supporting materials (BSM). The cyanobacterium,Scytonema sp. TISTR 8208, which produces an antibiotic, was immobilized onto PU-strips or -blocks. The free-floating PU-blocks could immobilize only about 70% of the total cells, while the anchored PU-strips could immobilize as much as 97%. PU-strips were chosen as the BSM and we named this type of reactor, seaweed-type bioreactor (STB). Optimal physical conditions for antibiotic production were determined in the STB. Inoculum density was 0.4 g l–1 and cells were sparged with air containing 5% CO2 circulated at the gas flow rate of 250 ml min–1 and illuminated at a light intensity of 200 mol photon m–2 s–1. Rapid lowering of the external pH (pH jump) enhances Cl influx in Chara. Experiments were conducted to distinguish between two factors which have previously been proposed to mediate in the response: raised cytoplasmic pH and lowered cytoplasmic Cl concentration. It is concluded that the latter alternative is more likely because: i) Cl influx is reduced at high external pH; ii) influx following the pH jump is never greater than that following pretreatment in Cl-free solution, which reduces cytoplasmic Cl concentration (Cl starvation); iii) the joint application of pH jump and Cl starvation does not result in a greater Cl influx than does Cl starvation alone; and iv) addition of NH 4 + , which increases cytoplasmic pH, does generate an additional stimulation of Cl influx following a pH jump. It is suggested that the increased cytoplasmic pH at the end of pretreatment at high external pH decays rapidly during the pH jump, and thus any effect on Cl influx is so transient as to be undetectable by the methods used. The results are discussed in terms of a reaction kinetic model for 2H+/Cl cotransport (Sanders, D. and Hansen, U.-P, 1981, J. Member. Biol. 58, 139–153) which describes quantitatively; i) the effects of NH 4 + on Cl influx in terms involving only a change in cytoplasmic pH; and ii) the combined effects of Cl starvation and NH 4 + in terms involving only changes in Cl concentration and cytoplasmic pH. Conversely, the combined effects of Cl starvation and pH jump cannot be described by the model if the effect of the pH jump is the consequence of increased cytoplasmic pH. Measurements of net ecosystem CO2 exchange by eddy correlation, incident photosynthetically active photon flux density (PPFD), soil temperature, air temperature, and air humidity were made in a black spruce (Picea mariana) boreal woodland near Schefferville, Quebec, Canada, from June through August 1990. Nighttime respiration was between 0.5 and 1.5 kg C ha–1 h–1, increasing with temperature. Net uptake of carbon during the day peaked at 3 kg C ha–1 h–1, and the daily net uptake over the experiment was 12 kg C ha–1 day–1. Photosynthesis dropped substantially at leaf-to-air vapor pressure deficit (VPD) greater than 7 mb, presumably as a result of stomatal closure. The response of ecosystem photosynthesis to incident PPFD was markedly non-linear, with an abrupt saturation at 600 mol m–2 s–1. This sharp saturation reflected the geometry of the spruce canopy (isolated conical crowns), the frequently overcast conditions, and an increase in VPD coincident with high radiation. The ecosystem light-use efficiency increased markedly during overcast periods as a result of a more even distribution of light across the forest surface. A mechanistic model of forest photosynthesis, parameterized with observations of leaf density and nitrogen content from a nearby stand, provided accurate predictions of forest photosynthesis. The observations and model results indicated that ecosystem carbon balance at the site is highly sensitive to temperature, and relatively insensitive to cloudiness.  相似文献

A field experiment in western Kenya assessed whether the agroforestry species Tithonia diversifolia (Hemsley) A. Gray, Tephrosia vogelii Hook f., Crotalaria grahamiana Wight & Arn. and Sesbania sesban (L) Merill. had access to forms of soil P unavailable to maize, and the consequences of this for sustainable management of biomass transfer. The species were grown in rows at high planting density to ensure the soil under rows was thoroughly permeated by roots. Soil samples taken from beneath rows were compared to controls, which included a bulk soil monolith enclosed by iron sheets within the tithonia plot, continuous maize, and bare fallow plots. Three separate plant biomass samples and soil samples were taken at 6-month intervals, over a period of 18 months. The agroforestry species produced mainly leaf biomass in the first 6 months but stem growth dominated thereafter. Consequently, litterfall was greatest early in the experiment (0–6 months) and declined with continued growth. Soil pH increased by up to 1 unit (from pH 4.85) and available P increased by up to 38% (1 g P g–1) in agroforestry plots where biomass was conserved on the field. In contrast, in plots where biomass was removed, P availability decreased by up to 15%. Coincident with the declines in litterfall, pH decreased by up to 0.26 pH units, plant available P decreased by between 0.27 and 0.72 g g–1 and Po concentration decreased by between 8 and 35 g g–1 in the agroforestry plots. Declines in Po were related to phosphatase activity (R2=0.65, P<0.05), which was greater under agroforestry species (0.40–0.50 nmol MUB s–1 g–1) than maize (0.28 nmol MUB s–1 g–1) or the bare fallow (0.25 nmol MUB s–1 g–1). Management of tithonia for biomass transfer, decreased available soil P by 0.70 g g–1 and Po by 22.82 g g–1. In this study, tithonia acquired Po that was unavailable to maize. However, it is apparent that continuous cutting and removal of biomass would lead to rapid depletion of P stored in organic forms.  相似文献

Effect of fruiting on carbon budgets of apple tree canopies   总被引:1,自引:0,他引:1  
Summary Carbon budgets were calculated from net photosynthesis and dark respiration measurements for canopies of field-grown, 3-year-old apple trees (Malus domestica Borkh.) with maximum leaf areas of 5.4 m2 in a temperature-controlled Perspex tree chamber, measured in situ over 2 years (July 1988 to October 1990) by computerized infrared gas analysis using a dedicated interface and software. Net photosynthesis (Pn) and carbon assimilation per leaf area peaked at respectively 8.3 and 7.7 mol CO2 m–2 s–1 in April. Net photosynthesis (Pn) and dark respiration (Rd) per tree peaked at 3.6 g CO2 tree–1 h–1 (Pn) and 1.2 g CO2 tree–1 h–1 (Rd), equivalent to 4.2 mol CO2 (Pn) and 1.4 mol CO2 (Rd) m–2 s–1 with maximum carbon gain per tree in August and maximum dark respiration per tree in October 1988 and 1989. In May 1990, a tree was deblossomed. Pn (per tree) of the fruiting apple tree canopy exceeded that of the non-fruiting tree by 2–2.5 fold from June to August 1990, attributed to reduced photorespiration (RI), and resulting in a 2-fold carbon gain of the fruiting over the non-fruiting tree. Dark respiration of the fruiting tree canopy progressively exceeded, with increasing sink strength of the fruit, by 51% (June–August), 1.4-fold (September) and 2-fold (October) that of the non-fruiting tree due to leaf (i. e. not fruit) respiration to provide energy (a) to produce and maintain the fruit on the tree and (b) thereafter to facilitate the later carbohydrate translocation into the woody perennial parts of the tree. The fruiting tree reached its optium carbon budget 2–4 weeks earlier (August) then the non-fruiting tree (September 1990). In the winter, the trunk respired 2–100 g CO2 month–1 tree–1. These data represent the first long-term examination of the effect of fruiting without fruit removal which shows increased dark respiration and with the increase progressing as the fruit developed.  相似文献

The influence of different light qualities on the photosynthetic rate, dark respiration, intracellular carbon and nitrogen content, and accumulation of photosynthetic pigments and cell-wall polysaccharides during short-term incubation (5 h) of the red algaGelidium sesquipedale was investigated. The same photon irradiance of 50mol m–2 s–2 below the light saturation point of photosynthesis was applied in each case. Blue light stimulated photosynthesis, dark respiration and the accumulation of chlorophyll and biliproteins, phycoerythrin in particular. The accumulation of internal carbon and nitrogen was greater under blue light than under the other light qualities. In contrast, the percentage of cell-wall polysaccharides was higher in red light. The content of cell-wall polysaccharides decreased during the time of incubation in all light treatments except in red light. The action of a non-photosynthetic photoreceptor in the control of cell-wall polysaccharide synthesis is suggested because the accumulation of cell-wall polysaccharides was not correlated with net photosynthesis in contrast to what occurred with carbon, chlorophyll and phycoerythrin accumulation.  相似文献

Oscillatoria amphigranulata is a fast-growing (3 doublings/day) cyanobacterium isolated from sulfide hot springs in New Zealand. Photosynthesis, as measured by incorporation of [14C]-HCO 3 - , was initially inhibited by 0.3–1.5 mM sulfide at pH 7.9–8.1. However, conversion to sulfide-dependent anoxygenic photosynthesis occurred in about 2 h or less under light intensities of 3–14 klx. Under the stimulation of higher light intensity (8–14 klx) a partial recovery of oxygenic photosynthesis also occurred. It was concluded that oxygenic photosynthesis was responsible for 21–42% of the total incorporation at sulfide concentrations of 1.0–0.3 mM, respectively. This contribution was suppressed at 1.5 mM sulfide and not elicited under lower light intensities (3–7 klx). As judged by the inhibitory effect of 10 g/ml chloramphenicol protein synthesis was required for attainment of both anoxygenic photosynthesis and photosystem II recovery. Sulfide could not be replaced by thiosulfate, elemental sulfur or dithionite as electron donors in photosynthesis, but elemental sulfur could serve as the sole assimilatory source of sulfur. Morphological and physiological measurements on individual leaves of Leucaena leucocephala seedlings were used to study acclimation to neutral shading. The light-saturated photosynthetic rate (Pn max) ranged from 19.6 to 6.5 mol CO2 m–2 s–1 as photosynthetic photon flux density (PPFD) during growth decreased from 27 to 1.6 mol m–2 s–1. Stomatal density varied from 144 mm–2 in plants grown in high PPFD to 84 mm–2 in plants grown in low PPFD. Average maximal stomatal conductance for H2O was 1.1 in plants grown in high PPFD and 0.3 for plants grown in low PPFD. Plants grown in low PPFD had a greater total chlorophyll content than plants grown in high PPFD (7.2 vs 2.9 mg g–1 on a unit fresh weight basis, and 4.3 vs 3.7 mg dm–2 on a unit leaf area basis). Leaf area was largest when plants were grown under the intermediate PPFDs. Leaf density thickness was largest when plants were grown under the largest PPFDs. This study examines the influence of current velocity in the toxiceffect of copper in diatom-dominated biofilms grown in artificial channels.Effects on community structure, algal biomass and photosynthesis (carbonincorporation) caused by 15 g L–1 of copperwere tested at contrasting (1 and 15 cm s–1)velocities. Moreover, a possible threshold on the effect of copper on algalbiomass and photosynthesis related to current velocity was examined by usingprogressively increasing current velocity (1 to 50 cms–1) at 15 g L–1 Cu.Chlorophyll-a decreased ca. 50% as a result of addition of15 g L–1 Cu. Chlorophyll decrease occurredearlier at 15 cm s–1 than at 1 cms–1 when adding 15 g L–1Cu. Copper also caused a remarkable decrease in carbon incorporation(from 30 to ca. 50%), which was produced earlier at 15 cms–1 (three days) than at 1 cms–1 (seven days). Some taxa were affected by thecombination of copper and current velocity. Both Achnanthesminutissima and Stigeoclonium tenue becomedominant at 15 cm s–1 in the presence of copper.Significant inhibition of algal growth in 15 g L–1Cu occurred at low (1 cm s–1) and highvelocities (50 cm s–1), but not at intermediatevelocity (20 cm s–1). The experiments indicatethat current velocity triggers the effect that copper has on diatom-dominatedbiofilms, and that the effect is more remarkable at low and high than atintermediate current velocities.  相似文献

It has been proposed that many marine macroalgae are able to utilize HCO 3 for photosynthesis and growth, and that energy-dependent ion pumping is involved in this process. We have therefore studied the light-dependent alkalization of the surrounding medium by two species of marine macroscopic brown algae,Fucus serratus L. andLaminaria saccharina (L.) Lamour. with the aim of investigating the role of extracellular carbonic anhydrase (EC 4.2.1.1.) in the assimilation of inorganic carbon from the seawater medium. In particular, the influence of membrane-impermeable or slowly permeable carbonic-anhydrase inhibitors on the rate of alkalization of the seawater has been investigated. Inhibition of the alkalization rate occurred in both species at an alkaline pH (pH 8.0) but no inhibition was observed at an acidic pH (pH 6.0). The alkalization was found to be light-dependent and inhibited by 3-(3,4-dichlorophenyl)-1, 1-dimethylurea and, thus, correlated with photosynthesis. Alkalization by macroalgae has previously been shown to be proportional to inorganiccarbon uptake. We suggest that alkalization of the medium at alkaline pH in both of the species examined is mainly the consequence of an extracellular reaction. The reaction is catalyzed by extracellular carbonic anhydrase which converts HCO 3 to OH and CO2; CO2 is then taken up through the plasmalemma. Using primary cultures of gill pavement cells from freshwater rainbow trout, a method is described for achieving confluent monolayers of the cells on glass coverslips. A continuous record of intracellular pH was obtained by loading the cells with the pH-sensitive flourescent dye 2,7-bis(2-carboxyethyl)-5(6)-carboxyfluorescein and mounting the coverslips in the flowthrough cuvette of a spectrofluorimeter. Experiments were performed in HEPES-buffered media nominally free of HCO3. Resting intracellular pH (7.43 at extracellular pH=7.70) was insensitive to the removal of Cl or the application of 4-acetamido-4-isothiocyanatostilbene-2,2-disulfonic acid (0.1 mmol·l–1), but fell by about 0.3 units when Na+ was removed or in the presence of amiloride (0.2 mmol·l–1). Exposure to elevated ammonia (ammonia prepulse; 30 mmol·l–1 as NH4Cl for 6–9 min) produced an increase in intracellular pH (to about 8.1) followed by a slow decay, and washout of the pulse caused intracellular pH to fall to about 6.5. Intracellular non-HCO 3 buffer capacity was about 13.4 slykes. Rapid recovery of intracellular pH from intracellular acidosis induced by ammonia prepulse was inhibited more than 80% in Na+-free conditions or in the presence of amiloride (0.2 mmol·l–1). Neither bafilomycin A1 (3 mol·l–1) nor Cl removal altered the intracellular pH recovery rate. The K m for Na+ of the intracellular pH recovery mechanism was 8.3 mmol·l–1, and the rate constant at V max was 0.008·s–1 (equivalent to 5.60 mmol H+·l–1 cell water·min–1), which was achieved at external Na+ levels from 25 to 140 mmol·l–1. Summary The response of net photosynthesis to temperature, moisture, and light was examined in thalli of the tropical basidiomycete lichen Cora pavonia from recent lahar flows on the volcanic summit La Soufrière (Guadeloupe, French West Indies). Although thalli of C. pavonia are typically exposed to only low light intensities and isothermal temperature conditions under prevailing cloud/shroud conditions on La Soufrière, their photosynthetic response matrix reveals an unexpected breadth of response. The temperature optimum of net photosynthetic uptake in C. pavonia rises from 6°C at a photon flux area density of 25 mol m–2 s–1 PAR to 27°C at 1000 mol m–2 s–1 PAR, with rates of maximal net photosynthetic uptake exceeding 25 mg CO2 g–1 h–1. Net photosynthesis was optimal at thallus moisture contents of 250 to 350 percent water content by weight, declining only slightly in fully saturated thalli. These response patterns pose an apparent paradox, as on most days they will act to severely restrict net photosynthetic uptake by thalli of C. pavonia on La Soufrière. This paradox is discussed in context of those selective pressures faced by lichen thalli in later successional stages as well as those imposed by brief periods of atypical weather conditions.  相似文献
