Rheum tanguticum (Rh. tanguticum) is a Chinese medicinal plant traditionally used in the treatment of constipation. As a byproduct, the seeds of this plant are rich in nutrients and phytochemicals. This study aimed to determine and assess seed germination ability, seed physical characteristics, soluble protein content, chemical constituents and antioxidant capacity from different breeding lines, to promote the development and utilization of seed resources. Significant differences were observed for the soluble protein content and antioxidant assays among the ten lines. The contents of aloe-emodin, rhein and catechins accumulated in seeds were extremely low and significantly different from those in roots. In contrast, emodin and chrysophanol were abundant in seeds, and significant differences were observed between seeds and roots. It was found that associations between gallic acid and catechins were not significant for either soluble protein or antioxidant capacity. There was a significantly positive correlation between the contents of four anthraquinones (aloe-emodin, rhein, emodin and chrysophanol) and soluble protein. Seeds have potent antioxidative capacity and relatively high levels of soluble protein content. The rich chemical composition of seeds can be widely used in the medical industry for further development.  相似文献

Examining variation in pollinator effectiveness and seed production resulting from single pollinator visits can provide a deeper understanding of how pollinators may influence reproduction in plant populations. When comparing populations, differences in the number of seeds produced from single pollinator visits to flowers may not always be attributable to differences in pollen deposition, but rather to differences in plant fecundity or resource availability. Pollinator effectiveness and seed production were studied for two populations over a 4-year period and were measured using single bee visit manipulations of flowers. No significant difference in pollinator effectiveness (pollen deposited on stigmas) was observed between the two populations. However, a significant difference between the two populations was observed in the number of seeds produced per flower. The Wellhouse population produced approximately three times as many seeds/flower from a single pollinator visit as did the Firefly Meadow population. Within each population, pollinators (Bombus pennsylvanicus and Apis mellifera) did not differ in the number of pollen grains deposited on stigmas or seeds produced per flower from single visit experiments. Differences in plant density, pollen viability, and ovules per flower also could not account for a significant amount of the variation. A resource augmentation experiment (water and fertilizer application) resulted in a decrease in seeds per flower per bee visit for the water treatment at the Wellhouse population only. For both populations, pollen deposition, pollen viability, and ovules per flower were unaffected by the resource augmentation. Alternative possibilities for the observed differences in seeds per flower per bee visit are discussed.  相似文献

The regulation of seed oil synthesis in rapeseed is largely unknown. In this study, we compared the gene expression during seed development between two lines of Brassica napus with a 10% difference in oil content. We isolated the immature seeds 15 and 25 days after flowering at periods preceding and including the major accumulation of storage oils and proteins. The differentially expressed gene clones between the two rape lines were isolated by subtractive suppression hybridization (SSH). All SSH clones were arrayed and screened by dot blot hybridization, followed by RT-PCR analysis for selected clones. A total of 217 cDNA clones corresponding to 30 genes were found to have a high expression in seeds with high oil content. Six genes were highly expressed in seeds with low oil content. Northern blot and enzyme activity analysis demonstrated a change in expression pattern of several genes. The results provide information on gene-encoding factors responsible for the regulation of oil synthesis. The possible role of these genes in seeds is discussed. The genes in this study may be suitable as novel targets for genetic improvement of seed oil content and may also provide molecular markers for studies of rape breeding.  相似文献

The directed dispersal hypothesis has two components: (1) non-random seed deposition by a predicted vector, which removes greater amounts of seeds to specific sites, and (2) higher seed survival and seedling establishment in these specific sites. Several studies suggest that ants perform both tasks. This study was designed to cover the processes from post-dispersed seeds to established juvenile plants of a typical ant-dispersed species. Our main objective was to determine whether Ricinus communis benefits from directed dispersal by ants to maintain its populations in previously colonized habitats. We examined whether there were differences between ant nest pile mounds and their vicinities in the: (1) densities of seeds with and without elaiosome, seedlings and juveniles; (2) performance of post-dispersed seeds (without elaiosome), which may be affected by seed density, a key feature attracting seed predators; (3) nutrient quantities; (4) number of germinated seeds and juvenile biomass measurements; and (5) ant protection of seedlings from herbivores. There were more seeds without elaiosome, seedlings and juveniles in pile mounds, and seeds with elaiosome were equally distributed. There was no difference in the number of non-removed seeds in pile mounds and in their vicinities, and there was no tendency for this difference to increase or decrease with time or with initial seed density. Apparently, there was no difference in nutrient contents in soils of pile mounds and their vicinities. Likewise, there was no difference in the number of seeds germinated and in the biomass measurements of juveniles in both soils. Ants did not provide differential protection for seedlings in pile mounds against potential herbivores. The dispersal of Ricinus seeds by ants had a marked effect on the distribution pattern of the seeds, seedlings and juveniles of this species. However, there were no additional advantages for the seeds, seedlings and juveniles in pile mounds and, therefore, Ricinus does not benefit from directed dispersal by ants to maintain its populations in the study sites.  相似文献

The ability of genetic manipulation to yield greatly increased concentrations of free amino acids (FAAs) in seeds of soybean was evaluated by introduction of a feedback-insensitive mutant enzyme of tryptophan (Trp) biosynthesis into two transformation-competent breeding lines deficient in major seed storage proteins. The storage protein-deficient lines exhibited increased accumulation of certain other seed proteins as well as of FAAs including arginine (Arg) and asparagine in mature seeds. Introduction of the gene for a feedback-insensitive mutant of an α subunit of rice anthranilate synthase (OASA1D) into the two high-FAA breeding lines by particle bombardment resulted in a >10-fold increase in the level of free Trp in mature seeds compared with that in nontransgenic seeds. The amount of free Trp in these transgenic seeds was similar to that in OASA1D transgenic seeds of the wild-type cultivar Jack. The composition of total amino acids in seeds of the high-FAA breeding lines remained largely unaffected by the expression of OASA1D with the exception of an increase in the total Trp content. Our results therefore indicate that the extra nitrogen resource originating from storage protein deficiency was used exclusively for the synthesis of inherent alternative nitrogen reservoirs such as free Arg and not for deregulated Trp biosynthesis conferred by OASA1D. The intrinsic null mutations responsible for storage protein deficiency and the OASA1D transgene affecting Trp content were thus successfully combined and showed additive effects on the amino acid composition of soybean seeds.  相似文献

Summary This experiment investigated the effect of parental nutrient shortage on the allocation of five nutrients to seeds and rhizomes in Sorghum halepense, a perennial, noxious weed, and to seeds in Sorghum bicolor, an annual, cultivated species. Plants from both species were grown from seeds and supplied with fertilizer at three concentrations. The allocation of biomass and nutrients (N, P, K, Ca and Mg) to reproductive and vegetative parts was determined. Relative biomass allocation to reproduction (either sexual or vegetative) remained constant in S. halepense in spite of large differences in total plant weight. In S. bicolor, however, biomass allocation to sexual reproductive structures decreased significantly with decreasing nutrient supply. Individual seed weight was not modified by parental nutrient supply in S. halepense, but it increased with decreasing nutrient availability in S. bicolor. Important differences in mineral allocation to seeds were found between the two species. While S. bicolor seeds were largely buffered from the differences in parental nutrient status, concentration of nutrients in S. halepense seeds decreased significantly with decreasing supply for all the nutrients analyzed except Ca. However, mineral nutrient concentration in S. halepense rhizomes remained remarkably constant despite differences in the external supply, evincing the priority given to vegetative reproduction at the expense of sexual reproduction. Overall, the pattern of nutrient allocation in S. bicolor seeds under different nutrient supply resembled the pattern observed in S. halepense rhizomes, but it had little resemblance to the pattern of nutrient allocation in S. halepense seeds. The results are discussed in terms of differences and similarities in the reproductive strategy of these two species.  相似文献

Enzymatic control of the accumulation of verbascose in pea seeds   总被引:4,自引:0,他引:4  
Verbascose, the pentasaccharide of the raffinose family of oligosaccharides, consists of galactose units joined to sucrose. In pea (Pisum sativum) seeds, the content of verbascose is highly variable. In a previous study on a high‐verbascose pea cultivar, the present authors have demonstrated that verbascose is synthesized by a multifunctional stachyose synthase (EC 2.4.1.67), which utilizes raffinose as well as stachyose as a galactosyl acceptor. Herein the results of a study of the cloning and functional expression of stachyose synthase from the low‐verbascose genotype SD1 are reported and it is demonstrated that this line contains a protein with a reduced ability to synthesize verbascose. Analysis of seeds from seven pea lines revealed a positive correlation between verbascose synthase activity and verbascose content. Among these genotypes, only the SD1 line showed low verbascose synthase activity when the data were normalized to stachyose synthase activity. These results suggest that differences in the level of verbascose synthase activity could be caused by mutations in the stachyose synthase gene as well as by variation in the amount of the protein. The lines were also analysed for activity of α‐galactosidase, a catabolic enzyme that could limit the extent of verbascose accumulation. No relationship was found between α‐galactosidase activity and the amount of raffinose family oligosaccharides.  相似文献

 Arcelin seed proteins of common bean (Phaseolus vulgaris L.) confer resistance to bruchid pests, and in vitro results suggested that greater resistance could be achieved by increasing the concentration of arcelin. We created backcross lines having arcelin alleles (SMARC lines), or alleles for the related protein phytohemagglutinin (PHA) (SMPHA lines), and a null allele for phaseolin to determine if seeds lacking phaseolin would contain increased quantities of arcelin or PHA proteins. To test the affects of genetically removing phaseolin, SMARC and SMPHA lines were derived as pairs of phaseolin-containing and phaseolin-null lines. Parental, SMARC, and SMPHA lines were grown in a replicated greenhouse trial and measured for days-to-flower, days-to-maturity, seed weight, and for quantities of phaseolin, arcelin dimer, PHA, and total proteins. There were no differences between pairs of phaseolin and phaseolin-null lines for days-to-flower, seed weight or total protein, and inconsistent differences for days-to-maturity. Arcelin concentrations were significantly increased in two of four pairs of SMARC lines, and PHA concentration was significantly greater in four of five pairs of SMPHA lines. To answer the question whether iron accumulation in transgenic rice seeds depends on the expression level of exogenous soybean ferritin, we generated two kinds of ferritin hyper-expressing rice lines by introducing soybean ferritin SoyferH-1 gene under the control of the rice seed storage glutelin gene promoter, GluB-1 and the rice seed storage globulin gene promoter, Glb-1, (GluB-1/SoyferH-1 and Glb-1/SoyferH-1, DF lines), and by introducing the SoyferH-1 gene under the control of Glb-1 promoter alone (Glb-1/SoyferH-1, OF lines). Ferritin expression was restricted to the endosperm in both lines and protein levels determined by western blot analysis were up to 13-fold higher than in a construct previously reported FK22 (GluB-1/SoyferH-1, in genetically Kitaake bachground); however, the maximum iron concentrations in seeds of both of the new lines were only about 30% higher than FK22. The maximum iron concentration in the OF and DF lines was about threefold higher than in the non-transformant. The mean Fe concentration in leaves of ferritin over-expressing lines decreased to less than half of the non-transformant while that the plant biomasses and seed yields of the ferritin-transformed lines were not significantly different from those of the non-transformant, suggesting that accumulation of Fe in seeds of hyper-expression ferritin rice did not always depend on the expression level of exogenous ferritin but may have been limited by Fe uptake and transport. No obvious differences were observed for other divalent-metal concentrations (Ca, Cd, Cu, Mg, Mn and Zn) in the seeds among all experimental lines and non-transformant.  相似文献

We examined, by two-dimensional polyacrylamide gel electrophoresis (2D-PAGE), near-isogenic lines of the r-gene in pea (Pisum sativum) which determines round (RR) vs. wrinkled (rr) seed. The study was undertaken to assess the number of protein changes resulting from a single gene substitution as a means of quantifying pleiotropic effects. A total of 636 to 770 resolvable polypeptides were identical in all respects between RR and rr for roots, shoots, leaflets, stipules, young ovaries, and young embryos. A single difference between the lines became evident about 21-23 days after anthesis in the embryos. Mature seeds of the two lines showed 62 spot differences in addition to differences in four clusters of spots, representing about 10% of the total number of spots visible on the gels. The protein differences are presumably involved in the many known physiological differences of the two seed types. 2-D PAGE analyses of near-isogenic lines are likely to be valuable in a number of quantitative and developmental genetic contexts.  相似文献

The seed deposition pattern created by a seed disperser is one of the components of the efficiency of a species as seed disperser, and ultimately may influence the recruitment of a plant species. In this study, we used the seeds of a bird-dispersed forest palm, Euterpe edulis, to investigate the effects of two distinct seed deposition patterns created by birds that defecate (clumped pattern) and regurgitate seeds (loose-clumped pattern) on the survival of seeds experimentally set in an E. edulis-rich site, and of seedlings grown under shade-house conditions. The study was conducted in the lowland forest of Parque Estadual Intervales, SE Brazil. Clumped and loose-clumped seeds were equally preyed upon by rodents and insects. Although clumped and isolated seedlings had the same root weight after 1 year, the isolated seedlings survived better and presented more developed shoots, suggesting intraspecific competition among clumped seedlings. Our results indicate that animals that deposit E. edulis seeds in faecal clumps (e.g. cracids, tapirs) are less efficient seed dispersers than those that regurgitate seeds individually (e.g. trogons, toucans). Intraspecific competition among seedlings growing from faecal clumps is a likely process preventing the occurrence of clumps of adult palms.  相似文献

Seed‐specific expression is an appealing alternative technology for the production of recombinant proteins in transgenic plants. Whereas attractive yields of recombinant proteins have been achieved by this method, little attention has been paid to the intracellular deposition and the quality of such products. Here, we demonstrate a comparative study of two antiviral monoclonal antibodies (mAbs) (HA78 against Hepatitis A virus; 2G12 against HIV) expressed in seeds of Arabidopsis wild‐type (wt) plants and glycosylation mutants lacking plant specific N‐glycan residues. We demonstrate that 2G12 is produced with complex N‐glycans at great uniformity in the wt as well as in the glycosylation mutant, carrying a single dominant glycosylation species, GnGnXF and GnGn, respectively. HA78 in contrast, contains additionally to complex N‐glycans significant amounts of oligo‐mannosidic structures, which are typical for endoplasmic reticulum (ER)‐retained proteins. A detailed subcellular localization study demonstrated the deposition of both antibodies virtually exclusively in the extracellular space, illustrating their efficient secretion. In addition, although a KDEL‐tagged version of 2G12 exhibited an ER‐typical N‐glycosylation pattern, it was surprisingly detected in protein storage vacuoles. The different antibody variants showed different levels of degradation with hardly any degradation products detectable for HA78 carrying GnGnXF glycans. Finally, we demonstrate functional integrity of the HA78 and 2G12 glycoforms using viral inhibition assays. Our data therefore demonstrate the usability of transgenic seeds for the generation of mAbs with a controlled N‐glycosylation pattern, thus expanding the possibilities for the production of optimally glycosylated proteins with enhanced biological activities for the use as human therapeutics.  相似文献

The fatty acid composition and content of membrane and storage lipids of two transgenic laurate-producing rape (Brassica napus L.) lines were monitored during seed development. The two lines, the medium-laurate (ML) line and the high-laurate (HL) line, accumulated 34 mol% and 55 mol% of laurate in their seed triacylglycerols, respectively. The diacylglycerols contained about 17 and 33 mol% of laurate in the ML- and HL-lines, respectively, from the mid-stage of seed development up to seed maturity. The ML-line showed a maximal relative laurate content in phosphatidylcholine (17 mol%) at the mid-stage of seed development whereafter the content decreased to 2.7 mol% with seed maturity. The laurate content in phosphatidylcholine was observed to remain high (26 mol%) in the HL-line from the mid-stage to the end of triacylglycerol deposition. Thereafter, the relative content decreased and reached 6.6 mol% in the mature seeds. There was an enhanced activity of lauroyl-phosphatidylcholine- metabolizing enzymes in the seed membranes from laurate-producing lines compared with control lines, which might explain the decrease seen in laurate content in phosphatidylcholine during seed maturation. A comparison of the laurate distribution in the lipids from developing laurate-producing rape seeds and developing seeds from three species naturally accumulating laurate at similar levels revealed differences in laurate metabolism compared with these species. The results suggest that phospholipids and triacylglycerols are synthesized from the same diacylglycerol pool in rape seeds and that rape lysophosphatidic acid acyltransferase and diacylglycerol acyltransferase do not have the same preference for laurate substrates as the corresponding enzymes in seed tissues naturally accumulating this acyl group. In addition, the mechanisms that specifically remove or exclude laurate from membrane lipids appear less effective in rape seed than in tissues naturally evolved to synthesize laurate-rich oils. The tropical rain forest landscape has been transformed to a mosaic composed of patches of crops, secondary vegetation and remnant forest fragments of different shapes and sizes. Isolation of patches and fragments is a critical issue in the maintenance of local species diversity. In this study we focus on the dispersal of propagules by birds to understant the movement of plants between landscape components. Seed deposition and the behavior of frugivorous birds were monitored at four isolated fig trees (Ficus yoponensis and F. aurea) in man-made pastures. Seed deposition was measured by trapping seeds under canopy trees for six months and by direct observation of bird visits to the four trees for one year. Seed deposition densities were 465, 614, 632 and 1097 seeds/m2 accumulated over six months under each of the four trees. We recorded 8268 seeds of 107 species under the trees, among them, 6726 seeds (81%) were of 56 species dispersed by vertebrate frugivores. Seeds of tree species accounted for 26% of the total species. Seventy-three species of birds perched in the observed trees, and 3344 visits were made by 47 species of frugivores. Frugivorous birds occurred in two groups: resident species nesting in the pastures and resident species nesting elsewhere. Propagule exchange between landscape components is clearly influenced by the behavior of these two groups. Structure and dynamics of the landscape depend on plant species availability within the mosaic. This availability is high and suggests possibilities for the management of the local species diversity of tropical rain forests.  相似文献

The probability that a seed will germinate depends on factors associated with genotype, maturation environment, post-maturation history, and germination environment. In this study, we examined the interaction among these sets of factors for 18 inbred lines from six populations of Bromus tectorum L., a winter annual grass that is an important weed in the semi-arid western United States. Seeds of this species are at least conditionally dormant at dispersal and become germinable through dry-afterripening under summer conditions. Populations and inbred lines of B. tectorum possess contrasting dormancy patterns. Seeds of each inbred line were produced in a greenhouse under one of three levels of maturation water stress, then subjected to immediate incubation under five incubation regimes or to dry storage at 20°C for 4 weeks, 12 weeks, or 1 year. Dry-stored seeds were subsequently placed in incubation at 20/30°C. Narrow-sense heritability estimates based on parent-offspring regressions for germination percentage of recently harvested seeds at each incubation temperature were high (0.518–0.993). Germination percentage increased with increasing water stress overall, but there were strong interactions with inbred line and incubation temperature. Inbred lines whose seeds were non-dormant over the full range of incubation temperatures when produced at low maturation water stress showed reaction norms characterized by little or no change as a function of increasing stress. For inbred lines whose dormancy status varied with incubation temperature, incubation treatments where seeds exhibited either very low or very high levels of dormancy showed the least change in response to maturation water stress. Inbred lines also varied in their pattern of dormancy loss during storage at 20°C, but maturation water stress had only a minor effect on this pattern. For fully afterripened seeds (1 year in storage at 20°C), inbred line and maturation water stress effects were no longer evident, indicating that differences in genotype and maturation environment function mainly to regulate dormancy and dormancy loss in B. tectorum, rather than to mediate response patterns of non-dormant seeds.  相似文献

 In order to investigate the relationship between the amino acid concentration in the phloem sap of leaves and the protein content in seeds, two Brassica napus genotypes and one B. carinata genotype with low, medium and high seed protein contents were analyzed. Phloem sap was collected from the B. napus winter rapeseed breeding line DSV15 with 19% protein of dry weight in the seeds, the spring cultivar ‘Duplo’ with 25% protein in the seeds and from the B. carinata line BRA1151/90 with 39% protein in the seeds by using the aphid-stylet technique. The total amino acid contents measured in the phloem varied considerably among the three genotypes analysed, and correlated positively with their respective seed protein contents. The total amino acid-to-sucrose ratio was lowest in B. napus line DSV15 which had the lowest seed protein content and highest in the B. carinata line BRA1151/90 which had the highest seed protein content. The amino-N translocation in the phloem during the light period was about 2-fold higher in the B. carinata line BRA1151/90 than in the B. napus lines Dulpo and DSV15. Predominant amino acids in the phloem were glutamine and glutamate, followed by serine, aspartate, and threonine. The amino acid patterns in the leaves resembled those in the phloem, although their absolute concentrations were higher in the phloem than in the cytosol of mesophyll tissue. Furthermore, the concentration gradient of amino acids between the cytosol of mesophyll cells and the phloem was higher in the B. carinata line BRA1151/90 than in the B. napus lines Duplo and DSV15. These results lead to the conclusion that the phloem translocation of amino-N and the phloem loading process of amino acids are decisive factors for the protein content in the seeds of Brassica species. More frequent deposition of seeds by frugivores beneath plants in fruit could impose spatial limits to the distribution of plants dispersed by animals and contribute to species coexistence. Also, differences in diet and use of microhabitats by seed dispersers could promote spatial variation in the combination of seed species deposited. We investigated patterns of seed deposition of Miconia fosteri and Miconia serrulata (Melastomataceae) by birds in the Amazon. The goal was to determine how distribution and abundance of fruiting plants, both con‐ and hetero‐specifics, affect the spatial variability in clumping and composition of multi‐specific seed deposition. We established two 9‐ha plots in undisturbed terra‐firme understory in the Ecuadorian Amazon. Seed rain was sampled with seed traps located in four microsites: below plants of the focal species, below Anthurium eminens (Araceae), and in randomly selected microsites. We examined seed deposition in these microsites in relation to habitat, fruiting neighborhood (fruit abundance, and distance to and density of plants of the target species), and crop size of M. fosteri or M. serrulata to determine if microsites differed in abundance and species composition of seeds. Seed traps below plants in fruit received more seeds than did randomly located traps. Seeds of the target species were, moreover, more commonly deposited below con‐ rather than hetero‐specific plants. Seed aggregation below fruiting plants increased in forest neighborhoods where the abundance of fruits and the combination of fruiting plant species contributed to the arrival of seeds. Microsites differed notably in the combination of seeds deposited by frugivores, and differences were less pronounced among microsites that received seeds of M. fosteri and M. serrulata than among all microsites where at least some seed species were deposited by birds. We demonstrate that two closely related, ecologically similar species possess many similarities in their patterns of seed deposition and in the factors that affect those patterns. The combination of seed species deposited below foci of dispersal depended on the fruiting plant species, and the spatial patterns of seed deposition varied with the location of the microsite and the combination of co‐dispersed species in the neighborhood. Similar species that share the same dispersers were confronted with different combinations of seeds depending on the microsite where they arrived, which could promote forest heterogeneity in the combination of plant species.  相似文献

1. Surface water is an important dispersal vector for wetland plant species. However, most previous studies on hydrochory (i.e. water dispersal) have focused on ecosystems with relatively rapid water flow. Therefore, there is a need to study such dispersal in slow‐flowing or stagnant waterbodies, such as drainage ditches, which might act as dispersal corridors between habitat patches. 2. To gain insight into the mechanisms by which seeds are transported in drainage ditches, the effect of the velocity of wind and water on the rate of transport of floating seeds of three wetland species (Carex pseudocyperus L., Iris pseudacorus L. and Sparganium erectum L.) was investigated. Furthermore, in release and retrace experiments with painted C. pseudocyperus seeds, a number of factors potentially determining the probability of seed deposition were investigated. 3. Net wind speed was found to be the main factor determining the rate at which seeds are transported in drainage ditches. No relation between water flow at middepth in the ditches and seed transport was found. Wind speed and flow at the water surface were positively related. The effect of wind speed on the rate of transport of floating seeds was greater for S. erectum seeds, because a greater ratio of their volume protrudes from the water, than for C. pseudocyperus and I. pseudacorus seeds. 4. The principal factors that determine seed deposition were aquatic plant cover, ditch slope and indentations in the ditch bank. Seeds changed direction if the wind direction changed, or if there was a bend in the ditch. The final pattern of deposition was related to mean net wind speed. Mean transport distance after 2 days varied between 34 and 451 m. 5. Unlike in rivers, seed transport in ditches was determined by wind speed and direction, enabling multidirectional seed dispersal. We conclude that in slow‐flowing waters, wind is a more important driver for hydrochorous seed transport than the flow of water. This sheds a new light on hydrochory and has important consequences for the management of otherwise fragmented wetland remnants.  相似文献

Summary The seed lectin of the tree legume,Bauhinia purpurea alba, was localized by electron microscopic immunocytochemistry. The pattern of lectin deposition and site of intracellular localization was examined in mid- to late-maturation seeds. The seed tissue was embedded in Lowicryl K4M, the use of which with seed tissues is discussed. Immunocytochemical labeling was accomplished with colloidal gold coupled to a second antibody. The immunocytochemical reaction was specific and sensitive. Protein bodies, Golgi apparatus and Golgi secretion vesicles were densely labeled. Golgi apparatus was oriented such that Golgi secretion vesicles were in close proximity to the protein bodies. The entire Golgi apparatus was labeled with no concentration gradient across the Golgi stack. These observations indicate that the final site of lectin deposition is the protein body, and that the Golgi apparatus plays an essential role in the deposition process.  相似文献
