摘要目的:探讨新疆地区维吾尔族、汉族脑啡肽酶(Neprilysin,NEP)基因单核苷酸多态性与阿尔茨海默病(Alzheimer’SdiseaseAD)的关系。方法:对新疆地区维吾尔族、汉族≥50岁8284名人群进行AD流行病学调查,参照ADRDA.NINCDS的标准,选取散发性阿尔茨海默病(sporadicAlzheimer’s disease,SAD)患者209例(AD组)与正常对照220例(对照组),应用聚合酶链反应一限制性片段长度多态性技术(PCR)检测NEP基因多态性,采用病例一对照的关联分析方法对NEP基因rs3736187位点进行基因型和等位基因频率分析。结果:(1)新疆维吾尔族、汉族两民族AD组与对照组间NEP基因基因型频率和等位基因频率分布差异均有统计学意义(P〈0.05)。携带T等位基因个体出现AD的危险性高于携带c等位基因的个体(0R=1.981,P〈0.05)。(2)新疆维吾尔族、汉族不同民族之间比较NEP基因基因型频率和等位基因频率分布差异均无统计学意义(P〉0.05),而在同一民族中AD组和对照组之间比较NEP基因等位基因频率分布差异均具有统计学意义(P〈0.05)。(3)两个年龄分段(〈65岁及≥65岁)之间NEP基因基因型频率和等位基因频率分布差异均无统计学意义(P〉0.05),而在同一年龄段内部AD组与对照组间等位基因频率分布差异具有统计学意义(P〈0.05)。(4)男性、女性之间NEP基因基因型频率和等位基因频率分布差异均无统计学意义,而在女性AD组与对照组间等位基因频率分布差异有统计学意义(P〈0.05)。结论:NEP基因rs3736187位点基因型频率和等位基因频率在新疆维吾尔族、汉族两民族间的分布相似;NEP基因的T等位基因是SAD的危险因素,在新疆维吾尔族、汉族两民族及女性SAD的发病中起重要作用。  相似文献

为探讨新疆克拉玛依市维吾尔族与汉族成年人体部体质特征。采用整群分层随机抽样原则,按照《国民体质测试指导站各指标的测试方法及判断标准》规定的方法,测量并获得新疆克拉玛依市维吾尔族、汉族20~59岁共4108个成年人的有效样本,并按Martin法对所测体部项目进行体质特征研究。新疆克拉玛依市维吾尔族:除肩型男性为中肩型、女性为窄肩型外,男女均为超中等身材、宽胸型、宽骨盆型、窄手型、矮胖型。新疆克拉玛依市汉族:除身材分型男性为高型、女性为超中等身材外,男女均为宽胸型、窄肩型、宽骨盆型、窄手型、中间胖瘦型。维吾尔族与汉族之间体质特征总体水平差异具有统计学意义(p<0.01),且差异主要存在于维吾尔族与汉族女性之间。克拉玛依市维吾尔族和汉族成年人体质特征均具有北方人群的体质特征。  相似文献

本文报告了中国北京、天津、济南、哈尔滨、沈阳、长春、上海、南京、杭州、武汉、广州及成都等12个城市汉族7—18岁儿童生长的长期趋势。 50年代至今,汉族儿童生长的长期趋势是明显的,30年来同年龄儿童身高每10年增长2.66(男)、2.40(女)厘米,体重每10年增长1.64(男)、1.14(女)公斤,近10年来的增长最为显著。生长突增高峰年龄大都提前1—2年。北京、上海、南京、广州、杭州等五所城市自30年代至今,生长长期趋势也是明显的,半个世纪来,每10年增长值,男童身高为1.12—2.66厘米,体重为0.56—1.29公斤,女童身高为1.42—2.67厘米,体重为0.65—1.18公斤。  相似文献

稻纵卷叶螟肠道细菌群落结构与多样性分析   总被引:1,自引:0,他引:1  
【目的】为明确水稻主要害虫稻纵卷叶螟Cnaphalocrocis medinalis(Guenée)幼虫肠道细菌的群落结构和多样性。【方法】利用Illumina Mi Seq技术对稻纵卷叶螟4龄幼虫肠道细菌的16S r DNA V3-V4变异区序列进行测序,应用USEARCH和QIIME软件整理和统计样品序列数和操作分类单元(operational taxonomic unit,OTU)数量,分析4龄幼虫肠道细菌的物种组成、丰度和多样性。【结果】稻纵卷叶螟4个数量不同的4龄幼虫样本(1,2,3和5头)共得165 386条reads,在97%相似度下可将其聚类为604个OTUs。总共注释到22个门,43个纲,82个目,142个科,204个属,244个种。其中在门水平上,主要优势菌为变形菌门(Proteobacteria)(相对丰度26%~34%)和放线菌门(Actinobacteria)(23%~32%);在纲水平上,主要优势菌为放线菌纲(Actinobacteria)(相对丰度23%~32%)、酸杆菌纲(Acidobacteria)(9%~11%)、α-变形菌纲(Alphaproteobacteria)(10%~13%)、β-变形菌纲(Betaproteobacteria)(6%~8%)和γ-变形菌纲(Gammaproteobacteria)(6%~12%);在科水平上,共有优势菌为类诺卡氏菌科(Nocardioidaceae)、丛毛单胞菌科(Comamonadaceae)、黄单胞菌科(Xanthomonadaceae)、鞘脂单胞菌科(Sphingomonadaceae)和芽单胞菌科(Gemmatimonadaceae)等。在属水平上,4个样本前5位优势属中,类诺卡氏属Nocardioides和鞘脂单胞菌Sphingomonas为共有优势属。稻纵卷叶螟肠道细菌Simpson指数、Shannon指数、Ace指数和Chao指数分别为0.16~0.65,0.94~3.22,212~488和210~490。【结论】稻纵卷叶螟幼虫肠道细菌多样性比较丰富,个体间微生物群落结构和多样性有差异。不同数量样本数据与总体数据有助于综合反映稻纵卷叶螟种群肠道微生物状况。本研究结果为进一步研究稻纵卷叶螟肠道微生物的功能及其在防治中的应用奠定了基础。  相似文献

目的探讨汉族与维吾尔族儿童封闭型不同龋敏感中变形链球菌乳酸脱氢酶活性与ldh基因多态性及龋病发生的关系。方法从前期实验分离的维吾尔族与汉族儿童口腔变形链球菌中筛选出:维吾尔族儿童封闭型口腔变形链球菌高龋组变形链球菌35株[龋失补牙数(dmft)≥5],无龋组变形链球菌37株[龋失补牙数(dmft)=0];27株表现乳酸脱氢酶高活性,24株低活性;汉族儿童口腔中分离的高龋和无龋变形链球菌各35株,高活性和低活性各25株,使用特异引物扩增细菌组DNA,获得结构基因ldh,进行聚合酶链反应-限制性内切酶多态性分析。结果维吾尔族儿童封闭型和汉族儿童MseⅠ酶切ldh-RFLP表现A、B两种基因型;而MnlⅠ、DdeⅠ、NlaⅢ和AluⅠ消化扩增产物未发现多态性。MseⅠ产生的B基因型菌株在维吾尔族儿童封闭型,汉族儿童高龋组的检出均高于无龋组(P0.05),A、B两种基因型在不同酶活性组的分布不同(P0.05),高酶活性组B基因型菌株的比例高于低酶活性组;A、B基因型在维吾尔族儿童封闭型和汉族高龋组、无龋组,高酶活性组和低酶活性组的检出比较差异无统计学意义(P0.05)。结论变形链球菌乳酸脱氢酶基因多态性在维吾尔族与汉族儿童并未表现出差异性,ldh基因多态性与变形链球菌乳酸脱氢酶活性及龋高敏感性相关。ldh基因分布与酶活性及乳牙龋具有正相关性。  相似文献

人类白细胞抗原(Human Leukocyte Antigen,HLA)基因复合物位于6p21.3,有220多个不同的功能基因,是人类基因组最复杂的遗传多态系统。HLA等位基因的变异在医学、法医学、人类学等领域具有重要的意义。自从1964年以来,HLA分型一直采用经典的微量淋巴细胞毒实验,但该方法是血清学水平的分,不能识别很多特异性的等位基因,而且高质量的抗体也不易获得。从20世纪90年代起,在国家自然科学基金的资助下,首先开展HLAⅡ类位点基因分研究及大规模群体多态性调查,所获得的中国主要民族基因数据已应用于多个领域。相比之下,HLAⅠ类基因数量更丰富,包含了A、B、C、E、F、G和假基因H、J、K、L等10个位点;基因分子结构更复杂,更具多态性。因此,HLAⅠ类DNA分型比HLAⅡ类分型及行多困难。直至目前中国人群HLA-A基因座基因多态性和分布频率的研究尚未充分进行。而任何DNA标记用于遗传分析、法医鉴定等领域之前,必须先进行群体调查,建立不同民族基因数据库,这是不可逾越的基础工作。鉴于此,采用灵敏而非同位素污染的PCR-SSOP基因分型技术,对165个汉族和162个维吾尔族个体的HLA-A基因座多态性进行调查。结果在汉族群体中发现22种等位基因,频率最高的是HLA-A*1101(19.7%),其次是*201(12.72%);在维族群体中发现22种等位基因,频率最高的是*2407(17.90%),等位基因*0101、*0201和*3301的频率均大于10%;HLA-A*0203、*0205、*0302、*2403和*3302仅在汉族群体中检出;HLA-A*0205、*0211、*2301、*2502、*68012和*6802仅在维族群体中检出。按照Hardy-Weinberg平衡定律检验,两个民族各等位基因型频率的预期值与实际观察值相吻合(P>0.05),证明了所获得汉族、维吾尔族HLA-A位点基因频率具有可靠性;同时也表明各等位基因的遗传特征符合符合孟德尔规律。经计算机统计分析,汉族群体HLA-A基因座杂合度(Heterozygosity,H)、个体识别率(Discrimination Power,DP)和非父排排率(Proba-bility of Paternity Exclusion,EPP)分别为0.9029、0.9776和0.8592;维族群体H、DP和EEP分别为0.9063、0.9379和0.7885。和其他遗传标记(如VNTR、STR、SNP)的单一位点相比,HLA-A具有高度的杂合率、个体识别率和非父排除率。因此,HLA-A等位基因在法医个体识别、亲权鉴定、基因诊断、人类学等领域具有重要的应用价值。  相似文献

目的:探讨新疆地区维吾尔族和汉族草酸钙结石与钙敏感受体(calcium sensitive receptor,Ca SR)基因多态性之间的关系。方法:选择398例临床确诊泌尿系草酸钙结石患者(200例维吾尔族,198例汉族)和399例正常对照者(200例维吾尔族,199例汉族),应用Sna Pshot方法对Ca SR基因两位点(rs1042636,rs1801726)的基因型及等位基因频率进行检测,并分析其与草酸钙结石发病的相关性以及对血钙、24 h尿钙水平的影响。结果:各组2个位点的基因型分布均符合Hardy-Weinberg平衡。汉族结石组与汉族对照组及维吾尔族结石族与维吾尔族对照组rs1042636、rs1801726位点基因型分布及基因频率差异均无统计学意义(P0.05)。维吾尔和汉族rs1042636基因型及等位基因频率比较差异有统计学意义(P0.05),且维吾尔族人群携带rs1042636等位基因A的风险高于汉族人群(病例组中OR值=2.145,%95CI=[1.602~2.866],P0.01;对照组中OR值=1.773,%95CI=[1.332~2.359],P0.01),其中维/汉病例组中等位基因频率分别为A=278(69.5%)/204(51.5%),G=122(30.5%)/192(48.5%);维/汉对照组中等位基因频率分别为A=264(66.0%)/208(52.3%),G=136(34.0%)/190(47.7%)。而病例组和对照组rs1801726基因型频率差异无统计学意义(P0.05);汉族病例组、对照组发现GG+AG基因型较AA基因型有较高的尿钙水平(病例组:P=0.007和对照组:P=0.006),维吾尔族人群该位点与两项指标无相关性。结论:Ca SR基因2个基因位点rs1042636、rs1801726可能不是新疆地区维吾尔族和汉族草酸钙结石发病的危险因子,两族rs1042636基因多态性分布存在差异,rs1042636位点基因多态性能影响汉族人群尿钙排泄,可能汉族调节钙排泄的遗传因素之一。  相似文献

调查了1506名朝鲜族,汉族4-7岁儿童体格发育情况。朝鲜族儿童平均身高和坐高皆低于汉族儿童相同年龄组,而平均体重却高于汉族同龄组;在头围、胸围指标中汉族男、女儿童4-5岁之间大于朝鲜族同龄组,但自5岁以后朝鲜族超过治疗组,可以看出,朝鲜族儿童体格为相对矮壮,而汉族儿童相对瘦高。又对朝、汉族儿童身同父母平均身高进行单变量相关分析显示出显著意义相关。延吉市朝、汉族儿童各项指标低于9185年全国九市效  相似文献

【目的】了解储粮害虫赤拟谷盗Tribolium castaneum (Herbst)成虫肠道细菌群落组成及多样性。【方法】利用Illumina MiSeq高通量测序技术对3个地理种群的赤拟谷盗雄虫肠道细菌16S rDNA的V3-V4变异区进行测序分析。【结果】从赤拟谷盗肠道样本中共测得465 293条reads,聚类为113个OTU,共注释到8门、15纲、33目、52科和79属的细菌。其中,优势菌门包括变形菌门(Proteobacteria)、厚壁菌门(Firmicutes)和放线菌门(Actinobacteria)等,优势菌属包括假单胞菌属(Pseudomonas)、肠杆菌科未鉴定属(Unclassified-f-Enterobacteriaceae)、不动杆菌属(Acinetobacter)、肠球菌属(Enterococcus)和根瘤菌属(Rhizobium)等。3个地理种群的赤拟谷盗肠道细菌的Shannon指数、Simpson指数、Ace指数和Chao指数分别为1.5-1.7、0.3、79.7-133.2和76.4-85.2。【结论】不同地理种群的赤拟谷盗成虫的肠道细菌群落组成存在显著差异。  相似文献

In steady state, the intestinal epithelium forms an important part of the gut barrier to defend against luminal bacterial attack. However, the intestinal epithelium is compromised by ionizing irradiation due to its inherent self-renewing capacity. In this process, small intestinal bacterial overgrowth is a critical event that reciprocally alters the immune milieu. In other words, intestinal bacterial dysbiosis induces inflammation in response to intestinal injuries, thus influencing the repair process of irradiated lesions. In fact, it is accepted that commensal bacteria can generally enhance the host radiation sensitivity. To address the determination of radiation sensitivity, we hypothesize that Paneth cells press a critical “button” because these cells are central to intestinal health and disease by using their peptides, which are responsible for controlling stem cell development in the small intestine and luminal bacterial diversity. Herein, the most important question is whether Paneth cells alter their secretion profiles in the situation of ionizing irradiation. On this basis, the tolerance of Paneth cells to ionizing radiation and related mechanisms by which radiation affects Paneth cell survival and death will be discussed in this review. We hope that the relevant results will be helpful in developing new approaches against radiation enteropathy.  相似文献

Objective: To explore the association between polymorphism of vitamin D receptor (VDR) and lead poisoning in Uygur and Han children in China.

Methods: The BsmI, ApaI and TaqI restriction sites of VDR genotyping were determined by polymerase chain reaction–restriction fragment length polymorphism in 443 Uygur and 469 Han children from Xinjiang province. The correlation between the polymorphism of VDR haplotypes and blood lead levels was explored.

Results: The genotype frequencies of VDR had significant differences in Han and Uygur children (p <0.01). According to VDR–BsmI, ApaI and TaqI haplotype analysis in Han children, haplotypes Atb and AtB were considerably decreased in the lead poisoning group (p <0.05) while haplotype aTb and ATb were significantly increased in the lead poisoning group (p <0.01). However, such results were not found in Uygur children (p?<?0.05).

Conclusion: A significant difference was seen in the frequency distribution of the VDR genotype among the different races. Haplotypes Atb and AtB might be protective factors while haplotypes ATb and aTb might be risk factors in Han children.  相似文献

目的:研究hTERC基因在新疆维吾尔族和汉族妇女宫颈脱落细胞中的表达情况及差异性。方法:收集2012年9月至2014年3月在新疆维吾尔自治区人民医院妇科门诊就诊或行宫颈癌机会性筛查的新疆维吾尔族和汉族妇女病例1160例,选择其中HPV感染阳性或TCT阳性或两项同时阳性的465例纳入研究队列,通过F1SH技术检测宫颈脱落细胞中hTERC基因的表达情况。结果:新疆维吾尔族与汉族妇女hTERC基因扩增的总阳性率无显著差异(P=0.620);维吾尔族与汉族妇女正常或慢性炎症组、CIN1组、CIN2组、CIN3组和SCC组hTERC基因扩增的阳性率亦均无显著差异(P=0.918,0.912,0.821,0.922,1.000),hTERC基因的表达率不存在民族差异;维吾尔族与汉族妇女正常或慢性炎症组/CIN1组和CIN2+组基因拷贝数的分布均无显著差异(P=0.925,0.862),且其正常或慢性炎症组/CIN1组和CIN2+组TERC:CEP比例分布亦均无显著差异(P=0.986,P=0.979),hTERC基因的扩增类型不存在民族差异;维吾尔族和汉族妇女hTERC基因扩增阳性率的大小顺序均为:SCC组≈CIN 3组CIN 2组CIN 1组≈正常或慢性炎症组,随着宫颈病变恶性程度的增加,各组中hTERC基因的扩增阳性率有增加的趋势,hTERC基因的表达与宫颈病变的恶性程度呈正相关(相关系数=0.648和0.712,P0.001)。结论:新疆维吾尔族与汉族妇女hTERC基因的表达率不存在民族差异;维吾尔族与汉族妇女hTERC基因的扩增类型不存在民族差异;随着宫颈病变恶性程度的增加,维吾尔族和汉族妇女hTERC基因的扩增阳性率有增加的趋势,hTERC基因的表达与宫颈病变的恶性程度呈正相关。  相似文献

Lipolytic enzymes are an important group of hydrolases that have found immense industrial application in biotechnology. In this study, the ability of gut bacteria isolated from the gut of the Eri silkworm, Samia ricini, to produce lipolytic enzymes was evaluated through qualitative and quantitative assays. The results of lipase screening showed that 28 isolates had lipolytic activity. The results of 16S ribosomal RNA sequencing indicated that the genus Bacillus comprised majority of the lipolytic bacterial isolates (71%) followed by Pseudomonas (15%); whilst Acinetobacter, Enterobacter and Enterococcus comprised 11%. Lipolytic activity was found in bacteria isolates identified from all the three gut compartments of S. ricini larvae with significant activity from isolates extracted from the foregut and midgut. The lipolytic index among the bacterial isolates ranged between 0.63 and 2.81 on Rhodamine B medium, and all isolates exhibited significant lipolytic activity with p-nitrophenyl butyrate (PNPB) with specific activity ranging from 0.52 to 0.82 μmol/min/mg. The effect of pH and temperature showed that lipase activity was optimum at 37 °C and pH 7–9. A phylogenetic relationship of lipase producing gut bacteria indicated high cluster stability for isolates from different stages (>50%) suggesting that the isolates persist across developmental stages of the host. The Eri silkworm is reared for its silk and the knowledge of its gut bacteria with the ability to produce lipases lies in the significance as far as boosting production of this insect via development of probiotics to enhance commercial Eri rearing. In addition, this insect may be a good resource for profiling novel lipolytic microbes for commercial production of lipases as lipases from microbial origin have assumed a great deal of importance as industrial enzymes due to their potential for use in biotechnology.  相似文献

Atomic absorption spectrometry is used to determine the contents of microelements (such as Zn, Cu, Ca, Mg and Fe) in the all-blood and hair of the Han, Uygur, and Kazak people in normal health state. The results show that the above mentioned elements in the hair arranged successively as Ca>Zn>Mg>Fe>Cu and those in the all-blood of the Han people arranged as Fe>Mg>Ca>Zn>Cu; of the Uygur and Kazak people, as Fe>Mg>Ca>Zn>Cu. This paper reports that the content of the microelement Mg in the all-blood and hair of the Uygur and Kazak people is obviously higher than that of the Han people (P<0.05).  相似文献

目的:判断细胞角蛋白19(cytokeratin19,CK19)作为循环肿瘤细胞的标志物的意义,探讨在肾癌病人外周血中CK19 的表达 情况和临床价值,并观察汉族与维吾尔族肾癌患者之间循环肿瘤细胞的差异。方法:50 例肾癌病人设为病例组,健康体检人群50 例设为对照组,对上述两组人群应用流式细胞仪技术检测外周血中CK19 的表达水平;并探讨肾癌病人外周血CK19 的表达与肾 癌病理分类、病理分期及组织学分级之间的关系;比较20 例汉族肾细胞癌患者及20 例维吾尔族肾癌患者外周血中CK19 的差异 表达情况。结果:组间比较,50例对照组中无阳性,50 例肾癌患者组中24 例阳性;组内比较,不同肾癌病理分类之间CK19 表达差 异没有统计学意义(P>0.05),不同肾癌病理分期及组织学分级之间CK19 表达差异具有有统计学意义(P<0.05)。维吾尔族与汉族 两个不同民族肾癌之间的血清循环肿瘤细胞存在着较明显差异(P<0.05)。结论:在肾癌患者外周血中的CK19 可以初步判断循环 肿瘤细胞的水平,对早期肾癌的诊断、预测具有一定的意义,值得进一步探索和研究,维吾尔族肾癌与汉族肾癌患者之间循环肿瘤 细胞可能存在差异。  相似文献

【目的】探究生活在相同环境下,同年龄段健康藏族人群和汉族人群肠道细菌组成的多样性特征及其可能的成因。【方法】入选同专业的20–24岁男性健康藏族学生和汉族学生各20名,同一时间段收集新鲜粪便,提取新鲜粪便细菌总DNA,通过高通量测序和生物信息学分析,结合对饮食习惯的问卷调查,研究健康藏族学生与汉族学生肠道细菌多样性与饮食习惯之间的关系。【结果】从40份样本中,测序共得到2880个OTU,alpha和beta多样性分析表明,生活在相同环境下,不同民族男性学生的肠道细菌组成有明显差异。Prevotella 9、Bacteroides和Lachnoclostridium等18个属为两民族学生共有,同时,Prevotella 9、Faecalibacterium和Succinivibrio等10个属细菌为对两民族学生肠道细菌组成差异具有显著性影响的标志性菌属。进一步聚类分析表明,所有学生样本分为ET F型、ET P型、ET F和ET B混合型3种不同肠型,随访调查结果显示不同类型分别对应以脂肪为主和以淀粉为主的不同饮食习惯。【结论】生活在相同环境下,健康藏族和汉族学生的肠道细菌组成特征及其差异可能与遗传因素及饮食习惯密切相关。  相似文献

Gut bacteria play a crucial role in the several metabolic activity of the insects. In the present work, effort has been made to decipher the gut microbiota associated with the developmental stages of Gynaikothrips uzeli a gall inducing thrips along with their predicted functional role. Further, an effort has been made to correlate the bacterial communities with plant pathogenesis and thelytoky behaviour of G. uzeli. Findings obtained revealed that genus Arsenophonus dominated the total bacterial diversity and was transmitted vertically through the developmental stages. Further, it was observed that the high abundance of genus Arsenophonus promotes the thelytoky behaviour in G. uzeli and results in the killing of males. Furthermore, strong connecting link between Arsenophonus abundance and gall induction in F. benjamina was observed in the current dataset. G. uzeli being in the category of phloem sucking insect was known for the induction of galls and the current findings for the first time unveiled the facts that high abundance of genus Arsenophonus a well-known plant pathogen may be one of the major reason for inducing galls in F. benjamina. Moreover, PICRUSt2 analysis revealed that predicted functional pathways like biosynthesis of amino acids, and metabolism of carbon, nitrogen, carbohydrates and amino acids (e.g. Arginine, Alanine, Aspartate, Glutamate, Proline, Cysteine, Methionine, Glycine, Threonine, and Serine) were frequently noticed in profiles associated with all the developmental stages of G. uzeli. More to this, the high abundance of Arsenophonus in G. uzeli suggest that representatives of this genus may be resistant and/or tolerant to different antibacterial agents, alkaloids, flavonoids, and glycosides (e.g. quercetin). The correlation of bacterial diversity in pathogenicity can be extrapolated in different pest and vector species of other arthropods.  相似文献
