The time constant of the process producing the delay in Na inactivation development as determined by the two pulse method (delay) was extracted and compared to that of the slowest Na activation process 3 for the I Na during the conditioning pulse of that same determination. delay and two pulse inactivation c values were computer generated using a nonlinear least squares algorithm. h and single pulse inactivation h values were independently generated for each determination also with the aid of the computer using the same non-linear least squares algorithm. In one determination at 2 mV, c was 4.68 and delay 0.494 ms while h was 4.70 and 3 0.491 ms for a c/h of 0.996 and a delay/3 of 1.006. Mean delay/3 from five determinations in four axons, both Cs and K perfused, and spanning a potential range of-27 to 2mV was 1.068. The precursor process to inactivation is channel opening. Some fraction of channels presumably inactivate via another route where prior channel opening is not required.  相似文献

Summary The kinetics of light-driven proton transport by bacteriorhodopsin has been studied in a model system consisting of a planar lipid bilayer membrane to which purple membrane fragments have been attached. After excitation with a 10-nsec laser flash a fast negative current-transient occurs, followed by a positive transient which decays to zero. The time course of the photocurrent may be represented by a sum of four exponentials with time constants 1= 1.2sec, 2= 17sec, 4= 57sec, 1= 950sec (at 25°C). In a D2O medium 2 and 3 are increased by a factor of 2.6 and 2.9, respectively, whereas 1 remains unaffected. The observed components of the photocurrent can be correlated to photochemical reaction steps inferred from flash-photometric experiments on the basis of the observed time constants, the activation energies, and the effects of pH and D2O. From the photocurrent signals information may be obtained on the magnitude of the charge displacement associated with the elementary transitions of the bacteriorhodopsin molecule.  相似文献

Synopsis Length-frequency data suggest Nile perch, Lates niloticus, from the Nyanza Gulf grew to a total length of 9 cm by age 118 days and 23 cm by age 287 days. A modified von Bertalanffy growth curve t = 1.35·L(1-e–K(t-t o)) with the parameters L = 93.1, K = 0.272 and to = 0.046, is suggested to describe growth up to 5 years of age and the relationship t = 1.35·(31.96 + 7.681t) for fish aged 6 years and above. Length-weight relationships were = 0.0234·-gt2.74 for fish between 7 and 15.9 cm total length, = 0.0151·2.94 for fish between 16 and 45.9 cm total length, and = 0.0023·3.44 for fish between 46 and 120 cm total length. Male Nile perch first matured between 50 and 55 cm total length when they were probably 2 years old; female Nile perch first matured between 80 and 85 cm total length when they were probably 4 years old. Small males were common, large males were rare, with the reverse holding for females. Sex change, from male to female, is a possible explanation for this size dimorphism.  相似文献

Summary By using a high field strength DC pulse of 15 kV/cm and a pulse duration of 5 ms for the transfection of E. coli by bacteriophage DNA, we obtained efficiencies of 1.1 × 106 (pfu/g bacteriophage , DNA). This represents a 100-fold improvement over the traditional CaCl2/heat shock method and is a viable alternative to the more costly in vitro packaging of recombinant bacteriophage DNA for the production of cDNA and genomic libraries.  相似文献

Kinetic and voltage-dependent characteristics of the inactivating action of incoming calcium currents were investigated in the somatic membrane of rat spinal ganglia neurons using an intracellular dialysis technique. It was shown that the "tail" of low-threshold calcium current could be reliably described by one exponent with a time constant of =1.2–1.8 msec at a repolarization potential of –90 mV. The "tail" of the high-threshold calcium current represented the sum of several exponents. The time constant of the main component which expressed inactivation of the high threshold calcium current was h=250–350 µsec. It was also shown that and h remained virtually unchanged for repolarization potentials in the subthreshold region; they increase, however, if the repolarizing potential is close to those potentials at which the corresponding component of calcium current is initially activated. A dependence was observed between the levels and h and duration of the depolarizing shift. Findings are discussed in the context of a three-tier model of calcium channels.A. A. Voltage dependence of the deactivation kinetics of calcium inward currents was investigated in the somatic membrane of murine spinal ganglia neurons. It was found that deactivation of high threshold calcium current has a slower component (=0.80–0.85 msec at a repolarizing potential of –80 mV) as well as the principal transient exponential component (130 sec at the same potential repolarizing level). A dissimilar relationship exists between amplitudes of the transient and slower exponential components, describing deactivation of high threshold calcium current and degree of activation of the depolarizing shift in membrane potential; the former dependence is expressed by a sigmoid and the latter by a V-shaped curve. The slower component of deactivation of high threshold current was inhibited substantially by perfusing the cell with a Tris-PO4-containing solution. Low-threshold calcium tail current undergoes slower deactivation (=1.1–1.2 msec) at a repolarizing potential of –160 mV. A relationship between the time constant of low threshold current deactivation and the type of penetrating cation used was observed. A kinetic model of calcium current deactivation is suggested, taking account of the three different types of calcium channels, (one low and two high threshold) present in the somatic membrane.A. A. Summary Electric impulses were found to cause transient permeability changes in the membranes of vesicles storing biogenic amines. Release of catecholamines induced by electric fields (of the order of 20 kV/cm and decaying exponentially with a decay time of about 150 sec) was studied, using the chromaffin granules of bovine adrenomedullary cells as a vesicular model system. Far-UV-absorption spectroscopy was applied to determine the amount of catecholamines released from suspended vesicles. A polarization mechanism is suggested for the induction of short-lived permeability changes caused by electric fields. Such transient changes in permeability may possibly represent a part of the sequence of events leading to stimulated neurohumoral secretion.  相似文献

Summary In separated outer medullary collecting duct (MCD) cells, the time course of binding of the fluorescent stilbene anion exchange inhibitor, DBDS (4,4-dibenzamido-2,2-stilbene disulfonate), to the MCD cell analog of band 3, the red blood cell (rbc) anion exchange protein, can be measured by the stopped-flow method and the reaction time constant, DBDS, can be used to report on the conformational state of the band 3 analog. In order to validate the method we have now shown that the ID50,DBDS,MCD (0.5±0.1 m) for the H2-DIDS (4,4-diisothiocyano-2,2-dihydrostilbene disulfonate) inhibition of DBDS is in agreement with the ID50,Cl ,MCD (0.94±0.07 m) for H2-DIDS inhibition of MCD cell Cl flux, thus relating DBDS directly to anion exchange. The specific cardiac glycoside cation transport inhibitor, ouabain, not only modulates DBDS binding kinetics, but also increases the time constant for Cl exchange by a factor of two, from Cl=0.30±0.02 sec to 0.56±0.06 sec (30mm NaHCO3). The ID50,DBDS,MCD for the ouabain effect on DBDS binding kinetics is 0.003±0.001 m, so that binding is about an order of magnitude tighter than that for inhibition of rbc K+ flux (K I,K +,rbc=0.017 m). These experiments indicate that the Na+,K-ATPase, required to maintain cation gradients across the MCD cell membrane, is close enough to the band 3 analog that conformational information can be exchanged. Cytochalasin E (CE), which binds to the spectrin/actin complex in rbc and other cells, modulates DBDS binding kinetics with a physiological ID50,DBDS,MCD (0.076±0.005 m); 2 m CE also more than doubles the Cl exchange time constant from 0.20±0.04 sec to 0.50±0.08 sec (30mm NaHCO3). These experiments indicate that conformational information can also be exchanged between the MCD cell band 3 analog and the MCD cell cytoskeleton.  相似文献

Elementary Na+ currents were recorded at 19°C in inside-out patches from cultured neonatal rat cardiocytes. In analyzing the sensitivity of chemically modified Na+ channels to several class 1 antiarrhythmic drugs, the hypothesis was tested that removal of Na+ inactivation may be accompanied by a distinct responsiveness to these drugs, open channel blockade.Iodate-modified and trypsin-modified cardiac Na+ channels are noninactivating but strikingly differ from each other by their open state kinetics, a O1–O2 reaction (open(1) 1.4±0.3 msec; open(2) 5.4±1.1 msec; at –40 mV) in the former and a single open state (open 3.0±0.5 msec; at –40 mV) in the latter. Lidocaine (150 mol/liter) like propafenone (10 mol/liter), diprafenone (10 mol/liter) and quinidine (20 mol/liter) in cytoplasmic concentrations effective to depress NP o significantly can interact with both types of noninactivating Na+ channels to reduce the dwell time in the conducting configuration. lodate-modified Na+ channels became drug sensitive during the O2 state. At –40 mV, for example, lidocaine reduced open(2) to 62±5% of the control without detectable changes in open(1). No evidence could be obtained that these inhibitory molecules would flicker-block the open Na+ pore. Drug-induced shortening of the open state, thus, is indicative for a distinct mode of drug action, namely interference with the gating process. Lidocaine proved less effective to reduce open(2) when compared with the action of diprafenone. Both drugs apparently interacted with individual association rate constants, alidocaine was 0.64×106 mol–1 sec–1 and adiprafenone 13.6×106 mol–1 sec–1. Trypsin-modified Na+ channels also appear capable of discriminating among these antiarrhythmics, the ratio adiprafenone/alidocaine even exceeded the value in iodate-modified Na+ channels. Obviously, this antiarrhythmic drug interaction with chemically modified Na+ channels is receptor mediated: drug occupation of such a hypothetical hidden receptor that is not available in normal Na+ channels may facilitate the exit from the open state.This work was supported by a grant of the Deutsche Forschungsgemeinschaft (Ko 778/2-4), Bonn.  相似文献

Structure–dynamics interrelationships are important in understanding protein function. We have explored the empirical relationship between rotational correlation times (c and the solvent accessible surface areas (SASA) of 75 proteins with known structures. The theoretical correlation between SASA and c through the equation SASA = Krc (2/3) is also considered. SASA was determined from the structure, c calc was determined from diffusion tensor calculations, and c expt was determined from NMR backbone13 C or 15N relaxation rate measurements. The theoretical and experimental values of c correlate with SASA with regression analyses values of Kr as 1696 and 1896 m2s-(2/3), respectively, and with corresponding correlation coefficients of 0.92 and 0.70.  相似文献

The effects of detergents on the electronic structure of the oxidized primary donor P+ and the time constant AP of the P+Q A charge recombination at ambient temperatures have been investigated in native and mutant reaction centers (RCs) from Rhodobacter sphaeroides. It is shown that N-lauryl-N,N-dimethyl-3-ammonio-1-propane sulfonate (SB12) induces a transition to a second distinct conformation of the RC. In the case of the wild type and the mutant FY(M197), in which a hydrogen bond is introduced to the 2-acetyl group of the dimer half of P that is associated with the M-subunit of the RC, the conformational change causes a more asymmetric spin density distribution between the two bacteriochlorophyll moieties of P+ in favor of the L-half. For both types of RCs the time constant AP depends on the SB12/RC ratio as does the position of the long-wavelength band of P, max. The increase of AP by 30 ms and the shift of max from 866 nm to 851 nm are indicative for the conformational change. In addition, a smaller linear increase of AP with increasing SB12/RC ratio is superimposed on the variation of AP due to the conformational change. Similar effects of SB12 on the optical spectra as well as on AP are also observed for the two heterodimer mutants HL(L173) and HL(M202), in which one of the bacteriochlorophylls of P is replaced by a bacteriopheophytin. There are no clear indications for a correlation of AP with the localization of the positive charge in P+. Furthermore, it is concluded from the dependence of AP on the SB12/RC ratio that the single-site mutations do not affect the standard free energy difference of the two conformations to a measurable extent.  相似文献

The effect of correlation between the kinetics of acetylcholine (ACh) quanta release and the kinetics of the postsynaptic membrane channel activity on end-plate currents (EPCs) was studied in model experiments. The presynaptic process was described by a transmitter secretion probability distribution (SPD), and the postsynaptic process by characteristicsof miniature EPC (MEPC) with standard amplitude and variable decay duration (MEPC). The SPD was represented by a curve with an S-like rise and exponential decay. The main portion of the data was analyzed after being adjusted to 20°C. The effect of the EPC amplitude loss (the difference between the EPC amplitudes in cases of synchronous and non-synchronous release of the ACh quanta) due to temporal dispersion of the transmitter secretion process was 43% at MEPC=1.0 msec and 20% at MEPC=4–5 msec under conditions when acetylcholinesterase (AChE) was inhibited.When MEPC varied over a wide range, the effect of the EPC amplitude loss could be described by a curve with steeply and gently sloping portions; the boundary between these two portions corresponded to approximately 1.0 msec MEPC, which is nearly the channel lifetime at resting membrane potential. The rise time of the EPC only slightly related to changes in MEPC. The rate of EPC decay exceeded that in MEPC only when the lifetimes of the ACh-activated ionic channels were lower than their physiological values. In this case, the kinetics of transmitter release became a factor determining the EPC decay time course.Three variants of an increase in temporal dispersion of the ACh quanta secretion were modeled for constant MEPC: one resulting from an increase in the SPD rising phase; a second resulting from equal increases in the SPD rising and declining phases, and a third resulting from an increase in the SPD declining phase. The results of the first variant corresponded most closely to experimental data on changes in EPC during a short-term period of rhythmical activity. This was also characterized by minimal effect of EPC amplitude loss.It has been suggested that the interrelation between the parameters of SPD and MEPC is an important factor regulating the amplitude and duration of the postsynaptic signal. Summary The passive electrical cable properties of ocellar L-neurons were determined by applying current steps and recording the voltage transients using a two-electrode intracellular current clamp system. Morphological data were obtained following intracellular staining with Lucifer yellow.Two groups of neurons were distinguished physiologically. In the first group both the membrane time constant m and the first equalizing time constant 1 could be determined. In the second group only m was measurable. The ratio of the physiological groups was equal to the ratio of the morphological types ML:(M1 plus M2) in the median ocellar nerve. Thus the first group probably consists of ML-type L-neurons. The passive cable properties of this group were calculated by combining the physiological and morphological data. The following values were obtained: electrotonic lengthL=1.35; membrane time constant m =7.6 ms; length constant =0.22 cm; membrane resistivityR m=2.0 · 103 · cm2; membrane capacitanceC m=3.8 F · cm–2; intracellular resistivityR i=24 · cm. Evidence is presented that the membrane parameters of the other types of L-neurons have the same values. By intracellular dialysis of isolated neurons of the mollusksHelix pomatia andLimnaea stagnalis and by a voltage clamp technique the characteristics of transmembrane ionic currents were studied during controlled changes in the ionic composition of the extracellular and intracellular medium. By replacing the intracellular potassium ions by Tris ions, functional blocking of the outward potassium currents was achieved and the inward current distinguished in a pure form. Replacement of Ringer's solution in the extracellular medium with sodium-free or calcium-free solution enabled the inward current to be separated into two additive components, one carried by sodium ions, the other by calcium ions. Sodium and calcium inward currents were found to have different kinetics and different potential-dependence: mNa=1±0.5 msec, mCa=3±1 msec, hNa=8±2 msec, hCa=115±10 msec (Vm=0), GNa=0.5 (Vm=–21±2 mV), GCa=0.5 (Vm=–8±2 mV). Both currents remained unchanged by tetrodotoxin, but the calcium current was specifically blocked by cadmium ions (2·10–3 M), verapamil, and D=600, and also by fluorine ions if injected intracellularly. All these results are regarded as evidence that the soma membrane of the neurons tested possesses separate systems of sodium and calcium ion-conducting channels. Quantitative differences are observed in the relative importance of the systems of sodium and calcium channels in different species of mollusks.A. A. Summary We have used magnetic resonance spectroscopy, both ESR and13C spin relaxation, to measure translational and rotational mobilities and partition coefficients of small nitroxide solutes in dipalmitoyl lecithin liposomes. Above the bilayer transition temperature,T c, the bilayer interior is quite fluid, as determined from the solutes' rapid rotational and moderately rapid translational motion; the rotational and translational viscosities within the bilayer are R <1cP and =6–10cP, respectively. and R are independent of molecular size for all solutes studied, but all were small compared to the size of the phospholipids. , and probably R , are relatively independent of temperature aboveT c, but both increase very sharply as temperature is lowered belowT c; at 32°C, R increases to 6cP and is greater than 1000 cP. Anisotropy of rotational motion increases gradually as temperature is lowered toT c, and changes little belowT c; anisotropy of translational motion was not investigated.13C nuclear spin relaxation measurements indicate that translational motion of nitroxide solutes is more rapid in the center of the bilayer than near the polar interface. It takes at least 100 nsec for a solute molecule to cross the bilayer/water interface. We estimate a lower limit of 2 sec/cm for the interfacial resistance to solute diffusion; this result indicates that interfacial resistance dominates permeation across the membrane. The relative solubility, or partition coefficient, is a strong function of solute structure, and decreases abruptly on cooling through the transition temperature. From the partition coefficient and its temperature dependence we calculate the free energy, enthalpy, and entropy of partition. Effects of cholesterol on partition and diffusion coefficients are compatible with the interpretation that bilayers containing cholesterol consist of two phases.  相似文献

Summary Elementary Na+ currents were recorded at 9°C in inside-out patches from cultured neonatal rat heart myocytes. In characterizing the sensitivity of cooled, slowly inactivating cardiac Na+ channels to several antiarrhythmic drugs including propafenone, lidocaine and quinidine, the study aimed to define the role of Na+ inactivation for open channel blockade.In concentrations (1–10 mol/liter) effective to depressNP o significantly, propafenone completely failed to influence the open state of slowly inactivating Na+ channels. With 1 mol/liter, open changed insignificantly to 96±7% of the control. Even a small number of ultralong openings of 6 msec or longer exceeding open of the whole ensemble several-fold and attaining open (at –45 mV) in cooled, (-)-DPI-modified, noninactivating Na+ channels proved to be drug resistant and could not be flicker-blocked by 10 mol/liter propafenone. The same drug concentration induced in(-)-DPI-modified Na+ channels a discrete block with association and dissociation rate constants of 16.1 ± 5.3 × 106 mol–1 sec–1 and 675 ± 25 sec–1, respectively. Quinidine, known to have a considerable affinity for activated Na+ channels, in lower concentrations (5 mol/liter) left open unchanged or reduced, in higher concentrations (10 mol/liter) open only slightly to 81% of the predrug value whereasNP o declined to 30%, but repetitive blocking events during the conducting state could never be observed. Basically the same drug resistance of the open state was seen in cardiac Na+ channels whose open-state kinetics had been modulated by the cytoplasmic presence of F ions. But in this case, propafenone reduced reopening and selectively abolished a long-lasting open state. This drug action is unlikely related to the inhibitory effect onNP o since hyperpolarization and the accompanying block attenuation did not restore the channel kinetics. It is concluded that cardiac Na+ channels cannot be flicker-blocked by antiarrhythmic drugs unless Na+ inactivation is removed.  相似文献

The role of postysynaptic potentiation (PSP) and asynchronous secretion of acetylcholine (ACh) in the generation of multiquantal currents and end plate currents (EPC) was investigated under voltage clamp conditions in transected sartorius muscle of the frog before and after 4-aminopyridine (4-AP) treatment. Compared with miniature EPC (MEPC), showing an average quantum content of 249, multiquantal EPC has a larger amplitude, longer rise-time, and longer decay-time (epc). Magnesium ions (6–10 mM) reduce the amplitude and spec of EPC without affecting its rise-time. Rhythmical stimulation (10 Hz for 60 sec) results in reduced amplitude and but increased rise-time of EPC. D-turbocurarine (5×10–7 M) and -bungarotoxin (1×10–5 gm/ml) diminishes the difference between epc and mepc. In the presence of 4-AP, all these effects are much more pronounced. It is proposed that asynchronous secretion of ACh from motor nerve teminals causes prolongation of the rise-time and reduction of the amplitude of EPC but has little or no effect upon the decay rate of EPC. The slow decay of multiquantal EPC, both in the absence and in the presence of 4-AP, is almost entirely due to postynaptic interaction of ACh quanta, i.e., PSP.Kurashov Meidcal Institute, Kazhan. The effective diffusivity of glucose in porous glass beads was determined using a transient method. Predictions for the intraparticle and surface concentrations were made by an analytical solution of the mass balance. The value of the diffusivity was expected to be lower than the value of the corresponding diffusion coefficient in water, but the opposite was observed. This effect results from intraparticle fluid flow, leading to high values of the apparent effective glucose diffusivity. To measure diffusion only and to prevent any internal convection during the diffusion experiment, the pores of the porous glass beads were filled with Ca-alginate gel. For these glass beads (internal porosity, , equal to 0.56), we found an effective glucose diffusivity of 2.2×10–10 m2/s at 30°C. Using the relationship to effective intraparticle diffusivity (Deff)=effective diffusivity in 1% Ca-alginate beads (Dgel) / (with the tortuosity factor) this gives =1.7. For known and measuring by the method described, the Deff can be calculated for other porous materials or diffusing substances. Knowledge of the exact value of the effective diffusivity is a necessity in bioreactor modelling and was demonstrated by prediction of the residence time distribution profiles in a packed-bed bioreactor containing immobilized yeast cells.  相似文献

Summary Dynamics of the backbone and some side chains of apo-neocarzinostatin, a 10.7 kDa carrier protein, have been studied from 13C relaxation rates R1, R2 and steady-state 13C-{1H} NOEs, measured at natural abundance. Relaxation data were obtained for 79 nonoverlapping C resonances and for 11 threonine C single resonances. Except for three C relaxation rates, all data were analysed from a simple two-parameter spectral density function using the model-free approach of Lipari and Szabo. The corresponding C–H fragments exhibit fast (e < 40 ps) restricted libration motions (S2=0.73 to 0.95). Global examination of the microdynamical parameters S2 and e along the amino acid sequence gives no immediate correlation with structural elements. However, different trends for the three loops involved in the binding site are revealed. The -ribbon comprising residues 37 to 47 is spatially restricted, with relatively large e values in its hairpin region. The other -ribbon (residues 72 to 87) and the large disordered loop ranging between residues 97–107 experience small-amplitude motions on a much faster (picosecond) time scale. The two N-terminal residues, Ala1 and Ala2, and the C-terminal residue Asn113, exhibit an additional slow motion on a subnanosecond time scale (400–500 ps). Similarly, the relaxation data for eight threonine side-chain C must be interpreted in terms of a three-parameter spectral density function. They exhibit slower motions, on the nanosecond time scale (500–3000 ps). Three threonine (Thr65, Thr68, Thr81) side chains do not display a slow component, but an exchange contribution to the observed transverse relaxation rate R2 could not be excluded at these sites. The microdynamical parameters (S2, e and R2ex) or (S infslow sup2 , S inffast sup2 and slow) were obtained from a straightforward solution of the equations describing the relaxation data. They were calculated assuming an overall isotropic rotational correlation time e for the protein of 5.7 ns, determined using standard procedures from R2/R1 ratios. However, it is shown that the product (1–S2e is nearly independent of e for residues not exhibiting slow motions on the nanosecond time scale. In addition, this parameter very closely follows the heteronuclear NOEs, which therefore could be good indices for local fast motions on the picosecond time scale.  相似文献

Summary Apical Na+ entry into frog skin epithelium is widely presumed to be electrodiffusive in nature, as for other tight epithelia. However, in contrast to rabbit descending colon andNecturus urinary bladder, the constant field equation has been reported to fit the apical sodium current (N Na)-membrane potential (mc) relationship over only a narrow range of apical membrane potentials or to be inapplicable altogether. We have re-examined this issue by impaling split frog skins across the basolateral membrane and examining the current-voltage relationships at extremely early endpoints in time after initiating pulses of constant transepithelial voltage. In this study, the rapid transient responses in mc were completed within 0.5 to 3.5 msec. Using endpoints to 1 to 25 msec, the Goldman equation provided excellent fits of the data over large ranges in apical potential of 300 to 420 mV, from approximately –200 to about +145 mV (cell relative to mucosa). Split skins were also studied when superfused with high serosal K+ in order to determine whether theI Na-mc relationship could be generated purely by transepithelial measurements. Under these conditions, the basolateral membrane potential was found to be –10±3 mV (cell relative to serosa, mean±se), the basolateral fractional resistance was greater than zero, and the transepithelial current was markedly and reversibly reduced. For these reasons, use of high serosal K+ is considered inadvisable for determining theI Na-mc relationship, at least in those tissues (such as frog skin) where more direct measurements are technically feasible. Analysis of theI Na-mc relationships under baseline conditions provided estimates of intracellular Na+ concentration and of apical Na+ permeability of 9 to 14mm and of 3 × 10–7 cm · sec–1, respectively, in reasonable agreement with estimates obtained by different techniques.  相似文献
