We present, for the first time, the oxygen response kinetics of mitochondrial respiration measured in intact leaves (sunflower and aspen). Low O2 concentrations in N2 (9–1500 ppm) were preset in a flow-through gas exchange measurement system, and the decrease in O2 concentration and the increase in CO2 concentration as result of leaf respiration were measured by a zirconium cell O2 analyser and infrared-absorption CO2 analyser, respectively. The low O2 concentrations little influenced the rate of CO2 evolution during the 60-s exposure. The initial slope of the O2 uptake curve on the dissolved O2 concentration basis was relatively constant in leaves of a single species, 1.5 mm s−1 in sunflower and 1.8 mm s−1 in aspen. The apparent K 0.5(O2) values ranged from 0.33 to 0.67 μ M in sunflower and from 0.33 to 1.1 μ M in aspen, mainly because of the variation of the maximum rate, V max (leaf temperature 22°C). The initial slope of the O2 response of respiration characterizes the catalytic efficiency of terminal oxidases, an important parameter of the respiratory machinery in leaves. The plateau of the response characterizes the activity of the mitochondrial electron transport chain and is subject to regulations in accordance with the necessity for ATP production. The relatively low oxygen conductivity of terminal oxidases means that in leaves, less than 10% of the photosynthetic oxygen can be reassimilated by mitochondria.  相似文献

Plasma cortisol levels of fingerling rainbow trout were measured as an index of the stress resulting from various procedures used for transport of the fish for stocking. When transported under 'normal' conditions, which included water at the hatchery acclimation temperature (10–11°C), O2 saturation or supersaturation, and neutral pH, there was a marked increase in plasma cortisol levels within 0.5 h, which was maintained over the next 4 h of transport; there was a significant decrease in plasma cortisol by 8 h of transport. It was found that the plasma cortisol levels at 4 and 8 h were not appreciably altered by transport under partial O2 desaturation, O2 saturation, O2 supersaturation, or 0.5% NaCl, or by anaesthesia with tricaine methanesulfonate (MS 222) prior to capture and transport in MS 222-free water or 0.5% NaCl. A 15 min exposure to an immobilizing dose of buffered or unbuffered MS 222, or 2-phenoxyethanol, caused an increase in plasma cortisol of about 2 h duration, indicating that anaesthetics are themselves stressful. Exposure to chilled water (1° C) caused a large increase in plasma cortisol levels by 4 h after initiation of exposure; plasma cortisol had decreased at 1 day, and by 2 days a constant level was reached which was above the level in fingerling trout under 'normal' hatchery conditions. Trout acclimated to chilled water for 24 h and transported in chilled water had an increase in plasma cortisol during transport. Anaesthesia prior to transport or addition of salt did not reduce the stress of transport as judged by plasma cortisol levels. The results indicate that stress from capture and transport during stocking cannot be avoided using present methods.  相似文献

Muscle cellularity at a developmental stage around the time of hatching was examined in rainbow trout which had been reared from the eyed stage at three different temperature regimes (5, 10 and 15° C) and different O2 tensions [70% of air saturation value (ASV) at 5° C, 100% of ASV at all temperatures, and 150% of ASV at 10 and 15° C]. It was found that, as has been shown for other species, there was a difference in muscle fibre numbers and fibre cross-sectional areas between some of the regimes. There was a decrease in fibre number at the intermediate and higher temperature, and a decrease in fibre size at the high temperature. The temperature effects observed were modified by the applied changes in O2 tension. An increased O2 tension at 10° C led to an increase in fibre size whereas a decrease in O2 tension at the low temperature resulted in a decrease in fibre number. The largest total white muscle cross-sectional area was achieved at 10° C under high O2 conditions. Temperature and O2 tension therefore had a clear effect on muscle cellularity and there was a significant interaction between the two parameters.  相似文献

Rates of oxygen consumption were measured in the geothermal, hot spring fish, Oreochromis alcalicus grahami by stopped flow respirometry. At 37° C, routine oxygen consumption followed the allometric relationship: V o2=0.738 M 0.75, where V o2 is ml O2 h −1 and M is body mass (g). This represents a routine metabolic rate for a 10 g fish at 37° C of 0.415 ml O2 g−1 h −1 (16.4 μmol O2 g −1 h −1). Acutely increasing the temperature from 37 to 42° C significantly elevated the rate of O2 consumption from 0.739 to 0.970 ml O2 g −1 h −1 ( Q 10=l.72). In the field, O. a. grahami was observed to be 'gulping' air from the surface of the water especially in hot springs that exceeded 40° C. O. a. grahami may utilize aerial respiration when O2 requirements are high.  相似文献

Smallmouth bass larvae became highly sensitive to oxygen deficiency on the second day after hatching and continued so to the 10th day. During this period they could not survive exposure to 1 mg O2 l–1 for 3 h at 20° C, and many were killed within 1 h. At 2 mg O2 l–1 half the larvae survived 3 h at 20° C; at 2.5 mg l–1 most survived, and at 3.5 mg l–1 all survived. Resistance to oxygen deficiency was regained by the 11th day, the majority of the larvae withstanding a 3-h exposure at 1 mg O2 l–1. At 25° C the effects of low oxygen concentration were intensified. At 3 and 4 mg O2 l–1 and 20° C the normally quiescent larvae became very active, even swimming to the surface 5 or 6 cm above the substrate. Increasing the temperature increased this response. Smallmouth larvae were more sensitive than large-mouth bass larvae to oxygen deficiency.  相似文献

Frost tolerance has been reported in the shoots of wild, tuberiferous potato species such as Solanum commersonii when the plants are grown in either field or controlled conditions. However, these plants can survive as underground tubers and avoid unfavorable environmental conditions altogether. As such, leaf growth and photosynthesis at low temperature may not be required for survival of the plants. In order to determine the temperature sensitivity of S. commersonii shoots, we examined leaf growth, development and photosynthesis in plants raised at 20/16°C (day/night). 12/9°C and 5/2°C. S. commersonii leaves grown at 5°C exhibited a marked decrease in leaf area and in total chlorophyll (Chl) content per leaf area when compared with leaves grown at 20°C. Furthermore, leaves grown at 5°C did not exhibit the expected decrease in either water content or susceptibility to low-temperature-induced photoinhibition that normally characterizes cold acclimation in frost-tolerant plants. Measurements of CO2-saturated O2 evolution showed that the photosynthetic apparatus of 5°C plants was functional, even though the efficiency of photosystem II photochemistry was reduced by growth at 5°C. A decrease in the resolution of the M-peak in the slow transients for Chl a fluorescence in leaves grown at 12 and 5°C and in all leaves exposed to high light at 5°C indicated that low temperature significantly affected processes on the reducing side of QA, the primary quinone electron acceptor in photosystem II. Thus S. commarsonii exhibits the characteristics of a plant that is limited by chilling temperatures. Although S. commersonii can tolerate light frosts, its sensitivity to chilling temperatures may result in shoot dieback in winter in its native habitat. The plants may avoid both chilling and freezing temperatures by overwintering as underground tubers.  相似文献

The effects of manganese (Mn) toxicity on photosynthesis in white birch ( Betula platyphylla var. japonica ) leaves were examined by the measurement of gas exchange and chlorophyll fluorescence in hydroponically cultured plants. The net photosynthetic rate at saturating light and ambient CO2 (Ca) of 35 Pa decreased with increasing leaf Mn concentrations. The carboxylation efficiency, derived from the difference in CO2 assimilation rate at intercellular CO2 pressures attained at Ca of 13 Pa and O Pa, decreased with greater leaf Mn accumulation. Net photosynthetic rate at saturating light and saturating CO2 (5%) also declined with leaf Mn accumulation while the maximum quantum yield of O2 evolution at saturating CO2 was not affected. The maximum efficiency of PSII photochemistry (Fv/Fm) was little affected by Mn accumulation in white birch leaves over a wide range of leaf Mn concentrations (2–17 mg g−1 dry weight). When measured in the steady state of photosynthesis under ambient air at 430 μmol quanta m−2 s−1, the levels of photochemical quenching (qP) and the excitation capture efficiency of open PSII (F'v/F'm) declined with Mn accumulation in leaves. The present results suggest that excess Mn in leaves affects the activities of the CO2 reduction cycle rather than the potential efficiency of photochemistry, leading to increases in QA reduction state and thermal energy dissipation, and a decrease in quantum yield of PSII in the steady state.  相似文献

Differences in mitochondrial membrane composition and ultrastructure were studied after storage of cauliflower ( Brassica oleracea , L., Botrytis group) for 5 days at 25°C in air or under controlled atmospheres: 3% O2, 21% O2+ 15% CO2 or 3% O2+ 15% CO2. In air, postharvest senescence involved a 20% decrease in mitochondrial phospholipid content. A large reduction in the relative abundance of phosphati-dylcholine (PC) and in the degree of unsaturation of PC and phosphatidyl ethanolamine (PE) was observed. However, the degree of unsaturation increased in cardiolipin (CL). Storage under 3% O2 did not prevent phospholipid breakdown. Low O2 prevented the relative decrease in PC observed during storage in air and the loss of linoleic acid from PC, but not from PE. This relative protection offered by the low O2 atmosphere was lost under 3% O2+ 15% CO2. The high CO2 atmospheres caused twice as much loss in phospholipids as that observed during storage in air. Extensive loss of mitochondrial protein, a marked decrease in phospholipid to protein ratio, and electron micrograph observations suggest structural alterations in the presence of high CO2.  相似文献

At 14° C, standard metabolic rate (75·1 mg O2 h−1 kg−1), routine metabolic rate (108.8 mg O2 h−1 kg−1), active metabolic rate ( c . 380 mg O2 h−1 kg−1), critical swimming speed (Ucrit 1·7 BL s−1), heart rate 47 min−1), dorsal aortic pressure (3·2 kPa) and ventilation frequency (63 min−1) for triploid brown trout Salmo trutta were within the ranges reported for diploid brown trout and other salmonids at the same temperature. During prolonged swimming ( c . 80% U crit), cardiac output increased by 2·3-fold due to increases in heart rate (1·8-fold) and stroke volume (1·2-fold). At 18° C, although standard and routine metabolic rates, as well as resting heart rate and ventilation frequency increased significantly, active metabolic rate and certain cardiorespiratory variables during exercise did not differ from those values for fish acclimated to 14° C. As a result, factorial metabolic scope was reduced (2·93-fold at 18° C v . 5·13-fold at 14° C). Therefore, it is concluded that cardiorespiratory performance in triploid brown trout was not unusual at 18° C, but that reduced factorial metabolic scope may be a contributing factor to the mortality observed in triploid brown trout at temperatures near 18° C.  相似文献

Leaves of the monoterpene emitter Quercus ilex were exposed to a temperature ramp with 5 °C steps from 30 to 55 °C while maintained under conditions in which endogenous emission of monoterpenes was allowed or suppressed, or under fumigation with selected exogenous monoterpenes. Fumigation with monoterpenes reduced the decline of photosynthesis, photorespiration and monoterpene emission found in non-fumigated leaves exposed to high temperatures. It also substantially increased respiration when photosynthesis and photorespiration were inhibited by low O2 and CO2-free air. These results indicate that, as previously reported for isoprene, monoterpenes may help plants cope with heat stress. Monoterpenes may enhance membrane stability, thus providing a rather non-specific protection of photosynthetic and respiratory processes. Monoterpene emission was maximal at a temperature of 35 °C and was inhibited at higher temperatures. This is likely to be the result of the temperature dependency of the enzymes involved in monoterpene synthesis. In contrast to other monoterpenes, cis- and trans- β -ocimene did not respond to exposure to high temperatures. Cis- β -ocimene also did not respond to low O2 or to fumigation. These results indicate that cis and trans- β -ocimene may have a different pathway of formation that probably does not involve enzymatic synthesis.  相似文献

Routine oxygen consumption rates of juvenile spot, Leiostomus xanthums , were measured over a range of temperatures, salinities and fish weights. As predicted, Q O2 increased with temperature and decreased with body weight. However, Q O2 decreased with decreasing salinity and did not show the expected minimum at isosmotic concentrations. The data are best described by the relationship: log10 Q O2 (mg O2 g−1 h−1) = 0.129 loglo salinity (%0) + 1.604 log10 temperature (°C)-0.1401og10(g)-2.767.  相似文献

Long-term fumigation with different mixtures of air pollutants, such as O3, NO2 and SO2, in combination with potassium deficiency in 4-year-old spruce trees shows clearly that the photosynthetic apparatus reacts very sensitively to environmental stress. Even when no change in photosynthetic oxygen evolution of the leaves was measurable, the content of D-1 protein in the reaction center of photosystem II already increased significantly at low stress conditions. With increasing concentrations of pollutants, the content of D-1 protein decreased considerably. This pattern was observed with all combinations of gases, although they differed in effectiveness. The most potent effect on the content of D-1 protein was found with a combined fumigation of SO2 plus NO2. Obviously, the ratio between synthesis and degradation of the D-1 protein is influenced by stress factors. However, the content of D-1 protein was not strongly correlated with photosynthetic oxygen evolution in whole needles nor with electron transport rate in isolated thylakoids. Under controlled potassium deficiency, effects similar to those observed upon ozone fumigation developed. The results suggest unspecific stress answers for the D-1 content and the photosynthetic reactions rather than specific responses.  相似文献

The role of high light stress in a natural environment was studied on spinach plants ( Spinacia oleracea L. cv. Wolter) grown in the field during the winter season. Fluorescence induction (at 293 K and 77 K) of leaves was used to characterize the stress effects. Night frost with minimum temperatures between – 1.5°C and –7.5°C (i.e. above the'frost killing point'at ca. –11.5°C) led to impaired photosynthesis. This was seen as increased initial fluorescence (Fo), decreased ratio of variable to maximum fluorescence (FV/FM) and lowered rates of O2 evolution. The freezing injury was reversible within several frostless days. Exposure to high light (about 900 mol m–2 s–1) at chilling temperatures in the field caused photoinhibition, manifested as decreased variable fluorescence (FV) and FV/FM ratio without changes in FO. The photoinhibitory fluorescence quenching was not stronger after frost than after frostless nights; synergism between light stress and preceding freezing stress was not observed. Fluorescence induction signals at 77 K showed that FV of photosystems I and II decreased to the same extent, indicating increased thermal deactivation of excited chlorophyll. Photoinhibition was fully reversible at +4°C within 1 h in low light, but only partially in moderate light. Preceding night frosts did not affect the recovery. The photoinhibition observed here is regarded as a protective system of thermal dissipation of excess light energy.  相似文献

When maize ( Zea mays L. cv. LG11) leaves are exposed to low temperatures and high light modifications to both photosystem 2 (PS2) and the light-harvesting chlorophyll a/b protein complex associated with photosystem 2 (LHC2) occur. This study examines the consequences of these modifications for phosphorylation of LHC2 and PS2 polypeptides and the associated changes in electron transport. Maize leaves were chilled at 5°C for 6 h under photon flux densities of 1 500 and 250 μmol m-2 s-1. Thylakoids were then isolated from the leaves and their abilities to phosphorylate LHC2 and PS2 polypeptides and modify electron transport activities were determined. Measurements of chlorophyll fluorescence induction in the thylakoids were also made. Thylakoids isolated from leaves chilled under high light and from leaves kept in the ambient growth environment had similar phosphoprotein profiles. However, polypeptide phosphorylation in thylakoids from the chilled leaves did not produce a decrease in PS2 electron transport. Chilling leaves under low light produced a decrease in the ability of isolated thylakoids to phosphorylate PS2, but not LHC2, polypeptides, which was not associated with any change in the phosphorylation-induced decrease in PS2 electron transport. Chilling under high, but not low, light appears to produce changes in membrane organisation that do not affect the ability of the thylakoids to phosphorylate PS2 and LHC2 polypeptides, but which do prevent the phosphorylation-induced decrease in excitation energy transfer from LHC2 to PS2.  相似文献

Respiration of steelhead trout sperm: sensitivity to pH and carbon dioxide   总被引:1,自引:0,他引:1  
Steelhead trout Oncorhynchus mykiss sperm held in seminal plasma or sperm-immobilizing buffer (pH 8·6) at 10° C consumed O2 at the rate of c . 2 nmol O2 min−1 10−9 sperm; the rate of O2 consumption was not different in sperm held for 4 or 24 h. Decreasing the extracellular pH from 8·5 to 7·5 either by diluting semen with buffer titrated with HCl or by increasing the partial pressure of CO2 in the incubation atmosphere resulted in c . a 40% decrease in the rate of sperm respiration. The data did not, however, support the hypothesis that the precipitous reduction in the capacity for sperm motility that occurs as external pH is reduced is a result of a decrease in cellular metabolism. The rate of O2 consumption of freshly collected semen from different males was not correlated to cellular ATP content or to the proportion of sperm that were motile upon activation; the initial ATP content and sperm motility were positively correlated. The rate of O2 consumption was not significantly increased following sperm activation or by the addition of an uncoupler of oxidative phosphorylation, carbonyl cyanide p -trifluoromethoxyphenylhydrazone, suggesting that these sperm have little, if any, capacity for increased oxidative metabolism.  相似文献

Goldsinny Ctenolabrus rupestris were subjected to rapid, environmentally realistic, reductions in temperature at 2° C increments from 10 to 4° C over a 3-day period in full-strength sea water. In separate experiments, oxygen uptake measurements and ultrasound recordings of heart rate and opercular motion were carried out at regular intervals over the same temperature regime. Mean oxygen uptake rates fell from 0.042 to 0.028 ml O2 g−1 h−1 between 10 and 6° C respectively (Q10=2.71). Between 6 and 4° C mean rates decreased from 0.028 to 0.008 ml O2 g−1 h−1 (Q10=542). Mean opercular motion and heart beat rates decreased from 49.5 and 60.3 beats min−1 respectively at 10° C to 18.7 and 18.0 beats min−1 respectively at 4° C. Most goldsinny subjected to 4° C were observed in a torpid state and would not react to external stimulation. Opercular motion was erratic at 4° C and would at times cease altogether for periods up to 1.3 min duration. Heart movement was diffcult to detect at 4° C and may also have ceased for prolonged periods. Q10 values for opercular motion and heart beat rates recorded between 6 and 4° C were 6.39 and 24.52 respectively compared with values of 2.42 and 2.93 respectively recorded between 10 and 8° C. Such large depressions in metabolism appear not to have been reported previously for a marine fish species. No goldsinny mortalities were recorded at any temperature. The possibility that hypometabolic torpor is an adaptive strategy for goldsinny survival at low environmental temperatures is discussed.  相似文献

Abstract. Third-instar larvae of the goldenrod gall fly ( Eurosta solidaginis Fitch) live inside ball galls on goldenrod plants from summer to the following spring.Because galls are highly exposed to the weather, larvae experience substantial variations in body temperature.This study documents the oxygen consumption of gall fly larvae with regard to the effects of ambient temperature, seasonal conditioning, and prior exposure to subzero temperature.The body mass of larvae doubles between the late summer and the autumn; it subsequently undergoes a modest decline by early winter.The O2, consumption of field-acclimatized larvae increases with ambient temperature, especially between 0 and 10°C (Q10= 2.6-3.4).The thermal sensitivity of metabolism declines at higher ambient temperatures, most notably during the autumn/early winter.After exposure to 15°C for 1 week, autumn and early winter larvae maintain much lower rates of O2 consumption than do late summer specimens.Prior exposure to -5°C for 24 h did not influence the O2 consumption of larvae.Low thermal sensitivity of O2 consumption, especially at higher ambient temperatures, is an energy-sparing mechanism during seasonal inactivity.Indeed, the persistence of this metabolic pattern in larvae exposed to 15°C suggests that they have entered a state of diapause.  相似文献

The oxygen uptake ( V O2), breathing frequency ( f R), breath volume ( V S.R), gill ventilation ( V G) and oxygen extraction (%) from the ventilatory current of four groups of Oreochromis niloticus during graded hypoxia were measured under the following acclimation temperatures: 20. 25. 30 and 35°C. The critical oxygen tensions ( P O2), determined from V O2 v. P O2 of inspired water at each experimental temperature were, respectively. 19±1±3±1. 18±0±4±9, 29±7± 4±1 and 30±2± 0.6 mmHg. The f R remained nearly constant during the reductions of O2 at all the temperatures studied. V G increased discretely from normoxic levels until the P O2 was reached, below which it assumed extremely high values (17-fold higher or more). The increases observed in V G resulted, at all the acclimation temperatures, in an elevation in V S.R rather than in f R. The extraction of O2 decreased gradually from normoxia until the P O2 was reached, below which an abrupt reduction of extraction was recorded, except at 35°C when fish showed a gradual reduction in extraction just below the tension of 80 mmHg.  相似文献

Standard metabolic rate (SMR), active metabolic rate (AMR) and critical oxygen saturation ( Scrit ) were measured in Atlantic cod Gadus morhua at 5, 10 and 15° C. The SMR was 35.5, 57.0 and 78.2 mg O2 kg−1 h−1 and Scrit was 16.5, 23.2 and 30.3%, at 5, 10 and 15° C, respectively. Previously reported SMR for Atlantic cod from arctic waters at 4° C was twice that measured at 5° C in the present study. A possible intraspecific latitudinal difference in the SMR is discussed. The AMR was 146.6, 197.9 and 200.4 mg O2 kg−1 h−1 and the critical swimming speed ( Ucrit ) was 1 6, 1.7 and 1.9 at 5, 10 and 15° C, respectively. The maximum oxygen consumption was found to be associated with exercise, rather than recovery from exercise as previously reported in another Study of Cod metabolism.  相似文献

Brassica rapa L. (rapid-cycling Brassica), was grown in environmentally controlled chambers to determine the interactive effects of ozone (O3) and increased root temperature (RT) on biomass, reproductive output, and photosynthesis. Plants were grown with or without an average treatment of 63 ppb O3. RT treatments were 13°C (LRT) and 18°C (HRT). Air temperatures were 25°C/15°C day/night for all RT treatments.
Ozone affected plant biomass more than did root temperature. Plants in O3 had significantly smaller total plant d. wt, shoot weight, leaf weight, leaf area and leaf number than plants grown without O3. LRT plants tended to have slightly smaller total plant d. wt, shoot weight, root weight, leaf weight, leaf area, and leaf number than HRT plants. For all variables, LRT plants grown in O3 had the smallest biomass, and plants grown in HRT without O3 had the largest biomass.
Ozone reduced both fruit weight and fruit number; LRT also reduced fruit weight but had no effect on fruit number. Ozone reduced photosynthesis but RT had no effect. Conductance and internal CO2 were unaffected by O3 or RT.
These studies indicate that plant growth with LRT might be more reduced in the presence of O3 than growth in plants with HRT, which might be able to compensate for O3-caused reductions in photosynthesis to avoid decreased biomass and reproductive output.  相似文献
