An Evaluation of the Gaspak System in the Culture of Anaerobic Bacteria   总被引:5,自引:5,他引:0  
S ummary : Comparable results were obtained with a commercially available disposable system for the production of H2 and CO2 in an anaerobic jar (the Gaspak system) and with a standard anaerobic jar procedure. Both systems were used with the Baird & Tatlock anaerobic jar and a room temperature catalyst. The test organisms included strict anaerobes such as Clostridium oedematiens type D and Cl. tetani. Comparative tests were also made with Bacteroides species from laboratory stock cultures and with freshly isolated strains of Bacteroides. On occasion, higher recoveries were obtained with the standard system when relatively demanding models were set up but it has limitations, having been developed initially for research purposes. The Gaspak system probably gives more reliable and more reproducible results in a laboratory in which experience with strict anaerobes is limited or sporadic.  相似文献

A specific effort was made to obtain monoclonal antibodies that bind to macromolecules that play a role in the development of the nervous system. It was considered that good candidates for such molecules were those that were only transiently present in the embryonic nervous system. Hybridomas were prepared from spleen cells taken from mice that had been immunized with nerve cords from cockroach embryos at the 43-50% stage of development. The hybridoma supernatants were screened for antibody binding to frozen sections of both embryonic and adult thoracic ganglia. Cell lines that produced monoclonal antibodies that transiently bound to the embryonic nervous system were saved and cloned. These developmental stage-specific monoclonal antibodies either did not bind to the adult nervous system or bound to it with a pattern very different from that in the embryonic nervous system. The developmental stage-specific antigens detected by these monoclonal antibodies were organized into four categories based on the part of the embryonic nervous system in which they were transiently localized. These include binding to the cell bodies of all neurons, cell bodies of subsets of neurons or neuroblasts, subsets of axons, and the neuropile. Preliminary biochemical characterization of the antigens showed that many of these antibodies were recognizing carbohydrate epitopes. Functions for these antigens, most of which are components of the cell surface, are tentatively proposed.  相似文献

Phosphate starvation derepresses a high-affinity phosphate uptake system in Saccharomyces cerevisiae strain A294, while in the same time the low-affinity phosphate uptake system disappears. The protein synthesis inhibitor cycloheximide prevents the derepression, but has no effect as soon as the high-affinity system is fully derepressed. Two other protein synthesis inhibitors, lomofungin and 8-hydroxyquinoline, were found to interfere also with the low-affinity system and with Rb+ uptake. After incubation of the yeast cells in the presence of phosphate the high-affinity system is not derepressed, but the Vmax of the low-affinity system has decreased from about 35%. Phosphate supplement after derepression causes the high-affinity system to disappear to a certain extent while in the meantime the low-affinity system reappears. The results are compared with those found in the yeast Candida tropicalis for phosphate uptake.  相似文献

An immunohistochemical method that uses anti-tubulin was utilized to observe the development of the enteric nervous system in chick embryonic duodenum. Neural crest cells, and enteric neuroblasts, or enteric ganglia, which derive from neural crest cells were clearly shown as sharp immunoreactive regions of tubulin. The distributions of enteric neuroblasts and enteric ganglia in chick duodena were in agreement with results of previous reports in which different techniques were used. The initial stage at which cells of neural crest origin were present in the duodenal walls (4-day-old embryos) was earlier than the initial stage (about 6-day-old embryos) reported earlier. This was verified by transmission electron microscopy. Also, the tubulin that is a component of the enteric nervous system was shown to be stable at a low temperature. This tubulin-immunostaining method provides a useful histochemical technique with which to study the development of the enteric ganglion and the function of tubulin as a component of the enteric nervous system.  相似文献

Relationship between bacteria and ciliates was investigated in continuous cultures with and without the recycling of flocculated biomass. Monocultures of ciliates of various ecological types were used together with mixed cultures of bacteria or bacteria and flagellates. The effect of ciliates on the distribution of flocculated and dispersed, biomass was tested with respect to characterizing an optimum mixed culture for activated-sludge treatment. Monoprotozoal populations were not successful in stabilizing the morphology of a bacterial community in a system with recycling of flocculated biomass. Stabilization was observed (with particular ecological types of ciliates) in the system with a high residence time without recycling and/or in the system with the recycling inoculated by both flagellates and ciliates.  相似文献

The sleep-wake cycle is virtually the most prominent circadian rhythm in mammals. In the timing system of sleep and wakefulness, the intrinsic neuropeptide systems of the suprachiasmatic nuclei (SCN) may play an important role. To elucidate this possible influence in the golden hamster, the immunoreactivity patterns of the suprachiasmatic gastrin-releasing peptide (GRP), vasoactive intestinal polypeptide (VIP), and arginine-vasopressin (AVP) systems were investigated in relation to the day-night and sleep-wake cycle by use of immunocytochemistry combined with semiquantitative planimetric analysis. For the GRP system, the highest level of immunoreactivity (expressed as area density) was observed in sleeping hamsters. Intermediate levels were found in awake, motorically active evening animals, whereas the lowest levels of immunoreactivity were detected in awake, motorically inactive hamsters studied in the morning. The immunoreactivity of the VIP system showed a completely opposite pattern, indicating highest area density in awake morning, intermediate area density in awake evening and lowest area density in sleeping golden hamsters. The immunoreactivity pattern of the AVP system, displaying highest levels in sleeping individuals, was virtually identical to that of the GRP system. Together with the related signs of neuronal activity, the present results favor an important role of these neuropeptide systems for the integration of central nervous information related to the sleep-wake cycle with photic information of the retinal input.  相似文献

The lactoperoxidase system (LPS), a natural bactericidal system in milk, was investigated for its activity against salmonellas in vivo and in vitro. In acidified raw milk, in which the LPS was supplemented with an exogenous supply of H2O2, the numbers of salmonellas decreased rapidly. Different salmonella serotypes were affected to the same extent; rough strains, however, were more susceptible than smooth strains. When calves were fed on fresh milk, containing the LPS, and challenged with Salmonella typhimurium in doses of either 10(9) or 10(10), the clinical findings and salmonella excretion patterns were similar to those of control calves fed on heated milk. It was concluded that further studies, perhaps in the field, are necessary to evaluate LPS as a possible non-antibiotic system to control salmonellosis.  相似文献

The localization of calcium in the membrane system of human platelets was determined by ultrahistochemical methods equipped with an electron probe x-ray microanalyzer. After potassium oxalate-glutaraldehyde treatment large amounts of electron opaque precipitates were observed around the membrane of the surface connected system. Electron probe x-ray microanalysis clearly defined that the precipitates were composed of calcium oxalate. The localization of calcium on the membrane of the surface connected system was also confirmed even after treatment of the platelets with potassium antimonate-OsO4. These results support a model which depicts the surface connected membrane system taking part in the store and the transport of calcium.  相似文献

The MicroFoss system was evaluated for its ability to detect Listeria species in environmental samples. The sensitivity and specificity of the MicroFoss were determined in relation to a standard culture method for Listeria detection. The sensitivities of both the MicroFoss and standard culture methods were similar (88.4%-MicroFoss, 90.7%-Culture) based on the total number of positive results obtained by both methods. The MicroFoss system detected Listeria spp. in 12 samples, which were not detected by culture, and the culture method detected Listeria spp. in 15 samples, which were not detected by the MicroFoss method. This was likely due to uneven distribution of low levels of Listeria organisms in the split sponge samples used to assess the performance of these test methods. The specificity value determined for the MicroFoss system was 92.7%. The majority of microbes causing false positive results in the MicroFoss system were Bacillus species, which were readily distinguishable from Listeria species by a simple Gram stain and morphological features. Listeria monocytogenes (89.4%-MicroFoss, 88.0%-Culture) and Listeria innocua (8.8%-MicroFoss, 7.7%-Culture) were the most common isolates of Listeria detected by the two test methods, with L. monocytogenes being the most predominant isolate detected. The highly comparable results and rapid nature of the MicroFoss system demonstrate its effectiveness as a detection system for species of Listeria in environmental samples. The fact that the sensitivity of the MicroFoss system was similar to that of the culture method and the Listeria results were obtained within 48 h of testing, support the use of the MicroFoss as an alternative rapid method for screening large numbers of environmental samples for Listeria spp.  相似文献

Cytokines, the polypeptide mediators of the immune system, were shown to exert numerous actions on endocrine functions. Bidirectional links based on the sharing of mediators and receptors between the immune and neuroendocrine systems lead to the concept of the immune-neuroendocrine system that seems to constitute an important and sophisticated regulatory system in the homeostasis. Several cytokines were found to be involved in the pathogenesis of diseases of the endocrine system. In this brief review, we attempt to present a general outline of the local actions of cytokines on cells of endocrine organs with an emphasis on disease etiology (pituitary tumours and autoimmune endocrine diseases in particular).  相似文献

The Australian Weed Risk Assessment system has been tested in a number of countries and geographical areas since its introduction in 1997, and is widely considered to be an accurate method of predicting the risk of invasiveness of new plant introductions. We evaluated the system against 152 plant species with at least a 50-year introduction history in Canada, including major and minor weeds and species which have not naturalized. Four questions that referred explicitly to Australian conditions were replaced with appropriate equivalent questions for Canada. The weediness of each species was independently rated by a panel of Canadian agricultural, botanical, and conservation experts. Using the standard cut-off scores, the system correctly rejected all major and 86% of minor weeds. However, it also incorrectly rejected 44% of non-weedy species. The diagnostic power of the system, as measured by the receiver operating characteristic curve, was similar to but somewhat lower than that found in other regions where the system has been evaluated. Answers to 23 of the 49 questions were not significantly associated with experts’ weediness ratings, indicating that a simplified system could give equally reliable results for Canada. Experts’ ratings were strongly related to cold-hardiness, suggesting that the system could be improved by making better use of data on climatic tolerances. We suggest that performance would also be improved by combining likelihood- and consequence-related scores in a multiplicative rather than additive way.  相似文献

Theoretical estimation of contribution of the electrostatic interactions to pre-orientation of ribonuclease subunits in process of complex formation was carried out. The subunit was considered as a multipole consisting of partial charges of all atoms of the molecule. The object of investigation was a system of two subunits with their centers of gravity fixed at some distance in vacuum. It was proposed that each subunit independently could rotate freely around its fixed center of gravity. The relative orientation states of the subunits in such system were searched at which the system has electrostatic energy minima (equilibrium states). In first approximation the equilibrium states were found using especially designed approximate method for electrostatic interaction energy calculation, which permitted to calculate and compare the energies of the system in 24(5) (approximately 8 10(6)) states with different mutual orientation of subunits. The angular coordinates of the found equilibrium states were further specified by calculation with gradient sliding method. Angular coordinates of the equilibrium states and the shapes of energy surface cuts along each coordinate angle were calculated also for the intersubunits distances diminished down to 50 angstroms. The dispersions of the angular coordinates of equilibrium states caused by heat movement (at T=300 degrees) and their changes with shortening the distance between centers of gravity of subunits were estimated. Mutual orientation of subunits in the equilibrium states of the system under consideration was found to be similar to their mutual orientations in complex. Also it was found that relaxation time of the system, caused by electrostatic interaction of subunits, after removing the system from an equilibrium state, is much less in vacuum than the mean time between their Brownian collisions at room temperature. It follows from these results that in the case of ribonuclease in vacuum the electrostatic interactions of its subunits must be strong enough to realize the effective pre-orientation of subunits during their Brownian approach from distances of the order 100 angstroms. Preliminary consideration taking into account the effect of surrounding water molecules on the electrostatic interactions of ribonuclease subunits showed that weakening of the interaction must be much less than in the case when one uses in its calculation the macroscopic dielectric permeability value equal to 80. So the results obtained for vacuum seem to be true for water solution also. More strict theoretical analysis of this problem will be carried out in the following publication.  相似文献

The hypothesis that the properties of the stretch reflex could be changed dependent on the appointed task of the motor system was tested by measuring the mechanical output impedance of the hand of a human subject. Small disturbing torques were exerted on the hand rotating in the wrist joint in palmar and dorsal direction. The angular position, velocity and acceleration were recorded during 180 msec after the start of the disturbing torque. Within the linear range differences were found between the situations that (1) the subject kept his hand in a fixed position (posture) and (2) the subject tracked a simple and slow moving target (tracking). A simple model of the peripheral part of the motor system with the spinal reflex mechanism was made to analyse the responses. With the help of this model the changes in the reflex sensors were distinguished from changes in the mechanical system of the hand with the muscles attached to it and from changes in the effect of an activation of the muscle. During posture the responses were affected by the attention of the subject. An important part of these changes was a shift in the delay time of the reflex. During tracking the responses changed markely dependent on the velocity of the hand. When this velocity is larger than about 0.05 rad.sec?1 then the strength of the reflex is approximately suppressed to about a half of the value during posture. Above this velocity the mechanical system of the hand with the muscles attached to it appeared to be less damped. The suppression of the reflex is caused by a decrease of the gain of the feedback sensors to about a third and an increase in the gain of the system of muscles with their load to about one and a half. It was concluded that the gain of the feedback sensors is controlled by signals from supraspinal levels.  相似文献

A new expert system developed on a Macintosh personal computer using a commercially available artificial intelligence shell was compared with four different discriminant functions (DFs) for the differentiation of microcytic anemia into etiologic categories. Several databases were used with a different composition but all contained at least some samples from thalassemic individuals and from patients with iron deficiency anemia. The DFs analyzed were those proposed by England and Fraser, Green and colleagues, Mentzer, and by Shine and Lal. None of the databases performed satisfactorily when used singly, whereas very high false-positive rates were obtained by one of them. The diagnostic efficiency was somewhat improved by combining several DFs. An expert system using an artificial intelligence "shell" with an "interference engine" was developed using cluster analysis and a set of learning examples. The input necessary for the system to achieve a conclusion consists of MCV, RBC, and RDW as well as a statement as to whether the patient has anemia. Based upon the values of these parameters, the expert system will give an "advice" regarding the probabilities for thalassemia, iron deficiency, and/or other probabilities such as previous transfusions, anemia of chronic disease, laboratory error, etc. In a prospective trial, the system functioned with an accuracy of better than 85%.  相似文献

目的:探索一种简便定量分析系统,通过检测HBV携带者血清中的HBV X区DNA,fRNA和trRNA拷贝数,为提高隐匿性感染期低复制状态HBV检测效果提供可能。方法:从血清中提取HBV DNA和RNA,对Core区、X区DNA进行PCR扩增,使用半巢式PCR法对fRNA、trRNA在同一离心管中进行反转录并扩增,选取大小相近的质粒作为竞争模板对其进行定量。并对拉米夫定干预治疗14周前后的患者血清中HBV XDNA、Core DNA、fRNA和trRNA拷贝数进行检测与比较。所有检测结果均通过southern杂交进行验证。结果:建立了针对DNA的定量分析系统及针对RNA的RT-PCR定量分析系统,并且阐明了阿米夫定治疗前后HBV DNA和X区RNA结构和数量的变化。治疗前治疗后DNA和RNA的拷贝数均下降。Core DNA下降显著,为103-104倍,而XDNA拷贝数下降102倍。而fRNA和trRNA仅有小幅下降,为10倍左右。结论:可以通过竞争性PCR方法对血清中HBV DNA和RNA进行定量检测,以期为HBV病毒的诊断提供更充足依据。  相似文献

Chronic experiments were carried out on five dogs with a defensive instrumental reflex (IR) associated with the maintenance of the flexor posture. Was studied the influence of carbocholine bilateral microinjections (0.05-0.10 mcg) into the nucleus accumbens (NAC) on the realization of the IR and on the behavioural differentiation of acoustic signals in a defensive situation. Cholinergic system of the NAC was shown to participate in both the motor and sensory mechanisms connected with the realization of motor responses to defensive and differentiation signals. Activation of the cholinergic system of the NAC led to an "improvement" of behavioural signal differentiation probably due to an increase of attention to significant stimuli. The cholinergic system of the NAC participated in the regulation of voluntary movements. Its influence was mainly excitatory and had unspecific and prolonged character.  相似文献

The effect of the neurodepressing hormone (NDH) was studied on different identified motoneurons in the abdominal ganglia of the crayfish Procambarus bouvieri (Ortmann). Although differences in sensitivity were apparent, all the neurons tested responded to NDH with a reduction in spontaneous firing rate, which lasted as long as NDH was present, and, depending on the concentration and time of action of the hormone, for even longer periods. NDH activity was determined in the various parts of the central nervous system of the crayfish, being highest in the eyestalk, gradually diminishing away from the eyestalk, with a cephalo-caudal gradient, being lowest in the abdominal ganglia. High levels of NDH activity were detected in the blood. After eyestalk ablation, NDH concentration steadily diminishes in the blood and central nervous system, until virtually disappearing after 4 days; from day 5 onwards, the activity is recovered up to its original levels. NDH synthesis takes place with a time constant of approximately 3 hr in cultured isolated segments of central nervous system, being highest in the eyestalk.  相似文献

By means of the method for revealing cholinesterases the alterations in the nervous system of Convoluta convoluta after mechanical lesion were studied. After a transversal transection of the animal into two pieces the anterior part of the body acquired the normal form within 3-4 days; the nervous system underwent inconsiderable transformations associated with a formation of a plexus in the injured area. The posterior parts of the body regenerated slowly. During the first days disintegration of the resting parts of the nervous system was observed. Later on the diffuse cholinesterase activity suggesting the beginning of the brain formation was manifested in the wound field. Plexus strands "grow" in the caudal direction from the brain in parallel with its development. Within three weeks the nervous system of the animals which had restored their mouth opening, body shape and normal behaviour consists of the brain and plexus not arranged in nervous trunks. The reaction of the nervous system to the ablation of a part of the brain and to a continuous mechanical injury were studied. The data obtained show a high morphological lability of the nervous system of Convoluta convoluta, its ability to radical structural rearrangements.  相似文献
