Metabolic syndrome is more prevalent in men than in women. In an experimental dietary model of metabolic syndrome, the high-fructose-fed rat, oxidative stress has been observed in males. Given that estradiol has been documented to exert an antioxidant effect, we investigated whether female rats were better protected than males against the adverse effects of a high-sucrose diet, and we studied the influence of hormonal status in female rats. Males and females were first fed a sucrose-based or starch-based diet for 2 weeks. In the males, the plasma triglyceride (TG)-raising effect of sucrose was accompanied by significantly lowered plasma alpha-tocopherol and a significantly lowered alpha-tocopherol/TG ratio (30%), suggesting that vitamin E depletion may predispose lipoproteins to subsequent oxidative stress. In males, after exposure of heart tissue homogenate to iron-induced lipid peroxidation, thiobarbituric reactive substances were significantly higher in the sucrose-fed than in the starch-fed rats. In contrast, in sucrose-fed females, neither a decrease in vitamin E/TG ratio nor an increased susceptibility of heart tissue to peroxidation was observed, despite both a significantly decreased heart superoxide dismutase activity (14%) and a significant 3-fold increase in plasma nitric oxide concentration compared with starch-fed females. The influence of hormonal status in female rats was then assessed using intact, ovariectomized, or estradiol-supplemented ovariectomized female rats fed the sucrose or starch diet for 2 weeks. After exposure of heart tissue to iron-induced lipid peroxidation, higher susceptibility to peroxidation was found only in ovariectomized females fed the sucrose diet compared with the starch group and not in intact females or ovariectomized females supplemented with estradiol. Thus, estrogens, by their effects on antioxidant capacity, might explain the sexual difference in the pro-oxidant effect of sucrose diet resulting in metabolic syndrome in rats.  相似文献

In humans, the relationship between the prenatal testosterone exposure and the ratio of the second and the fourth digits (2D:4D) has been extensively studied. Surprisingly, data on this relationship have thus far been lacking in experimental animals such as rats. We studied the effect of maternal testosterone enhancement during pregnancy on the digit ratio and open field activity of adult progeny in Wistar rats. Elevated levels of maternal testosterone resulted in lower 2D:4D ratios and an elongated 4D on the left and right forepaws in both males and females. We found no sex difference in 2D:4D in control animals. In the open field test, control females were more active than control males and testosterone females, while the activity of testosterone females did not differ from that of control males. We found a positive correlation between motor activity and the right forepaw 2D:4D ratio of control males and females. Prenatal exposure to testosterone resulted in the disappearance of this correlation in both males and females. Our results show that elevated levels of testosterone during the prenatal period can influence forepaw 4D length, 2D:4D ratio, and open field motor activity of rats, and that these variables are positively correlated. Thus, this approach represents a noninvasive and robust method for evaluating the effects of prenatal testosterone enhancement on anatomical and physiological parameters.  相似文献

To determine the chronic toxicity of diesel exhaust, 2,150 F 344 rats were made to inhale various dilutions of exhaust under specific pathogen-free conditions for 16 hours a day, 6 days a week, for 30 months. Ten experimental groups, each with 120 male and 95 female 5 week-old rats, were set up and 14 males and 9 females were usually sacrificed for measurement of the 51 hematological and clinico-biochemical parameters at 7, 13, 19, and 25 months of age, respectively. These parameters were also examined in all surviving rats after 30 months of exhaust inhalation. In this experiment, no remarkable change in any of the parameters was observed in the periodic examinations. Therefore, tendencies of change in parameters were examined by comparing histograms for control and experimental groups. For the reference histograms, we used the values obtained from 144 males and 116 females aged 13-31 months. The first histogram was based on data from 1,352 rats, and the range from the minimum to the maximum value was divided into eight parts. When almost all the values were centered on a peak in the first histogram, this peak was divided into a further eight parts in the second histogram. Histogram evaluation clarified the differences between the control and experimental groups better than general statistical methods such as Student's t-test.  相似文献

The study was designed to examine the effect of oleanolic acid on cauda epididymal sperm motion using a computer-aided sperm analysis system and to elucidate the relationship between sperm motion and fertility, as a tool for contraceptive studies. Oleanolic acid-polyvinylpyrrollidone suspension was orally administered to adult male Wistar rats for 30 days, followed by a 14-day drug withdrawal from half of the rats in the group. Control rats received only polyvinylpyrrollidone. All males were mated with untreated females. Treated males failed to impregnate females, whereas control and oleanolic acid withdrawn males achieved 100% pregnancies. Sperm motion analysed on the Sperm Motility Quantifier (SMQ) showed significant differences in linearity (P < 0.001) and wobble (P < 0.01) between control and treated groups. However, the curvilinear velocities were not significantly different (P > 0.05) among all the groups. Sperm motility patterns verified differences among kinematic parameters.  相似文献

This research was carried out to examine the hypothesis that the absence of fathers promotes a different spacing behaviour in juveniles Calomys musculinus at the beginning of the breeding period. The study was carried out in four 0.25-ha enclosures (two control and two experimental), in a natural pasture, between November 2003 and February 2004. In this study the fathers were removed from the experimental enclosures after juveniles were born. Home-range size depended on sex of juveniles and treatment (father removal). In control and experimental enclosures, female home-range sizes were always smaller than male home-ranges. Male home-ranges were always larger in experimental enclosures than in control enclosures. Treatment and overlap type (intra- and inter-sexual) were not independent. The overlap proportions of male home-ranges were greatest in experimental enclosures than in control enclosures, in both the overlap types (male/male, males/females). The intra- (females/females) and inter-sexual (females/males) overlap proportions of female home-ranges were independent of treatment. In C. musculinus, at the beginning of the breeding period and in absence of adult males, juvenile males increase their home-range size and therefore the degree of inter- and intra-sexual home-range overlap as a mechanism for enlarging the number of receptive females that they encounter.  相似文献

Summary Feeding the secondary plant compound 6-Methoxybenzoxazolinone (6-MBOA) during winter to a free living population of Microtus townsendii accelerated breeding in females. Both the recruitment of young and sexual maturation of these young were advanced by four weeks in comparison with a control population. Overwintered females supplied with 6-MBOA matured at a lower body weight than control females. But there was no such weightat-maturity difference between the grids for males. During April and May 72% of all juveniles captured came from the experimental population. Winter reproduction and early recruitment of young stimulated by 6-MBOA could have important population consequences for these voles.  相似文献

Muscovy ducks display marked sexual dimorphism for body weight. The aim of our study was to analyse the possible relationships between this dimorphism and plasma levels of T3, GH and IGF-I from hatch to 15 weeks of age. We found no significant effect of sex or age on plasma T3 which remained relatively stable about 6 nM x L(-1). Plasma GH levels were highest (14-24 ng x mL(-1)) but highly variable during the first four weeks in females and during the first seven weeks in males. Then, they decreased and remained low (3-4 ng x mL(-1)) until the age of 15 weeks. A trend for higher GH concentrations in males compared to females was observed across the experimental period. The difference was maximum around 6-7 weeks of age. Plasma IGF-I levels rose sharply between 2 and 3 weeks of age and remained high until 6-7 weeks of age for the females and 13 weeks of age for the males then started to decrease. For the whole experimental period, significantly higher IGF-I levels were measured in males. The differences between sexes were significant between 7 and 12 weeks of age and at 14 weeks of age. The differences in circulating GH and IGF-I levels between sexes suggest that the somatotrophic axis is implicated in Muscovy duck sexual dimorphism.  相似文献

In the open-field test, male rats usually defecate more and ambulate less than females. A strain was studied in which the males defecate less than females, while still ambulating less. Infants of this strain were castrated (males) or injected with testosterone propionate (females) and tested in the open field as adults. There were significant effects of these neonatal treatments on ambulation, but not on defecation, in contrast to previous reports for the latter measure in rats showing a sex difference in defecation taking the usual direction. It is suggested that absence of the usual direction of a sex difference normally under the developmental control of androgen may indicate a genotype which has escaped from such control. Finally, a number of 'sibling' effects' were observed, i.e. effects on the adult behaviour of the members of one sex in a litter produced by treatments administered in infancy to members of the other sex.  相似文献

The ventromedial hypothalamus (VMH) is one of several sexually dimorphic nuclei that regulate mating behavior, and is rich in steroid hormone receptors and aromatase activity. We looked at the contribution of the androgen receptor (AR) to the volume of the VMH in rats by measuring each of the four subdivisions of the VMH in 90 day old male, female, and XY male rats carrying a mutant AR allele (tfm), which renders animals largely unresponsive to androgens. Confirming published reports, total VMH volume was greater in wild-type males than in females (P<0.01). The mean total volume of the VMH in TFM males was intermediate, but not significantly different from either females or males (Ps>0.10). The sex difference in VMH volume was primarily accounted for by the ventrolateral subdivision (VMHvl), which in both females and TFM males was significantly smaller than in wild-type males (Ps<0.005). There was no significant sex difference in the volume of the other three subdivisions of the VMH. Neuronal somata were larger in males than females in VMHvl, central VMH (VMHc) and the dorsomedial VMH (VMHdm), with TFM males having feminine neuronal somata in the VMHdm and VMHc. These data suggest that AR plays a role during sexual differentiation of the VMH, imparting its greatest effect in the VMHvl. ARs may regulate aromatase expression or activity to affect estrogen receptor activation, or may act independently of estrogen receptors to influence VMH morphology.  相似文献

The aim of this study was to find out how a long-term shielding of the geomagnetic field (GMF) affected the immune system of rats. Male and female Wistar rats were kept up to an age of 2 months in a natural GMF (about 37 microT). Afterwards, the rats were divided into four groups (males and females separately): control rats were maintained in ambient GMF, while experimental animals were housed under conditions of a weakened GMF (below 12 microT) achieved with steel cages. After 6 months, the rats were sacrificed by decapitation. Spleens and thymuses were isolated and weighed. Peritoneal cells were eluted and cultured in vitro to study their ability to produce nitric oxide (NO) and to synthesize superoxide anion (O2(-)), important microbicidal molecules of macrophages. The number of macrophages was estimated by a crystal violet staining method. We found that the long-term shielding of the GMF could influence the functioning of the immune system in a sex-dependent manner. The deprivation of the GMF delayed physiological thymus involution, that effect being more strongly expressed in females. The weakening of the GMF resulted in an increased number of peritoneal macrophages, especially in males. The shielding of the GMF diminished the ability of macrophages to release NO and to synthesize O2(-), those effects being more powerfully expressed in males and females, respectively. It is proposed that the observed changes in the immune system occur as a consequence of the protective effect of GMF shielding on the circadian rhythm-dependent level of melatonin.  相似文献

We studied demographic effects of the mitochondria-targeted antioxidant SkQ1 on free-breeding Campbell dwarf hamsters (Phodopus campbelli, Thomas, 1905, Rodentia, Cricetidae) in an outdoor vivarium with seasonally varying day length and temperatures. The animals were kept in pairs from their young age. We removed litters from parental cages at their age of 25 days. Experimental hamsters received daily 50 nmol/kg SkQ1 with water by oral dosing, whereas control animals received water. SkQ1 had no effect on the lifespan of either males or females in reproductive pairs. Mortality among females was higher than among males irrespective of SkQ1 treatment, this being related to higher costs of reproduction in females. However, SkQ1 accelerated breeding in pairs in the first half of the reproductive period of a year. Although there were no statistical differences in body mass of males and females between experimental and control animals during most of their life, SkQ1-receiving males had higher body mass at the end of their life. The opposite tendency was characteristic for old females. One-year-old males and females of the experimental and control groups showed no difference in intensity of immune response to sheep red blood cells. The dermal hypersensitivity response to phytohemagglutinin (test for T-cell immunity) was significantly higher in SkQ1-treated 1- and 1.5-year-old males. This was not true for females. There was a tendency toward increased density of the neutrophil population in blood in 1-year-old SkQ1-treated males. However, experimental males showed no difference from control males in the activity of the “peroxidase-endogenous hydrogen peroxide system” of neutrophils. The background level of stress estimated by the concentration of cortisol in blood serum was significantly lower in the SkQ1-treated males during autumn adaptive adjustment of the organism. A similar trend was also observed during the January frosts, when the background level of stress was rather high. We observed no differences between cortisol concentration in experimental and control animals during the reproductive period in early spring and mid-summer. We tend to interpret the absence of geroprotective effect of SkQ1 on free-breeding dwarf hamsters by its ability to intensify breeding. The objective of this study was to investigate the sex-dependent regulation of skeletal muscle uncoupling protein (UCP)3 mRNA expression in response to overweight and its relationship with serum levels of free fatty acids, leptin, and insulin. Two obesity models were used: rats made obese by feeding them with a cafeteria diet for 14 wk, and postcafeteria overweight rats fed a chow diet for 10 wk after consuming the cafeteria diet for 14 wk. The effects of 24-h fasting were studied in postcafeteria rats and their age-matched controls. The cafeteria rats ate a high-fat diet and attained an excess body weight that was higher in females (+59%) than in males (+39%). A trend to higher induction of abdominal muscle UCP3 mRNA in male rats than in females after cafeteria diet was apparent (+116% increase vs. +26% increase). Postcafeteria male but not female rats still showed the tendency to have increased UCP3 mRNA levels relative to their age-matched controls. A linear regression analysis showed a significant positive correlation of the UCP3 mRNA levels with overweight and with serum levels of leptin and insulin in males, but not in females, and no correlation with serum free fatty acid levels. A subsequent correlation analysis and a multiple linear regression analysis showed that overweight was the only parameter actually related to UCP3 mRNA levels in males. Fasting-induced upregulation of muscle UCP3 mRNA levels was higher in males (5- to 7-fold) than in females (3- to 4-fold). Our results point to the existence of sex-associated differences in the control of muscle UCP3 expression in response to overweight and fasting, with an impaired induction in female rats under both conditions. The correlation of abdominal muscle UCP3 mRNA expression with overweight in males could be related to their relative resistance to gain weight after chronic overeating of a cafeteria diet, by the purported role of UCP3 in the regulation of lipid utilization.  相似文献

The effects of novel or relevant (a single exposure to experimental chamber) and irrelevant (20 exposures to experimental chamber) stimuli on the levels of serotonin (5-HT) and its metabolite 5-hydroxyindoleacetic acid (5-HIAA) in the frontal cortex, striatum, and nucleus accumbens in the left and right hemispheres were studied in male and female rats. It was found that 5-HT and 5-HIAA contents in the frontal cortex changed in response to neither relevant nor irrelevant stimuli. However, there were hemispheric difference in 5-HT and 5-HIAA in the frontal cortex of intact animals. The level of 5-HT in males and the level of 5-HIAA in females were higher in the left frontal cortex. In females, the level of 5-HIAA in the left striatum decreased in response to the novel stimulus. Sex differences in: a) 5-HT metabolism (increase in the level of 5-HIAA in males and increase in 5-HT in females) and b) lateralization (the striatal 5-HT metabolism in males changed bilaterally and only in the left hemisphere in females) were observed in reactions to irrelevant stimuli. Both in male and female rats, serotonin content in the nucleus accumbens changed only in response to the irrelevant stimuli. The 5-HT level increased in the left and right hemispheres independently of sex, but hemispheric difference was revealed only in females, in which the serotonin level was higher in the left nucleus accumbens. It is concluded that serotonergic neurotransmitter mechanisms are involved in hemispheric and sex differences in selective attention.  相似文献

The sex difference observed in frequency of rats exhibiting the serotonin behavioral syndrome induced by pargyline/1-tryptophan depends on hormonal state. Castration eliminated the sex difference in drug response in adult and prepubertal males, whereas ovariectomy had little effect. Dihydrotestosterone administration to males (10-30 days) reinstated the sex difference, but had little effect in females. Testicular feminized mutants (Tfm/y), deficient in androgen receptors, respond like females. Estrogen administration has no effect in either sex. Manipulation of the hormonal environment on postnatal days 0-7 (blockade of aromatization in males or estradiol administration to females) has no effect on the expression of the sex difference when the animals were tested as adults. Therefore, androgens acting via androgen receptors appear to mediate this subsensitivity of male rats to the drug challenge. The results of these experiments indicate that sex and hormonal environment are important variables in determining the experimental and perhaps clinical responses to drugs.  相似文献

Human adolescents exhibit higher levels of novelty-seeking behaviour than younger or older individuals, and novelty-seeking is higher in males than females from adolescence onwards. Gonadal hormones, such as testosterone and estradiol, have been suggested to underlie age and sex difference in response to novelty; however, empirical evidence in support of this hypothesis is limited. Here, we investigated whether suppressing gonadal hormone levels during adolescence affects response to novelty in laboratory rats. Previously, we have shown that male adolescent Lister-hooded rats (postnatal day, pnd, 40) exhibit a stronger preference than same-aged females for a novel object compared to a familiar object. In the current study, 24 male and 24 female Lister-hooded rats were administered with Antide (a gonadotrophin-releasing hormone antagonist), or with a control vehicle solution, at pnd 28. Antide provided long-term suppression of gonadal hormone production, as confirmed by ELISA assays and measurement of internal organs. Response to novel objects was tested at pnd 40 in Antide-treated and control subjects using a ‘novel object recognition’ task with a short (2-minute) inter-trial interval. In support of previous findings, control males exhibited a stronger preference than control females for novelty when presented with a choice of objects. Antide-treated males exhibited a significantly lower preference for novel objects compared to control males, whilst Antide-treated females did not differ significantly from control females in their preference for novelty. Antide treatment did not affect total time spent interacting with objects. We discuss how gonadal hormones might influence sex differences in preference for novelty during adolescence.  相似文献

The platelet biosynthesis of total lipids, lipid fractions and fatty acids was determined by incorporation of [14C]acetate in normal and castrated rats of both sexes. Comparison was made between animals fed laboratory chow alone, and animals receiving, in addition, for 4 days by stomach tube a saturated (cream) or polyunsaturated (sunflower seed oil) fat. In male rats, the polyunsaturated fat increased slightly the total platelet lipid biosynthesis. The saturated fat drastically reduced it by 43% in comparison to the polyunsaturated fat-fed animals. Normal female rats did not exhibit a similar difference in the platelet lipid synthesis. However, the inhibitory effect of saturated fat on lipid synthesis could be observed in castrated females, although it was less pronounced (27% reduction) than in castrated or normal males (43%). Administration to castrated males of estradiol for 1 month almost completely inhibited the difference induced by the feeding of the different fats in the lipid platelet synthesis of male rats. This difference in the platelet lipid biosynthesis between male and female rats, normal and castrated, was observed mostly in the phospholipid (especially phosphatidylcholine), monoacylglycerol and triacylglycerol fractions and affected primarily the synthesis of the three main saturated fatty acids, 14:0, 16:0 and 18:0. Thus, it seems that, in rat, the short-term administration of a saturated fat induces drastic changes in the platelet lipid biosynthesis, but only in males. Mechanisms producing inhibition of remating in mated female Mediterranean fruit flies Ceratitis capitata, were investigated by matings with surgically altered males. Comparison of remating by females mated with either intact control males or males with a shortened penis, showed that ejaculate or a physical stimulus of penis insertion caused remating inhibition for at least 10 days after first mating. Remating frequency at two days after mating was significantly higher in females mated to castrated (spermless) males than in females mated to sham-operated control males. This difference disappeared by day four after mating, indicating that sperm cause a shorter-term inhibition of remating than does a normal first mating. Other factors in addition to sperm must therefore play a role in inhibition of remating.  相似文献

Effects of gonadal steroids on conspecific odor preference for either (1) sexually active male or active female, (2) sexually active or gonadectomized (gdx) males, (3) sexually active or gdx females, and (4) gdx males or gdx females were determined in male and female rats in a three-chamber apparatus. For the first test, gdx females were made sexually active by treatments with estradiol benzoate (EB) and progesterone (P), and sexually active males were selected by prior screening. Sexually active males and females preferred opposite-sex odor over same-sex odor. Odor of sexually active opposite-sex conspecifics was preferred over that of inactive ones. Immediately after the completion of the first test, sexually active males were gdx and females were left without hormonal treatment. Second and third tests were carried out 2 and 5 weeks after the first test. In the second test, gdx males preferred odor of sexually active males rather than that of receptive females (male-directed preference); in the third test, both males and females showed no preference when tested with four stimulus pairs. The final tests were carried out in gdx males with EB and P, and gdx females with 2-week exposure to testosterone (T). Males with EB and P showed a male-directed preference again, whereas T-treated females kept their own female preference. Injection of EB alone to gdx males did not induce any preference. The present study clearly demonstrated sex difference in conspecific odor preference. Although both male and female preferences depend on their circulating sex steroids, the direction of male preference is more susceptible to their hormonal states, compared to that of females.  相似文献
