The in vivo effects of a subcutaneously-administered subcurative dose (200 mg/kg body weight of mouse) of praziquantel on the structure of Schistosoma mansoni was investigated. Worms were collected at varying times post-treatment and processed for both SEM and TEM examination. Praziquantel caused extensive structural changes to both male and female worms within 15 min of treatment although variations in the amount of drug-induced damaged was observed between male and female worms. In female worms although some tegumental vacuolation was observed within 15 min, the major structural change was an often extensive vacuolisation of the subtegumental tissues followed by varying degrees of structural disruption to the subtegumental and gastrodermal musculature. In male worms the initial effects were a vacuolisation of parts of the dorsal tegument and loss of tegumental cytoplasm due to the pinching off of evaginations of the outer surface. With increasing time post-treatment there was an increase in the amount of tegumental damage, particularly in male worms, with total disruption of parts of the outer surface occurring. Also in male worms there was an increase in the amounts of vacuolisation of the parenchymal tissues and in the degree of structural damage to the musculature. In those female worms where subtegumental damage was not extensive changes in the structure of the differentiating vitelline cells were noted.  相似文献

The distribution of Ca2+-dependent adenosine triphosphatase (EC 3.6.1.3.) and nonspecific (Na-K-Mg) adenosine triphosphatase activity in the tegument and subtegumental tissues of Schistosoma mansoni from both mixed and single sex infections was investigated cytochemically. Differences in the distribution of tegumental Ca-adenosine triphosphatase activity in 60- to 70-day-old female worms were found which could be related to the degree of sexual development in the two types of females, with little or no tegumental activity being found in 70-day-old females from single sex infections. In contrast, 28-day-old females from single sex infections showed low levels of tegumental Ca-adenosine triphosphatase activity, suggesting that the lack of tegumental activity in 70-day-old single sex females may be due to a loss or suppression of activity as a consequence of the failure of females in single sex infections to pair and develop to full sexual maturity. No differences in the distribution of nonspecific (Na-K-Mg) adenosine triphosphatase activity between females from mixed and single sex infections were found. The sexual status or age of male worms appeared to have little or no effect on the distribution of tegumental adenosine triphosphatases.  相似文献

A detailed temporal examination was made of alterations induced by artemether in the tegument of adult Schistosoma mansoni worms using scanning electron microscopy (SEM). Mice infected with S. mansoni cercariae 42 days previously were treated intragastrically with artemether at a single dose of 400 mg/kg. Groups of 3 mice were killed at 24 hr, 72 hr, and 7 days after treatment; the worms were collected by perfusion and examined by SEM. Twenty-four hours after artemether treatment, focal damage to the tubercles on the tegumental surface of male worms was seen. In both male and female worms, there was focal swelling and fusion of tegumental ridges, and sometimes peeling. After 72 hr, the damage to the tegument had increased, especially in female worms, with extensive swelling, fusion, and peeling of the tegumental ridges. In the most severely damaged worms, host leukocytes were seen to be adhered to the damaged tegument. Damage to the oral sucker was also occasionally seen in both male and female worms. Seven days after treatment, the appearance of the tegument had returned to normal in some male and female worms, whereas others still showed apparent damage. The results demonstrate that artemether damages the tegument of adult S. mansoni, and the intensity of damage is more severe in female worms than in males.  相似文献

The present study was performed to observe tegumental ultrastructure of Echinoparyphium recurvatum according to developmental stages. Worms (1, 3, 5 and 15-day old) were recovered from chicks experimentally infected with metacercariae from Radix auricularia coreana. One-day old worms were elongated and ventrally concave, and covered with peg-like tegumental spines except the adjacent areas of the head crown and excretory pore. Type I sensory papillae were distributed on the lip of the oral sucker, and grouped ciliated papillae were around the oral sucker. Peg-like tegumental spines were densely distributed on the anterior surface of the ventral sucker level. The ventral sucker had an aspinous tegument and no sensory papillae. Tegumental spines on the posterior surface of the ventral sucker level were sparsely distributed and disappeared posteriorly. In 3 and 5-day old worms, the tegument around the oral sucker was aspinose and wrinkled concentrically. The ventral sucker had a wrinkled tegument and many bulbous papillae. Type I sensory papillae were distributed between the bulbous papillae. Tegumental spines were spade-shaped with a terminal tip. A total of 45 collar spines including 4 end group ones on both ventral corners was alternately arranged in 2 rows. The 15-day old worms were very stout and their tegumental spines were tongue-shaped without a terminal tip. From the above results, it is confirmed that the surface ultrastructure of E. recurvatum was generally similar to that of other echinostomatid flukes. However, some features, i.e., morphological change of tegumental spines and appearance of sensory papillae on the ventral sucker according to development, and number, shape and arrangement of collar spines, were characteristic, which may be of taxonomic and bioecological significance.  相似文献

Experimental adults and natural metacercariae of Brachylaima sp. (Digenea: Brachylaimidae) were processed for transmission and scanning electron microscopy, for tetramethyl rhodamine isothiocyanate phalloidin fluorescence, conventional and confocal microscopy and for monoclonal anti-actin antibody immunoelectron microscopy, to describe the ultrastructural features of the tegument and to localize actin in several regions of the body. The general body tegument is spinous and contains membrane-bound disc-shaped biconcave vesicles in adults and metacercariae. Spines are flat and apically indented in the adult. A thick glycocalyx covers the body of the metacercaria. Direct fluorescence revealed filamentous actin in the spines and the subtegumental musculature of the adult. The subtegumental musculature comprises two systems consisting of an outer circular layer and an inner layer composed of cross-linked longitudinal and diagonal fibres. Immunoreactivity in adults and metacercariae revealed the presence of actin in the spines and subtegumental musculature but not in the tegumental matrix, interstitial fibrous material nor the parenchyma. Actin was detected in the dense collar and bulb matrix of the tegumental ciliated sensilla and, for the first time, in the apical cytoplasmatic projections of the digestive cell of the adult.  相似文献

Progress has been made over the last decade with the development and clinical use of artemether as an agent against major human schistosome parasites. The tegument has been identified as a key target of artemether, implying detailed studies on ultrastructural damage induced by this compound. We performed a temporal examination, employing a transmission electron microscope to assess the pattern and extent of ultrastructural alterations in adult Schistosoma mansoni harboured in mice treated with a single dose of 400 mg/kg artemether. Eight hours post-treatment, damage to the tegument and subtegumental structures was seen. Tegumental alterations reached a peak 3 days after treatment and were characterized by swelling, fusion of distal cytoplasma, focal lysis of the tegumental matrix and vacuolisation. Tubercles and sensory organelles frequently degenerated or collapsed. Typical features of subtegumental alterations, including muscle fibres, syncytium and parenchyma tissues, were focal or extensive lysis, vacuolisation and degeneration of mitochondria. Severe alterations were also observed in gut epithelial cells and vitelline cells of female worms. Our findings of artemether-induced ultrastructural alterations in adult S. mansoni confirm previous results obtained with juvenile S. mansoni and S. japonicum of different ages.  相似文献

The surface topography and ultrastructural changes of Schistosoma mansoni worms developed from ultraviolet-irradiated cercariae were compared with those from non-irradiated cercariae. The tegument of worms developed from irradiated cercariae showed a variety of changes including the occurrence of oedemata and most of the tubercles being torn at the tip and having lost their spines on the dorsal surface of some male worms. In females, parts of the tegument were devoid of spines and surface lesions were present. The range and extent of these changes differed not only among individual worms but also between different regions of individual worms resulting in generalized deformities in those worms originating from irradiated cercariae.  相似文献

The tegument of the adult blood fluke Schistosoma japonicum is in direct contact with the host blood and immune systems. A comprehensive understanding of the ultrastructure of the tegument is crucial to the understanding of how the parasite maintains itself within the mammalian host. Important functions such as nutritional uptake and immune evasion are suspected functions of the tegument and this review discusses these aspects and presents some insights into some of these crucial functions. Transmission electron microscopy has allowed the identification of ultrastructural features of the adult S. japonicum, some of which differ from the reported features of other schistosome species. Morphological differences within the tegument of the adult S. japonicum are noted between sexes, among different regions of the worms and between aspects along the length of the parasite. Differences included variations in the ultrastructure, size and number of tegumental bodies and mitochondria within the matrix, and differences in the relative area of the apical surface of the tegument. Functions of the various components of the tegument matrix and specialised functions of different regions of the male and female parasites are discussed based on ultrastructural findings and previously reported biochemical and molecular data.  相似文献

Electron microscopic examination of the epidermal basement membrane region of the rat tapeworm, Hymenolepis diminuta, has revealed specialized connective tissue structures that appear to anchor the epidermis, or tegument, to the parenchymal tissues of the helminth, as well as interconnect subtegumental muscle fibers, tegument, and parenchyma. Anchoring fibrils-cross-banded bundles of ca. 3 nm diameter filaments--were observed to directly interlink tegument and muscle, muscle and muscle, and tegument, muscle, and parenchymal connective tissue. Anchoring fibrils therefore appear to mechanically integrate epidermal tissue movements in response to subtegumental muscle contraction. A well-developed stratum of microfibrils, forming the lamina reticularis of the tegumental basement membrane, may also help anchor the tegument as well as to serve as a flexible, reinforcing sheath that protects parenchymal tissues from excessive radial displacement due to muscle contraction.  相似文献

The ventral sucker of Schistosoma mansoni cercaria is a cupshaped structure that is attached to the ventral surface of the organism by a homogeneous connective tissue that surrounds the acetabular glands. The sucker consists of an extensive complex of circular and longitudinal muscles. The longitudinal muscles extend outwoard in a radial pattern to form the cup of the organ. Intermingled with the muscles are nerve bundles and subtegumental cells (cytons). Dendritic nerve fibers connect to sensory papillae which are found on the surface tegument. Two types of sensory papillae are present: a commonly found unsheathed uniciliated papilla, and a previously unidentified tegumental encapsulated structure. The tegumental ultrastructure of juvenile and adult Himasthla alincia (Digenea: Echinostomatidae) was observed by scanning electron microscopy. One-, 5- (juveniles) and 20-day-old worms (adults) were harvested from chicks experimentally fed metacercariae from a bivalve, Mactra veneriformis. The juvenile worms were elongated and curved ventrally. The head crown bore 31 collar spines, arranged in a single row. The lip of the oral sucker had 12 paired, and 3 single type I sensory papillae, and the ventral sucker had about 25 type II sensory papillae. The anterolateral surface between the two suckers was densely packed with tegumental spines with 4-7 pointed tips. The adult worms were more elongated and filamentous, and had severe transverse folds over the whole body surface. On the head crown and two suckers, type I and II sensory papillae were more densely distributed than in the juvenile worms. Retractile brush-like spines, with 8-10 digits, were seen on the anterolateral surface, whereas claw-shaped spines, with 2-5 digits, were sparsely distributed posteriorly to the ventral sucker. The cirrus characteristically protruded out, and was armed with small spines distally. The surface ultrastructure of H. alincia was shown to be unique among echinostomes, especially in the digitation of its tegumental spines, the distribution of sensory papillae and by severe folds of the tegument.  相似文献

Adult Schistosoma mansoni were studied radioautographically in order to ascertain the effect of exposures to a fixed concentration of colchicine (5 × 10?4M) for varying time intervals upon the incorporation of [3H]proline in the tegument. Additionally, a study was made on the effect of varying time exposures of colchicine on the cytochemical localization of alkaline phosphatase (EC 3.1.3.1) in the tegumental invaginations. Worms exposed to colchicine for more than 2 hr preceding addition of the labeled amino acid displayed significant changes in the pattern of distribution. The most profound change was noted in the male tegument where a statistically significant decrease was observed in treated worms. Female worms, on the other hand, failed to display any effect of the drug on the distribution pattern for the times utilized. The distribution of alkaline phosphatase activity was much reduced in the teguments of both sexes. Morphological effects of the drug included disappearance of microtubules from the cytoplasmic connectives, a stacking of RER in the subtegumental cells, and accumulation of discoid granules and membranous bodies in the subtegumental cells. It is hypothesized that the amino acid is associated with the discoid granule at the subtegumental cell level and is ultimately translocated, with the aid of microtubules in the cytoplasmic connectives, to the tegument. Alkaline phosphatase activity is assumed to be associated with the membranous body.  相似文献

International Journal for Parasitology3: 13–25. The tegumental outer membrane of the cercaria is trilaminate: the adult worm, however, has a seven-layered membrane. Formation of the heptalaminate membrane commences immediately after the cercaria has penetrated the vertebrate host: multilaminate membrane-bounded vacuoles are passed from subtegumental cells into the tegument where they enlarge, join to the outer membrane and open to the exterior. The heptalaminate limiting membrane of the vacuole thus becomes the outer membrane of the tegument. At the same time the original trilaminate tegumental membrane is formed into microvilli which are cast off and thus the cercarial outer membrane is lost. Schistosomula usually have a heptalaminate outer membrane within three hours of penetration. After this time the large vacuoles are replaced by smaller membraneous bodies which presumably contribute to the outer membrane during growth of the schistosomulum. The membraneous bodies are also present in the tegument of the adult worms and there is some evidence that the outer membrane is continually renewed.  相似文献

The effect of normal and immune serum on Taenia crassiceps larvae in vitro was assessed by Evans blue dye uptake and electron microscopy. Normal guinea pig, rabbit, goat, and fetal calf serum did not have any significant detrimental effects upon the larvae after 7 days of culture in vitro. Culture for 7 days in normal mouse serum resulted in some loss of tegumental microtriches but the tegument itself remained intact. Culture in hyperimmune rabbit serum resulted in complete loss of the tegument and disruption of subtegumental structures within 48 hr. The effects of immune mouse serum in vitro closely paralleled those previously seen during early immune damage in vivo. Immune serum taken 2 to 4 weeks after secondary intraperitoneal infection with T. crassiceps metacestodes caused loss of the larval tegument and degeneration of the subtegumental tissues after 7 days in culture, whereas immune mouse serum taken 6 weeks after secondary infection caused only minor ultrastructural changes and appeared to be less toxic to larvae than normal mouse serum. Although complement appeared to increase the number and severity of the tegumental lesions, the presence of heat-labile components of complement was not essential for mediation of tegumental damage by immune mouse serum.  相似文献

The fine–structural characteristics of the basement membrane of the tegument of F. hepatica were examined following extraction fixations and tannic acid infiltration. The basement membrane was shown to consist of three layers: lamina lucida, lamina densa, and lamina reticularis. The lamina densa appeared amorphous and homogeneous with tannic acid impregnation. The lamina reticularis appeared as a dense network of 10–12 nm fibrils. Anchoring fibrils cross this layer and form loops. Along their length they contact hemidesmosomes of muscles, thus connecting muscle to muscle and to tegument. The tegument/basement membrane contact is enhanced by extensions of the lamina densa into infoldings of the tegumental basal membrane. Where tegumental spines reach the basement membrane, the contact is reinforced by hemidesmosomes that connect to anchoring fibrils reaching toward the underlying muscles. The basement membrane thus seems to be a complex structure integrating the distal tegumental layer with underlying tissues and transducing muscle contractions to the tegument and its spines.  相似文献

International Journal for Parasitology10: 275–280. Validity of the tumulus, a new organ recently described on the tegument of Eubothrium salvelini, is confirmed. Spherical, dense inclusions approximately 0.23–0.29 μm were associated with the tumulus and with subtegumental cell bodies. Origin of these inclusions within subtegumental cell bodies and their transport via ducts to the tumulus is described. Inclusions are synthesized within granular endoplasmic reticulum and packaged by Golgi apparatus prior to transport. Inclusions were observed only in association with the tumulus within the tegumental distal cytoplasm.  相似文献

A scanning electron microscopic study was performed to observe the tegumental ultrastructures of Paragonimus iloktsuenensis according to its developmental stages. The metacercariae were obtained from the liver of the brackish water crab, Sesarma dehaani. Juvenile and adult P. iloktsuenensis were recovered from the experimental rats on 2, 4 and 8 weeks after infection. The findings were summarized as follows: 1. The excysted metacercariae were characteristically gourd-shape, with their whole body surface beset with numerous spade-shape spines. The large, type II sensory papillae (non-ciliated round swellings) were arranged along the rim of the oral and ventral suckers, 11-12 and 6-8 in numbers respectively. 2. Two-week old juvenile worms, recovered chiefly from the liver of the experimental rats, were slender in body shape, with their ventral sucker near the anterior one-third level. The distribution of tegumental spines was less dense than in the excysted metacercariae. The spines were with 1-2 pointed tips and 3-4 longitudinal splits. Numerous ciliated knob-like, type I papillae were observed in both sides of the oral sucker, and 6 large, type II papillae were arranged along the rim of the ventral sucker. 3. Four-week old worms, recovered from the thoracic cavity and/or lung parenchyme of the experimental rats, were thicker than wide in body configuration, and their ventral sucker was located near the anterior one-fourth level. The tegumental spines at ventral surface were grouped, each group with 3-5 aggregated ones. The type I and type II papillae (small-sized) were distributed chiefly around the rim of two suckers. 4. Adult (eight-week old) worms, recovered from the capsules in the lung parenchyme, were very stout, and covered densely with bearfoot-like spines. At dorsal surface, cobblestone-like cytoplasmic processes were well-developed, with many tegumental spines embedded in them. It was observed in this study that the tegument of P. iloktsuenesis continued to change and differentiate as the worms grew to be adults.  相似文献

Amoscanate, when administered orally as an aqueous or “formulated” preparation, induced pronounced ultrastructural abnormalities in male and female Schistosoma haematobium. Higher dose levels of the aqueous suspension (300 mg/kg body wt) had to be administered to achieve the full range of effects induced by formulated doses of 2.5–8 mg/kg body wt. Worms were recovered from hamsters between 1 and 120 hr after treatment. Although the amount of amoscanate-induced damage varied considerably between worms, an overall pattern of damage emerged. Initially, 1 hr after treatment, amoscanate caused tegumental vacuolation and oedema. As the drug treatment period was extended to 24 hr, blebbing, exudation, collapse of sensory organelle bases, and abnormal mitochondria became increasingly evident. With exposure to higher drug doses (50–300 mg/kg body wt), the tegument became further distorted with the appearance of necrotic structures and myelin whorls, which appeared to represent various stages in lysosomal formation and digestion. Eventually, erosion of surface layers resulted in the breakdown of tegumental integrity. The caeca and vitellaria were also adversely affected by drug treatment. Basal vacuolation and the formation of myelin whorls occurred in the gastrodermis. In the mature S4 vitelline cells, coalesced vitelline droplets and myelin whorls were evident.  相似文献
