琥珀酸放线杆菌发酵培养基的优化   总被引:4,自引:0,他引:4  
对琥珀酸放线杆菌(Actinobacillus succinogenes)CGMCC1593发酵产生琥珀酸培养基的主要成分,及其含量进行优化。通过单因素试验,得出发酵培养基中葡萄糖、酵母膏和玉米浆的含量对产生琥珀酸有显著影响;采用响应面法(RSM),得出多元二次回归方程拟合的三种因素与琥珀酸含量间的函数关系,并根据优化结果与实验,CGMCC1593产琥珀酸达到41.69g/L。  相似文献

丁二酸是一种重要的C4化合物平台,可以合成一系列重要化合物。文中对产琥珀酸放线杆菌Actinobacillus succinogenes GXAS137发酵生产丁二酸培养基成分进行优化。通过单因素和Plackett-Burman试验设计筛选出影响丁二酸发酵的重要参数,采用最陡爬坡实验逼近最大丁二酸生产区域后,利用Box-Behnken设计确定重要参数的最佳水平。筛选结果表明,影响丁二酸产量的重要参数是葡萄糖、酵母提取物和碱式碳酸镁浓度。最佳条件为(g/L):葡萄糖70.00,酵母提取物9.20,碱式碳酸镁58.10。优化后丁二酸产量达到47.64 g/L。与初始条件 (36.89 g/L) 相比,丁二酸浓度提高了30 %。在最佳工艺条件下得到的试验结果与模型预测值很吻合,说明建立的模型是有效的。  相似文献

为提高琥珀酸放线菌Actinobacillus succinogenes CGMCC1593厌氧发酵产丁二酸的水平。研究了以葡萄糖为C源,发酵液中不同氧化还原电位(VORP)对A.succirtogenes CGMCC1593生长和代谢产物分布的影响。结果表明:菌体生长和丁二酸积累的较佳VORP分别为-220mV和-270mV;利用代谢流分析法,比较VORP在-220mV和-270mV时发酵对数生长期(8h)和稳定期(20h)的代谢通量分布,以及发酵过程中磷酸烯醇式丙酮酸(PEP)、丙酮酸(Pyr)节点,NADH通量分配的变化,由此得出在VORP为-270mV时,NADH总通量和丁二酸方向代谢通量增幅明显。在发酵过程中,通过降低VORP至-270mV,使丁二酸的产率从70%提高到85%。  相似文献

This study presents the production of succinic acid (SA) by Actinobacillus succinogenes using high-sugar-content beverages (HSCBs) as feedstock. The aim of this study was the valorization of a by-product stream from the beverage industry for the production of an important building block chemical, such as SA. Three types of commercial beverages were investigated: fruit juices (pineapple and ace), syrups (almond), and soft drinks (cola and lemon). They contained mainly glucose, fructose, and sucrose at high concentration—between 50 and 1,000 g/L. The batch fermentation tests highlighted that A. succinogenes was able to grow on HSCBs supplemented with yeast extract, but also on the unsupplemented fruit juices. Indeed, the bacteria did not grow on the unsupplemented syrup and soft drinks because of the lack of indispensable nutrients. About 30–40 g/L of SA were obtained, depending on the type of HSCB, with yield ranging between 0.75 and 1.00 gSA/gS. The prehydrolysis step improved the fermentation performance: SA production was improved by 6–24%, depending on the HSCB, and sugar conversion was improved of about 30–50%.  相似文献

In this work, production of succinic acid by Actinobacillus succinogenes CGMCC1593 using cane molasses as a low cost carbon source was developed. In anaerobic bottles fermentation, succinic acid concentration of 50.6+/-0.9 g l(-1) was attained at 60 h using an optimum medium containing molasses pretreated with sulfuric acid, resulting in a succinic acid yield of 79.5+/-1.1% and sugar utilization of 97.1+/-0.6%. When batch fermentation was carried out in a 5-l stirred bioreactor with pretreated molasses, 46.4 g l(-1) of succinic acid was attained at 48 h and faster cells growth was also observed. Fed batch fermentation was performed to minimize the substrate (sugar) inhibition effect, giving 55.2 g l(-1) of succinic acid and 1.15 g l(-1)h(-1) of productivity at 48 h. The present study suggests that the inexpensive cane molasses could be utilized for the economical and efficient production of succinic acid by A. succinogenes.  相似文献

In this work, straw hydrolysates were used to produce succinic acid by Actinobacillus succinogenes CGMCC1593 for the first time. Results indicated that both glucose and xylose in the straw hydrolysates were utilized in succinic acid production, and the hydrolysates of corn straw was better than that of rice or wheat straw in anaerobic fermentation of succinic acid. However, cell growth and succinic acid production were inhibited when the initial concentration of sugar, which was from corn straw hydrolysate (CSH), was higher than 60 g l?1. In batch fermentation, 45.5 g l?1 succinic acid concentration and 80.7% yield were attained after 48 h incubation with 58 g l?1 of initial sugar from corn straw hydrolysate in a 5-l stirred bioreactor. While in fed-batch fermentation, concentration of succinic acid achieved 53.2 g l?1 at a rate of 1.21 g l?1 h?1 after 44 h of fermentation. Our work suggested that corn straw could be utilized for the economical production of succinic acid by A. succinogenes.  相似文献

Continuous anaerobic fermentations were performed in a novel external-recycle, biofilm reactor using d-glucose and CO2 as carbon substrates. Succinic acid (SA) yields were found to be an increasing function of glucose consumption with the succinic acid to acetic acid ratio increasing from 2.4 g g−1 at a glucose consumption of 10 g L−1, to 5.7 g g−1 at a glucose consumption of 50 g L−1. The formic acid to acetic acid ratio decreased from an equimolar value (0.77 g g−1) at a glucose consumption of 10 g L−1 to a value close to zero at 50 g L−1. The highest SA yield on glucose and highest SA titre obtained were 0.91 g g−1 and 48.5 g L−1 respectively. Metabolic flux analysis based on the established C3 and C4 metabolic pathways of Actinobacillus succinogenes revealed that the increase in the succinate to acetate ratio could not be attributed to the decrease in formic acid and that an additional source of NADH was present. The fraction of unaccounted NADH increased with glucose consumption, suggesting that additional reducing power is present in the medium or is provided by the activation of an alternative metabolic pathway.  相似文献

Rapeseed meal was evaluated for succinic acid production by simultaneous saccharification and fermentation using Actinobacillus succinogenes ATCC 55618. Diluted sulfuric acid pretreatment and subsequent hydrolysis with pectinase was used to release sugars from rapeseed meal. The effects of culture pH, pectinase loading and yeast extract concentration on succinic acid production were investigated. When simultaneous saccharification and fermentation of diluted acid pretreated rapeseed meal with a dry matter content of 12.5% (w/v) was performed at pH 6.4 and a pectinase loading of 2% (w/w, on dry matter) without supplementation of yeast extract, a succinic acid concentration of 15.5 g/L was obtained at a yield of 12.4 g/100g dry matter. Fed-batch simultaneous saccharification and fermentation was carried out with supplementation of concentrated pretreated rapeseed meal and pectinase at 18 and 28 h to yield a final dry matter content of 20.5% and pectinase loading of 2%, with the succinic acid concentration enhanced to 23.4 g/L at a yield of 11.5 g/100g dry matter and a productivity of 0.33 g/(Lh). This study suggests that rapeseed meal may be an alternative substrate for the efficient production of succinic acid by A. succinogenes without requiring nitrogen source supplementation.  相似文献

Fermentation and succinic acid production by Actinobacillus succinogenes YZ0819 was inhibited by high NaCl. To enhance the resistance of this strain to osmotic stress, an NaCl-tolerant mutant strain of A. succinogenes (CH050) was screened and selected through a continuous culture using survival in 0.7 M NaCl as the selection criterion. Using Na2CO3 as the pH regulator and glucose as the carbon source in batch fermentation, the isolated osmo-resistant stain, A. succinogenes CH050, produced up to 66 g/l succinic acid with a yield of 73.37% (w/w). The concentration of succinic acid and mass yield were increased by 37.5 and 4.37%, respectively, compared to the parent strain. The dry cell weight reached 10.1 g/l, which is 37% higher than that of the parent strain. The high tolerance of A. succinogenes CH050 to osmotic stress increased improved the succinic acid production from batch fermentation.  相似文献

Carbon sources with different oxidation states were used to investigate the possibility increasing the availability of NADH and the NADH/NAD+ ratio and to determine the effect of this manipulation on the distribution of metabolites in Actinobacillus succinogenes NJ113. The sugars glucose, sorbitol and gluconate were each used at an initial concentration of 40 g/L.The yield of succinic acid (0.75) and the ratio of succinic acid to acetic acid (5.06) were both higher for sorbitol than the values obtained with glucose (0.66 and 2.68, respectively). In contrast, with gluconate as the carbon source the yield of succinic acid was 0.54 and the ratio of succinic acid to acetic acid was only 1.70. This work showed that different levels of NADH availability and the NADH/NAD+ ratio can be achieved by using carbon sources that have different oxidation states.Highly reduced sorbitol was examined as a possible carbon substrate for maximizing the redox potential during the production of succinic acid.  相似文献

An industrial fermentation system using lignocellulosic hydrolysate, waste yeast hydrolysate, and mixed alkali to achieve high-yield, economical succinic acid production by Actinobacillus succinogenes was developed. Lignocellulosic hydrolysate and waste yeast hydrolysate were used efficiently as carbon sources and nitrogen source instead of the expensive glucose and yeast extract. Moreover, as a novel method for regulating pH mixed alkalis (Mg(OH)2 and NaOH) were first used to replace the expensive MgCO3 for succinic acid production. Using the three aforementioned substitutions, the total fermentation cost decreased by 55.9%, and 56.4 g/L succinic acid with yield of 0.73 g/g was obtained, which are almost the same production level as fermentation with glucose, yeast extract and MgCO3. Therefore, the cheap carbon and nitrogen sources, as well as the mixed alkaline neutralize could be efficiently used instead of expensive composition for industrial succinic acid production.  相似文献

The effects of redox potential used as a control parameter on the process of succinic acid production in batch cultures of Actinobacillus succinogenes NJ113 have been investigated. In batch fermentation, cell growth and metabolite distribution were changed with redox potential levels in the range of ?100 to ?450 mV. From the results, the ORP level of ?350 mV was preferable, which resulted in high succinic acid yield (1.28 mol mol?1), high succinic acid productivity (1.18 g L?1 h?1) and high mole ratio of succinic acid to acetic acid (2.02). The mechanism of redox potential regulation was discussed by metabolic flux analysis and the ratio of NADH/NAD+. We expected that redox potential can be used as a valuable parameter to monitor and control much more anaerobic fermentation production.  相似文献

Fermentation at low pH is an efficient way to improve the competitiveness of biological succinic acid-producing process. Actinobacillus succinogenes shows good performance of succinic acid production under anaerobic conditions, but its succinic acid production capability at the low-pH is inefficient due to the poor acid resistance. Herein, a mutant A. succinogenes BC-4 with improved cell growth and succinic acid production under weak acid conditions was obtained by adaptive evolution. The specific growth rate and succinic acid production of BC-4 reached 0.13 g/L/h and 20.77 g/L, which were increased by 3.25- and 2.95- fold, respectively compared with the parent strain under anaerobic condition at pH 5.8. The activities of specific enzymes with ATP generation were significantly enhanced under weak acidic conditions, resulting in 1.28-fold increase in the maximum ATP level. Membrane fatty acid composition analysis demonstrated that the ratio of saturated to unsaturated fatty acids was decreased from 1.62 to 1.44 in mutant BC-4, leading to improved intracellular pH homeostasis. Furthermore, the change from long-chain to median-chain fatty acid might lower the permeability of H+ into cytoplasm for survival under acid stress. These results indicated that A. succinogenes BC-4 is a promising candidate for succinic acid production under weak acid condition.  相似文献

Due to environmental issues and the depletion of fossil-based resources, ecofriendly sustainable biomass-based chemical production has been given more attention recently. Succinic acid (SA) is one of the top value added bio-based chemicals. It can be synthesized through microbial fermentation using various waste steam bioresources. Production of chemicals from waste streams has dual function as it alleviates environmental concerns; they could have caused because of their improper disposal and transform them into valuable products. To date, Actinobacillus succinogenes is termed as the best natural SA producer. However, few reviews regarding SA production by A. succinogenes were reported. Herewith, pathways and metabolic engineering strategies, biomass pretreatment and utilization, and process optimization related with SA fermentation by A. succinogenes were discussed in detail. In general, this review covered vital information including merits, achievements, progresses, challenges, and future perspectives in SA production using A. succinogenes. Therefore, it is believed that this review will provide platform to understand the potential of the strain and tackle existing hurdles so as to develop superior strain for industrial applications. It will also be used as a baseline for identification, isolation, and improvement of other SA-producing microbes.

Immense interest has been devoted to the production of bulk chemicals from lignocellulose biomass. Diluted sulfuric acid treatment is currently one of the main pretreatment methods. However, the low total sugar concentration obtained via such pretreatment limits industrial fermentation systems that use lignocellulosic hydrolysate. Sugarcane bagasse hemicellulose hydrolysate is used as the carbon and nitrogen sources to achieve a green and economical production of succinic acid in this study. Sugarcane bagasse was ultrasonically pretreated for 40 min, with 43.9 g/L total sugar obtained after dilute acid hydrolysis. The total sugar concentration increased by 29.5 %. In a 3-L fermentor, using 30 g/L non-detoxified total sugar as the carbon source, succinic acid production increased to 23.7 g/L with a succinic acid yield of 79.0 % and a productivity of 0.99 g/L/h, and 60 % yeast extract in the medium could be reduced. Compared with the detoxified sugar preparation method, succinic acid production and yield were improved by 20.9 and 20.2 %, respectively.  相似文献

对Actinobacillus succinogenes130Z厌氧发酵产丁二酸的培养条件进行了初步研究。研究了不同有机氮源,不同碳、氮源浓度配比、CO2供体、培养温度,培养基起始pH值对菌株生长和产酸的影响,并在5 L发酵罐中进行了放大试验。结果表明最佳培养基配方为(g/L):葡萄糖10,酵母膏5,NaHCO310,Na2HPO40.3,NaH2PO4.2H2O 9.6,K2HPO43,MgCl20.2,MnCl20.2,NaCl 0.1;pH7.0。在最佳条件下,血清瓶37℃培养24 h,丁二酸产量达到8.3 g/L,在5 L发酵罐中培养,葡萄糖质量浓度分别为10和100 g/L时,丁二酸产量分别达到8.2和45.6 g/L,收率分别为80%和65%。  相似文献

以Actinobacillus succinogenes NJ113为出发菌株,经硫酸二乙酯(DES)诱变,在含8~20 g/L硫酸铵平板中筛选到一株耐铵型突变株YZ25,该菌株在含8 g/L硫酸铵培养基中厌氧发酵,琥珀酸产量达32.68 g/L,比出发菌提高了180.5%,对葡萄糖收率达65.4%,副产物乙酸、甲酸产量分别下降3.5%、28.7%,琥珀酸/乙酸比值由0.63提高到2.5。在7.5 L发酵罐中,用氨水调节pH分批实验,发酵34 h琥珀酸产量达27.13 g/L,较出发菌株提高了85.3%。  相似文献

经硫酸二乙酯(DES)诱变,在含61~242mmol/LNH4+梯度平板中,筛选到一株耐铵型突变株YZ25,该菌株在含121mmol/LNH4+发酵培养基中,琥珀酸产量达32.68g/L,转化率为65.4%,比出发菌提高了180.5%。进一步考察了不同形态铵盐对YZ25生长的影响,结果表明添加少量铵盐能够提高突变菌的生长速率,但当超过一定量后菌株生长受到抑制,不同铵盐对菌株的抑制程度不同,硫酸铵、碳酸氢铵、氯化铵和硝酸铵对突变株YZ25的半抑制浓度分别为:215mmol/L、265mmol/L、235mmol/L、210mmol/L。为了考察铵离子对YZ25发酵产琥珀酸的影响过程,在3.0L发酵罐以氨水作为pH的调控剂发酵,结果表明在稳定期前菌株生长基本不受铵离子抑制,生物量能够达到正常水平,但是进入稳定期后铵离子抑制作用越来越明显,导致菌株生长提前结束,耗糖不完全,产酸受阻。最后结合产琥珀酸放线杆菌Actinobacillus succinogenes代谢途径分析了铵离子对菌株抑制作用的机理。  相似文献

The culture conditions in CO(2) fixation by Actinobacillus succinogenes for succinic acid production were investigated by a model of available CO(2) in a 3-l fermentor. The results from the model analysis showed that the available CO(2) for succinic acid production in the fermentation broth is the sum of HCO(3) (-), CO(3) (2-), and CO(2) influenced by external culture conditions such as medium components, CO(2) partial pressures, and temperature. The optimized conditions for CO(2) supply in a 3-l fermentor were determined as follows: CO(2) partial pressure and stirring speed were maintained at 0.1 MPa and 200 r min(-1), respectively, with a pH of 6.8 and a temperature of 37°C; 0.15 mol l(-1) NaHCO(3) was added. Under the optimized conditions, a CO(2) fixation rate of 0.57 g l(-1) h(-1) was obtained, and a succinic acid concentration of 51.6 g l(-1) with a yield of 75.8% was reached. These results suggest that optimized conditions of CO(2) supply are effective in high succinic acid production and thus have potential applications in succinic acid production and CO(2) fixation.  相似文献
