Abstract The kinetics of bacterial adherence to hexadecane were measured using disposable polystyrene cuvettes. The rate of adherence was exponential, and was itself linearly dependent on the water:hexadecane ratio employed. The dependency of the rate of adherence on the water:hexadecane ratio, termed the removal coefficient, varied from 4.7 min−1 for Streptococcus pyogenes to 72 min−1 for Acinetobacter calcoaceticus . The removal coefficient of Serratia marcescens was a function of growth temperature, 48 min−1 following growth at 30°C as opposed to 5.8 min−1 for 35°C-grown cells.  相似文献

The interaction of temperature and fish size on growth of juvenile halibut   总被引:3,自引:0,他引:3  
Growth rate of individually tagged juvenile halibut was influenced significantly by the interaction of temperature and fish size. The results suggest an optimum temperature for growth of juvenile halibut in the size range 5–70 g between 12 and 15° C. Overall growth rate was highest at 13° C (1·62% day −1). At c. 5 g at the beginning of the experiment, fish at 16° C had the highest growth rate (3·2% day −1), but reduced this rate as they grew bigger. At 9 and 11°p C, growth rates were equal or only slightly lower during the later stages of the experiment, while the fish at 6° C showed significantly lower overall growth rate (0·87% day−1). Optimal temperature for growth decreased rapidly with increasing size, indicating an ontogenetic reduction in optimum temperature for growth. Moreover, a more flattened parabolic regression curve between growth and temperature as size increased indicated reduced temperature dependence with size. Although individual growth rates varied significantly at all times within the experimental temperatures, significant size rank correlations were maintained during the experiment. This indicated an early establishment of a stable size hierarchy within the fish groups. Haematocrit was highest at the highest temperature while Na+/K+-ATPase activity was inversely related to temperature. There was no difference in plasma Na+, Cl and K+ concentrations among the temperature groups.  相似文献

N. ONISHI, I. KIRA AND K. YOKOZEKI. 1996. Galacto-oligosaccharide (Gal-OS) was produced from lactose by a yeast, Sirobasidium magnum CBS6803. With toluene-treated resting cells, 136 mg ml−1 of Gal-OS was produced from 360 mg ml−1 of lactose at 50°C for 42 h. Then, the yield of Gal-OS was increased by a culture method in which cell growth followed the enzymatic reaction : 224 mg ml−1 of Gal-OS was produced at 30°C for 60 h. Finally, combination of the toluene-treated resting cells and glucose oxidase plus catalase was applied to improve productivity by the removal of a by-product, glucose, which inhibits the Gal-OS production, from the reaction mixture. In this case, 242 mg ml−1 4-galactosyl-lactose. of Gal-OS was produced at 50°C for 42 h without cell growth. The structure of the major product ws identified as 4-galactosyl-laetos.  相似文献

At 14° C, standard metabolic rate (75·1 mg O2 h−1 kg−1), routine metabolic rate (108.8 mg O2 h−1 kg−1), active metabolic rate ( c . 380 mg O2 h−1 kg−1), critical swimming speed (Ucrit 1·7 BL s−1), heart rate 47 min−1), dorsal aortic pressure (3·2 kPa) and ventilation frequency (63 min−1) for triploid brown trout Salmo trutta were within the ranges reported for diploid brown trout and other salmonids at the same temperature. During prolonged swimming ( c . 80% U crit), cardiac output increased by 2·3-fold due to increases in heart rate (1·8-fold) and stroke volume (1·2-fold). At 18° C, although standard and routine metabolic rates, as well as resting heart rate and ventilation frequency increased significantly, active metabolic rate and certain cardiorespiratory variables during exercise did not differ from those values for fish acclimated to 14° C. As a result, factorial metabolic scope was reduced (2·93-fold at 18° C v . 5·13-fold at 14° C). Therefore, it is concluded that cardiorespiratory performance in triploid brown trout was not unusual at 18° C, but that reduced factorial metabolic scope may be a contributing factor to the mortality observed in triploid brown trout at temperatures near 18° C.  相似文献

The Cape golden mole, Chrysochloris asiatica is an insectivore which excavates superficial foraging burrows as it searches for its food. It has a mean (±S.D.) resting metabolic rate (RMR) when newly captured of 1–17±0.17 cm3 O2g-1 h-1 ( n = 14), within the thermoneutral zone (TNZ) of 30–32°C.
The body temperature (Tb) of the mole in the TNZ is low 32.9 ± 0.36 ( n = 14) and remains stable at ambient temperatures (Tas) from 28–32°C. Above 32°C (range 34–37°C), Tb increases albeit slightly to 36 ± 1.75°C ( n = 14). The conductance is high 0.27 ± 006cm3 O2g-1 h-l°C-1 ( n = 46) at the lower limit of thermoneutrality. The mean RMR at 9°C (the lowest Ta tested) was 4.82±11 cm3 O2g-1h-1, which is 4.1 times that of the RMR in the TNZ.
At an ambient temperature of 9°C, three of the golden moles entered a state of torpor where the RMR was reduced from 5.9±0.56 to 10 1.0 ± 0.69cm3O2g-1h-1.  相似文献

Dark grown leaves of wheat were irradiated with red light of different intensities, at a temperature close to 0°C. The rate of photoreduction of the protochlorophyllide 650-form into chlorophyllide 684-form was measured. On continued irradiation the chlorophyllide 684-form was photodecomposed. By comparing the rates of the two processes the quantum yield for photooxidation of the chlorophyllide 684-form was calculated. The quantum yield was 2°10-5 at an intensity of 2200 W m-2, and increased with decreasing light intensity to 3.2°10-5 at an intensity of 170 W m-2.  相似文献

Abstract. Cyperus longus L. , which has a widespread but disjunct distribution throughout Europe and extends northwards into Britain, was found to be a C4 species based upon its Kranz leaf anatomy, low CO2 compensation point and the labelling of malate as an early product of 14CO2 fixation. The photosynthetic characteristics of C. longus are similar to many other C4 species with a high maximum rate of photosynthesis (> 1.5 mg CO2 m −2 s −1) and a relatively high temperature optimum (30–35°C), but unlike many C4 species the rate of photosynthesis does not decline rapidly below the optimum temperature and a substantial rate (0.6 mgCO2 m−2s−1)occursat 15°C. Leaf extension is very slow at 15°C and shows a curvilinear response to temperatures between 15 and 25°C. Leaves extend at a rate of almost 4 cm d−1 at 25°C.  相似文献

In ten experiments on commercial sugar-beet crops grown on organic soils in 1984–86, a Genstat programme was used to examine the relationship between the initial population of Heterodera schachtii and sugar-beet root yield using the equation

Fixing T = 200 eggs + juveniles 100 g-1 soil and ZT= 0.95, estimated values of Ymax varied from 49.2–67.1 t ha-1 (129– 155% of the national average root yield for the years in which the experiments were carried out) and estimates of Ymin varied from 14.5–53.9 t ha-1 (27–94% of Ymax). Callus of 'Marsh' grapefruit ( Citrus paradisi Macf.) albedo tissue was used to investigate the effect of preconditioning temperature on the rate of chilling - stimulated K+ leakage. Callus grew most rapidly at 30°C and attained a weight of about 1 g after 30 days. The rate of K.+ leakage from nonchilled callus tissue decreased as temperature decreased from 20 to 7.5°C, but no measurable change in rate was observed between 7.5 and 0°C. When calli were held for 40 days at 01 2.5 or 5°C, K+ leakage increased 200%, 60% or 0%) respectively. Holding callus for 5 days at 10 or 15°C prior to chilling for 40 days at 0°C prevented the increase in K+ leakage observed in callus receiving no preconditioning treatment. Preconditioning at 7.5 and 20°C was less effective in reducing chilling - induced leakage. Preconditioning at 10°C for 5, 2 or 1 day reduced chilling – induced leakage after 40 days at 0°C by 50%, 33% and 15%. respectively.  相似文献

Photosynthetic response of Eragrostis tef to temperature   总被引:1,自引:0,他引:1  
Photosynthetic characteristics of leaves of tef, Eragrostis tef (Zucc.) Trotter, plants, grown at 25/15°C (day/night), were measured at temperatures from 18 to 48°C. The highest carbon exchange rates (CER) occurred between 36 and 42°C. and averaged 27 μmol m−2 s−1. At lower or higher temperatures, CER was reduced, but the availability of CO2 to the mesophyll, measured as internal CO2 concentration, was highest when temperatures were above or below the optimum for CER. In addition, CER and stomatal conductance were not correlated, but residual conductance was highly correlated with CER (r = 0.98). In additional experiments, relative 13C composition for leaf tissue grown at 25, 35 and 45°C averaged -14.4 per mille, confirming that tef is a C4 grass species. Dry matter accumulation was higher at 35 than at 25, and lowest at 45°C. Leaf CER rates increased hyperbolically with increased light when measured from 0 to 2000 μmol m−2 s−1 PPFD. The highest CER, 31.8 μ-mol m-2 s−1, occurred at 35°C and 2000 μmol m−2 s−1 PPFR. At high light, CER at 25 and 35°C were nearly equal because of higher stomatal conductance at 25°C. Residual conductance was, however, clearly highest at 35°C compared to 25 and 45°C treatments. Stomatal conductance and residual conductance were not correlated in either set of experiments, yet residual conductance was always highest when temperatures were between 35 and 42°C across experiments, suggesting that internal leaf photosynthetic potential was highest across that temperature range.  相似文献

Growth and proteinase production by Micrococcus sp. INIA 528 in a batch-operated laboratory fermentor were investigated, with trypticase soy broth as the basal medium for studies on optimum temperature, pH and medium composition. Maximum growth was recorded at 34°C and pH 715, whereas optimum temperature and pH for proteinase production were 31°C and pH 6.25. Maximum rate of enzyme production occurred during the late log and early stationary phases of growth. Addition of 5.0 g 1-1 yeast extract, 1.0 g 1-1 glucose, 1.0 g 1-1 MgSO4 or 1.0 g 1-1 K2HPO4 to basal medium resulted in a lower enzyme yield, but supplementation of basal medium with 2.5 g 1-1 (NH4)2SO4 increased enzyme production by 45%. A high initial biomass added to fresh broth supplemented with 2.5 g 1-1 (NH4)2SO4 only increased enzyme activity by 19%, compared to the maximum enzyme activity achieved with the standard inoculum.  相似文献

Aerobic plate counts (APC 37°C and APC 25°C) and Escherichia coli enumerations (Petrifilm) were used to determine sources of bacterial contamination during sheep dressing, determine the hygienic efficacy of hand wash and knife 'sterilization'procedures and compare the hygiene efficiency of conventional and inverted sheep dressing systems. The major slaughterline sources of microbial contamination were: fleece > workers' hands > faecal pellets > knife blades. Aerobic plate counts (APC 37°C) exceeding log 4.4 cfu cm-2 were considered indicative of direct fleece contact, whereas E. coli numbers exceeding log 3.3 cfu cm-2 were considered indicative of direct faecal contact. A 44°C water hand rinse removed 90% of the microbial contamination from workers' hands, but rinsed hands, particularly those contacting the fleece, still carried a microbial population exceeding log 4.0 cfu cm-2. A 44°C rinse followed by an 82°C water dip reduced the contamination on knife blades to less than log 3.0 cfu cm-2. Inverted dressing systems produced carcasses with a lower contamination level than conventional systems. With both systems little increase in contamination occurred after pelt removal. The areas of highest contamination were the forequarter region with inverted dressing and the hindquarter with conventional dressing. In both cases these regions are the sites where cuts are made through the skin. With both systems contamination around these cuts was entirely consistent with direct fleece contact resulting from 'rollback'.  相似文献

Portulaca oleracea , a C4 species, is reported to be a serious weed in 45 crops in 81 countries. Experiments were conducted in the laboratory, the screenhouse and the field to determine the influence of environmental factors on seed germination and seedling emergence of P. oleracea . In the laboratory, germination in the dark was low and was not influenced by the tested temperatures (35/25°C, 30/20°C and 25/15°C alternating day/night temperatures). In the light/dark regime, however, germination was lower at 25/15°C and 35/25°C than at 30/20°C (70%, 75% and 81% germination, respectively). In conditions of 106 mM sodium chloride or −0.34 MPa osmotic potential, seeds germinated to only 50% of maximum germination of the control. Germination was not influenced by buffered pH solutions ranging from 5 to 9. In the screenhouse, germination was greatest for seeds placed on the soil surface, but emergence declined with increasing seed burial depth in soil; no seedlings emerged from the depth of 2 cm. Seedling emergence and seedling dry matter were markedly reduced by the addition of rice residue to the soil surface at rates equivalent to 4 to 6 t ha−1. In the field, seedling emergence of P. oleracea was greater under zero till (ZT) (17–20%) than under minimum tillage (6–10%), a likely reflection of low seed burial and exposure of seeds to light with a ZT system. This study identifies some of the factors enabling P. oleracea to be a widespread weed in the humid tropics, and the information could contribute to improved control strategies.  相似文献

Turbot Scophthalmus maximus maximum oxygen uptake following feeding and exhaustive exercise increased from 107 mg O2 kg−1 h−1 at 6° C to c . 218 mg O2 kg−1 h−1 at 18° C, then increased slightly from 18 to 22° C to 224 mg O2 kg−1 h−1. Standard oxygen uptake increased exponentially as a function of temperature from 11 mg O2 kg −1 h−1 at 6° C to 66 mg O2 kg−1 h−1 at 22° C. Gradual reduction in oxygen concentration to 87–90% air saturation at 6, 10. 18° C and <80% at 14 and 22° C limited the maximum metabolic rate but, supersaturation (>100% saturation) had little effect. Metabolic scope attained a maximum of 176 mg O2 kg−1 h−1 at 18° C. Interpolation of the results showed that this value changed little between 16 and 20° C. It is suggested that this temperature range is optimal for turbot of c . 500 g. A comparison with a previous study on feeding demand in intensive farming conditions showed a linear relationship between appetite and metabolic scope. It is concluded that the ability of a fish to supply energy (including the energy requirement of digestive metabolism) above a standard level is a limiting factor in the manifestation of its feeding demand.  相似文献

Daily food consumption and alimentary canal evacuation rates for common carp, Cyprinus carpio L., were estimated at 4-week intervals from April to October 1979. Both the rate of food evacuation and its relationship to temperature were described by an exponential function. Gut evacuation rates ranged from 6.0% of gut contents per hour at 9°C to 32.7% h−1 at 26.5°C. Temperature accounted for 72–91% of the variation in gut evacuation rate among months. Daily food consumption varied from a low of 3.87 mg g−1 of fish wet wt per day in April at 14°C (0.39% of wet body wt) to a high of 40.75 mg g−1 (4.08%) in August at 26.5°C. Although a greater quantity of food appeared to be consumed during the morning in spring and early summer and a greater amount in the evening after July, there was not a statistically significant time of day for peak feeding activity. The field method described here seemed to provide reasonable estimates of food consumption compared with other field and laboratory studies.  相似文献

Spirogyra Link (1820) is an anabranched filamentous green alga that forms free-floating mats in shallow waters. It occurs widely in static waters such as ponds and ditches, sheltered littoral areas of lakes, and stow-flowing streams. Field observations of its seasonal distribution suggest that the 70-μm-wide filament form of Spirogyra should have a cool temperature and high irradiance optimum for net photosynthesis. Measurements of net photosynthesis and respiration were marie at 58 combinations of tight and temperature in a controlled environment facility. Optimum conditions were 25°C and 1500 μmol photons m−2 s−1, at which net photosynthesis averaged 75.7 mg O2 gdm−1 h−1. Net photosynthesis was positive at temperatures from 5° to 35°C at most irradiances except at combinations of extremely low irradiances and high temperatures (7 and 23 μmol photons m−2 s−1 at 30°C and 7, 23, and 35 μmol photons m−2 s−1 at 35°C). Respiration rates increased with both temperature and prior irradiance. Light-enhanced respiration rates were significantly greater than dark respiration rates following irradiances of 750 μmol photons m−2 s−1 or greater. Polynomials were fitted to the data to generate response surfaces; such response surfaces can be used to represent net photosynthesis and respiration in ecological models. The data indicate that the alga can tolerate the cool water and high irradiances characteristic of early spring but cannot maintain positive net photosynthesis under conditions of high temperature and low light (e.g. when exposed to self-shading ).  相似文献

Microclimate recordings were made over four years on the Rock and Pillar Range, New Zealand. These are used in conjunction with mortality and freezing data derived in the laboratory to make inferences about the winter thermal ecology of the freeze-tolerant alpine cockroach Celatoblatta quinquemaculata (Dictyoptera: Blattidae). Threshold temperatures are identified through laboratory experiments at ecologically relevant cooling rates: 0°C; −3.1°C (FP5: the temperature at which 5% of cockroaches are expected to be frozen); and −4.5°C (FP50). The maximum cooling rate in the microhabitat across any of the thresholds was 0.06°C min−1, considerably slower than the 1°C min−1 normally used in laboratory studies. Freeze-thaw events occurred regularly in the field, and temperature minima occasionally fell to temperatures lethal to C. quinquemaculata . Variability in snow cover contributed to interannual variation in microclimate temperatures. Decreased snow cover is predicted with climate change scenarios, and this will probably result in more freeze-thaw cycles and more extreme minimum temperatures in this environment. It is concluded that the limited environmental tolerances of the animals living in this habitat make the few degrees of interannual variation ecologically relevant.  相似文献

The growth properties of juvenile spotted wolffish Anarhichas minor reared at 4, 6, 8 and 12° C, and a group reared under 'temperature steps', (T‐step) i.e . with temperature reduced successively from 12 to 9 and 6° C were investigated. Growth rate and feed efficiency ration was significantly influenced by temperature and fish size. Overall growth rate was highest at 6° C (0·68% day−1) and lowest at 12° C (0·48% day−1), while the 4 and 8° C, and the T‐step groups had similar overall growth rates, i.e . 0·59, 0·62 and 0·51% day−1 respectively. Optimal temperature for growth ( T opt G ) and feed efficiency ratio (Topt FCE) decreased as fish size increased, indicating an ontogenetic reduction in T opt G and T opt FCE. The results suggest a T opt G of juvenile spotted wolffish in the size range 135–380 g, dropping from 7·9° C for 130–135 g to 6·6° C for 360–380 g juveniles. The T opt FCE dropped from 7·4° C for 120–150 g to 6·5° C for 300–380 g juveniles. A wider parabolic regression curve between growth, feed efficiency ratio and temperature as fish size increased, may indicate increased temperature tolerance with size. Individual growth rates varied greatly at all time periods within the experimental temperatures, but at the same time significant size rank correlations were maintained and this may indicate stable size hierarchies in juvenile spotted wolffish.  相似文献

Goldsinny Ctenolabrus rupestris were subjected to rapid, environmentally realistic, reductions in temperature at 2° C increments from 10 to 4° C over a 3-day period in full-strength sea water. In separate experiments, oxygen uptake measurements and ultrasound recordings of heart rate and opercular motion were carried out at regular intervals over the same temperature regime. Mean oxygen uptake rates fell from 0.042 to 0.028 ml O2 g−1 h−1 between 10 and 6° C respectively (Q10=2.71). Between 6 and 4° C mean rates decreased from 0.028 to 0.008 ml O2 g−1 h−1 (Q10=542). Mean opercular motion and heart beat rates decreased from 49.5 and 60.3 beats min−1 respectively at 10° C to 18.7 and 18.0 beats min−1 respectively at 4° C. Most goldsinny subjected to 4° C were observed in a torpid state and would not react to external stimulation. Opercular motion was erratic at 4° C and would at times cease altogether for periods up to 1.3 min duration. Heart movement was diffcult to detect at 4° C and may also have ceased for prolonged periods. Q10 values for opercular motion and heart beat rates recorded between 6 and 4° C were 6.39 and 24.52 respectively compared with values of 2.42 and 2.93 respectively recorded between 10 and 8° C. Such large depressions in metabolism appear not to have been reported previously for a marine fish species. No goldsinny mortalities were recorded at any temperature. The possibility that hypometabolic torpor is an adaptive strategy for goldsinny survival at low environmental temperatures is discussed.  相似文献

Production and food intake by an herbivorous pupfish population ( Cyprinodon nevadensis amargosae ) living in the outflow of a thermal artesian well (Tecopa Bore) near Death Valley, California is described. Water issues from the ground at 47.5° C and cools 8.12° C before leaving the study area 300 m from the source. High stream temperatures restricted the pupfish population to some 41 % of the study area, with a resulting mean density of 89 individuals m−2 (range = 13 to 196m−2). Biomass estimates ranged from 7 kcal m−2 to 42 kcal m−2. The mean annual standing crop of pupfishes (24 kcal m−2) turned over about five times annually. Growth rates were highest in juveniles (♂= 9.5% day−1) and slowest in large adults (♀= 08% day−1). Monthly production ranged from 22 kcal m−2 in September to 3 kcal m−2 in July and August. Pupfish in Tecopa Bore fed on algae and detritus, ingesting 1941 kcal m−2 yr−1 or 17.5% of the annual net primary production. 119 kcal m−2 yr−1 was deposited in growth. This latter value is approximately ten times greater than values previously reported for large carnivorous fishes but is comparable lo values reported for herbivorous fishes under pond culture.  相似文献
