Juvenile hormone III (JH) is synthesized by the corpora allata (CA) and plays a key role in mosquito development and reproduction. A decrease in JH titer during the last instar larvae allows pupation and metamorphosis to proceed. As the anti-metamorphic role of JH comes to an end, the CA of the late pupa once again synthesizes JH, which plays an essential role in orchestrating reproductive maturation. In spite of the importance of Aedes aegypti as a vector, a detailed study of the changes of JH hemolymph titers during the gonotrophic cycle has never been performed. In the present studies, using a high performance liquid chromatography coupled to a fluorescent detector (HPLC–FD) method, we measured changes in JH levels in the hemolymph of female mosquitoes during the pupal and adult stages. Our results revealed tightly concomitant changes in JH biosynthesis and JH hemolymph titers during the gonotrophic cycle of female mosquito. Feeding high sugar diets resulted in an increase of JH titers, and mating also modified JH titers in hemolymph. In addition these studies confirmed that JH titer in mosquitoes is fundamentally determined by the rate of biosynthesis in the CA.  相似文献

Hydrazino and aminooxy derivatives of β-alanine were found to cause blackening of Manduca sexta pupal cuticle when they were injected into pharate pupae at the onset of pre-ecdysial tanning. One of these compounds, ethyl hydrazinoacetate (EHA), was used for further study. It was effective if injected up to about 4 hr before pupal ecdysis. These melanized cuticles contained excessive amounts of dopamine and decreased amounts of N-β-alanyldopamine (NBAD) and N-acetyldopamine (NADA). Furthermore, EHA induced elevated dopamine and lowered β-alanine levels in the hemolymph. Similar blackening occurred when 20 mg/animal dopamine was injected. Injection of excess β-alanine rescued the normal brown color, irrespective of the concentration of EHA. Also, EHA caused melanization in vitro in the presence of dopamine, whereas the addition of β-alanine and NBAD allowed normal pupal coloration in vitro. These hydrazino and aminooxy compounds likely interfere with β-alanine synthesis or mobilization and thus with N-acylation of the catecholamines to form NBAD and N-β-alanylnorepinephrine.  相似文献

Analysis of extracts of hemolymph obtained from sexually mature alate females of Solenopsis invicta from monogyne colonies resulted in identification of juvenile hormone III (JH III). The average amount of JH III was 0.32±0.04 pmol/μmolof hemolymph. Topical application of 0.038 pmol of JH III was sufficient to stimulate alates to shed their wings in the presence of the queen. The time in which alates were induced to dealate decreased linearly with increasing concentrations of JH III from 0.038 to 3.8 pmol. However, higher JH III concentrations deviated from linearity and did not reach dealation times comparable with those that occur after mating flights. Thus, it appears that the mechanism of dealation that occurs when female alates are out of the influence of their queen is different from the one associated with mating flights. Application of 0.42 μmol of precocene II inhibited dealation of alates in queenless colonies. However, this inhibition was reversed after applying 38 pmol JH III to precocene-treated alates. The sizes of corpora allata (CA) from sexuals treated with JH III did not differ from those of controls. However, the sizes of CA were reduced in alates treated with precocene II. The results indicated that JH was important to dealation.  相似文献

In the present study, we report that contact with isolated female antenna significantly increases both the pheromone 3-hydroxy-2-butanone (3H-2B) release and the hemolymph JH III level in all examined aggressive posture-adopting (AP) and NP (non-AP-adopting) socially naïve males, with significantly faster concomitant pre-mating wing-raising behavior in AP as compared to NP males. 3H-2B release and JH III level were significantly increased after mating in both AP and NP males. A positive correlation was observed between mating experience and dominant status. Furthermore, mated-AP males initiated fights more rapidly and fought for a significantly longer duration than mated-NP males; retention with the paired female for 24 h did not affect this increase. JH III level and 3H-2B release were significantly increased in dominant males as compared to subordinates. These results suggest that prior mating experience in invertebrates may enhance aggression in subsequent male–male encounters, with accompanying physiological (hormone and pheromone) responses.  相似文献

Determining the specific molecular pathways through which dopamine affects behavior has been complicated by the presence of multiple dopamine receptor subtypes that couple to different second messenger pathways. The observation of freely moving adult bees in an arena was used to investigate the role of dopamine signaling in regulating the behavior of the honey bee. Dopamine or the dopamine receptor antagonist flupenthixol was injected into the hemolymph of worker honey bees. Significant differences between treated and control bees were seen for all behaviors (walking, stopped, upside down, grooming, flying and fanning), and behavioral shifts were dependent on drug dosage and time after injection. To examine the role of dopamine signaling through a specific dopamine receptor in the brain, RNA interference was used to reduce expression levels of a D1-like receptor, AmDOP2. Injection of Amdop2 dsRNA into the mushroom bodies reduced the levels of Amdop2 mRNA and produced significant changes in the amount of time honey bees spent performing specific behaviors with reductions in time spent walking offset by increases in grooming or time spent stopped. Taken together these results establish that dopamine plays an important role in regulating motor behavior of the honey bee.  相似文献

To explore the role of tyramine in the transformation of reproductive states of honeybee workers, brain levels of tyramine and N-acetyltyramine were measured in both normal and queenless workers. Queenless workers had higher tyramine levels and lower N-acetyltyramine levels than normal workers did. Intermediate reproductive workers that were transferred into a normal colony from a queenless colony had intermediate levels of tyramine and N-acetyltyramine. Elevation of tyramine in the queenless workers occurred at an earlier adult stage than elevation of dopamine. Tyramine levels in intermediate reproductive workers returned to the levels seen in normal workers, but dopamine levels in intermediate reproductive workers remained elevated at the same level as in queenless workers. Thus, brain tyramine may be regulated by the colony condition with or without a queen. Injection of an amine uptake inhibitor, reserpine, depleted tyramine and elevated N-acetyltyramine. Distributions of tyramine and dopamine within the brain were distinctively different, whereas distributions of N-acetyltyramine and N-acetyldopamine were similar, suggesting that each functional amine is stored in specific neurosecretory cells and released to the relevant receptor sites but that metabolism into each N-acetylmetabolite is determined by diffusion.  相似文献

Phenol oxidase (PO) and dopachrome conversion enzyme (DCE) were partially purified from Aedes aegypti larvae by ammonium sulfate fractionation. PO from A. aegypti functions in the hydroxylation of monophenols (e.g., tyrosine and tyramine) to their related o-diphenols, and the oxidation of o-diphenols (e.g., l-dopa, dopamine, N-acetyldopamine) to their respective o-quinones. Partially purified DCE showed high specificity toward dopachrome generated from dopa with the l-configuration. The combined effects of PO and DCE significantly accelerated melanization pathways when l-dopa was used as substrate. Significant DCE activity also was detected in hemolymph samples from adult, female A. aegypti, and undoubtedly plays a role in melanotic encapsulation reactions.  相似文献

N-acetyldopamine is rapidly and extensively converted to N-acetyldopamine 3-O-phosphate and N-acetyldopamine 3-O-sulphate in the newly ecdysed cockroach, Periplaneta americana. Dopamine 3-O-sulphate also serves as a naturally occurring precursor of N-acetyldopamine 3-O-sulphate in vivo.Radioisotope experiments revealed that the N-acetyldopamine moiety of these esters is incorporated into the cuticle during sclerotization and that the phosphate and sulphate moieties are not (not, at least, as the intact esters). It is suggested that N-acetyldopamine 3-O-phosphate and N-acetyldopamine 3-O-sulphate may be the forms in which the eventual cuticular sclerotizing agent (N-acetyldopamine?) is transported from the blood into the epidermis by a ‘carrier’ protein.  相似文献

The oriental fruit fly, Bactrocera dorsalis (Hendel) (Diptera: Tephritidae), is considered a major economic threat in many regions worldwide. In order to better understand the flight capacity of B. dorsalis and its physiological basis, the functions and regulatory roles of juvenile hormone (JH) in the flight muscle of B. dorsalis were studied under a controlled environment. JH titer of B. dorsalis varied with age and sex. Females have higher JH titers and better flight capabilities than males, given that the increase in JH also corresponded to the ovarian development and maturation in females. The flight duration and distance of both males and females increased with the gradual increase of JH titer after adult emergences. Both JH titer and flight capability peaked in 15-d-old adult and declined subsequently with age. Flight activity stimulated the production of JH as adults flown for 24 h on the flight mills have the highest JH titers compare to adults flown on shorter flight durations. Adults treated with 0.5 µg and 5 µg of JH III were able to fly long durations and long distances, nevertheless when JH titer was too low or too high, it would restrict the flight ability of the fly. The mutual reinforcement of JH and flight activity provides fundamental understanding on the physiological aspects of the flight capability and dispersal, which facilitates strategies for the long-term control of this destructive pest.  相似文献

N-β-Alanyldopamine (NBAD) and other diphenols in tissues of the fifth larval instar of the tobacco hornworm, Manduca sexta (L.), were analyzed by HPLC with electrochemical detection. NBAD accumulated in the integument during the intermolt feeding period, with maximal levels in the wandering stage (6 mmol/g). It then declined to a low level during apolysis and endocuticle digestion, while hemolymph NBAD increased during the same interval to a peak concentration (3 mM) shortly before pupal ecdysis. Trachea and foregut contained lesser amounts of NBAD (0.5 mmol/g), perhaps associated with cuticle, whereas fat body, muscle, midgut and hindgut had 0.1 mmol/g or less. Dopamine (DA), N-acetyldopamine and 3,4-dihydroxyphenylalanine (DOPA) were at least 10-fold less abundant than NBAD in the integument. NBAD synthase, which catalyzes the formation of NBAD from DA and β-alanine, was assayed in both integument and fat body. Highest activity was detected in the integument, where two peaks were observed, one at day 3 near the end of larval feeding and the other at day 9 as pupal cuticle tanning was initiated. Fat body enzyme was substantially less and was detected only in the pharate pupa. Maximal NBAD synthesis by integument cultured in vitro was dependent upon DA supplementation of at least 1.4 mM. 20-Hydroxyecdysone did not alter NBAD synthesis in vitro in either the integument or the fat body, even though injection of this hormone into isolated larval abdomens induced synthesis and/or transport of integumental NBAD back into the hemolymph. The rate-limiting steps in the NBAD biosynthetic pathway appear to be the production of DOPA and DA, because β-alanine occurs in the hemolymph at relatively high levels throughout larval-pupal development.  相似文献

By injection of azadirachtin (AZA) to 5th instar hoppers of Locusta migratoria migratorioides, “over-aged” nymphs were produced, some of which survived for long periods without moulting to adults. Such over-aged male nymphs exhibited mating behaviour belatedly, though at a lower intensity than normal adults. Juvenile hormone (JH) markedly intensifies male sexual behaviour in crowded adult (6th instar) Locusta; thus, the lower intensity of sexual behaviour of the over-aged nymphs might have been caused by a low endogenous JH titre, or by a weaker response to the hormone. To clarify this point, chemically allatectomized over-aged 5th instar nymphs were produced by combined treatment with precocene and AZA. These exhibited a very low intensity of mating behaviour. Injections of exogenous JH III intensified the mating behaviour of the chemically allatectomized over-aged 5th instar male nymphs in a dose-dependent way, but the effect was weaker than in chemically allatectomized adult males. AZA-induced over-aged 4th instar male Locusta hoppers also exhibited mating behaviour. By injection of AZA, 5th instar over-aged hoppers of Schistocerca gregaria were also produced. Males showed mating behaviour belatedly, despite the fact that JH completely controls male sexual behaviour in this species. The intensity of this behaviour was again lower than in adults. Interrelations between integumental morphogenesis, “ethogenesis” of male sexual behaviour, and endocrine factors in locusts are discussed.  相似文献

Diflubenzuron (DFB) has been known to prevent metamorphosis of silkworm, Bombyx mori, from larval to pupal stage at low dose exposure. To explain this inhibitory action of DFB, a hypothesis was raised that DFB acts like juvenile hormone (JH) or DFB inhibits JH esterase to increase endogenous JH titer. A JH bioassay using isolated abdomen clearly indicates that DFB does not act as JH analog because DFB did not induce vitellogenesis in the isolated female abdomen, while endogenous JHs did significantly. General esterase activities in hemolymph were lower in DFB-treated fifth instar larvae than in the control larvae, but there was no difference between fat body esterase activities in both groups. Two hemolymph esterases (‘E1’ and ‘E2’) of the fifth instar larvae were separated and visualized by α-and β-naphthyl acetate. From in vitro incubation experiment, the cathodal esterase (‘E1’) was sensitive to DFB at its nanomolar range. Considering the fact that early fifth instar larvae have high level of JH esterase in the hemolymph, these results suggest that DFB inhibit larval to pupal metamorphosis by blocking JH degradation, which increases endogenous JH titer especially at the critical period when the larvae determine metamorphic development at the following molt.  相似文献

Hemolymph of Danaus plexippus (monarch butterfly) was analyzed for juvenile hormone titer by gas liquid chromatography-mass spectrometry (GC-MS) in the selected ion monitoring mode. Laboratory reared, reproductively active, adult males and females contained JH I, II and III. JH II predominated with titers ranging from 2.3 to 11 ng/ml hemolymph. Titers of JH I and JH III were approx. 1 order of magnitude lower than those of JH II. JH I, II and III acids were also detected, but at levels generally lower than the corresponding JHs. JH titers in field collected reproductively inactive, gregarious adult monarchs, were 1–2 orders of magnitude lower than those of laboratory reared reproductively active monarchs. JH II was again the predominant JH in these animals.  相似文献

The effect of 20-hydroxyecdysone (20E) and juvenile hormone (JH) on the glutathione pathway of the greater wax moth Galleria mellonella (Lepidoptera: Pyralidae) was determined by investigating glutathione peroxidase (GSH-Px), glutathione S-transferases (GST), and glutathione reductase (GR) activities as well as reduced and oxidized glutathione (GSH and GSSG) content with respect to developmental stage. The continuous decreases of GSH-Px and GST activities dependent on the growth period of G. mellonella occurred in JH and 20E groups over and under their controls, respectively. While the GR activities of G. mellonella showed increases in young pupa (YP) for both control and in old larvae (OL) for the 20E groups after the minimum at these periods, they also increased after old pupa (OP) for the JH group with a maximum in OL period. Although GR activity levels in the JH group were significantly higher compared with controls and 20E groups up to OP period, the activity levels for the control and 20E groups were higher than those of the JH group at adult (AD) and old pupa (OP) periods, respectively. In spite of increases in the GR activity of 20E and control groups of G. mellonella, decreased GSH and increased GSSG levels were observed at aging period. GSH levels in the JH group reached a maximum at prepupa (PP) and then decreased with non-significant changes from OL to AD period. According to the results, GSH and GSSG levels, as well as GSH/GSSG ratios, were below and over control levels in 20E and JH groups, respectively, during all of the investigated developmental stages. On the contrary, the LPO levels were higher than the control for 20E and lower for the JH groups during the developmental period. These results show that while ecdysone hormone has a negative effect on the glutathione-related detoxication capacity of G. mellonella, the juvenile hormone has a positive effect on this process.  相似文献

The effects of dopamine (DA) on juvenile hormone (JH) metabolism and fitness (estimated as fecundity and viability levels under heat stress (38 °C)) in Drosophila virilis have been studied. An increase of DA level obtained by feeding with DA reduced fitness of wild-type (wt) flies under stress, and decreased JH degradation in young wt females while increasing it in sexually mature wt females. A decrease in DA levels resulted from 3-iodo-tyrosine treatment and caused a decrease in JH degradation in sexually mature wt and heat sensitive (hs) mutant females (DA level in hs females is twice as high in wt females). A dramatic decrease in viability under stress and fecundity under normal conditions in wt, but not hs, females was observed. 3-iodo-tyrosine treatment also reduced the number of oocytes at stages 8-14, delayed oocyte transition to stage 10 and resulted in the accumulation of mature eggs in wt females. It delayed maturation of wt, but not hs, males as well, but did not affect their fertility. This advances our understanding of the regulation of JH metabolism by DA in Drosophila and suggests a crucial role for the basal DA level in fitness.  相似文献

Juvenile hormone (JH) is necessary for the production of vitellogenin (Vg) in the boll weevil, Anthonomus grandis. Occurrence of Vg in this species is typically restricted to reproductively competent females, and is not detected in untreated males. However, the JH analog, methoprene stimulates Vg production in intact males and in the isolated abdomens of both male and female boll weevils (where in each case no Vg is detected without treatment), suggesting that males are competent to produce Vg but are normally not stimulated to do so. Preliminary work indicating that male boll weevil corpora allata (CA) produced little or no JH in vitro suggested that failure of males to produce Vg might be due to very low JH levels compared to females. This study re-examines the question of JH in male boll weevils by determining in vitro production of JH III by male CA during the first 10 days after adult emergence, determining hemolymph JH esterase activity during this same time period and hemolymph JH III titers in adults of both sexes. We also re-examine the ability of isolated male abdomens to produce Vg in response to hormonal stimulation, analyzing the effect of a wide range of methoprene and JH III dosages. Results indicate that male A. grandis have circulating JH titers and JH production similar to females. JH esterase activity is slightly but significantly higher in males than females. Vg production by isolated abdomens of both sexes after stimulation with methoprene or JH III was confirmed. Dose response studies indicated that high levels of methoprene were less effective than intermediate doses in stimulating Vg synthesis in both sexes. We conclude that the sexually dimorphic effect of JH on Vg synthesis is not due to differences in JH production or differences in JH titer between the sexes.  相似文献

Tryptophan, 5-hydroxytryptophan, 5-hydroxytryptamine, a 5-hydroxyindolacetic acid-like substance, N-acetyl-5-hydroxytryptamine, dopamine, N-acetyldopamine and octopamine were assayed simultaneously in the central nervous system of virgin and fed females Blaberus craniifer Burm.2. Amounts were quantified by HPLC and electrochemical detection in both crowded and isolated females during the first weeks of imaginal life, from days 0–30.3. Amine contents were modified by housing conditions, and in general, higher levels were found in the isolated individuals, with the most pronounced differences observed at D10 and D30.4. The relationship between these fluctuations and the speeding up of ovarian maturation by isolation is discussed.  相似文献

Abstract Dopamine has been suggested to be involved in physiological and/or behavioural changes triggered by mating in European honeybee (Apis mellifera) queens but its specific role remains unclear. In the present study, the amounts of dopamine (DA) and its metabolite, N‐acetyldopamine (NADA) are measured, in queens of various ages to clarify the association with locomotor activity. The effects of DA receptor agonist/antagonist drugs on locomotor activity are further investigated. Brain levels of DA and NADA are relatively constant during the period before mating when locomotor activity reportedly increases with age but decreases in 1‐year‐old laying queens with low locomotor activity. Reduced DA and NADA levels are also found in haemolymph of 1–3‐month‐old laying queens. When a DA receptor agonist or antagonist is injected into 6‐day‐old virgin queens, locomotor activity levels increase significantly with 2‐amino‐6,7‐dihydroxy‐1,2,3,4‐tetrahydronaphthalene (agonist), and decreased with cis(Z)‐flupenthixol (antagonist). These results suggest that DA systems are involved in the motor control of honeybee queens, and that the decline in DA levels reduces locomotor activity after mating but increased locomotor activity before mating may be independent of DA levels.  相似文献
