Background: C14:0, C15:0, C17:0 and trans-C16:1(n-7) are often used as biomarkers for dairy fat intake. Trans-C18:1(n-7) and CLA, two fatty acids which are also present in dairy, have hardly been explored. We investigated whether trans-C18:1(n-7) and CLA can enrich the existing biomarker portfolio.

Methods: Data were obtained from Lifelines (n?=?769). Dairy fat intake was determined by FFQ. Fatty acids were measured in fasting plasma triglycerides (TG), phospholipids (PL) and cholesterol esters (CE).

Results: Median (25th–75th percentile) intakes of dairy and dairy fat were 322(209–447) and 12.3(8.4–17.4) g/d respectively. A pilot study showed that trans-C18:1(n-7) and CLA were only detectable in TG and PL. Of the established markers, TG C15:0 was most strongly associated with dairy fat intake (standardized β (std.β)?=?0.286, R2?=?0.111). Of the less established markers, TG trans-C18:1(n-7) was most strongly associated with dairy fat intake (Std.β?=?0.292, R2?=?0.115), followed by PL CLA (Std.β?=?0.272, R2?=?0.103) and PL trans-C18:1(n-7) (Std.β?=?0.269, R2?=?0.099). In TG, a combination of C15:0 and trans-C18:1(n-7) performed best (R2?=?0.128). In PL, a combination of C14:0, C15:0, trans-C18:1(n-7) and CLA performed best (R2?=?0.143).

Conclusion: Trans-C18:1(n-7) and CLA can be used as biomarkers of dairy fat intake. Additionally, combining established with less established markers allowed even stronger predictions for dairy fat intake.  相似文献

Objective: The objective was to investigate blood-based biomarkers of type I (PRO-C1), III (PRO-C3) and VI (PRO-C6) collagen formation in systemic sclerosis (SSc) patients and examine their correlation to modified Rodnan skin score (mRSS).

Methods: Limited (lSSc, n?=?76) and diffuse SSc (dSSc, n?=?41) fulfilling the ACR/EULAR 1980 and 2013 classification criteria for SSc and asymptomatic controls (n?=?9) were included. PRO-C1, PRO-C3 and PRO-C6 were measured in serum.

Results: LSSc compared to dSSc were significantly older, had longer disease duration and lower mRSS. PRO-C3 was higher in early dSSc compared to early lSSc (mean [95 percentile], 27.4 [13.1–39.1] ng/mL vs 14.9 [8.2–28.8] ng/mL, p?=?0.006). PRO-C6 levels were higher in early dSSc compared to early lSSc and late dSSc (early dSSc: 28.2 [10.4–92.3] ng/ml vs early lSSc: 11.0 [6.9–28.5] ng/ml; p?=?0.006 and late dSSc: 12.6 [6.5–25.3] ng/mL, p?=?0.04). No difference was observed with PRO-C1. PRO-C3 and PRO-C6 were moderately correlated with mRSS with R-partials of 0.36 (p?<?0.001) and 0.29 (p?=?0.002), respectively

Conclusion: Measures of type III and VI collagen formation are potential objective biomarkers of fibrosis in systemic sclerosis. These biomarkers could be useful in monitoring the disease and efficacy of treatment.  相似文献

Purpose: The main objective of the present study was to evaluate the effects of laterality discrimination training on neck joint position sense and cervical range of motion (ROM) in patients with chronic non-specific neck pain (NSCNP).

Materials and methods: Forty-eight patients with NSCNP were randomly assigned to the neck group (NG) that observed neck images or the foot group (FG) that observed foot images. Response time, response accuracy, cervical ROM, and joint position error (JPE) were the main variables. The secondary outcome measures included psychosocial variables.

Results: Differences between groups in the cervical ROM for flexion (p?=?.043) were obtained, being NG group the one which obtained greater values. NG showed an improvement in right rotation (p?=?.018) and a decrease in flexion was found in the FG (p?=?.039). In JPE, differences between groups were obtained in the left rotation (p?=?.021) and significant changes were found in the NG for flexion, extension, and left rotation movements (p?<?.05). Moderate associations were found between left and right accuracy regarding to post-intervention flexion and right rotation (r?=?0.46, r?=?0.41; p?<?.05) in NG.

Conclusion: Improvements in cervical range of motion and joint position sense are obtained after the performance of the laterality discrimination task of images of the neck but not the feet. Visualization of images of the painful region presents moderate correlations with the accuracy and response time in the movements of flexion and right rotation.  相似文献

Differences between two maize and two triticale genotypes grown in low soil compaction (LSC), moderate soil compaction (MSC) and severe soil compaction (SSC) and with a limited (D) or excess (W) soil water content were observed as a decrease in shoot (S) and root (R) biomass, leaf greening (SPAD) and increase in membrane injury (LI), root and leaf water potential (ψ), photosynthesis (Pn), transpiration (E) and stomata conductance (gS). Close correlations between ψL and ψR, and between differences ψL and ψRψ) were found. Drought or waterlogging with LSC conditions in both maize genotypes resulted in higher WUE than in control plants (LSC C), but under the SSC WUE declined. However, for triticale differences in WUE, between treatments were small and insignificant. In general, changes in markers were greater for genotypes sensitive to the soil compaction (Ankora, CHD-12) than in resistant ones (Tina, CHD-247) and were higher in seedlings grown under SSC conditions.

The process of combating neoplasms and mononuclear cells, and during H. pylori infection, several pro-inflammatory and anti-inflammatory cytokines are synthesized. In view of the involvement of the IL-6 law and the presence of H. pylori in the development of gastric diseases, the present study aimed to characterize the promoter-region polymorphism −597 (G/A) (rs1800797), −572 (C/G) (rs1800796), and −174 (G/C) (rs1800795) by PCR-RFLP in 375 gastric biopsy specimens from patients with peptic symptoms. A total of 375 samples were analyzed: 87 patients (without lesion without gastric tissue); 236 patients with gastritis and 52 patients with gastric cancer analyzed the PCR-RFLP techniques. All the results were normalized in relation to the presence of H. pylori. The frequencies of the three polymorphisms were compared in the Control vs Gastritis groups and a statistically significant test observed: −174 (G/C) (OR: 1.27; 95% CI: 0.84–1.93; P = 0.26), 572 (C/G) (OR: 1.42; 95% CI: 0.78–2.59; P = 0.25), and 597 (G/A) (OR: 0.98; 95% CI, 0.64–1.52; P = 0.94). Similar results were obtained when the gastric cancer group was compared to the control group: −174 (G/C) (OR: 1.27; 95% CI: 0.66–2.47; P = 0.47), −572 (C/G) (OR: 1.07; 95% CI: 0.43–2.68; P = 0.88), and −597 (G/A) (OR: 1.01; 95% CI, 0.5–0.9; P = 0.99). The haplotypes were and were not observed statistically significant differences. In conclusion, we found no correlations between any of the three polymorphisms in the IL-6 gene analyzed in this study and a higher risk of gastritis or gastric cancer.  相似文献

Introduction. Heterophyllium albicans Thér., H. henryi Tixier, H. micro-alare (Broth. & Paris) Broth. and H. tonkinense (Broth. & Paris) Broth. were all either poorly known species or known only from the type collections since their inception.

Methods. Examination of the type material confirmed that the alar cell structure in these species do not conform to the generic concept of Heterophyllium (Schimp.) Kindb. The study also revealed some new features previously unreported in these species, i.e., the filamentous pseudoparaphyllia, one of the characteristic features of the genus Isopterygium Mitt.

Key Results & Conclusions. Three new combinations, Isopterygium albicans (Thér.) Y.Jia & S.He, I. micro-alare (Broth. & Paris) Y.Jia & S.He, and I. tonkinense (Broth. & Paris) Y.Jia & S.He are proposed. Heterophyllium henryi Tixier is treated as a new synonym of Brotherella henonii (Duby) M.Fleisch.  相似文献

An efficient and reproducible protocol is described for the regeneration of Astragalus melilotoides protoplasts isolated from hypocotyl-derived embryogenic calli. Maximum protoplast yield (11.74±0.6×105/g FW) and viability (87.07±2.8%) were achieved using a mixture of 2% (w/v) Cellulase Onozuka R10, 0.5% (w/v) Cellulase Onozuka RS, 0.5% (w/v) Macerozyme R10, 0.5% (w/v) Hemicellulase, and 1% (w/v) Pectinase, all dissolved in a cell protoplast wash (CPW) salt solution with 13% (w/v) sorbitol. First divisions occurred 3–7 days following culture initiation. The highest division frequency (9.86±0.68%) and plating efficiency (1.68±0.05%) were obtained in solid-liquid medium (KM8P) supplemented with 1.0 mg/l 2,4-dichlorophenoxyacetic acid, 0.5 mg/l 6-benzylaminopurine (BA), 0.2 mg/l kinetin, 0.2 M glucose, 0.3 M mannitol and 500 mg/l casein hydrolysate. Upon transfer to MS medium with 0.5 mg/l -naphthaleneacetic acid and 1-2 mg/l BA, the protoplast-derived calli produced plantlets via somatic embryogenesis (56.3±4.1%) and organogenesis (21.6±0.6%). Somatic embryos or adventitious shoots developed into well-rooted plantlets on MS medium without any plant growth regulators or supplemented with 3.0 mg/l indole-3-butyric acid, respectively. Context: Liposomes have been shown to improve human red blood cell (RBC) in vitro quality by minimizing membrane damage occurring during 42-d hypothermic storage. Small animal models are necessary to evaluate novel blood products and guide future clinical studies.

Objectives: The aim of this study was to assess the effect of liposome treatments on rat RBC hypothermic storage lesion (HSL) and to examine in vivo outcomes of transfusing liposome treated RBCs in a rat model.

Materials and methods: Unilamellar liposomes were synthesized which contained saturated (DPPC:CHOL, 7:3?mol%), unsaturated (DOPC:CHOL, 7:3?mol%), saturated charged (DPPC:CHOL:PS, 6:3:1?mol%), and unsaturated charged (DOPC:CHOL:PS, 6:3:1?mol%) phospholipids. After liposome treatment, rat RBC quality was assessed by percent hemolysis, deformability, aggregation, hematological indices, microvesiculation, and cholesterol/phospholipid concentrations. An anemic rat model of myocardial ischemia and reperfusion (I/R) was used to evaluate the outcomes of transfusing liposome-treated RBCs.

Results: All four liposome treatments resulted in significant decreases in hemolysis, with the most prominent effect seen with DOPC-liposomes (DOPC: 1.6?±?0.1% versus control: 3.1?±?0.2%, p?=?0.015). RBCs treated with uncharged liposomes had lower hemolysis compared with charged liposomes (3.4?±?0.2% versus 3.9?±?0.4%, p?=?0.010). The in vivo study showed no significant difference in the hemoglobin levels and infarct size (53.3?±?13.1% versus 45.3?±?8.4%, p?=?0.223) between liposome and control groups.

Discussion and conclusion: Liposome treatment improved in vitro quality of stored rat RBCs. However, the changes observed in vitro were not sufficient to improve the in vivo outcomes of myocardial I/R in anemic rats transfused with liposome-treated RBCs.  相似文献

Background: The presence of exotic plants increases the heterospecific pollen (HP hereafter) received by native plants and reduces their reproductive output.

Aims: We assessed whether the exotic herb Echium vulgare (Boraginaceae) increased HP and reduced seed output of the native plants Phacelia secunda (Boraginaceae) and Stachys albicaulis (Lamiaceae) in an Andean locality, central Chile.

Methods: The presence of HP was studied in native plants growing with and without co-existence with the exotic E. vulgare. A complementary hand pollination experiment was carried out to assess whether E. vulgare pollen reduced the reproductive success of native plants.

Results: In the presence of E. vulgare, 17.3% and 3.7% of P. secunda and S. albicaulis individuals that coexisted with the exotic species received HP. For P. secunda, the number of conspecific pollen grains decreased in invaded patches compared with non-invaded patches; no differences were observed for S. albicaulis. The pollen of E. vulgare negatively affected the reproductive success of S. albicaulis but not that of P. secunda.

Conclusions: The presence of HP cannot be predicted from the presence of exotic plants alone, and other factors, such as flower morphology, could explain the greater HP transfer in P. secunda (actinomorphic flowers) than in S. albicaulis (zygomorphic flowers). A higher negative effect of E. vulgare pollen on P. secunda versus S. albicaulis could be related to the phylogenetic resemblance between the exotic donor and native recipient plant because pollen-stigma compatibility may be evolutionary conserved through common lineages.  相似文献

Objectives: In this study, an improved lyophilized PEGylated liposomal formulation of docetaxel (DOC) has been developed.

Methods: PEGylated docetaxel liposome (PL-DOC) was prepared by thin-film evaporation method and lyophilization. The effect of various components of the lipids and their compatibility with DOC on the entrapment efficiency (EE) of liposome was investigated. The lyophilized PL-DOC was characterized by morphology, particle size, zeta potential, EE, release in vitro and stability. Pharmacokinetics and biodistribution in vivo of lyophilized PL-DOC were also investigated.

Results: The optimal liposome formulation was egg phosphatidylcholine (EPC):cholesterol (CH):DSPE-PEG2000:DOC?=?56:40:4:4 (molar ratio). Sucrose and mannitol were chosen as cryoprotectant in the lyophilization (cryoprotectant-to-lipid (C/L) mass ratio = 8:1). The size of lyophilized PL-DOC was 152.3?±?1.0?nm with negative charge and the EE was 89.75?±?1.79%. Compared with nonlyophilized PL-DOC, the lyophilized PL-DOC was more stable at 4?°C for six months. The lyophilized PL-DOC also showed the good stability after reconstituted by 5% glucose injection. In vitro release study of PL-DOC showed that PL-DOC had a sustained release effect. After i.v. administration at the dose of 10?mg/kg in rats, a significant increase in the AUC0-∞, MRT0-∞ and t1/2 was observed in PL-DOC group compared with conventional docetaxel liposome (CL-DOC) and DOC injection (DOC-I) group. Biodistribution studies in mice showed that PL-DOC significantly decreased the uptake by the organs of mononuclear phagocytic system (MPS), such as liver and spleen, while prolonging the retention time of DOC in the plasma.

Conclusion: Our PEGylated liposome formulation reported in this study could potentially produce viable clinical strategies for improved delivery of DOC for the treatment of human cancer.  相似文献

Introduction. The present study investigates the taxonomy of European members of Weissia Hedw. subgenus Astomum Hampe., a group considered taxonomically difficult and including species of high conservation concern.

Methods. A broad set of samples were subject to DNA sequencing and morphological analysis, plus a review of type material was undertaken.

Key Results and Conclusions. Three taxonomic additions and changes are supported. (1) Weissia longifolia var. angustifolia (Baumgartner) Crundw. & Nyholm is raised to species rank, W. angustifolia (Baumgartner) D.A.Callaghan, comb. et. stat. nov. (2) The type of W. multicapsularis (Sm.) Mitt. comprises an intermix of Phascum cuspidatum Hedw. and W. longifolia Mitt., and the protologue refers to unique features of P. cuspidatum but no such features of W. longifolia. Weissia multicapsularis is therefore described as a new synonym of P. cuspidatum. (3) Plants that have been named as W. multicapsularis by modern authors comprise an undescribed species, here named W. wilsonii D.A.Callaghan, sp. nov. An illustrated key to European species of Weissia subgenus Astomum is provided.  相似文献

Raphidophyte algae (Raphidophyceae) can be divided according to pigment composition and plastid ancestry into two categories, brown‐ and green‐pigmented taxa. We sought to examine if there are any biochemical differences in plastid lipid composition between the two groups. To this end, the composition and positional distribution of fatty acids of the chloroplast lipids, mono‐ and digalactosyldiacylglycerol (MGDG and DGDG, respectively), were examined using positive‐ion electrospray/mass spectrometry (ESI/MS) and electrospray/mass spectrometry/mass spectrometry (ESI/MS/MS). Brown‐pigmented strains from the genera Chattonella, Fibrocapsa, and Heterosigma primarily consisted of 20:5/18:4 (sn‐1/sn‐2) MGDG and 20:5/18:4 DGDG, while isolates of the green‐pigmented raphidophyte Gonyostomum semen (Ehrenb.) Diesing contained these as well as 18:3/18:4 MGDG and DGDG, thus underscoring its green algal plastid lineage. Although previously unseen without the regiochemical information provided by ESI/MS/MS, Chattonella subsalsa Biecheler possessed 20:5/18:3 DGDG as a major form, a potential biosynthetic intermediate in the production of 20:5/18:4 DGDG. These results provide a modern interpretation of the fatty acid regiochemistry of MGDG and DGDG.  相似文献

Thirty-three oil samples isolated from aerial parts of Myrtus communis L. harvested in seven localities, from Northern to Central Morocco, have been analyzed by combination of chromatographic and spectroscopic techniques. The 33 compositions have been subjected to statistical analysis, hierarchical cluster analysis (HCA) and principal component analysis (PCA). Two groups have been differentiated on the basis of their myrtenyl acetate and α-pinene contents and each one was sub-divided in two sub-groups according to the contents of 1,8-cineole and linalool. The compositions of our 33 myrtle oil samples may be named as follow by their main components: sub-group IA (13/33): α-pinene/1,8-cineole/linalool; sub-group IB (6/33): 1,8-cineole/α-pinene; sub-group IIA (10/33): 1,8-cineole/myrtenyl acetate; sub-group IIB (4/33): myrtenyl acetate.  相似文献

Sesamin is a specific inhibitor of Δ5 desaturation, the conversion from dihomo-γ-linolenic acid (20: 3, n-6) to arachidonic acid (AA, 20: 4, n-6). Previously, we reported that sesamin inhibited Δ5 desaturation of n-6 fatty acids in rat hepatocytes but not that of n-3 fatty acids, from 20: 4 (n-3) to eicosapentaenoic acid (EPA, 20: 5, n-3). In this study, we investigated the interaction of sesamin and EPA on Δ5 desaturation of both series and the n-6/n-3 fatty acids ratio by measuring actural fatty acid contents in vivo. Rats were fed three types of dietary oils; 1) linoleic acid (LA, 18: 2, n-6): linolenic acid (LLA, 18: 3, n-3) = 3: 1, n-6/n-3 ratio of 3: 1 (LA group), 2) LA: LLA =1: 3, n-6/n-3 ratio of 1: 3 (LLA group), 3) LA: LLA: EPA =1: 0.5: 3, n-6/n-3 ratio of 1: 3.5 (EPA group) with or without sesamin (0.5% w/w) for 4 weeks. In all groups, sesamin administration increased the content of dihomo-γ-linolenic acid (20: 3, n-6) in the liver and decreased the Δ5 desaturation index of n-6 fatty acid, the ratio of 20: 4/20: 3 (n-6). On the contrary, the Δ5 desaturation index of n-3 fatty acid, the ratio of 20: 5 + 22: 5 + 22: 6/20: 4 (n-3), was increased by the administration of sesamin. These results suggest that sesamin inhibits the A5 desaturation of n-6 fatty acid, but not that of n-3 fatty acid in rat livers. Sesamin administration decreased incorporation of EPA (n-3) and simultaneously increased the AA (n-6) content in the liver. The n-6/n-3 ratio in the liver was increased by administering sesamin under n-3 rich conditions, i.e., the LLA and EPA groups.  相似文献

Abstract

Objective: To evaluate whether urinary phospholipids could be regarded as biomarkers of chronic kidney disease.

Materials and methods: Thirteen healthy volunteers and 26 consecutive chronic kidney disease patients were included. Urinary phospholipids were quantified by high-performance liquid chromatography coupled with electrospray ionization tandem mass spectrometry.

Results: Urinary phosphatidylcholines concentrations (PC 16:0/16:0, 16:0/22:3, 16:0/18:1 and 16:0/18:2) were significantly higher both in glomerulonephritis group (all p?<?0.001) and in tubulointerstitial injury group (all p?<?0.05) than in healthy control group. Meanwhile, sphingomyelin concentrations (SM 18:1/16:0 and 18:1/18:0) in glomerulonephritis group were significantly higher than those in healthy control group (all p?<?0.001). Urinary PCs and SMs were positively correlated with proteinuria but negatively correlated with serum albumin. Meanwhile, PCs were positively correlated with serum creatinine.

Conclusion: Our work first demonstrated that urinary phospholipids might be biomarkers for the chronic kidney disease patients. Increased urinary phospholipids in chronic kidney disease patients might result from proteinuria, damaged kidney function or proteinuria induced hypoalbuminemia or lipotoxicity.  相似文献

Context: Troponin (hs-TnT) levels predict mortality after acute exacerbation of COPD (AECOPD). Whether this is independent of heart failure (HF) is not established.

Material and methods: Prospectively included AECOPD patients adjudicated for acute HF categorized into three groups: (A) AECOPD, but acute HF the primary cause for hospitalization; (B) AECOPD the primary cause, but co-existing myocardial dysfunction and (C) AECOPD without myocardial dysfunction.

Results: About 103 AECOPD patients; 18% A, 27% B and 54% C. Hs-TnT level differed between the groups: (ng/l, median) A: 41, B: 25 and C: 15, p?=?0.03 for A versus B and p?=?0.005 for B versus C. During a median 826 days, 47% died. In Cox analysis, hs-TnT levels remained associated with mortality (hazard ratio per 10?ng/l 1.3, p?<?0.0001).

Conclusion: hs-TnT levels are influenced by myocardial dysfunction/HF in AECOPD, but provide independent prognostic information. The prognostic merit of hs-TnT cannot be attributed to HF alone.  相似文献

Cedrelopsis grevei H. Baillon bark, endemic plant from Madagascar, is used in folk medicine for the treatment of rheumatism, muscular pain, and for its antifungal and antibiotic activities. In this paper, the phytochemical composition, antioxidant, antimicrobial, and cytotoxic activities of C. grevei bark essential oil (EO), its non-polar (I and II) and polar (III) fractions and its main compounds (ishwarane, β-elemene and α-copaene) were investigated.

The GC–MS analysis pointed out the presence of 36 components, representing about 80% as semi-quantitative characterization of the total. The presence of ishwarane, β-elemene and α-copaene as the main constituents highlighted its peculiar composition as a sesquiterpene-rich phytocomplex. Moreover, the quantification was performed for the first time by means of the experimental and predicted response factors (ERFs and PRFs, respectively).

As regards the biological activity, C. grevei EO and its fractions showed weak antioxidant activity against Trolox. The whole EO demonstrated instead considerable antimicrobial activity against Staphylococcus aureus and Staphylococcus epidermidis, while its polar-fraction evidenced an interesting bioactivity against Pseudomonas aeruginosa and Candida albicans. Finally, C. grevei EO and its fractions exhibited an interesting cytotoxic activity on human lung cancer cells (A549) and human colorectal cancer cells (CaCo-2).  相似文献
