The level of ACTH in bovine pituitary intraglandular colloid (IGC/ACTH), the holocrine secretion of the marginal half of the intermediate lobe (IL) cells, was found to correlate with the concentration of this peptide in blood plasma (BP/ACTH), cerebrospinal fluid (CSF/ACTH), the paraventricular nuclei (PVN/ACTH) and the arcuate nuclei (AN/ACTH). The ACTH content in all sites was measured by radioimmunoassay (RIA). Although the IL is virtually avascular, the intraglandular lumen housing the IGC communicates nonetheless with the venous cavernous sinuses as well as the subarachnoid cerebral spinal fluid (CSF). Therefore, we content that the IGC serves as a transport medium whereby IL materials gain access to these extrapituitary sites.  相似文献

ACTH-lipid interactions were investigated by: (1) lipid-monolayer studies using several zwitterionic and anionic phospholipids and gangliosides, (2) permeability experiments by following the swelling rate of liposomes in isotonic glycerol solutions by light scattering, using liposomes of synthetic lipids and liposomes made of lipids extracted from light synaptic plasma membranes, and (3) by steady-state fluorescence anisotropy measurements on liposomes derived from light synaptic plasma membranes employing 1,6-diphenyl-1,3,5-hexatriene as fluorescent probe. (1) The monolayer experiments demonstrated an interaction with gangliosides GT1, GM1, dioleoylphosphatidic acid and phosphatidylserine, but little or no interaction with phosphatidylcholine or sphingomyelin. The interaction with monolayers of GT1 or phosphatidic acid decreased for ACTH1-13-NH2 and ACTH1-10. (2) The liposome experiments showed that 2·10?5 M ACTH1–24 increased the glycerol permeability by 20% and decreased the activation energy only when liposomes derived from light synaptic plasma membranes were used. Treatment of the liposomes with neuraminidase abolished the ACTH-induced permeability increase. (3) Steady-state fluorescence depolarization measurements revealed that ACTH1–24, ACTH1-16-NH2 and ACTH1–10 did not change the fluidity of liposomes derived from light synaptic plasma membranes as sensed by diphenylhexatriene. It is concluded that ACTH1–24 can bind to negatively charged lipids and can form an amphipathic helix aligned parallel to the membrane surface involving the N-terminal residues 1 to 12, possibly to 16. Polysialogangliosides will favorably meet the condition of a high local surface charge density under physiological circumstances. It is suggested that ACTH-ganglioside interactions will participate in ACTH-receptor interactions.  相似文献

After incubations autoradiography was made. In comparison, "conventional" immunostaining was carried out on adjacent sections and parallel cultures. The morphology of nucleated cells in the peripheral blood of rats after single dose irradiation with a dose of 5.5 Gy was followed during 28 days after irradiation. During profound agranulocytopenia and granulocytopenia the number of lymphocyte-like mononuclear cells was increased from the days 7-10 after irradiation and the number of monocyte-like mononuclear cells increased from day 14. The cell population discussed in the paper differed markedly from typical lymphocytes and monocytes in particular cytomorphologic parameters.  相似文献

The melanocortin-2 (MC2) receptor is a G protein-coupled receptor that mediates responses to ACTH. The MC2 receptor acts in concert with the MC2 receptor accessory protein (MRAP) that is absolutely required for ACTH binding and signaling. MRAP has a single transmembrane domain and forms a highly unusual antiparallel homodimer that is stably associated with MC2 receptors at the plasma membrane. Despite the physiological importance of the interaction between the MC2 receptor and MRAP, there is little understanding of how the accessory protein works. The dual topology of MRAP has made it impossible to determine whether highly conserved and necessary regions of MRAP are required on the intracellular or extracellular face of the plasma membrane. The strategy used here was to fix the orientation of two antiparallel MRAP molecules and then introduce inactivating mutations on one side of the membrane or the other. This was achieved by engineering proteins containing tandem copies of MRAP fused to the amino terminus of the MC2 receptor. The data firmly establish that only the extracellular amino terminus (Nout) copy of MRAP, oriented with critical segments on the extracellular side of the membrane, is essential. The transmembrane domain of MRAP is also required in only the Nout orientation. Finally, activity of MRAP-MRAP-MC2-receptor fusion proteins with inactivating mutations in either MRAP or the receptor was rescued by co-expression of free wild-type MRAP or free wild-type receptor. These results show that the basic MRAP-MRAP-receptor signaling unit forms higher order complexes and that these multimers signal.  相似文献

Previous studies showed that a 6-week pretreatment of rats with testosterone plus estradiol enhanced the recovery of spermatogenesis 9 weeks after gamma irradiation, resulting in a dose-modifying factor (DMF) of about 2. To test whether the effect of the hormone treatment was mediated through changes in oxygen tension, thiol levels or DNA repair, we irradiated the testes of rats with neutrons, which depend less on these factors than does low-LET radiation for their cytotoxic action. Control rats and rats treated with testosterone plus estradiol were irradiated with 0.7-2.7 Gy of cyclotron-generated high-energy neutrons. The recovery of spermatogenesis was assessed 9 weeks after irradiation by testis weights, sperm counts and the tubule repopulation indices. Greater recovery of spermatogenesis was observed for all end points, with a DMF of about 2 for rats treated with testosterone plus estradiol compared to the irradiated, cholesterol-treated rats. The equal protection factors for neutrons and gamma rays indicate that oxygen, thiols and repair of DNA damage are unlikely to be involved in the protective effect of the hormone treatment.  相似文献

Twenty-four hours after isolation of the pituitary by surgical removal of the medial hypothalamus, i.e. in rats with pituitary island, E. coli endotoxin significantly increased the plasma corticosterone level. Atrophy of the neural lobe, due to pituitary stalk section performed one month prior to removal of the medial hypothalamus, did not prevent the increase of ACTH release by E. coli endotoxin. E. coli endotoxin-induced ACTH release in MBH-deprived animals does not appear to be a function of mechanisms operating only in the innervated lobe.  相似文献

Adult male Wistar rats were exposed for 2 h a day, 7 days a week for up to 30 days to continuous 2450 MHz radiofrequency microwave (rf/MW) radiation at a power density of 5–10 mW/cm2. Sham-exposed rats were used as controls. After ether anesthesia, experimental animals were euthanized on the final irradiation day for each treated group. Peripheral blood smears were examined for the extent of genotoxicity, as indicated by the presence of micronuclei in polychromatic erythrocytes (PCEs). The results for the time-course of PCEs indicated significant differences (P<0.05) for the 2nd, the 8th and the 15th day between control and treated subgroups of animals. Increased influx of immature erythrocytes into the peripheral circulation at the beginning of the experiment revealed that the proliferation and maturation of nucleated erythropoietic cells were affected by exposure to the 2450 MHz radiofrequency radiation. Such findings are indicators of radiation effects on bone-marrow erythropoiesis and their subsequent effects in circulating red cells. The incidence of micronuclei/1000 PCEs in peripheral blood was significantly increased (P<0.05) in the subgroup exposed to rf/MW radiation after eight irradiation treatments of 2 h each in comparison with the sham-exposed control group. It is likely that an adaptive mechanism, both in erythrocytopoiesis and genotoxicity appeared in the rat experimental model during the subchronic irradiation treatment.  相似文献
