用荧光染色法观察了合成的大鼠降钙素基因相关肽(CGRP)对大鼠心肌细胞内游离Ca ̄(2+)含量的影响。结果证明,CGRP能明显增加心肌细胞内Ca ̄(2+)含量,小、中和大剂量(10 ̄(-9)、10 ̄(-8)、10 ̄(-7)mol/L)的CGRP使Ca ̄(2+)含量分别增加至276.88±6.31、364.997±12.70、576.397±15nmol/L与对照组(136.28±7.24nmol/L)相比差异非常显著(P<0.01),且随着CGRP剂量的增加而作用明显加强,呈现剂量—效应关系。30μmol/L的维拉帕米对CGRP所致的细胞内Ca ̄(2+)增加有抑制作用,对小、中、大剂量CGRP作用的抑制率分别为48%、44%和18%。我们推测,CGRP可能直接作用于心肌细胞。低浓度CGRP的正性肌力作用主要是促进Ca ̄(2+)经Ca ̄(2+)通道内流,使心肌细胞内Ca ̄(2+)含量增加的结果。在大剂量CGRP的正性肌力作用中Ca ̄(2+)内流也起到一定作用。  相似文献

本实验观察了冠脉内注射降钙素基因相关肽(CGRP0.3μg/kg)对正常及不同程度冠脉狭窄犬的心功能的影响。结果表明正常犬冠脉内注射CGRP后,平均动脉压(MAP)下降1.2kPa(P<0.05),同时,心率(HR)、心输出量(CO)、左室收缩压峰值(LVSP)均不同程度增加;左室舒张末压(LVEDP)轻度降低。在中度狭窄30min后,冠脉内注射CGRP对HR、MAP无明显影响;而重度狭窄后注射CGRP,MAP由狭窄时降低逐渐增高,HR由增快而变慢。CO、LVSP均显著增高,LVEDP降低,此作用较冠脉狭窄前更为明显。提示CGRP扩张冠脉动脉,增加冠脉血流量和心排血量,增强心肌收缩力,对缺血心脏功能有保护作用。  相似文献

在肠缺血/再灌注(I/R)损伤的大鼠模型中,用放射免疫法分析血管和离体肺灌洗液中降钙素基因相关肽(CGRP)。结果发现I/R损伤后血浆CGRP水平较自身伤前增高161.3%(P〈0.01),伤前预先用血小板激活因子受阻断剂(SR2741A),血浆CGRP仍然高于自身伤前117.8%(P〈0.01)。而假手术组没有明显变化。另外,I/R损伤组大鼠离体肺灌洗液同正常对照相比,CGRP还下降了17.4%  相似文献

本工作在麻醉大鼠冠状动脉口注射内皮素1(ET1)900pmol/kg能引起室性早搏(PVC)、室速(VT)、室颤(VF)等严重心律失常,心律失常评分(AS)为5.6±1.0;冠状动脉口单独注射降钙素基因相关肽(CGRP)300-1200pmol/kg仅引起血压一过性下降,此后逐渐恢复,无心律失常发生,心律失常评分为0。用CGRP300pmol/kg预处理后再注射ET1900pmol/kg,心律失常发生率减少,严重程度降低,AS为1.6±1.6。CGRP1200pmol/kg+ET1组心律失常评分显著低于ET1对照组(P<0.01)。本实验结果表明,CGRP的抗心律失常作用很可能有部分是通过拮抗内皮素的致心律失常作用来实现的。  相似文献

应用免疫组化技术亲和组化法和ABC法,检测了106例乳腺癌组织中ER、PR和CEA水平。其阳性率依次为83%、81.1%和88.7%。其中79.2%的乳腺癌ER和PR表达一致。在98例ER和/或PR阳性乳腺癌中有92例(93.9%)呈CEA阳性,8例ER和PR阴性乳腺癌中6例(75%)为CEA阴性,89%乳腺癌CEA与ER和PR表达一致。在癌的分级表达中,随着癌的组织学分级增高CEA阳性率增高,而ER和PR阳性率减低。结果表明,乳腺癌ER、PR和ChA表达可反映肿瘤的不同生物学特征。同时检测三者对选择内分泌治疗及判断预后更有意义。  相似文献

为探讨正常大鼠与吞噬功能缺陷大鼠视网膜色素上皮(RPE)和视杆细胞外节(ROS)膜表面的甘露糖受体的存在和分布,本实验用胶体金标记物对12只Wistar大鼠和12只RCS大鼠的RPE细胞及临近其顶部的ROS表面的Con-A受体进行标记.结果表明。两种大鼠RPE细胞及ROS膜表面均有胶体金标记物.胶体金标记物在两种RPE细胞微绒毛上分布密度较低,而在两种大鼠ROS膜表面分布密度则高。说明两种大鼠RPE细胞和ROS膜表面均有Con-A受体分布。在两种大鼠ROS被RPE细胞吞入后,均无胶体金标记物。  相似文献

研究发现在有或者没有磷酸盐的条件下,葡萄糖均抑制昆明白小鼠l-4细胞期胚胎的体外发 育。在不含葡萄糖和磷酸盐的HECM-l中,桑椹率为40.05%(74/168),而对照G-HECM-1仅为 8.14%(7/86);不含葡萄糖含有磷酸盐的CZB中,桑椹率为67.11%(93/152),而对照TALP仅 6· 67%(6/90)。用不含葡萄糖而含有1. 0mmo1/L谷氨酸肢和0. 11mmol/L EDTA的CZB液,与兔输 卵管上皮单层培养细胞(ROEC)协同培养小鼠1细胞胚,73.33%(110/150)胚胎发育至桑椹胚, 但没有观察到囊胚形成、用上述CZHROEC系统培养小鼠1细胞胚48小时(3-4细胞),再移入 TCM199+10%FCS+ROEC系统,有76.74%(67/86)胚胎发育至桑椹胚,96/小时后,40.70% (35/86)发育至囊胚。  相似文献

采用自制的壳聚糖为载体对单宁酶(TA)固定化,TA与壳聚糖配比1:2.5,30℃固定2h,活力回收达23.6%~33.1%;偶联效率为84.9%~88.0%。固定化单宁酶(ITA)的表观Km值(以没食子酸丙酯为底物)为22.2×10-6mol/L,TA的Km值(以没食子酸丙酯为底物)为10×10-6mol/L,TA和ITA的最适反应温度分别为40℃和50℃;60℃处理15min,残存活性分别为13.6%和60.3%。TA和ITA的最适pH值分别为5.8和6.4;TA在pH4.8~7.8活力稳定,而ITA活力稳定范围在pH4.8~6.8.ITA作用于EGCG的半衰期为78.7h,EGCG水解率达90.3%。对茶多酚提取物进行水解,其所含的酯型儿茶素EGCG和ECG水解率分别为96.4%和96.8%,非酯型儿茶素EGC和EC的含量显著增加。  相似文献

应用真彩色医学图像分析技术,对51例角结膜缘上皮良、恶性病变(其中上皮增生19例,不典型增生6例,原位癌14例,鳞状细胞癌12例)进行了DNA原位定量分析研究.结果显示:原位癌和鳞状细胞癌DNA含量明显增加,多为高倍异倍体细胞,DNA直方圆明显右移,峰值主要位于>5C处,≥5C细胞分别占78.89%和78.31%;上度增生病变DNA含量较低,多为低倍整倍体细胞,DNA直方图峰值位于2C~4C处,2C~4C细胞占88.59%;上皮不典型增生病变DNA含量介于良性病变和癌之间,DNA直方图逐渐右移,2C~4C细胞占58.62%,≥5C细胞占41.38%。以上数据经统计学处理各组间有显著性差异。表明DNA倍性程度与肿瘤的增殖程度呈正相关,高倍异倍体细胞随肿瘤恶性程度的增高而增多。作者认为DNA原位图像定量分析可为角结膜缘上皮良、恶性病变的诊断、分级及早期发现癌变趋势提供一个可靠的参考指标。  相似文献

观察了hFPIL6/2对6.5Gyγ线照射NIH小鼠第10天造血功能恢复的影响。结果表明:照射小鼠连续4d给予hFPIL6/2250μg·kg-1·d-1,其脾重、CFU-8、骨髓有核细胞数及CM-CFU分别比对照组增加59.0%、278.5%、57.9%和138.2%,统计学处理均有显著差异;对此四项指标的改善也明显优于25μg组。另外,250μg剂量组小鼠外周血象30d的动态观察结果表明,hFPIL6/2不但能明显提高红细胞和血红蛋白的最低值,而且能使血小板的恢复提前。提示hFPIL6/2在促进血小板生成和促进红系造血方面可能具有良好的应用前景。  相似文献

Purpose

Epidemiologic findings are inconsistent concerning the associations between C-reactive protein (CRP), interleukin 6 (IL-6) and lung cancer risk. We conducted a meta-analysis of epidemiologic studies to examine these associations.

Methods

A systematic literature search up to October 2011 was performed in MEDLINE and EMBASE. Study-specific risk estimates were pooled using a random-effects model.

Results

The 10 studies on CRP involved a total of 1918 lung cancer cases. The pooled RR of lung cancer for one unit change in natural logarithm (ln) CRP was 1.28 (95% CI 1.17–1.41). There was no statistically significant heterogeneity among studies (P = 0.116; I2 = 36.6%). We also found that CRP was significantly associated with increased risk of lung cancer among men (RR 1.18, 95% CI 1.09–1.28) but not among women. The 5 studies on IL-6 involved a total of 924 lung cancer cases. The pooled RR of lung cancer for one unit change in ln IL-6 was 1.28 (95% CI 0.92–1.79), however, statistically significant heterogeneity was found. After excluding the study contributing most to the heterogeneity, the summary estimate was essentially unchanged.

Conclusion

CRP was associated with increased risk of lung cancer, especially among men. There was no significant association between IL-6 and lung cancer risk.  相似文献

The correlation between rs1205, rs2808630 variants of C-reactive protein (CRP) gene and susceptibility of cancer has been assessed previously, but with conflicting results. We adopted odds ratios (ORs) with 95% confidence intervals (CIs), in silico tools and enzyme-linked immunosorbent assay (ELISA) analysis to evaluate this association. Totally, 10,614 cancer subjects and 33,294 controls were involved in the pooled analysis. When all the studies were pooled, no significant correlation was indicated between the two variants and cancer risk. However, in stratification analysis by ethnicity, we found that CRP rs1205 C>T polymorphism was associated with an elevated risk of cancer in Asians (T-allele vs. C-allele, OR = 1.20, 95% CI = 1.06–1.36, pheterogeneity = .226; TT vs. CC, OR = 1.48, 95% CI = 1.14–1.93, pheterogeneity = .089). Similar findings were observed for rs2808630 variant. In silico tools showed that lung adenocarcinoma participants with high CRP expression may have shorter overall survival time than low expression group. ELISA analysis indicated that CRP expression in prostate adenocarcinoma subjects with TT + TC genotypes was statistically higher than in those with CC genotypes. CRP rs1205 C>T and rs2808630 T>C polymorphism may be associated with cancer risk, especially for Asians.  相似文献

The objective of this study was to measure the serum concentrations of C-reactive protein (CRP) in cancer patients and compare with those of immune disease patients and healthy individuals for discriminatory analysis. For this purpose, automatic systems for special protein analysis (Type: Drcon Diognostica Tarbox) was used to measure serum CRP concentrations in 276 cancer patients (Group A), 110 immune disease patients (Group B), 161 phlogistic patients (Group C), and 125 age-matched healthy individuals (Group D). Our data show that serum CRP concentrations in Group A were significantly higher than those in Groups B and D, whereas CRP concentrations in Group B were higher than those in Group D. The differences of serum CRP concentrations between Groups A and B as well as between Groups B and D were significant (P?<?0.01). We, therefore, concluded that the measurement of serum CRP concentrations was a fast and accurate method to distinguish between cancer and immune disease patients.  相似文献

摘要目的:4g讨C反应蛋白(CRr')在结直肠癌患者中的表达及与预后的关系。方法:采用免疫速率散射比浊法检测血清CRP,并与健康对照组进行比较,分析CRP与临床病理分期之间的关系,评价术前、术后CRP表达与预后的关系。结果:研究组术前CRP为(28.64±7.15)mg/L,术后降低为(7.83±1.03)mg/L,差异有统计学意义(P〈0.05),都高于对照组的(1.13±0.28)mg/L,差异有统计学意义(P〈0.05)。CRP表达与年龄和性别中的表达差异无统计学意义(P〉0.05),与T、N、M分期和分化程度有关(P〈0.05)。术前高、低CRP组的5年生存率分别为79.17%和95.24%,差异有统计学意义(P〈0.05);术后高、低CRP组的5年生存率分别为85.00%和88.00%,差异无统计学意义(P〉0.05)。结论:结直肠癌与炎症反应密切相关,CRP在其发生、发展中发挥着重要作用,术前CRP水平对疾病的预后具有一定的预测价值。  相似文献

摘要 目的:探讨色素内镜联合C反应蛋白(C-reactive protein,CRP)和转铁蛋白(Transferrin,TRF)在青海地区早期食管癌患者的诊断价值。方法:2018年6月至2020年3月选择在本院进行食管癌筛查的青海地区患者250例,所有患者都给予色素内镜检查,同时给予血浆CRP和TRF检查并判断诊断价值。结果:在250例患者中,病理诊断为原位癌28例,高级别上皮内瘤变10例,低级别上皮内瘤变12例,慢性炎症200例;其中早期食管癌38例。卢戈液色素内镜显示不染色47例,淡染色95例,浓染色108例。早期食管癌患者的血浆CRP值都高于非早期食管癌患者,TRF值低于(P<0.05)。Pearson相关性分析显示早期食管癌与色素内镜卢戈液不染色、CRP、TRF值存在相关性(P<0.05)。色素内镜联合CRP和TRF在青海地区早期食管癌的诊断敏感性与特异性分别为97.4 %和100.0 %。结论:色素内镜联合CRP和TRF在青海地区早期食管癌患者诊断中的应用具有很好的敏感性与特异性,能有效预测早期食管癌发生情况,对早期诊治早期食管癌具有一定的指导意义。  相似文献

C-Reactive protein (CRP) is a well-known inflammatory marker, and elevated CRP levels has been reported to be associated with the risk of various cancers. To date, no study has investigated the association between elevated CRP and incidents of cancer among Mexican Americans. In the current prospective cohort study, we measured pre-diagnostic CRP levels in serum samples and evaluated their relationships with demographic characteristics and health behaviors associated with cancer risk among 2753 Mexican Americans selected from the Mano-A-Mano Mexican American Cohort Study. At baseline, median levels of serum CRP significantly differed by demographics (sex, age category, marital status, and education levels) and health behaviors (cigarette smoking status, alcohol drinking status, BMI category, and physical activity levels). In the multivariable analysis, the study participants who were women, older, never drinking alcohol, overweight or obese, and physically inactive had increased likelihood of having high CRP levels (≥ median levels among all study participants) compared to their counterparts. A total of 177 cancer cases were identified during the follow-up with a median follow-up time of 127 months. In the quartile analysis, study participants in the 4th quartile with highest CRP levels had significantly 1.88 fold increased risk of cancer (hazard ratio (HR) = 1.88, 95%CI: 1.12, 3.13) compared to those in the 1st quartile with lowest CRP levels. The association was further confirmed in analyses using clinical CRP levels. In summary, our findings suggested that serum CRP levels have potential to serve as a predictive marker of cancer risk in Mexican Americans.  相似文献

Emerging evidence suggests that there is interplay between the frequency and circadian timing of eating and metabolic health. We examined the associations of eating frequency and timing with metabolic and inflammatory biomarkers putatively associated with breast cancer risk in women participating in the National Health and Nutrition Examination 2009–2010 Survey. Eating frequency and timing variables were calculated from 24-hour food records and included (1) proportion of calories consumed in the evening (5pm-midnight), (2) number of eating episodes per day, and (3) nighttime fasting duration. Linear regression models examined each eating frequency and timing exposure variable with C-reactive protein (CRP) concentrations and the Homeostatic Model Assessment of Insulin Resistance (HOMA-IR). Each 10 percent increase in the proportion of calories consumed in the evening was associated with a 3 percent increase in CRP. Conversely, eating one additional meal or snack per day was associated with an 8 percent reduction in CRP. There was a significant interaction between proportion of calories consumed in the evening and fasting duration with CRP (p = 0.02). A longer nighttime fasting duration was associated with an 8 percent lower CRP only among women who ate less than 30% of their total daily calories in the evening (p = 0.01). None of the eating frequency and timing variables were significantly associated with HOMA-IR. These findings suggest that eating more frequently, reducing evening energy intake, and fasting for longer nightly intervals may lower systemic inflammation and subsequently reduce breast cancer risk. Randomized trials are needed to validate these associations.  相似文献

The importance of inflammation is increasingly noticed in cancer. The aim of this study was to analyze the prognostic influence of pre-operative serum C-reactive protein (CRP) in a cohort of 148 lymph node-negative breast cancer patients. The prognostic significance of CRP level for disease-free survival (DFS), metastasis-free survival (MFS) and overall survival (OS) was evaluated using univariate and multivariate Cox regression, also including information on age at diagnosis, tumor size, tumor grade, estrogen receptor (ER), progesterone receptor (PR) and human epidermal growth factor receptor 2 (HER2) status, proliferation index (Ki67) and molecular subtype, as well as an assessment of the presence of necrosis and inflammation in the tumor tissue. Univariate analysis showed that CRP, as a continuous variable, was significantly associated with DFS (P = 0.002, hazard ratio [HR]  = 1.04, 95% confidence interval [CI]  = 1.02–1.07) and OS (P = 0.036, HR  = 1.03, 95% CI  = 1.00–1.06), whereas a trend was observed for MFS (P = 0.111). In the multivariate analysis, CRP retained its significance for DFS (P = 0.033, HR  = 1.01, 95% CI  = 1.00–1.07) as well as OS (P = 0.023, HR  = 1.03, 95% CI  = 1.00–1.06), independent of established prognostic factors. Furthermore, large-scale gene expression analysis by Affymetrix HG-U133A arrays was performed for 72 (48.6%) patients. The correlations between serum CRP and gene expression levels in the corresponding carcinoma of the breast were assessed using Spearman''s rank correlation, controlled for false-discovery rate. No significant correlation was observed between CRP level and gene expression indicative of an ongoing local inflammatory process. In summary, pre-operatively elevated CRP levels at the time of diagnosis were associated with shorter DFS and OS independent of established prognostic factors in node-negative breast cancer, supporting a possible link between inflammation and prognosis in breast cancer.  相似文献

目的:探究血清降钙素原(PCT)、C-反应蛋白(CRP)与可溶性人髓系细胞触发受体-1(sTREM-1)在肺癌患者术后肺部感染中表达及其诊断价值。方法:选择2016年2月至2019年10月期间在我院行肺癌根治术的420例肺癌患者作为研究对象,根据术后患者是否发生肺部感染进一步划分为380例未感染组和40例感染组。感染组根据治疗结局进一步划分为29例治疗好转亚组与11例未好转亚组。采用酶联免疫吸附法检测各组的血清PCT、CRP与s TREM-1水平,采用受试者工作特征(ROC)曲线分析血清PCT、CRP和s TREM-1对肺癌患者术后肺部感染的预测价值。结果:与未感染组相比,感染组手术后血清PCT、CRP和s TREM-1水平均明显升高(P0.05)。与治疗好转亚组相比较,治疗未好转亚组手术后以及感染后血清PCT、CRP和s TREM-1水平均明显升高(P0.05)。ROC曲线显示,PCT的曲线下面积(AUC)为0.713,最佳截断值为1.23 ng/mL,灵敏度、特异度分别为0.81、0.79,准确度为0.82;CRP的AUC为0.752,最佳截断值为36.07 mg/L,灵敏度、特异度分别为0.83、0.81,准确度为0.83;s TREM-1的AUC为0.792,最佳截断值为20.58 pg/mL,灵敏度、特异度分别为0.86、0.84,准确度为0.85;PCT、CRP联合s TREM-1预测肺癌患者术后肺部感染的AUC为0.884,灵敏度、特异度分别为0.89、0.91,准确度为0.92。结论:肺癌根治术后肺部感染发生与患者血清PCT、CRP和s TREM-1水平相关,早期联合检测血清PCT、CRP和s TREM-1有助于预测肺癌根治术患者肺部感染发生风险,在肺癌根治术后肺部感染的预测和诊断中具有一定临床价值。  相似文献

Background

Inflammation-based prognostic scores such as the neutrophil lymphocyte ratio (NLR), platelet lymphocyte ratio (PLR), Glasgow prognostic score (GPS), and modified GPS (mGPS) have been reported to have prognostic value in patients with many types of cancer, including esophageal squamous cell carcinoma (ESCC). However, the role of the C-reactive protein/Albumin (CRP/Alb) ratio in ESCC has not yet been evaluated.

Methods

A total of 468 patients suffering from histologically proven ESCC were enrolled between January 2000 and July 2010. The GPS, mGPS, NLR, PLR and CRP/Alb ratios were tested together with established prognostic factors in univariate and multivariate Cox regression analyses of overall survival (OS).

Results

The optimal cutoff level for the CRP/Alb ratio was 0.50. The CRP/Alb ratio (continuous) had higher AUC values at 12 months (0.796), 24 months (0.805), and 36 months (0.815) than the NLR, GPS and mGPS. In univariate analysis, the 5-year OS rate for patients with a CRP/Alb ratio > 0.50 was 43.4%, while the rate for patients with a CRP/Alb ratio ≤ 0.50 was 17.7% (P < 0.0001). In multivariate analysis, patients with a CRP/Alb ratio > 0.50 had worse survival than patients with a CRP/Alb ratio ≤ 0.50 (HR: 2.44; 95% CI: 1.82–3.26; P < 0.0001).

Conclusion

In summary, to the best of our knowledge, this is the first study to identify the CRP/Alb ratio as a novel inflammation-based prognostic factor in a large group of ESCC patients. The prognostic value of the CRP/Alb ratio needs to be verified in prospective multicenter studies.  相似文献
