Summary Eight thermophilic fungi were tested for production of mannanases and galactanases. Highest mannanase activities were produced byTalaromyces byssochlamydoides andTalaromyces emersonii. Mannanases from all strains tested were induced by locust bean gum except in the case ofThermoascus aurantiacus, where mannose had a greater inducing effect. Locust bean gum was also the best inducer of -mannosidase and galactanase except in the case ofT. emersonii where galactose was a better inducer of both these enzymes. Highest mannanase activity was produced byTalaromyces species when peptone was used as nitrogen source whereas sodium nitrate promoted maximum production of this enzyme byThielavia terrestris andT. aurantiacus. The pH optima of mannanases from the thermophilic fungi were in the range 5.0–6.6 and contrasted with the low pH optimum (3.2) of the enzyme fromAspergillus niger. Galactanases had pH optima in the range 4.3–5.8. The mannanase fromT. emersonii and the galactanase fromT. terrestris were most thermostable, each retaining 100% activity for 3 h at 60°C.  相似文献

Summary The yeast-like fungusAureobasidium is a promising source of xylanase (EC 3.2.1.8) with an exceptionally high specific activity. For enzyme production in volumes of several liters, xylose was the preferred carbon source and inducer. Xylanase in clarified cultures was concentrated by reversible adsorption to cation-exchange matrix to 5% of the initial volume, and recovered at nearly 2 million IU/1. Selective conditions permitted 97% recovery of xylanase with a 1.8-fold enrichment in specific activity, to 70% of purity. The predominant xylanase species (20 kDa) was subsequently purified to >99% of homogeneity by gel filtration chromatography. Purified enzyme exhibited an isoelectric point of 8.5, and specific activity of 2100 IU/mg under optimal conditions, determined to be pH 4.5 and 45°C. Summary Extracellular mannanase activity produced bySporotrichum cellulophilum was purified into two components using acetone precipitation, SP-Sephadex C50 ion exchange chromatography and preparative polyacrylamide gel electrophoresis. The purified mannanse components, M1 and M2, had molecular weights of 108 000–112 000 and 32 200–36 000 respectively. Component M1 was shown to contain 2 subunits having molecular weights of 62 000 and 50 000. M1 and M2 had similar pH-activity profiles with pH optima of 5.5 and 6.0 respectively. M1 was more thermostable than M2: half lives of the enzymes at 70°C were 30 and 9 min for M1 and M2 respectively.  相似文献

An extracellular, debranching isoamylase fromHendersonula toruloidea ATCC 64930, grown on starch, was purified 12-fold to an electrophoretically homogeneous state. The purified enzyme (estimated mol wt 83000) was optimally active at pH 6.0 and 50°C and remained active when held at 70°C (30 min) and at pH 6 to 8 for 24 h. Na+, Fe2+ and Ba2+ (at 5mm) enhanced enzyme activity while Hg2+, Zn2+ and Cu2+ (at 5mm) were inhibitory. The enzyme hydrolysed amylopectin (Km, 0.25 mg/ml), forming maltose, maltotriose and maltotetraose and hydrolyzed glycogen (Km, 0.29 mg/ml) and soluble starch (Km, 0.42 mg/ml) forming maltotriose and maltotetraose. A novel chymotrypsin inhibitor of the potato I protease inhibitor family from the earthworm Lumbricus terrestris was purified. The inhibitor, named LTCI, was isolated by methanol extraction, affinity chromatography on immobilized methylchymotrypsin, and ion exchange chromatography followed by RP–HPLC. The 7076 Da inhibitor consists of a single polypeptide chain of 64-amino-acid residues without disulfide bridges. LTCI is the first of the potato I protease inhibitors with Tyr in position P1 of the reactive site. cDNA analysis revealed that LTCI is produced as a 86-amino-acid precursor with a 22-amino-acid secretory signal peptide. RT–PCR analysis demonstrates that LTCI mRNA is expressed in body wall, intestine, and coelomocytes. The recombinant LTCI was produced in Escherichia coli as a fusion protein with intein and chitin binding domain using IMPACT™–CN system.  相似文献

An endopolygalacturonase of Rhizopus sp. strain LKN, one of several isolates from tempe starter (ragi), was purified 235-fold by CM-Sephadex C-50, DEAE-Sephadex A-50 ion exchange chromatographies and Sephadex G-75 gel filtration. The purified enzyme was homogeneous by SDS-PAGE with a M r of 38.5 kDa. Its K m value for pectic acid was 2 mg/ml. It was stable at pH 4.5 to 11 and up to 50°C, with optimum activity at pH 4.5 to 4.75 and 55 to 60°C. Summary The larval drift of the fire salamander was investigated over a period of three years in a mountain brook (Niederbergisches Land, F.R. Germany), as well in a laboratory water channel. The rate of larval drift fluctuated between 19% and 41% of the total population of larvae in a defined section of the brook during these three years. Most (83%) of the drifting larvae were hatchlings or very young stages. The drift was dependent on the strength of the current, the number of spawning females, the presence of suitable hiding places, sufficient space and adequate food. Hungry larvae drifted more often than satiated animals. The drift behaviour of hatchlings differed distinctly from that of older larvae. The significance of ecological factors on larval drift is discussed. It is evidently a more important factor in selection than has hitherto been recognized.  相似文献

Summary The zone of endosperm breakdown in the germinated date seed (Phoenix dactylifera L.) is a narrow area immediately adjacent to the surface of the enlarging cotyledon, or haustorium. The zone width is correlated with the amount of cell division in the adjacent region of the haustorium. The sequence of endosperm breakdown is: 1. protein bodies vacuolate, 2. storage cell walls become electron-transparent immediately adjacent to the protoplast of each endosperm cell, 3. all remaining cytoplasm and lipid bodies disappear, and 4. the remaining cell walls become electron-transparent and collapse against the haustorium surface. Two cell wall hydrolases are present—endo-mannanase (EC3.2.1.78) and -mannosidase (EC3.2.1.25). -mannosidase is detectable in the endosperm before germination. At germination, the major portion of activity is found in the softened endosperm. -mannanase is only detectable from germination and there is always hundreds of fold greater activity in the softened endosperm than elsewhere. Proteinase is detectable in trace amounts at germination in the softened endosperm but is also found in the haustorium at later stages. Isolated haustoria, incubated in extracted ivory nut (Phytelephas macrocarpa) mannan in buffer, cause no mannan breakdown. Haustoria, incubated in a solution of locust bean galactomannan, cause no decrease in galactomannan viscosity. Our observations suggest that although haustoria probably regulate mannan breakdown in the endosperm, they do not seem to secrete the hydrolytic enzymes concerned.  相似文献

Summary InB. terrestris diploid males develop normally into adults (Duchateau et al., 1994). The diploid males are similar in appearance to the haploid males, except that they are smaller. The size of the testis of diploid males, relative to the length of the radial cell, is smaller than that of haploid males. There is overlap in the frequency distribution with respect to body size and testis size. The spermatozoa of diploid males are larger than those of the haploids and the vasa deferentia contain fair less spermatozoa than those of haploid males of the same age. Countings and measurements of the spermatozoa, therefore, can give the best indication about the ploidy of the males. Diploid males are successful in mating. They mate at a younger age than haploid males and they die sooner. The number of vial offspring of diploid males, however, is very low. No queen that mated with a diploid male produced a colony, but a few queens did produce some progeny. These might have been triploid males and workers. InB. terrestris higher ploidy results in smaller individuals, whereas in several other species of the Hymenoptera it has been found to result in larger individuals.  相似文献

The effects of neurotensin (NT) and cholecystokinin (CCK) were studied on isolated crop-gizzard preparations of Lumbricus terrestris suspended in a smooth muscle organ bath. Changes in the amplitude and frequency of contractions associated with spontaneous motility were observed in response to neurotransmitters known to have an excitatory effect (acetylcholine) and an inhibitory effect (serotonin); and to the hormones NT and CCK, which in vertebrate models have both been shown to inhibit gastric motility. The overall contractile amplitude and frequency of crop-gizzard contractions were decreased in response to increasing concentrations of NT and CCK. In general, both hormone-induced responses were similar when compared at equal molar concentrations. Cholecystokinin, however, did exhibit a greater reduction in contractile frequency than NT. It is speculated that possible desensitization of earthworm NT-receptors to higher hormone concentrations resulted in a depressed maximal response in the concentration–response curve. Despite that possibility, the overall hormonal inhibition was statistically significant. These results infer that NT- and CCK-induced inhibition of crop-gizzard motility may have a modulatory role in the transport of nutrients and overall efficiency of worm metabolism.  相似文献

Ayurvedic medicine, which uses decoctions made of medicinal plants, is used to cure diseases in many Asian countries including Sri Lanka. Although proper storage facilities for medicinal plants are unavailable in Sri Lanka, neither the potential for growth of toxigenic fungi nor their ability to produce mycotoxins in stored medicinal plants has been investigated. We isolated three Fusarium species, F. culmorum, F. acuminatum and F. graminearum from the medicinal plant Tribulus terrestris. Culture extracts of the 3 Fusarium spp. were cytotoxic to mammalian cell lines BHK-21 and HEP-2. Three toxic metabolites produced by Fusarium spp; T-2 toxin, zearalenone, and diacetoxyscirpenol were also cytotoxic to the same mammalian cell lines. The 3 Fusarium spp. grown on rice media produced zearalenone. Plant material destined for medicinal use should be stored under suitable conditions to prevent growth of naturally occurring toxigenic fungi prior to its use.  相似文献

The giant, 3.6-MDa hexagonal bilayer hemoglobin (Hb) of Lumbricus terrestris consist of twelve 213-kDa globin subassemblies, each comprised of three disulfide-bonded trimers and three monomer globin chains, tethered to a central scaffolding of 36–42 linkers L1–L4 (24–32 kDa). It is known to contain 50–80 Ca and 2–4 Cu and Zn; the latter are thought to be responsible for the superoxide dismutase activity of the Hb. Total reflection X-ray fluorescence spectrometry was used to determine the Ca, Cu, and Zn contents of the Hb dissociated at pH 2.2, the globin dodecamer subassembly, and linker subunits L2 and L4. Although the dissociated Hb retained 20 Ca2+ and all the Cu and Zn, the globin subassembly had 0.4 to 3 Ca2+, depending on the method of isolation, and only traces of Cu and Zn. The linkers L2 and L4, isolated by reversed-phase high-pressure liquid chromatography at pH 2.2, had 1 Ca per mole and very little Cu and Zn. Electrospray ionization mass spectrometry of linker L3 at pH 2.2 and at neutral pH demonstrated avid binding of 1 Ca2+ and additional weaker binding of 7 Ca2+ in the presence of added Ca2+. Based on these and previous results which document the heterogeneous nature of the Ca2+-binding sites in Lumbricus Hb, we propose three classes of Ca2+-binding sites with affinities increasing in the following order: (i) a large number of sites (>100) with affinities lower than EDTA associated with linker L3 and dodecamer subassembly, (ii) 30 sites with affinities higher than EDTA occurring within the cysteine-rich domains of linker L3 and dodecamer subassembly, and (iii) 25 very high affinity sites associated with the linker subunits L1, L2, and L4. It is likely that the low-affinity type (i) sites are the ones involved in the effects of 1–100 mM Group IIA cations on Lumbricus Hb structure and function, namely increased stability of its quaternary structure and increased affinity and cooperativity of its oxygen binding.  相似文献

Summary Mycelial inocula of the two ectomycorrhizal fungi,Pisolithus tinctorius (Pt) andThelephora terrestris (Tt) were artificially introduced into fumigated nursery soil before seed sowing. At the end of the first growing season, Pt stimulated the height growth ofPinus densiflora, P. thunbergii, P. rigida, andP. rigida×taeda by 55, 36, 69, and 37%, respectively, compared with control seedlings (with no fumigation and no inoculation). When growth stimulation was expressed by dry weight, Pt increased dry weight ofP. densiflora andP. rigida×taeda by 143% and 128%, respectively, over control seedlings. Total absorption (per plant top) of nitrogen and phosphorus of inoculatedP. densiflora seedlings was also increased by 188 and 144%, respectively, even though the concentration of these two elements in the tissue was not significantly different from that of control seedlings.Tt failed to stimulate growth of the inoculated plants possibly due to its lower level of infection (40–60% mycorrhizal tips) compared with the relatively high infection (over 80%) of Pt.Pinus koraiensis did not respond to inoculation during the first growing season, probably due to its inherent characteristics of producing a small number of short roots and its slow shoot growth during the first few years after germination.This study was supported by the International Foundation for Science in Stockholm, Sweden.  相似文献

Direct regeneration of shoots and roots through juvenile expiants has been achieved inTribulus terrestris. Cotyledonary leaves along with epicotyl segment from young seedlings were cultured on MS medium containing various concentrations of auxin with cytokinin and glutamine. A combination of 0.2 mgL−1 NAA, 0.5 mgL−1 BAP and 50 mgL−1 glutamine induced high frequency of shoot and root differentiation in 10 weeks. The callus also could be induced on the above medium from the cut end of radical segments. Morphogenic response such as per cent shoot and root differentiation was recorded at regular intervals.  相似文献

Summary The localisation and distribution of 10 vertebrate-derived neuropeptides in the earthworm, Lumbricus terrestris, have been determined by an indirect immunofluorescence technique. The peptides are pancreatic polypeptide (PP), peptide tyrosine tyrosine (PYY), neuropeptide Y (NPY), glucagon (C-terminal), vasoactive intestinal polypeptide (VIP), peptide histidine isoleucine (PHI), gastrinreleasing peptide (GRP), calcitonin gene-related peptide (CGRP), neurotensin (NT), and met-enkephalin. For 6 of the peptides — PYY, NPY, PHI, glucagon, GRP and CGRP — this is the first demonstration of their presence in any annelid, and NT has not previously been described in an oligochaete. Cell bodies and nerve fibres immunoreactive to the 10 peptides occur throughout the CNS. In the PNS, epidermal sensory cells displayed immunoreactivities to PP and PYY, and PP-, PYY-, NPY-, PHI- and GRP-like immunoreactivities occurred in nerve fibres supplying the main body muscles. Nerve fibres immunoreactive to PP and PYY are also associated with the innervation of the gut (pharynx, oesophageal glands, and mid and posterior regions of the intestine). No endocrine cells immunoreactive for any of the antisera tested could be identified in the gut epithelium, suggesting that dual location of peptides in the brain and gut epithelium is a phenomenon that occurred at a later stage in evolution. No immunoreactive elements were detected in any of the organs and ducts of the reproductive and excretory systems.  相似文献

Summary A fine structural investigation was performed on receptor cells lying at the base of the epidermis in the earthworm, Lumbricus terrestris. Two types of receptor cells with many similarities, but also with major differences, were discriminated.One receptor is of the microvillar receptor type, that appears to be identical with the photoreceptor cell described earlier by Röhlich et al. (1970). Proximal to the nucleus is a large vacuole (phaosome, Binnenkörper) with many daughter cavities containing a large number of microvilli and several cilia with the 9 × 2 + 0 microtubular pattern. The intracellular cavity has no connection with the surface membrane, in contrast to that in hirudineans (White and Walther, 1969) and pogonophores (Nørrevang, 1974).The other receptor is the ciliated receptor type, that is presently described for the first time. This receptor also has a comparatively large uniform cavity, few microvilli and about 20 cilia with the 9 × 2 + 2 microtubular pattern. The cilia leave the cell in the proximal part through a wide opening, make a turn of 180 °, and proceed toward the epidermal surface. Receptors of a similar type have been described by Golding and Whittle (1975) in the cerebral ganglion of four limicole oligochaete annelids; they presumed that these cells have an osmoreceptor function. The new epidermal receptor type described in the present investigation probably has a chemoreceptor function of hitherto unknown kind.This investigation was supported by the Royal Physiographic Society at Lund, Sweden. Summary The distribution patterns of serotonin-immunoreactive somata in the cerebral and subpharyngeal ganglion, and in the head and tail ganglia of the nerve cord of Lumbricus terrestris are described from whole-mount preparations. A small number of serotonin-immunoreactive neurons occurs in the cerebral ganglion, in contrast to the large population of serotonin-immunoreactive neurons that exists in all parts of the ventral nerve cord. From the arrangement of serotonin-immunoreactive somata in the subpharyngeal ganglion, we suggest that this ganglion arises from the fusion of two primordial ganglia. In head and tail ganglia, the distribution of serotonin-immunoreactive somata resembles that in midbody segments. Summary The effect of workers size frequency distribution on colony development was studied in 12 young colonies ofB. terrestris. By replacing the original workers with workers of determined size, colonies constituting small, large or mixed size nursing workers were created. The nursing workers size frequency distribution did not influence the average size of the newly emerged workers, nor their size frequency distribution. In contrast, the number of emerging workers and number of egg cells constructed by the queen in colonies with large workers were higher than in colonies with small workers. The small number of emerging workers is explained by prolonged duration of larval time in response to sub-optimal feeding in colonies of small workers. The higher number of egg cells constructed by the queens is supposed to be in response to the number of new cocoons available, or to better condition of the brood.  相似文献

Zusammenfassung Im Dehydrations-Experiment tolerieren Regenwürmer Flüssigkeitsverluste bis zu 70% ihres Körpergewichts. Hierbei ergeben sich hohe Anforderungen an die Anpassungsfähigkeit von Nephridien, Darm und Integument, die im Dienste des Wasser-und Elektrolythaushalts stehen. Um die Abhängigkeit dieser Organe von zentralnervösen Einflüssen zu studieren, wurden bei Lumbricus terrestris Ober-, Unterschlundganglion sowie ein Bauchmarkabschnitt zwischen dem 10.–15. Segment hinter dem Clitellum entfernt. In diesen Segmenten wurden 30, 60 und 120 min nach Versuchsbeginn der Schleimgehalt von Epidermiszellen, die Kerngröße von Darmepithel- und Chloragogen-Zellen sowie der innere und äußere Tubulusumfang von Nephridien cytometrisch bestimmt. In gleichen Intervallen wurde im Oberschlundganglion der Nucleinsäure- und Proteingehalt Gomoripositiver Neurone mikrospektrographisch gemessen. Aus dem Vergleich von cytometrischen und mikrospektrographischen Daten geht hervor: 1. Der nephridiale Stäbchenkanal steht am stärksten unter neuroendokrinem Einfluß. 2. Darmepithel- und Chloragogen-Zellen hängen sowohl von zentraler, neuroendokriner als auch von segmentaler, nervöser Kontrolle ab. 3. Die Schleimsekretion von Epidermiszellen wird wahrscheinlich über segmentale Nerven gesteuert; sie zeigt jedoch die ausgeprägteste Autonomie. Somit stehen die an der Regulation des Wasser- und Elektrolythaushalts beteiligten Organe von Lumbricus terrestris unter einer doppelten Kontrolle durch peptiderge sekretorische Neurone des Oberschlundganglions und segmentale Neurone des Bauchmarks.

Summary Earthworms tolerate loss of fluids up to 70% of their total body weight. The adaptive capacity must thus be very high in organs which control the exchange of water and electrolytes in nephridia, the intestine and skin. In order to study the influence of the central nervous system on adaption, the supra- and suboesophageal ganglion and the 10th–15th segments behind the clitellum were removed. In segments deprived of ventral nerve cord the mucous content of epidermal cells, the nuclear area of intestinal epithelial cells and chloragogen cells together with the circumference of nephridial tubules were measured 30, 60 and 120 min after placing Lumbricus terrestris in a standardized dry environment. Protein and nucleic acid contents of Gomori-positive neurons in the supraoesophageal ganglion were measured at these times. Comparison of the results of cytometric and microspectrographic measurements shows that 1) the distal segment of the wide nephridial tube is under neuroendocrine control, 2) intestinal epithelial cells and chloragogen cells are influenced by segmental neurons and central secretory nervous elements, 3) mucous secretion is autonomous but may be controlled by segmental nerve impulses. Therefore, water- and electrolyte exchange in Lumbricus terrestris is regulated by secretory peptidergic neurons of the supraoesophageal ganglion and by nerve cell complexes in the ventral nerve cord.
