Emerging aquatic insects, including mosquitoes, are known to transfer to terrestrial ecosystems specific essential biochemicals, such as polyunsaturated fatty acids (PUFA). We studied fatty acid (FA) composition and contents of dominant mosquito populations (Diptera: Culicidae), that is, Anopheles messeae, Ochlerotatus caspius, Oc. flavescens, Oc. euedes, Oc. subdiversus, Oc. cataphylla, and Aedes cinereus, inhabited a steppe wetland of a temperate climate zone to fill up the gap in their lipid knowledge. The polar lipid and triacylglycerol fractions of larvae and adults were compared. In most studied mosquito species, we first found and identified a number of short‐chain PUFA, for example, prominent 14:2n‐6 and 14:3n‐3, which were not earlier documented in living organisms. These PUFA, although occurred in low levels in adult mosquitoes, can be potentially used as markers of mosquito biomass in terrestrial food webs. We hypothesize that these acids might be synthesized (or retroconverted) by the mosquitoes. Using FA trophic markers accumulated in triacylglycerols, trophic relations of the mosquitoes were accessed. The larval diet comprised green algae, cryptophytes, and dinoflagellates and provided the mosquitoes with essential n‐3 PUFA, linolenic, and eicosapentaenoic acids. As a result, both larvae and adults of the studied mosquitoes had comparatively high content of the essential PUFA. Comparison of FA proportions in polar lipids versus storage lipids shown that during mosquito metamorphosis transfer of essential eicosapentaenoic and arachidonic acids from the reserve in storage lipids of larvae to functional polar lipids in adults occurred.  相似文献

Increasing sea surface temperatures (SST) and blooms of lipid‐poor, filamentous cyanobacteria can change mesozooplankton metabolism and foraging strategies in marine systems. Lipid shortage and imbalanced diet may challenge the build‐up of energy pools of lipids and proteins, and access to essential fatty acids (FAs) and amino acids (AAs) by copepods. The impact of cyanobacterial blooms on individual energy pools was assessed for key species temperate Temora longicornis and boreal Pseudo‐/Paracalanus spp. that dominated field mesozooplankton communities isolated by seasonal stratification in the central Baltic Sea during the hot and the cold summer. We looked at (a) total lipid and protein levels, (b) FA trophic markers and AA composition, and (c) compound‐specific stable carbon isotopes (δ13C) in bulk mesozooplankton and in a subset of parameters in particulate organic matter. Despite lipid‐poor cyanobacterial blooms, the key species were largely able to cover both energy pools, yet a tendency of lipid reduction was observed in surface animals. Omni‐ and carnivory feeding modes, FA trophic makers, and δ13C patterns in essential compounds emphasized that cyanobacterial FAs and AAs have been incorporated into mesozooplankton mainly via feeding on mixo‐ and heterotrophic (dino‐) flagellates and detrital complexes during summer. Foraging for essential highly unsaturated FAs from (dino‐) flagellates may have caused night migration of Pseudo‐/Paracalanus spp. from the deep subhalocline waters into the upper waters. Only in the hot summer (SST>19.0°C) was T. longicornis submerged in the colder subthermocline water (~4°C). Thus, the continuous warming trend and simultaneous feeding can eventually lead to competition on the preferred diet by key copepod species below the thermocline in stratified systems. A comparison of δ13C patterns of essential AAs in surface mesozooplankton across sub‐basins of low and high cyanobacterial biomasses revealed the potential of δ13C‐AA isoscapes for studies of commercial fish feeding trails across the Baltic Sea food webs.  相似文献

SUMMARY 1. Transfer of carbon from freshwater to terrestrial ecosystems can occur through predation on adult aquatic insects, but the significance of this trophic pathway to the energetics of riparian communities is poorly understood. We used stable isotopes of carbon and nitrogen to explore linkages between aquatic insect production and the nutrition of web‐building and free‐living spiders alongside two streams in the North Island of New Zealand. 2. δ13C values for riparian tree leaves (means for each site = ?32.2 and ?30.3‰) were distinct from those of lichens collected from stream channel rocks and instream algae, both of which were similar (?23.4 to ?22.4‰). δ15N values for leaves were similar at both sites (?3.4 and ?2.7‰), but algae were considerably more depleted in δ15N atonesite suggesting significant differences in instream nitrogen sources between the twostreams. 3. Isotope values for potential aquatic prey of spiders indicated that aquatic algal production was their primary carbon source at both sites. Terrestrial invertebrates collected and assumed to be potential prey reflected a range of carbon sources and represented several trophic levels. 4. At one site, δ13C values indicated a primarily algae‐aquatic insect pathway of carbon transfer to both web‐building and free‐living spider guilds. The other site appeared to have a primarily terrestrial carbon pathway for the free‐living spider guild, and a mixed aquatic‐terrestrial pathway for the web‐building guild. 5. Overall, web‐building spiders were estimated to obtain around 61% of their body carbon from aquatic production compared with 55% for free‐living spiders. Our findings suggest that consumption of prey derived from aquatic sources can provide significant nutrition for spiders living along some stream channels. This pathway may represent an important feedback mechanism contributing to the energetics of riparian communities at sites where aquatic insect production is high.  相似文献

A nearly 40-year debate on the origins of carbon supporting animal production in lotic systems has spawned numerous conceptual theories emphasizing the importance of autochthonous carbon, terrestrial carbon, or both (depending on river stage height). Testing theories has been hampered by lack of adequate analytical methods to distinguish in consumer tissue between ultimate autochthonous and allochthonous carbon. Investigators initially relied on assimilation efficiencies of gut contents and later on bulk tissue stable isotope analysis or fatty acid methods. The newest technique in amino acid, compound specific, stable isotope analysis (AA-CSIA), however, enables investigators to link consumers to food sources by tracing essential amino acids from producers to consumers. We used AA-CSIA to evaluate nutrient sources for 5 invertivorous and 6 piscivorous species in 2 hydrogeomorphically contrasting large rivers: the anastomosing Upper Mississippi River (UMR) and the mostly constricted lower Ohio River (LOR). Museum specimens we analyzed isotopically had been collected by other investigators over many decades (UMR: 1900–1969; LOR: 1931–1970). Our results demonstrate that on average algae contributed 58.5% (LOR) to 75.6% (UMR) of fish diets. The next highest estimated contributions of food sources were from C3 terrestrial plants (21.1 and 11.5% for the LOR and UMR, respectively). Moreover, results from 11 individually examined species consistently demonstrated the importance of algae for most fish species in these trophic guilds. Differences among rivers in relative food source availability resulting from contrasting hydrogeomorphic complexity may account for relative proportions of amino acids derived from algae.  相似文献

Pigment contents and proximate compositions are important traits to evaluate the nutritional value of microalgae. The environmental condition and nutrient availability in the culture medium are hypothesised to regulate algal colour, nutritional composition and production. This study aimed to compare the pigment and proximate compositions in Tisochrysis lutea under phototrophic and mixotrophic conditions in an attempt to improve algal product quality. Algae were grown in the phototrophic and mixotrophic conditions at 50 μmol photons m?2 s?1 for 12 h daily, but 50 mM glycerol was added as a carbon source in the mixotrophic culture. The pigment contents in algae significantly increased in the mixotrophic condition, and the contents of chlorophylls a and c and carotenoids increased nearly 60 % compared with those in the phototrophic condition. The contents of saturated and polyunsaturated fatty acids in algae were significantly higher while the content of monounsaturated fatty acids was significantly lower in the mixotrophic condition than those in the phototrophic condition. Although the contents of protein, lipid and carbohydrate in T. lutea were not different, their overall production was significantly enhanced in the mixotrophic condition compared to the phototrophic condition. This study indicates that mixotrophic culture promotes pigment and proximate production and the change of fatty acid profile in algae depends on the addition of organic carbon to the culture medium.  相似文献

Fatty acid (FA), total lipid, protein, amino acid, carbon, nitrogen, and phosphorus content was analyzed in 24 samples of freshwater microalgae. The samples originated from batch, continuous, or mass cultures in various growth phases and from net samples from lakewater. FA were analyzed quantitatively by using an internal standard in a GLC system and expressed as mg·g?1 dry weight (DW). The FA of one group of blue-greens (e.g. Oscillatoria and Microcystis) were similar to those of the greens with higher amounts of 18C acids of the ω3 type compared to the ω6 type, whereas the other group (e.g. Anabaena and Spirulina) contained mostly ω6 acids. The flagellates, a taxonomically diverse group, were characterized by high amounts of long-chained (20–22 C) polyunsaturated FA (PUFA), particularly of the ω3 type. The ω3/ω6 ratio appears to be highest in algae in the exponential growth phase. The increased lipid content in stressed algae was mostly due to increased saturated fatty acids and ω6 acids, whereas the valuable ω3 acids were unchanged or even decreased. Amino acid composition (% of total amino acids) did not vary much betaken species, but when analyzed quantitatively (mg-g?1 DW), varied considerably between species and within species in different growth phases. The nitrogen and phosphorus contents were variable in all three algal groups. The relationship between PUFA and phosphorus content differed among the algal groups. The data suggest that PUFA in the phospholipids consist mostly ω3 acids.  相似文献

Symbiotic algae incubated in host tissue homogenate of the coral Plesiastrea versipora for 2 h in the light released at least four and a half times as much photosynthetically fixed carbon (range 13.8±3.1 to 158±9.5 nmol C/106 algae) as algae incubated in seawater (range 1.4±0.3 to 10.8±0.6 nmol C/106 algae) indicating the presence of ‘host release factor’. When algae were incubated in a low molecular weight fraction of homogenate containing partially purified ‘host release factor’ they also released more carbon (range 62.2±3.7 to 279±11.4 nmol C/106 algae) than algae incubated in seawater. This low molecular weight fraction contained free amino acids. We tested the hypothesis that the free amino acids in this fraction were responsible for ‘host release factor’ activity. Algae incubated in a mixture of free amino acids equivalent to those found in this fraction, released more fixed carbon (range 2.4±0.3 to 25.2±0.2 nmol C/106 algae) than algae incubated in seawater but in each experiment, release was much lower than when algae were incubated in host tissue homogenate. These data indicate that the stimulation of release of photosynthetically fixed carbon from the symbiotic algae of Plesiastrea versipora incubated in partially purified host release factor is not primarily due to the presence of free amino acids. We are continuing further studies to determine the exact nature of the active compound.  相似文献

  1. According to the River Continuum Concept, headwater streams are richer in allochthonous (e.g. terrestrial leaves) than autochthonous (e.g. algae) sources of organic matter for consumers. However, compared to algae, leaf litter is of lower food quality, particularly ω-3 polyunsaturated fatty acids (n-3 PUFA), and would constrain the somatic growth, maintenance, and reproduction of stream invertebrates. It may be thus assumed that shredders, such as Gammarus, receive lower quality diets than grazers, e.g. Ecdyonurus, that typically feed on algae.
  2. The objective of this study was to assess the provision of dietary PUFA from leaf litter and algae to the shredder Gammarus and the grazer Ecdyonurus. Three different diets (algae, terrestrial leaves, and an algae–leaf litter mix) were supplied to these macroinvertebrates in a flume experiment for 2 weeks. To differentiate how diet sources were retained in these consumers, algae were isotopically labelled with 13C.
  3. Both consumers became enriched with 13C in all treatments, demonstrating that both assimilated algae. For Gammarus, n-3 PUFA increased, whereas n-6 PUFA stayed constant. By contrast, the n-3 PUFA content of Ecdyonurus decreased as a consequence of declining algal supply.
  4. Results from compound-specific stable isotope analysis provided evidence that the long-chain n-3 PUFA eicosapentaenoic acid (EPA) in both consumers was more enriched in 13C than the short-chain n-3 PUFA α-linolenic acid, suggesting that EPA was taken up directly from algae and not from heterotrophic biofilms on leaf litter. Both consumers depended on algae as their carbon and EPA source and retained their EPA from high-quality algae.

The River Continuum Concept implies that consumers in headwater streams have greater dietary access to terrestrial basal resources, but recent studies have highlighted the dietary importance of high-quality algae. Algae provide consumers with physiologically important omega-3 (n-3) polyunsaturated fatty acids (PUFA), particularly eicosapentaenoic acid (EPA). However, terrestrial plants and most benthic stream algae lack the long-chain (LC) n-3 PUFA docosahexaenoic acid (DHA, 22:6n-3), which is essential for neural development in fish and other vertebrates. We sampled subalpine streams to investigate how the PUFA composition of neural (brain and eyes), muscle, and liver tissues of freshwater fish is related to their potential diets (macroinvertebrates, epilithon, fresh and conditioned terrestrial leaves). The PUFA composition of consumers was more similar to epilithon than to terrestrial leaves. Storage lipids of eyes most closely resembled dietary PUFA (aquatic invertebrates and algae). However, DHA and arachidonic acid (ARA, 20:4n-6) were not directly available in the diet but abundant in organs. This implies that algal PUFA were selectively retained or were produced internally via enzymatic PUFA conversion by aquatic consumers. This field study demonstrates the nutritional importance of algal PUFA for neural organs in aquatic consumers of headwater regions.

Herbivory is a dominant feeding strategy among animals, yet herbivores are often protein limited. The gut microbiome is hypothesized to help maintain host protein balance by provisioning essential macromolecules, but this has never been tested in wild consumers. Using amino acid carbon (δ13C) and nitrogen (δ15N) isotope analysis, we estimated the proportional contributions of essential amino acids (AAESS) synthesized by gut microbes to five co-occurring desert rodents representing herbivorous, omnivorous and insectivorous functional groups. We found that herbivorous rodents occupying lower trophic positions (Dipodomys spp.) routed a substantial proportion (~40%–50%) of their AAESS from gut microbes, while higher trophic level omnivores (Peromyscus spp.) and insectivores (Onychomys arenicola) obtained most of their AAESS (~58%) from plant-based energy channels but still received ~20% of their AAESS from gut microbes. These findings empirically demonstrate that gut microbes play a key functional role in host protein metabolism in wild animals.  相似文献

Benthic food web dynamics and carbon flow were examined in the littoral zone of Lake Coleridge, a large deep oligotrophic lake, using radioactive and stable isotope techniques in conjunction with analyses of stomach contents of the fauna. We specifically address two hypotheses: (1) that macrophytes only contribute to the carbon flow to higher trophic levels when they have decayed; and (2) that epiphytic algae is the major source of carbon for macroinvertebrates, and thus fish, with only minor contributions from phytoplankton or terrestrial sources. Epiphytic diatoms were a major component of the stomach contents of the gastropod snail Potamopyrgus antipodarum, and of chironomids. Animal remains were also common in the diet of some chironomids, while amorphous organic matter predominated in the stomachs of oligochaetes. A variety of epiphytic algal taxa was found in trichopteran larvae. Feeding rate of P. antipodarum measured with radioactive tracers increased by 10× on decayed macrophytes (Elodea) compared with live material, while feeding rates on characean algae increased by a factor of 3 when decayed material was presented. However, assimilation rates were less than 20% on decayed material compared with 48–52% on live material. Potential carbon sources were easily distinguished based on their 13C values, although isotopic ratios showed significant variation among sites. Epiphytic algae showed less variation among sites than macrophytes and were depleted by 4–5 compared with macrophytes. Detrital material, organic matter in the sediments and plankton were significantly depleted in 13C relative to macrophytes and slightly depleted relative to epiphytic algae. Most macroinvertebrate taxa showed a similar pattern among sites to macrophytes and epiphytic algae. P. antipodarum and chironomids were slightly enriched compared with epiphytic algae. Ratios for the common bully (Gobiomorphus cotidianus) were generally consistent with a diet dominated by chironomids, while there was some evidence for terrestrial inputs for koaro (Galaxias brevipinnis) and juvenile brown trout. Epiphytic algae appear to underpin much of the production in the littoral zone of this oligotrophic lake, with trichopteran and chironomid larvae mediating carbon flows from algae to fish. Macrophytes do not make a major contribution directly to carbon flow to higher trophic levels even when decayed. The lack of a direct link between macrophytes and higher trophic levels is due to the faunal composition, including a lack of large herbivores.  相似文献

We looked at how three ecological tracers may influence the characterization and interpretation of trophic structures in a tropical marine system, with a view to informing tracer(s) selection in future trophic ecology studies. We compared the trophic structures described by stable isotope compositions (carbon and nitrogen), the total mercury concentration (THg) and levels of essential fatty acids (EFA) at both the individual and species level. Analyses were undertaken on muscle tissue samples from fish and crustacean species caught in the waters surrounding the Seychelles. The carbon isotope composition (δ13C) correlated to the proportion of arachidonic acid (ARA), whereas the nitrogen isotope composition (δ15N) correlated to the proportion of docosahexaenoic acid (DHA) and THg. At the individual level, trophic position obtained with these three last tracers are similar. In contrast, the eicosapentaenoic acid (EPA) was not clearly correlated to any of the tracers. At the species level, the use of EFA (ARA and DHA), as compared to stable isotopes, resulted in slight structural modifications, mainly in the middle trophic levels. For example, the EFA overestimated the trophic positions of Thunnus alalunga and Etelis coruscans but underestimated the trophic positions of other snappers and groupers. While ARA mainly originates from coastal/benthic areas, DHA is conserved throughout the food web and may be used as a proxy indicator of trophic position. However, metabolic disparities can affect ecological tracers and in turn, distort the trophic structures derived from their results. This is especially true for species with close trophic ecologies. Despite these caveats, we think that analysing at the individual level the wealth of ARA, DHA and THg data that has already been obtained through earlier nutrition or food security studies would enhance our understanding of trophic structures.  相似文献

How the abundant pelagic life of the Southern Ocean survives winter darkness, when the sea is covered by pack ice and phytoplankton production is nearly zero, is poorly understood. Ice‐associated (“sympagic”) microalgae could serve as a high‐quality carbon source during winter, but their significance in the food web is so far unquantified. To better understand the importance of ice algae‐produced carbon for the overwintering of Antarctic organisms, we investigated fatty acid (FA) and stable isotope compositions of 10 zooplankton species, and their potential sympagic and pelagic carbon sources. FA‐specific carbon stable isotope compositions were used in stable isotope mixing models to quantify the contribution of ice algae‐produced carbon (αIce) to the body carbon of each species. Mean αIce estimates ranged from 4% to 67%, with large variations between species and depending on the FA used for the modelling. Integrating the αIce estimates from all models, the sympagic amphipod Eusirus laticarpus was the most dependent on ice algal carbon (αIce: 54%–67%), and the salp Salpa thompsoni showed the least dependency on ice algal carbon (αIce: 8%–40%). Differences in αIce estimates between FAs associated with short‐term vs. long‐term lipid pools suggested an increasing importance of ice algal carbon for many species as the winter season progressed. In the abundant winter‐active copepod Calanus propinquus, mean αIce reached more than 50% in late winter. The trophic carbon flux from ice algae into this copepod was between 3 and 5 mg C m?2 day?1. This indicates that copepods and other ice‐dependent zooplankton species transfer significant amounts of carbon from ice algae into the pelagic system, where it fuels the food web, the biological carbon pump and elemental cycling. Understanding the role of ice algae‐produced carbon in these processes will be the key to predictions of the impact of future sea ice decline on Antarctic ecosystem functioning.  相似文献

1. It is often assumed that lakes highly influenced by terrestrial organic matter (TOM) have low zooplankton food quality because of elemental and/or biochemical deficiencies of the major particulate organic carbon pools. We used the biochemical [polyunsaturated fatty acids, especially eicosapentaenoic acid (EPA) – 20:5ω3] and elemental (C : P ratio) composition of particulate matter (PM) as qualitative measures of potential zooplankton food in two categories of lakes of similar primary productivity, but with contrasting TOM influence (clear water versus humic lakes). 2. C : P ratios (atomic ratio) in PM were similar between lake categories and were above 400. The concentration (μg L−1) and relative content (μg mg C−1) of EPA, as well as the particulate organic carbon concentration, were higher in the humic lakes than in the clear‐water lakes. 3. Our results show high fatty acid quality of PM in the humic lakes. The differences in the biochemical quality of the potential zooplankton food between lake categories can be attributed to the differences in their phytoplankton communities. 4. High biochemical quality of the food can result in high efficiency of energy transfer in the food chain and stimulate production at higher trophic levels, assuming that zooplankton are able to ingest and digest the resource available.  相似文献

  1. Chytrid fungal parasites convert dietary energy and essential dietary molecules, such as long-chain (LC) polyunsaturated fatty acids (PUFA), from inedible algal/cyanobacteria hosts into edible zoospores. How the improved biochemical PUFA composition of chytrid-infected diet may extend to zooplankton, linking diet quality to consumer fitness, remains unexplored.
  2. Here, we assessed the trophic role of chytrids in supporting dietary energy and PUFA requirements of the crustacean zooplankton Daphnia, when feeding on the filamentous cyanobacterium Planktothrix.
  3. Only Daphnia feeding on chytrid-infected Planktothrix reproduced successfully and had significantly higher survival and growth rates compared with Daphnia feeding on the sole Planktothrix diet. While the presence of chytrids resulted in a two-fold increase of carbon ingested by Daphnia, carbon assimilation increased by a factor of four, clearly indicating enhanced carbon transfer efficiency with chytrid presence.
  4. Bulk carbon (δ13C) and nitrogen (δ15N) stable isotopes did not indicate any treatment-specific dietary effects on Daphnia, nor differences in trophic position among diet sources and the consumer. Compound-specific carbon isotopes of fatty acids (δ13CFA), however, revealed that chytrids bioconverted short-chain to LC-PUFA, making it available for Daphnia. Chytrids synthesised the ω-3 PUFA stearidonic acid de novo, which was selectively retained by Daphnia. Values of δ13CFA demonstrated that Daphnia also bioconverted short-chain to LC-PUFA.
  5. We provide isotopic evidence that chytrids improved the dietary provision of LC-PUFA for Daphnia and enhanced their fitness. We argue for the existence of a positive feedback loop between enhanced Daphnia growth and herbivory in response to chytrid-mediated improved diet quality. Chytrids upgrade carbon from the primary producer and facilitate energy and PUFA transfer to primary consumers, potentially also benefitting upper trophic levels of pelagic food webs.

Isocitrate lyase is a key enzyme of the glyoxylate cycle. This cycle plays an essential role in cell growth on acetate, and is important for gluconeogenesis as it bypasses the two oxidative steps of the tricarboxylic acid (TCA) cycle in which CO2 is evolved. In this paper, a null icl mutant of the green microalga Chlamydomonas reinhardtii is described. Our data show that isocitrate lyase is required for growth in darkness on acetate (heterotrophic conditions), as well as for efficient growth in the light when acetate is supplied (mixotrophic conditions). Under these latter conditions, reduced acetate assimilation and concomitant reduced respiration occur, and biomass composition analysis reveals an increase in total fatty acid content, including neutral lipids and free fatty acids. Quantitative proteomic analysis by 14N/15N labelling was performed, and more than 1600 proteins were identified. These analyses reveal a strong decrease in the amounts of enzymes of the glyoxylate cycle and gluconeogenesis in parallel with a shift of the TCA cycle towards amino acid synthesis, accompanied by an increase in free amino acids. The decrease of the glyoxylate cycle and gluconeogenesis, as well as the decrease in enzymes involved in β–oxidation of fatty acids in the icl mutant are probably major factors that contribute to remodelling of lipids in the icl mutant. These modifications are probably responsible for the elevation of the response to oxidative stress, with significantly augmented levels and activities of superoxide dismutase and ascorbate peroxidase, and increased resistance to paraquat.  相似文献

Algae have been explored for renewable energy, nutraceuticals, and value-added products. However, low lipid yield is a significant impediment to its commercial viability. Genetic engineering can improve the fatty acid profile of algae without compromising its growth. This study introduced the diacylglycerol acyltransferase (BnDGAT) gene from Brassica napus into Chlorella sorokiniana-I, a fast-growing and thermotolerant natural strain isolated from wastewater, which increased its intracellular lipid accumulation. Hygromycin-resistant cells were selected, and enhanced green florescence protein fluorescence was used to distinguish pure transgenic cell lines from mixed cultures. Compared to the wild type, BnDGAT expression in transgenic C. sorokiniana-I caused a threefold increase in non-polar lipid and a twofold increase in polyunsaturated fatty acids. Nile red staining reaffirmed the presence of higher intracellular lipid bodies in transgenic cells. There was a substantial alteration in the fatty acid profile of transgenic alga expressing BnDGAT. The non-essential omega 9 (C18: 1) fatty acid decreased (5%–7% from 18%), while alpha-linolenic acid, an essential omega 3 fatty acid (C18: 3), was increased (23%–24% from 11%). This study substantiates a valuable strategy for enhancing essential omega-3 fatty acids and neutral lipids to improve its nutritional value for animal feed. The increased lipid productivity should reduce the cost of producing fatty acid methyl esters (FAME). Improved FAME quality should address the clouding issues in cold regions.  相似文献
