As a step toward greater understanding of the genetics of verticillium wilt resistance in plants, we report the sequencing of a candidate wilt resistance gene, mVe1, from the mint diploid model species, Mentha longifolia (Lamiaceae). mVe1 is a putative homolog of tomato (Solanum lycopersicum L.) verticillium wilt (Ve) resistance genes. The mVe1 gene has a coding region of 3,051 bp. The predicted mVe1 protein contains a leucine-rich repeat domain, a common feature of plant disease resistance proteins. We compared 13 mVe1 alleles from three mint species. These alleles shared 96.2–99.6% nucleotide identity. We analyzed four M. longifolia populations segregating with respect to mVe1 alleles and wilt resistance versus susceptibility and found one association between mVe1 genotype and wilt phenotype. We conclude that mVe1 may play a role in mint verticillium wilt resistance, but variation for resistance in our segregating progenies is likely polygenic. Therefore, further investigations of mVe1 and identification of additional candidate genes are both warranted.  相似文献

Commercial peppermint (P) (Mentha × piperita L. ev. Black Mitcham), native spearmint (NS) (M. spicata L.) and Scotch spearmint (SS) (M. × gracillis Sole cv Baker) petioles and orange mint (OM) (M. citrata Ehrh.) leaf disks were cocultivated with a number of Agrobacterium tumefaciens strains. P, SS and OM initiated tumor-like callus tissue on growth regulator-free MS medium after cocultivation with strain A281, a hypervirulent agropine strain containing Ti plasmid pTiBo542. Callus did not initiate from explants cocultivated with strain C58, a virulent nopaline strain; with A 136, a plasmidless strain, or from uninoculated controls. A281-derived callus was maintained on growth regulator-free medium in the absence of antibiotics for up to two years with no bacterial outgrowth. No shoots regenerated from any of the tumors on regeneration medium. Five of seven OM callus lines assayed gave a positive signal for agropine. DNA extracted from OM tumor tissue hybridized to a DNA probe specific to the T-DNA region of pTi plasmid. Genomic Southern analysis of DNA from tumors of P and SS indicated that one to a few copies of the T-DNA integrated into the mint chromosomes. PCR amplification of genomic DNA with primers specific for one of the T-DNA encoded genes yielded fragments that, when analyzed by restriction enzyme mapping and on Southern blots, corresponded to the cytokinin biosynthesis gene ipt. Summary Nine Triticum durumT. monococcum amphiploids (AABBAmAm) were synthesized by chromosome doubling of sterile triploid F1 hybrids involving nine T. durum (AABB) cultivars and a T. monococcum (AmAm) line. The triploid F1 hybrids had a range of 4–7 bivalents and 7–13 univalents per PMC. The synthetic amphiploids, however, showed a high degree of preferential pairing of chromosomes of the A genomes of diploid and tetraploid wheats. The amphiploids were meiotically stable and fully fertile. Superiority of four amphiploids for tiller number per plant, 100-grain weight, protein content and resistance to Karnal bunt demonstrated that these could either be commercially exploited as such after overcoming certain inherent defects or used to introgress desirable genes into durum and bread wheat cultivars. Methods for improvement of these amphiploids are discussed.  相似文献

Tomato gray mold (Botrytis cinerea Pers.) is a common disease worldwide, and often causes serious production loss by infecting leaves, stems, flowers and fruits. Presently, no resistant cultivars are available. To find new breeding materials for gray mold resistance, assessment for resistance of the leaflet and stem in six tomato cultivars, 44 wild tomato accessions and a Solanum lycopersicoides accession was performed. Although no correlation was observed (r=−0.127ns) between resistance of the leaflet and the stem, L. peruvianum LA2745, L. hirsutum LA2314 and L. pimpinellifolium LA1246 showed high resistance both in the leaflet and in the stem. Particularly, in the leaves of LA2745, no lesions were observed even more than two weeks after the inoculation with conidia, and F1s between a cultivated tomato and LA2745 also showed high resistance as observed in LA2745. From these results, LA2745 is thought to be a promising material for breeding gray-mold resistant cultivars.  相似文献

Diseases affecting strawberry (Fragaria × ananassa Duch.) have been of major concern in recent years because of their widespread occurrence and potential for yield loss. Anthracnose, caused by the fungus Colletotrichum acutatum, is one of the most serious diseases of strawberry worldwide. Tissue-culture induced (somaclonal) variation provides one strategy for generating disease-resistant genotypes. As part of a program to generate strawberry germplasm resistant to anthracnose, an in vitro screening system was used to evaluate several commercial cultivars, Chandler, Delmarvel, Honeoye, Latestar, Pelican and Sweet Charlie propagated in vitro, and shoots regenerated from leaf explants of these cultivars for resistance to C.␣acutatum isolate Goff (highly virulent). Regenerants with increased levels of resistance were identified from all of the cultivars. The greatest increases in disease resistance were observed for regenerants from leaf explants of cultivars Pelican and Chandler that exhibited 17.5- and 6.2-fold increases in resistance, respectively. The highest levels of anthracnose resistance (2 to 6% leaf necrosis) were exhibited by regenerants from explants of cultivars Pelican and Sweet Charlie. These studies suggest that generating somaclonal variation may be a viable approach to obtaining strawberry plants with increased levels of anthracnose resistance.  相似文献

The extent to which resistance to leek rust (Puccinia allii) in commercially useful cultivars of leek (Allium ampeloprasum) could be developed by selection within current commercial cultivars or their recent predecessors was investigated. Differences in resistance among commercial cultivars was not great. However, a wide range of resistance levels was found in both half‐sib and inbred progeny derived from these cultivars. In a crop simulation, one of the breeding lines showed a reduction in the rate of new pustule appearance of 44% compared to cv. Wintra. It is concluded that selection within modern cultivar level material is a potentially productive strategy for the commercial development of leek rust resistance in the short to medium term.  相似文献

Recombination rate data are presented for three populations of grape based on framework genetic linkage maps developed with simple-sequence repeat markers. These linkage maps were constructed from different Vitis species and represent three genetic backgrounds. The first population is pure Vitis vinifera, derived from a cross of the European cultivars Riesling and Cabernet Sauvignon. The second is an interspecific cross between two commercially used rootstock cultivars of different North American Vitis species parentage, Ramsey (Vitis champinii) and Riparia Gloire (Vitis riparia). The third population, D8909-15 (Vitis rupestris × (Vitis arizonica/Vitis girdiana form)) × F8909-17 (V. rupestris × (V. arizonica/Vitis candicans form)), is an F1 from two half-sibs. Genome-wide and chromosome-wide recombination rates varied across the three populations and among the six Vitis parents. Global recombination rates in the parents of the third F1 population, with a complex Vitis background, were significantly reduced. In the first and third populations, the recombination rate was significantly greater in the male parent. Specific genome locations with frequent heterogeneity in recombination were identified, suggesting that recombination rates are not equal across the Vitis genome. The identification of regions with suppressed or high recombination will aid grape breeders and geneticists who rely on recombination events to introgress disease resistance genes from the genomes of wild Vitis species, develop fine-scale genetic maps, and clone disease resistance genes. A study was conducted to determine whether the cuticles in two genera of the family Cruciferae are effective barriers to infection by Rhizoctonia solani, and whether differences in cuticle and epidermal cell wall thickness and morphology of epicuticular wax exist between resistant and susceptible cultivars. As Canola/rapeseed (Brassica napus) and mustard (Sinapis alba) plants develop from 1 to 3 weeks of age, they become increasingly resistant to R. solani AG2-1 seedling root rot. Seven-day-old seedlings of S. alba cultivars are invariably more resistant than B. napus cultivars. Brassica napus cultivars do not show an obvious cuticle layer at 1 week but at 3 weeks the presence of a cuticle is seen through autofluorescence with a concomitant increase in resistance to R. solani. Removal of the cuticle from 3-week-old hypocotyls by chloroform treatment results in a decrease in cuticular autofluorescence and a significant increase in disease severity in both resistant and susceptible cultivars. Three-week-old plants of S. alba have a much lower percent disease rating and a significantly (p=0.05) thicker cuticle layer than similar-age plants of B. napus. The results suggest that the cuticle plays an important role in the resistance of S. alba and older plants of B. napus to infection by R. solani.  相似文献

Status and Perspectives of Clubroot Resistance Breeding in Crucifer Crops   总被引:3,自引:0,他引:3  
Clubroot disease is a major threat to crops belonging to the Brassicaceae. It is controlled most effectively by the use of resistant cultivars. Plasmodiophora brassicae, the causal agent, shows a wide variation for pathogenicity, which can be displayed by using differential host sets. Except for Brassica juncea and B. carinata, resistant accessions can be found in all major crops. Most resistance sources are race-specific, despite some race-independent resistant accessions which can be found in B. oleracea. European field isolates from P. brassicae display great variation and show a tendency to overcome different resistance sources from either B. rapa or B. oleracea. At present, resistance genes from stubble turnips (B. rapa) are most effective and most widely used in resistance breeding of different Brassica crops. Resistance to P. brassicae from turnips was introduced into Chinese cabbage, oilseed rape, and B. oleracea. Although most turnips carry more than one resistance gene, the resistant cultivars from other crops received primarily a single, dominant resistance gene having a race-specific effect. Populations of P. brassicae that are compatible against most of the used resistance sources have been present in certain European areas for many decades. Such pathogen populations appeared in Japanese Chinese cabbage crops only a few years after the introduction of resistant cultivars. As the spread of virulent P. brassicae pathotypes seems to be slow, resistant cultivars are still a very effective method of control in many cropping areas. Mapping studies have revealed the presence of several clubroot-resistance genes in the Brassica A and C genomes; most of these genes are showing race specificity. Only in B. oleracea was one broad-spectrum locus detected. Two loci from the A genome confer resistance to more than one pathotype, but not to all isolates. Progress made in the determination of resistance loci should be used to formulate and introduce an improved differential set. Future efforts for breeding P. brassicae resistance will focus on durability by broadening the genetic basis of clubroot resistance by using either natural variation or transgenic strategies.  相似文献

Background  

Pre-harvest infection of peanuts by Aspergillus flavus and subsequent aflatoxin contamination is one of the food safety factors that most severely impair peanut productivity and human and animal health, especially in arid and semi-arid tropical areas. Some peanut cultivars with natural pre-harvest resistance to aflatoxin contamination have been identified through field screening. However, little is known about the resistance mechanism, which has slowed the incorporation of resistance into cultivars with commercially acceptable genetic background. Therefore, it is necessary to identify resistance-associated proteins, and then to recognize candidate resistance genes potentially underlying the resistance mechanism.  相似文献

The use of genetically diverse resistance sources is important in breeding for durable disease resistance. Detection and evaluation of resistance genes by conventional inheritance experiments, however, often require laborious screening and genetic testing. In the present study, a marker-assisted screening for resistance sources was initiated in soybean [Glycine max (L.) Merr] using one DNA microsatellite and two RFLP markers tightly linked to a soybean mosaic virus (SMV) resistance gene (Rsv1). The three marker loci were used to screen 67 diverse soybean cultivars, breeding lines, and plant introductions. Five variants were found at the microsatellite locus (HSP176L), and the two RFLP loci (pA186 and pK644a) near Rsv1 show a remarkably higher level of restriction polymorphism than Rsv1-independent RFLP loci. Several specific variants at the three marker loci were found to be correlated with virus resistance, among which HSP176L-2 can be detected by PCR, thus may be useful for germplasm screening. The grouping of the 67 accessions according to their multilocus marker variants agrees with the available pedigree information. When all, or most, of the cultivars within a given group with the same Rsv1-linked marker variant are resistant, their SMV resistance is most likely conferred by Rsv1. These putatively Rsv1-carrying groups contain a total of 38 SMV-resistant lines including six differential cultivars that are known to carry Rsv1. The remaining seven resistant accessions (Columbia, Holladay, Peking, Virginia, FFR-471, PI 507403, and PI 556949) do not carry resistance marker variants, and at least some of them could be sources of resistance genes independent of Rsv1.  相似文献

Effect of gene Lr34 in the enhancement of resistance to leaf rust of wheat   总被引:1,自引:0,他引:1  
Summary Leaf rust resistance gene Lr34 is present in many wheat cultivars throughout the world that have shown durable resistance to leaf rust. Fourteen pair-wise combinations of Lr34 and seedling leaf rust resistance genes were developed by intercrossing near isogenic Thatcher lines. In both seedling and adult plant tests homozygous paired combinations of specific resistance genes with Lr34 had enhanced resistance relative to either parent to different numbers of isolates that were avirulent to the additional resistance genes. The TcLr34, 18 line also expressed enhanced resistance to specific isolates virulent to Lr18 in seedling and adult plant stages. In rust nursery tests, homozygous lines were more resistant than either parent, if the additional leaf rust gene conditioned an effective of resistance when present singly. The ability of Lr34 to interact with other genes conditioning effective resistance may contribute to the durability of leaf rust resistance in cultivars with Lr34. Crossing blocks of Mentha arvensis cv Kalka (menthol mint without carvone) and Mentha spicata cv Neera (carvone mint without menthol) in alternate rows (2:1::Kalka:Neera), and pollination of florets of Kalka with that from Neera, yielded a carvone-rich variant among the open-pollinated seedlings. The variant possesses the hybrid phenotype, including the vigorous M. arvensis growth habit and the synthesis of rich oil aroma supplemented with a menthol tinge (carvone 64%–76% against 58% for the normal carvone mint cv Neera), and thus a novel combination of the essential oil. Chromosome counts and random amplified polymorphic DNA analysis confirmed that this spearmint variant, designated as the variety Neerkalka, is a unique interspecific hybrid (2n=5x=60) of M. arvensis cv Kalka (2n=8x=96) and M. spicata cv Neera (2n=2x=24). Vegetative multiplication of the hybrid was facilitated by its underground sucker-reproducing ability which is otherwise absent in spearmints. The per cent improvement in the variant ranged from 31–97 for herbage yield and 95–317 for oil yield over the standard spearmint varieties (MSS-5, Arka and Neera), with per hectare oil yields of 125.0 kg, 139.0 kg and 65.0 kg, respectively. The use of herbs in pharmaceutical preparation is ever increasing, and the demand for pesticides free material by the concern industries is on the rise. Consequently the need to grow disease-free plants using non-chemical fertilizers and pesticides is the need of the hour. Mentha arvensis cv. kosi is highly infested with Meloidogyne incognita (Kofoid and White) Chitwood, and severe oil yield loss occurs due to this nematode pest. Employing ecofriendly ways of nematode management, the mutualistic endophytes (Trichoderma harzianum strain Thu, Glomus intraradices) and plant growth promoting rhizobacteria (Bacillus megaterium and Pseudomonas fluorescens) were assessed individually and in combination on plant biomass, oil yield of menthol mint (M. arvensis cv. kosi), reproduction potential and population development of root knot nematode, M. incognita under glasshouse conditions. These microbes enhanced the plant biomass and percent oil yield both with and without M. incognita inoculation. Dual application of mutualistic fungal endophytes and Plant Growth Promoting Rhizobacteria (PGPRs) may be a wise option for enhancing the oil yield and tolerance of menthol mint against M. incognita infection.  相似文献

为鉴定引起四川盆地地区核桃黑斑病的病原菌,采用组织分离法对病原菌进行分离,利用柯赫氏法则验证其致病性,依据菌株形态学和基于16S rDNA基因序列分析对病原菌进行鉴定;同时,利用分离的菌株对18个栽培品种(无性系)进行抗病性评价。结果表明,分离菌株的菌落形态与黄单胞杆菌属(Xanthomonas)相似,其16S rDNA序列与树生黄单胞杆菌(X. arboricola)的(登录号为KP340804.1)同源性高达99%,因此,将引起四川盆地地区核桃黑斑病的病原菌鉴定为树生黄单胞杆菌。18个核桃栽培品种(无性系)的田间侵染发病率和病情指数分别为35.07%~78.57%和17.71%~51.96%,变异系数分别为17.62%和28.78%,并以此为基础评价出5个高抗病品种(无性系)。这为核桃黑斑病致病机理研究和抗病新品种的选育奠定基础。  相似文献

Powdery mildew significantly affects grain yield and end-use quality of winter wheat in the southern Great Plains. Employing resistance resources in locally adapted cultivars is the most effective means to control powdery mildew. Two types of powdery mildew resistance exist in wheat cultivars, i.e., qualitative and quantitative. Qualitative resistance is controlled by major genes, is race-specific, is not durable, and is effective in seedlings and in adult plants. Quantitative resistance is controlled by minor genes, is non-race-specific, is durable, and is predominantly effective in adult plants. In this study, we found that the segregation of powdery mildew resistance in a population of recombinant inbred lines developed from a cross between the susceptible cultivar Jagger and the resistant cultivar 2174 was controlled by a major QTL on the short arm of chromosome 1A and modified by four minor QTLs on chromosomes 1B, 3B, 4A, and 6D. The major QTL was mapped to the genomic region where the Pm3 gene resides. Using specific PCR markers for seven Pm3 alleles, 2174 was found to carry the Pm3a allele. Pm3a explained 61% of the total phenotypic variation in disease reaction observed among seedlings inoculated in the greenhouse and adult plants grown in the field and subjected to natural disease pressure. The resistant Pm3a allele was present among 4 of 31 cultivars currently being produced in the southern Great Plains. The genetic effects of several minor loci varied with different developmental stages and environments. Molecular markers associated with these genetic loci would facilitate incorporating genetic resistance to powdery mildew into improved winter wheat cultivars.  相似文献

Monogenically-inherited resistance to Soil-borne cereal mosaic virus (SBCMV) in hexaploid bread wheat cultivars ‘Tremie’ and ‘Claire’ was mapped on chromosome 5D. The two closest flanking markers identified in the Claire-derived mapping population, Xgwm469-5D and E37M49, are linked to the resistance locus at distances of 1 and 9 cm, respectively. Xgwm469-5D co-segregated with the SBCMV resistance in the Tremie-derived population and with the recently identified Sbm1 locus in the cv. Cadenza. This suggested that Tremie and Claire carry a resistance gene allelic to Sbm1, or one closely linked to it. The diagnostic value of Xgwm469-5D was assessed using a collection of SBCMV resistant and susceptible cultivars. Importantly, all susceptible genotypes carried a null allele of Xgwm469-5D, whereas resistant genotypes presumably related to either Claire and Tremie or Cadenza revealed a 152 or 154 bp allele of Xgwm469-5D, respectively. Therefore, Xgwm469-5D is well suited for marker assisted selection for SBCMV resistance.  相似文献

The present study is the first investigation of the volatile‐oil variability and insecticidal properties of the endemic Moroccan mint Mentha suaveolens subsp. timija (mint timija). The yield of essential oils (EOs) obtained from different wild mint timija populations ranged from 0.20±0.02 to 1.17±0.25% (v/w). GC/MS Analysis revealed the presence of 44 oil constituents, comprising 97.3–99.9% of the total oil compositions. The main constituents were found to be menthone (1.2–62.6%), pulegone (0.8–26.6%), cis‐piperitone epoxide (2.9–25.5%), piperitone (0.3–35.5%), trans‐piperitone epoxide (8.1–15.7%), piperitenone (0.2–9.6%), piperitenone oxide (0.5–28.6%), (E)‐caryophyllene (1.5–11.0%), germacrene D (1.0–15.7%), isomenthone (0.3–7.7%), and borneol (0.2–7.3%). Hierarchical‐cluster analysis allowed the classification of the EOs of the different mint timija populations into four main groups according to the contents of their major components. This variability within the species showed to be linked to the altitude variation of the mint timija growing sites. The results of the insecticidal tests showed that all samples exhibited interesting activity against adults of Tribolium castaneum, but with different degrees. The highest toxicity was observed for the EOs belonging to Group IV, which were rich in menthone and pulegone, with LC50 and LC90 values of 19.0–23.4 and 54.9–58.0 μl/l air in the fumigation assay and LC50 and LC90 values of 0.17–0.18 and 0.40–0.52 μl/cm2 in the contact assay.  相似文献

Striga hermonthica is a serious root parasite of sorghum in the semiarid tropics. Successful parasitism is dependent on interactions of Striga seeds and host roots. Several sorghum cultivars have been found which resist parasitism. The basis of resistance is not well known. One possible method for reducing the chances of parasitism is by restricted host root development. This research was conducted to evaluate this hypothesis in sorghum known to possess resistance to parasitism by Striga.Root length density of 21-day-old pot-grown resistant cultivars, Framida, N-13, IS-9830, Tetron and P-967083, were compared to that of the susceptible check, Dabar, using the line intercept method of measuring root length. There was no significant difference between resistant cultivars and the susceptible cultivar Dabar. The RLD of resistant P-967083 however was significantly less than Framida, another resistant cultivar.The RLD of Dabar was compared to that of Framida and P-967083 in USA and Niger field trials. Root length density was determined on soil cores taken at flowering with a Giddings Soil Sampler. Each core was divided into 10-cm fractions for estimating RLD by the line intercept method. In the USA Dabar had significantly greater RLD than the two resistant cultivars in the upper 10-cm portion of the soil profile, but only significantly greater than P-967083 in the 10–20-cm portion. Significant differences in RLD between susceptible and resistant cultivars were not found at depths between 20–60 cm. In field trials in Niger, RLD of Dabar was significantly greater than either resistant cultivar in the (0 to 30 cm) portion of the soil core. These results suggest that part of the Striga resistance of P-967083 and perhaps Framida may be a result of avoiding interactions between parasitic seeds and host roots.  相似文献
