青海高寒草甸土壤放线菌区系研究   总被引:10,自引:1,他引:9  
2001~2002年从海北高寒草甸生态系统采集土样,用不同方法从中分离放线菌300余株,根据其形态和分类特征,分别归入小单孢菌属(Micromonospora)、诺卡氏菌属(Nocardia)、糖多孢菌属(Saccharopolyspora)、原小单孢菌属(Promicromonospora)和链霉菌属(Streptomyces),并将链霉菌归入7个类群。同时对230株中温菌和110株低温菌的部分酶活性及其对真菌和细菌的拮抗性进行了测定,发现链霉菌不仅具有许多酶活性,而且对真菌和细菌有拮抗性。  相似文献

以采集自四个红树林地点的16份混合土壤为研究材料,选用7种选择性培养基,共分离获得330株放线菌。其中217株菌经16SrRNA基因序列分析,发现近75%菌株属于小单孢菌属(Micromonospora),其他还包括多形态孢菌属(Polymorphospora),疣孢菌属(Verrucosispora)等小单孢菌科的2个属和非小单孢菌科的9个属。采用美蓝酶标仪法对所分离到的放线菌进行抗菌活性检测,共50株菌表现出对金黄色葡萄球菌(Staphylococcus aureus ATCC 51650)、大肠杆菌(Escherichia coli ATCC 25922)和白色念珠菌(Candida albicans ATCC 10231)有不同程度抗性。然后利用高效液相色谱(HPLC)和液质联用技术(LC-MS)对有生物活性的菌株进行化学筛选,最后确定了5株可能产新颖化合物的小单孢菌。  相似文献

弯孢类炭疽菌菌株的RAPD分析与分类研究   总被引:7,自引:0,他引:7  
通过RAPD分析对38个不同寄主来源的弯孢类炭疽菌菌株的系统发育及分类进行研究。结果表明:许多引物的RAPD扩增带型在种内是相似或一致的,而种间差异较大。UPGMA聚类分析的结果表明:38个弯孢类炭疽菌菌株被聚为6个类群,群内菌株之间遗传相似性较高,而群间遗传相似性较低;群与群之间分界明显,表明种的分界相当明显。因此,RAPD分析所反映的种间亲缘关系,将有助于疑难种分类地位的确定和近似种的区分。RAPD分析的结果还揭示了一些近似种的分类关系,如按传统方法分别鉴定为Colletotrichumtruncatum、C.circinans和辣椒炭疽菌C.capsici的许多菌株聚类在同一群内,有很高的遗传相似性,实验结果支持它们为同一个种。实验中还发现5条辣椒炭疽菌的特有片断,分别由引物OPE-14、OPH-15(2条)、OPM-12和OPM-20扩增产生。这些特异带的发现对该菌的快速检测和鉴定具有重要意义。  相似文献

稗草致病菌——尖角突脐孢菌菌株RAPD指纹图谱的分析   总被引:8,自引:1,他引:7

以我国主要稻区的稗草植株上分离的17株尖角突脐孢菌菌株为试验材料,采用改良的SDS法提取其基因组DNA,并运用优化的RAPD分析体系对其进行了分子标记遗传差异研究。从25个随机引物中筛选出20个扩增效果好的引物,对全部试验材料进行了RAPD扩增,共得到239条有效带,其中多态性带229条(占95.8%)。依据扩增结果建立了17株尖角突脐孢菌基因型的DNA指纹图谱并对其进行了有效区分。根据RAPD分析结果计算了菌株间的遗传距离,分析了它们的遗传差异并进行了聚类分析,结果表明,RAPD分子标记技术是能够用于杂草致病菌资源的鉴定的,并可以进一步应用于特定性状的基因标记研究。  相似文献

瓜类保护地土壤镰孢菌种群及UP-PCR多样性分析   总被引:4,自引:0,他引:4  
对采自辽宁省部分地区瓜类保护地的36份土壤样品进行镰孢菌(Fusarium)分离培养,共获得112株镰孢菌,采用传统形态学分类和现代分子生物学方法,确定属于11个种.对其中25株镰孢菌及3株对照镰孢菌进行了通用引物PCR(UP-PCR)多样性分析.结果表明:6条引物扩增出73条带,其中多态性条带66条,占总条带数的90.4%.对供试菌株进行UP-PCR聚类图谱分析,当相似系数为0.736时,可将其划分为8个类群,其中14株尖孢镰孢菌聚为一类.UP-PCR分析体现了镰孢菌菌株间的亲缘关系及差异性,可以作为镰孢菌分类的辅助方法.  相似文献

【目的】对一株分离自植物根际土壤的具有抗真菌活性的链霉菌IMS002进行菌株分类鉴定,通过活性追踪分离纯化并鉴定有机相中的活性物质。【方法】通过16S rDNA和5个不同基因(atpD,gyrB,recA,rpoB,trpB)串联聚类分析以及生理生化实验分析,对链霉菌IMS002进行菌株分类鉴定,用扫描电子显微镜观察该株链霉菌的菌丝及孢子形态,以尖孢镰刀菌(Fusarium oxysporum)为指示菌进行生物活性追踪,通过硅胶柱层析、凝胶柱层析及高压液相色谱(HPLC)对活性物质进行分离和纯化,使用液质联用高分辨质谱仪、500 MHz核磁共振波谱仪以及圆二色光谱仪确定该物质的化学结构。【结果】IMS002经初步鉴定与产二素链霉菌(Streptomycesambofaciens)具有较近的亲缘关系,其发酵液对尖孢镰刀菌具有良好的抑菌效果,经分离和纯化以及现代波谱技术分析,确定有机相中的抑菌活性组分为Borrelidin。【结论】链霉菌IMS002能够产生化合物Borrelidin,该化合物对尖孢镰刀菌具有抑制活性。  相似文献

从云南程海湖分离到473株淡水真菌,用滤纸片法测定其抗菌活性,活性菌株经显微形态观察进行分类鉴定。结果显示有290株菌对至少1种病原菌具有抗菌活性,占总菌株数的61.31%;高抗菌活性菌株59株(抑菌圈直径≥15 mm),占总菌株的12.47%。将具有抗菌活性的菌株鉴定为46个属,优势属为青霉属(Penicillium)、曲霉属(Aspergillus)和枝孢属(Cladosporium)。  相似文献

采用组织块分离方法对五味子根内生真菌进行分离鉴定,并以人参疫病病菌[Phytophthora cactorum(Leb.et Coh.)Schroet.]、人参黑斑病病菌(Alternaria panax Whetz.)为供试病原菌对分离的内生真菌进行了抑菌活性筛选。试验共分离得到74株内生真菌,根据形态特征鉴定出33株,分别隶属于半知菌亚门丛梗孢科的轮枝孢属(Verticillium)、柱隔孢属(Ramularia)和瘤座孢科的镰刀菌属(Fusarium)。通过抑菌试验筛选出5个内生真菌有抑制作用,其中3个内生真菌的发酵产物、乙醇提取液对2种病原菌均有一定的抑菌活性。  相似文献

All five examined strains ofCoprinus cinereus could be clearly discriminated from the strains of five otherCoprinus species by RAPD patterns with 12 of 13 primers. Also one specimen of unknownCoprinus strain was identified to beC. cinereus by this method. The RAPD patterns were similar among the strains in the same species; many common DNA fragments were recognized as well as some strain-specific DNA fragments. Thus all seven strains ofC. cinereus and all four strains ofC. angulatus examined could be distinguished individually. Diakryotic strains showed the combined RAPD patterns of the two monokaryotic strains constituting the dikaryon. The combined RAPD markers observed in the dikaryons were segregated in their basidiospore progeny. All 18 randomly picked progeny showed different combinations of RAPD markers from the parental strains.  相似文献

Three molecular techniques, ITS sequence analysis, random amplified polymorphic DNA (RAPD) and amplified fragment length polymorphism (AFLP) were used to study phylogenetic and genotypic relationships among strains of the genus Fellomyces. In the analyses were included strains isolated predominantly from epiphytic lichens collected in Indonesia, China and Mexico. The polyphasic approach indicated that the Fellomyces isolates are genotypically heterogeneous and that lichens represent a specific environment for selection of large number of the sterigmatoconidia producing species. The phylogenetic and genotypic analysis confirmed the existence of 11 currently accepted Fellomyces species and indicated that several species may be the new representatives of the genus. The RAPD and AFLP analyses demonstrated a higher potential in distinguishing the Fellomyces strains than the ITS regions. Since the sequence analysis showed low or no divergence among several strains, both RAPD and AFLP fingerprinting indicated that the strains may be discriminated at the species level.  相似文献

We performed random amplification of polymorphic DNA (RAPD) analysis on five strains of Alexandrium tamarense and nine strains of Alexandrium minutum. Arbitrary 10-mer oligonucleotides were used as primers for the PCR. Electrophoresis on denaturing acrylamide gels improved RAPD reproducibility and increased the band number. Eight of the 20 primers assayed gave reproducible results and the band profiles generated by them were used for constructing a similarity matrix. Analyses were performed independently for the strains of each species and jointly for all the strains of both species. Results for A. tamarense showed the highest similarity for two distinct clones isolated from the same water sample in the Baltic Sea during a bloom (KAC01 and KAC02). The highest similarity among A. minutum clones was found for three strains (AL1V, AL2V and AL3V) isolated in the Ria de Vigo in NW Spain. The results show a high genetic diversity within a single species. We have shown the potential of the RAPD technique to discriminate between two conspecific strains, as well as for establishing similarities that are related to the biogeographic origin of the strains.  相似文献

根据ITS1-5.8S-ITS2区域的特异核酸序列变化,加特隐球酵母Cryptococcus gattii(≡新型隐球酵母加特变种Cryptococcu neoformans var.gattii)可分为6种基因型。本研究通过检测加特隐球酵母的IGS基因,发现其IGS序列有着更丰富的变异和信息位点。通过结合加特隐球酵母RAPD(随机扩增的多态性DNA)图谱比较研究,与IGS和ITS的序列分析结果大体一致,说明新近发现的加特隐球酵母ITS8型确实有别于以前报道过的其他加特隐球酵母ITS基因型。研究证明IGS1及IGS2基因片段分析可以作为加特隐球酵母基因分类鉴定中有效的辅助鉴别的分子生物学方法,联合多种基因分类鉴定的方法可以更有效地揭示新型隐球酵母加特变种种内不同基因亚型间的遗传进化关系。  相似文献

应用随机扩增多态性DNA技术(RAPD)对48株假丝酵母菌临床菌株进行PCR扩增,并对扩增产物的指纹图谱进行分析,结果显示:48株假丝酵母菌中白假丝酵母菌35株,非白假丝酵母菌13株,引物1和引物2将来源不同的35株白假丝酵母菌分成4型和6型,且RAPD技术可将白假丝酵母菌鉴定至种,并可分辩同种菌的不同型别,是假丝酵母菌致感染、追踪病原的分子流行病学研究有效方法。  相似文献

AIMS: To evaluate the effectiveness of two independent methods in differentiating a large population of lactic acid bacteria (LAB) isolated from wheat flours and sourdoughs and to correlate eventual differences/similarities among strains with their geographical origin and/or process parameters. METHODS AND RESULTS: One hundred fifty strains belonging to Lactobacillus spp. and Weissella spp., plus eight type strains, one for each species, and two unidentified isolates, were characterized by randomly amplified polymorphic DNA (RAPD) and SDS-PAGE of cell-wall proteins. The RAPD analysis separated the eight type strains but did not always assign all the strains of a species to the same group, while SDS-PAGE cell-wall protein profiles were species-specific. Frequently, strains isolated from sourdoughs of the same geographical origin or produced by similar raw material/process parameters showed similar RAPD and/or cell-wall profiles. CONCLUSIONS: The combined use of the RAPD and cell-wall protein analysis represents a useful tool to classify large adventitious microbial populations and to discriminate the diversity of the strains. SIGNIFICANCE AND IMPACT OF THE STUDY: This study represents a typing of a large collection of flour/sourdough LAB and provides evidence of the advantage of using two independent methods in the classification and traceability of microorganisms.  相似文献

本研究选用15 种东北藓类植物, 其中塔藓、赤茎藓、垂枝藓、拟垂枝藓、羽平藓、木藓、万年藓等为北半球广泛分布的大型藓类, 而褶叶藓、粗枝藓、球蒴金发藓等则是东亚特有种。在野外调查和采集的基础上, 进行RAPD 分析, 旨在从DNA 水平上对它们的亲缘关系和分类问题加以研究, 为它们的分类问题和亲缘关系提供分子证据。结果:从分子水平上进一步验证了15 种东北藓类植物形态分类的正确性。不同种类分别聚在金发藓目(Polytrichinales), 变齿藓目(Isobryales)和灰藓目(Hypnobryales)中, 与现采用分类系统一致。东北特产属褶叶藓属(Okamuraea)和粗枝藓属(Gollania)与垂枝藓科(Hyitidiaceae)植物关系较近, 而赤茎藓(Pleurozium)与绢藓(Entodon)有一定亲缘关系。因此, 将赤茎藓置于塔藓科(Hylocomiaceae)是合适的。  相似文献

The intraspecific genetic diversity of Oenococcus oeni, the key organism in the malolactic fermentation of wine, has been evaluated by random amplified polymorphic DNA (RAPD), ribotyping, small-plasmid content, and sequencing of RAPD markers with widespread distribution among the strains. Collection strains representing the diversity of this species have been studied together with some new isolates, many of which were obtained from wines produced by spontaneous malolactic fermentation. The RAPD profiles were strain specific and discerned two main groups of strains coincident with clusters obtained by macrorestriction typing in a previous work. Ribotyping and the conservation of RAPD markers indicates that O. oeni is a relatively homogeneous species. Furthermore, identical DNA sequences of some RAPD markers among strains representative of the most divergent RAPD clusters indicates that O. oeni is indeed a phylogenetically tight group, probably corresponding to a single clone, or clonal line of descent, specialized to grow in the wine environment and universally spread.  相似文献

用RAPD和AFLP的方法对中国卤虫(Artemia)种及亲缘关系的研究   总被引:18,自引:0,他引:18  
利用RAPD(随机扩增多态DNA)和AFLP(扩增片段长度多态性)技术对不同种及种群卤虫的关系进行分析。 101个随机引物对4种卤虫Afranciscana、A urmiana、A sinica和A.parthenogenelica基因组DNA进行扩增,平均每个种获得751条带,其中458条带为多态性标记,每个引物提供平均74个标记信息,聚类结果表明A.sinica是不同于其他旧大陆两性生殖卤虫的一个独立的种。对来自 15个种及品系的卤虫的 AFLP分析显示了非常好的遗传多态性,采用 12对引物检测到 594条带,其中 480个为多态性标记。聚类结果表明来自西藏的两性生殖卤虫为不同于中国内陆两性生殖卤虫的新种。而孤雌生殖卤虫在进化过程中可能是多源的,中国内陆和沿海的孤雌生殖卤虫是沿着不同的途径进化的,内陆和沿海的孤雌生殖卤虫可能为不同的种。  相似文献

Biotransformation of rifamycin B to rifamycin S using two strains of C. lunata namely NCIM 716 and NMU grown on various solid substrates viz., grass, paper, jowar/wheat straw, bran and bagasse was studied. Almost complete biotransformation efficiency of rifamycin B at 0. 06 mM concentration was observed within 24 hr. Among these two strains, C. lunata NMU showed 90% of biotransformation and higher rate of cellulose utilization on solid substrates vis-à-vis reference strain. Cellulase activity of both strains was also studied for exoglucanase, endoglucanase and beta-glucosidase. Column bioreactor studies with bagasse revealed further improvement in biotransformation efficiency of C. lunata NMU.  相似文献
