Background

Increasing incidences of insecticide resistance in malaria vectors are threatening the sustainable use of contemporary chemical vector control measures. Fungal entomopathogens provide a possible additional tool for the control of insecticide-resistant malaria mosquitoes. This study investigated the compatibility of the pyrethroid insecticide permethrin and two mosquito-pathogenic fungi, Beauveria bassiana and Metarhizium anisopliae, against a laboratory colony and field population of West African insecticide-resistant Anopheles gambiae s.s. mosquitoes.

Methodology/Findings

A range of fungus-insecticide combinations was used to test effects of timing and sequence of exposure. Both the laboratory-reared and field-collected mosquitoes were highly resistant to permethrin but susceptible to B. bassiana and M. anisopliae infection, inducing 100% mortality within nine days. Combinations of insecticide and fungus showed synergistic effects on mosquito survival. Fungal infection increased permethrin-induced mortality rates in wild An. gambiae s.s. mosquitoes and reciprocally, exposure to permethrin increased subsequent fungal-induced mortality rates in both colonies. Simultaneous co-exposure induced the highest mortality; up to 70.3±2% for a combined Beauveria and permethrin exposure within a time range of one gonotrophic cycle (4 days).

Conclusions/Significance

Combining fungi and permethrin induced a higher impact on mosquito survival than the use of these control agents alone. The observed synergism in efficacy shows the potential for integrated fungus-insecticide control measures to dramatically reduce malaria transmission and enable control at more moderate levels of coverage even in areas where insecticide resistance has rendered pyrethroids essentially ineffective.  相似文献

Background

Pyrethroid insecticide-treated bed nets (ITNs) help contribute to reducing malaria deaths in Africa, but their efficacy is threatened by insecticide resistance in some malaria mosquito vectors. We therefore assessed the evidence that resistance is attenuating the effect of ITNs on entomological outcomes.

Methods and Findings

We included laboratory and field studies of African malaria vectors that measured resistance at the time of the study and used World Health Organization–recommended impregnation regimens. We reported mosquito mortality, blood feeding, induced exophily (premature exit of mosquitoes from the hut), deterrence, time to 50% or 95% knock-down, and percentage knock-down at 60 min. Publications were searched from 1 January 1980 to 31 December 2013 using MEDLINE, Cochrane Central Register of Controlled Trials, Science Citation Index Expanded, Social Sciences Citation Index, African Index Medicus, and CAB Abstracts. We stratified studies into three levels of insecticide resistance, and ITNs were compared with untreated bed nets (UTNs) using the risk difference (RD). Heterogeneity was explored visually and statistically. Included were 36 laboratory and 24 field studies, reported in 25 records. Studies tested and reported resistance inconsistently. Based on the meta-analytic results, the difference in mosquito mortality risk for ITNs compared to UTNs was lower in higher resistance categories. However, mortality risk was significantly higher for ITNs compared to UTNs regardless of resistance. For cone tests: low resistance, risk difference (RD) 0.86 (95% CI 0.72 to 1.01); moderate resistance, RD 0.71 (95% CI 0.53 to 0.88); high resistance, RD 0.56 (95% CI 0.17 to 0.95). For tunnel tests: low resistance, RD 0.74 (95% CI 0.61 to 0.87); moderate resistance, RD 0.50 (95% CI 0.40 to 0.60); high resistance, RD 0.39 (95% CI 0.24 to 0.54). For hut studies: low resistance, RD 0.56 (95% CI 0.43 to 0.68); moderate resistance, RD 0.39 (95% CI 0.16 to 0.61); high resistance, RD 0.35 (95% CI 0.27 to 0.43). However, with the exception of the moderate resistance category for tunnel tests, there was extremely high heterogeneity across studies in each resistance category (chi-squared test, p<0.00001, I 2 varied from 95% to 100%).

Conclusions

This meta-analysis found that ITNs are more effective than UTNs regardless of resistance. There appears to be a relationship between resistance and the RD for mosquito mortality in laboratory and field studies. However, the substantive heterogeneity in the studies'' results and design may mask the true relationship between resistance and the RD, and the results need to be interpreted with caution. Our analysis suggests the potential for cumulative meta-analysis in entomological trials, but further field research in this area will require specialists in the field to work together to improve the quality of trials, and to standardise designs, assessment, and reporting of both resistance and entomological outcomes. Pyrethroid resistance is becoming a major problem for vector control programs, because at present, there are few suitable chemical substitutes for pyrethroids, as when used on bed nets the insecticide must have low mammalian toxicity as well as high activity to mosquitoes. Pyriproxyfen (PPF) is one of the most active chemicals among the juvenile hormone mimic (JHM) group. Sterilizing mosquitoes by using PPF could be a potential control measure for pyrethroid-resistant malaria vectors. We investigated the sterilizing effects of two types of PPF-impregnated bed nets – a 1% PPF-impregnated net and a 1% PPF +2% permethrin-impregnated net (Olyset Duo) – to pyrethroid-resistant wild population of Anopheles gambiae s.s. in western Kenya. High mortality of blood-fed mosquitos was observed 3 days post-collection, in the houses where PPF-impregnated nets were used, indicating the effect of PPF on the longevity of mosquitos that came in contact with the net. Reduction in the number of ovipositing females, number of eggs, and number of progeny per female were also observed in the houses in which both Olyset Duo and PPF-impregnated nets were used. This is the first field study showing the high sterilizing efficacy of PPF against wild pyrethroid-resistant An. gambiae s.s. population. In addition, we recognized the necessity of combined use of permethrin with PPF, in order to reduce the risk of mosquito bites and provide a level of personal protection. Further studies on wild pyrethroid-resistant mosquito populations such as An. arabiensis and An. funestus s.s. would provide more information on the practical use of the PPF-impregnated bed nets.  相似文献

Mosquito larvae of the sibling species Anopheles arabiensis Patton and An. gambiae Giles sensu stricto (Diptera: Culicidae) were investigated for interspecific competition. Single-species and mixed-species populations were reared at 27 degrees C from the first instar to pupation at different densities (100, 200 or 400 larvae/200 cm2 tray) with a constant amount of food, 0.2 mg/larva/day. Pupae obtained from mixed populations were identified to species using PCR. Both species had a 1:1 sex ratio at pupation. Development time to pupation averaged about one day less for An. arabiensis compared to An. gambiae, ranging from 0.93-1.49d for males and from 0.44-0.84 d for females in single populations. In mixed species populations the difference for males ranged from 0.99-1.58d and for females from 0.93-1.62d. Survival rates of An. gambiae s.s. were significantly higher than those of An. arabiensis in both the single-species and mixed-species populations. Mixed-species rearing did not have an effect on the survival of An. gambiae, whereas the mortality rate of An. arabiensis was significantly higher in mixed populations than when only this species was reared at the same densities, suggesting a competitive disadvantage for An. arabiensis in mixed populations. High proportions of larvae (4-35%) were lost during development; these losses could not be accounted for by corpses found in the rearing pans. The possibility of cannibalism was investigated by rearing each species separately in small containers (five per 50 ml), inspected every 6h, but no cannibalism was detected at any stage of development in either species. It was concluded that, under these experimental circumstances, interspecific competition between both species did occur but with a detrimental effect on An. arabiensis only. Relevance of these findings to the ecology of both species in the field is discussed briefly.  相似文献

Malaria vector control relies on toxicity of insecticides used in long lasting insecticide treated nets and indoor residual spraying. This is despite evidence that sub–lethal insecticides reduce human–vector contact and malaria transmission. The impact of sub–lethal insecticides on host seeking and blood feeding of mosquitoes was measured. Taxis boxes distinguished between repellency and attraction inhibition of mosquitoes by measuring response of mosquitoes towards or away from Transfluthrin coils and humans. Protective effective distance of coils and long-term effects on blood feeding were measured in the semi–field tunnel and in a Peet Grady chamber. Laboratory reared pyrethroid susceptible Anopheles gambiae sensu stricto mosquitoes were used. In the taxis boxes, a higher proportion of mosquitoes (67%–82%) were activated and flew towards the human in the presence of Transfluthrin coils. Coils did not hinder attraction of mosquitoes to the human. In the semi–field Tunnel, coils placed 0.3 m from the human reduced feeding by 86% (95% CI [0.66; 0.95]) when used as a “bubble” compared to 65% (95% CI [0.51; 0.76]) when used as a “point source”. Mosquitoes exposed to coils inside a Peet Grady chamber were delayed from feeding normally for 12 hours but there was no effect on free flying and caged mosquitoes exposed in the semi–field tunnel. These findings indicate that airborne pyrethroids minimize human–vector contact through reduced and delayed blood feeding. This information is useful for the development of target product profiles of spatial repellent products that can be used to complement mainstream malaria vector control tools.  相似文献

Male hybrids between Anopheles gambiae and An. arabiensis suffer from hybrid sterility, and inviability effects are sometimes present as well. We examined the genetic basis of these reproductive barriers between the two species, using 21 microsatellite markers. Generally, recessive inviability effects were found on the X chromosome of gambiae that are incompatible with at least one factor on each arabiensis autosome. Inviability is complete when the gambiae and arabiensis inviability factors are hemi- or homozygous. Using a QTL mapping approach, regions that contribute to male hybrid sterility were also identified. The X chromosome has a disproportionately large effect on male hybrid sterility. Additionally, several moderate-to-large autosomal QTL were found in both species. The effect of these autosomal QTL is contingent upon the presence of an X chromosome from the other species. Substantial regions of the autosomes do not contribute markedly to male hybrid sterility. Finally, no evidence for epistatic interactions between conspecific sterility loci was found.  相似文献

The response to phosphate of samples of natural local populationsof T. repens, collected from soils of different phosphate contents,was determined over a wide range of phosphate levels in sandculture. A marked correlation (r = +0.96) was found betweenresponse of populations to phosphate at low phosphate levelsand the amount of phosphate in the native soil of the population;populations from soils high in phosphate showed larger reductionsin growth at low phosphate levels than did those from soilslow in phosphate. It is concluded that response to phosphateis one aspect of the physiological adaptation of the naturalpopulations to particular soil conditions. Populations alsodiffered in response to very high levels of phosphate, and inthe range of phosphate levels tolerated; the possible adaptivesignificance of this is discussed in relation to the phosphatebuffering effects of the native soils. Populations tolerantof low phosphate levels had higher concentrations of phosphorusin the shoot material, and a higher uptake of phosphorus perunit weight of roots, than did intolerant populations. The differencesbetween populations in response to phosphate are therefore believedto be due to difference in uptake rather than to differencein metabolic requirement of phosphate.  相似文献

SYNOPSIS A microsporidan found parasitic in Anopheles gambiae in Tanzania was identified as Nosema algerae Vavra & Undeen, 1970. Its development was traced in laboratory-bred Anopheles stephensi. Spores were oval, 3.0-4.3 μ× 1.8-2.5 μ and had filaments 40-60 μ long; schizonts were uni-, bi- or tetranucleate; elongate sporonts gave rise to 2 sporoblasts. The parasite attacked almost every tissue of the body of the larvae, pupae and adults including the ova, but transovarial transmission was thought to be unimportant as infected ova did not develop into viable larvae. Hosts reacted to the presence of the parasite by encapsulating some spores in a coat of melanin.
The parasite was lightly infective to several species of culicine mosquitoes, but the infection was limited mainly to the nervous system, mid-gut epithelium and Malpighian tubules.

Host-seeking behaviour of the anthropophilic malaria vector Anopheles gambiae sensu stricto (Diptera: Culicidae) is mediated predominantly by olfactory cues. Several hundreds of odour components have been identified from human emanations, but only a few have been proven to act as attractants or synergists in the host-seeking behaviour of female An. gambiae. In previous work, aromatics, alcohols and ketones in human odours were found to elicit electrophysiological activity in antennal olfactory neurons of female An. gambiae. However, the behavioural effects of these compounds have not been investigated. In this study, behavioural responses of female An. gambiae to components of human breath, urine and sweat at a series of concentrations, or a single concentration in the case of acetone, were examined in combination with ammonia and L-lactic acid in a dual-choice olfactometer. The results showed that at specific concentrations 4-ethylphenol, indole, 3-methyl-1-butanol and two ketones inhibited the attractive effect of a mixture of ammonia and lactic acid. Acetone on its own was not attractive; however, when combined with lactic acid, the binary mixture was attractive. When combined with ammonia, acetone inhibited the attractiveness exerted by ammonia alone. Dodecanol and dimethyldisulphide did not affect the attraction exerted by ammonia and lactic acid at any of the concentrations tested. By contrast, a human-specific armpit odour, 7-octenoic acid, augmented the attraction exerted by the combination of ammonia and lactic acid at a specific dosage.  相似文献

Behavioural and electrophysiological responses of Anopheles gambiae Giles sensu stricto (Diptera: Culicidae) to human skin emanations collected on glass beads were studied using a dual-port olfactometer and electroantannography. Glass beads to which skin emanations from human hands had been transferred elicited a level of attraction similar to a human hand. The attractiveness of these handled glass beads faded away 4 h after transfer onto the beads. Storage at -20 degrees C for up to 8 weeks showed a decreased but still attractive effect of the beads. In a choice test between one individual and four others, the emanations from the reference individual were significantly more attractive in three out of four cases. The headspace of handled glass beads elicited a dose-dependent EAG response. The substances causing EAG activity could be removed partially by dichloromethane, ethanol and pentane-ether. Glass beads provide a suitable neutral substrate for the transfer of human odour to enable chemical analysis of the human skin emanations for identification of kairomones of anthropophilic mosquitoes.  相似文献

In Madagascar we used odour-baited entry traps (OBETs) for host choice tests of wild female anopheline mosquitoes (Diptera: Culicidae) at representative localities on the East and West sides of the island (villages Fenoarivo and Tsararano, respectively) and at the southern margin of the central plateau (Zazafotsy village, 800 m altitude). No insecticide house-spraying operations have been undertaken at these villages. Odours from a man and a calf of similar mass, concealed in different tents, were drawn by fans into separate OBETs set side by side. Traps were alternated to compensate for position effects, and different pairs of individual baits were employed for successive replicates. Totals of 266 An. funestus Giles sensu stricto and 362 An. gambiae Giles sensu lato were collected in 48 trap nights during March-June 1999. For each mosquito species the 'index of anthropophily' was defined as the proportion of females caught in the human-baited trap. For An. funestus this index was found to be consistently greater than 0.5 (value for random choice between traps/hosts), indicating that this species 'preferred' human to calf odour (index=0.83). Conversely, the index of anthropophily for An. gambiae s.l. indicated they 'chose' calf in preference to human odour (index=0.26). No significant differences of relative preference for calf or man were detected between villages; geographical variance accounted for <8% of the total experimental variance. Molecular identifications of 181 specimens of the An. gambiae complex (approximately 50% of the samples) revealed only An. arabiensis Patton at Tsararano and Zazafotsy, but >97% An. gambiae Giles sensu stricto at Fenoarivo, in accordance with prior knowledge of the differential distributions of these sibling species on the island. Predominant zoophily (i.e. intrinsic 'preference' for cattle odours) by both An. arabiensis and An. gambiae s.s. in Madagascar contrasts with their greater anthropophily in continental Africa.  相似文献

The relationship between mosquito 4th instar larval desiccation and survival to adulthood was explored by three methods in the laboratory. Two colonies of Anopheles arabiensis and one of Anopheles gambiae were studied. We found significant differences in tolerance to desiccation among all three stocks suggesting an intra‐ and interspecific genetic component to desiccation tolerance. An. arabiensis KGB, originating from Zimbabwe about 1975, had a much‐reduced desiccation tolerance compared to An. gambiae G3, colonized in the Gambia in 1975, and An. arabiensis DONGOLA which originated in Sudan in 2004. Individuals of the G3 stock survived desiccation of times up to 40 min with survival of 0.52. The degree of difference in tolerance between G3 and DONGOLA was smallest and was detected by one of three experimental methods. Mass losses of individuals that were weighed individually and survived to adulthood averaged 27% and 29% for G3 and DONGOLA and 20% for the less tolerant KGB stock, respectively. Such differences in survival in transiently dry larval habitats may account in part for differences in the distribution of these species and karyotypes.  相似文献

Prolonged bed rest may cause changes in the autonomic nervous system that are related to cognition and emotion. This study adopted an emotional flanker task to evaluate the effect of 45 days -6° head-down bed rest (HDBR) on executive functioning in 16 healthy young men at each of six time points: the second-to-last day before the bed rest period, the eleventh, twentieth, thirty-second and fortieth day during the bed rest period, and the eighth day after the bed rest period. In addition, self-report inventories (Beck Anxiety Inventory, BAI; Beck Depression Inventory, BDI; Positive Affect and Negative Affect Scale, PANAS) were conducted to record emotional changes, and the participants’ galvanic skin response (GSR), heart rate (HR) and heart rate variability (HRV) were assessed as measures of physiological activity. The results showed that the participants’ reaction time on the flanker task increased significantly relative to their responses on the second-to-last day before the period of bed rest, their galvanic skin response weakened and their degrees of positive affect declined during the bed rest period. Our results provide some evidence for a detrimental effect of prolonged bed rest on executive functioning and positive affect. Whether this stems from a lack of aerobic physical activity and/or the effect of HDBR itself remains to be determined.  相似文献

Predictions of the effects of climate change on the extent of forests, savannas and deserts are usually based on simple response models derived from actual vegetation distributions. In this review, we show two major problems with the implicitly assumed straightforward cause–effect relationship. Firstly, several studies suggest that vegetation itself may have considerable effects on regional climate implying a positive feedback, which can potentially lead to large‐scale hysteresis. Secondly, vegetation ecologists have found that effects of plants on microclimate and soils can cause a microscale positive feedback, implying that critical precipitation conditions for colonization of a site may differ from those for disappearance from that site. We argue that it is important to integrate these nonlinearities at disparate scales in models to produce more realistic predictions of potential effects of climate change and deforestation.  相似文献

Of eleven tomato cultivars ‘Baby’ was the most susceptible to Phytophthora infestans. The sporangium concentration of the fungus and the methods of fungicide application had a significant effect on the efficacy of oxadixyl, mancozeb and cymoxanil when tested singly or in mixture against sensitive and phenylamide resistant strains of P. infestans on tomato. Mancozeb was the least effective fungicide, its activity decreased significantly with increasing numbers of sporangia used for inoculation (increasing disease pressure) and its efficacy was dependent on the method of application. Oxadixyl and cymoxanil showed much stronger antifungal activities (except oxadixyl against resistant strains) which did not depend on the number of sporangia or method of fungicide application. The mixture of oxadixyl, mancozeb and cymoxanil was equally active and was independent of application type, sporangium concentration and level of sensitivity of the fungus. Mixtures of the single components showed synergistic interactions up to levels of 20, depending on the activity of the individual fungicide and the different disease intensities. Three-way mixtures containing oxadixyl, mancozeb, and cymoxanil represent therefore promising possibilities in practice for strong disease pressure, even when resistance problems are expected.  相似文献

Sugar feeding by male mosquitoes is critical for their success in mating competition . However, the facets of sugar source finding under natural conditions remain unknown. Here, evidence obtained in Western Burkina Faso indicated that the distribution of An. gambiae s.s. (M and S molecular forms) males across different peri‐domestic habitats is dependent on the availability of potential sugar sources from which they obtain more favorable sites for feeding or resting. Among field‐collected anophelines, a higher proportion of specimens containing fructose were found on flowering Mangifera indica (Anacardiaceae), Dolonix regia (Fabaceae), Thevetia neriifolia (Apocynaceae), Senna siamea, and Cassia sieberiana (both Fabaceae) compared to that recorded on other nearby plants, suggesting that some plants are favored for use as a sugar source over others. Y‐tube olfactometer assays with newly‐emerged An. gambiae s.s. exposed to odors from individual plants and some combinations thereof showed that males use odor cues to guide their preference. The number of sugar‐positive males was variable in a no‐choice cage assay, consistent with the olfactory response patterns towards corresponding odor stimuli. T hese experiments provide the first evidence both in field and laboratory conditions for previously unstudied interactions between males of An. gambiae and natural sugar sources.  相似文献

Abstract.Laboratory colonies of the human malaria vectors Anopheles gambiae Giles and An. arabiensis Patton have distinct flight tones. If flight tone similarly distinguishes natural populations of these sympatric sibling species, it may play a role in reproductive isolation of swarms that are otherwise behaviourally identical. To assess the fidelity of flight tone differences in natural populations, flight tone was measured in the F1 progeny of mosquitoes of both species captured in western Kenya. Flight tone distributions of wild An . gambiae and An. arabiensis were similar to their laboratory conspecifics. However, interspecies comparisons of flight tone of wild mosquitoes revealed significantly different but overlapping distributions for both sexes. Furthermore, when the effect of body size on flight tone was determined, there was a positive correlation between wing length and flight tone for both sexes of An. gambiae and An. arabiensis , suggesting that mosquito size is a significant variable affecting flight tone. Although these findings diminish any practical benefit of flight tone as a diagnostic tool in species identification, its potential role in pre‐mating species recognition needs further investigation.  相似文献
