Single clover plants were grown in the vegetative state, at 20 ± 1°, 85 ± 5% relative humidity, 320 ± 10 ppm CO2, 12-hour day, with Hoagland nutrient in Perlite, and 100 w · m−2 of photosynthetically active radiation (0.4-0.7 μ) from mercury-fluorescent lamps. Each plant was confined within a circle 18 cm in diameter by means of a wire framework. The CO2 exchange rate of the whole plant was measured every second day for 3 months. There was no optimum leaf area index for the net photosynthesis rate. The respiration rate was determined mainly by the gross photosynthesis rate and only partly by the amount of non-photosynthetic or heavily shaded tissue. At the maximum leaf area index, when leaves were dying as fast as they were being produced, both photosynthesis and respiration remained at or near their maximum rates. At the end of 3 months, the whole plant was harvested and the dry weight and carbon content determined. The measured dry weight was close to that calculated from the total CO2 uptake and a constant ratio of carbon content to dry weight of 39%. Optimum leaf area indices observed in field experiments are attributed to the failure to include the material which dies between harvests, and to decreases in the gross photosynthesis rate caused by climate changes or lack of nutrient, for example. The rate of photorespiration of Douglas-fir seedlings was measured under different light intensities by: (1) extrapolating the curve for CO2 uptake in relation to atmospheric CO2 content to zero CO2 content, and (2) measuring CO2 evolution of the plants into a CO2-free airstream. Different results, obtained from these techniques, were believed to be caused by a severe restriction of the photosynthetic activity when the latter was used. With the first method, CO2 evolution was lower than the dark respiration rate at low light intensity. For all temperatures studied (6°, 20°, 28°) a further increase in light intensity raised the CO2 evolution above dark respiration before it leveled off. The rate of CO2 evolution was stimulated by increase in temperature at all light intensities. With the CO2-free air method, CO2 evolution in the light was less than dark respiration at all light intensities.  相似文献

Whole-plant diurnal C exchange analysis provided a noninvasive estimation of daily net C gain in transgenic tobacco (Nicotiana tabacum L.) plants deficient in leaf cytosolic pyruvate kinase (PKc−). PKc− plants cultivated under a low light intensity (100 μmol m−2 s−1) were previously shown to exhibit markedly reduced root growth, as well as delayed shoot and flower development when compared with plants having wild-type levels of PKc (PKc+). PKc− and PKc+ source leaves showed a similar net C gain, photosynthesis over a range of light intensities, and a capacity to export newly fixed 14CO2 during photosynthesis. However, during growth under low light the nighttime, export of previously fixed 14CO2 by fully expanded PKc− leaves was 40% lower, whereas concurrent respiratory 14CO2 evolution was 40% higher than that of PKc+ leaves. This provides a rationale for the reduced root growth of the PKc− plants grown at low irradiance. Leaf photosynthetic and export characteristics in PKc− and PKc+ plants raised in a greenhouse during winter months resembled those of plants grown in chambers at low irradiance. The data suggest that PKc in source leaves has a critical role in regulating nighttime respiration particularly when the available pool of photoassimilates for export and leaf respiratory processes are low.  相似文献

Uptake of NO3 by nonnodulated soybean plants (Glycine max L. Merr. cv Ransom) growing in flowing hydroponic culture at 22 and 14°C root temperatures was measured daily during a 31-day growth period. Ion chromatography was used to determine removal of NO3 from solution during each 24-hour period. At both root-zone temperatures, rate of NO3 uptake per plant oscillated with a periodicity of 3 to 5 days. The rate of NO3 uptake per plant was consistently lower at 14°C than 22°C. The lower rate of NO3 uptake at 14°C during the initial 5 to 10 days was caused by reduced uptake rates per gram root dry weight, but with time uptake rates per gram root became equal at 14 and 22°C. Thereafter, the continued reduction in rate of NO3 uptake per plant at 14°C was attributable to slower root growth.  相似文献

Ocean acidification (OA) due to atmospheric CO2 rise is expected to influence marine primary productivity. In order to investigate the interactive effects of OA and light changes on diatoms, we grew Phaeodactylum tricornutum, under ambient (390 ppmv; LC) and elevated CO2 (1000 ppmv; HC) conditions for 80 generations, and measured its physiological performance under different light levels (60 µmol m−2 s−1, LL; 200 µmol m−2 s−1, ML; 460 µmol m−2 s−1, HL) for another 25 generations. The specific growth rate of the HC-grown cells was higher (about 12–18%) than that of the LC-grown ones, with the highest under the ML level. With increasing light levels, the effective photochemical yield of PSII (Fv′/Fm′) decreased, but was enhanced by the elevated CO2, especially under the HL level. The cells acclimated to the HC condition showed a higher recovery rate of their photochemical yield of PSII compared to the LC-grown cells. For the HC-grown cells, dissolved inorganic carbon or CO2 levels for half saturation of photosynthesis (K1/2 DIC or K1/2 CO2) increased by 11, 55 and 32%, under the LL, ML and HL levels, reflecting a light dependent down-regulation of carbon concentrating mechanisms (CCMs). The linkage between higher level of the CCMs down-regulation and higher growth rate at ML under OA supports the theory that the saved energy from CCMs down-regulation adds on to enhance the growth of the diatom.  相似文献

The relationships between various carbohydrate pools of the soybean (Glycine max [L.] Merrill) fruit and growth rate of seeds were evaluated. Plants during midpod-fill were subjected to various CO2 concentrations or light intensities for 7 days to generate different rates of seed growth. Dry matter accumulation rates of seeds and pod wall, along with glucose, sucrose, and starch concentrations in the pod wall, seed coat, and embryo were measured in three-seeded fruits located from nodes six through ten. Seed growth rates ranged from 4 to 37 milligrams·day−1·fruit−1. When seed growth rates were greater than 12 milligrams·day−1·fruit−1, sucrose concentration remained relatively constant in the pod wall (1.5 milligrams·100 milligrams dry weight−1), seed coat (8.5 milligrams·100 milligrams dry weight−1), and embryo (5.0 milligrams·100 milligrams dry weight−1). However, sucrose concentrations decreased in all three parts of the fruit as growth rate of the seeds fell below 12 milligrams·day−1·fruit−1. This relationship suggests that at high seed growth rates, flux of sucrose through the sucrose pools of the fruit was more important than pool size for growth. Starch concentration in the pod wall remained relatively constant (2 milligrams·100 milligrams dry weight−1) at higher rates of seed growth but decreased as seed growth rates fell below 12 milligrams·day−1·fruit−1. This suggests that pod wall starch may buffer seed growth under conditions of limiting assimilate availability. There was no indication that carbohydrate pools of the fruit were a limitation to transport or growth processes of the soybean fruit.  相似文献

Excised green leaves of mung bean (Phaseolus aureus L. var. Mungo) were used to determine the effect of light on the rate of endogenous respiration via the tricarboxylic acid cycle. Illumination with white light at an intensity of 0.043 gram calories cm−2min−1 (approximately 8600 lux) of visible radiation (400-700 nm) gave a rate of apparent photosynthesis, measured as net CO2 uptake, of 21 mg CO2 dm−2hr−1 which was about 11-fold greater than the rate of dark respiration. The feeding of 14CO2 or 14C-labeled acids of the tricarboxylic acid cycle in the dark for 2 hours was established as a suitable method for labeling mitochondrial pools of cycle intermediates.  相似文献

Ucides cordatus is an abundant mangrove crab in Brazil constructing burrows of up to 2 m depth. Sediment around burrows may oxidize during low tides. This increase in sediment-air contact area may enhance carbon degradation processes. We hypothesized that 1) the sediment CO2 efflux rate is greater with burrows than without and 2) the reduction potential in radial profiles in the sediment surrounding the burrows decreases gradually, until approximating non-bioturbated conditions. Sampling was conducted during the North Brazilian wet season at neap tides. CO2 efflux rates of inhabited burrows and plain sediment were measured with a CO2/H2O gas analyzer connected to a respiration chamber. Sediment redox potential, pH and temperature were measured in the sediment surrounding the burrows at horizontal distances of 2, 5, 8 and 15 cm at four sediment depths (1, 10, 30 and 50 cm) and rH values were calculated. Sediment cores (50 cm length) were taken to measure the same parameters for plain sediment. CO2 efflux rates of plain sediment and individual crab burrows with entrance diameters of 7 cm were 0.7–1.3 µmol m−2 s−1 and 0.2–0.4 µmol burrows−1 s−1, respectively. CO2 released from a Rhizophora mangle dominated forest with an average of 1.7 U. cordatus burrows−1 m−2 yielded 1.0–1.7 µmol m−2 s−1, depending on the month and burrow entrance diameter. Laboratory experiments revealed that 20–60% of the CO2 released by burrows originated from crab respiration. Temporal changes in the reduction potential in the sediment surrounding the burrows did not influence the CO2 release from burrows. More oxidized conditions of plain sediment over time may explain the increase in CO2 release until the end of the wet season. CO2 released by U. cordatus and their burrows may be a significant pathway of CO2 export from mangrove sediments and should be considered in mangrove carbon budget estimates.  相似文献

Needles from phosphorus deficient seedlings of Pinus radiata D. Don grown for 8 weeks at either 330 or 660 microliters CO2 per liter displayed chlorophyll a fluorescence induction kinetics characteristic of structural changes within the thylakoid chloroplast membrane, i.e. constant yield fluorescence (FO) was increased and induced fluorescence ([FP-FI]/FO) was reduced. The effect was greatest in the undroughted plants grown at 660 μl CO2 L−1. By week 22 at 330 μl CO2 L−1 acclimation to P deficiency had occurred as shown by the similarity in the fluorescence characteristics and maximum rates of photosynthesis of the needles from the two P treatments. However, acclimation did not occur in the plants grown at 660 μl CO2 L−1. The light saturated rate of photosynthesis of needles with adequate P was higher at 660 μl CO2 L−1 than at 330 μl CO2 L−1, whereas photosynthesis of P deficient plants showed no increase when grown at the higher CO2 concentration. The average growth increase due to CO2 enrichment was 14% in P deficient plants and 32% when P was adequate. In drought stressed plants grown at 330 μl CO2 L−1, there was a reduction in the maximal rate of quenching of fluorescence (RQ) after the major peak. Constant yield fluorescence was unaffected but induced fluorescence was lower. These results indicate that electron flow subsequent to photosystem II was affected by drought stress. At 660 μl CO2 L−1 this response was eliminated showing that CO2 enrichment improved the ability of the seedlings to acclimate to drought stress. The average growth increase with CO2 enrichment was 37% in drought stressed plants and 19% in unstressed plants.  相似文献

Individual leaves of potato (Solanum tuberosum L. W729R), a C3 plant, were subjected to various irradiances (400-700 nm), CO2 levels, and temperatures in a controlled-environment chamber. As irradiance increased, stomatal and mesophyll resistance exerted a strong and some-what paralleled regulation of photosynthesis as both showed a similar decrease reaching a minimum at about 85 neinsteins·cm−2·sec−1 (about ½ of full sunlight). Also, there was a proportional hyperbolic increase in transpiration and photosynthesis with increasing irradiance up to 85 neinsteins·cm−2·sec−1. These results contrast with many C3 plants that have a near full opening of stomata at much less light than is required for saturation of photosynthesis.  相似文献

A leaf disk assay for photorespiration has been developed based on the rate of release of recently fixed 14CO2 in light in a rapid stream of CO2-free air at 30° to 35°. In tobacco leaves (Havana Seed) photorespiration with this assay is 3 to 5 times greater than the 14CO2 output in the dark. In maize, photorespiration is only 2% of that in tobacco.

The importance of open leaf stomata, rapid flow rates of CO2-free air, elevated temperatures, and oxygen in the atmosphere in order to obtain release into the air of a larger portion of the 14CO2 evolved within the tissue in the light was established in tobacco. Photorespiration, but not dark respiration, was inhibited by α-hydroxy-2-pyridinemethanesulfonic acid, an inhibitor of glycolate oxidase, and by 3-(4-chlorophenyl)-1,1-dimethylurea (CMU), an inhibitor of photosynthetic electron transport, under conditions which did not affect the stomata. These experiments show that the substrates of photorespiration and dark respiration differ and also provide additional support for the role of glycolate as a major substrate of photorespiration. It was also shown that at 35° the quantity of 14CO2 released in the assay may represent only 33% of the gross 14CO2 evolved in the light, the remainder being recycled within the tissue.

It was concluded that maize does not evolve appreciable quantities of CO2 in the light and that this largely accounts for the greater efficiency of net photosynthesis exhibited by maize. Hence low rates of photorespiration may be expected to be correlated with a high rate of CO2 uptake at the normal concentrations of CO2 found in air and at higher light intensities.

Global climate change is expected to affect how plants respond to their physical and biological environments. In this study, we examined the effects of elevated CO2 ([CO2]) and low soil moisture on the physiological responses of mountain maple (Acer spicatum L.) seedlings to light availability. The seedlings were grown at ambient (392 µmol mol−1) and elevated (784 µmol mol−1) [CO2], low and high soil moisture (M) regimes, at high light (100%) and low light (30%) in the greenhouse for one growing season. We measured net photosynthesis (A), stomatal conductance (g s), instantaneous water use efficiency (IWUE), maximum rate of carboxylation (V cmax), rate of photosynthetic electron transport (J), triose phosphate utilization (TPU)), leaf respiration (R d), light compensation point (LCP) and mid-day shoot water potential (Ψx). A and g s did not show significant responses to light treatment in seedlings grown at low soil moisture treatment, but the high light significantly decreased the C i/C a in those seedlings. IWUE was significantly higher in the elevated compared with the ambient [CO2], and the effect was greater at high than the low light treatment. LCP did not respond to the soil moisture treatments when seedlings were grown in high light under both [CO2]. The low soil moisture significantly reduced Ψx but had no significant effect on the responses of other physiological traits to light or [CO2]. These results suggest that as the atmospheric [CO2] rises, the physiological performance of mountain maple seedlings in high light environments may be enhanced, particularly when soil moisture conditions are favourable.  相似文献

Portulacaria afra, a succulent plant, shifts from a predominantly C3 mode of gas exchange to a typical Crassulacean acid metabolism type CO2 uptake in response to water or NaCl stress. Control plants in the absence of water stress assimilated CO2 during the light (about 7-8 mg CO2 dm−2 hr−1), transpiration (about 1.5 g dm−2 hr−1) was predominantly during the day, stomates were open during the day, and there was little diurnal organic acid fluctuation. Stressed plants showed only dark CO2 uptake and dark water loss, nocturnal stomatal opening, and an increased diurnal fluctuation of titratable acidity. Within 2 weeks after rewatering, stressed plants returned to the control acid fluctuation levels indicating that the response to stress was reversible.  相似文献

In the intact eye, the transition from light to dark alters pH, [Ca2+], and [K] in the subretinal space (SRS) separating the photoreceptor outer segments and the apical membrane of the retinal pigment epithelium (RPE). In addition to these changes, oxygen consumption in the retina increases with a concomitant release of CO2 and H2O into the SRS. The RPE maintains SRS pH and volume homeostasis by transporting these metabolic byproducts to the choroidal blood supply. In vitro, we mimicked the transition from light to dark by increasing apical bath CO2 from 5 to 13%; this maneuver decreased cell pH from 7.37 ± 0.05 to 7.14 ± 0.06 (n = 13). Our analysis of native and cultured fetal human RPE shows that the apical membrane is significantly more permeable (≈10-fold; n = 7) to CO2 than the basolateral membrane, perhaps due to its larger exposed surface area. The limited CO2 diffusion at the basolateral membrane promotes carbonic anhydrase–mediated HCO3 transport by a basolateral membrane Na/nHCO3 cotransporter. The activity of this transporter was increased by elevating apical bath CO2 and was reduced by dorzolamide. Increasing apical bath CO2 also increased intracellular Na from 15.7 ± 3.3 to 24.0 ± 5.3 mM (n = 6; P < 0.05) by increasing apical membrane Na uptake. The CO2-induced acidification also inhibited the basolateral membrane Cl/HCO3 exchanger and increased net steady-state fluid absorption from 2.8 ± 1.6 to 6.7 ± 2.3 µl × cm−2 × hr−1 (n = 5; P < 0.05). The present experiments show how the RPE can accommodate the increased retinal production of CO2 and H2O in the dark, thus preventing acidosis in the SRS. This homeostatic process would preserve the close anatomical relationship between photoreceptor outer segments and RPE in the dark and light, thus protecting the health of the photoreceptors.  相似文献

Inoculated pea plants (Pisum sativum L.) were grown with N-free nutrients in a controlled environment room and rates of respiratory CO2 evolution and C2H2 reduction by the intact nodulated roots were determined. Experiments followed changes related to diurnal cycles, light and dark treatments, partial defoliation, aging of plants and NH4NO3 addition. In all experiments, changes in C2H2 reduction were associated with parallel changes in the respiration rate, although in all but the defoliation experiment there was a basal level of respiration which was independent of the rate of C2H2 reduction. In conditions which affected growth or plant size as well as C2H2 reduction, respiration changed by an average of 0.42 mg CO2 (μmol C2H2 reduced)−1. However, some treatments decreased C2H2 reduction without greatly changing the growth and in these conditions respiration was decreased by an average of 0.27 mg CO2 (μmol C2H2 reduced)−1. While this value may also include some respiration associated with other processes, it is proposed that it more closely estimates respiration directly associated with energy utilization for acetylene reduction; whereas the higher value includes respiration related to maintenance and growth processes as well.  相似文献

Soybean (Glycine max. L. Merr.) plants were grown in an experiment with a 3 × 3 factorial design using different levels of light (170, 350, and 700 μE·m−2·s−1) and P as factors. Plants were grown in a greenhouse in pot cultures using a soil low in plant-available P under three P regimes: no additional P, P added as KH2PO4, or P uptake enhanced by colonization of the host plant with the vesicular-arbuscular mycorrhizal (VAM) fungus Glomus fasciculatum (Thaxt. sensu Gerd.) Gerd. and Trappe. Development of the VAM fungal endophyte and of plants under all three P regimes was depressed by limiting light. However, the growth response of VAM plants to increasing light relative to non-VAM plants in the absence of additional P increased while the response relative to non-VAM plants with additional P decreased slightly. The highly significant interaction between the factors (P < 0.001) of the experiment was due to differences in the magnitude and direction of simple effects of the factors. The implications of these differences in terms of source-sink relationships of the symbionts and the value of different non-VAM controls in interpreting VAM effects are discussed.  相似文献

Elevated CO2 interactions with other factors affects the plant performance. Regarding the differences between cultivars in response to CO2 concentrations, identifying the cultivars that better respond to such conditions would maximize their potential benefits. Increasing the ability of plants to benefit more from elevated CO2 levels alleviates the adverse effects of photoassimilate accumulation on photosynthesis and increases the productivity of plants. Despite its agronomic importance, there is no information about the interactive effects of elevated CO2 concentration and plant growth regulators (PGRs) on potato (Solanum tuberosum L.) plants. Hence, the physiological response and source-sink relationship of potato plants (cvs. Agria and Fontane) to combined application of CO2 levels (400 vs. 800 µmol mol−1) and plant growth regulators (PGR) [6-benzylaminopurine (BAP) + Abscisic acid (ABA)] were evaluated under a controlled environment. The results revealed a variation between the potato cultivars in response to a combination of PGRs and CO2 levels. Cultivars were different in leaf chlorophyll content; Agria had higher chlorophyll a, b, and total chlorophyll content by 23, 43, and 23%, respectively, compared with Fontane. The net photosynthetic rate was doubled at the elevated compared with the ambient CO2. In Agria, the ratio of leaf intercellular to ambient air CO2 concentrations [Ci:Ca] was declined in elevated-CO2-grown plants, which indicated the stomata would become more conservative at higher CO2 levels. On the other hand, the increased Ci:Ca in Fontane showed a stomatal acclimation to higher CO2 concentration. The higher leaf dark respiration of the elevated CO2-grown and BAP + ABA-treated plants was associated with a higher leaf soluble carbohydrates and starch content. Elevated CO2 and BAP + ABA shifted the dry matter partitioning to the belowground more than the above-media organs. The lower leaf soluble carbohydrate content and greater tuber yield in Fontane might indicate a more efficient photoassimilate translocation than Agria. The results highlighted positive synergic effects of the combined BAP + ABA and elevated CO2 on tuber yield and productivity of the potato plants.  相似文献

The data reported were obtained by noting the response of pepper plants to all combinations of the following treatments: high light, 1.5 × 106 ergs per square centimeter per second; low light, 3.0 × 104 ergs per square centimeter per second; three levels of CO2: 50, 268, and 730 parts per million; nutrient solution osmotic potentials of −0.5, −5.0, −7.5, and −9.5 bars.  相似文献

The effects of Mg deficiency on the photosynthesis and respiration of sugar beets (Beta vulgaris L. cv. F58-554H1) were studied by withholding Mg from the culture solution and by following changes in CO2 and water vapor exchange of attached leaves. Leaf blade Mg concentration decreased from about 1200 to less than 200 meq kg−1 dry matter without change in the rate of photosynthetic CO2 uptake per unit leaf area, while from 200 to 50 meq kg−1 the rate decreased to one-third. Rates of photorespiratory evolution of CO2 into CO2-free air responded to Mg like those of photosynthetic CO2 uptake, the rates decreasing to one-half, below 200 meq kg−1. Respiratory CO2 evolution in the dark increased almost 2-fold in low Mg leaves. Magnesium deficiency had less effect on leaf (mainly stomatal) diffusion resistance (r1) than on mesophyll resistance (rm); in Mg-deficient plants rm increased from 2.9 to 7.1 sec cm−1, whereas r1 became significantly greater than the control value only in the most severe instances of Mg deficiency.  相似文献
