The pericarp of the dormant sugarbeet fruit acts as a storage reservoir for nitrate, ammonium and -amino-N. These N-reserves enable an autonomous development of the seedling for 8–10 d after imbibition. The nitrate content of the seed (1% of the whole fruit) probably induces nitrate-reductase activity in the embryo enclosed in the pericarp. Nitrate that leaks out of the pericarp is reabsorbed by the emerging radicle. Seedlings germinated from seeds (pericarp was removed) without external N-supply are able to take up nitrate immediately upon exposure via a low-capacity uptake system (vmax = 0.8 mol NO 3 - ·(g root FW)–1·h–1; Ks = 0.12 mM). We assume that this uptake system is induced by the seed nitrate (10 nmol/seed) during germination. Induction of a high-capacity nitrate-uptake system (vmax = 3.4 mol NO 3 - ·(g root FW)–1·h–1; Ks = 0.08 mM) by externally supplied nitrate occurs after a 20-min lag and requires protein synthesis. Seedlings germinated from whole fruits absorb nitrate via a highcapacity uptake mechanism induced by the pericarp nitrate (748 nmol/pericarp) during germination. The uptake rates of the high-capacity system depend only on the actual nitrate concentration of the uptake medium and not on prior nitrate pretreatments. Nitrate deprivation results in a decline of the nitrate-uptake capacity (t1/2 of vmax = 5 d) probably caused by the decay of carrier molecules. Pathways of K+ movement across the erythrocyte membrane of frog Rana temporaria were studied using 86Rb as a tracer. The K+ influx was significantly blocked by 0.1 mmol·l-1 ouabain (by 30%) and 1 mmol·l-1 furosemide (by 56%) in the red cells incubated in saline at physiological K+ concentration (2.7 mmol·l-1). Ouabain and furosemide had an additive effect on K+ transport in frog red cells. The ouabain-sensitive and furosemide-sensitive components of K+ influx saturated as f(K+)e with apparent K m values for external K e + concentration of 0.96±0.11 and 4.6±0.5 mmol·l-1 and V max of 0.89±0.04 and 2.8±0.4 mmol·l cells-1·h-1, respectively. The residual ouabain-furosemide-resistant component was also a saturable function of K e + medium concentration. Total K+ influx was significantly reduced when frog erythrocytes were incubated in NO - 3 medium. Furosemide did not affect K+ transport in frog red cells in NO 3 - media. At the same K e + concentration the ouabain-furosemide-insensitive K+ influx in Cl- medium was significantly greater than that in NO - 3 medium. We found no inhibitory effect of 1 mmol·l-1 furosemide on Na+ influx in frog red cells in Cl- medium. K+ loss from the frog erythrocytes in a K+-free medium was significantly reduced (mean 58%) after replacement of Cl- with NO - 3 . Furosemide (0.5 mmol·l-1) did not produce any significant reduction in the K+ loss in both media. The Cl--dependent component of K+ loss from frog red cells was 5.7±1.2 mmol·l-1·h-1. Extrusion of H+ by intact barley (Hordeum vulgare L.) roots was automatically titrated. Simultaneously, uptake of K+ into the roots, transport of K+ through the roots, and (as a residual term) accumulation of K+ within the root tissue were determined. When no monovalent cation was present in the medium the steady rate of H+ release was close to zero. Addition of K+ stimulated H+ extrusion within less than 1 min. The stimulation of H+ release was apparently limited only by the movement of K+ through the apoplast of the roots. The steady rate of H+ extrusion depended on the availability of external K+ and saturated at a K+ concentration of about 100 mol· dm-3. Half-maximum rates of net K+ uptake and H+ extrusion were reached at a K+ concentration of about 10 mol·dm-3. With (slowly absorbable) sulfate as the only anion present, the stoichoimetry between H+ release and net K+ uptake was one. In conclusion, the uptake of K+ across the plasmalemma of the cells of the root cortex is electrically coupled to H+ extrusion.  相似文献

Isolated posterior gills (no. 7) of shore crabsCarcinus maenas acclimated to brackish water of a salinity of 10 S were bathed and perfused with 50% sea water (200 mmol·l-1 Na+), and the internal perfusate collected during subsequent periods of 5 min. During a single passage through the gill the pH of the perfusion medium decreased from ca. 8.1 to ca. 7.7, a result implying that the gill possesses structures which recognize unphysiologically high pH values in the haemolymph and regulates them down to physiological values of ca. 7.7. The calculated apparent proton fluxes from the epithelial cells into the haemolymph space amounted to 17.9 mol·g fw-1·h-1, a value of only 3.8% of net Na+ fluxes observed under comparable conditions. When 0.1 mmol·l-1 KCN, an inhibitor of mitochondrial cytochrome oxidase, or 5 mmol·l-1 ouabain, a specific inhibitor of Na+/K+-ATPase were applied in the internal perfusate, down-regulation of pH was no longer observed and the gill was completely depolarized, i.e. transepithelial potential differences dropped from-7.8 to 0 mV (haemolymph space negative to bath). Regulation of pH was completely inhibited by antagonists of carbonic anhydrase (0.1 mmol·l-1 acetazolamide or 0.01 mmol·l-1 ethoxyzolamide) applied in the perfusate. Inhibitors of Na+/H+ exchange, 0.1 mmol·l-1 amiloride applied in the external bathing medium or in the internal perfusate, and symmetrical 0.01 mmol·l-1 5-(N-ethyl-N-isopropyl)amiloride, as well as inhibitors of Cl-/HCO3 - exchange and Na+/HCO3 - cotransport, 0.5 mmol·l-1 4,4-diisothiocyanatostilbene-2,2-disulphonate or 0.3 mmol·l-1 4-acetamido-4-isothiocyanatostilbene 2,2-disulphonate applied on both sides of the gill, and inhibitors of H+-ATPase, 0.05 mmol·l-1 N-ethylmaleimide and 0.1 mmol·l-1 N,N-dicyclohexylcarbodiimide —applied on both sides of the gill — did not alter the acidification of the perfusate observed in controls. Using artificial salines buffered to pH 8.1 with 0.75 mmol·l-1 tris (hydroxymethyl) aminomethane instead of 2 mmol·l-1 HCO3 -, apparent proton fluxes were reduced to 11% of controls, a result suggesting that pH regulation by crab gills needs the presence of HCO3 -. The findings obtained suggest that pH regulation by crab gills depends on the oxidative metabolism of the intact branchial epithelium and that carbonic anhydrase plays a central role in this process. Na+/H+ exchange, anion exchange or cotransport and active proton secretion seem not to be involved. The inhibition of K+ uptake through the plasma membrane resulting from injury caused by cutting, or from application of polyamines (PAs), has been investigated in root segments of maize (Zea mays L.) and pea (Pisum sativum L.). It was found, for both treatments, that K+ uptake recovered if the segments were washed for 2 h. The K+ uptake inhibited by cutting and that inhibited by spermidine treatment were stimulated to the same extent by fusicoccin. In addition, there was a correlation between the extent of the recovery of K+ uptake caused by washing and the distribution, along the root axis, of both PAs and the activities of enzymes responsible for PA degradation. In apical segments of maize, where the PA content and the activity of the degradative enzyme polyamine oxidase (EC 1.5.3.3) were higher than in the more distal segments, the recovery of K+ uptake caused by washing was also higher. On the other hand, the opposite trend was observed in root segments of pea, where the PA content and the activity of the degradative enzyme diamine oxidase (EC 1.4.3.6) were higher in distal segments in which K+ uptake was greatly stimulated by washing. The effect of the amine-oxidase inhibitor, aminoguanidine, indicates that the degradation products of PAs are involved in the mechanism of inhibition of K+ uptake by PAs. Spinach (Spinacia oleracea L.) and barley (Hordeum vulgare L.) were grown in hydroponic culture with varying levels of orthophosphate (Pi). When leaves were fed with 20 mmol·l–1 Pi at low CO2 concentrations, a temporary increase of CO2 uptake was observed in Pi-deficient leaves but not in those from plants grown at 1 mmol·l–1 Pi. At high concentrations of CO2 (at 21% or 2% O2) the Pi-induced stimulation of CO2 uptake was pronounced in the Pi-deficient leaves. The contents of phosphorylated metabolites in the leaves decreased as a result of Pi deficiency but were restored by Pi feeding. These results demonstrate that there is an appreciable capacity for rapid Pi uptake by leaf mesophyll cells and show that the effects of long-term phosphate deficiency on photosynthesis may be reversed (at least temporarily) within minutes by feeding with Pi.Abbreviation Pi orthophosphate  相似文献

About 92% of the taurine influx in flounder erythrocytes at physiological conditions in vitro (330 mosmol·l-1, 145 mmol·l-1 Na+, 0.30 mmol·l-1 taurine) is Na+-dependent. This influx is highly specific for taurine. The -amino compounds hypotaurine and -alanine were the only compounds which mimicked the inhibitory effect of taurine on influx of [14C]taurine, the former more than the latter. Counterexchange of taurine was also mediated by the taurine transporters. Reduction of osmolality per se did not affect the activity of these transporters. Non-linear regression analysis of the influx values revealed the presence of two different influx systems: a system with high affinity and low capacity and another with low affinity and high capacity. However, we cannot exclude the possibility that this influx of taurine was mediated by only one transporter which operated in different modes depending on the extracellular Na+ concentration. On the assumption that the Na+-dependent influx was mediated by two separate systems, the maximal velocity of the low capacity system was 2.55 nmol·g dry weight-1·min-1 at 145 mmol·ll-1 extracellular Na+. This capacity was about 50% lower than that of the high capacity system. The Michaelis constants were 0.013 and 1.34 mmol·l-1, respectively. Reduction of the extracellular Na+ concentration reduced maximal velocity and the affinity to taurine of both transport systems. At 10 mmol·l-1 Na+ or lower concentrations the high capacity system did not seem to operate. The activation method suggested that each taurine molecule transported by the high capacity system was accompanied by two Na+. The stoichiometry of the low capacity system was 1 taurine: 1 Na+. Within 1 min of transfer from coelomic fluid to fresh water, eggs of rainbow trout (Oncorhynchus mykiss) underwent a transient loss of Na+ and K+ coupled with an elevation of the chorionic envelope. Both mechanisms were blocked by adding a monovalent cation Li+ or K+ (140 mmol·l-1) to the fresh water, but the divalent ion Mg2+ (100 mmol MgCl2·l-1) or elevating the osmotic pressure to 300 mOsmol·l-1 with glycine had no inhibitory effect. The blocking of Na+ loss occurred at external monovalent cation (LiCl) concentrations above 70 mmol·l-1. A 20-s exposure of eggs to fresh water was sufficient to trigger Na+ loss and chorion elevation, even when the eggs were subsequently transferred to fresh water containing 140 mmol LiCl·l-1. Eggs placed in a medium containing 140 mmol LiCl·l-1 and 2 mmol Ca(NO3)2·l-1 showed chorion elevation and associated Na+ loss after addition of calcium ionophore (20 mol·l-1 A.23187). The dorsal skin of the leech Hirudo medicinalis was used for electrophysiological measurements performed in Ussing chambers. The leech skin is a tight epithelium (transepithelial resistance = 10.5±0.5 k· cm-2) with an initial short-circuit current of 29.0±2.9 A·cm-2. Removal of Na+ from the apical bath medium reduced short-circuit current about 55%. Ouabain (50mol·l-1) added to the basolateral solution, depressed the short-circuit current completely. The Na+ current saturated at a concentration of 90 mmol Na+·l-1 in the apical solution (K M=11.2±1.8 mmol·l-1). Amiloride (100 mol·l-1) on the apical side inhibited ca. 40% of the Na+ current and indicated the presence of Na+ channels. The dependence of Na+ current on the amiloride concentration followed Michaclis-Menten kinetics (K i=2.9±0.4 mol·l-1). The amiloride analogue benzamil had a higher affinity to the Na+ channel (K i=0.7±0.2 mol·l-1). Thus, Na+ channels in leech integument are less sensitive to amiloride than channels known from vertebrate epithelia. With 20 mmol Na+·l-1 in the mucosal solution the tissue showed an optimum amiloride-inhibitable current, and the amiloride-sensitive current under this condition was 86.8±2.3% of total short-circuit current. Higher Na+ concentrations lead to a decrease in amiloride-blockade short-circuit current. Sitmulation of the tissue with cyclic adenosine monophosphate (100 mol·l-1) and isobutylmethylxanthine (1 mmol·l-1) nearly doubled short-circuit current and increased amiloride-sensitive Na+ currents by 50%. By current fluctuation analysis we estimated single Na+ channel current (2.7±0.9 pA) and Na+ channel density (3.6±0.6 channels·m-2) under control conditions. After cyclic adenosine monophosphate stimulation Na+ channel density increased to 5.4±1.1 channels·m-2, whereas single Na+ channel current showed no significant change (1.9±0.2 pA). These data present a detailed investigation of an invertebrate epithelial Na+ channel, and show the similarities and differences to vertebrate Na+ channels. Whereas the channel properties are different from the classical vertebrate Na+ channel, the regulation by cyclic adenosine monophosphate seems similar. Plant roots accumulate K+ over a range of external concentrations. Root cells have evolved at least two parallel plasma-membrane K+ transporters which operate at millimolar and micromolar external [K+]: high-affinity K+ uptake is energised by symport with H+, while low-affinity uptake is assumed to occur via ion channels. To determine the role of ion channels in low-affinity K+ uptake, a characterisation of the principal K+-selective ion channels in the plasma membrane of Arabidopsis thaliana (L.) Heynh. cv. Columbia roots was undertaken. Two classes of K+-selective channels were frequently observed: one inward (IRC) and one outward (ORC) rectifying with unitary conductances of 5 pS, 20 pS (IRCs) and 15 pS (ORC), measured in symmetrical 10 mM KCl. The dominant IRC (5 pS) and ORC (15 pS) were highly cation-selective (PCl PK < 0.025) but less selective amongst monovalent cations (PNaPK0.17–0.3). Both the IRC and the ORC were blocked by Ba2+, Cs+ and tetra-ethyl-ammonium, whereas 4-aminopyridine and quinidine selectively inhibited the ORC. The ORC open probability was steeply voltage-dependent and ORC activation potentials were close to the potassium equilibrium potential (EK+), enabling ORCs to conduct mainly outward, but occasionally inward, K+ current. By contrast, gating of the 5-pS IRC was weakly voltageependent and IRC gating was invariably restricted to membrane potentials more negative than EK+, ensuring K+ transport was always inwardly directed. Studies on channel activity were conducted for a large number of root cells grown at two levels of external [K+], one where K+ uptake is likely to be principally through channels (6 mM K+) and one where it must be energised (100 M K+). Shifting growth conditions from high to low K+ did not affect single-channel properties such as conductance and selectivity, nor the manifestation of the ORC and 20-pS IRC, but led to enhanced activity of the 5-pS IRC. The concentration dependence of the influx ofl-lysine in excised roots ofArabidopsis thaliana seedlings was analyzed for the wild-type (WT) and two mutants,rlt11 andraec1, which had been selected as resistant to lysine plus threonine, and to S-2-aminoethyl-l-cysteine, respectively. In the WT three components were resolved: (i) a high-affinity, low-capacity component [K m = 2.2 M;V max = 23 nmol·(g FW)–1·h–1]; (ii) a low-affinity, high-capacity component [K m = 159 M;V max = 742 nmol·(g FW)–1·h–1]; (iii) a component which is proportional to the external concentration, with a constant of proportionalityk = 104 nmol·(g FW)–1 h–1];·mM–1. The influx ofl-lysine in the mutants was lower than in the WT, notably in the concentration range 0.1–0.4 mM, where it was only 7% of that in the WT. In both mutants the reduced influx could be fully attributed to the absence of the low-affinity (high-K m ) component. This component most likely represents the activity of a specific basic-amino-acid transporter, since it was inhibited by several other basic amino acids (arginine, ornithine, hydroxylysine, aminoethylcysteine) but not byl-valine. The high-affinity uptake ofl-lysine may be due to the activity of at least two general amino acid transporters, as it was inhibitable byl-valine, and could be further dissected into two components with a high affinity (K i = 1–5 M; and a low affinity (K i = 0.5–1mM) forl-valine, respectively. Summary The lachrymal salt glands of hatchlings of the green sea turtle (Chelonia mydas) secrete a hyperosmotic (up to 2000 mosmol·kg–1) NaCl solution. X-ray microanalysis of frozen-hydrated glands showed that during secretion intracellular Na+ concentration in the principal cells increased from 13 to 34 mmol·l–1 of cell water, whilst Cl and K+ concentrations remained unchanged at 81 mmol·l–1 and 160–174 mmol·l–1, respectively. The high Cl concentration and the change in Na+ concentration are consistent with the prevailing paradigm for secretion by the structurally and functionally similar elasmobranch rectal gland. Concentrations of Na+, Cl and K+ in the lumina of secretory tubules of secreting (Na+ 122, Cl 167, K+ 38 mmol·l–1) and non-secreting (Na+ 114, Cl–1 174, K+ 44 mmol·l–1) glands were similar and the fluid was calculated to be approximately isosmotic with blood. In the central canals Na+ and Cl concentrations were similar but K+ concentration was lower (11–15 mmol·l–1). It is concluded that either a high transepithelial NaCl gradient in secretory tubules and central canals is very rapidly dissipated during the short time between gland excision and freezing, or that ductal modification of an initial isosmotic secretion occurs.  相似文献

Maceration with pectinase (4.5h) of Commelina benghalensis L. leaves stripped at either side yielded isolated vein networks consisting of four to five secondary veins and tertiary cross veins (=minor veins). Examination with Evans Blue and injection of Fluorescein F showed that 80% of the veins were viable. Proof of normal functioning of isolated minor veins was that [14C]sucrose fed to an apical vein network attached to the remaining intact part of the leaf was absorbed and finally arrived in the petiole. Sucrose uptake by veins obeyed Michaelis-Menten kinetics (K m 5·10-4 mol l-1; V max (light) 3.2 mol h-1 g-1 fresh weight, V max (dark) 1.5 mol h-1 g-1 fresh weight). A linear component, not inhibited by carbonylcyanide m-chlorophenylhydrazone and p-chloromercuribenzenesulfonic acid, was present. Maximal uptake took place at 5 mmol l-1 K+; concentrations of K+ higher than 10 mmol l-1 decreased the rate of uptake. The uptake rates by isolated veins and veins in situ (in disks) were in the same order of magnitude. Despite the large number of studies of nitrate metabolism in plants, it remains undetermined to what extent this key plant system is controlled by overall plant N nutrition on the one hand, and by the nitrate ion itself on the other hand. To investigate these questions, V max for nitrate uptake (high-affinity range), and nitrate reductase (NR) mRNA and activity, were measured in roots of N-limited barley (Hordeum vulgare L. cv. Golf) grown under conditions of constant relative addition of nitrate, with the seminal roots split between two culture compartments. The total amount of nitrate added per unit time (0.09·d-1) was distributed between the two root parts (subroots) in ratios of 1000, 982, 955, 9010, 8020, and 5050. These nitrate-addition ratios resulted in nitrate fluxes ranging from 0 to 23 mol nitrate·g-1 DW root·h-1, while the external nitrate concentrations varied between 0 and 1.2 M. The apparent V max for net nitrate uptake showed saturation-type responses to nitrate flux maintained during preceding growth. The flux resulting in half-maximal induction of nitrate uptake was approximately 4 mol nitrate·g-1 DW root·h-1, corresponding to an external nitrate concentration of 0.7 M. The activity of NR and levels of NR mRNA did not saturate within the range of nitrate fluxes studied. None of the parameters studied saturated with respect to the steady-state external nitrate concentration. At the zero nitrate addition — the 0%-root — initial uptake activity as determined in short-term 15N-labelling experiments was insignificant, and NR activity and NR mRNA were not detectable. However, nitrate uptake was rapidly induced, showing that the 0%-root had retained the capacity to respond to nitrate. We examined transepithelial transport of Ca2+ across the isolated opercular epithelium of the euryhaline killifish adapted to fresh water. The opercular epithelium, mounted in vitro with saline on the serosal side and fresh water (0.1 mmol·l–1 Ca2+) bathing the mucosal side, actively transported Ca2+ in the uptake direction; net flux averaged 20–30 nmol·cm–2·h–1. The rate of Ca2+ uptake varied linearly with the density of mitochondria-rich cells in the preparations. Ca2+ uptake was saturable, apparent K 1/2 of 0.348 mmol·l–1, indicative of a multistep transcellular pathway. Ca2+ uptake was inhibited partially by apically added 0.1 mmol·l–1 La3+ and 1.0 mmol·l–1 Mg2+. Addition of dibutyryl-cyclic adenosine monophosphate (0.5 mmol·l–1)+0.1 mmol·l–1 3-isobutyl-l-methylxanthine inhibited Ca2+ uptake by 54%, but epinephrine, clonidine and isoproterenol were without effect. Agents that increase intracellular Ca2+, thapsigargin (1.0 mol·l–1, serosal side), ionomycin (1.0 mol·l–1, serosal side) and the calmodulin blocker trifluoperazine (50 mol·l–1, mucosal side) all partially inhibited Ca2+ uptake. In contrast, apically added ionomycin increased mucosal to serosal unidirectional Ca2+ flux, indicating Ca2+ entry across the apical membrane is rate limiting in the transport. Verapamil (10–100 mol·l–1, mucosal side), a Ca2+ channel blocker, had no effect. Results are consistent with a model of Ca2+ uptake by mitochondria rich cells that involves passive Ca2+ entry across the apical membrane via verapamil-insensitive Ca2+ channels, intracellular complexing of Ca2+ by calmodulin and basolateral exit via an active transport process. Alkaloid uptake into vacuoles isolated from a Fumaria capreolata L. cell suspension culture was investigated. The uptake is carrier-mediated as shown by its substrate saturation, its sensitivity to metabolic inhibitors and especially by its exclusive preference for the (S)-forms of reticuline and scoulerine while the (R)-enantiomers which do not occur in this plant species were strictly discriminated. The carrier has a high affinity for (S)-reticuline with a K m=0.3 M. The rate of alkaloid uptake was 6 pmol·h-1·l-1 vacuole, and 0.03 mg alkaloid·mg-1 vacuolar protein were taken up. Transport was stimulated five-to seven-fold by ATP and was inhibited by the ATPase inhibitors N,N-dicyclohexylcarbodiimide and 4-4-diisothiocyanatostilbene-2,2 disulfonic acid, as well as by the protonophore carbonyl cyanide m-chlorophenylhydrazone. A number of alkaloids did not compete with labelled (S)-reticuline for uptake into vacuoles. The uptake system is absolutely specific for alkaloids indigenous to the plant from which the vacuoles were isolated. Slight modifications of the topography of an alkaloid molecule even with full retention of its electrical charge results in its exclusion. Alkaloid efflux was also shown to be mediated by a highly specific energy-dependent carrier. These results contradict the previously proposed ion-trap mechanism for alkaloid accumulation in vacuoles. The renal response to infusion of three different saline solutions was studied in chicks of Leach's storm petrel (Oceanodroma leucorhoa). Each of the solutions (125 mmol·1-1 NaCl at 5.3 ml·h-1, 250 mmol·l-1 NaCl at 2.6 ml·h-1, and 550 mmol·l-1 NaCl at 1.2 ml·h-1) provided the same delivery of Nacl but in different volumes of water. Birds infused with 125 mmol·l-1 NaCl had a glomerular filtration rate of 25.7 ml·h-1, a urine flow rate of 4.4 ml·h-1, and excreted 71% of the infused Na+ load in the urine. With infusion of 250 mmol·l-1 NaCl, the glomerular filtration rate was unchanged (23.3 ml·h-1), but urine flow rate was reduced to 0.93 ml·h-1 and only 35% of the Na+ load was excreted in the urine. Infusion of 550 mmol·l-1 NaCl induced a sharp decrease in glomerular filtration rate (to 3.8 ml·h-1) and urine flow rate (to 0.08 ml·h-1), and only 1.4% of the infused Na+ was excreted in the urine. The contribution of different nephron populations to filtration was assessed by the pattern of staining of glomeruli by alcian blue infused during the last 30 min of the saline infusion. The numbers of stained glomeruli did not differ between birds infused with 125 and 250 mmol·l-1 NaCl (59000 and 55000 glomeruli per kidney, respectively), and the patterns of staining were similar for birds in these two groups. Birds infused with 550 mmol·l-1 NaCl had lighter staining overall and fewer stained glomeruli (37000 per kidney). This absence of staining was predominant in the smaller size classes of glomeruli, suggesting a selective shutdown of smaller (reptilian-type) nephrons during times of osmotic challenge in these birds. This work was devoted to the study of the structure-affinity relationships in neutral amino acid transport by intestinal brush border of marine fish (Dicentrarchus labrax). The effects of the length of the side chain on kinetics of glycine, alanine, methionine and amino isobutyric acid were investigated. In the presence of K+ two components were characterized: one is saturable by increased substrate concentrations, whereas the other can be described by simple diffusion mechanism. Simple diffusion, a passive, non-saturable, Na+-independent route, contributes largely to the transport of methionine and to a much lesser extend to alanine, glycine or alphaaminoisobutyric acid uptakes. If a branched chain is present, as in the case of amino isobutyric acid, diffusion is low. A Na+-independent, saturable system has been fully characterized for methionine, but not for branched amino acids such as amino isobutyric acid. In the presence of Na+ saturable components were shown. Two distinct Na+-dependent pathways have been characterized for glycine uptake, with low and high affinities. For alanine and methionine only one Na+-dependent high affinity system exists with the same half-saturation concentration and the same maximum uptake at saturable concentrations. Glycine high affinity system has the same half-saturation concentration as methionine or alanine uptake, whereas maximum uptake is lower. The substitution of the hydrogen by a methyl group results in a severe decrease of uptake (aminoisobutyric acid). Mutual inhibition experiments indicate that the same carriers could be responsible for methionine and alanine uptakes and probably glycine Na+-dependent uptake. The influence of Na+ concentrations (100-1 mol·l-1) on amino acid uptake was examined. Glycine, alanine, methionine and amino isobutyric acid transport can be described by a hyperbolic function, with a saturation uptake which is highly increased for methionine. However, the half-saturation concentration does not seem to be strongly affected by the amino acid structure. The effect of Na+ concentration (25 and 100 mmol·l-1) on the kinetics of methionine uptake have been also examined.
