Aminoacid composition and hydrophobicity index of mitochondrial polypeptides

The aminoacid composition of protein stained bands in polyacrylamide gels, after electrophoresis of proteins from inner mitochondrial membranes, was investigated hydrolyzing directly the gel slices. The Hydrophobicity Index of 18 prominent polypeptide bands was calculated after their aminoacid analysis. The polypeptides less related to the membrane have low hydrophobicity as inferred from their Hydrophobicity Indexes.     

Effect of Sclerospora graminicola infection on total nitrogen content and free aminoacid composition of bajra plants

Summary Studies on the total nitrogen content and free aminoacid composition of healthy Bajra (Pennisetum typhoides Stapf &Hubb.) plants and those infected withSclerospora graminicola (Sacc.)Schroet. were made. Total nitrogen content of the diseased tissues increased as a result of infection. Considerable difference existed in free aminoacid composition of healthy and diseased tissues. Tryptophane was one of the aminoacids which was constantly isolated from the extracts of diseased tissues only.     

Hb Sinai,a new α chain mutant a 47 His

Summary A new chain mutant Hb-Sinai ₄₇ ^His is described. The aminoacid composition of all tryptic peptides has been determined, with the exception of the insoluble core. In the fingerprint peptide T 6 which normally migrates between T 3 and T 1, moves now between T 3 and T 7. The aminoacid composition of peptide T 6 indicates a change in the aminoacid composition from Asp- to His⁺ in position 47.This work was supported by Grant No. GM 13714 U.S.P.H.S.     

NH2-terminal sequence of calf fetuin

Calf fetuin, one of the 3 major known fetal proteins has been isolated by a two-step purification procedure and characterized by aminoacid composition. The purified glycoprotein, which consisted of a single chain, was submitted to 47 steps of automatic aminoacid sequencing, allowing to determine 44 positions. This section of the molecule was devoid of carbohydrates. Comparison of this sequence with a variety of detectable potentially related protein did not allow to point to any detectable homology.     

Various physico-chemical properties of the Yersinia pestis capsular protein

Some properties of the structure of Y. pestis capsular antigen macromolecules have been studied. The aminoacid composition of F1 protein, the aminoacid sequence of the N-terminal fragment of antigen polipeptide chain were determined. Some peculiarities in the dissociation of capsular antigen macromolecules have been studied. The formation of the product resulting from unterminated thermodissociation of F1 protein oligomeric form, consisting of four subunits, has been registered. The aspects of F1 protein association are discussed.     

Preparation and immunoephelometric quantitation of fibrinogen in rabbits]

Pure rabbit fibrinogen was prepared by a method involving two ammonium sulfate precipitations, one 2 M phosphate buffer precipitation, one DEAE cellulose chromatography and lastly one Sepharose 6 B chromatography. The aminoacid composition was determined and an immunonephelemetric assay was proposed. This assay followed an accurate determination of fibrinogen concentration in a rabbit with inflammatory reaction.

Characterization of enzymatic deficiencies of branched chain amino-acid catabolism in human fibroblasts by genetic complementation

Leucine and Isoleucine metabolism in cultured skin fibroblasts from patients with leucinosis, beta-Ketothiolase deficiency, propionic, methylmalonic and isovaleric acidemia, was compared with that in normal fibroblasts. A simple assay was developed using (U14C) Isoleucine and (U14C) Leucine as substrates. Radioactive incorporation into protein aminoacids were measured. The (U14C) branched chain aminoacid incorporation into proteins provides an estimation of the protein synthesis and the incorporation ratio (14C) Aspartate + (14C) Glutamate/(14C) branched chain aminoacid, measures the integrity of the metabolic pathway. Complementation tests permits to characterize the genetic defect. The incorporation ratio was significantly decreased in blocked pathways, namely in leucinosis and isovaleric acidemia in the presence of (U14C) Leucine and in Leucinosis, beta-Ketothiolase deficiency, propionic and methylmalonic acidemia in the presence of (U14C) Isoleucine. There was a significant restoration of activity in mutant strains with distinct genetic defects after polyethylene-glycol fusion. This assay provides a valuable tool to screen for enzymatic deficiencies of branched chain aminoacid catabolism.     

Differential D-glucose requirements of the general amino-acid permease and protein synthesis in Saccharomyces cerevisiae var. ellipsoideus

The dependence of the general aminoacid permease and protein synthesis on the availability of D-glucose as energy source was studied. Stimulation by the sugar was immediate once added to the cell suspensions and seems to be mediated by energy derived directly from glycolysis. The general aminoacid permease was saturated linearly with D-glucose whereas protein synthesis was saturated sigmoidealy requiring much higher concentration of the sugar than the general aminoacid permease.     

氨基酸组成聚类、蛋白质结构型和结构型的预测

用信息聚类方法对蛋白质的氨基酸组成进行聚类,发现存在梯级成团（大集团分解成小集团）现象,６４５个蛋白质可分成１５个小集团,每一个小集团与蛋白质二级结构含量决定的结构型有一定相关性,但与蛋白质五大结构型相关性不明显。指出了由氨基酸成分和二级结构含量预测结构型的方案中存在的问题。提出了由蛋白质二级结构序列预测蛋白质结构型的新方法,并给出了预测蛋白质结构型的简明预测规则     

落叶果树花粉中的氨基酸含量及其评价

<正> 蜜源植物蜂花粉,在营养学上被称作“蛋白质和氨基酸的浓缩体”。落叶果树中的苹果花粉,也被确认为营养价值极好级别的植物花粉之一。但是,对于落叶果树花粉中的氨基酸含量,树种、品种间的区别,迄今国内外尚少系统研究和报道。为了认识、评价和选择利用落叶果树的花粉资源,于1987～1989年间,对10个落叶果树树种、82个品种花粉中的氨基酸含量,进行了系统的测定、分析。现将结果整理报告于下。     

Isolation and characterization of a lectin from the seeds of Dioclea lehmanni.

Affinity chromatography of the globulin fraction from the seeds of Dioclea lehmanni on Sephacryl S-200 yielded two lectins, one slightly retarded and another strongly bound. The latter, which was a glucose/mannose specific lectin, was purified and the following properties were determined: pI, Mr of subunits, carbohydrate content, A, aminoacid composition, hemagglutination and inhibition patterns, N-terminal sequence and mitogenic activity. These properties of the lectin were very similar to those of the Con A and Dioclea grandiflora lectins.     

Collagen from human joint cartilage in deforming arthritis

Type II collagen of human cartilage has been comparatively studied in norm and at deforming arthrosis. Collagen preparations are characterized by aminoacid composition and electrophoretic mobility in polyacrylamide gel. It is supposed that at deforming arthrosis the degenerative-dystrophic processes in the articular cartilage of man are caused by appearance of procollagen.     

SECRETOOL: integrated secretome analysis tool for fungi

The secretome (full set of secreted proteins) has been studied in multiple fungal genomes to elucidate the potential role of those protein collections involved in a number of metabolic processes from host infection to wood degradation. Being aminoacid composition a key factor to recognize secretory proteins, SECRETOOL comprises a group of web tools that enable secretome predictions out of aminoacid sequence files, up to complete fungal proteomes, in one step. Structure of the human alpha 1-acid glycoprotein gene: sequence homology with other human acute phase protein genes.

We have determined the sequence coding for human alpha 1-acid glycoprotein from two independently isolated cDNA clones and a genomic clone. The aminoacid sequences deduced from the three clones, deriving from three different individuals, are identical. Southern blot analysis on human DNA indicates that there are at least two genes coding for alpha 1-AGP. We propose that alpha 1-AGP found in plasma is a mixture of the products of these two different genes. This is the simpler explanation for the heterogeneity in the aminoacid composition in purified alpha 1-AGP observed by Schmid et al. (1). DNA sequence comparison with cDNA clones coding for human alpha 1-antitrypsin and haptoglobin shows a conserved sequence within the 5' untranslated region which may play a role in the acute phase response.     

Neocarzinostatin: spectral characterization and separation of a non-protein chromophore.

Chromatographically purified neocarzinostatin exhibits absorption, fluorescence, magnetic circular dichroic and circular dichroic spectral characteristics above and below 300 nm atypical for a protein with its reported aminoacid composition, indicating the presence of a non-protein chromophore. The drug complex, stable at acidic pH, can be dissociated by treatment with reducing or denaturing agents at neutral or basic pH. Chromatography of the dissociated complex, or more conveniently, methanol extraction of the lyophilized drug, separates a protein with an amino-acid composition identical to neocarzinostatin and a highly fluorescent chromophore free of amino-acids.     

Addition of amino acids and oligopeptides to the diet of piglets. I. Effects on productive performance and development of intestinal microbial flora

The effects in connection with the addition of small quantities of aminoacid and oligopeptide pools to balanced diets were studied on 36 male piglets, divided into three groups (1: control; 2: aminoacid pool; 3: aminoacid and oligopeptide pool). Piglets receiving the aminoacid pool (group 2) exhibited better performances: an increased weight gain resulting in higher feed efficiency and an increased overall microbial count with a predominance of the useful population.     

Effect of different oxygen pressure levels on the transport of alpha-aminoisobutyric acid in myocardial cell cultures

The activity of aminoacid transport, as measured by alpha-aminoisobutyrate uptake, has been studied in confluent myocardial cell cultures exposed to different oxygen tensions. The results obtained indicate that the rate of cellular uptake and accumulation of the inert aminoacid increase with time as the fraction of oxygen is reduced. When alpha-aminoisobutyrate was added in presence of all other aminoacids of the medium, the effect of oxygen was also evident, suggesting a mechanism which overcomes the competitive action of the other aminoacids assigned to the same transport system of alpha-aminoisobutyrate (A system). The modulation of aminoacid transport activity may represent one of the possible mechanisms by which environmental oxygen affect the rate of cellular protein synthesis.     

Further characterization of the orange carotenoid protein extracted from the marine demosponge Axinella verrucosa

The orange coloration of the marine sponge A. verrucosa is provided by some carotenoids widespread in the ectosome and mesohyl of this sponge. These carotenoids are bound to a glyco(lipo)protein forming a non covalent complex. Six carotenoids are bound to the glyco(lipo)protein, but only alpha-carotene was identified by HPLC. The aminoacid composition is quite different from those previously reported in Porifera. The content of Ser and Gly and the total polar residues are high. The presence of Met and Pro was not evidenced. Some unusual aminoacids were detected, of which only Taurine was probably identified.
