Changes in the temperature dependence of the photosynthetic rate depending on growth temperature were investigated for a temperate evergreen tree, Quercus myrsinaefolia . Plants were grown at 250 μ mol quanta m–2 s–1 under two temperature conditions, 15 and 30 °C. The optimal temperature that maximizes the light-saturated rate of photosynthesis at 350 μ L L–1 CO2 was found to be 20–25 and 30–35 °C for leaves grown at 15 and 30 °C, respectively. We focused on two processes, carboxylation and regeneration of ribulose-1,5-bisphosphate (RuBP), which potentially limit photosynthetic rates. Because the former process is known to limit photosynthesis at lower CO2 concentrations while the latter limits it at higher CO2 concentrations, we determined the temperature dependence of the photosynthetic rate at 200 and 1000 μ L L–1 CO2 under saturated light. It was revealed that the temperature dependence of both processes varied depending on the growth temperature. Using a biochemical model, we estimated the capacity of the two processes at various temperatures under ambient CO2 concentration. It was suggested that, in leaves grown at low temperature (15 °C), the photosynthetic rate was limited solely by RuBP carboxylation under any temperature. On the other hand, it was suggested that, in leaves grown at high temperature (30 °C), the photosynthetic rate was limited by RuBP regeneration below 22 °C, but limited by RuBP carboxylation above 22 °C. We concluded that: (1) the changes in the temperature dependence of carboxylation and regeneration of RuBP and (2) the changes in the balance of these two processes altered the temperature dependence of the photosynthetic rate.  相似文献

The major β-1,4-endoglucanase (EG) of the thermophilic actinomycete, Thermomonospora curvata , contributed over 80% of the total EG activity recovered from cell-free culture fluid after growth on cellulose. The enzyme was purified to electrophoretic homogeneity by ammonium sulphate precipitation, ion-exchange chromatography and size exclusion HPLC. This monomeric enzyme had a specific activity of 750 IU mg−1 when assayed with 2.5% (w/v) carboxymethyl cellulose (CMC) at 70°C, pH 6.0. Highest activity was observed on CMC with a degree of polymerization of 3200. The EG was stable for 48 h at 60°C, pH 6.0 and had a half-life of 30 min at 80°C; temperature and pH optima were 70–73°C and 6.0–6.5, respectively. The mol. wt was 100000 and the pI was 4.0. The K m and V max values were 7.33 mg ml−1 and 833 μmol min−1, respectively. EG activity was inhibited by Fe2 +, Hg2 +, Ag+ and Pb2 +, and enhanced by dithiothreitol and Zn2 +. The first 12 amino acid residues at the N -terminus were: Asp-Glu-Val-Asp-Glu-Ile-Arg-Asn-Gly-Asp-Phe-Ser. Glutamic and aspartic acid constituted 24% of the total amino acid composition; no amino sugar was found.  相似文献

Soybean ( Glycine max cv. Clark) was grown at both ambient (ca 350 μmol mol−1) and elevated (ca 700 μmol mol−1) CO2 concentration at 5 growth temperatures (constant day/night temperatures of 20, 25, 30, 35 and 40°C) for 17–22 days after sowing to determine the interaction between temperature and CO2 concentration on photosynthesis (measured as A, the rate of CO2 assimilation per unit leaf area) at both the single leaf and whole plant level. Single leaves of soybean demonstrated increasingly greater stimulation of A at elevated CO2 as temperature increased from 25 to 35°C (i.e. optimal growth rates). At 40°C, primary leaves failed to develop and plants eventually died. In contrast, for both whole plant A and total biomass production, increasing temperature resulted in less stimulation by elevated CO2 concentration. For whole plants, increased CO2 stimulated leaf area more as growth temperature increased. Differences between the response of A to elevated CO2 for single leaves and whole plants may be related to increased self-shading experienced by whole plants at elevated CO2 as temperature increased. Results from the present study suggest that self-shading could limit the response of CO2 assimilation rate and the growth response of soybean plants if temperature and CO2 increase concurrently, and illustrate that light may be an important consideration in predicting the relative stimulation of photosynthesis by elevated CO2 at the whole plant level.  相似文献

Rice ( Oryza sativa L. cv. IR72) was grown at three different CO2 concentrations (ambient, ambient + 200 μmol mol−1, ambient + 300 μmol mol−1) at two different growth temperatures (ambient, ambient + 4°C) from sowing to maturity to determine longterm photosynthetic acclimation to elevated CO2 with and without increasing temperature. Single leaves of rice showed a cooperative enhancement of photosynthetic rate with elevated CO2 and temperature during tillering, relative to the elevated CO2 condition alone. However, after flowering, the degree of photosynthetic stimulation by elevated CO2 was reduced for the ambient + 4°C treatment. This increasing insensitivity to CO2 appeared to be accompanied by a reduction in ribulose-1.5-bisphosphate carboxylase/oxygenase (Rubisco) activity and/or concentration as evidenced by the reduction in the assimilation (A) to internal CO2 (C1) response curve. The reproductive response (e.g. percent filled grains, panicle weight) was reduced at the higher growth temperature and presumably reflects a greater increase in floral sterility. Results indicate that while CO2 and temperature could act synergistically at the biochemical level, the direct effect of temperature on floral development with a subsequent reduction in carbon utilization may change sink strength so as to limit photosynthetic stimulation by elevated CO2 concentration.  相似文献

Abstract Acremonium persicinum grown in batch culture with ammonium tartrate as the nitrogen source possessed an NADP+-dependent glutamate dehydrogenase and a glutamine synthetase. Glutamate synthase was not detected under the culture conditions used. Kinetic studies of the NADP+-dependent glutamate dehydrogenase at 25°C and pH 7.6 revealed an apparent K m of 3.2 × 10−4 M for 2-oxoglutarate and an apparent K m of 1.0 × 10−5 M for ammonium ions, with corresponding apparent V max values of 0.089 and 0.13 μmol substrate converted/min/mg of protein, respectively. Glutamine synthetase was measured by the γ-glutamyl transferase reaction at 30°C and pH 7.55. This transferase reaction of glutamine synthetase had a higher rate at 30°C than at 25°C or 37°C.  相似文献

Nickel and rubidium uptake by oat plants ( Avena sativa L. cv. Victory) were examined in relation to solution temperature, solution concentrations, metabolic inhibitors, anaerobic root conditions, transpiration and time. Over a 4-h period, uptake rates for both Ni2+ and Rb+ remained constant at 23°C. Decreasing temperatures to 2°C, 20 μ M concentrations of 2,4-dinitrophenol (DNP), or anaerobic root conditions decreased Ni2+ and Rb+ uptake rates by 97 to 86% in whole plants. Treatment of excised roots with 20 μ M DNP decreased Ni2+ uptake by 93%. Nickel and Rb+ uptake rates measured as a function of the external solution concentration followed a typical parabolic curve. Km (0.012 m M ) and Vmax [2.72 μmol (g dry weight)-1 h-1] values for Ni2+ were nearly 7 times lower than those for Rb+ [0.09 m M and 19.2 μmol (g dry weight)-1 h-1]. In all experiments, Ni2+ and Rb+ showed qualitatively similar uptake patterns, but Rb+ uptake was quantitatively more sensitive than Ni2+ to experimental manipulations.  相似文献

The effects of photon flux density and temperature on net photosynthesis and transpiration rates of mature and immature leaves of three-year-old Japanese larch Larix kaempferi (Lamb.) Sarg. trees were determined with an infrared, differential open gas analysis system. Net photosynthetic response to increasing photon flux densities was similar for different foliage positions and stage of maturity. Light compensation was between 25 and 50 μmol m−2 s−1. Rates of photosynthesis increased rapidly at photon flux densities above the compensation level and became saturated between 800 and 1000 μmol m−2 s−1. Transpiration rates at constant temperature likewise increased with increasing photon flux density, and leveled off between 800 and 1000 μmol m−2 s−1. Photosynthetic response to temperature was determined in saturating light and was similar for all foliage positions; it increased steadily from low temperatures to an optimum range betweeen 15 and 21°C and then decreased rapidly above 21°C. Transpiration rate, however, increased continuously with rising temperature up to the experimental maximum. CO2 compensation concentrations for mature foliage varied between 58 and 59 μl l−1; however, foliage borne at the apex of the terminal leader compensated at 75 μl l−1. None of these data support the claim that Japanese larch possesses C4 photosynthetic characteristics.  相似文献

Ananas comosus L. (Merr.) (pineapple) was grown at three day/night temperatures and 350 (ambient) and 700 (elevated) μ mol mol–1 CO2 to examine the interactive effects of these factors on leaf gas exchange and stable carbon isotope discrimination ( Δ ,‰). All data were collected on the youngest mature leaf for 24 h every 6 weeks. CO2 uptake (mmol m–2 d–1) at ambient and elevated CO2, respectively, were 306 and 352 at 30/20 °C, 175 and 346 at 30/25 °C and 187 and 343 at 35/25 °C. CO2 enrichment enhanced CO2 uptake substantially in the day in all environments. Uptake at night at elevated CO2, relative to that at ambient CO2, was unchanged at 30/20 °C, but was 80% higher at 30/25 °C and 44% higher at 35/25 °C suggesting that phosphoenolpyruvate carboxylase was not CO2-saturated at ambient CO2 levels and a 25 °C night temperature. Photosynthetic water use efficiency (WUE) was higher at elevated than at ambient CO2. Leaf Δ -values were higher at elevated than at ambient CO2 due to relatively higher assimilation in the light. Leaf Δ was significantly and linearly related to the fraction of total CO2 assimilated at night. The data suggest that a simultaneous increase in CO2 level and temperature associated with global warming would enhance carbon assimilation, increase WUE, and reduce the temperature dependence of CO2 uptake by A. comosus .  相似文献

1. Hyalella montezuma is endemic to Montezuma Well, Arizona, and is exposed to minimal diel and seasonal temperature fluctuations in the pelagic zone (21 ± 4 °C). Juvenile H . montezuma feed in the pelagic zone during the day and migrate into the littoral vegetation at night, while adults remain primarily in the littoral vegetation.
2. Oxygen consumption ( V O2) of adult and juvenile H . montezuma was measured at 20, 25 and 30 °C. The V O2 of both adult and juvenile H . montezuma increased with temperature. However, the V O2 of juveniles was significantly greater than that of adults at all temperatures, with greatest divergence at 30 °C where mean juvenile V O2 (6.31 μl mg–1 dry weight (DW) h–1) was almost twice that of adults (3.60 μl mg–1 DW h–1).
3. Survivorship of juveniles was significantly lower (54%) at 30 °C than at 27.5 °C (95%) after 4 h, whereas adults showed at least a 93% survivorship at both temperatures.
4. Our data suggest that temperature may have been the proximate cue that elicited the diel horizontal migration of juvenile H . montezuma in Montezuma Well, with the behaviour maintained and enhanced by intensive invertebrate predation in the pelagic and littoral zones.  相似文献

Photosynthetic characteristics of leaves of tef, Eragrostis tef (Zucc.) Trotter, plants, grown at 25/15°C (day/night), were measured at temperatures from 18 to 48°C. The highest carbon exchange rates (CER) occurred between 36 and 42°C. and averaged 27 μmol m−2 s−1. At lower or higher temperatures, CER was reduced, but the availability of CO2 to the mesophyll, measured as internal CO2 concentration, was highest when temperatures were above or below the optimum for CER. In addition, CER and stomatal conductance were not correlated, but residual conductance was highly correlated with CER (r = 0.98). In additional experiments, relative 13C composition for leaf tissue grown at 25, 35 and 45°C averaged -14.4 per mille, confirming that tef is a C4 grass species. Dry matter accumulation was higher at 35 than at 25, and lowest at 45°C. Leaf CER rates increased hyperbolically with increased light when measured from 0 to 2000 μmol m−2 s−1 PPFD. The highest CER, 31.8 μ-mol m-2 s−1, occurred at 35°C and 2000 μmol m−2 s−1 PPFR. At high light, CER at 25 and 35°C were nearly equal because of higher stomatal conductance at 25°C. Residual conductance was, however, clearly highest at 35°C compared to 25 and 45°C treatments. Stomatal conductance and residual conductance were not correlated in either set of experiments, yet residual conductance was always highest when temperatures were between 35 and 42°C across experiments, suggesting that internal leaf photosynthetic potential was highest across that temperature range.  相似文献

The induction of freezing tolerance by abscisic acid (ABA) or cold treatment in suspension cultured cells of Solanum commersonii was studied. Both ABA (50–100 μ M ) at 23°C and low temperature (4°C) increased freezing tolerance in cultured Solanum commersonii cells from a LT50 (freezing temperature at which 50% cells were killed) of —5°C (control) to —11.5°C in 2 days. Cold-induced freezing tolerance reached its maximum at 2 days and remained constant throughout the cold acclimation period of 11 days. The freezing tolerance induced by ABA, however, showed a rapid decline 2 to 5 days after initiation of ABA treatments. Addition of ABA (100 μ M ) to the culture medium at the inception of low temperature treatment did not enhance freezing tolerance of the cells beyond the level attainable by either treatment singly. Poly(A+)-RNA was isolated from the respective treatments, translated in a rabbit reticulocyte lysate cell free system, and the translation products were resolved by two dimensional polyacrylamide gel electrophoresis (ID-PAGE). Analysis of the in vitro translated products revealed changes in the abundance of approximately 26 products (encoding for polypeptides with M, of 14 to 69 kDa and pl of 4.90 to 6.60) in ABA-treated cells 12 h after treatment, and 20 (encoding for polypeptides with Mr of 12 to 69 kDa, with pl of 4.80 to 6.42) in cells exposed to 4°C for 12 h. There were only 5 novel translation products observed when the ABA-treated cells reached the highest level of freezing tolerance (2 days after the initiation of ABA treatment). Changes in translatable RNA populations during the induction of freezing tolerance in cells treated with either ABA or low temperature are discussed.  相似文献

The floating angiosperm Lemna gibba L. was exposed for 2 h to various combinations of photosynthetic photon flux densities and temperature. The extent of photoinhibition of photosynthesis was assayed by measuring the net CO2 uptake before and after a photoinhibitory treatment, and the time course for photoinhibition was studied. It was found that the maximum quantum yield and the light-saturated rate of CO2 uptake were affected by the interaction between light and temperature during the photoinhibitory treatment. At a constant photon flux density of 650 μmol m−2 s−1 the extent of photoinhibition increased with decreasing temperature showing that even a chilling-resistant plant like L. gibba is much more susceptible to photoinhibition at chilling temperatures. About 60% photoinhibition of the quantum yield for CO2 uptake could be obtained either by a high photon flux density of 1 750 μmol m−2 s−1 and 25°C or by a moderate photon flux density of 650 μmol m−2 s−1 and 3°C. The time courses of recovery from 60% photoinhibition produced by either of these two treatments were similar, indicating that the nature of the photoinhibition was intrinsically similar. The extent of photoinhibition was related to the amount of light absorbed in excess to what could be handled by photosynthesis at that temperature. The vital importance of photosynthesis in alleviating photoinhibition is discussed.  相似文献

This paper describes a method for the isolation of highly purified sarcoplasmic reticulum from plaice fast muscle. The interrelationships of pH, KCL, Ca2+, Mg2+, ADP and temperature have been investigated. Protein composition of plaice white muscle sarcoplasmic reticulum was found to be comparable to that described for rabbit fast muscle, with a major component of 100 000 daltons. Arrhenius plots of the Ca2+-AT Pase are linear over the range 0–30°C. Activation enthalpy (60±1.5 kJ/mol) was found to be independent of KCl concentration. The calcium concentration required to give half maximal activation of the AT Pase (KCa) was found to decrease with increasing temperature, from a maximum of 1.7 μm at 0°C to 0.55 at 20°C.  相似文献

The objective of the study was to investigate the interactive effects of elevated atmospheric carbon dioxide concentration, [CO2], and temperature on the wood properties of mature field-grown Norway spruce ( Picea abies (L.) Karst.) trees. Material for the study was obtained from an experiment in Flakaliden, northern Sweden, where trees were grown for 3 years in whole-tree chambers at ambient (365 μmol mol−1) or elevated [CO2] (700 μmol mol−1) and ambient or elevated air temperature (ambient +5.6 °C in winter and ambient +2.8 °C in summer). Elevated temperature affected both wood chemical composition and structure, but had no effect on stem radial growth. Elevated temperature decreased the concentrations of acetone-soluble extractives and soluble sugars, while mean and earlywood (EW) cell wall thickness and wood density were increased. Elevated [CO2] had no effect on stem wood chemistry or radial growth. In wood structure, elevated [CO2] decreased EW cell wall thickness and increased tracheid radial diameter in latewood (LW). Some significant interactions between elevated [CO2] and temperature were found in the anatomical and physical properties of stem wood (e.g. microfibril angle, and LW cell wall thickness and density). Our results show that the wood material properties of mature Norway spruce were altered under exposure to elevated [CO2] and temperature, although stem radial growth was not affected by the treatments.  相似文献

Lactobacillus plantarum was found to produce extracellular polygalacturonase (EC 3.2.1.15.). Maximum enzyme production was obtained in a medium containing 0.5% glucose and 1.5% low methyl-pectin as inducer at 27°C at an initial pH of 6.8. Enzyme production was strongly inhibited by 5 μmol/l NiCl2, 5 μmol/l CoCl2, 5 μmol/l CuSO4, and 10 μmol/l ZnCl2. MnSO4 and MgSO4 at 200 μmol/l and 50 μmol/l respectively seemed to enhance enzyme biosynthesis. The optimal pH and temperature for enzyme activity were 4.5 and 30°C respectively. Spirogyra Link (1820) is an anabranched filamentous green alga that forms free-floating mats in shallow waters. It occurs widely in static waters such as ponds and ditches, sheltered littoral areas of lakes, and stow-flowing streams. Field observations of its seasonal distribution suggest that the 70-μm-wide filament form of Spirogyra should have a cool temperature and high irradiance optimum for net photosynthesis. Measurements of net photosynthesis and respiration were marie at 58 combinations of tight and temperature in a controlled environment facility. Optimum conditions were 25°C and 1500 μmol photons m−2 s−1, at which net photosynthesis averaged 75.7 mg O2 gdm−1 h−1. Net photosynthesis was positive at temperatures from 5° to 35°C at most irradiances except at combinations of extremely low irradiances and high temperatures (7 and 23 μmol photons m−2 s−1 at 30°C and 7, 23, and 35 μmol photons m−2 s−1 at 35°C). Respiration rates increased with both temperature and prior irradiance. Light-enhanced respiration rates were significantly greater than dark respiration rates following irradiances of 750 μmol photons m−2 s−1 or greater. Polynomials were fitted to the data to generate response surfaces; such response surfaces can be used to represent net photosynthesis and respiration in ecological models. The data indicate that the alga can tolerate the cool water and high irradiances characteristic of early spring but cannot maintain positive net photosynthesis under conditions of high temperature and low light (e.g. when exposed to self-shading ).  相似文献

The temperature dependence of C3 photosynthesis may be altered by the growth environment. The effects of long-term growth in elevated CO2 on photosynthesis temperature response have been investigated in wheat ( Triticum aestivum L.) grown in controlled chambers with 370 or 700 μmol mol−1 CO2 from sowing through to anthesis. Gas exchange was measured in flag leaves at ear emergence, and the parameters of a biochemical photosynthesis model were determined along with their temperature responses. Elevated CO2 slightly decreased the CO2 compensation point and increased the rate of respiration in the light and ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) Vcmax, although the latter effect was reversed at 15°C. With elevated CO2, Jmax decreased in the 15–25°C temperature range and increased at 30 and 35°C. The temperature response (activation energy) of Vcmax and Jmax increased with growth in elevated CO2. CO2 enrichment decreased the ribulose 1,5-bisphosphate (RuBP)-limited photosynthesis rates at lower temperatures and increased Rubisco- and RuBP-limited rates at higher temperatures. The results show that the photosynthesis temperature response is enhanced by growth in elevated CO2. We conclude that if temperature acclimation and factors such as nutrients or water availability do not modify or negate this enhancement, the effects of future increases in air CO2 on photosynthetic electron transport and Rubisco kinetics may improve the photosynthetic response of wheat to global warming.  相似文献

Oxygen consumption of Oreochromis niloticus at different stages of development was studied in relation to salinity, temperature and time of day, using a Warburg apparatus. The oxygen consumption of newly hatched (0–14 h) larvae was 3.40 μl O2 larva−1 h−1, of older yolk sac larvae 10.09 μl O2 larva−1 h−1, and of one-month-old fry 32.99 μl O2 larva−1 h−1. The QO2 values showed a decrease with development and growth, ranging from 21.2–26.0 μl O2 mg−1 h−1 in newly hatched larvae to 2.97 μl mg−1 h−1 in one-month-old fry. Changes in oxygen consumption occurred with salinity, the highest being at 17%o. Active larvae (12-24 mm T.L.) showed a doubling of consumption with a 10° C rise in temperature, and their Q10 factor increased from 2.25 to 3.43 with increasing size. Day-old yolk-sac larvae, late yolk-sac larvae (5 days old) and fry of 12 14 mm length all showed a depression in oxygen consumption at midnight followed by a dawn rise.  相似文献

Oxygen uptake rates and yolk-inclusive dry weiGhts were measured during the egg and yolk-sac larval stages of milkfish, Chanos chanos (Forsskal). Oxygen uptake by eggs and yolk-sac larvae was measured to assess the effects of four salinities (20,25,30,35 ppt) at 28°C. The effects of three temperatures (23,28,33°C) on oxygen uptake by yolk-sac larvae were determined at a salinity of 35 ppt. Dry weights were measured throughout embryonic development at 28°C and the yolk-sac stage at 23.28 and 33°C.
Oxygen uptake rates of eggs increased more than fivefold during embryogenesis (0.07±0.03 to 0.40 ± 03 μl O2 egg −1 h −1;blastula to prehatch stage). Larval oxygen uptake did not change with age but was affected by rearing temperature (0.33 ± 0.08, 0.44 ± 0.07 and 0.63 ± 0.13 μl O2 larva −1 h−1 at 23, 28 and 33°C, respectively; Q10= 1.93). Acute temperature changes from 28 to 33°C caused significant increases in oxygen uptake by embryos (Q 10= 1.69–3.58) and yolk-sac larvae (Q 10=2.55). Salinity did not affect metabolic rates.
Dry weight of eggs incubated at 28°C decreased 13% from fertilization to hatching. Incubation temperatures from 23–33°C did not affect dry weights at hatching. Rearing temperatures significantly affected the rate of larval yolk absorption (Q 10= 2.25).  相似文献
