Proteolytic activity was demonstrated when a strain of Microsporum canis was cultured in broth with human hair, but not in the medium without hair. The extracellular proteinase (keratinase) was isolated and purified by chromatography. Disc electrophoresis showed one protein band of extracellular proteinase, and the antibody against this enzyme gave a single precipitin line in agar diffusion. The IgG fraction completely neutralized the proteinase activity. The proteinase of M. canis may play a role in infection caused by this fungus, by affecting keratinized tissue such as stratum corneum and hair. 相似文献

Strepin P-l, a new proteinase inhibitor, is a low molecular weight peptide isolated from the culture fluid of Streptomyces tanabeensis (SAB-934). Strepin P-l strongly inhibited not only cysteine proteinases, calpain and papain, but also trypsin. The purification procedures included HP-20 adsorption chromatography, DEAE-cellulose, Amberlite CG-50, Sephadex LH-20 and G-25 column chromatography. The yield was 12mg from 8 liters of culture fluid. The proteinase inhibitor thus prepared was a peptide composed of tyrosine, valine and argininal, that reacted positively with Sakaguchi and Pauly reagents on TLC. The N-terminal amino acid, tyrosine, was blocked with an isovaleryl group and the structure was elucidated to be AMsovaleryl-tyrosyl-valyl-argininal. The amino acid sequence-inhibitory activity relationships of strepin P-l, leupeptin and antipain toward calpain and papain are also discussed. 相似文献

Respiratory Chain of a Pathogenic Fungus, Microsporum gypseum: Effect of the Antifungal Agent Pyrrolnitrin 下载免费PDF全文
Pyrrolnitrin has been reported to inhibit Bacillus megaterium primarily by forming complexes with phospholipids and to block electron transfer of Saccharomyces cerevisiae between succinate or reduced nicotinamide adenine dinucleotide (NADH) and coenzyme Q. We found that pyrrolnitrin inhibited respiration of conidia of Microsporum gypseum. In mitochondrial preparations, pyrrolnitrin strongly inhibited respiration and the rotenone-sensitive NADH-cytochrome c reductase. The rotenone-insensitive NADH-cytochrome c reductase, the succinate-cytochrome c reductase, and the reduction of dichlorophenolindophenol by either NADH or succinate were inhibited to a lesser extent. However, the activity of cytochrome oxidase was not affected by pyrrolnitrin. The extent of reduction of flavoproteins by NADH and succinate, measured at 465 - 510 nm, was unaltered; however, the reduction of cytochrome b, measured at 560 - 575 nm, was partially inhibited by pyrrolnitrin. The level of totally reduced cytochrome b was restored with antimycin A. We, therefore, concluded that the primary site of action of this antifungal antibiotic is to block electron transfer between the flavoprotein of the NADH-dehydrogenase and cytochrome b segment of the respiratory chain of M. gypseum. 相似文献

Along the coastal belt of Makran, on the southeastern margin of Iran, there are several active and inactive mud volcanoes with different morphologies. Ain—an actively bubbling eye-shaped mud pool—is one of these unique geological phenomena. Present study indicates the first overview of the bacterial diversity of this mud volcano obtained by culture techniques. For this purpose, two samples were collected at two different depths of the mud pool and were cultivated on two different nutrient culture media. A total of 13 isolates were randomly chosen for further phenotypic and genotypic characterization. Growth of the isolates occurred at 25–42°C, pH 8–10 and 0–10% NaCl, indicating that most of them were haloalkaliphiles. Phylogenetic analysis based on 16S rRNA gene sequences indicated that the bacterial isolates belonged to three major taxa: Gammaproteobacteria (genera: Marinobacter, Nitrincola, Stentrophomonas), Actinobactera (genera: Kocuria, Brevibacterium, Dietzia) and Firmicutes (genus Planomicrobium). Gammaproteobacteria were the most abundant, following Actinobacteria and Firmicutes. Most of the strains isolated in the present study had the ability to produce extracellular hydrolytic enzymes such as lipases, amylases, proteases and DNases, making them important biotechnologically. 相似文献

从病死老虎肺脏中分离到1株肠球菌,并对该菌做了生理生化鉴定、药敏试验,致病性试验。本菌对多种抗生素高度耐药,对小白鼠有强致病性,其LD50为2.7×109.2cfu。并用PCR方法扩增分离菌株16S rDNA基因,获得1 415 bp片段,该片段核苷酸序列提交GenBank,登陆号为HM346186,将分离株的16S rDNA核苷酸序列与GenBank上其他肠球菌进行同源性分析。结果表明,分离株的16S rDNA核苷酸序列与肠球菌(EU285587)的同源性为100%,因此该分离菌株被鉴定为致病性肠球菌,命名为YN-1株(云南-1株)。 相似文献

In this study, bronchoalveolar lavage (BAL) specimens from 247 immunocompromised patients were investigated for the incidence of opportunistic fungi. In the direct examination and culture of the specimens, 5 (2.02%) of filamentous fungi and 55 (22.26%) yeasts were isolated and identified as follows: Aspergillus fumigatus (2), A. terreus (1), A. nidulans (1), Mucorsp. (1), Candida albicans (29), C. glabrata (3), C. Parapsilosis (1), Trichosporon beigelii (1), Candida sp. (13) and unknown yeasts (8). 为明确深圳某海马养殖基地线纹海马(Hippocampus erectus)突发性死亡病因,本研究从患病死亡线纹海马病灶组织分离到一株优势菌SZVA20190621,随后对分离菌的形态特征、生理生化特性、回归感染、16S rRNA基因进化及药物敏感性进行研究.结果显示,该菌葡萄糖产气、甘露醇、覃糖、蔗糖、氧化酶活性和MR等生理生化鉴定为阳性;与NCBI上溶藻弧菌(Vibrio alginolyticus)KT986151具有98.16%的同源性;回归感染实验表明菌株SZVA20190621对线纹海马具有强致病性,LD50为3.73×105CFU/mL.综上结果,鉴定该菌为致病性溶藻弧菌.药敏结果显示,该菌株对头孢唑林、头孢噻吩、哌拉西林、大观霉素、阿米卡星、克拉霉素、克林霉素及呋喃妥因耐药;对氨曲南、头孢西丁、头孢呋辛、头孢曲松、卡那霉素、红霉素中度敏感;对头孢他啶、头孢噻肟、头孢哌酮、头孢吡肟、链霉素、妥布霉素、庆大霉素、复方新诺明、氯霉素等17种抗生素高度敏感.本研究为线纹海马养殖中溶藻弧菌病的防控提供一定的参考依据. 相似文献

Escherichia coli serotype O157:H7 was detected among bacteria collected from the Ganges River. O157:H7 isolates tested positive for stx1, stx2, and eae gene sequences. A method is reported for the isolation of nuclei from the dermatophytic fungus,Microsporum gypseum. The yield ranged from greater than 90% in crude preparations to less than 20% in highly purified preparations as judged by the recovery of DNA. Nuclei isolated in this manner appeared intact in electron microscopic observations and possessed chemical compositions, i.e. low protein/DNA and RNA/DNA ratios, which indicated a low degree of cytoplasmic contamination. 相似文献

目的:建立一种从小鼠表皮组织提取高质量RNA的方法。方法:用热击法分离小鼠表皮,用TRIzol法提取RNA,用紫外分光光度计测定RNA的产率和纯度,用琼脂糖电泳和RT-PCR检测RNA的质量和完整性。结果:采用新方法提取的小鼠表皮总RNA,其D260nm/D280nm值为1.8~2.0,大于1.5,且RNA产率高于100μg/g;琼脂糖电泳出现5S、18S和28S等3条清晰的rRNA条带,而且28SrRNA条带的亮度约为18S的2倍;用新方法制备的总RNA可成功地用于RT-PCR实验。结论:采用热击法分离表皮并结合TRIzol法可提取到高质量、完整性好的小鼠表皮总RNA,并能用于相关的分子生物学实验。 相似文献
