The relationship between number of holes in a grain probe trap body and capture of stored-grain pests was determined in laboratory tests using adults of the rice weevil, Sitophilus oryzae (L.), the sawtoothed grain beetle, Oryzaephilus surinamensis (L.), and the red flour beetle, Tribolium castaneum (Herbst). Polyvinylchloride (PVC) probe bodies were attached to electronic sensor heads, and insect captures were recorded electronically using an Electronic Grain Probe Insect Counter (EGPIC) system. In comparisons among PVC probe trap bodies with 60-492 holes, tested at 71 insects per kg in 2.8 kg of soft wheat in cylindrical mini-silos, sawtoothed grain beetle and rice weevil captures were directly related to number of holes in the probe trap body, but there was no relationship for red flour beetle capture. Subsequent tests were conducted comparing sawtoothed grain beetle and rice weevil captures in a PVC probe body with 210 holes over a 40-cm long trapping surface with two commercially available probe traps, a polycarbonate (Lexan) probe trap with 180 holes over a 14-cm long trapping surface and a polyethylene (WBII) probe trap with 750 holes over a 34-cm long trapping surface. The highest percentage capture of both species was in the WBII probe trap, but the 210-hole PVC probe body was as effective as the Lexan probe body for rice weevils and sawtoothed grain beetles at 71 and 17 insects per kg of wheat, respectively.  相似文献

An environmental enrichment device was designed and evaluated for captive chimpanzees. The puzzleboard consisted of a 61-cm, square piece of clear plastic with holes of various sizes drilled into the center. The puzzleboard was bolted to the roof of a chimpanzee enclosure. Flavored primate treats were arranged on top of the board, and the chimpanzee used a finger to manipulate the food across the board to be retrieved through the larger holes. The behavior and puzzle use of 29 chimpanzees housed in eight groups were evaluated while the device was empty and while filled with food. For each group, four 1-hr scan samples were recorded for each condition. Data showed that an average of 40 min elapsed from the time the puzzleboard was filled to the last recorded contact by a subject. Mean individual use ranged from 4.8 to 23.8 min/hr. Female chimpanzees used the puzzle significantly more often than did males. Aggressive, affiliative, inactive, and self-directed behaviors were significantly reduced when the puzzle was filled. In addition, the puzzleboard was inexpensive, sturdy, and easily constructed, mounted, and operated.  相似文献

The purpose of the present investigation was to examine strength performance of 6 common resistance training exercises using free weight bars of different thickness. Eleven resistance-trained men (8.2 +/- 2.6 years of experience; age: 22.1 +/- 1.6 years; body mass: 90.5 +/- 8.9 kg) underwent 1 repetition maximum (1RM) strength testing on 6 occasions in random order for the deadlift, bent-over row, upright row, bench press, seated shoulder press, and arm curl exercises under 3 conditions using: (a) a standard Olympic bar (OL), (b) a 2-inch thick bar (5.08 cm grip span), and (c) a 3-inch thick bar (7.62 cm grip span). Significant (p < 0.05) interactions were observed for the "pulling" exercises. For the deadlift and bent-over row, highest 1RM values were obtained with OL, followed by the 2- and 3-inch bar. Significant 1RM performance decrements for the 2- and 3-inch bars were approximately 28.3 and 55.0%, respectively, for the deadlift; decrements for the 2- and 3-inch bars were approximately 8.9 and 37.3%, respectively, for the bent-over row. For the upright row and arm curl, similar 1RMs were obtained for OL and the 2-inch bar. However, a significant performance reduction was observed using the 3-inch bar (approximately 26.1% for the upright row and 17.6% for the arm curl). The reductions in 1RM loads correlated significantly to hand size and maximal isometric grip strength (r = -0.55 to -0.73). No differences were observed between bars for the bench press or shoulder press. In conclusion, the use of 2- and 3-inch thick bars may result in initial weight reductions primarily for pulling exercises presumably due to greater reliance on maximal grip strength and larger hand size.  相似文献

The relationship between decreasing spermatogenic activity and fertility, pregnancy outcome and the progeny is poorly understood. To study this relationship a model where testosterone is given by a sustained release device is used. Adult male Sprague-Dawley rats received empty or testosterone-filled implants measuring 0.5, 1.0, 2.0, 3.0, 4.0 and 8.0 cm. On Day 90 and again on Day 104 each male was exposed to two females in proestrus. Twenty days later the females were killed. Corpora lutea, implantation sites, resorptions and live normal and abnormal fetuses were counted. Sperm counts in the caput-corpus region of the epididymis in the 3.0-, 4.0- and 8.0-cm testosterone treatment groups were 12.6%, 3.0% and 29.9% of control, while those in the caudal region were 19.8%, 4.0% and 50.8% of control, respectively. The number of females with spermatozoa in the vagina after breeding was significantly diminished only in animals treated with the 4.0-cm testosterone implants (control, 95.8%; 4.0-cm, 50%) while the number of pregnant females per sperm-positive females was markedly reduced in the females mated with both the 3.0-cm and 4.0-cm testosterone implants (control, 82.6%; 3.0-cm, 10.0%; 4.0-cm, 7.7%). There was no effect on the numbers of corpora lutea, on the incidence of pre- or post-implantation loss, malformations, or on the numbers of pups/litter. Individual animals with a decrease in caudal epididymal spermatozoal reserves to less than 5 million, however, are infertile. A decrease in epididymal spermatozoal reserves mediated by testosterone does not cause an increase in teratogenicity in the resultant progeny.  相似文献

Microscopic characterization of charge carriers in solar cells is useful for high‐performance cell fabrication because the formation and accumulation of charges in cells greatly affect the device performance. Electron spin resonance (ESR) is suitable for such characterization because it can directly observe charge carriers with spins in these cells. In this work, the ESR method is applied to organic thin‐film solar cells to investigate charge formation in such devices. Heterojunction cells of indium tin oxide (ITO)/poly(3,4‐ethylenedioxythiophene):poly(4‐styrenesulfonate) (PEDOT:PSS)/pentacene/C60/bathocuproine (BCP)/Al are investigated. Clear ESR signals are observed by inserting a typical PEDOT:PSS hole buffer layer. From analysis of the dependence of the ESR characteristics on the external magnetic field direction, the bias voltage, and the duration of solar‐simulated irradiation, the charges (mobile holes) in pentacene layers are successfully identified and it can be deduced that these holes are formed at the PEDOT:PSS/pentacene interface during device fabrication. This ESR analysis provides useful knowledge for understanding device operation and improving device performance at the microscopic level.  相似文献

The preparation of ultrathin sections of high quality with a Porter-Blum microtome requires that the drive wheel be rotated at a slow, uniform rate during actual cutting. A mechanical device to control the motion of the microtome drive wheel consists of a synchronous motor connected to a drive shaft (4 rev/min), which can be intermittently coupled to the rim of the microtome wheel by a friction drive of 1/2-inch diameter. The microtome drive wheel is rotated manually at a rapid rate until the specimen is about to come into contact with the knife; then the mechanical drive is engaged for slow movement during cutting. When the section has been cut, the drive is disengaged by a lever operated with the left hand; after which, the microtome wheel is turned quickly back to the cutting position with the right hand.  相似文献

A new drilling device for the correct localization of drilling holes has been developed for the fixation of cruciate ligament prostheses. We describe how the device is employed under surgical operating conditions. Its accuracy in application is demonstrated in a cadaver knee. The device has also been used clinically with success. The aim of this study was to develop spheronized microparticulates as a drug delivery system using the 1-step closed rotor disk fluid-bed technology, and to scale up the batch spheronization process. Ibuprofen was used as the model drug and microcrystalline cellulose/sodium carboxymethyl cellulose hydrocolloid (Avicel(R) RC-581 or CL-611) was present as the diluent/binder. The mixture, in 1:1 ratio, was blended with and without 1% sodium lauryl sulfate (SLS) and spheronized with the rotor disk insert, using either water or hydroxypropylmethyl cellulose (HPMC) as binder. Fluid-bed machines (Vector/Freund Flo-Coater model) FLM-1 (with 9-inch rotor insert for 0.75 kg) and FLM-15 (with a 12-inch and 19-inch rotor inserts for 1 kg and 5, 10 kg, respectively) were used. The critical process parameters included inlet air temperature, rotor disk speed and configuration, air flow, and rate of binder application. The 1 kg batch containing SLS that was made with 12-inch smooth stainless steel or waffle teflon plates rotating at 500 rpm had desirable characteristics. The sphericity values were 0.88 and 0.91, with percent yield of 85.4 and 91.2 and drug content values of 94.47% and 91.44%, respectively. The spheroids showed good flow properties with respective rapid drug release (Q20 = 83.27 and 91.75). No difference was seen in the Avicel RC-581 and CL-611. Based on the 1 kg data, Avicel RC-581 and smooth stainless steel and waffle teflon plates (12 inch and 19 inch), the batch was scaled up to 5 and 10 kg. The scale-up parameters included rotor speed (124 -300 rpm) and spray rate (90-140 g/min). The scale-up batches had similar flow characteristics, release rate, and size distribution. The geometric mean diameter increased as batch size increased, and slightly bigger spheroids were obtained using the waffle teflon plate. Ibuprofen spheres with very good physical characteristics were developed using the rotor disk fluid-bed technology, a 1-step closed process that did not require additional unit processes.  相似文献

The mechanical properties of certain bone screws have been examined. The torque developed during insertion of 4 mm (5/32″) non-self-tapping and self-tapping screws and their holding power in bone have been measured. Screws should be inserted into tapped holes using a torque limiting device if failures of screws at insertion are to be prevented. A simple device has been developed for the simultaneous cooling of up to 9120 cm2 of allograft skin in a flat package format. The device, named an insulated alternating-offset heat sink device, is composed of a stack of interleaved layers of 2.0-mm-thick packets of skin and 3.18-mm-thick aluminum heat sinks (each 33.0 cm long by 22.9 cm wide). Four skin packets are placed in a single layer on each heat sink plate, and the number of plates can be varied to accommodate different numbers of skin packets. Every heat sink protrudes 6.3 cm of its 33.0-cm length beyond the skin packets to make a fin for heat convection, but adjacent plates alternate the direction of their fin protrusion so that the layers of plates alternate in their 6.3-cm offset. Insulation layers of 2.54-cm-thick expanded polystyrene are placed on the exposed surfaces of the top and bottom heat sinks in the stack, and the stack is held together by rubber bands. The device is cooled in a -70 degrees C mechanical refrigerator. Maximal cooling rates of -1.8 degrees C min-1 are obtained for both 6- and 11-plate devices, and -3.0 degrees C min-1 for a 2-plate device. The exothermic temperature plateaus associated with skin cooled in these devices are 1.5-1.8 min in duration. Skin cooled by this technique maintains levels of glucose oxidation similar to those associated with skin cooled by liquid nitrogen vapor at a controlled rate of -1 degree C min-1, provided rapid warming is employed after -70 degrees C storage. The development of this device provides a method for the simple, low-cost cryopreservation of the large amounts of allograft skin obtained from a cadaveric donor.  相似文献

Single particle electron cryomicroscopy (cryo-EM) is often performed using EM grids coated with a perforated or holey layer of amorphous carbon. Regular arrays of holes enable efficient cryo-EM data collection and several methods for the production of micropatterned holey-carbon film coated grids have been described. However, a new generation of direct detector device (DDD) electron microscope cameras can benefit from hole diameters that are smaller than currently available. Here we extend a previously proposed method involving soft lithography with a poly(dimethylsiloxane) (PDMS) stamp for the production of holey-carbon film coated EM grids. By incorporating electron-beam (e-beam) lithography and modifying the procedure, we are able to produce low-cost high-quality holey-carbon film coated EM grids with ∼500 nm holes spaced 4 μm apart centre-to-centre. We demonstrate that these grids can be used for cryo-EM. Furthermore, we show that by applying image shifts to obtain movies of the carbon regions beside the holes after imaging the holes, the contrast transfer function (CTF) parameters needed for calculation of high-resolution cryo-EM maps with a DDD can be obtained efficiently.  相似文献

The mucosa of the small and large intestine of goat fetuses exhibited villi which had disappeared after the 32.5-cm curved crown rump (CVR) stage. At places, the stratified epithelial lining persisted among the normal columnar epithelium with goblet cells. The concentration of goblet cells increased with age, while the thickness of the epithelium decreased. The crypts of Lieberkühn were tortuous at the base. Brunner's glands appeared at the 14.2-cm CVR stage. Peyer's patches appeared at the 24.5-cm CVR stage in the ileum. The muscularis mucosae differentiated in the large intestine in group II (16.2- to 24.5-cm CVR length) and progressed caudocranially. The striated border of the intestinal epithelium presented with alkaline phosphatase activity; this border and the goblet cells also stained for mucin. Glycogen was demonstrable in the epithelium with greater concentrations in the duodenum and jejunum in group I (11.5- to 14.6-cm CVR length), and in the ileum and large intestine in group III (30.8- to 39.5-cm CVR length).  相似文献

This study assessed the efficacy of using visual and auditory biofeedback while immersed in a tridimensional videogame to practice a stress management skill (tactical breathing). All 41 participants were soldiers who had previously received basic stress management training and first aid training in combat. On the first day, they received a 15-minute refresher briefing and were randomly assigned to either: (a) no additional stress management training (SMT) for three days, or (b) 30-minute sessions (one per day for three days) of biofeedback-assisted SMT while immersed in a horror/first-person shooter game. The training was performed in a dark and enclosed environment using a 50-inch television with active stereoscopic display and loudspeakers. On the last day, all participants underwent a live simulated ambush with an improvised explosive device, where they had to provide first aid to a wounded soldier. Stress levels were measured with salivary cortisol collected when waking-up, before and after the live simulation. Stress was also measured with heart rate at baseline, during an apprehension phase, and during the live simulation. Repeated-measure ANOVAs and ANCOVAs confirmed that practicing SMT was effective in reducing stress. Results are discussed in terms of the advantages of the proposed program for military personnel and the need to practice SMT.  相似文献

The systematic insertion of thin films of P3HT and PCBM at the electron‐ and hole‐collecting interfaces, respectively, in bulk‐heterojunction polymer solar cells results in different extents of reduction in device characteristics, with the insertion of P3HT at the electron‐collecting interface being less disruptive to the output currents compared to the insertion of PCBM at the hole‐collecting interface. This asymmetry is attributed to differences in the tail state‐assisted charge injection and recombination at the active layer‐electrode interfaces. P3HT exhibits a higher density of tail states compared to PCBM; holes in these tail states can thus easily recombine with electrons at the electron‐collection interface during device operation. This process is subsequently compensated by the injection of holes from the cathode into these tail states, which collectively enables net current flow through the polymer solar cell. The study presented herein thus provides a plausible explanation for why preferential segregation of P3HT to the cathode interface is inconsequential to device characteristics in P3HT:PCBM bulk‐heterojunction solar cells.  相似文献

The bleach continuum in the 1900-1800-cm(-1) region was reported during the photocycle of bacteriorhodopsin (bR) and was assigned to the dissociation of a polarizable proton chain during the proton release step. More recently, a broad band pass filter was used and additional infrared continua have been reported: a bleach at >2700 cm(-1), a bleach in the 2500-2150-cm(-1) region, and an absorptive behavior in the 2100-1800-cm(-1) region. To fully understand the importance of the hydrogen-bonded chains in the mechanism of the proton transport in bR, a detailed study is carried out here. Comparisons are made between the time-resolved Fourier transform infrared spectroscopy experiments on wild-type bR and its E204Q mutant (which has no early proton release), and between the changes in the continua observed in thermally or photothermally heated water (using visible light-absorbing dye) and those observed during the photocycle. The results strongly suggest that, except for the weak bleach in the 1900-1800-cm(-1) region and >2500 cm(-1), there are other infrared continua observed during the bR photocycle, which are inseparable from the changes in the absorption of the solvent water molecules that are photothermally excited via the nonradiative relaxation of the photoexcited retinal chromophore. A possible structure of the hydrogen-bonded system, giving rise to the observed bleach in the 1900-1800-cm(-1) region and the role of the polarizable proton in the proton transport is discussed.  相似文献

A simple and practical 6.8-cm-diameter (36.30-cm(2) cross-sectional-area) preparative disk gel electrophoresis device, based on the design of M. Hayakawa et al. (Anal. Biochem. 288 (2001) 168), in which the elution buffer is driven by an electroosmotic buffer flow through the membrane into the elution chamber from the anode chamber was constructed. We have found that the dialysis membranes employed provide suitable flow rates for the elution buffer, similar to those of an earlier 3.6-cm-diameter device, resulting in the prevention of excess eluate dilution. The efficiency of this device was demonstrated by the fractionation of a bovine serum albumin (BSA) Cohn V fraction into monomer, dimer, and oligomer components using nondenaturing polyacrylamide gel electrophoresis (native-PAGE). The maximum protein concentration of the eluate achieved was 133 mg/ml of BSA monomer, which required a dilution of the eluate for subsequent analytical PAGE performance. As a practical example, the two-dimensional fractionation of soluble dipeptidyl peptidase IV (sDPP IV) from 50 ml fetal bovine serum (3.20 g protein) per gel is presented. The sDPP IV enzyme protein was recovered in a relatively short time, utilizing a 6.5% T native-PAGE and subsequential sodium dodecyl sulfate-PAGE system. This device enhances the possibility of continuous electrophoretic fractionation of complex protein mixtures on a preparative scale.  相似文献

A method is described for the efficient concentration of viruses from large volumes of tap water in relatively short time periods. Virus in acidified tap water in the presence of aluminum chloride is adsorbed to a 10-inch (ca. 25.4 cm) fiberglass depth cartridge and a 10-inch pleated epoxy-fiberglass filter in series at flow rates of up to 37.8 liters/min (10 gallons/min). This filter series is capable of efficiently adsorbing virus from greater than 19,000 liters (5,000 gallons) of treated tap water. Adsorbed viruses are eluted from the filters with glycine buffer (pH 10.5) and the eluate is reconcentrated using an aluminum flocculation process. Viruses are eluted from the aluminum floc with glycine buffer (pH 11.5). Using this procedure, viruses in 1,900 liters (500 gallons) of tap water can be concentrated 100,000-fold in 3 h with an average recovery of 40 to 50%.  相似文献

A simple, low cost device for electrotransformation has been designed and constructed. The cost of the circuit was only $150. Maximum field strength of 12,000 V/cm with an actual time constant up to 11 msec was obtained with a newly designed circuitry and a 0.1-cm electrode gap cuvette. Eschericia coli strains DH1, DH5 alpha, and LE392 were transformed at an efficiency of 10(9)/micrograms DNA with plasmids pUC18 and pBR322. E. faecalis strain OG1X was transformed at an efficiency of 0.9 x 10(5)/micrograms DNA without treatment with glycine.  相似文献
