Background  

Murray's Law states that, when a parent blood vessel branches into daughter vessels, the cube of the radius of the parent vessel is equal to the sum of the cubes of the radii of daughter blood vessels. Murray derived this law by defining a cost function that is the sum of the energy cost of the blood in a vessel and the energy cost of pumping blood through the vessel. The cost is minimized when vessel radii are consistent with Murray's Law. This law has also been derived from the hypothesis that the shear force of moving blood on the inner walls of vessels is constant throughout the vascular system. However, this derivation, like Murray's earlier derivation, is based on the assumption of constant blood flow.  相似文献

Current epidemiological approaches to radon dosimetry yield a dose conversion factor (DCF) of 4 mSv WLM−1 while the dosimetric approaches give a value closer to 13 mSv WLM−1. The present study investigated whether the application of compartment models for the bronchial (BB) and bronchiolar (bb) regions, rather than more anatomically realistic airway tube models, has brought the dosimetric DCF to the higher values. The airway tube model of the tracheo-bronchial tree was used to calculate the effective dose per unit radon exposure. All other elements of the human respiratory tract from the reports of the ICRP or NRC were adopted. A dosimetric derivation of the radon DCF using the airway tube model yielded a value of 14.2 mSv WLM−1. This value is slightly larger than, but not significantly different from, the result obtained through the ICRP 66 approach. It is concluded that utilization of the airway tube model instead of the regional ICRP 66 compartmental model cannot reconcile the gap between dose conversion factors derived from epidemiological and dosimetric approaches.  相似文献

A coalescent argument is used to derive the effective size in simple models with recurrent local extinctions. Several alternative methods of derivation of this result are given and compared to earlier analyses of this problem. The different methods described in this paper all give the same result, which differs from earlier ones. For two published sets of estimates of demographic parameters, metapopulation structure appears to result in a moderate reduction of effective size relative to total adult population size.  相似文献

We examined the distribution of intermediate filaments in early quail embryos in order to determine whether these cytoskeletal proteins play a role in the epithelial-mesenchymal transitions that commonly occur during embryogenesis, e.g., the separation of neural-crest cells from the neural epithelium. The distribution of cytokeratins, vimentin, and desmin was examined in frozen sections of quail embryos at stages during which dramatic reorganizations of tissues take place. All embryonic tissues were found to contain either vimentin or cytokeratins, but the distribution of these cytoskeletal proteins was characteristic neither of the cellular organization (e.g., epithelium vs. mesenchyme) nor of the germ-layer derivation of the tissues. Cytokeratin monoclonal antibodies stained most embryonic epithelia (defined here as being sheet-like tissue with an underlying basement membrane), including epidermis and extraembryonic membranes derived in part from the ectoderm, splanchnopleure and kidney tubules derived from mesoderm, and endoderm. Cytokeratin antibodies did not stain some epithelia, including the neural tube, neural plate, and dermatome/myotome. Whereas the cytokeratin antibodies exclusively stained epithelia, the vimentin antibodies labeled both epithelial (the neural tube, dermatome/myotome, and somatic and splanchnic mesoderm) and mesenchymal tissues (the sclerotome and neural-crest cells), regardless of their germ-layer derivation. In early embryos, antibodies against desmin only stained the myotome and, in 4-day embryos, the heart and mesenchyme around the pharynx. As the distribution of intermediate-filament types did not reflect tissue organization or germ-layer derivation, we propose that the distribution of intermediate filaments in early avian embryos reflects the motile capacity of an embryonic cell and/or the presence of specialized cell junctions, i.e., desmosomes.  相似文献

Recent work by Volz (J Math Biol 56:293–310, 2008) has shown how to calculate the growth and eventual decay of an SIR epidemic on a static random network, assuming infection and recovery each happen at constant rates. This calculation allows us to account for effects due to heterogeneity and finiteness of degree that are neglected in the standard mass-action SIR equations. In this note we offer an alternate derivation which arrives at a simpler—though equivalent—system of governing equations to that of Volz. This new derivation is more closely connected to the underlying physical processes, and the resulting equations are of comparable complexity to the mass-action SIR equations. We further show that earlier derivations of the final size of epidemics on networks can be reproduced using the same approach, thereby providing a common framework for calculating both the dynamics and the final size of an epidemic spreading on a random network. Under appropriate assumptions these equations reduce to the standard SIR equations, and we are able to estimate the magnitude of the error introduced by assuming the SIR equations.  相似文献

We utilize a novel, transgenic cell-labeling system to assess the embryonic derivation of cartilages in the post-metamorphic skull of anuran amphibians. Many of these cartilages form de novo at metamorphosis and have no obvious precursors within the larval skeleton. Most adult cartilages are derived from mandibular- or hyoid-stream neural crest, either individually or in combination; branchial-stream neural crest makes a modest contribution. Each stream also contributes to at least one cartilage in the middle ear or external ear. Four cartilages are composite elements; each is derived from at least two distinct cell populations. Many boundaries between adjacent neural-crest territories are cryptic insofar as they do not coincide with anatomical boundaries. The system of adult cranial segmentation revealed by these fate-mapping results differs in important respects from both the segmentation of the ontogenetically earlier larval skull and the cranial segmentation in amniotes. Most striking is the rostral "inversion" of neural-crest-derived cartilages in Xenopus, such that mandibular stream-derived elements are deployed caudal to those derived from the hyoid stream, which predominate anteriorly. This novel pattern of rostral segmentation may be a consequence of the complex, biphasic life history that is characteristic of most species of living amphibians, and especially anurans, in which cranial architecture is significantly reconfigured at metamorphosis. Neural-crest derivation of the vertebrate skull is not invariant; instead, embryonic derivation of individual components of the cranial skeleton may vary widely among species.  相似文献

Induced pluripotent stem(iPS)cells can be derived from human somatic cells by cellular reprogramming.This technology provides a potential source of non-controversial therapeutic cells for tissue repair,drug discovery,and opportunities for studying the molecular basis of human disease.Normally,mouse embryonic fibroblasts(MEFs)are used as feeder layers in the initial derivation of iPS lines.The purpose of this study was to determine whether SNL fibroblasts can be used to support the growth of human iPS cells reprogrammed from somatic cells using lentivirai expressed reprogramming factors.In our study,iPS cells expressed common pluripotency markers,displayed human embryonic stern cells(hESCs)morphology and unmethylated promoters of NANOG and OCT4.These data demonstrate that SNL feeder cells can support the derivation and maintenance of human iPS cells.  相似文献

A model is presented for the motion of individual molecules of DNA undergoing pulse field gel electrophoresis (PFGE). The molecule is represented by a chain of charged beads connected by entropic springs, and the gel is represented by a segmented tube surrounding the beads. This model differs from earlier reptation/tube models in that the tube is allowed to leak in certain places and the chain can double over and flow out of the side of the tube in kinks. It is found that these kinks often lead to the formation of U shapes, which are a major source of retardation in PFGE. The results of computer simulations using this model are compared with real DNA experimental results for the following cases: steady field motion as seen in fluorescence microscopy, mobility in steady fields, mobility in transverse field alternation gel electrophoresis (TFAGE), mobility in field inversion gel electrophoresis (FIGE), and linear dichroism (LD) of DNA in agarose gels during PFGE. Good agreement between the simulations and the experimental results is obtained.  相似文献

The preservation of non-mineralized tissues in the fossil record is extremely rare. The Lower Devonian Hunsrück Slate of Germany has long been known for the preservation of non-mineralized tissues in pyrite but whether or not these remnants represent true soft tissues has been questioned. This is especially true for struetures visible only on radiographs that are too delicate for excavation by traditional methods. Here we report the discovery of well-preserved pyritized tube feet in six fully prepared specimens of the protasterid brittle starBundenbachia beneckei from the Hunsrück Slate. This discovery represents the first report of fossilized ophiuroid tube feet in the fossil record. The successful excavation of the delicate tube feet was made possible by improved airbrasive techniques developed by German fossil collectors. The relatively large size of the fossil tube feet inBunden-bachia beneckei is consistent with earlier inferences on size based on the presence of large podial basins. Protasterid ophiuroids lack the specialized arm musculature and articulations that provide increased flexibility and strength to the arms of modern ophiuroids with typically reduced tube feet. How-ever, tube foot form and perhaps function inBundenbachia might have been similar to those of living asteroids in which large tube feet are used primarily for locomotion and food-manipulation thus compensating for a lack of specialized arm musculature and articulation. Hence, feeding and life mode of protasterid ophiuroids was not necessarily limited to sedentary, infaunal microphagy as traditionally suggested. Two Hunsrück protasterid ophiuroids,Bundenbachia benecki andPalaeophiomyxa grandis are redescribed and compared.   相似文献

We explore the transmission process for sexually transmitted diseases (STDs). We derive the classical frequency-dependent incidence mechanistically from a pair-formation model, using an approximation that applies to populations with rapid pairing dynamics (such as core groups or non-pair-bonding animals). This mechanistic derivation provides a framework to assess how accurately frequency-dependent incidence portrays the pair-based transmission known to underlie STD dynamics. This accuracy depends strongly on the disease being studied: frequency-dependent formulations are more suitable for chronic less-transmissible infections than for transient highly transmissible infections. Our results thus support earlier proposals to divide STDs into these two functional classes, and we suggest guidelines to help assess under what conditions each class can be appropriately modelled using frequency-dependent incidence. We then extend the derivation to include situations where infected individuals exhibit altered pairing behaviour. For four cases of increasing behavioural complexity, analytic expressions are presented for the generalized frequency-dependent incidence rate, basic reproductive number (R0) and steady-state prevalence (i infinity) of an epidemic. The expression for R0 is identical for all cases, giving refined insights into determinants of invasibility of STDs. Potentially significant effects of infection-induced changes in contact behaviour are illustrated by simulating epidemics of bacterial and viral STDs. We discuss the application of our results to STDs (in humans and animals) and other infectious diseases.  相似文献

We had earlier reported the derivation and characterization of two new sibling human embryonic stem cell lines BJNhem19 and BJNhem20, from discarded grade III embryos of Indian origin. We report here the characteristics of the two sibling cell lines after long-term continuous culture for over 2 yr during which they have been passaged over 200 times. We show that both cell lines adapt well to culture on various mouse and human feeders as well as in feeder-free conditions. The cells show normal diploid karyotype and continue to express all pluripotency markers. Both cell lines differentiate to derivatives of all three germ layers in vitro. However as reported earlier, BJNhem19 is unable to generate teratomas in nude or SCID mice or differentiate to beating cardiomyocytes when tested over several passages during long-term stable culture. On the other hand, the cardiac differentiation capacity of BJNhem20 is greatly increased, and it can generate beating cardiomyocytes that proliferate when isolated and cultured further. In conclusion, the two cell lines have maintained a stable phenotype for over 2 yr and are indeed immortal. Their derivation from grade III embryos does not seem to have any adverse effect on their long-term phenotype. Genetic reprogramming of human somatic cells to induced pluripotent stem cells (iPSCs) could offer replenishable cell sources for transplantation therapies. To fulfill their promises, human iPSCs will ideally be free of exogenous DNA (footprint-free), and be derived and cultured in chemically defined media free of feeder cells. Currently, methods are available to enable efficient derivation of footprint-free human iPSCs. However, each of these methods has its limitations. We have previously derived footprint-free human iPSCs by employing episomal vectors for transgene delivery, but the process was inefficient and required feeder cells. Here, we have greatly improved the episomal reprogramming efficiency using a cocktail containing MEK inhibitor PD0325901, GSK3β inhibitor CHIR99021, TGF-β/Activin/Nodal receptor inhibitor A-83-01, ROCK inhibitor HA-100 and human leukemia inhibitory factor. Moreover, we have successfully established a feeder-free reprogramming condition using chemically defined medium with bFGF and N2B27 supplements and chemically defined human ESC medium mTeSR1 for the derivation of footprint-free human iPSCs. These improvements enabled the routine derivation of footprint-free human iPSCs from skin fibroblasts, adipose tissue-derived cells and cord blood cells. This technology will likely be valuable for the production of clinical-grade human iPSCs.  相似文献

This is expressed by the transition from irregular to genomically programmed morphologies of their calcareous shells that guarantee a stable horizontal resting position. This paper describes the derivation and use of predictive retention coefficients for the reversed-phase high-performance liquid chromatography of peptides. The use of predicted elution positions in the isolation of peptides is illustrated by two examples where peptides, whose existence was postulated from cDNA sequence data, have been successfully isolated. The combination of the powerful chromatographic technology and the ability to predict the elution positions of peptides based on their composition provides a very potent method for the isolation of peptides from biological tissues.  相似文献

A general derivation is presented for the scaling laws governing the size and number of capillary blood vessels in mammals. The derivation is based on the assumption of three idealized similarity principles known to apply, at least approximately, to resting mammals: (i) size-invariant blood pressure; (ii) size-invariant fraction of blood in the capillaries; and (iii) size-invariant oxygen consumption and uptake, per unit of body mass, during each heart cycle. Results indicate that the radius and length of capillaries, and the number that are open and active in the resting state, should scale with mammal mass to the powers 1/12, 5/24 and 5/8, respectively, consistent with earlier work by the author. Measurements are presented supporting the results. Physiological changes accompanying strenuous exercise are accounted for by a change in the scaling law for capillary number, from scaling exponent 5/8 to 3/4.  相似文献
