Background

Resource partitioning is facilitated by adaptations along niche dimensions that range from morphology to behaviour. The exploitation of hidden resources may require specially adapted morphological or sensory tools for resource location and utilisation. Differences in tool diversity and complexity can determine not only how many species can utilize these hidden resources but also how they do so.

Methodology and Principal Findings

The sclerotisation, gross morphology and ultrastructure of the ovipositors of a seven-member community of parasitic wasps comprising of gallers and parasitoids developing within the globular syconia (closed inflorescences) of Ficus racemosa (Moraceae) was investigated. These wasps also differ in their parasitism mode (external versus internal oviposition) and their timing of oviposition into the expanding syconium during its development. The number and diversity of sensilla, as well as ovipositor teeth, increased from internally ovipositing to externally ovipositing species and from gallers to parasitoids. The extent of sclerotisation of the ovipositor tip matched the force required to penetrate the syconium at the time of oviposition of each species. The internally ovipositing pollinator had only one type of sensillum and a single notch on the ovipositor tip. Externally ovipositing species had multiple sensilla types and teeth on their ovipositors. Chemosensilla were most concentrated at ovipositor tips while mechanoreceptors were more widely distributed, facilitating the precise location of hidden hosts in these wasps which lack larval host-seeking behaviour. Ovipositor traits of one parasitoid differed from those of its syntopic galler congeners and clustered with those of parasitoids within a different wasp subfamily. Thus ovipositor tools can show lability based on adaptive necessity, and are not constrained by phylogeny.

Conclusions/Significance

Ovipositor structure mirrored the increasingly complex trophic ecology and requirements for host accessibility in this parasite community. Ovipositor structure could be a useful surrogate for predicting the biology of parasites in other communities.  相似文献

The structure of the ovipositor sclerites and muscles was studied in the tephritid fly Oxyna parietina (Linnaeus, 1758) inducing galls on the stems of Artemisia vulgaris. Adaptations of the ovipositor structure associated with changes of the oviposition substrate due to new larval habits are analyzed. The ovipositor muscles of Oxyna parietina are compared to those of Campiglossa. The genera Oxyna Robineau-Desvoidy, 1830 and Campiglossa Rondani, 1870 belong to the same genus-group of Tephritini, but differ in their host associations and oviposition habits: the larvae of Campiglossa plantaginis (Haliday, 1833) develop in the capitula of Aster tripolium and other asters (Asteraceae). The ovipositor sclerites and muscles are similar in the two genera. Progression of hemolymph into the membranous sheath of the ovipositor and protraction of the aculeus are necessary conditions for laying eggs into the capitula of Asteraceae (Campiglossa plantaginis), as well as for insertion of eggs into the axils of Artemisia leaves (Oxyna parietina). The most significant characters of Oxyna and Campiglossa include the presence of the hyaline apodeme and associated muscles MVM 6 and MVM 7 (instead of the median apodeme in Urophora). Thus, in other tephritid flies similarity in the structure of the ovipositor muscles may reflect similar morphofunctional adaptations to laying eggs into similar substrates, but similarity of Campiglossa and Oxyna in the structure of the ovipositor muscles is due to their close relations.  相似文献

We have studied the restriction fragment length polymorphisms (RFLPs) found in the germline T-cell receptor genes of 25 inbred Mus musculus strains and 8 wild Mus species. Included in the inbred mice tested were several strains which spontaneously develop systemic autoimmune disease. Extensive polymorphism was evident for the variable (V) gene segments of the gene family for both the inbred strains and wild mouse species. Changes in the total number of bands hybridizing with probes for V gene segments suggest that members of a V gene segment subfamily are not closely linked, but are interspersed with members of other subfamilies; that expansion and contraction of the multimembered subfamilies may be an important diversifying factor. Our data obtained with gene probes revealed genomic diversity that is much more limited than that seen for the locus. Analysis of inbred mice with probes for the gene locus revealed some RFLPs, but little evidence of expansion or contraction in the numbers of gene segments. Among the autoimmune mice, NZW, NZB, and BXSB/MpJ all display distinctive differences with gene probes. NZW mice have a large deletion of the gene family, which has been reported previously. We found no differences to distinguish the MRL/MpJ lpr/lpr mice from non-autoimmune strains.  相似文献

The Bcl-2 family proteins plays a central role in apoptosis. The pro- or anti-apoptotic activities of Bcl-2 family are dependent on the Bcl-2 homology (BH) regions. Bcl-rambo, a new pro-apoptotic member, is unusual in that its pro-apoptotic activity is independent of its BH domains. However, the mechanism underlying Bcl-rambo-induced apoptosis is largely unknown. Mitochondrial localization is indispensable for the pro-apoptotic function of Bcl-rambo. Bcl-rambo interacts physically with the adenine nucleotide translocator (ANT), suppresses the ADT/ATP-dependent translocation activity of ANT. Collectively, our data indicate Bcl-rambo is a pro-apoptotic member of the Bcl-2 family, induces the permeability transition via interaction with ANT.

Background

X chromosome inactivation (XCI) is an important gene regulation mechanism in females to equalize the expression levels of X chromosome between two sexes. Generally, one of two X chromosomes in females is randomly chosen to be inactivated. Nonrandom XCI (XCI skewing) is also observed in females, which has been reported to play an important role in many X-linked diseases. However, there is no statistical measure available for the degree of the XCI skewing based on family data in population genetics.

Results

In this article, we propose a statistical approach to measure the degree of the XCI skewing based on family trios, which is represented by a ratio of two genotypic relative risks in females. The point estimate of the ratio is obtained from the maximum likelihood estimates of two genotypic relative risks. When parental genotypes are missing in some family trios, the expectation-conditional-maximization algorithm is adopted to obtain the corresponding maximum likelihood estimates. Further, the confidence interval of the ratio is derived based on the likelihood ratio test. Simulation results show that the likelihood-based confidence interval has an accurate coverage probability under the situations considered. Also, we apply our proposed method to the rheumatoid arthritis data from USA for its practical use, and find out that a locus, rs2238907, may undergo the XCI skewing against the at-risk allele. But this needs to be further confirmed by molecular genetics.

Conclusions

The proposed statistical measure for the skewness of XCI is applicable to complete family trio data or family trio data with some paternal genotypes missing. The likelihood-based confidence interval has an accurate coverage probability under the situations considered. Therefore, our proposed statistical measure is generally recommended in practice for discovering the potential loci which undergo the XCI skewing.

The structure of the ovipositor sclerites and musculature was investigated in Timia erythrocephala Wiedemann, 1824 (Ulidiidae), whose larvae are saprophagous. The ovipositor musculature of this species is compared with that of the previously investigated Tephritidae. Based on morphological analysis, characters common to Ulidiidae and primitive Tephritidae and supposedly plesiomorphic for Tephritidae are revealed. Abdominal segment VII in Timia is compact and consists of tergite and stemite VII fused into a syntergostemite with a visible suture. The preserved suture between the stemite and tergite of segment VII should be regarded as an intermediate state in the process of syntergostemite formation in tephritoid flies, which was completed already in Lenitovena.  相似文献

Background

The serine/threonine mammalian Ste-20 like kinases (MSTs) are key regulators of apoptosis, cellular proliferation as well as polarization. Deregulation of MSTs has been associated with disease progression in prostate and colorectal cancer. The four human MSTs are regulated differently by C-terminal regions flanking the catalytic domains.

Principal Findings

We have determined the crystal structure of kinase domain of MST4 in complex with an ATP-mimetic inhibitor. This is the first structure of an inactive conformation of a member of the MST kinase family. Comparison with active structures of MST3 and MST1 revealed a dimeric association of MST4 suggesting an activation loop exchanged mechanism of MST4 auto-activation. Together with a homology model of MST2 we provide a comparative analysis of the kinase domains for all four members of the human MST family.

Significance

The comparative analysis identified new structural features in the MST ATP binding pocket and has also defined the mechanism for autophosphorylation. Both structural features may be further explored for inhibitors design.

Background

Ca2+-binding proteins are important for the transduction of Ca2+ signals into physiological outcomes. As in calmodulin many of the Ca2+-binding proteins bind Ca2+ through EF-hand motifs. Amongst the large number of EF-hand containing Ca2+-binding proteins are a subfamily expressed in neurons and retinal photoreceptors known as the CaBPs and the related calneuron proteins. These were suggested to be vertebrate specific but exactly which family members are expressed outside of mammalian species had not been examined.

Findings

We have carried out a bioinformatic analysis to determine when members of this family arose and the conserved aspects of the protein family. Sequences of human members of the family obtained from GenBank were used in Blast searches to identify corresponding proteins encoded in other species using searches of non-redundant proteins, genome sequences and mRNA sequences. Sequences were aligned and compared using ClustalW. Some families of Ca2+-binding proteins are known to show a progressive expansion in gene number as organisms increase in complexity. In contrast, the results for CaBPs and calneurons showed that a full complement of CaBPs and calneurons are present in the teleost fish Danio rerio and possibly in cartilaginous fish. These findings suggest that the entire family of genes may have arisen at the same time during vertebrate evolution. Certain members of the family (for example the short form of CaBP1 and calneuron 1) are highly conserved suggesting essential functional roles.

Conclusions

The findings support the designation of the calneurons as a distinct sub-family. While the gene number for CaBPs/calneurons does not increase, a distinctive evolutionary change in these proteins in vertebrates has been an increase in the number of splice variants present in mammals.  相似文献

Background

Alkyl hydroperoxidase activity provides an important antioxidant defense for bacterial cells. The catalytic mechanism requires two peroxidases, AhpC and AhpD, where AhpD plays the role of an essential adaptor protein.

Results

The crystal structure of a putative AhpD from Pseudomonas aeruginosa has been determined at 1.9 Å. The protein has an all-helical fold with a chain topology similar to a known AhpD from Mycobacterium tuberculosis despite a low overall sequence identity of 9%. A conserved two α-helical motif responsible for function is present in both. However, in the P. aeruginosa protein, helices H3, H4 of this motif are located at the N-terminal part of the chain, while in M. tuberculosis AhpD, the corresponding helices H8, H9 are situated at the C-terminus. Residues 24-62 of the putative catalytic region of P. aeruginosa have a higher sequence identity of 33% where the functional activity is supplied by a proton relay system of five residues, Glu36, Cys48, Tyr50, Cys51, and His55, and one structural water molecule. A comparison of five other related hypothetical proteins from various species, assigned to the alkyl hydroperoxidase D-like protein family, shows they contain the same conserved structural motif and catalytic sequence Cys-X-X-Cys. We have shown that AhpD from P. aeruginosa exhibits a weak ability to reduce H2O2 as tested using a ferrous oxidation-xylenol orange (FOX) assay, and this activity is blocked by thiol alkylating reagents.

Conclusion

Thus, this hypothetical protein was assigned to the AhpD-like protein family with peroxidase-related activity. The functional relationship of specific oligomeric structures of AhpD-like structural family is discussed.

This paper describes patterns of annual and diurnal activity in the endangered endemic cyprinid Aulopyge huegeli from Croatia. The species is largely nocturnal. Spawning in the laboratory begins at temperatures of 20°C. Aulopyge huegeli is a rock and gravel brood hider. The female deposits eggs in various fissures with a special ovipositor. The eggs are whitish and have a diameter between 1.5 and 2mm. The zona radiata is relatively thin (3.2–5.5m) and provides the eggs with the necessary plasticity. The surface is covered with an attaching substance and with short attachment extension around the vegetal pole. This correlates with features of the spawning microhabitats. Aulopyge huegeli has a very small micropyle which consists of a pit with a diameter of 7.5m and a canal with a diameter of 2.5m (micropyle type I).  相似文献

Background

The widely expressed Dps proteins, so named after the DNA-binding properties of the first characterized member of the family in Escherichia coli, are considered major players in the bacterial response to stress.

Scope of review

The review describes the distinctive features of the “ferritin-like” ferroxidation reaction, which uses hydrogen peroxide as physiological iron oxidant and therefore permits the concomitant removal of the two reactants that give rise to hydroxyl radicals via Fenton chemistry. It also illustrates the structural elements identified to date that render the interaction of some Dps proteins with DNA possible and outlines briefly the significance of Dps–DNA complex formation and of the Dps interaction with other DNA-binding proteins in relation to the organization of the nucleoid and microbial survival.

General significance

Understanding in molecular terms the distinctive role of Dps proteins in bacterial resistance to general and specific stress conditions.

Major conclusions

The state of the art is that the response to oxidative and peroxide-mediated stress is mediated directly by Dps proteins via their ferritin-like activity. In contrast, the response to other stress conditions derives from the concerted interplay of diverse interactions that Dps proteins may establish with DNA and with other DNA-binding proteins.  相似文献

Relation of the ovipositor structure and the egg-laying habit with the larval biology in the family Scathophagidae was investigated. The ovipositor was examined and illustrated for 29 species including Phrosia albilabris (F.), Cordilura umbrosa (Lw.), Spathephilus breviventris (Lw.), Scathophaga stercoraria (L.), Chaetosa punctipes (Mg.), Pogonota barbata (Ztt.), Cochliarium cuneiventris (Ztt.), and Delina nigrita (Fll.). Species with different bionomics and different larval feeding tissues, i.e., different substrates for oviposition, were distinguished. Comparative morphological analysis of the author’s and literature data on 29 species belonging to 15 genera has shown the constancy of the ovipositor structure within the genera examined. Two principal types of the ovipositor structure were distinguished: (a) the elongate ovipositor with the distal position of the terminal part of the abdomen (the proctiger), this type is characteristic of phytophagous species (the delinine type); and (b) the shortened ovipositor with the proctiger shifted dorsally (the scathophagine type), this type is typical of predatory and saprophagous species. The delinine type of the ovipositor is adapted to inserting eggs into hard plant tissues. Changes in the feeding habits of the larvae were found to be associated with changes in the ovipositor structure. The dependence of the ovipositor structure on the egg-laying habit and on the larval biology enables hypothesizing biological features of those species for which the relevant data are not available yet. The morphogenesis of the ovipositor is speculated based on the data on its structure.  相似文献

Classification and morphological and molecular evidence supporting relationships of Mymarommatidae (Mymarommatoidea) and the 20 families of Chalcidoidea are reviewed. Five autapomorphies support monophyly of Mymarommatoidea, at least two autapomorphies support monophyly of Chalcidoidea, and three synapomorphies support a sister-group relationship between Mymarommatoidea and Chalcidoidea. Mymaridae are indicated as the likely sister group of all other Chalcidoidea by: two features of the ovipositor, the unique structure of a muscle between the mesofurca and axillary lever, and sequence data from the 28s rDNA gene. Structure of the upper valvulae of the ovipositor could indicate Rotoitidae as the second-most basal clade of Chalcidoidea. Chalcididae, Elasmidae, Encyrtidae, Eulophidae, Eurytomidae, Leucospidae, Mymaridae, Ormyridae, Rotoitidae, Signiphoridae, Torymidae and Trichogrammatidae are each indicated as monophyletic by at least one putative synapomorphy, but could render other families paraphyletic. Aphelinidae, Eupelmidae, Pteromalidae, and Tetracampidae are not demonstrably monophyletic. Agaonidae is monophyletic only if restricted to Agaoninae, and Eucharitidae is monophyletic only if restricted to Eucharitinae + Oraseminae. Eupelmidae may be paraphyletic with respect to Tanaostigmatidae and Encyrtidae, and Tanaostigmatidae including Cynipencyrtus may be paraphyletic relative to Encyrtidae. Perilampidae (Perilampinae + Chrysolampinae) are either polyphyletic or paraphyletic with respect to Eucharitidae + Akapalinae + Philomidinae. No cladistic hypotheses of familial relationships based on character evidence have considered the superfamily in its entirety.  相似文献

Background

Genomic DNA frequently undergoes rearrangement of the gene order that can be localized by comparing the two DNA sequences. In mitochondrial genomes different mechanisms are likely at work, at least some of which involve the duplication of sequence around the location of the apparent breakpoints. We hypothesize that these different mechanisms of genome rearrangement leave distinctive sequence footprints. In order to study such effects it is important to locate the breakpoint positions with precision.

Results

We define a partially local sequence alignment problem that assumes that following a rearrangement of a sequence F, two fragments L, and R are produced that may exactly fit together to match F, leave a gap of deleted DNA between L and R, or overlap with each other. We show that this alignment problem can be solved by dynamic programming in cubic space and time. We apply the new method to evaluate rearrangements of animal mitogenomes and find that a surprisingly large fraction of these events involved local sequence duplications.

Conclusions

The partially local sequence alignment method is an effective way to investigate the mechanism of genomic rearrangement events. While applied here only to mitogenomes there is no reason why the method could not be used to also consider rearrangements in nuclear genomes.

This paper aims to describe in detail the mechanism by which eggs are moved along the length of the ovipositor of insects. A series of posteriorly orientated scales, located along the inside of the ovipositor valves, catch the surface of the egg as it emerges from the oviduct and move it along the ovipositor as the valves oscillate back and forth. The morphology of ovipositor scales is examined by scanning electron microscopy, and is compared for 22 species in 20 Families of insects. The mechanism is also confirmed by direct manipulation of the ovipositor of an anaesthetized insect. Ovipositor scales vary in length from 1 to 30 μm and can be spine-like, comb-like or scale-like in structure in different species. Caddis-flies (Trichoptera) were the only group examined which did not possess ovipositor scales. One species, Philanisus plebeius Walker, has longitudinal ridges along the inside surface of its ovipositor valves. The lack of ovipositor scales in the caddisfly species examined in this study is discussed in relation to their behaviour and possible oviposition sites.  相似文献

From the perspective of medical anthropology and comparative research, American oncology appears as a unique variant of international biomedical culture, particularly when contrasted with oncological practice in societies such as Japan and Italy. Based on interviews with 51 oncologists in Harvard teaching hospitals, this paper argues that American oncological practice draws on distinctive cultural meanings associated with hope and is infused with popular notions about the relationship between psyche and soma, the progressive efficacy of biotechnical interventions, truth-telling, and the nature of the physician-patient relationship.  相似文献

Background

Mixed epithelial and stromal tumor (MEST) is a distinctive benign composite neoplasm of the kidney predominantly seen in females mostly in the perimenopausal period. Although these tumors are known to arise from renal pelvis, our case was distinct in that it had no intrapelvic component growing in exophytic fashion.

Case report

A 35 year old female patient presented to us with vague abdominal pain. She had undergone excision of bilateral ovarian cystic masses for cystic teratoma twelve years earlier. A computed tomography scan of abdomen and pelvis showed a 9 × 7 cm uniformly solid mass with poor contrast enhancement situated in the inferomedial aspect of the left kidney. On exploration, the mass was arising from the inferior and anterior aspect of left renal pelvis, and was attached to it with a narrow pedicle. There was no adherence or attachment to the renal parenchyma. The mass was excised preserving the kidney. Microscopically, the tumor was composed of large collagenized areas containing bundles of spindle cells and several 'microcysts' lined by cuboidal epithelium suggestive of a benign mixed epithelial stromal tumor.

Discussion

Mixed epithelial tumors usually present in perimenopausal women as a partially cystic mass. Tumors are composed of irregular mixtures of cystic and solid areas, glands with variable complexity and distribution and the stromal component is characterized by a spindle cell proliferation. Commonly, it arises from the renal parenchyma and pelvis and nephrectomy is advocated to manage these tumors.

Conclusion

MEST is a distinctive benign tumor of the kidney that should be distinguished from other renal neoplasms. MEST arising from the renal pelvis and growing exophytically is a rare entity. The overall prognosis is favorable.

Introduction

Loquat leaf extract (LLE) is commonly used in China for a variety of ailments including diabetes. Several recent reports implicate LLE and a sesquiterpene glycoside, one of its components, as being an anti-hyperglycemic agent. However, the underlying mechanism of action of this anti-hyperglycemic agent has not been reported.

Objective

We have conducted a tracer-based metabolomics study to investigate the effects of sesquiterpene and loquat extract on the balance of flux of central glucose metabolism in HepG2 cells and to compare with those of “insulin sensitizers”, metformin and rosiglitazone.

Methods

Human hepatoma HepG2 cells in confluence culture were incubated in Dulbecco’s modified Eagle’s medium containing 50% [1, 2 13C2]-glucose in the presence of rosiglitazone, metformin, LLE or pure sesquiterpene. Cells were harvested in 48 h. Mass isotopomers of metabolites (glycogen, ribose, deoxyribose, glutamate and palmitate) were determined.

Results

13C labeling in metabolic intermediates were summarized in a mass isotopomer matrix. Treatment with loquat extract/sesquiterpene, metformin and rosiglitazone each produced distinctive mass isotopomer patterns reflecting disparate effects on the contribution of glucose to various metabolites production, and on several metabolic flux ratios. The overall effect of LLE and sesquiterpene on glucose metabolism is clearly different from those of the known “insulin sensitizers”.

Conclusion

Our study demonstrates the utility of isotopomer matrix in summarizing metabolic actions of LLE on the balance of fluxes occurring within the central glucose metabolism in HepG2 cells. 13C carbon tracing (tracer-based metabolomics) is a useful systems biology tool to elucidate glucose metabolic pathways affected by diabetes and its treatment.
