The abnormal pistils widely occur in Japanese apricot (Prunus mume Sieb. et Zucc) and seriously affect the fruit production. In this study, a CCoAOMT homologue, PmCCoAOMT, was cloned in Japanese apricot and the bioinformatics software analyzed the structural characteristics. The PmCCoAOMT protein was detected to be located in the cell cytoplasm by onion transient expression experiment. Analysis of the real-time PCR data showed that PmCCoAOMT gene expressed in the prophase development of pistil and the expression level in ‘Daqiandi’ was higher than ‘Longyan.’ The expression level in ‘Longyan’ was higher than ‘Daqiandi’ in the late period development of pistil, and the expression level of perfect flower (perfect pistil) was higher than imperfect flower (pistil deformity and no pistil). Compared with the control, the over-expression of PmCCoAOMT transgenic tobacco lines showed bigger flowers, darker petals. The lignin monomer composition in transgenic tobacco lines was also measured, and the results showed that transgenic tobacco lines had a higher S (Syringyl)/G (Guaiacyl) ratio (22.3 %) than control lines (11.8 %). Also, the perfect flower buds contained more S/G ratio (92.62 %) than imperfect flower buds (83.55 %) in ‘Daqiandi.’ Our results indicated that the PmCCoAOMT gene might have function in lignin accumulation, which contributed to pistil development in Japanese apricot.  相似文献

The cardiomyopathies are defined as: ‘a myocardial disorder in which the heart is structurally and functionally abnormal, in the absence of coronary artery disease, hypertension and congenital heart disease sufficient to cause the myocardial abnormality.’1 With the advent of molecular cardiology and cardiovascular imaging (including MRI and echocardiography) in the last decade, much insight has been gained into the different clinical presentations, its familial and genetic causes and pathophysiology in explaining left ventricular function and prognosis. doi: 10.1111/j.1741‐2358.2012.00652.x Oral healthcare issues in rural residential aged care services in Victoria, Australia Objectives: To identify major issues in providing and accessing oral health care in Victorian rural residential aged care services from the perspectives of dentists, aged care staff and residents. Methods: Structured interviews were conducted with five dentists, nine aged care staff and six residents. Three focus groups were conducted with aged care staff. These data were thematically analysed independently by two researchers. Results: The challenges reported by dentists included complexity of care, infrastructure needs and need for skill development. Aged care staff reported lack of skills and confidence in providing oral hygiene care, especially in residents with natural teeth, and an increasing burden on their daily workload. Residents reported concern and shame regarding their declining oral health status and increased challenges accessing appropriate oral health care. Conclusion: These findings indicate the need to build and sustain aged care ‘oral health teams’ who are able to provide daily oral hygiene care for residents and mentor other staff. Rural dentists need access to gerodontic training, portable equipment and appropriate workspaces in aged care services. Aged care and oral health services need to establish clear referral and communication pathways.  相似文献

Traits related to root architecture are of great importance for yield performance of crop species, although they remain poorly understood. The present study is aimed at identifying the genomic regions involved in the control of root morphological traits in durum wheat (Triticum durum Desf.). A set of 123 recombinant inbred lines derived from the durum wheat cross of cvs. ‘Creso’ × ‘Pedroso’ were grown hydroponically to two growth stages, and were phenotypically evaluated for a number of root traits. In addition, meta-(M)QTL analysis was performed that considered the results of other root traits studies in wheat, to compare with the ‘Creso’ × ‘Pedroso’ cross and to increase the QTL detection power. Eight quantitative trait loci (QTL) for traits related to root morphology were identified on chromosomes 1A, 1B, 2A, 3A, 6A and 6B in the ‘Creso’ × ‘Pedroso’ segregating population. Twenty-two MQTL that comprised from two to six individual QTL that had widely varying confidence intervals were found on 14 chromosomes. The data from the present study provide a detailed analysis of the genetic basis of morphological root traits in wheat. This study of the ‘Creso’ × ‘Pedroso’ durum-wheat population has revealed some QTL that had not been previously identified.  相似文献

Based on 10 months of fieldwork in the Acholi region of northern Uganda among youth and adults who were forcefully recruited into the Lord’s Resistance Army (LRA) during the war, this article provides qualitative details to research on ‘appetitive aggression.’ Through two case-stories the article unfolds first person articulations of how ‘appetitive aggression’ is experienced as ‘the urge to kill’ and how it relates to the emic Acholi spiritual concept of ‘cen’; a local Luo expression used to describe places and human beings possessed by evil spirits. The analysis illuminates what the individual and social implications of ‘the urge to kill’ and ‘cen’ entail for two Acholi men; first in a militia and then in a civil post-war context. The analysis then relates these findings to soldier experiences across cultures and time periods. While our analysis supports the findings in ‘appetitive aggression’ studies that appetitive aggression serves as a resilient protective factor against developing post-traumatic stress disorder (PTSD), this study documents that once the former forcefully recruited return to civilian life, ‘appetitive aggression’ and ‘the urge to kill’ precipitate individual and at times lethal social and moral complications in a fragile post-war community. Thus, the article argues that appetitive aggression and the emic perceptions and experiences of it among the local population are essential to consider in studies, processes and programs targeting demobilization, rehabilitation, reconciliation and re-integration.  相似文献

Current scholarship in the social sciences and humanities, including the study of religion, shows a marked appraisal of bodily sensations, emotions, and experiences as eminently social and politico‐aesthetic phenomena (rather than reducing them to a matter of mere individual psychology). How to grasp the genesis of shared perceptions and feelings, and even some kind of ‘wow’ effect, in relation to a posited ‘beyond’ has become a central issue for scholars of religion today. Placed against the horizon of the material turn in the study of religion, R.R. Marett's approach to religion as an ‘organic complex of thought, emotion, and behaviour’ and his concept of awe gain renewed topicality. Engaging with Marett's ideas in the context of broader debates about religious experience, in this article (which is based on my 2014 Marett lecture) I call attention to the surplus generated in the interplay of religious things and bodily sensations and explore its role in politics and aesthetics of religious world‐making. My central point is that Marett's work offers valuable resources for an approach to religion that neither takes for granted the existence of a god or transcendental force (as in ontological approaches), nor invests in unmasking it as an illusion (as in critiques of religion as irrational), but instead undertakes a close study of the standardized methods that yield the fabrication of some kind of excess that points to a ‘beyond’ and yet is grounded in the here and now.  相似文献

Sudden cardiac arrest or death (SCA/SCD) in athletes has a low event rate. Pre-participation or eligibility screening is a widely accepted method of primary prevention of SCA/SCD in athletes. Most European countries and international sports governing bodies perform ECG-inclusive screening. However, implementation of a resting 12-lead ECG in pre-participation or eligibility cardiac screening is still a topic of debate. Recently, the ‘International recommendations for electrocardiographic interpretation in athletes’ was published in three leading international medical journals. These international ECG criteria are based on studies with detailed information on resting 12-lead ECG of Caucasian and Afro-Caribbean athletes or on consensus in case evidence was lacking. Normal, borderline and abnormal ECG findings in young athletes (age 12–35 years) are clearly described and illustrated to assist the screening physician in interpreting ECGs of athletes correctly.In this ‘point of view paper’ we will discuss whether these new ECG criteria actually help prevent SCA/SCD in athletes.  相似文献

I argue that the concept ‘physician‐assisted suicide’ covers two procedures that should be distinguished: giving someone access to humane means to end his own life, and taking co‐responsibility for the safe and effective execution of that plan. In the first section I explain the distinction, in the following sections I show why it is important. To begin with I argue that we should expect the laws that permit these two kinds of ‘assistance’ to be different in their justificatory structure. Laws that permit giving access only presuppose that the right to self‐determination implies a right to suicide, but laws that permit doctors to take co‐responsibility may have to appeal to a principle of mercy or beneficence. Actually this difference in justificatory structure can to some extent be found in existing regulatory systems, though far from consistently. Finally I argue that if one recognizes a right to suicide, as Oregon and other American states implicitly do, and as the European Court of Human Rights has recently done explicitly, one is committed to permit the first kind of ‘assistance’ under some conditions.  相似文献

A real-time activity monitoring system within an Android based smartphone is proposed and evaluated. Motion and motionless postures may be classified using principles of kinematical theory, which underpins hierarchical rule-based algorithms, based on accelerometer and orientation data. Falls detection was implemented by analyzing whether the postures classified as ‘lying’ or ‘sit-tilted’ posture are deemed normal or abnormal, based on the analysis of time, users’ current position and posture transition. Experimental results demonstrate that the approach can detect various types of falls efficiently (i.e., in real-time within a smart phone processor) and also correctly (95 % and 93 % true positives for falls ending with ‘lying’ and ‘sit-tilted’ respectively). The approach is reliable for different subjects and different situations, since it is not only based on empirical thresholds and subject-based training models, but in addition it is underpinned by theory.  相似文献

The development of single nucleotide polymorphism (SNP) markers in Japanese pear (Pyrus pyrifolia Nakai) offers the opportunity to use DNA markers for marker-assisted selection in breeding programs because of their high abundance, codominant inheritance, and potential for automated high-throughput analysis. We developed a 1,536-SNP bead array without a reference genome sequence from more than 44,000 base changes on the basis of a large-scale expressed sequence tag (EST) analysis combined with 454 genome sequencing data of Japanese pear ‘Housui’. Among the 1,536 SNPs on the array, 756 SNPs were genotyped, and 609 SNP loci were mapped to linkage groups on a genetic linkage map of ‘Housui’, based on progeny of an interspecific cross between European pear (Pyrus communis L.) ‘Bartlett’ and ‘Housui’. The newly constructed genetic linkage map consists of 951 loci, comprising 609 new SNPs, 110 pear genomic simple sequence repeats (SSRs), 25 pear EST–SSRs, 127 apple SSRs, 61 pear SNPs identified by the “potential intron polymorphism” method, and 19 other loci. The map covers 22 linkage groups spanning 1341.9 cM with an average distance of 1.41 cM between markers and is anchored to reference genetic linkage maps of European pears and apples. A total of 514 contigs containing mapped SNP loci showed significant similarity to known proteins by functional annotation analysis.  相似文献

Despite ‘abnormal’ motion being considered a risk factor for low back injury, the current understanding of ‘normal’ spine motion is limited. Identifying normal motion within an individual is complicated by the considerable variation in movement patterns amongst healthy individuals. Therefore, the purpose of this study was to characterize sources of variation in spine motion among a sample of healthy participants. The second objective of this study was to develop a multivariate model capable of predicting an expected movement pattern for an individual. The kinematic shape of the lower thoracic and lumbar spine was recorded during a constrained dynamic trunk flexion movement; as this is not a normal everyday movement task, movements are considered ‘typical’ and ‘atypical’ for this task rather than ‘normal’ and ‘abnormal’. Variations in neutral standing posture accounted for 85% of the variation in spine motion throughout the task. Differences in total spine range of flexion and a regional re-weighting of range of motion between lower thoracic and lumbar regions explained a further 9% of the variance among individuals. The analysis also highlighted a difference in temporal sequencing of motion between lower thoracic and lumbar regions which explained 2% of the total movement variation. These identified sources of variation were used to select independent variables for a multivariate linear model capable of predicting an individuals’ expected movement pattern. This was done as a proof-of-concept to demonstrate how the error between predicted and observed motion patterns could be used to differentiate between ‘typical’ and ‘atypical’ movement strategies.  相似文献

A method of making three-dimensional molds of the vagina with the materials used by dentists for oral impressions gives considerably more information about normal and abnormal anatomic features than other methods of study, including direct viewing, palpation and contrast x-ray studies. The molds are of value in the demonstration of anatomic patterns in relation to normal and abnormal functions, in evaluation of surgical and other therapeutic techniques, in teaching, and in explaining the abnormalities and the aims of treatment to patients.The use of such molds is being applied to the study of pelvic prolapse, urinary stress incontinence, sexual function of the vagina and birth injuries.  相似文献

This article explores how carpentry apprentices learn to handle uncertain and potentially dangerous situations in the working environment at construction sites. The analysis focuses on how these situations are negotiated at the workplaces in relation to bodily sensations of pain, relations with their co-workers and the importance placed on ‘getting the work done’. It shows that instead of working to eliminate potentially dangerous situations through the use of precautionary measures such as lifting equipment or personal protection gear, the apprentices learn to understand risk and uncertainties as a part of the job through general learning processes at the workplace.  相似文献

Translucent tissue defect (TTD) is an undesirable postharvest disorder of potato tubers characterized by the development of random pockets of semi-transparent tissue containing high concentrations of reducing sugars. Translucent areas turn dark during frying due to the Maillard reaction. The newly released cultivar, Premier Russet, is highly resistant to low temperature sweetening, but susceptible to TTD. Symptoms appeared as early as 170 days after harvest and worsened with time in storage (4–9 °C, 95 % RH). In addition to higher concentrations of glucose, fructose and sucrose, TTD resulted in lower dry matter, higher specific activities of starch phosphorylase and glc-6-phosphate dehydrogenase, higher protease activity, loss of protein, and increased concentrations of free amino acids (esp. asparagine and glutamine). The mechanism of TTD is unknown; however, the disorder has similarities with the irreversible senescent sweetening that occurs in tubers during long-term storage, where much of the decline in quality is a consequence of progressive increases in oxidative stress with advancing age. The respiration rate of non-TTD ‘Premier Russet’ tubers was inherently higher (ca. 40 %) than that of ‘Russet Burbank’ tubers (a non-TTD cultivar). Moreover, translucent tissue from ‘Premier Russet’ tubers had a 1.9-fold higher respiration rate than the average of non-translucent tissue and tissue from non-TTD tubers. Peroxidation of membrane lipids during TTD development resulted in increased levels of malondialdehyde and likely contributed to a measurable increase in membrane permeability. Superoxide dismutase and catalase activities and the ratio of oxidized to total glutathione were substantially higher in translucent tissue. TTD tubers also contained twofold less ascorbate than non-TTD tubers. TTD appears to be a consequence of oxidative stress associated with accelerated aging of ‘Premier Russet’ tubers.  相似文献

The presence, diversity and abundance of non-native plant species in natural vegetation are common condition indicators used to determine conservation status, with consequences for management strategies and investment. The rationale behind non-native species metrics as condition indicators is the assumption that non-natives have negative consequences on native biodiversity and habitat condition. The case against non-native species is not so clear-cut, with some studies reporting neutral or even facilitative interactions, often depending on spatial scale. Observational and experimental evaluations of the impact of particular non-native species on biodiversity provide a vital evidence-base for general conservation management strategies. Unintentionally though, many studies that quantify the impacts of non-native species have resulted in a publication bias in which species with known impacts are selected for investigation far more often than benign species. Here we argue that meta-analyses of the impacts of individual non-native species on natives, no matter how meticulous or objective, should not be generalized beyond the set of ‘training’ species. The likelihood of such extrapolation is increased when meta-analyses are reported with little qualification as to the skewed sampling towards problematic species, and because alternative findings such as non-native assemblages having positive interactions with native biodiversity, are under-reported. To illustrate, we discuss two meta-analyses that make general conclusions from impact studies skewed towards ‘transformers’, the most extreme invaders. We warn that if generic non-native species management strategies were to be based on these conclusions, they could not only fail to meet objectives but in some instances harm native biodiversity.  相似文献

This article examines the role co-ethnic youth basketball leagues play in shaping ethnic community among third- and fourth-generation Japanese American youth. With dwindling rates of Japanese immigration, increased rates of out-marriage, and fewer cultural hubs available, finding a thriving ethnic community has become a particular challenge for later-generation Japanese Americans. Drawing from ethnographic data, I argue that even among highly ‘assimilated’ Japanese Americans, youth basketball leagues serve as an active space for constructing and preserving ethnic community and social networks. I demonstrate how through social, cultural, and spatial interactions facilitated by sports, some Japanese Americans have found a sense of ethnic ‘connectedness’ within basketball leagues while performing their own renditions of ethnic identity that is simultaneously attentive to fluid local and global connections. These findings illuminate the role and importance of sport through which Japanese Americans challenges the racial contours of American-ness and belonging while claiming their place, locally, nationally, and transnationally.  相似文献

Objectives: The purpose of this study was to use the oral health impact profile (OHIP‐14) to evaluate the impact of oral disease on the quality of life of a group of independently‐living elderly persons in an urban area of Japan. Subjects: A total of 1244 participants of the Senior Citizen's College, who attended the lectures once a week. They were community‐dwelling, independently‐living people over 60 years of age. Measurements: Japanese version of the short‐form OHIP‐14. Results: Internal reliability for the 14 items overall was very high (Cronbach's α = 0.95). Report of ‘painful aching’ and ‘uncomfortable to eat’ were the two most highly scored items using the mean sum OHIP‐14 score. A multiple logistic regression analysis indicated that the sum OHIP‐14 score had significant associations with self‐assessment of general health, dental status, and a perceived need for dental treatment. However, age, gender, dissatisfaction with financial status or education level was not significantly associated with the sum OHIP‐14. Compared with that of other countries, the items were ranked similarly, whereas the perceived magnitudes of the problems were quite different from other population. Conclusions: The OHIP‐14 in Japanese had a high internal reliability, was significantly associated with dental status and comparable ranking for items when compared with studies from other countries.  相似文献

This paper is part of an investigation dedicated to indoor pollen from a forensic point of view. Previous studies documented the ‘natural’ pollen content of a flat and how it was ‘artificially’ altered by ornamental plants. The present study investigated if this ‘artificial pollen trace’ can be transferred to shoes. Brand new sneakers were worn in the flat and afterwards examined for pollen on their soles. The results showed that the shoes gathered pollen and that some pollen types correlated to those found in the flat. Afterwards, some of these shoes were worn while walking through the city for approximately 1.8 km. On these shoes, flat pollen was also detectable, but in low abundances. This study shows that, from a palynological point of view, shoes are worth a look at when a crime is investigated. Uncommon pollen grains on shoes can point to an indoor location with the corresponding ornamental plants.  相似文献
